To investigate the effect of dietary folic acid on fish flesh quality, muscle antioxidant status and the potential mechanism, young grass carp (Ctenopharyngodon idella) were fed diets containing 0.10–3.12 mg/kg diet of folic acid for 8 weeks. The results showed that optimum folic acid increased muscle contents of protein, lipid, pH, shear force, amino acids, unsaturated fatty acids, hydroxyproline and glutathione, whereas contents of moisture, cooking loss, lactate and saturated fatty acids and cathepsin L activity showed an opposite trend (p < .05). Moreover, optimum folic acid elevated antioxidant enzyme activities and mRNA levels, as well as NF‐E2‐related factor 2 and casein kinase 2 mRNA levels (p < .05). However, optimum folic acid decreased reactive oxygen species (ROS) content and Kelch‐like ECH‐associated protein 1a (Keap1a) and Keap1b mRNA levels (p < .05). Interestingly, excess folic acid induced negative effects on above‐mentioned parameters (p < .05). Summarily, this study indicated that optimum folic acid improved fish flesh quality and muscle antioxidant system associated with Nrf2‐Keap1 pathway. Based on muscle cathepsin L activity and ROS content, the folic acid requirements for young grass carp were 1.87 and 1.80 mg/kg diet, respectively, regarding to the flesh quality and antioxidant status.  相似文献

The objective of this study was to compare the effects of four astaxanthin preparations with different ratio of E/Z (trans/cis) isomers and different coating materials on the pigmentation and antioxidation properties of rainbow trout. Five diets were designed as basal diet (without astaxanthin supplementation) and four astaxanthin diets (100 mg/kg diet) supplemented with four astaxanthin products, BASF (79% all‐E, gelatin coated), Wisdom‐B (85% all‐E, gelatin coated), Wisdom‐C (94% all‐E, carrageenan coated), and Wisdom‐D (94% all‐E, gelatin coated). After 4 wk feeding, the flesh astaxanthin content, redness, and yellowness of astaxanthin‐supplemented groups were higher than those of control group at the second and fourth week (P < 0.05). Among these astaxanthin‐supplemented groups, the Wisdom‐B group had the highest flesh redness and astaxanthin content. The astaxanthin‐supplemented groups also had the lower flesh frozen loss and malondialdehyde level than control group at the fourth week (P < 0.05). The results indicated that dietary 100 mg/kg astaxanthin could improve the flesh redness and antioxidation abilities of rainbow trout. The ration of E/Z isomers and properties of coating materials of astaxanthin preparations influenced the pigmentation and antioxidation properties, and Wisdom‐B had a better pigmentation effect on the flesh than other astaxanthin preparations.  相似文献

The effects of dry or wet brining, different liquid smoke (LS) dipping schedules, and different LS on the weight, water activity, view area, and texture attributes of mussels were compared. Dry brining resulted in significant weight loss (about 30%), whereas liquid brining (4% salt) increased the original weights. With LS dippings, wet brined mussels lost weight, while dry brined samples gained weight. For the same brining and dipping schedule, there was no significant difference between dipping in hickory or in maple LS (p > 0.05). The R2 of linear, power, and quadratic polynomial fits to weight versus view area (calculated by image analysis) was 0.8, too low for practical applications. The change in individual mussel weight versus view area was different depending on the brining method. The weight versus view area of wet brined mussels increased and decreased during treatment steps, while dry brined mussels showed a regular decrease in weight versus view area. Texture, measured by texture profile analysis, and water activity also changed significantly upon dry brining (p < 0.05).  相似文献

The potential effect of body mass (m), maturity stage (ms), food type (ft), food protein (p) and lipid (li) content, and food protein‐to‐energy ratio, P/E (pe) on Specific Growth Rate (SGR, % day−1), Absolute Feeding Rate (AFR, g day−1), Feed Efficiency (FE, %), Assimilation Efficiency (AE, %), and Protein Retention Efficiency (PRE, %) in the common octopus was investigated. Six food types were provided ad libitum: shrimp, squid, hake, mussel, sardine and artificial one (gels made of hydrated squid flour agglutinated with sodium alginate). Estimated SGRs, AFRs, FEs, AEs and PREs were modelled with General Linear Models based on an information theory approach, using m, ms, ft, p, li and pe as potential predictor variables. SGR decreased when m increased; octopuses fed on shrimps showed the highest SGRs and the ones fed on mussels showed the lowest SGRs. AFR increased with m. Maximum and minimum FEs were observed, when food provided was shrimps and mussels, respectively. Maximum PRE was performed by octopuses fed on shrimps or sardines and minimum PRE by octopuses fed on mussels. Octopuses fed on artificial diet reached satisfactory levels of SGR (0.50% day−1) and FE (12.3%).  相似文献

To investigate effects of iron (Fe) on growth, haematological parameters, flesh quality and antioxidant status in muscle, young grass carp (Ctenopharyngodon idella) (292.0 ± 3.2 g) were fed graded levels of Fe (20.7, 38.4, 52.8, 79.3, 98.0 and 120.0 mg kg?1 diet) for 8 weeks. Per cent weight gain (PWG) and feed intake were improved with Fe levels up to 52.8 mg kg?1 diet. Serum Fe, erythrocyte counts, haemoglobin (Hb), haematocrit and mean cell haemoglobin increased with optimal Fe levels (38.4–79.3 mg kg?1 diet) (< 0.05). The muscle protein and lipid contents were increased by dietary Fe, whereas moisture, liquid loss, shear force and hydroxyproline contents followed opposite trends. Malondialdehyde and protein carbonyl contents in muscle were the lowest in fish fed the 52.8 or 79.3 mg Fe kg?1 diet, respectively, while superoxide dismutase, catalase, glutathione‐S‐transferase, glutathione peroxidase and glutathione reductase activities, and glutathione content were increased by Fe levels up to 52.8–79.3 mg kg?1 diet. Results indicated that the optimal Fe improved growth, flesh quality and muscle antioxidant defence of young grass carp. Dietary Fe requirements for PWG, serum Fe and Hb of young grass carp (292–695 g) were 73.5, 72.8 and 69.0 mg kg?1 diet, respectively.  相似文献

To investigate the effects of dietary vitamin C on growth, flesh quality and antioxidant capacity of juvenile golden pompano Trachinotus ovatus, a 56‐day feeding trial with five graded levels of dietary VC (D1: 11.69, D2: 34.89, D3: 59.10, D4: 114.26 and D5: 227.93 mg VC per kg of diet) was performed on 375 fish (triplicate groups of 25 fish per diet, initial weight 13.57 ± 0.09 g). Results showed that fish of D3 group exhibited the maximum specific growth rate (SGR) and the highest liver enzymatic activities of catalase (CAT) and glutathione peroxidase (GSH‐PX), which were consistent with the expression levels of cat and gsh‐px. Besides, the D3 group also showed higher contents of protein and lipid, and lower cooking loss, drip loss and malondialdehyde content in muscle than D1 group. The docosahexaenoic acid proportion in muscle increased with increasing dietary VC levels. Furthermore, the lowest expression levels of carnitine palmitoyltransferase1 (cpt1) and peroxisome proliferator‐activated receptor α (pparα) were detected in livers of D3 group. The optimum dietary VC level was 49.73 mg/kg from the broken‐line analysis based on the SGR, in which better growth performance, antioxidative ability and flesh quality were observed in T. ovatus juveniles.  相似文献

In order to test seasonal, rearing, maturing and anaerobic muscle activity effect on the flesh quality of turbot (Scophthalmus maximus) a total of 80 farmed turbot from three different strains from reared under natural or continuous light were killed by a percussive blow to the head in November (winter, Icelandic strain), March (spring, Portuguese strain) and June (summer, domesticated strain (France turbot)). To test the effect of anaerobic muscle activity, 10 fish were on each occasion pre rigor filleted, where one fillet was used as a control, while the other fillet was electrically stimulated using a squared 5 Hz, 10 V pulsed DC for 3 min. All pre rigor fillets were measured for pH, weighed, wrapped in aluminum foil and stored in polystyrene boxes with ice. After 7 days of storage the fillets were measured instrumentally for pH, drip loss, colour (CIE L* a* b*) and texture properties such as hardness and shear force, while fillet shrinkage and colour (RBG) were evaluated with computer imaging on photographs from a standard lightbox. Results showed that softness of the flesh was mainly influenced by factors associated with growth, such as season, photoperiod and maturation. Anaerobic muscle activity simulated with electrical stimulation caused an increase in drip loss (<1%) and loss of shear force (<4%), but had no effect on hardness or fillet shrinkage. Computer imaging revealed that muscle contractions related to the electrical stimulus forced out blood from the fillet causing less reddishness for the entire storage period. We conclude that a pH drop upon slaughter associated with anaerobic muscle activity has a minor effect on the flesh quality in the short run, while seasonal/alternatively genetic effects are predominant.  相似文献

The production and livelihood of aquaculture facilities depend on increased growth of species being harvested. Only 15%-25% of nitrogen and phosphorus are retained in farmed fish even when all feed is consumed. In both marine and freshwater aquaculture systems, bivalves have proven to be an inexpensive method for controlling algal growth through suspension feeding. With 25% of the native U.S. freshwater mussel species under federal protection, their use in aquaculture ponds could lead to further propagation and restoration of natural populations. Elliptio complanata is an important species in Delaware rivers and connected estuaries; a recent two-year study by the United States Geological Survey found that 98% of mussels found in the Delaware River were Elliptio complanata. The objective of this study was to calculate the clearance rate of E. complanata in a laboratory study. This would help us determine if mussels can serve as an addition to chemical and mechanical filtration regiments and thrive in an aquaculture setting. Using feeding chambers we investigated the effect of E. complanata on algae grown from an aquaculture pond and on a cultured alga over a four-hour period. Temperature, dissolved oxygen, and pH were monitored, and samples were than analyzed for total chlorophyll-α. The average of both the single cell using Chlorella vulgaris and mixed assemblage treatments chlorophyll-α clearance rate was 0.187 liters per hour per mussel. Prior to this experiment, these mussels were held for a period for two months in the holding system in the laboratory. The resulting low clearance rate as compared to the previous studies in the field is possibly due to the stress from holding these mussels. However, our study demonstrated that mussels have the potential to be used in an aquaculture setting and/or recreational ponds to help reduce algae populations that can occur as a result of eutrophication.  相似文献

A study was conducted to determine the effect of feeding with diets containing kefir on growth performance and fatty acid profile of rainbow trout (Oncorhynchus mykiss). Four isonitrogenous (450 g protein kg?1) and isocaloric (4325 kcal kg?1) diets were prepared in trout feed to contain 0 (control), 20, 50 and 100 g kg?1 kefir. Fish, initial weight of 46 g, were randomly distributed into triplicate 520‐L fibreglass tanks in freshwater flow‐through system. Fish were fed at 4% of the body weight thrice a day for 12 weeks. The results indicated that survival rate ranged from 97.14 to 100% without significant difference among treatments (p > .05). Whole‐body moisture and lipid composition were significantly affected by diets containing different levels of kefir (p < .05), but no differences were determined in protein and ash. Furthermore, the fatty acid profile of flesh showed differences among the groups. The percentages of saturated fatty acid in the flesh lipid decreased, while 18:3n‐3 and polyunsaturated fatty acids were increased at higher substitution levels of kefir grain. The present study showed that up to 100 g kg?1 supplementation of kefir in diets could be improve the fatty acid profile, especially PUFA, in fish flesh without adverse effect on the growth, feed utilization and survival rate of rainbow trout.  相似文献

Bivalves have been proven to be an inexpensive method for removing suspended solids, dissolved nutrients, and controlling algal growth through suspension feeding. The freshwater mussel, Elliptio complanata, is one of the most abundant species in Delaware, and it is additionally favorable for this experiment because of its hardiness against environmental stress and its filtration efficiency. This study examines the possibility that biomechanical filters such as E. complanata can supplement existing chemical and mechanical filtration regiments in aquaculture pond management. Twelve earthen aquaculture ponds located at the Delaware State University were stocked at a density of 4,000 catfish (Ictalurus punctatus) per hectare. Four different concentrations (0, 75, 150, and 300) of E. complanata were placed in trays within the ponds with three replicates of each concentration. The trays were suspended 15 cm below the surface of the water for a period of five months. Fish were fed a 32% protein diet twice daily at a rate of 1% of their body weight. Water quality was analyzed weekly while fish and mussel growths were measured biweekly. Results from water quality parameters varied significantly and were not conclusive based on the data obtained in this study. Water quality did not improve within the mussel treatment ponds during the course of this study and was found to be significantly below that of control ponds for all of the water chemistry parameters measured, with the exception of ammonia. Catfish in the 150 mussel treatment ponds grew the fastest; the 75 mussel treatment provided the most growth in mussels; and the 300 mussel group maintained the highest mussel survivorship. Although there were differences between ponds, fish growth and survivorship were not significantly different between treatments. While our mussel densities may have not been sufficient, their placement within the water column may have added additional stress. Being removed from the sediment can result in a decreased clearance rate in a benthic species such as E. complanata. This study may not have been conclusive in proving that mussels can be used to maintain water quality in aquaculture ponds, but we did see positive growth and survivorship of mussels and fish that indicated that the mussels were thriving in the aquaculture pond setting. This study may show that aquaculture ponds provide a suitable habitat in which propagated mussels may be held until their release into native areas.  相似文献

ABSTRACT

Obscure puffer fish (Takifugu obscurus) flesh, often eaten as raw fish fillet, is susceptible to softening. To investigate the mechanisms for the softening of flesh and changes in firmness, the proliferation of total microbial flora, Enterobacteriaceae, Pseudomonas spp., and lactic acid bacteria (LAB), total volatile basic nitrogen (TVBN), trimethylamine (TMA), and pH were periodically assayed during storage at 0, 4, and 8ºC. Myofibril extraction and separation of puffer fish flesh was carried out and the length of the separated myofibrils was measured periodically using an optical microscope during the entire storage time. Transmission electron microscopy was also used to inspect the changes in myofibrillar characteristics of the fish flesh at the beginning and end of the storage period. The results indicated that lower chilled temperature can contribute to reduced proliferation of microbial flora and less variation in TVBN, TMA, and pH and further maintain the integrity of myofibrils and retain firmness of puffer fish flesh during storage.  相似文献

  • 2. In Bear Creek, overall density dropped from 56.0–6.9 mussels m?2, and in Battle Creek, overall density dropped from 80.7–13.4 mussels m?2 in 2006.
  • 3. Large numbers of empty shells were found in Bear Creek in 2006, indicating high levels of mortality. A disproportionate loss of large, old mussels resulted in a forward shift in modal age class, from 51–60 years to 31–40 years.
  • 4. In Battle Creek, survival appeared to be higher, although significant numbers of shells were found in places. A lack of juvenile mussels indicated recruitment problems, possibly due to habitat degradation following colonization of the stream corridor by beavers and/or a lack of migratory host fish.
  • 5. Effective remedial actions for the Bear Creek and Battle Creek M. falcata populations are required within the next 5–10 years and 50 years, respectively, in order to ensure their long‐term survival.

Antioxidants have been widely used as additives to provide protection against oxidative degradation of foods by free radicals. The effect of thyme essence (Zataria multiflora Boiss) on the rancidity development in cobia (Rachycentron canadum) fillets during frozen storage was studied. Cobia fillets were treated with thyme essence (250 and 500 ppm) and then stored at??18°C for up to 6 months. Rancidity development was measured by several biochemical indices including free fatty acids (FFA), peroxide value (PV), thiobarbituric acid (TBA), and complemented by sensory analysis (flesh odor, consistency, and appearance). Also, pH and expressible moisture were measured during 6-month storage. Proximate composition was also determined in the first day. TBA, PV, and FFA levels increased in all treatments due to lipid oxidation. Thyme essence showed an antioxidative effect in cobia fillets during frozen storage as indicated by TBA, PV, and FFA levels. Results showed that FFA, primary and secondary oxidation products, expressible moisture (EM), and pH of thyme essence treated samples were significantly lower than those of the control samples (p < 0.05). Thyme essence retarded oxidative changes in frozen cobia fillets, and the best oxidation inhibition was obtained using thyme essence at 500 ppm.  相似文献

In order to determine the prevalence of pathogenic Escherichia coli, a total number of 155 E. coli isolates from aquaculture, clinical and veterinary sources were screened for seven pathogenic virulence markers and a house‐keeping gene by a polymerase chain reaction. The targeted virulence genes included eaeA of enteropathogenic E. coli, elt and est of enterotoxigenic E. coli (ETEC), ipaH of enteroinvasive E. coli, pCVD432 of enteroaggregative E. coli, stx, hlyA and eaeA of shigatoxigenic E. coli (STEC) and Enterohaemorrhagic E. coli. All the isolates were positive for phoA, the house‐keeping gene for E. coli. Among the 155 isolates, seven numbers (4.5%) harboured the virulence markers belonging to the pathogenic group ETEC and STEC. The virulent genes detected in these groups were elt, est, hlyA and stx. The sources of these virulence genes were fish (hlyA), shrimp (elt), feeder canal water (hlyA and elt) of aquaculture origin and from diarrhoea affected cow (hlyA, est and stx). The isolates with pathogenic traits belonged to the serogroups O6 or O29 and the remaining could not be typed. They showed resistance to two to four antibiotics out of the 12 antibiotics tested. Biotyping revealed that three isolates belonged to a single biotype (7333) and the remaining isolates were of diverse types. In conclusion, a molecular tool such as PCR proves as more effective tool for detection of this pathogen than the conventional methods. Detection of these emerging pathogens in aquaculture samples warrants for strict adherence to hygienic handling at retail outlets and proper cooking by the consumer before consumption.  相似文献

Pathogenic microorganisms may be incorporated into biofilms found in aquaculture systems, causing recurring exposure to potential disease agents. Aerobic plate counts, the presence of Enterobacteriaceae, and the presence of Escherichia coli, modified to express a green fluorescent protein (GFP E. coli), was used to evaluate the effectiveness of various sanitizers in decreasing bacterial incorporation into newly generated biofilms in recirculating aquaculture systems. Disks of Buna-N rubber, polyvinyl chloride (PVC), chlorinated PVC, glass, fiberglass, and stainless steel were placed in aquariums stocked with Nile tilapia, Oreochromis niloticus. The effectiveness of water, an alkaline cleanser, sodium hypochlorite, a quaternary ammonium compound, or peracetic acid as a sanitizer was evaluated on each substrate by enumerating total plate counts, GFP E. coli, and Enterobacteriaceae. Sodium hypochlorite and peracetic acid were the most effective sanitizers, with an overall percentage reduction of GFP E. coli of approximately 2 logs10. The quaternary ammonium compound was moderately effective, 1 log10, against the target organisms. Water demonstrated a 2 log10 reduction of the total plate count, suggesting that some mechanical cleaning was achieved. The type of material used as substrate for the biofilm had no significant (P?>?0.05) effect on the effectiveness of the sanitizers.  相似文献

A combination composed of Fructus mume extract and stable chlorine dioxide was investigated for its antibacterial activity, mechanism of action, and its application against Escherichia coli and Staphylococcus aureus in mackerel (Scomber japonicus) fillets. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values against E. coli and S. aureus ranged from 0.9–1.2 and 1.3–1.6 mg/mL for F. mume extract and 0.004–0.006 and 0.007–0.010 mg/mL for stable chlorine dioxide, respectively. We observed a mechanism of action against E. coli that involved a pore-formation role that led to cell membrane damage. In the case of S. aureus, the mechanism of action is believed to be mediated by the combination’s activity as a metabolite disrupting agent. Here, the amount of bacteria was greatly reduced in mackerel fillets treated with the combination. Moreover, the flavor and texture properties of fillets were not significantly affected by this treatment (p < 0.05). These findings suggested that the combination has promise in being used as an additive to a broader class of foodstuffs.  相似文献

The objective of this study was to assess sperm DNA longevity in blue mussels (Mytilus edulis) using a dynamic assessment of sperm DNA fragmentation (SDF) after sperm activation. Mature blue mussels (n = 57) in Vigo (Galicia, Spain) were obtained, specifically rope farmed blue mussels (n = 38) and wild blue mussels (n = 19). After the sperm collection, a subsample was assessed for SDF (0 h), while the rest of the sample was incubated for 6, 9, 12, 24 and 48 h at 15°C, assessing each time point using the Sperm‐Halomax kit (Halotech DNA, Madrid, Spain). The Kaplan–Meier estimator, log‐rank (Mantel–Cox) test and Mann–Whitney U‐test were used for statistical analyses (spss v. 16.0), α = 0.05. The rate of SDF (r‐SDF) between rope farmed and wild blue mussels over 0–6 h incubation was not significantly different (P = 0.278), but was for 6–24 h (P = 0.004). Differences in r‐SDF were observed when comparing the means between the two groups (P < 0.0001). Individual differences in r‐SDF existed among the rope farmed (P < 0.0001) and wild blue mussels (P < 0.0001). Wild blue mussels presented a higher DNA longevity than the farmed blue mussels. Selection of blue mussel males with a low level of sperm DNA damage and greater sperm DNA longevity may result in better fertilization and seed production.  相似文献

The effects of location and density on the growth and survival of juvenile freshwater pearl mussel, Chamberlainia hainesiana, 150–360 d after metamorphosis were investigated to determine the optimal ranges for commercial farming in Thailand. Juveniles were reared outdoors in two locations (the Mae Klong Reservoir and the Khwae Yai River, Kanchanaburi Province). Three density levels (25, 50, and 100 mussels/basket) were established at each location, with three replicates per density level. Phytoplankton from the water column in the natural habitats served as feed; no supplemental feed was provided. The mussels cultured in the reservoir had significantly higher growth rates than those cultured in the river (P < 0.01). The density level of 25 mussels/basket exhibited the highest average growth rates (in weight, length, height, and width per day). The density levels of 25 and 50 mussels/basket did not show significant differences in growth from each other (P > 0.05), but both showed significant differences in growth from the density level of 100 mussels/basket (P < 0.01).  相似文献
