A study was conducted to determine the feasibility of using sclerotia ofSclerotinia sclerotiorum for producing conidia ofConiothyrium minitans in liquid culture. The medium (SST) was made of water containing 2.0, 1.5, 1.0 or 0.5% (w/v) ground sclerotia ofS. sclerotiorum and 100 μgl −1 thiamine hydrochloride (HCl). One ml of conidial suspension (2 × 107 conidia ml−1) ofC. minitans LRC 2534 was inoculated into 100 ml of SST medium or control (thiamine HCl in water) and incubated at 20 ± 2°C on a shaker at 200 rpm. Subsamples were removed periodically and examined under a compound microscope. Conidia in the SST media germinated within 24 h, developed into branched hyphae within 48 h, produced pycnidia after 3–4 days, and the pycnidia released mature conidia after 7 days. Production of conidia varied with the concentration of sclerotia in the SST medium. It was lower (3.6 × 106 conidia ml−1) at 0.5% but higher (1.2 × 108 conidia ml−1) at 2%. The new conidia were viable and the colonies developing from them showed the original morphological characteristics. Field studies were conducted near Lethbridge, Alberta, Canada, in 2001, 2004 and 2005 to determine the efficacy of the antagonistic fungusUlocladium atrum for control of white mold of bean caused bySclerotinia sclerotiorum. Results of the 3 years of field trials showed that, compared with the untreated control, foliar application of a spore suspension ofU. atrum (300 ml m−2 of 106 spores ml−1 suspension) significantly reduced incidence and severity of white mold, increased seed yield and reduced contamination of bean seed by sclerotia ofS. sclerotiorum. The level of control of white mold observed in the treatment ofU. atrum was similar to that of the mycoparasitic fungusConiothyrium minitans, but lower than the fungicide treatments of Ronilan (vinclozolin) at the rate of 1200 g ha−1 per application in 2001, or Lance (boscalid) at the rate of 750 g ha−1 per application in 2004 and 2005. Significant antibacterial activity was observed in the essential oil (E.O.) ofRosa damascena Mill. and the Minimum Bactericidal Concentration (MBC) of the E.O. was determined as 1386.5 μg ml−1 forErwinia amylovora, the causal agent of fire blight disease. Ooze formation on immature pears and lesion formation in artificially inoculated shoots were completely (100%) prevented by the essential oil ofR. damascena (1500 μg ml−1), essential oil ofThymbra spicata var.spicata (500 μg ml−1) and streptomycin (100 μg ml−1). Copper oxychloride plus maneb significantly reduced ooze formation and lesion formation, but the control was less than that obtained with the essential oils or streptomycin. Leaves ofPelargonium domesticum andRuscus hypoglossum infected byBotrytis cinerea Pers., produced 12 and 1.5 nl ethylene/h/g, respectively, 34 days after inoculation; wounded or healthy leaves and phyloclades of them produced much lower amounts. When the fungus was grown on dead leaves it produced negligible amounts of the gas even when supplemented with methionine. Exogenous ethylene enhanced gray mold development in both hosts. Silver thiosulfate, aminooxyacetic acid (AOA) and aminoethoxyvinylglycine significantly inhibited disease development in pelargonium, and the latter two compounds inhibited ethylene production. AOA inhibited disease development and ethylene production in cut rose flowers; calcium ions inhibited disease development whereas the chelator EGTA [ethylene glycol bis-(β-aminoethyl ether)N,N,N′,N′-tetraacetic acid] enhanced it. Disease suppression by an excess of Ca2+ was correlated with repression of ethylene production by the flowers, whereas deficiency in Ca2+ increased disease severity. A study was conducted in two greenhouses with a history of Fusarium crown and root rot (Fusarium oxysporum f.sp.radicis-lycopersici, Forl) and root-knot nematodes (Meloidogyne javanica andM. incognita). During the 2005–06 growing season, the effectiveness of soil disinfestation by solarization in combination with low doses of metham-sodium (500, 750, 1000 and 1250l ha−1) or dazomet (400 g ha−1), was tested against soilborne pathogens and nematodes in an attempt to find a suitable alternative to methyl bromide, which is soon to be phased out. Solarization alone was not effective in the greenhouse with a high incidence ofForl. In the greenhouse with a low level ofForl, all the treatments tested reduced disease incidence, and were therefore considered to be applicable for soil disinfestation. In addition, root-knot nematode density decreased with all the treatments tested in both of the greenhouses.  相似文献

The effect of microclimate variables on development ofClonostachys rosea and biocontrol ofBotrytis cinerea was investigated on rose leaves and crop residues. C.rosea established and sporulated abundantly on inoculated leaflets incubated for 7–35 days at 10°, 20° and 30°C and then placed on paraquat—chloramphenical agar (PCA) for 15 days at 20°C. On leaflets kept at 10°C, the sporulation after incubation on PCA increased from 60% to 93% on samples taken 7 to 21 days after inoculation, but decreased to 45% on material sampled after 35 days. A similar pattern was observed on leaves incubated at either 20° or 30°C. The sporulation ofC. rosea on leaf disks on PCA was not affected when the onset of high humidity occurred 0, 4, 8, 12 or 16 h after inoculation. However, sporulation was reduced to 54–58% on leaflets kept for 20–24 h under dry conditions after inoculation and before being placed on PCA. The fungus sporulated on 68–74% of the surface of leaf disks kept for up to 24 h at high humidity after inoculation, but decreased to 40–51% if the high humidity period before transferral to PCA was prolonged to 36–48 h. The growth ofC. rosea on leaflets was reduced at low inoculum concentrations (103 and 104 conidia/ml) because of competition with indigenous microorganisms, but at higher concentrations (105 and 106 conidia/ml) the indigenous fungi were inhibited. Regardless of the time of application ofC. rosea in relation toB. cinerea, the pathogen’s sporulation was reduced by more than 99%. The antagonist was able to parasitize hyphae and conidiophores ofB. cinerea in the leaf residues. AsC. rosea exhibited flexibility in association with rose leaves under a wide range of microclimatic conditions, and in reducingB. cinerea sporulation on rose leaves and residues, it can be expected to suppress the pathogen effectively in rose production systems.  相似文献

Essential oils extracted fromEucalyptus blakelyi (1,8-cineole, 77.5%),Melaleuca fulgens (1,8-cineole, 56.9%) and 1,8-cineole were shown to have fumigant toxicity against different development stages ofSitophilus oryzae. The eggs ofS. oryzae were the most tolerant, followed by pupae, larvae and adults in that order.M. fulgens oil,E. blakelyi oil and 1,8-cineole at 100 μl per liter of air gave, respectively, LT50 values of 16.2, 17.4 and 9.1 h for adults, 31.1, 19.3 and 17.5 h for larvae, 55.6, 75.2 and 39.7 h for pupae, and required >7 days for eggs. SeveralAlternaria cassiae isolates were recovered from diseased sicklepod plants (Senna obtusifolia) in the southern regions of Brazil. A representative isolate (Cenargen CG593) was tested for its host range under greenhouse conditions. The fungus promoted symptoms in sicklepod, cassava (Manihot dulce), tomato (Lycopersicon esculentum) and eggplant (Solanum melongena) when tested at a spore concentration of 106 spores ml−1. When the plants were inoculated with a suspension of 105 spores ml−1 and held at a dew period of 12 h (cassava) or 18 h (tomato and eggplant), the plants showed symptoms of the disease, but they recovered and continued their normal vegetative growth. The development time and parasitization rate ofDiaeretiella rapae (M’Intosh) onBrevicoryne brassicae (L.) feeding on differentBrassica cultivars was studied in the laboratory at 20°C. The shortest development time from egg to adult parasitoid was 11.6 days on cabbage cv. ‘Yalova 1’ and the longest was 12.1 days on turnip cv. ‘Antep’ and rapeseed cv. local variety. Females lived significantly longer than males on the host plants used in the study. Females and males had the shortest longevity on rapeseed at 11.1 and 5.1 days, respectively. The highest percent parasitism ofB. brassicae byD. rapae was found on cabbage (40.20%), and the lowest was recorded on turnip (32.64%). Our results demonstrate that parasitism rate could be influenced by the plant quality, probably due to the nutritional status of the aphids or to toxic compounds ingested through the plant. Potential antagonists ofFusarium solani f. sp.pisi (Fsp) were selected from soil samples with varying degrees of receptivity to this pathogen. They were tested against Fsp isolate 48 (Fs48), in increasingly complex systems. Most species testedin vitro were able to antagonize Fs48. No relation could be establishedin vitro between the receptivity of the soil from which an isolate originated and its antagonism to Fs48. In soils naturally infested with pea root rot pathogens, which were stored humid at 4°C for a period longer than a year, various isolates ofFusarium, Gliocladium andPenicillium spp. were able to reduce root rot. After sterilization of these soils, onlyGliocladium roseum isolates, added at 105 conidia g–1 dry soil, significantly reduced disease severity and prevented root weight losses caused by Fs48 at 104 conidia g–1 dry soil. In soils in which the biota were activated by growing peas before the assays, doses of 106 and 107 ofG. roseum were required to reduce root rot. In these soils, the antagonistic effects of fluorescent pseudomonad strains from soil of low receptivity to Fsp were variable. Some strains of fluorescent pseudomonads, from soil moderately receptive to Fsp and from highly infested soils, were also able to reduce root rot. Disease suppression by pseudomonad strains was more evident in the absence than in the presence ofAphanomyces euteiches in the root rot pathogen complex. The role of receptiveness of the soil with regard to potential antagonists is discussed.  相似文献

Ageratum conyzoides L. is an annual herb in the tropics and subtropics whose extracts are known to possess pharmacological and biocidal activity. We report on the bioactivity of a secondary metabolite (a chromene) isolated from the shoots ofA. conyzoides against some plant pathogenic fungi. Organic solvent extracts from the shoots were tested for antifungal activity against the plant pathogenic fungiRhizoctonia solani, Sclerotium rolfsii, Botryodiplodia theobromae, Phomopsis theae andFusarium species growingin vitro on potato dextrose agar medium. The cruden-hexane extract completely inhibited the growth ofR. solani andS. rolfsii. Then-hexane extract was chromatographed over a column of silica gel followed by activity-guided fractionation to give an antifungal principle. Structure elucidation by detailed analysis of1H,13C NMR and mass spectroscopy identified the active compound as precocene II. The growth ofR. solani andS. rolfsii was completely inhibited by precocene II at a concentration of 80–100 ppm. The sclerotia ofR. solani andS. rolfsii were also completely suppressed by 150 ppm of precocene II. Sub-culture of these inhibited fungi onto precocene II-free medium restored growth of the fungus, indicating that precocene II is fungistatic. Information on persistence ofBacillus thuringiensis (Bt) is needed to improve the microbial pest management programs against the pine processionary mothThaumetopoea wilkinsoni in pine forests in Israel. The persistence of the microbe under natural conditions of rain and sunlight was evaluated and is documented here for the first time. Pine saplings were sprayed with three commercialBt products, Foray 48B, Delfin WG and Dipel DF, all used at 32,000 IU mg−1 in a formulation with 1% (w/v) of condensed milk. In experiments conducted in November and December of 2004, the saplings were either exposed to rain and sunlight or were sheltered to avoid these environmental factors. The lowest rainfall recorded in the 8-day experiments was 16.5 mm (test 2) and the heaviest was 71.1 mm (test 1). Solar irradiation ranged from 9.4 to 10.9 MJ m−2. The minimum temperature was close to 10°C and the maximum was less than 23°C. Needles of the treated saplings and their controls were sampled after 0, 1, 5 and 8 days, and were fed to 1 st or 2 nd instar larvae. Dipel DF persisted better than Delfin WG and still retained its initial activity of 80–100% mortality on day 8 at low rainfall (test 2). Dripping ofBt from upper to lower branches was quantified with the larval bioassays. 为探究绿色环保的诱抗剂对苎麻生长发育以及抗苎麻夜蛾Arcte coerula特性的影响,选取4种常见且经济的诱抗剂水杨酸、几丁寡糖、壳寡糖1500和壳寡糖1000,分别设置8个浓度处理苎麻后喂食苎麻夜蛾,观察其取食叶面积,在田间处理区半人工释放苎麻夜蛾,观察其为害程度,并在收获时测量苎麻株高和原麻产量。结果表明,4种诱抗剂在适宜浓度下均能显著诱导苎麻对苎麻夜蛾的抗性,田间试验和室内试验结果的一致性较好。其中,80×10-6几丁寡糖稀释液处理对苎麻的诱抗效果最高,苎麻夜蛾为害程度指数仅为8.38,其次是120×10-6壳寡糖1000稀释液处理,为害程度指数为16.71,3.0 g/L壳寡糖1500处理和1.2 mmol/L水杨酸处理对苎麻的诱抗效果最低,为害程度指数分别为27.21和28.07。4种诱抗剂在适宜浓度下都能显著增加苎麻株高和原麻产量,但各诱抗剂处理的增产量无显著差异,原麻产量以40×10-6壳寡糖1000稀释液处理最高,其次为2.0 g/L壳寡糖1500处理,最后为120×10-6几丁寡...  相似文献

During a nematode survey, severe infections of tobacco feeder roots and heavy soil infestations byMeloidogyne incognita race 1 were found in S. Miguel (Azores islands, Portugal). This is the first record ofM. incognita infection of tobacco in Azores. Morphology of various life stages, analysis of the esterase electrophoretic pattern and differential host tests were used for nematode characterization and identification. Nematode-induced mature galls were spherical and/or ellipsoidal and usually contained more than one female, males and egg masses with eggs. Feeding sites were characterized by the development of giant cells that contained granular cytoplasm and many hypertrophied nuclei. Giant cell cytoplasm was aggregated along a thickened cell wall. Vascular tissues within galls appeared disorganized. The relationship between the initial nematode population density and growth of tobacco plants was tested in a glasshouse experiment in which inoculum levels varied from 0 to 512 eggs and juveniles (J2) cm−3 of soil. Seinhorst’s model was fitted to height and top fresh weight data of the inoculated and control plants. Tolerance limits with respect to plant height and fresh top weight of tobacco cv. ‘Erzegovina’ plants toM. incognita race 1 were estimated as 1.25 eggs and J2 cm−3 of soil. Five experimental trials were carried out to test different biological control agents against Fusarium wilt of lettuce, cause byFusarium oxysporum f.sp.lactucae. In the presence of a very high disease incidence, the best results in terms of disease control as well as increased growth response were shown byTrichoderma harzianum T 22 (RootShield), which, at 3 gl −1 of substrate, provided very consistent results.F. oxysporum IF 23 gave good disease control but in two out of five trials reduced the biomass produced. Less consistent, but still significant results were provided byF oxysporium MSA 25, at 3 gl −1 of substrate, and byTrichoderma viride TV 1. The twoF. oxysporum agents Fo 251/2 and Fo 47 and the mixture ofT. harzianum + T. viride (Remedier) partially reduced disease incidence but were less effective than the above mentioned. Less interesting results were offered byStreptomyces griseoviridis (Mycostop). The results obtained show that biological control can play a role in the management of Fusarium wilt of lettuce.  相似文献

Fusarium proliferatum can occur on a wide range of economically important vegetable plants but its role in disease is not always well established. In 2000 and 2001, from forty-one field samples of wilting onion and garlic plants in Serbia, F. proliferatum as the predominant fungal species was isolated from root and bulbs. Seventy isolates were firstly characterized for their sexual fertility and were shown to be mostly members of Gibberella intermedia (sixty-seven of seventy isolates, the remaining three isolates were unfertile), the sexual stage of F. proliferatum (syn. mating population D of G. fujikuroi complex). A selected set of eleven F. proliferatum isolates from both hosts were also tested for their pathogenicity and toxigenicity. Although onion and garlic plants were susceptible to all isolates, onion plants showed a significantly higher disease severity index. Six of the eleven isolates of F. proliferatum produced fumonisin B1 from 25 to 3000 μg g−1, and beauvericin from 400 to 550 μg g−1; ten isolates produced fusaric acid from 80 to 950 μg g−1 and moniliformin from 50 to 520 μg g−1. Finally, all isolates produced fusaproliferin up to 400 μg g−1. These results confirm F. proliferatum as an important pathogen of garlic and onion in Europe and that there is a potential mycotoxin accumulation risk in contaminated plants of both garlic and onion.  相似文献

The development and reproductive rates ofTetranychus cinnabarinus Boisduval (Acari: Tetranychidae), the carmine spider mite, were studied on two strawberry cultivars (Fragaria ×ananassa Duch.; ‘Camarosa’ and ‘Sweet Charlie’) at three NaCl concentrations, 1760, 2400 and 3040 mgl −1, and control. The effects of NaCl application on the contents of plant nutrients, chlorophyll, proline, peroxidase activity (POX) and proteins were assessed. On Camarosa,T. cinnabarinus had a faster development rate with applications of NaCl than without, except for female development at the highest salinity level. The total development time of females increased with the NaCl concentration, whereas total development time of males decreased. The oviposition period and female longevity on Camarosa was significantly longer in the control than with NaCl, whereas daily and total fecundity were significantly higher at all NaCl concentrations than in the control, except for total fecundity at the lowest salinity level. The total development time, oviposition period, female longevity, daily and total fecundity on Sweet Charlie did not differ significantly between NaCl salinity levels and the control. The intrinsic rate of natural increase (r m ) on Camarosa and Sweet Charlie were significantly higher with NaCl than in the control. However, the contents of Na, Cl, P, chlorophyll, proline, POX and protein in the two strawberry cultivars changed depending on NaCl concentration. Chitosan (β-1,4-linked glucosamine oligomer) derived from crab shells conferred a high protection of grapevine leaves against grey mould caused by Botrytis cinerea. Under controlled conditions, it was shown to be an efficient elicitor of some defense reactions in grapevine leaves and to inhibit directly the in vitro development of B. cinerea. Treatment of grapevine leaves by chitosan led to marked induction of lipoxygenase (LOX), phenylalanine ammonia-lyase (PAL) and chitinase activities, three markers of plant defense responses. Dose-response curves show that maximum defense reactions (PAL and chitinase activities) and strong reduction of B. cinerea infection were achieved with 75–150 mg l−1 chitosan. However, greater concentrations of chitosan did not protect grapevine leaves with the same efficiency, but inhibited mycelial growth in vitro. Present results underlined the potency of chitosan in inducing some defense responses in grapevine leaves which in turn might improve resistance to grey mould.  相似文献

Bemisia tabaci (Gennadius), the sweetpotato whitefly, has only one known entomophthoralean pathogen,Erynia radicans (Entomophthoraceae). Two new pathogens have been isolated recently from a glasshouse population of this pest:Conidiobolus coronatus and another, undescribed species ofConidiobolus (Entomophthorales: Ancylistaceae). Artificial inoculation experiments revealed that eggs ofB. tabaci are practically immune to infection by either species. Second-instar larvae are highly resistant, only high doses of conidia causing between <1% and 4.6% mortality. Adults were found to be much more susceptible. Doses of 60 conidia/mm2 ofC. coronatus caused average mortalities ofca 95%. The maximum mortality of adults caused byConidiobolus sp. was much lower,ca 30%, at a dose of 210 conidia/mm2. The incubation period (inoculation to death) for both species, under our experimental conditions, is very short: 18–24 h forC. coronatus and 30 h forConidiobolus sp. Both fungi produced loricoconidia (conidia metamorphosed into resting spores) on cotton leaves and other dry surfaces. This ability allowedConidiobolus sp. to remain viable for 17–21 days on cotton leaves, in glasshouse conditions and in the absence of hosts, whileC. coronatus persisted for 10–14 days.  相似文献

The effect of different soil amendments and biological control agents on soil-borne root diseases that cause significant economic losses in organic and other soil-based tomato production systems (Pyrenochaeta lycopersici and Verticillium albo-atrum) was compared. Organic matter inputs (fresh Brassica tissue, household waste compost and composted cow manure) significantly reduced soil-borne disease severity (measured as increased root fresh weight) and/or increased tomato fruit yield, with some treatments also increasing fruit number and/or size. Soil biological activity also increased with increasing organic matter input levels and there were significant positive correlations between soil biological activity, root fresh weight and fruit yield. This indicates that one mechanism of soil-borne disease control by organic matter input may be increased competition by the soil biota. Chitin/chitosan products also significantly reduced soil-borne disease incidence and increased tomato fruit yield, number and/or size, but had no effect on soil biological activity. Biological control products based on Bacillus subtilis and Pythium oligandrum and commercial seaweed extract (Marinure) and fish emulsion (Nugro)-based liquid fertilisers had no positive effect on soil-borne disease incidence and fruit yield, number and size. The use of ‘suppressive’ organic matter inputs alone or in combination with chitin/chitosan soil amendments can therefore be recommended as methods to control soil-borne diseases in organic and other soil-based production systems.  相似文献
