为明确辽宁省水稻稻瘟病菌Magnaporthe oryzae对稻瘟灵的抗性水平及变化动态,本研究于2017—2019年自辽宁省6个市的6个稻区采集并分离获得187株稻瘟病菌,利用菌丝生长速率法测定这187株稻瘟病菌菌株对稻瘟灵的敏感性及抗性,并采用茎叶喷雾法测定稻瘟灵对水稻稻瘟病的田间防治效果以及该药剂与其它药剂的交互抗性。结果表明:供试187株稻瘟病菌菌株对稻瘟灵的EC50范围为0.94~34.96 μg/mL,左侧正态分布峰内EC50平均值为5.05 μg/mL,稻瘟病菌对稻瘟灵的敏感性分布频率不符合正态分布,表明菌群中已经出现对稻瘟灵敏感性下降的亚群体。187株稻瘟病菌菌株对稻瘟灵的平均抗性倍数为3.70,抗性菌株出现频率为55.61%,平均抗性指数为0.40,抗性水平呈逐年增加趋势。2017—2019年稻瘟灵对水稻叶瘟病的防治效果分别为82.97%、78.86%和81.36%,对水稻穗颈瘟的防治效果分别为79.81%、77.68%和80.20%,说明在现有防控手段下未产生高水平抗性群体。稻瘟灵与肟菌酯、戊唑醇之间无交互抗性。与不同药剂轮换使用的前提下,稻瘟灵仍可作为主要药剂防治稻瘟病。  相似文献

 近几年我国山东省、黑龙江省、辽宁省等多地稻瘟病菌(Magnaporthe oryzae)已对稻瘟灵杀菌剂产生了抗药性。江苏省是我国水稻生产大省,为明确江苏省稻瘟病菌对稻瘟灵的敏感性,本研究分离了2019年采自江苏省6个地区的稻瘟病菌单孢菌株101株,监测到13株稻瘟病菌为稻瘟灵的抗药性菌株,全省稻瘟病菌对稻瘟灵的抗性频率为12.9%。除仪征市未监测到抗药性菌株外,其他5个地区徐州市、淮安市、连云港市、东台市以及宜兴市稻瘟病菌对稻瘟灵的抗性频率分别为40.0%、33.3%、11.5%、5.0%和5.9%。利用菌丝生长速率法对其进行了敏感性测定,结果表明,这13株抗药性菌株的EC50值为15.0~20.6 mg·L-1,其抗性水平在3.6~5.0之间,均表现为低抗水平。进一步对筛选到的稻瘟灵抗性菌株适合度分析发现,只有2株抗性菌株2019-33和2019-45适合度降低,其余抗性菌株均无明显变化。这13株抗性菌株与嘧菌酯、咪鲜胺以及稻瘟酰胺均不存在交互抗性,且抗性相关基因MoIRR未发生任何位点突变。  相似文献

稻瘟病菌对稻瘟灵、异稻瘟净和三环唑的敏感性   总被引:3,自引:1,他引:2  
采用菌丝生长速率法分别测定了57株稻瘟病菌对稻瘟灵和79株稻瘟病菌对异稻瘟净的敏感性,分别建立了敏感基线,并检测了141株大田稻瘟病菌对稻瘟灵和异稻瘟净的抗药性水平;采用活体植株法测定了223株稻瘟病菌对三环唑的敏感性。57株稻瘟病菌对稻瘟灵的平均EC50值为1.4312±0.0867μg/mL,其敏感性频率分布呈连续性单峰曲线,1.4312μg/mL可作为稻瘟病菌对稻瘟灵的敏感基线,在141株大田稻瘟病菌中敏感、低抗和中抗菌株出现的频率分别为21.99%、73.76%和4.26%;79株稻瘟病菌对异稻瘟净的平均EC50值为6.6393±0.2662μg/mL,其敏感性频率分布也呈连续性单峰曲线,6.6393μg/mL可作为稻瘟病菌对异稻瘟净的敏感基线,141株大田稻瘟病菌中敏感、低抗和中抗菌株出现的频率分别为90.07%、7.80%和2.13%。稻瘟病菌对三环唑的EC50值最大为47.6144μg/mL,最小为1.0850μg/mL,供试菌株的EC50值呈近似正态分布,未出现敏感性下降的抗药性亚群体。  相似文献

水直播条件下黑龙江省不同稻区稻瘟病菌致病性分析   总被引:1,自引:0,他引:1  
为明确水直播条件下黑龙江省不同稻区稻瘟病菌Magnaporthe oryzae的致病性分化情况,以24个抗瘟单基因系品种为寄主,来源于2017—2018年黑龙江省水直播稻田的242株稻瘟病菌菌株为接种体,采用离体划伤方法接种,记录病斑反应型,计算有效致病菌株率和抗性频率,并进行聚类分析。结果显示,在水直播条件下,2017年,黑龙江省南部和中东部稻区稻瘟病菌菌株对抗瘟单基因系品种的有效致病菌株率介于8.33%~95.83%和20.83%~95.83%之间,无毒基因出现频率分别为575次和622次;2018年,南部和中东部稻区稻瘟病菌菌株对鉴别体系的有效致病菌株率介于29.17%~95.83%和20.83%~91.67%之间,无毒基因出现频率分别为536次和571次。2017年,黑龙江省南部和中东部稻区稻瘟病菌菌株的致病性相似系数介于0.15~1.00和0.14~1.00之间,以致病性相似系数0.40为阈值,可将菌株分别划分为5个类群和6个类群;2018年,南部和中东部稻区菌株的致病性相似系数介于0.15~0.93和0.26~1.00之间,以致病性相似系数0.40为阈值,可将菌株分别划分为5个类群和4个类群。2017年,抗瘟单基因系品种对黑龙江省南部和中东部稻区稻瘟病菌菌株的抗性频率介于11.29%~88.71%和10.77%~86.15%之间,其中抗瘟单基因系品种IRBL9-W(Pi-9)和IRBLz5-CA(Pi-z5)抗性表现最好;2018年,抗瘟单基因系品种对南部和中东部稻区菌株的抗性频率介于10.34%~82.67%和15.79%~85.96%之间,其中抗瘟单基因系品种IRBL9-W(Pi-9)和IRBLz5-CA(Pi-z5)抗性表现最好。表明水直播条件下黑龙江省稻瘟病菌致病性分化剧烈,稻瘟病菌整体致病力较强,但仍有部分水稻种质抗性较好且相对稳定,基因聚合后抗性会得到进一步提升。  相似文献

辽宁省稻瘟病菌对稻瘟灵的敏感性监测   总被引:2,自引:0,他引:2  
 利用菌丝生长速率法,测定了自辽宁省主产稻区2009-2010年分离的87株稻瘟病菌对稻瘟灵的敏感性。结果表明,稻瘟灵的抑制中浓度EC50值为0.35~7.01 μg/mL,敏感性差异达20倍;且2009年稻瘟病菌对稻瘟灵的敏感性高于2010年。根据野生敏感型病原群体对药剂敏感性呈正态分布的原理,辽宁省稻瘟病菌对稻瘟灵的敏感性基线定为(1.77±0.80)μg/mL,辽宁省主要稻区均有抗稻瘟灵菌株出现,除2010年在铁岭稻产区监测到1株中抗菌株外,大部分稻瘟病菌株都表现为低抗水平。稻瘟病菌不同生理小种群对稻瘟灵的敏感性无明显差别。  相似文献

1990~1997年,用中国稻瘟病菌鉴别寄主鉴定了广东省稻瘟病菌单孢菌株1574份,共鉴定出8群43个生理小种。其中ZC群一直是广东省的优势种群,其出现频率平均为50.2%;ZC13一直是优势小种,其出现频率平均为27.4%,其次是ZG(18.8%)、ZC15(14.5%)和ZB13(9.1%)小种。不同地区不同年份优势小种的类型及其组成不同。  相似文献

 对来自贵州省20个县(市)200余份稻瘟病标样上的146个单孢菌株采用生长速率法测定了稻瘟病菌对稻瘟灵的抗药性。结果表明,贵州省主要稻区都有抗稻瘟灵菌株出现,但大部分抗性菌株的抗药性水平表现为低抗水平。黔东南、贵阳、遵义地区抗性菌株频率明显高于其它几个地区,抗性菌株频率分别为10.96%、9.59%和7.53%。  相似文献

川东南田间稻瘟病菌群体生理小种组成除ZA群外,还包含ZB、ZC、ZD、ZE、ZF、ZG共6群,ZB群略占优势,其致病力有逐年增强的趋势。各小种群体间的变化因主推品种的变化而变复杂。稻瘟病菌群体对所有已知抗病基因均能致病,对Pi-k、Pi-k基因致病力强。通过对杂交稻亲本致病性监测结果表明,杂交稻亲本恢复系密阳46、8480抗瘟性较好,明恢63抗性有增强趋势,多系1号和不育系K17A抗瘟性基本丧失。大面积推广示范显示,汕优多系1号、K优5号等抗性丧失快,岗优22有较强的耐瘟性。  相似文献

分析了三环唑和多菌灵对水稻稻瘟病毒力及对品种抗病性的影响,结果表明三环唑和多菌灵对水稻品种与稻瘟病菌互作有重要影响。当喷雾0.2μg/mL三环唑于13个日本已知抗性基因品种时,原对ZE3和ZG1混合菌株表现抗性的爱知旭表现感病,当喷雾2μg/mL三环唑时,爱知旭则表现抗病,而在0、0.2μg/mL三环唑时均表现抗病的露明则在2μg/mL三环唑时表现感病;喷雾0.2μg/mL多菌灵于13个日本已知抗性基因品种时,ZB1、ZE3、ZG1混合菌株在0μg/mL多菌灵时表现抗性的爱知旭则表现感病,当多菌灵浓度为2μg/mL时,露明又表现为抗病,而在0、0.2μg/mL多菌灵时,对ZG1表现为抗病的草笛则表现感病;13个已知抗性基因品种中其他品种在使用了三环唑和多菌灵后抗性也发生了变化,这说明稻瘟病菌的分布和组成与这类杀菌剂的长期使用可能有一定关系。  相似文献

A technique is described for testing isolates of Septoria tritici from winter wheat for resistance to benzimidazole fungicides. Secondary spores from 23 isolates were tested on Czapek Dox V-8 agar amended with benomyl at 1–10 μg ml. Twenty-one isolates were recovered from eight crops in England in 1984 and two (PBI isolates) were obtained in 1973. Thirteen isolates, including both PBI isolates, were sensitive to benomyl at 1 μg ml?1 and nine were resistant at 10 μg ml?1. The remaining isolate had a low proportion (1:3.7 x 104) of resistant spores. The minimum inhibitory concentration for sensitive isolates was 0.2–0.4 μg ml 1 benomyl and for resistant isolates was more than 1000 μg ml 1. Benomyl-resistant isolates were cross-resistant to carbendazim, thiabendazole and thiophanate-methyl, but not to 12 other fungicides with different modes of action. Field isolates of Botrytis cinerea with moderate levels of resistance to dicarboximide fungicides (ED50 1.0–4.9 μg ml?1) and to dicloran were obtained from glasshouses where vinclozolin and iprodione failed to control grey mould. From sensitive and moderatcly-resistant cultures, laboratory isolates were selected on dicarboximide-amended medium, which were highly resistant to these fungicides (ED50 125->3000 μg ml?1). Conidia of all the resistant isolates germinated well on media amended with 100 μg ml?1 of the dicarboximides vinclozolin, iprodione, procymidone and myclozolin and with 5 μg ml?1 of metomeclan. However, the spores of the moderately resistant isolates did not germinate on 100 μg ml?1 metomeclan while the spores of the highly resistant isolates germinated well. Using media with 100 μg ml?1 of metomeclan to distinguish between the two phenotypes, no highly resistant strain was detected among 312 resistant samples from five cucumber glasshouses with a high frequency of moderately resistant strains. From air-borne inoculum of five glasshouses with 100% resistant populations, 1604 colonies were recovered on vinclozolin-amended (100 μg ml?1) medium and none on metomeclan-amended (100 μg ml?1) medium. It is concluded that strains of B. cinerea highly resistant to dicarboximides are absent from field populations.  相似文献

Mutants of Botrytis cinerea and Ustilago maydis highly resistant to fludioxonil were isolated at a high frequency, after nitrosoguanidine or UV mutagenesis, respectively, and selection on media containing fludioxonil. Tests on the response of mutant strains to high osmotic pressure resulted in the identification of two fludioxonil-resistant phenotypes (FLDosm/s and FLDosm/r), regarding the sensitivity to high osmolarity. Approximately 95% of fludioxonil-resistant mutants were found to be more sensitive to high osmotic pressure than the wild-type parent strains. Genetic analysis of phenylpyrrole-resistance in the phytopathogenic basidiomycete U. maydis, showed that fludioxonil-resistance was coded by three unlinked chromosomal loci (U/fld-1, U/fld-2 and U/fld-3), from which only the U/fld-1 mutation coded an osmotic sensitivity similar to that of the wild-types. Cross-resistance studies with fungicides from other chemical groups showed that the mutations for resistance to phenylpyrroles affect the sensitivity of mutant strains to the aromatic hydrocarbon and dicarboximide fungicides, but not to the benzimidazoles, anilinopyrimidines, phenylpyridinamines, hydroxyanilides or the sterol biosynthesis inhibiting fungicides. A study of fitness parameters in the wild-type and fludioxonil-resistant mutants of B. cinerea, showed that all osmotic sensitive (B/FLDosm/s) isolates had significant reductions in the characteristics determining saprophytic fitness such as mycelial growth, sporulation, conidial germination and sclerotial production. Contrary to that, with the exception of mycelial growth, the fitness parameters were unaffected or only slightly affected in most of the osmotic resistant (B/FLDosm/r) isolates. Tests on cucumber seedlings showed that the osmotic-sensitive strains were significantly less pathogenic compared with the wild-type and B/FLDosm/r strains of B. cinerea. Preventative applications of the commercial products Saphire 50 WP (fludioxonil) and Rovral 50 WP (iprodione) were effective against lesion development on cotyledons by the wild-type and the mutant strains of B. cinerea that were resistant to the anilinopyrimidine cyprodinil (B/CPL-27) and to the hydroxyanilide fenhexamid (B/FNH-21), but ineffective, even at high concentrations, against disease caused by the fludioxonil-resistant isolates (B/FLD) and a mutant strain resistant to the dicarboximide iprodione (B/IPR-1). Experiments on the stability of the fludioxonil-resistant phenotype showed a reduction of resistance, mainly in osmotic-sensitive isolates, when the mutants were grown on inhibitor-free medium. A rapid recovery of the high resistance was observed after mutants were returned to the selection medium. Studies on the competitive ability of mutant isolates against the wild-type parent strain of B. cinerea, by applications of a mixed conidial population, showed that, in vitro, all mutants were less competitive than the wild-type strain. However, the competitive ability of osmotic-resistant mutants was higher than the osmotic-sensitive ones. Furthermore, competition tests, in planta, showed a significant reduction of the frequency of both phenylpyrrole-resistant phenotypes, with a respective increase in the population of the wild-type strain of the pathogen.  相似文献

番茄叶霉病菌对氟硅唑敏感基线及抗药性风险研究   总被引:2,自引:0,他引:2  
从辽宁省不同地区未使用过氟硅唑的保护地中采集番茄叶霉病叶,经单孢分离获得43株叶霉病菌。采用茵丝生长速率法测定其对氟硅唑的敏感性。结果表明,菌株间存在较大差异,EC50值最低为0.151 9μg/mL,最高达4.647 5 μg/mL。所有菌株的EC50总平均值为1.253 1μg/mL,可作为番茄叶霉病菌对氟硅唑的敏感基线。野生菌株经紫外线照射,获得两个抗药性突变菌株,其抗药性倍数分别为6.0和7.6。突变菌株的菌丝生长速率及产孢能力显著低于原野生菌株。  相似文献

水稻稻瘟病菌对烯肟菌胺的抗性风险评估及抗性机制初探   总被引:2,自引:0,他引:2  
 采用菌丝生长速率法测定了100株采自我国主要水稻产区的水稻稻瘟病菌对烯肟菌胺的敏感性, 结果表明, 其EC50分布于0.011 1~0.295 6 μg·mL-1, 平均EC50=(0.078 6±0.056 1) μg·mL-1。供试菌株对烯肟菌胺的敏感性分布呈单侧峰曲线, 未出现抗药性亚群体, 可将该曲线作为稻病瘟菌对烯肟菌胺的敏感性基线。通过室内药剂驯化获得了7株抗药突变体, 突变频率为1.11×10-4, 其中2株高抗突变体NJ0811-I和A10的抗性水平大于1 000倍, 抗药性性状能稳定遗传, 致病力显著弱于其亲本菌株;5株低抗突变体抗性水平在2.05~4.55倍之间, 抗药稳定性差, 适合度与亲本无显著性差异。交互抗药性结果表明, 烯肟菌胺与嘧菌酯存在正交互抗药性, 与田间防治稻瘟病常用药剂稻瘟灵、异稻瘟净无交互抗药性。综合分析表明, 稻瘟病菌对烯肟菌胺可能存在低到中等抗性风险。进一步克隆了抗药突变体及其亲本的cytb基因, CYTB氨基酸序列比对结果表明, 2株高抗突变体均在143位由甘氨酸突变为丝氨酸(G143S), 建立了高抗菌株的AS-PCR分子检测方法;而5株低抗突变体cytb基因未发生点突变, 推测可能存在其他的抗性分子机制。  相似文献

The baseline sensitivity ofFusarium graminearum Schwade [teleomorph =Gibberella zeae (Schweinitz) Petch] to the fungicide JS399-19 (development code no.) [2-cyano-3-amino-3-phenylacrylic acetate] and the assessment of risk to JS399-19 resistancein vitro are presented. The mean EC50 values for JS399-19 inhibiting mycelial growth of three populations of wild-typeF. graminearum isolates were 0.102±0.048, 0.113±0.035 and 0.110±0.036 μg ml−1, respectively. Through UV irradiation and selection for resistance to the fungicide, we obtained a total of 76 resistant mutants derived from five wild-type isolates ofF. graminearum with an average frequency of 1.71 × 10−7% and 3.5%, respectively. These mutants could be divided into three categories of resistant phenotypes with low (LR), moderate (MR) and high (HR) level of resistance, determined by the EC50 values of 1.5–15.0 μg ml−1, 15.1–75.0 μg ml−1 and more than 75.0 μg ml−1, respectively. There was no positive cross-resistance between JS399-19 and fungicides belonging to other chemical classes, such as benzimidazoles, ergosterol biosynthesis inhibitors and strobilurins, suggesting that JS399-19 presumably has a new biochemical mode of action. Although the resistant mutants appeared to have comparable pathogenicity to their wild-type parental isolates, they showed decreased mycelial growth on potato-sucrose-agar plates and decreased sporulation capacity in mung bean broth. Nevertheless, most of the resistant mutants possessed fitness levels comparable to their parents and had MR or HR levels of resistance. Mutants of Botrytis cinerea with moderate and high resistance to pyraclostrobin, a Qo inhibitor of mitochondrial electron transport at the cytochrome bc 1 complex, were isolated at a high mutation frequency, after nitrosoguanidine mutagenesis and selection on medium containing pyraclostrobin and salicylhydroxamate (SHAM), a specific inhibitor of cyanide-resistant (alternative) respiration. Oxygen uptake in whole cells was strongly inhibited in the wild-type strain by pyraclostrobin and SHAM, but not in the mutant isolates. Cross-resistance studies with other Qo and Qi inhibitors (QoIs and QiIs) of cytochrome bc 1 complex of mitochondrial respiration showed that the mutation(s) for resistance to pyraclostrobin also reduced the sensitivity of mutant strains to other QoIs as azoxystrobin, fluoxastrobin, trifloxystrobin and picoxystrobin, but not to famoxadone and to the QiIs cyazofamid and antimycin-A. An increased sensitivity of pyraclostrobin-resistant strains to the carboxamide boscalid, an inhibitor of complex II, and to the anilinopyrimidine cyprodinil, a methionine biosynthesis inhibitor, was observed. Moreover, no effect of pyraclostrobin resistance mutation(s) on fungitoxicity of the hydroxyanilide fenhexamid, the phenylpyrrole fludioxonil, the benzimidazole benomyl, and to the phenylpyridinamine fluazinam, which affect other cellular pathways, was observed. Study of fitness parameters in the wild-type and pyraclostrobin-resistant mutants of B. cinerea showed that most mutants had a significant reduction in the sporulation, conidial germination and sclerotia production. Experiments on the stability of the pyraclostrobin-resistant phenotype showed a reduction of resistance, mainly in moderate resistant strains, when the mutants were grown on inhibitor-free medium. However, a rapid recovery of the resistance level was observed after the mutants were returned to a selective medium. Studies on the competitive ability of mutant isolates against the wild-type parent strain, by applications of a mixed conidial population, showed that, in vitro, all mutants were less competitive than the wild-type strain. However, the competitive ability of high resistant mutants was higher than the moderate ones. Pathogenicity tests on cucumber seedlings showed that all mutant strains tested exhibited an infection ability similar with the wild-type parent strain. Preventive applications of the commercial product of F-500 25EC (pyraclostrobin) were effective against lesion development on cotyledons by the wild-type, but ineffective, even at high concentrations, against disease caused by the pyraclostrobin-resistant isolates. Boscalid (F-510 50WG) was found equally effective against the disease caused by the wild-type or pyraclostrobin-resistant mutants. This is the first report indicating the appearance of B. cinerea strains resistant to QoI fungicides by the biochemical mechanism of site modification and the risk for field resistance.  相似文献

In Calabria (southern Italy), control of crown and root rot of capsicum caused by Phytophthora capsici has relied primarily on soil drenches of metalaxyl. However, severe outbreaks occur every year in glasshouse crops, in which the practice of using plastic mulch and furrow irrigation favours the disease. Single‐hypha isolates of P. capsici collected in Calabria in 1992/1998 were tested in vitro for their level of sensitivity to metalaxyl. Isolates of other species of Phytophthora were used as reference. Fungicide sensitivity was determined by plating mycelial plugs onto potato dextrose agar amended with metalaxyl, at final concentrations ranging from 0.1 to 1000μg mL?1 a.s. Inhibition of radial growth (%) was determined when colonies on unamended medium had covered approximately two‐thirds of the plate. The ED50 values for inhibition of mycelial growth of P. capsici isolates ranged from 1.41 to44.6μg mL?1 a.s. More than 80% of the P. capsici isolates from commercial plastic‐house crops in Calabria showed a moderate level of resistance as they were inhibited less than 60% at 5 μg mL?1 but more than 60% at 100μg mL?1  相似文献

 Laboratory mutants of Cochliobolus heterostrophus resistant to iprodione were obtained after chemical mutageneses. All the mutants were able to grow on the medium amended with iprodione 100 μg/ml. They showed positive cross-resistance to procymidone and fludioxonil and were sensitive to high osmolarity. Crosses between the mutant and a wild-type strain revealed that the fungicide resistance and osmotic sensitivity traits were inherited by their offspring in a 1 : 1 mutant/wild type ratio, indicating that the mutant phenotypes in these strains were due to alteration at a single gene locus. Results from allelism tests indicated that three genes (Dic1, Dic2, Dic3) conferred the mutant phenotypes. Among them, Dic1 mutant strains were classified into three types on the basis of their phenotypes. The first type was moderately resistant to the fungicides and less sensitive to osmotic stress than the other Dic1 mutant strains. The second type showed moderate fungicide resistance, but growth was inhibited under lower osmotic stress (50 mM KCl). The other Dic1 mutant strains grew well on medium containing iprodione and fludioxonil even at a concentration of 100 μg/ml and were highly sensitive to osmotic stress. The Dic2 and Dic3 mutant strains had moderate resistance to the fungicides with low-level osmotic sensitivity. The Dic1 gene was epistatic to Dic2 and Dic3 for fungicide resistance and hypostatic to them for osmotic sensitivity. These results suggest that the osmoregulatory system is involved in fungicide resistance in laboratory mutants of C. heterostrophus. Received: March 14, 2002 / Accepted: August 13, 2002  相似文献
