Induction of the microsomal oxidase system by dietary phenobarbital and β-naphthoflavone was examined in three blowflies, Phormia regina (Mg.), Lucilia illustris (Mg.), and Eucalliphora lilica (Walk.). Responses were similar in adults and larvae of all species. Phenobarbital increased cytochrome P-450 levels up to 9-fold and aldrin epoxidase up to 138-fold. Increases in cytochrome P-450 and aldrin epoxidase caused by β-naphthoflavone were minor relative to those produced by phenobarbital. In toxicity experiments with carbaryl and propoxur tolerance was associated with the amount of microsomal oxidase activity. Using piperonyl butoxide to synergize carbaryl and propoxur there was no clear indication for the use of either the synergist ratio or synergist difference as an indicator of microsomal oxidase activity.  相似文献

A mitogen-activated protein kinase (MAPK) pathway has been demonstrated as a key pathway in plant defense against pathogen attacks. With proteomics approaches, we specifically studied activation events downstream of a MAPK kinase, tMEK2, in tomato. Overexpression of a constitutively activated tomato MAPK kinase gene (tMEK2MUT) enhanced resistance of transgenic tomato lines to the virulent bacterial pathogen Pseudomonas syringae pv. tomato. Pathogenesis-related genes, PR1b1, β-1,3-glucanase, and endochitinase were up-regulated by tMEK2MUT. Two-dimensional electrophoresis and matrix-assisted laser desorption/ionisation-time-of-flight-mass spectrometry analysis of total soluble leaf proteins indicated that β-1,3-glucanase and endochitinase are among the up-regulated proteins in these transgenic plants. Co-expression studies using a transient gene expression system have indicated that β-1,3-glucanase and endochitinase genes up-regulated by tMEK2MUT were down-regulated by different specific phosphatases through dephosphorylation of certain downstream signaling molecules. Our observations indicate that increased products of β-1,3-glucanase and endochitinase genes downstream of tMEK2 may play an important role in achieving disease resistance.  相似文献

Risk assessment of herbicides towards non-target plants in Europe is currently based solely on tests on algae and floating aquatic plants of Lemna sp. Effects on terrestrial non-target species is not systematically addressed. The purpose of the present study was to compare combination effects of herbicide mixtures across aquatic and terrestrial test systems, and to test whether results obtained in the traditional aquatic test systems can be extrapolated to the terrestrial environment. This was done by evaluating ten binary mixtures of nine herbicides representing the seven most commonly used molecular target sites for controlling broadleaved weeds. Data were evaluated statistically in relation to the concentration addition model, and for selected concentrations to the independent action model. The mixtures were tested on the terrestrial species Tripleurospermum inodorum (L.) Schultz-Bip. (Scentless Mayweed) and Stellaria media (L.) Vill. (Common Chickweed), and on the aquatic species Lemna minor L. (Lesser duckweed) and the alga Pseudokirchneriella subcapitata (Korschikov) Hindak. For the two mixtures of herbicides with the same molecular site of action, the joint effect was additive. For the eight mixtures of herbicides with different sites of action, two of the mixtures were consistently antagonistic across species, while for the remaining six mixtures the joint effect depended on the species tested. This dependence was, however, not systematic, in the sense that none of the species or test systems (terrestrial versus aquatic) had a significantly higher probability of showing synergistic or antagonistic joint effects than others. Synergistic interactions were not observed, but approximately 70% of the mixtures of herbicides with different sites of action showed significant antagonism. Hence, the concentration addition model can be used to estimate worst-case effects of mixtures of herbicides on both terrestrial and aquatic species. Comparing the sensitivity of the species to a 10% spray drift event showed that the terrestrial species were more vulnerable to all herbicides compared with the aquatic species, emphasising the importance of including terrestrial non-target plants in herbicide risk assessment.  相似文献

Overgrazing is regarded as one of the key factors of vegetation and soil degradation in the arid and semi-arid regions of Northwest China.Grazing exclusion(GE)is one of the most common pathways used to restore degraded grasslands and to improve their ecosystem services.Nevertheless,there are still significant controversies concerning GE’s effects on grassland diversity as well as carbon(C)and nitrogen(N)storage.It remains poorly understood in the arid desert regions,whilst being essential for the sustainable use of grassland resources.To assess the effects of GE on community characteristics and C and N storage of desert plant community in the arid desert regions,we investigated the community structure and plant biomass,as well as C and N storage of plants and soil(0-100 cm depth)in short-term GE(three years)plots and adjacent long-term freely grazing(FG)plots in the areas of sagebrush desert in Northwest China,which are important both for spring-autumn seasonal pasture and for ecological conservation.Our findings indicated that GE was beneficial to the average height,coverage and aboveground biomass(including stems,leaves and inflorescences,and litter)of desert plant community,to the species richness and importance values of subshrubs and perennial herbs,and to the biomass C and N storage of aboveground parts(P<0.05).However,GE was not beneficial to the importance values of annual herbs,root/shoot ratio and total N concentration in the 0-5 and 5-10 cm soil layers(P<0.05).Additionally,the plant density,belowground biomass,and soil organic C concentration and C storage in the 0-100 cm soil layer could not be significantly changed by short-term GE(three years).The results suggest that,although GE was not beneficial for C sequestration in the sagebrush desert ecosystem,it is an effective strategy for improving productivity,diversity,and C and N storage of plants.As a result,GE can be used to rehabilitate degraded grasslands in the arid desert regions of Northwest China.  相似文献

Four annual experiments with garlic (Allium sativum) were established in fields naturally infested with Sclerotium cepivorum, causal agent of garlic white rot, to investigate the effect of time of planting on yield. Initial stands were unaffected by time of planting. Final stands and yields were highest when garlic was planted approximately 1 month earlier (28 January-15 February) when soils were warmer than at the traditional time of planting (15 March-15 April). When garlic was planted at the traditional time, final stands and yields were reduced 56-92% primarily from the attack of S. cepivorum. A 100% loss in yield occurred in some crops planted in cool soils in late March. With an early planting (15 January), final stands were similar to the plantings of 28 January and 15 February, but yields were reduced by half because of a much lower mean weight of the harvested bulbs. In a late planting (15 May), the final stand was 75% lower with correspondingly lower yield than that obtained with the early plantings. The largest bulbs (21.6 g) were obtained from the planting on 28 February; however, the yields from this planting were reduced by half from the planting 2 weeks earlier because of the 50% reduction in final stand. Early planting of garlic is recommended as an important management strategy to avoid white rot in areas with soils infested with sclerotia of S. cepivorum.  相似文献

Recently, Elsinoë piri has become an important pathogen in unsprayed and organic apple orchards. The increasing demand for organic apples as well as the limited use of fungicides imposed by European legislation has turned this pathogen, which has been known for over a century in Europe, into a resurgent phytosanitary threat. It also represents a new trait to be taken into account in apple breeding programmes. As E. piri is extremely difficult to isolate and causes symptoms that can be confused with those caused by other pathogens, a quantitative PCR (qPCR) test has been developed based on the internal transcribed spacer (ITS) region of the rDNA gene. This test, which displays a low limit of detection, allows the early detection of the fungus in apple leaves. As a quantitative method, it is a promising tool for breeding purposes as it can be used on leaves to assess the level of quantitative resistance of apple cultivars to the disease. The molecular test is also useful to gain knowledge on the epidemiology of this re-emerging fungal disease. Spore trapping conducted in 2015, 2016 and 2020 using Rotorod devices and the qPCR test showed that airborne inoculum is released at the end of the growing season. Tests were also carried out to demonstrate that the pathogen was detected in buds during winter. A comparison of 18 E. piri isolates based on the sequencing of five genome regions highlighted a high genotypic diversity. All these isolates were detected with the qPCR test developed in this study.  相似文献

There were indications that endo-1,3--glucanase (1,3-(1,3;1,4)--D-Glucan 3(4)-glucanohydrolase (EC 3.2.1.6)) and cellulase (1,4-(1,3;1,4)--D-Glucan 4-glucanohydrolase (EC 3.2.1.4)) activity of Trichoderma harzianum Rifai isolate T3 were induced in sphagnum peat moss cultivations and dual culture experiments by the presence of Pythium ultimum. Further, P. ultimum stimulated the germination of Trichoderma conidia. Endo-1,3--glucanase and cellulase were purified from T. harzianum isolate T3, known to control Pythium damping-off of cucumber seedlings. The enzymes were purified from the culture filtrate of the fungus by gel filtration and isoelectric focusing. The purified endo-1,3--glucanase was a small protein with a molecular mass of 17 kilodaltons and a pI of 5.0. Two cellulases were purified to homogeneity and had molecular masses of 40 and 45 kilodaltons respectively, and pI's of 6.4 and 7.6 respectively. Germination of encysted zoospores and elongation of germ tubes of a plant pathogenic Pythium isolate were inhibited by low concentrations of the purified enzymes. A strong synergistic effect was observed on the inhibition of cyst germination by a combination of the endo-1,3--glucanase and the fungicide Fongarid. Finally, a time-course study of colonization of the rhizosphere of cucumber seedlings showed that the active fungal mycelial biomass of a GUS-transformant of T. harzianum isolate T3 increased over four weeks. Trichoderma appeared to colonize healthy roots only superficially, whereas the mucilage of the root hairs and of distal parts of wounded areas or broken parts of the roots, were extensively colonized.  相似文献

UV irradiation enhanced the resistance of grapefruit against the development of green mold décay caused byPenicillium digitatum, the main postharvest pathogen of citrus fruit, and significantly inhibited the fungus’ growth at the fruit wound sites. Immunoblotting analysis using specific citrus chitinase and β-1,3-endoglucanase antibodies, showed that UV irradiation, wounding of the fruit, or a combination of these two treatments, induced the accumulation of a 25 kD chitinase protein in the fruit’s peel tissue. On the other hand, UV irradiation or wounding of the fruit alone was unable to induce the accumulation of 39 and 43 kD β-1,3-endoglucanase proteins, but the combination of the two treatments increased these protein levels. It is suggested that both chitinase and β-1,3-endoglucanase may play a role in the UV-induced resistance of grapefruit againstP. digitatum. The effects of short-term nutrient additions (NPKμ, 10 g·m- 2) and disturbances on the species diversity, functional traits, soil properties, and community assembly mechanisms of the herbaceous community in the Urat desert steppe were examined. A mixed linear model, principal component analysis, and null model methods were utilized and both disturbance and interaction treatments were found to significantly reduce species richness and β diversity. Short-term nutrient additions significantly reduced soil pH but increased soil electrical conductivity. Six functional traits of the dominant species (i.e. Stipa glareosa, Peganum harmala, Salsola collina, Corispermum mongolicum) and six community-weighted mean significant changes under the disturbance and nutrient addition treatments were identified. The disturbance and nutrient additions promoted the transformation of the dominant species in the community from conservative strategies to acquisitive strategies. The results of the two null models revealed that community assembly under the control tended to be a stochastic process, while the disturbance and interaction treatments tended to be deterministic processes, and that of the nutrient addition treatments tended to be weaker deterministic processes. Plant communities in the desert steppe responded and adapted to the nutrient additions and disturbances through changes in the dominant species and their crucial functional traits. © 2023 Chinese Academy of Sciences. About 900.000 ha of Egypt’s agricultural lands are suffering from salinity build-up problem. Therefore, two field experiments were conducted in split-split plot design with four replications to study α?tocopherols foliar application with 100 and 200 ppm and hand hoeing (two and three hand hoeing, plus unweeded) and their interactive effects on growth, osmoprotectants, mineral contents, seed yield, yield components, and nutritive value of the lupine plants and its associated weeds grown under salt-affected soil at 1.30, 3.20 and 4.23 dS m?1. Three hand hoeing treatment achieved the highest weed depression expressed in the lowest number and dry matter of broad-leaved, narrow-leaved and total weeds, and reduction % in dry matter were 80.75, 78.25 and 79.26 % compared with unweeded, respectively. Salinity stress significantly decreased total chlorophyll content, leaf area index and plant dry weight, while α?tocopherol and hoeing increased in all previous traits relative to corresponding controls. Lupine seed yield, yield components, and oils, carbohydrates, proteins, carotenoids and alkaloids in the yielded seeds were significantly reduced by increasing salinity levels, meanwhile hoeing and α?tocopherol treatments improved them. Total essential amino acids were decreased by increasing salinity levels and increased by increasing α?tocopherol concentrations and number of hoeing treatments. It could be concluded that α?tocopherol especially at 200 ppm partially alleviated the harmful effects of salinity stress on the growth of lupine plants and nutritive value of the yielded seeds. Three hand hoeing treatment was the optimum and safe method in controlling weeds associated with lupine plant grown under salinity stress.  相似文献

Potato and tomato are the two major hosts for Phytophthora infestans causing late blight. The susceptibility of leaves and whole plants of Solanum nigrum, S. villosum, and S. scabrum to infection by P. infestans was tested under laboratory conditions. Out of 39 plants representing 38 different S. nigrum accessions, 16 were highly resistant (seven accessions did not show any symptoms of infection, nine were highly resistant showing necrotic lesions in the place of infection), and 23 plants of S. nigrum were colonized by, at least, 1 of the 2 isolates of P. infestans (17 accessions were infected with two P. infestans isolates, and 6 accessions showed different reactions depending on the isolate used for inoculation). Three accessions of S. villosum, and one accession of S. scabrum were tested and did not show any symptoms of infection. The majority of S. nigrum accessions infected by P. infestans in a detached leaf assay were also infected in the whole plant assay. The reaction of field- and greenhouse-grown plants to inoculation with P. infestans in detached leaf assays was similar, but in some cases leaves from field-grown plants reacted as resistant in comparison with the leaves from greenhouse-grown plants, which were susceptible.  相似文献

The objective of the present study was to investigate the role of α tocopherol and selenium on malathion induced hepatic damage, and antioxidant defense in chicks. The chicks were divided into three groups. First group received malathion 10 mg/kg BW, orally for 60 days, the second group received the same dose of malathion but was supplemented with α tocopherol and selenium for 60 days and the third group served as the control. A compromised antioxidant capacity as evidenced by increased levels of erythrocytic lipid peroxidation and decreased concentration of vitamin E and decreased activity of glutathione peroxidase was observed in chicks following the administration of malathion. An improved antioxidant status was observed in chicks of second group with α tocopherol and selenium supplementation including higher concentration of vitamin E, increased activity of glutathione peroxidase and lower levels of lipid peroxidation. Histopathological studies of liver in the chicks which received malathion exhibited, moderate to severe degenerative and necrotic changes in the hepatocytes. The correlation of decreased antioxidant status of chicks with degenerative changes in liver suggests that lipid peroxidation may be one of the important mechanism in the chronic toxicity of malathion. The results indicate that α tocopherol and selenium were effective in partially alleviating degenerative changes induced by malathion in the liver of chicks by attenuating processes leading to lipid peroxidation.  相似文献

Abstract

Cohorts of newly laid egg masses of two pod‐sucking bugs, Nezara viridula and Piezodorus hybneri (Hemiptera: Pentatomidae), were exposed to naturally occurring predators and parasitoids on soybean in unsprayed fields at four sites in Northern Sumatra. The fate of the young stages was recorded until the late first instar, before the nymphs start to disperse and feed. Partial life tables showed that mortality of N. viridula until the late first instar was 50–87%, of which 18–65% occurred during the egg stage and was mainly caused by predation; 2–26% of the eggs died of parasitism. P. hybneri which was present at only two sites, showed a 73–89% mortality until the late first instar, of which 69–85% was during the egg stage and, again, mainly caused by predation; 15–24% of the eggs died of parasitism. To evaluate the role of individual egg predator species in the field, we conducted hourly observations of exposed egg masses of the two pests. Dolichoderus sp., Solenopsis geminata (Hymenoptera: Formicidae), Paederus sp. (Coleoptera: Staphylinidae) and several gryllids (Orthoptera) were most commonly encountered feeding on the eggs. An index of ‘predation pressure”; was calculated from the densities of predators in the field and from an estimate of their attack rate based on field observations of predator feeding. Two sites with low predator pressures had low predation rates, while the two sites with high predation pressures had considerably higher predation rates. The implications of the findings for IPM training in soybean are discussed.  相似文献

Thirty-seven cassava genotypes from Benin, including advanced breeding lines, were tested for their reaction to bacterial blight in the forest–savanna transition, wet savanna and dry savanna zones of Benin. Sixteen genotypes were repeated in 12 environments. In year 1998, genotypes RB92164, RB92022, TMS30572, BEN86004, RB92033 and Dangbo2, and in year 2000, genotypes RB92202, RB92151, RB92132 and TMS30572 were resistant in one ecozone. Among the more resistant genotypes, CAP94030, BEN86040, RB89509, RB92132 and TMS30572 showed low interaction across environments and were most stable in disease reaction. Ten genotypes were classified as high yielding across environments. Among the more resistant group of genotypes, only TMS30572 and RB89509 were high yielding, with RB89509 being unstable in yield across environments. Selection of genotypes proved reliable only after artificial inoculation. Comparing environments, artificially inoculated treatments in the wet savanna zone and in the forest–savanna transition zone with stable high symptom severity proved most suitable for screening of genotypes, while the wet savanna zone with low natural infection in year 1998 was suitable for production of propagation material, and the site in the dry savanna zone with natural infection in year 1998 was the best environment for cassava production. The correlation between disease severity and root yield was significant only for the non-inoculated treatment in the dry savanna zone in year 2000 (R = –0.58), but not in any other environment. Among the 37 genotypes tested, several genotypes can be recommended to farmers in specific ecozones, and genotype TMS30572 revealed as relatively stable in disease resistance and in high yield across ecozones.  相似文献
