日粮不同营养水平对母猪妊娠早期胚胎存活的影响   总被引:3,自引:0,他引:3  
试验以63头长白-大白杂交初产母猪为试验动物,研究不同营养水平对母猪妊娠早期繁殖性能的影响。将配种后母猪随机分到3个营养水平组(高、中、低)中,各营养水平通过每天供给2、1.2和0.6倍维持需要的饲粮,分别在妊娠12、25和35d将母猪称重、屠宰。结果表明:各水平组母猪妊娠12、25和35d的母体总增重和净增重有极显著(P<0.01)差异。中水平妊娠25d的子宫及内容物重量和两侧子宫角长度均显著(P<0.05)高于高水平和低水平;中水平妊娠35d的子宫及内容物重量和右侧子宫角长度显著(P<0.05)高于高水平。中水平妊娠12 d的总活胚数显著(P<0.05)高于高水平。中水平妊娠12、25和35d的胚胎存活率显(著P<0.05)高于高水平;中水平妊娠25d和35d的胚胎存活率显著(P<0.05)高于低水平。中水平妊娠35 d的活胚重显著(P<0.05)高于高水平,极显著(P<0.01)高于低水平。以上试验表明,在妊娠35d内给母猪饲喂1.2倍维持需要能使子宫发育更良好,并具有较高的胚胎存活率,而高水平饲喂和严重限饲均会导致母猪妊娠早期的胚胎存活率降低。  相似文献

A RIA was developed for porcine intrauterine folate binding protein (FBP). Displacement of [125I]FBP caused by increasing dilutions of uterine flushings collected from either d-15 pregnant or nonpregnant gilts or media from culture of endometrial tissue from d-15 pregnant or nonpregnant gilts was parallel to the displacement caused by the standard curve. Addition of known amounts of purified allantoic fluid FBP to dilutions of either intrauterine flushings or endometrial culture medium were measured accurately with the RIA. To test specificity, 2-mL samples of uterine flushings collected from d-15 pregnant and nonpregnant gilts were preincubated with 10 microCi of [3H]folic acid and then chromatographed using Sephadex G-100 (Sigma Chemical Co., St. Louis, MO). The fractions were subsequently assayed for radioactivity by liquid scintillation spectrophotometry and for FBP by RIA. The [3H]folic acid and FBP peaks coincided, indicating that the RIA is specific for FBP. Uterine flushings were collected on d 10, 11, 12, 13, 14, and 15 of the cycle or pregnancy from 1) White crossbred, 2) progesterone-treated White crossbred (200 mg of progesterone at 48 and 72 h after estrus), and 3) Meishan gilts and assayed for FBP. Total FBP increased 140-fold from d 10 to 15, and the pattern of change across day did not differ between pregnant and nonpregnant gilts. Progesterone treatment increased intrauterine FBP content on d 10 and 11. No difference in FBP concentrations was detected between White crossbred and Meishan gilts. These results indicate that the RIA for FBP is valid, allowing measurement of this protein in uterine flushings and endometrial culture medium. The onset of FBP secretion by the uterus between d 10 and 15 of the cycle or pregnancy is influenced by the timing of onset of progesterone influence in a manner similar to the endometrial proteins uteroferrin and retinol binding protein. In contrast to these endometrial proteins, FBP concentrations are similar in Meishan and White crossbred gilts.  相似文献

A total of 59 gilts (BW = 137.7 kg) from 3 breeding groups were used to assess the effects of feeding l-carnitine during gestation on gilt growth characteristics, blood metabolites, and uterine and chorioallantoic expression of IGF axis components at d 40, 55, and 70 of gestation. Experimental treatments were arranged in a 2 x 3 factorial, with main effects of added l-carnitine (0 or 50 ppm) and day after initial breeding (d 40, 55, or 70 of gestation). All gilts received a constant feed allowance of 1.75 kg/d and a top-dress containing 0 or 50 ppm of l-carnitine beginning on the first day of breeding through the assigned day of gestation. No dietary treatment differences were observed for gilt BW, backfat, or estimated protein or fat mass at any day of gestation. No differences were observed in circulating total and free carnitine at breeding, but concentrations increased (P < 0.01) as day of gestation increased for gilts fed diets containing l-carnitine compared with those fed the control diet. Maternal IGF-I concentration decreased (P < 0.01) from d 0 to 70 for all gilts, with no differences between treatments. Insulin-like growth factor binding protein-3 mRNA (P = 0.05) and IGFBP-5 mRNA (P = 0.01) increased in the endometrium of gilts supplemented with l-carnitine. These data demonstrate that l-carnitine supplementation and day of gestation alter the expression of the IGF axis by changing the expression of IGFBP at the fetal-maternal interface in swine. These changes in the IGF axis at the fetal maternal interface may aid in determining the reasons for the effects of l-carnitine on reproductive traits.  相似文献

The interrelationships among d-11 conceptus size, d-105 placental weight, placental efficiency (the ability of the placenta to support fetal growth and development), fetal erythropoiesis, and uterine capacity were examined in 1/2 Meishan, 1/2 White crossbred gilts that were unilaterally ovariohysterectomized at 90 to 100 d of age. In Exp. 1, gilts were mated after at least one normal estrous cycle and then slaughtered at 105 d of gestation and number of fetuses and CL, placental weights, fetal weights, hematocrits, fetal plasma iron, and fetal plasma folate were measured. In Exp. 2, gilts were mated and plasma progesterone was measured on d 2 and 3 of gestation. On d 11, the length of the remaining uterine horn was recorded and the uterine horn was flushed with minimal essential medium. Number of CL, conceptus number, conceptus diameters, and total uterine flush retinol-binding protein (tRBP), acid phosphatase (tAP), and folate-binding protein (tFBP) were measured. Gilts were mated again and slaughtered at 105 d of pregnancy and the same traits measured in Group 1 were recorded. Plasma progesterone concentrations on d 2 and 3 were correlated with average conceptus diameter on d 11 (r = 0.60, P < 0.01, for each day). In contrast, tRBP (r = 0.49, P < 0.01), tAP (r = 0.53, P < 0.01), and tFBP (r = 0.51, P < 0.01) in uterine flushings on d 11 were only correlated with d-3 plasma progesterone concentrations. No correlations between d-11 average conceptus diameter or d-11 uterine length with d-105 uterine capacity were observed. Uterine capacity was negatively correlated with placental weight, fetal weight and fetal hematocrit (r = -0.36, P < 0.01; r = -0.44, P < 0.01; r = -0.32, P < 0.01; respectively). Hematocrits were correlated with fetal plasma iron (r = 0.50, P < 0.01) and folates (r = 0.44, P < 0.01). Hematocrit, plasma iron, and plasma folate were each correlated with residual fetal weights after adjusting for placental weight (a measure of placental efficiency), and accounted for 11% of the variation in this trait. These data suggest that conceptus diameter and uterine protein secretion on d 11 may be influenced by the onset of progesterone secretion by the CL, but do not support an influence of conceptus growth during early pregnancy on uterine capacity. These results also suggest that reducing placental and fetal weights will likely result in increased uterine capacity.  相似文献

Two estrogen antagonists (keoxifene and clomiphene) and two aromatase inhibitors (LY56110 and 4-hydroxyandrostenedione, 4-OHA) were utilized to determine the role of conceptus estrogen in trophoblastic elongation and maintenance of pregnancy in the pig. Pregnant gilts were unilaterally hysterectomized on day 10.5, and infused via a uterine arterial catheter with 200 mg of keoxifene or vehicle. The remaining uterine horn was removed based on time estimated for conceptus elongation. In a second study, pregnant gilts were injected daily with 200 mg (i.m.) of clomiphene or vehicle during pregnancy (days 10-16) and hysterectomized on day 30. A third study assessed in vitro aromatase inhibition by 4-OHA and LY56110 using trophoblastic microsomes incubated with [1 beta, 2 beta-3H]-androstenedione for 6 hr. In a fourth study, in vivo inhibition of aromatase activity was determined. For this study pregnant gilts, unilaterally hysterectomized on day 10.5, received either 4-OHA, LY56110, or vehicle. Conceptus development and uterine estrogens were quantified. None of the estrogen antagonists and aromatase inhibitors interferred with conceptus elongation. Uterine protein, calcium and acid phosphatase were similar (P greater than .10) between keoxifene- and vehicle-treated gilts. Embryonic survival of clomiphene- and vehicle-treated gilts was similar (91.5 vs 87.4%). In vitro, 4-OHA and LY56110 had 50% inhibitory concentrations of 0.1 microM and 13 nM. Treatment of gilts with 4-OHA reduced total estrogens in uterine flushings by 57% (P less than .02), whereas treatment with LY56110 did not significantly lower total estrogen content in uterine flushings. Estrogen antagonists were not effective in blocking conceptus elongation and maintenance of pregnancy. Although estrogen synthesis can be inhibited in vitro, dosages of aromatase inhibitors used were not totally effective in vivo.  相似文献

It has been shown that zearalenone disrupts early pregnancy in swine without altering intrauterine content of estradiol 17 beta or progesterone, embryo migration, or estradiol-17 beta synthesis by blastocysts. However, serum concentrations of progesterone were reduced 2 to 3 weeks after mating in gilts that ingested zearalenone. Therefore, progesterone was administered to gilts during early pregnancy to determine whether it could counteract the detrimental actions of zearalenone on embryonic development. Thirty-two crossbred gilts (Hampshire x Chester White x Yorkshire x Duroc) were assigned randomly to a 2 x 2 factorial arrangement of treatments: zearalenone (Z); zearalenone plus progesterone (ZP); progesterone (P); or control (C). From postmating days 4 to 15, Z- and ZP-treated gilts were fed 1 mg of Z/kg of body weight, and P-treated and C gilts were fed ethanol as vehicle in a corn-soybean diet. On postmating days 3 to 15, P- and ZP-treated gilts were injected IM with 100 mg of progesterone, and C and Z-treated gilts were injected with progesterone carrier (15% ethanol, 15% benzyl alcohol, 70% propylene glycol). Blood was collected from gilts by puncture of the jugular vein daily from days 3 to 15, on alternate days from days 17 to 31, and then twice weekly until the end of the experiment. Fetal development was assessed in Z- and ZP-treated gilts on postmating day 47.6 +/- 2.9 by cesarean section and in P-treated and C gilts at slaughter on postmating days 51.2 +/- 3.2. Serum concentrations of progesterone in P-treated gilts were greater on days 7 to 8, 10 to 15, 17, and 19 than in C gilts. Serum concentrations of progesterone were greater on days 8, 10, and 12 in ZP-treated than in C gilts. Swine uterine capacity affects litter size, and it could be used as a selection parameter of reproductive performance. Although there are some controversial results, evidences show that the catheter penetration length is positively correlated with litter size, and it could be used as a tool for predicting selection methods. The aim of this study was to determine whether there is any association between the prenatal survival rate and placental size at 70 days of gestation, the vaginal length [catheter penetration length during artificial insemination (AI)] and the uterine capacity in a homogeneous group of gilts. Sixty-six commercial-line gilts in pre-pubertal phase had their oestrus induced by hormonal treatment [600 UI of Equine Chorionic Gonadtrophin (eCG) i.m. and after a 72-h period 5 mg of luteinizing hormone (LH) i.m.], but only 40 gilts showed cyclicity after induction. The AI catheter penetration length was tested on these 40 gilts at the moment of AI using a calibrated AI catheter. Four gilts returned to oestrus and the other 36 were killed at around day 69 of pregnancy. The uterine length and weight showed a significant and positive correlation with the prenatal survival rate (p <0.05). The catheter penetration length was unable to predict the conceptus survival rate on 70 days of gestation; however, the uterine size influenced the survival rate positively. The mean placental area was positively correlated with the mean placental weight (p <0.0001), and both with the mean foetal weight (p <0.0001 and p <0.001, respectively). The analysis of the results obtained showed that neither did the catheter penetration length measurement during AI, nor the prenatal survival rate on day 70 of pregnancy predict the uterine capacity, but the uterine and placental size had a significant influence on the prenatal survival and foetus weight, respectively.  相似文献

Modern sows are younger and leaner at time of mating and probably have poorer appetites than sows of 10 to 15 years ago. Therefore, feeding strategies should aim to minimize weight loss and maintain a sow's body condition throughout her reproductive life. The efficiency with which gilts are introduced into the breeding herd is as important in economic terms as is the efficiency with which the sow returns to estrus after weaning. Gilts should be selected at 50 to 60 kg, and fed a 16% protein diet ad libitum until mated at their second estrus, when they weigh 115 to 120 kg and have 17 to 20 mm backfat. Flushing gilts before the onset of second or third estrus increases ovulation rate of restricted gilts to the levels achieved by gilts fed ad libitum. During gestation, maintenance represents 75 to 85% of total energy requirements. The aim should be to achieve 20 to 25 mm backfat at farrowing. Increased feed intake from day 2 to 3 after mating will not increase embryo mortality. Feeding an extra 1 kg feed/sow/day for the last 10 days of gestation increases piglet birth weight slightly and prevents a loss of 1.5 to 2.0 mm of sow backfat. Wherever possible, sows should be fed ad libitum from the day after farrowing until weaning. Reduced feed intake by lactating sows, for whatever reason, results in excessive weight and condition loss. Excessive weight loss in lactation causes extended remating intervals, a lower percentage of sows returning to estrus within 10 days of weaning, reduced pregnancy rate, and reduced embryo survival. Ovulation rate is not affected by level of feed intake in lactation. It has been suggested that sows will have minimum weaning-to-service intervals when they weigh 150 kg or more at weaning. It is likely that the sow must be anabolic for about 10 days before she will exhibit postweaning estrus. The decision when to rebreed is made some time prior to weaning and is mediated by a host of substrates, hormones, and neurotransmitters. Sows with a delayed return to estrus also have a lower pregnancy rate and smaller subsequent litters. If sows lose considerable weight or condition during lactation, a high level of feeding in the postweaning period will improve embryo survival.  相似文献

In cattle and sheep, a progestogenated uterus is susceptible to infections, but this is not well documented for pigs. Therefore, the effects of day of the estrous cycle and progesterone on the susceptibility to uterine infections were evaluated. Gilts (n = 5 per group) were assigned to treatments in 2 x 2 factorial arrays. In Exp. 1, day of cycle and bacterial challenge were main effects. On d 0 or 8, uteri were inoculated with either 70 x 10(7) cfu of Escherichia coli and 150 x 10(7) cfu of Arcanobacterium pyogenes in PBS or with PBS. In Exp. 2, ovariectomy (OVEX) and progesterone treatment were main effects. On d 0, gilts were ovariectomized or a sham procedure was performed. After surgery, gilts received i.m. injections of progesterone (10 mg/5 mL) or 5 mL of safflower oil diluent twice daily. On d 8, gilts were inoculated with the same doses of bacteria as in Exp. 1. In Exp. 1 and 2, vena caval blood was collected for 4 d, after which uteri were collected. Sediment and ability to culture E. coli and A. pyogenes from uterine flushings were used to diagnose infections. Differential white blood cell counts and lymphocyte response to concanavalin A (Con A) and lipopolysaccharides (LPS) were used to measure lymphocyte proliferation. Progesterone, estradiol-17beta, prostaglandin F2alpha, (PGF2alpha), and prostaglandin E2 (PGE2) were measured in vena caval blood. In Exp. 1, d-8 gilts receiving bacteria developed infections, but d-0 gilts receiving bacteria did not. Daily percentages of neutrophils and lymphocytes changed (P < 0.05) with cycle day and bacterial challenge. Basal- and Con A-stimulated lymphocyte proliferation were greater (P < 0.05) for d-0 than for d-8 gilts. Concentrations of PGF2, (P < 0.01) and PGE2 (P < 0.05) increased after bacterial challenge, regardless of stage of the estrous cycle at the time of inoculation. In Exp. 2, OVEX decreased (P < 0.001) and progesterone treatment increased (P < 0.001) progesterone concentrations, and OVEX decreased (P < 0.01) estradiol-17beta. Gilts with ovarian and/or exogenous progesterone developed infections. Daily percentages of neutrophils and lymphocytes changed in response to OVEX, and neutrophils changed (P < 0.05) in response to endogenous and exogenous progesterone. Lymphocyte proliferation in response to Con A and LPS increased (P < 0.05) with OVEX and decreased (P < 0.05) with progesterone treatment. We conclude that endogenous and exogenous progesterone reduce the ability of the uterus in gilts to resist infections.  相似文献

The impact of different patterns of feed restriction between d 1 and 15 of the estrous cycle on subsequent reproductive performance of 23 trios of littermate gilts was tested. Some gilts were fed a high plane of nutrition (HH gilts) throughout the cycle, in contrast to HR gilts, which were restricted from d 8 to 15, and RH gilts, which were restricted from d 1 to 7. During feed restriction, weight gain in RH gilts (2.5 +/- .7 kg) was lower (P = .006) between d 1 and d 7 than in their HH and HR littermates (5.6 +/- .7 and 5.6 +/- .8 kg, respectively) and it was lower (P = .0001) in HR gilts (5.5 +/- .5 kg) between d 8 to d 15 than in their HH and RH counterparts (8.5 +/- .4 and 9.4 +/- .5 kg, respectively). There were no differences in backfat changes among groups. Embryonic survival in HR gilts at d 28 of gestation (68.3 +/- 4.8%) was lower (P < .05) than in HH and RH gilts (83.6 +/- 4.3 and 81.7 +/- 4.5%, respectively). Plasma progesterone concentrations in HR gilts were lower (P < .05) at 48 and 72 h after onset of standing estrus (.82 +/- .2 and 3.6 +/- .5 ng/mL, respectively) than in HH and RH gilts (1.44 +/- .2 and 1.24 +/- .2 ng/mL, 5.0 +/- .4 and 5.0 +/- .5 ng/mL, respectively at 48 and 72 h). No differences in ovulation rate were observed among treatments. Placental area was positively correlated to embryo size at d 28 (embryo size = .0003 x (area) + 18.35; r = .28, P = .03) but placental volume was negatively correlated to the number of embryos in utero (placental volume = -4.317 x (number) + 207.55, r = -.39, P = .002). These data demonstrate that the timing of feed restriction during follicular development has important consequences for subsequent embryo survival, possibly mediated by differences in progesterone concentrations in early pregnancy.  相似文献

This experiment was designed to determine the effects of variations in dietary energy intake on reproductive performance and gene expression of luteal and endometrium tissues in Large White(LW)and Meishan(MS) gilts during early and middle pregnancy. After insemination, 32 LW gilts were assigned to high and low(HE_Land LE_L, 14.23 and 12.56 MJ DE/kg, respectively) diet treatment groups,while 32 MS gilts were allocated to HE_M and LE_M(12.56 and 10.88 MJ DE/kg) groups. Gilts were slaughtered on days 35, 55 and 90 of gestation. The fetal survival and luteal progesterone(P_4) concentration in the HE_Lgroup were higher on day 35 but lower on day 90 of gestation compared with the LE_L group(P 0.05) for LW gilts. However, fetal survival and luteal P_4 concentration on day 35 of gestation were greater(P 0.05) in the LE_M group than in the HE_M group for MS gilts, but no significant difference in mid-gestation was showed. The fetal weights of both breeds were higher for the high energy diets compared with the respective control group on day 90 of gestation(P 0.05). In addition,the m RNA levels of P_4 synthesis-related proteins had correlated with luteal P_4 concentration in both breeds. Further, endometrial levels of uteroferrin(ACP5), retinol-binding protein 4(RBP_4) and secreted phosphoprotein 1(SPP1) m RNA were upregulated in the HE_Lgroup on day 35 of gestation but ACP5 and SPP1 were downregulated on day 55 of gestation compared with the LE_Lgroup(P 0.05) for LW gilts. In MS gilts, diet only affected the expression of SPP1(P 0.05). Our results revealed the differential sensitivity of LW and MS breeds to variations in dietary energy intake. For LW gilts, the HE_Lgroup improved fetal survival on day 35 but a sustained high energy diet decreased fetal survival on day 90 of gestation. The differences in dietary energy intake did not influence fetal survival on day 90 of gestation but the higher energy diet did increase fetal weight in the MS breed compared with the lower energy intake diet. These results may be due to differential luteal secretion activity and endometrium gene expression in these two breeds.  相似文献

This study was conducted to evaluate the effect of estrus induction with gonadotropins on endometrial and conceptus expression of HoxA10, transforming growth factor (TGF) β1, leukemia inhibitory factor (LIF), and prostaglandin H synthase-2 (PGHS-2) during early pregnancy in pigs. Twenty-four prepubertal gilts received 750 IU of pregnant mare serum gonadotropin (PMSG) and 500 IU of human chorionic gonadotropin (hCG) 72 h later. Gilts in the control group (n = 23) were observed daily for estrus behavior. Endometrial tissue samples, conceptuses, blood serum, and uterine luminal flushings (ULFs) were collected on days 10, 11, 12, and 15 after insemination. There was no effect of estrus induction on estradiol content in ULFs, or on ovulation and fertilization rates in studied gilts. However, the content of progesterone in the blood serum was greater in naturally ovulated gilts in comparison to gonadotropin-treated animals on day 12 of pregnancy (P < 0.05). HoxA10 expression was up-regulated in the endometrium of pregnant gilts, with natural ovulation on days 12 (P < 0.05) and 15 (P < 0.001) in comparison to days 10 and 11. When compared to control gilts, administration of PMSG/hCG resulted in decreased expression of endometrial HoxA10, TGFβ, LIF, and PGHS-2 on day 12 of pregnancy (P < 0.05). Conceptus expression of studied factors was not affected by gonadotropin treatment. Overall, these results suggest improper endometrial preparation for implantation in prepubertal gilts induced to ovulate with PMSG/hCG.  相似文献

Synchronization of development between the embryo and uterus is required for successful pregnancy establishment.Transfer of early embryos requires synchrony with the recipient uterus of 2 days or less in sheep,because asynchrony of 3 days or more results in failure of pregnancy recognition signaling for maintenance of corpus luteum (CL) and progesterone (P4) production and/or uterine support of the embryo.The objective was to determine if P4 treatment of recipient ewes would obviate the need for pregnancy recognition signaling and maintain a uterine environment conducive to embryo survival after asynchronous transfer,thereby establishing a universal recipient.Embryos (morulae/blastocysts) were recovered on day 6 from super-ovulated donor ewes.Recipient ewes received 25 mg P4 daily from day 6 post-estrus until 60 days after embryo transfer.Embryos were transferred into recipients on day 6,9,12,18,or 30 post-estrus.The pregnancy rate on day 22 post-transfer was 60% for synchronous transfers to day 6 ewes,44% and 22% for asynchronous transfers to day 9 and 12 ewes,and 0% for asynchronous transfers to day 18 and 30 ewes.On day 39 post-transfer,pregnancy rates remained 60% for day 6 ewes,33% for day 9 ewes,and 0% for day 12,18,and 30 ewes.The P4 treatment did extend the window of uterine receptivity to early embryos in ewes by one day,but did not create a universal recipient.Available results support the idea that a window of uterine receptivity to the conceptus exists in sheep that is independent of pregnancy recognition signaling.  相似文献

Our objective was to study the effects of dietary energy source (fat or starch) on periestrus hormone profiles and embryonal survival in primiparous sows. During lactation, 48 primiparous sows were fed either a starch-rich or a fat-rich diet, at either a high (44 MJ NE/d) or a low (33 MJ NE/d) feeding level. After weaning, all sows received the same amount of feed (31 MJ NE/d from weaning to estrus and 17.5 MJ NE/d from breeding to slaughter) of the same dietary energy source fed during lactation. Around estrus, blood samples were taken to analyze the preovulatory LH surge, estradiol (E2), and progesterone (P4). Sows were inseminated on each day of standing estrus. On d 35 after last insemination, all 35 pregnant sows were slaughtered and their reproductive tracts were removed. The number, weight, and length of the embryos and placentas were determined as well as the weight and length of the uterus. The LH, E2, and P4 profiles were similar for the treatment groups, except for the E2 levels at 16, 12, and 8 h before the LH surge, which were lower in the sows fed the fat-rich diet at a low level. Ovulation rate tended to be higher in sows fed the high compared to the low feeding level during lactation (18.0 vs. 16.2; P = .09), but the number of total and viable embryos as well as embryonal survival rate were not influenced by the treatments. Neither uterine length and weight nor length and weight of the embryos and placentas were affected by treatments. However, after removal of the embryo-placental units, uterine weight was greater in sows fed the high than in those fed the low feeding level during lactation (1.8 vs. 1.6 kg; P = .03). Plasma insulin concentration during lactation was not related to any of the uterine, placental, or embryo traits. Mean progesterone concentration between 24 and 250 h after the LH surge was positively correlated with embryonal survival. Differences in progesterone concentration between sows with high and low embryonal survival were evident from 172 h after the LH surge. From the present study, we conclude that altering feeding level during lactation or dietary energy source from farrowing until d 35 of subsequent pregnancy did not affect embryonic development and embryonal survival.  相似文献

Experiments were performed to determine the effects of components of the GH-IGF axis on conceptus development and postnatal growth in sheep. In Exp. 1, ewes received one of the following treatments: 1) sustained release GH at breeding, 2) sustained release GH at breeding and estradiol-17beta at d 5 and 6, 3) only estradiol-17beta at d 5 and 6, or 4) no treatment. Uteri were flushed on d 7, and flushings were analyzed for content of IGF-I. A single injection of sustained-release bovine GH at breeding increased IGF-I content in uterine luminal flushings compared with control ewes (P < 0.05). Treatment with estradiol-17beta on d 5 and 6 after breeding did not alter IGF-I content compared with control ewes, and it blocked the effect of GH on uterine luminal IGF-I content. In Exp. 2, sustained release GH or no treatment was administered at breeding, and gravid uteri were collected at d 25, 80, or 140 of gestation. On d 80, GH-treated ewes had smaller chorioallantoic weights (P < 0.05) and tended to have more efficient placentae (fetal weight/total placental weight; P = 0.052), with a higher percentage of placental weight as cotyledons (P = 0.068) compared with control ewes. In Exp. 3, ewes were treated with or without sustained release GH at progesterone withdrawal. Lambs from GH-treated ewes were heavier at birth (P < 0.05). Lambs from GH-treated ewes reared as singles, but not lambs reared as multiples, were heavier at 30, 60 (P < 0.05), and 75 d (P = 0.075) of age than lambs from control ewes. In conclusion, ewes treated with sustained-release GH at breeding developed smaller, more efficient placentas, and had larger lambs at birth.  相似文献

Eighty crossbred gilts were assigned randomly to treatments: 1) removal of an ovary and ipsilateral uterine horn (UHO) at 130 d of age and removal of the remaining ovary and uterine horn 12 d post-puberty; 2) UHO at 130 d of age, mated and reproductive tracts recovered when slaughtered at 30 d of gestation; 3) UHO 12 d post-puberty, mated and slaughtered at 30 d of gestation and 4) unoperated controls that were mated and slaughtered at 30 d of gestation. Age of puberty was not affected by treatments. Gilts in treatment 1 had a mean ovulation rate at the pubertal estrus comparable to gilts in treatment 3. But, gilts in treatments 2 and 3 had 16% fewer (P less than .01) corpora lutea at 30 d of gestation than control gilts. Length and weight of the remaining uterine horn at 12 d post-puberty for gilts treated at 130 d of age were similar to the averages of gilts left intact. Gilts with one uterine horn had 2.2 fewer live embryos at 30 d of gestation than control gilts (P less than .01). But, the proportion of corpora lutea represented by live embryos did not differ significantly among treatments. Gilts with one uterine horn had 1.1 fewer live embryos (P less than .15) after adjustment for number of corpora lutea, less uterine space occupied by each embryo (P less than .01) and less total placental membrane per embryo (P less than .05) than control gilts.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The relative importance of factors causing a decrease in sow productivity with systems involving weaning of piglets under 3 weeks of age was studied. The observed reduction in litter size has been attributed to the ovulation rate or to embryo mortality. Subjects were 45 female pigs. 3 lactation lengths were tried: 7, 21, and 42 days. All sows were remated at the 1st postweaning estrus and slaughtered 20 days later to determine ovulation rate and embryo survival. All were fed 1.8 kg/day during gestation. During lactation, the feeding level was increased to a maximum of 6.3 kg/day depending on the number of piglets. Feed level from weaning to remating was 2.7 kg/day. Blood samples taken on 5 occasions from weaning to slaughter were assayed for progesterone. The weaning to estrous interval increased from 6.1 to 8.2 days when lactation length was reduced from 42 to 7 days. Ovulation rates, as determined by luteal count, were similar for the different periods of lactation. Numbers of viable embryos decreased significantly (p less than .05) as lactation length was reduced from 42 to 7 days. The survival of those embryos present decreased also (p less than .01). Embryo mortality increased from 17.3 to 40.4% when the lactation period was reduced from 42 to 7 days. Ovarian weights, uterine weights, and average embryo spacing were the same in all 3 groups. Plasma progesterone levels were low at weaning and remating, higher at 2 days postcoitum, and maximum at 10 days postcoitum. Between treatment groups, plasma progesterone levels were similar but varied markedly within treatment groups. Since ovulation rates were shown to be similar, this factor was eliminated. The interval from weaning to estrus was not considered important. Litter size was shown to be a major limiting factor. Embryo survival during the early period of gestation was shown to be the most important factor in limiting productivity.  相似文献

Influence of initial length of uterus available to each embryo on its subsequent survival and development was determined by systematic restriction of the length available to each potential embryo. Fifty-seven pregnant crossbred gilts were laparotomized at d 3 of gestation, length of uterine horns was measured in situ and corpora lutea (CL) were counted. In Exp. 1, uterine space available to each potential embryo was restricted by ligating one uterine horn 5 cm from the tip per CL. Uteri were examined at d 20, 25 or 50. In Exp. 2, one uterine horn was ligated on d 3 at 10, 20 or 30 cm from the tip per CL and uteri were examined at d 50. Embryos in the restricted section (RS) had a specific mean uterine length available to each potential embryo of 5, 10, 20 or 30 cm. Embryos in the nonrestricted section (NRS) had a variable mean uterine length available to each potential embryo of 44 +/- 4 cm. When embryos were restricted to 5 cm, the proportion of surviving fetuses at d 20, 25 and 50 was 61, 12 and 8%, respectively, whereas in combined NRS it was 82%. When the uterus was examined at d 50 after restricting embryos to 10, 20 or 30 cm/CL, 25, 33 and 52% of fetuses survived; in combined NRS survival was 71%. Each fetus surviving to d 50 in RS was associated with 36 cm of initial uterine length but fetal survival was not associated with number of CL.
