1. The objective of this study was to evaluate the effect of maternal stress (MS) induced by supplementing the hen’s diet with 2 mg/hen/d dietary corticosterone (CORT) on embryonic development, biochemical blood parameters and hatching performance of broiler chicks.

2. A total of 200 Ross broiler breeder hens at 42 weeks of age were randomly divided into two groups: MS or control. Hens in the MS were fed 2 mg/hen/d CORT for 14 d. Eggs (648 and 635 eggs for MS and control, respectively) were collected from d 3 to 14 of dietary CORT supplementation and incubated. Weights of embryo, chicks and organs and body composition were determined during incubation and at hatch. Biochemical blood parameters were measured at internal pipping stage and day of hatch. Hatching performance and embryonic mortalities were recorded.

3. Hens fed a diet supplemented with CORT had lighter body weight and produced less eggs at the end of the 14-d treatment period. Although MS embryos were heavier than control from 12 to 18 d of incubation, chick weight was similar at the day of hatch. Lower relative weights for yolk sac and bursa were observed at 12 d of incubation for MS chicks compared to control. Chicks from both groups had similar body content in spite of higher fat content of MS embryos on d 18 of incubation.

4. MS had no effect on the duration of incubation or hatching performance but increased mortality at the pipping stage.

5. The results suggest that hormone-mediated MS might affect embryonic development during incubation without adverse effect on chick weight and body composition.  相似文献

The present study was conducted to assess the effects of dietary supplementation of Zingiber officinale and Zingiber zerumbet and to heat‐stressed broiler chickens on heat shock protein (HSP) 70 density, plasma corticosterone concentration (CORT), heterophil to lymphocyte ratio (HLR) and body temperature. Beginning from day 28, chicks were divided into five dietary groups: (i) basal diet (control), (ii) basal diet +1%Zzerumbet powder (ZZ1%), (iii) basal diet +2%Zzerumbet powder (ZZ2%), (iv) basal diet +1%Z. officinale powder (ZO1%) and (v) basal diet +2%Z. officinale powder (ZO2%). From day 35–42, heat stress was induced by exposing birds to 38 ± 1 °C and 80% RH for 2 h/day. Irrespective of diet, heat challenge elevated HSP70 expression, CORT and HLR on day 42. On day 42, following heat challenge, the ZZ1% birds showed lower body temperatures than those of control, ZO1% and ZO2%. Neither CORT nor HLR was significantly affected by diet. The ZO2% and ZZ2% diets enhanced HSP70 expression when compared to the control groups. We concluded that dietary supplementation of Z. officinale and Zzerumbet powder may induce HSP70 reaction in broiler chickens exposed to heat stress.  相似文献

Twenty healthy ostriches (ten cocks and ten hens), and twenty birds with tibiotarsal rotation (nine cocks and 11 hens) (14 months old) were isolated, hooded and weighed. A run (50 m x 2.5 m) was divided into sections marked 5 m, 10 m, 15 m and 20 m. Time taken for each bird to pass these points was recorded and speed computed. The degree of tibiotarsal rotation in the right foot was mean +/- SEM, 156 +/- 2.69 degrees. Comparisons between left and right foot length in healthy birds showed no significant differences. Foot length was significantly lower in tibiotarsal rotation (P = 0.03). The right foot in tibiotarsal rotation was significantly shorter than the left foot. The number of strides per each 5 m division were significantly (P < 0.05) greater in tibiotarsal rotation by comparison with healthy birds. At 20 m, healthy cocks had more strides than hens. The stride length in hens was significantly (P < 0.05) greater than cocks at 5, 10 and 15 m, respectively, but lower throughout in tibiotarsal rotation (P = 0.001). The speed of hens was significantly (P < 0.05) greater than cocks. Tibiotarsal rotation resulted in significantly (P < 0.05) reduced speeds. Hens may be able to escape danger faster than cocks. The occurrence of tibiotarsal rotation necessitates consideration of genetics, management, sex, nutrition and growth rates.  相似文献

1. The effects of corticosterone (CORT) on diet selection of broiler chickens offered a choice of a high protein concentrate (381 g CP/kg, 17.5 MJ/kg ME) and whole wheat (113 g CP/kg, 15.9 MJ/kg ME) in relation to age were examined in two experiments.

2. Daily intramuscular injections of 2 and 4 mg/kg of CORT for a 5‐d period in both 2‐ and 5‐week‐old chickens resulted in increases in total food, protein and energy intakes. This led to a decrease in protein accretion in older but not younger chicks, an increase in total lipid contents of the carcase at both ages, and produced changes in internal organs.

3. CORT significantly reduced body weight gain of young but not old chickens, suggesting that mature birds respond better than young ones to the physiological changes caused by treatment, by making subsequent appropriate food choices.

4. Administration of CORT in young chicks increased wheat intake at 2 and 4 h after injection, while in older birds a similar increase was maintained up to 24 h after injection. Intake of HP food was decreased by both doses of CORT in young chicks but there was no significant effect in older chickens.

5. Changes in energy: protein ratio in the chosen diet appeared at 4 h after treatment in old chickens and at 24 h in younger chicks.

6. The results suggest that birds are able to detect metabolic changes caused by CORT administration and attempt to redress them by modifying their food pattern. The time course of the response of birds to these changes is age related. However, the food selection pattern did not completely compensate for the physiological defects.  相似文献

It has been previously established that growth and carcass qualities of chicks are modified by genotype and dietary amino acid supply. In this study, we evaluated the effects of lysine deficiency and genetic selection on muscle protein metabolism. Chicks originating from an experimental line selected for breast development (QL) and its control line (CL) were provided ad libitum access to isoenergetic diets containing 20% crude protein but differing in their lysine content (0.75 or 1.01%). Protein fractional synthesis rates (FSR) were measured in vivo in the pectoralis major and sartorius muscles of 3-wk-old chickens (flooding dose of [3H]phenylalanine). Fractional breakdown rates (FBR) were estimated as the difference between synthesis and deposition. Lysine deficiency reduced (P < 0.001) growth performance and muscle weights and increased (P < 0.05) muscle FSR, capacity for protein synthesis (muscle RNA:Protein, Cs) and FBR. Although QL birds grew faster and had heavier pectoralis major muscles than CL birds (P < 0.05), there was no line difference in sartorius weight (P = 0.15). No difference between the lines was observed in sartorius protein metabolism (P > 0.14). In the pectoralis major muscle, chicks of both lines receiving an adequate lysine intake also exhibited similar protein turnover rates. However, in chicks fed the lysine-deficient (0.75% lysine) diet, FSR and Cs were higher in QL than in CL chicks (P < 0.05), and FBR tended (P = 0.07) to be higher in QL chicks. This increased protein turnover in the QL birds on the lysine-deficient diet suggests that the responsiveness of muscle protein metabolism to amino acid supply is modified by genetic selection for breast development.  相似文献

The effect of selection for high stress response on adrenocortical function was examined by measuring the corticosterone response of adrenocortical cells isolated from random-bred Japanese quail and quail selected for high serum corticosterone response to immobilization (high-stress). Highly enriched adrenocortical cells were incubated with various concentrations of ACTH1-24 (ACTH), 8-bromo-cyclic AMP (8Br-cAMP) and pregnenolone for 2 hr. Corticosterone production was measured by radioimmunoassay. Basal corticosterone production values by cells from random-bred and high-stress birds were not different. In contrast, the average maximal ACTH- and 8Br-cAMP-induced corticosterone production by cells from high-stress quail was 89% greater than that of cells from random-bred quail. However, the average pregnenolone-supported corticosterone production by cells from high-stress birds was 34% less than that of cells from random-bred birds. Thus, the data suggest that although random-bred quail cells had a greater potential capacity for corticosterone production, high-stress quail cells had a greater ability to couple ACTH, ACTH-transmembrane-signaling factors and subsequent second messengers with the available steroidogenic enzyme pool. The magnitude of the differences in function between cells from high-stress and random-bred birds was greater for female cells compared to male cells. In addition to differences in cellular function, there were also differences in adrenal and relative adrenal weights between random-bred and high-stress quail. The average, adrenal and relative (mg/100 g body weight) adrenal weights of high-stress quail were 14-16% greater than those of random-bred quail. It is concluded that the enhanced serum corticosterone response of the high-stress quail line is, in part, due to an increase in relative adrenal weight and an increase in adrenocortical cell responsiveness to ACTH.  相似文献

Research was focussed on investigating the influence of different quantities (0, 10, 20 and 30%) of rapeseed meal (RM), 00-cultivar Silvia on production results, as well as on the morphological and pathohistological changes in the internal organs of chicks during the course of the experiment, measured on the 21st and 42nd days of the 42-day experiment. The experiment involved 120 chicks divided into four groups: a control group (C) and three experimental groups (E1-3). It was found that chicks in Groups C, E1 and E2 realised significantly (P < 0.05) higher gains than those in Group E3, both after the test period and at the end of the experiment. No significant differences with regard to feed conversion were found between groups of chicks. Throughout the experiment chicks in Groups E1-3 were found to have a significantly (P < 0.05) heavier liver. Compared to Groups E1-3, Group C chicks had a significantly (P < 0.05) heavier gizzard after 21 days, but following the finisher diet Group E3 had a significantly (P < 0.05) lighter gizzard. Compared with birds in Group C, those in Groups E2-3 had significantly heavier (P < 0.05) unevacuated intestines when fed the starter diet, and those in Groups E1-2 showed a similar result when fed the finisher diet. Chicks in Group E3 had a significantly lower grill weight than those in Groups C, E1 and E2, both in the first half of the experiment and at the end. The starter diet did not result in any differences in the quantity of abdominal fat among groups, but following the finisher diet Groups E2-3 showed significantly less abdominal fat in comparison to Groups C and E1 (P < 0.05). Group C showed a significantly higher carcass yield than Groups E1-3 (P < 0.05) in the first half of the experiment and at the end. Groups E1-3 manifested a slight to medium hypertrophy of thyroid epithelial tissue as well as slight thymus hypertrophy and slight atrophy of the cloacal bursa follicles.  相似文献

为探讨蛋鸡攻击行为与五羟色胺、皮质酮的相关性,选用1日龄京白939雏鸡96只,随机分为6个重复,每重复16只,采用瞬时观察法和连续观察法筛选不同周龄攻击行为有差异的蛋鸡,结合ELISA和qRT-PCR方法测定蛋鸡血清激素水平和5-HT1A受体mRNA表达量的变化。结果显示:蛋鸡的攻击行为随周龄的增加呈现逐渐上升的趋势(P<0.05);整体看,血清五羟色胺(5-HT)含量随周龄的增加呈下降趋势,皮质酮(CORT)呈现上升趋势(P<0.05),而下丘脑5-HT1A受体mRNA表达量随周龄增加有上升的趋势(P<0.05或P<0.01)。Pearson相关分析结果显示,蛋鸡攻击行为与血清五羟色胺(5-HT)呈极显著负相关(R=-0.741,P<0.01),与皮质酮(CORT)呈显著正相关(R=0.751,P<0.01)。综上所述,血清激素水平与蛋鸡攻击行为有一定联系,5-HT1A受体基因可能参与调控蛋鸡的攻击行为。  相似文献

Summary

A comparative study was undertaken regarding the extractability of glucosaminoglycans and mucoproteins in the tendon tissue of birds highly susceptible to synovitis, viz. broiler breed cocks (BB cocks), and of birds highly resistant to synovitis, viz. White Leghorn hens (WL hens).

It .was shown that in the case of WL hens this extractability decreased in accordance with ageing. In the case of BB cocks such a decrease was not observed. This observation is in support of a working hypothesis which supposes a relatively high degree of interaction between the several components of tendon tissue (e.g. collagen and glucosaminoglycans/mucoproteins) in the case of WL hens, and a relatively low degree of this interaction in the case of BB cocks.

Moreover the results of this study account for the observation that the tendon tissue of WL hens is more resistant to tensile stress than that of BB cocks, and they indicate that the above interaction is a determinant in the aetiology of synovitis.  相似文献

The relationships between the severity of lameness, the presence of tibial dyschondroplasia, and the frequency of dustbathing behaviour and duration of tonic immobility were studied in 96 broiler chicks kept from day-old in groups of four in wire-floored cages. Dustbathing was observed when the birds were given access for one hour to a tray containing sand or straw. The duration of tonic immobility and the severity of lameness were scored during weeks 3, 4, 5 and 6. After slaughter at 41 or 45 days of age the birds were examined for tibial dyschondroplasia; 34 birds were classified as having tibial dyschondroplasia and 54 had detectable lameness problems. Birds with tibial dyschondroplasia had a higher lameness score (P < 0.001), dustbathed on fewer days (P < 0.0001), and had longer periods of tonic immobility (P < 0.03) at six weeks than birds which did not have the condition. However, almost all the chicks, including those with tibial dyschondroplasia dustbathed on day 27 after they had not been given access to the tray for three days, significantly more than on days when they had not been deprived of the tray (P < 0.0001). Furthermore, lame birds and birds with tibial dyschondroplasia also dustbathed less with age. Studies of the vertical wing-shaking element of dustbathing confirmed that tibial dyschondroplasia reduced dustbathing (P < 0.05) and also showed that during the one-hour tests, sand resulted in more dustbathing than straw (P < 0.01).  相似文献

We reported that meat chicks have either a greater capability to acclimatize to novel environments, or a blunted hypothalamic‐pituitary‐adrenal response to novel environments compared with layer chicks in a commercial base. The present study compared the differences in behavior and plasma corticosterone concentrations under isolation‐induced stress between neonatal meat and layer Nagoya chicks which had been separated from the same population. Both types of neonatal chicks reared in groups were individually separated and their spontaneous activity and distress‐induced vocalizations were monitored for 10 min. The responses of the two types were remarkably different, with the meat chicks being less active than the layer chicks. Distress‐induced vocalizations were fewer in the meat than in the layer chicks. The meat chicks spent more time in a sleeping posture during isolation‐induced stress. Plasma corticosterone concentrations measured at the end of the test tended to be higher in the layer chicks than in meat ones, but not significantly. In conclusion, the selection of Nagoya chickens for meat and layer may have trends similar to those observed in commercial chickens in relation to stress susceptibility.  相似文献

This study was designed to examine the physiology and behavior of pigs whose dams were snared and then injected with ACTH during gestation. Administration of ACTH to dams during pregnancy has been shown to replicate the effects of prenatal stress in other species. Control sows (n = 8) were given no treatment, whereas the treatment sows (ACTH, n = 8) were immobilized by snaring the snout and then administered an i.v. injection of ACTH (1 IU/kg BW) weekly from 6 to 12 wk of gestation. A pig was killed from each sow at 1, 30, and 60 d of age. The hypothalamus, pituitary gland, adrenal glands, and liver were immediately obtained to determine the amounts of corticotropin-releasing hormone (CRH), beta-endorphin, and mRNA for pro-opiomelanocorticotropin (POMC), the ACTH receptor (ACTH-R), and insulin-like growth factor I (IGF-I). Pituitary corticotrope and somatotrope cell numbers and adrenal cortex-to-medulla area ratios (CORT:MED) were also determined. Pigs' behaviors were recorded at 6 and 8 wk of age. At 75 d of age, a blood sample was taken and a biopsy puncture was created on one pig from each litter, then pigs were stressed by mixing. Blood was sampled every other day for 10 d to determine plasma cortisol concentrations and differential leukocyte counts. Biopsy damage was evaluated for healing. At 1 d of age, control pigs tended to weigh more (P = .09), have a lower expression of ACTH-R mRNA (P = .01) and IGF-I mRNA (P = .01), and a lower CORT:MED (P = .04) than ACTH pigs. At 30 d of age, control pigs had a greater concentration of beta-endorphin (P = .01) and tended to have a lower concentration of CRH (P = .09) and IGF-I mRNA (P = .10) than ACTH pigs. At 60 d of age, control pigs tended to have lighter pituitary glands (P = .08), a lower expression of POMC mRNA (P = .02), and a CORT:MED (P = .003) than ACTH pigs. At 8 wk of age, control pigs performed a higher frequency of belly nosing (P = .07) and oral vice behaviors (P = .01) than ACTH pigs. In response to mixing stress, control pigs had lesser concentrations of plasma cortisol (P = .03) and healed faster (P = .006) than ACTH pigs. Thus, exogenous ACTH and restraint during gestation alters the HPA axis of the sow's offspring, and during stressful situations later in life health, and therefore welfare, may be compromised.  相似文献

In an 18-month study the domestic fowl in 42 households in three traditional management systems were recorded to determine reproduction, growth and mortality parameters. There were significant differences for all systems combined in the number of clutches started per month with the lowest number being laid in the cool dry season of January and February. Mean number of eggs per clutch was 8.8; the number of eggs increased significantly throughout the career of the hen but there were no significant systems differences. Egg weight was 34.4g there being no significant differences due to age of hen or system. Intervals between clutches were 92 days, annual egg output was 35 per hen (assuming that birds were present in the flock for a year) equivalent to about 118% of mean adult hen body weight: hens actually in the flocks during the study had produced 2.1 clutches on average. Hatchability of eggs was 69.1% there being significant seasonal and systems differences in this parameter, lowest hatchability being in the hot dry season (March to June) and in the rainfed millet system. Mortality rates to eight weeks were 56% of chicks hatched. Growth to 10 weeks averaged 4g/d. Mature cocks weighted 1.60 kg and mature hens 1.02 kg: hen weights were significantly affected by reproductive state, laying hens being heavier than both incubating hens and those rearing chicks, while those rearing chicks were heavier than those incubating. The effect of ACTH (16 units) on plasma cortisol and corticosterone concentrations in healthy psittacine birds was evaluated. Plasma corticosterone significantly increased (P less than 0.01) from a mean (+/- SD) basal concentration of 3.25 +/ 3.6 ng/ml to 26.47 +/- 9.25 (one hour after ACTH administration) and 25.69 +/- 13.23 ng/ml (2 hours after ACTH administration). For maximal increase in plasma corticosterone as measured by radioimmunoassay (RIA), heat denaturation was necessary to release corticosteroids from steroid-binding proteins. As measured by RIA, plasma cortisol concentrations did not increase, whether or not the heat denaturation step was included. Addition of cortisol to avian plasma did not prevent accurate quantification of cortisol as measured by RIA. Plasma corticosterone concentrations in cockatoos, macaws, Amazon parrots, conures, and lorikeets before and after ACTH administration indicated that the ACTH stimulation test could be used to evaluate adrenal secretory capacity in psittacine birds.  相似文献

Birds tend to adjust their behavior and physiology to changes in food availability in their environment. Seasonal fluctuation of food resources may act as an energetic challenge, augmenting hypothalamus–pituitary–adrenal axis (HPA axis) activity, leading to an increase in corticosterone concentrations and promoting the metabolism of energy stores. Plant invasions may alter seasonal food fluctuations by providing a food supply during scarce seasons. This could attenuate the energetic challenge, reducing HPA axis activity and the metabolism of reserves. Using a system with seasonal fluctuation in food availability, we tested if fruit supplementation by the invasive fleshy‐fruited Pyracantha angustifolia during the season of native fruit scarcity decreases the consumption of energy stores through activity attenuation of the HPA axis. We measured changes in body condition and circulating corticosterone (CORT) concentration in Turdus chiguanco occurring at sites both invaded and not invaded by P. angustifolia over 3 time periods that correspond to the periods prior to, during and after highest fruit production of the plant. Fruit scarcity in the ecosystem appears as an energetic challenge for T. chiguanco, given that body mass, fat score and residuals of body mass/tarsus length decreased during winter in a site not invaded by the exotic shrub. Conversely, the presence of the invasive plant seemed to attenuate the metabolism of energetic reserves, as we did not record changes in body condition in birds inhabiting the invaded site. Unexpectedly, plasma CORT concentration did not vary between sites or periods. Further evaluation is required to elucidate how enhanced body condition, resulting from the consumption of a fleshy‐fruited invasive plant, affects survivorship and reproductive performance in T. chiguanco.  相似文献

1. Alterations in carbohydrate metabolism in terms of tissue glycogen contents, phosphorylase (EC 2.4.1.1) activity, hepatic glucose-6-phosphatase (6-6-Pase: EC 3.1.3.9) activity and blood glucose have been evaluated in 30-d-old White Leghorn chicks under induced chronic hypocorticalism (by dexamethasone: DXM) and hypercorticalism (by corticosterone: CORT). 2. DXM treatment showed increased tissue glycogen contents and hypoglycaemia with decreased phosphorylase activity while CORT treatment produced a reverse set of changes. 3. Both steroid treatments increased hepatic G-6-Pase activity. These observations have been taken to indicate a definite role for glucocorticoids in regulating carbohydrate metabolism in neonatal chicks. 4. It is suggested that hypo- or hyper-corticalism could influence carbohydrate metabolism by affecting the secretory/activity ratio of pancreatic hormones.  相似文献

ABSTRACT

1. The purpose of the present study was to determine if an intraperitoneal injection of two toll-like receptor-7 (TLR7) agonists, imiquimod and resiquimod, affect feed intake, voluntary activity, cloacal temperature, crop-emptying rate, plasma corticosterone (CORT) and glucose concentrations, and splenic gene expression of cytokines in chicks (Gallus gallus).

2. Although intraperitoneal injection of 100 µg imiquimod significantly increased splenic gene expression of interleukin-1β (IL-1β), IL-6, IL-8, and interferon-γ (IFN-γ), it did not affect feed intake, voluntary activity, cloacal temperature, crop-emptying rate or plasma constituents.

3. Intraperitoneal injection of 100 µg resiquimod significantly decreased feed intake, voluntary activity, cloacal temperature, crop-emptying rate and increased plasma corticosterone concentrations.

4. Intraperitoneal injection of resiquimod significantly increased splenic gene expression of IL-1β, IL-6, IL-8, IFN-γ, and tumour necrosis factor-like cytokine 1A.

5. The results showed that activation of TLR7 is associated with anorexia, hypoactivity, hypothermia, disturbance of feed passage in the digestive tract and the response to stress in chicks.  相似文献

Sixty 7‐d‐old male broiler chicks were housed in individual metabolism cages and offered one of six purified diets for 14 d. The diets were formulated to have the same ME contents but had different nitrogenous supplements: diet A, was the basal diet, containing PRG essential amino acid (EAA) mixture equivalent to 15 g N/kg; B, basal+12% glutamic acid (GA); C, basal plus EAA mixture equivalent to EAA content of 20.09% DPM; D, basal + EAA+12% GA; E, basal + 20.09% autoclaved DPM; F, basal + 20.09% unautoclaved DPM. The rank order of final body weights at 21 d of chicks fed on these diets were D(292 g) > B(258 g), (F254 g), E(253 g) > C(206 g) > A(180 g), (P<0.05). Chicks fed on diet C had better food conversion efficiencies (FCE) than those fed on diet A (0.449 and 0.374 respectively) and D was better than B (0.592 and 0.533 respectively).

In a second experiment, carried out under field trial conditions using commercial‐type diets formulated to contain DPM, 24 groups, each of about 40 male or female broiler chicks, were given diets containing 0% (P) or 5% (Q) DPM from 1 d of age for 4 weeks. From 4 to 8 weeks one of three diets with 0% (R), 5% (S) or 10% (T) DPM was fed according to a 2 (sexes) x 2 x 3 factorial design. Between o and 4 weeks feeding DPM had no significant effect on body weight or food consumption, but birds fed diet Q had an improved FCE (P<0.05). Between 4 and 8 weeks birds fed on S and T were heavier than the controls at 6 weeks, but this increase was not significant at 8 weeks (P> 0.05). Food consumption was not affected, but the FCE improved with increasing inclusion of DPM. At 8 weeks of age birds fed on Q, were heavier than birds fed on diet P.  相似文献
