The Eucalyptus gall waspOphelimus maskelli (Ashmead) (Hymenoptera: Chalcidoidea: Eulophidae: Ophelimini) and its introduced parasitoidClosterocerus chamaeleon (Hymenoptera: Chalcidoidea: Eulophidae: Entedoninae) are recorded for the first time in the Mediterranean and Aegean Regions of Turkey. Some morphological characters of adult and larva ofO. maskelli and larva ofC. chamaeleon are described and illustrated. Distributions of the two wasps collected in Turkey are given. Results indicate that the biological control agent,C. chamaeleon, has spread over 1,300 km in the 16 months since its initial release.  相似文献

Ophelimus maskelli (Ashmead) (Hymenoptera: Eulophidae), an invasive species in the Mediterranean area that occurs naturally onEucalyptus camaldulensis at Wagga Wagga in New South Wales, was studied in Israel on the same host plant. The most distinctive characteristic ofO. maskelli as compared with other congeners is the presence of only a single seta on the submarginal vein.O. maskelli shows a clear tendency to oviposit in developed, immature leaves, and it prefers to oviposit on an area of the leaf blade near the petiole. The female lays an average of 109 eggs when maintained on water alone. The egglaying distribution tended towards aggregation, and the same tendency has also been observed within and among trees. The wasp prefers to attack the lower canopy. The gall diameter ranged from 1.2 to 0.9 mm, and gall density from 11.5 to 36.0 galls cm−2, respectively. The typical color of the gall appeared as soon as third instar larva developed. Galls on shaded leaves remained green or greenish-yellow, whereas those that were exposed to the sun were reddish.O. maskelli produces three generations per year in Israel. High flight occurs among the spring generations, starting in early March. The wasps were attracted to green sticky plates much more than to the other tested colors. Heavy galling byO. maskelli results in premature shedding of the leaves, soon after the emergence of the wasps. Among the 84 eucalyptus species tested, the following 14 species were found to be suitable hosts:E. botryoides, E. bridgesiana, E. camaldulensis, E. cinerea, E. globulus, E. gunii, E. nicholii, E. pulverulenta, E. robusta, E. rudis, E. saligna, E. tereticornis andE. viminalis. This study focused on morphological and development characters that can clearly distinguishO. maskelli fromOphelimus eucalypti Gahan, and on examining the effect of crossing between eucalyptus species on the development possibilities of the wasps. Fusarium species are soil-borne fungal pathogens that produce a variety of disease symptoms when attacking crop plants. The mode of root colonization of Eucalyptus viminalis seedlings by a pathogenic F. oxyporum strain (Foeu1) at the ultrastructural level and changes in cell wall pectin during host pathogen interactions are described. Root systems of E. viminalis plants were inoculated with F. oxysporum in an in vitro model system. Hyphae of F. oxysporum adhered to the outer epidermal cell walls through fibrillar material, and after penetration they spread into the internal tissues. They developed intercellularly and intracellularly in the root cortex and invaded vascular tissues. Papillae were induced, and the host plasma membrane ruptured in colonized cells, causing rapid host tissue and cell damage. Changes in distribution and occurrence of nonesterified and methyl-esterified pectins were evaluated after root colonization by F. oxysporum using two monoclonal antibodies, JIM 5 and JIM 7, respectively. Nonesterified pectin in control roots was mainly localized in the epidermal cell walls and middle lamellae in parenchymal cortex, whereas methyl-esterified pectin accumulated more in primary cell walls of the cortex and phloem. Decreases in immunodetected nonesterified and methyl-esterified pectins were associated with extensive plant tissue degradation after root colonization by the pathogenic fungus.  相似文献

Nonpathogenic isolates of Fusarium oxysporum can be successful antagonists of pathogenic forms of the same fungal species that commonly attacks crop plants. The characteristics that distinguish nonpathogenic from pathogenic forms are not well understood. In this study, the mode of root colonization of Eucalyptus viminalis seedlings by a nonpathogenic F. oxysporum strain is described at the ultrastructural level. Root systems of E. viminalis plants were inoculated with nonpathogenic F. oxysporum strain Fo47 in an in vitro model system. Changes in the occurrence of nonesterified and methyl-esterified pectins in colonized E. viminalis roots were evaluated by in situ immunolabeling using two monoclonal antibodies, JIM 5 and JIM 7. Modes of penetration and root colonization patterns in E. viminalis seedlings by the nonpathogenic fungus were similar to those described for pathogenic forms of F. oxysporum. However, root interactions differed in that the nonpathogenic fungus did not induce host tissue damage. No papilla-like appositions were observed in host cells in response to invading hyphae, which did not disrupt the host plasma membrane in many cases, suggesting that a biotrophic relationship was established. Root colonization by the nonpathogenic strain did not induce alteration in JIM 7 labeling of methyl-esterified pectin in E. viminalis cell walls, whereas nonesterified pectin was detected to a significantly greater extent in cell walls of roots colonized by the fungus. Pectin components decreased slightly only at points of hyphal contact with host cells. Because nonpathogenic strains utilize pectin in pure culture, host control over enzyme activity or production by the fungi may at least partly explain their compatible interactions with host tissues.  相似文献

Essential oils extracted fromEucalyptus blakelyi (1,8-cineole, 77.5%),Melaleuca fulgens (1,8-cineole, 56.9%) and 1,8-cineole were shown to have fumigant toxicity against different development stages ofSitophilus oryzae. The eggs ofS. oryzae were the most tolerant, followed by pupae, larvae and adults in that order.M. fulgens oil,E. blakelyi oil and 1,8-cineole at 100 μl per liter of air gave, respectively, LT50 values of 16.2, 17.4 and 9.1 h for adults, 31.1, 19.3 and 17.5 h for larvae, 55.6, 75.2 and 39.7 h for pupae, and required >7 days for eggs. Mycosphaerella leaf disease (MLD) is caused by species of Mycosphaerella and several anamorphic form genera that have been connected to Mycosphaerella. Until recently, MLD of eucalypts was largely ignored in Portugal. However, serious damage to Eucalyptus globulus has been reported since 1999 when frequent and severe defoliation of young trees was observed. The severity of this disease prompted a preliminary study of the Mycosphaerella species associated with major symptoms of a leaf blotch disease in commercial plantations of E. globulus in Portugal, which is presented here. The species were identified by molecular methods based on the ITS1-5.8S-ITS2 cluster, together with morphological characters. In addition to confirming the species previously recorded, Mycosphaerella vespa is reported for the first time from Portugal, while the status of Mycosphaerella grandis remains to be resolved.  相似文献

Four parasitoids emerged from leaves galled by the waspOphelimus maskelli (Ashmead) that were collected in Wagga Wagga, NSW, Australia:Closterocerus chameleon (Girault) and an unidentified Tetrastichinae species (Eulophidae); andStethynium ophelimi Huber andStethynium breviovipositor (Huber (Mymaridae).C. chamaeleon andS. ophelimi were released in eucalyptus plantations infested withO. maskelli in Israel. The recovery of the parasitoids, as well as several aspects of their possible role in control of the galls, were studied. In view of the parasitism of the galler in Wagga Wagga, we suggest that the proportion ofS. ophelimi in the Israeli population ofO. maskelli will increase markedly as soon as the gall wasp population density reaches a very low level.  相似文献

Fourteen species in two families of the order Hymenoptera: Ichneumonidae (1 sp.) and Eulophidae (13 spp.), were recorded during the study ofCameraria ohridella parasitoids at eight localities in Serbia. Among the observed parasitoids, the most significant species wereMinotetrastichus frontalis, Pediobius saulius, Pnigalio agraules andClosterocerus trifasciatus. In addition, at some localities the speciesMinotetrastichus platanellus andCirrospilus talitzkii were also significant. Leaf galls causing severe damage were observed on Eucalyptus camaldulensis in the Gharb‐Maâmora region in 2002 due to Ophelimus maskelli (Ashmead) (Hymenoptera: Eulophidae) reported for the first time in Morocco. Since then, this pest has spread to other eucalyptus stands nationwide. During field investigations, the authors noted that the degree of infestation of Eucalyptus camaldulensis differs from one region to another and were classified on a three‐level scale: low, moderate or high. The study of the life cycle of the insect in the field and in the laboratory revealed the presence of two generations per year. This work has shown that Closterocerus chamaeleon (Hymenoptera: Eulophidae), a parasitoid used successfully in other EPPO countries as a biological control agent, is present in Morocco. The rate of parasitism was very high, which consequently resulted in the lack of damage caused by the gall pest on Eucalyptus camaldulensis in Morocco.  相似文献

The biology ofLixus bardanae (F.) (Coleoptera: Curculionidae) on curly dock (Rumex crispus L.) in northeastern Anatolia (Bayburt, Erzurum and Kars Provinces), Turkey, was studied during the years 2000 and 2001.L. bardanae completes one generation in a year, overwintering as an adult. It feeds on leaves of the host plant. Females lay eggs individually into stems and the young larvae create galleries in stems while feeding. Pupation occurs inside stems in cells fashioned from frass. In late September, adults move into soil and overwinter in an upright position around the roots of the host plant. Infestation levels were found to range between 34% and 84%. The ability of selected strains of fluorescent Pseudomonas spp. to cause induced systemic resistance (ISR) in Eucalyptus urophylla against bacterial wilt caused by Ralstonia solanacearum was investigated. Four of the five strains used can produce salicylic acid (SA) in vitro and, therefore, chemical SA, that is known to induce resistance in many plant species, was used as a reference treatment. Whereas a soil drench with SA did induce systemic resistance in E. urophylla, infiltration of SA into leaves did not. None of the fluorescent Pseudomonas spp. strains caused ISR against bacterial wilt when applied to the soil, but two strains, P. putida WCS358r and P. fluorescens WCS374r triggered ISR when infiltrated into two lower leaves 3–7 days before challenge inoculation. A mutant of strain WCS358r defective in the biosynthesis of the fluorescent siderophore pseudobactin, did not cause ISR, while the purified siderophore of WCS358r did, suggesting that pseudobactin358 is the ISR determinant of WCS358. A siderophore-minus mutant of WCS374r induced the same level of disease resistance as its parental strain, but the purified siderophore induced resistance as well, indicating that both the siderophore and another, unknown, inducing determinant(s) of WCS374r can trigger ISR in Eucalyptus. A possible role of WCS374r-produced SA remains uncertain. Transformation of a siderophore-minus mutant of WCS358 with the SA biosynthetic gene cluster from WCS374 did not enable this transformant to cause ISR in E. urophylla.  相似文献

The clustered hrp genes encoding the type III secretion system in the Japanese strains MAFF301237 and MAFF311018 of Xanthomonas oryzae pv. oryzae were sequenced and compared. The strains differ in their pathogenicity, location, and year of isolation. A 30-kbp sequence comprising 29 open reading frames (ORFs) was identical in its structural arrangement in both strains but differed from X. campestris pv. campestris, X. axonopodis pv. citri, and X. axonopodis pv. glycines in certain genes located between the hpaB-hrpF interspace region. The DNA sequence and the putative amino acid sequence in each ORF was also identical in both X. oryzae pv. oryzae strains as were the PIP boxes and the relative sequences. These facts clearly showed that the structure of the hrp gene cluster in X. oryzae pv. oryzae is unique.  相似文献

Severe rot of leaves, peduncles and flowers caused by Gibberella zeae (anamorph: Fusarium graminearum) was found on potted plants of hyacinth (Hyacinthus orientalis), a liliaceous ornamental, in greenhouses in Kagawa Prefecture, Japan, in January 2001. This disease was named “Fusarium rot of hyacinth” as a new disease because only the anamorph, F. graminearum, was identified on the diseased host plant. The authors contributed equally to this work. The fungal isolate and its nucleotide sequence data obtained in this study were deposited in the Genebank, National Institute of Agrobiological Sciences and the DDBJ/EMBL/GenBank databases under the accession numbers MAFF239499 and AB366161, respectively.  相似文献

In some areas of Japan, yellow spots with white pustules on leaves, stems, petioles, peduncles and calyces were found on Ipomoea nil, I. triloba, I. lacunosa and I. hederacea var. integriuscula. We demonstrated that the diseases on I. nil, I. triloba and I. lacunosa were caused by host-specific strains of Albugo ipomoeae-panduratae and defined three forma speciales of the fungus, respectively, for the three Ipomoea species: “f. sp. nile”, “f. sp. trilobae” and “f. sp. lacunosae”. Because the diseases were new to Japan, we coined the Japanese name “shirosabi-byo”, which means white rust. We also showed that the disease on I. hederacea var. integriuscula was caused by A. ipomoeae-hardwickii. We named this new disease “white rust (shirosabi-byo in Japanese)”.  相似文献

Yeast-like fungi were isolated from lesions on azuki bean (cv. Shin-Kyotodainagon) seeds that had been sucked by bean bugs in Kyoto Prefecture, Japan. On the basis of morphological and physiological characteristics and sequence data of the internal transcribed spacer (ITS) regions including the 5.8S rDNA, these yeasts were identified as Eremothecium coryli and E. ashbyi. Pathogenicity of those yeasts was confirmed by a reinoculation test. To our knowledge, this is the first report of the occurrence of yeast spot in azuki bean in Japan. The nucleotide sequence data reported are available in the GeneBank/EMBL/DDBJ database as accessions AB478291–AB478309 for E. coryli AZC1–19 and AB478310–AB478317 for E. ashbyi AZA1–8.  相似文献

Among the factors affecting the quality and yield of garlic production, blue mold caused by -- Penicillium spp. -- is responsible for economical losses in many countries. Allicin, present in garlic bulbs, has been suggested as having antifungal activity against some Penicillium species. This study was conducted to evaluate the response of garlic accessions against Penicillium hirsutum infection and to compare this response with bulb allicin content. Twelve garlic accessions were inoculated with P. hirsutum, and assayed in greenhouse and growth chamber experiments. Plant growth parameters and the fungal production of conidia were evaluated. Significant differences were found among the accessions. Accessions Castaño and Morado were most resistant whereas AR-I-125 and Fuego were always severely affected by the disease. A low correlation was found (r = 0.17) between allicin content and tolerance, indicating that allicin is not the main factor involved in the resistance against P. hirsutum.  相似文献

Actinidia chinensis and A. deliciosa plants from China, showing a range of symptoms, including vein clearing, interveinal mottling, mosaics and chlorotic ring spots, were found to contain ~300 nm rod-shaped virus particles. The virus was mechanically transmitted to several herbaceous indicators causing systemic infections in Nicotiana benthamiana, N. clevelandii, and N. occidentalis, and local lesions in Chenopodium quinoa. Systemically- infected leaves reacted with a Tobacco mosaic virus polyclonal antibody in indirect ELISA. PCR using generic and specific Tobamovirus primers produced a 1,526 bp sequence spanning the coat protein (CP), movement protein (MP), and partial RNA replicase genes which showed a maximum nucleotide identity (88%) with Turnip vein clearing virus and Penstemon ringspot virus. However, when the CP sequence alone was considered the highest CP sequence identity (96% nt and 98% aa) was to Ribgrass mosaic virus strain Kons 1105. The morphological, transmission, serological and molecular properties indicate that the virus is a member of subgroup 3 of the genus Tobamovirus.  相似文献

The aim of this research was to study levels of resistance to Fusarium basal rot in onion cultivars and related Allium species, by using genetically different Fusarium isolates. In order to select genetically different isolates for disease testing, a collection of 61 Fusarium isolates, 43 of them from onion (Allium cepa), was analysed using amplified fragment length polymorphism (AFLP) markers. Onion isolates were collected in The Netherlands (15 isolates) and Uruguay (9 isolates), and received from other countries and fungal collections (19 isolates). From these isolates, 29 were identified as F. oxysporum, 10 as F. proliferatum, whereas the remaining four isolates belonged to F. avenaceum and F. culmorum. The taxonomic status of the species was confirmed by morphological examination, by DNA sequencing of the elongation factor 1-α gene, and by the use of species-specific primers for Fusarium oxysporum, F. proliferatum, and F. culmorum. Within F. oxysporum, isolates clustered in two clades suggesting different origins of F. oxysporum forms pathogenic to onion. These clades were present in each sampled region. Onion and six related Allium species were screened for resistance to Fusarium basal rot using one F. oxysporum isolate from each clade, and one F. proliferatum isolate. High levels of resistance to each isolate were found in Allium fistulosum and A. schoenoprasum accessions, whereas A. pskemense, A. roylei and A. galanthum showed intermediate levels of resistance. Among five A. cepa cultivars, ‘Rossa Savonese’ was also intermediately resistant. Regarding the current feasibility for introgression, A. fistulosum, A. roylei and A. galanthum were identified as potential sources for the transfer of resistance to Fusarium into onion.  相似文献

Mycosphaerella leaf disease on Eucalyptus is well known in Uruguay but none of the more serious Mycosphaerella spp. and Teratosphaeria spp. causing this disease have yet been found. In the autumn of 2007, more severe defoliation than has been known in the past and associated with symptoms resembling Mycosphaerella infections was observed on Eucalyptus globulus. Isolations and identifications based on morphology and DNA sequence comparisons showed that the causal agent of the defoliation is the well known and serious pathogen Teratosphaeria nubilosa (=Mycosphaerella nubilosa). This is the first record of the pathogen in South America. Using ten microsatellite loci previously developed for T. nubilosa, only one multilocus haplotype was found from 46 T. nubilosa collected isolates. Interestingly, this haplotype was the same as one previously found in Portugal and Spain. The results suggest that T. nubilosa has recently been introduced into Uruguay and that it most likely originated from the Iberian Peninsula where E. globulus is widely planted.  相似文献
