于 2003 年研究了自然脱水过程中七叶树种子生理特性的变化。结果表明:七叶树种子对脱水高度敏感,为顽拗性种子。种子成熟采收时含水量高达 60.3%,种子在室温下自然干燥 30 天后,含水量下降到 30.2%,此时种子生活力完全丧失。在脱水初期,种子发芽率有所上升,但随后种子发芽率迅速降低。随着含水量的下降,种子浸出液相对电导率上升,但当含水量在 53.7% 到 50.9%之间时,种子浸出液相对电导率出现异常升高。同时种子超氧化物歧化酶(SOD)的活性随脱水时间的延长而下降,只有种子含水量在 53.7% 到 50.9%之间时出现例外。丙二醛(MDA)含量在脱水初期缓慢上升,在含水量下降到低于 50.9%时,丙二醛(MDA)含量快速上升。可溶性糖含量随脱水时间的延长缓慢上升。当种子含水量为 47%–60%时,种子发芽率较高,说明这一含水量区间有利于种子生活力的保持。图 6 参 13。  相似文献

Intact seeds from freshly harvested fruits of Myrica rubra (Sieb et Zucc.) were dormant and required 8 weeks of warm stratification followed by 12 weeks of cold stratification for germination. Exogenous application of gibberellic acid (GA(3)) to intact fresh seeds was effective in breaking dormancy, with > 70% of seeds germinating when treated with 5.2 mM GA(3) and incubated at a day/night temperature of 30/20 degrees C for 20 weeks. Removing the hard endocarp or endocarp plus seed coat of fresh seeds promoted germination, and addition of GA(3) to the embryo accelerated germination. The gibberellins GA(1) and GA(4) were more effective than GA(3) in promoting germination of seeds with the endocarp removed. Endogenous contents of GA(1), GA(3), GA(4), GA(7) and GA(20) were quantified by gas chromatography-mass spectrometry-selected ion monitoring in the endocarps, seed coats and embryos of fresh seeds treated with 5.2 mM GA(3). The content of GA(3) decreased in the endocarp during incubation, whereas GA(1) contents increased in the endocarp and seed coat. A high GA(1) content was detected in the endocarps and embryos of newly germinated seeds. We speculate that GA(3) was converted to GA(1) during incubation and that GA(1) is involved in seed germination. Endogenous abscisic acid (ABA) contents were measured in fresh seeds and in warm and cold stratified seeds. The ABA content in fresh seeds was distributed in the order endocarp > seed coat > embryo, with the content in the endocarp being about 132-fold higher than in the seed coat and embryo. Total ABA content of seeds subjected to warm or cold stratification, or both, was 8.7- to 14.0-fold lower than that of fresh seeds. Low contents of endogenous GA(1), GA(3), GA(7) and GA(20), but elevated contents of GA(4), were found in the seed coats and endocarps of warm plus cold stratified seeds and in the seed coats and embryos of newly germinated seeds. These observations, coupled with the finding that GA stimulated germination of dormant Myrica seeds, provide evidence that endogenous ABA inhibited release of dormancy and that endogenous gibberellins, especially GA(4) or GA(1), or both, are involved in germination.  相似文献

Twenty percent of of the world's flowering plants produce recalcitrant seeds (i.e., seeds that cannot withstand drying or freezing). We investigated whether the embryonic axis from the normally recalcitrant seeds of silver maple (Acer saccharinum L.) can be made tolerant to desiccation (10% water content) and low temperature (-196 degrees C, cryopreservation) by pretreatment with ABA or the compound tetcyclacis, which enhances endogenous ABA concentrations. Pretreatment of axes with both ABA and tetcyclacis increased germination after desiccation and freezing to 55% from a control value of zero. Pretreatment of axes with ABA and tetcyclacis increased the ABA content of the axes, as measured by enzyme-linked immunoassay, and stimulated the synthesis of storage and dehydrin-like proteins, believed to have a role in the desiccation tolerance of orthodox seeds.  相似文献

A study was conducted to determine the effects of electrostatic field (ESF) treatment on seed germination and seedling growth of Sorbus pohuashanesis. The experiments were arranged by uniform design computed by the Data Processing System (DPS), including three levels of seeds soaking time, four levels of ESF intensity and four levels of ESF treatment time, with 12 treatments. Ten seeds were used in each treatment with three replicates. Seed vigor, seed germinating ability, emergence rate of seedling, survival rate of seedling, and seedling height and diameter, as well as the change in activities of superoxide dismutase (SOD), soluble protein contents, total chlorophyll contents, soluble total sugar contents in leaves of S. pohuashanensis seedlings were measured after ESF treatments. The experiment results show that ESF treatment could improve the water absorption ability of dry seeds of S. pohuashanensis, resulting in fast germination at room temperature under light conditions. Combined treatment of ESF with cold stratification could increase seed germination percentage significantly (to 42.20%), promote seedling height growth, affect leaf SOD activity, and could raise contents of total chlorophyll, soluble protein, and total soluble sugar in leaves. Seed soaking time had a significant effect on seed relative electroconductivity, seed germination under light, SOD activity, soluble protein content and total soluble sugar content of seedling leaves. ESF intensity exerted a moderate effect on these indexes. ESF treatment time only had significant effect on total chlorophyll contents, no evident effect on other indexes.  相似文献

通过采集不同发育阶段种子并对其进行自然干燥处理,以含水量、发芽率和ABA含量变化为指标,研究了色木槭和拧筋槭种子在自然发育和干燥过程中的脱水耐性变化.结果表明,色木槭和拧筋槭种子虽经历了成熟脱水,但种子仍不耐脱水;其发芽率和活力指数随发育进程而逐渐增加,但随自然脱水进程而逐渐降低;色木槭和拧筋槭种子最大脱水耐性分别出现在花后100 d和120 d;在整个发育过程中,两树种种子的ABA(脱落酸)含量均较低;拧筋槭种子中ABA含量在脱水过程中呈下降趋势,而色木槭种子中ABA含量在脱水过程中虽有升高但变化不显著.推测ABA含量低可能为色木槭和拧筋槭种子不耐脱水的主要原因之一.  相似文献

We studied effect of desiccation on Knema attenuata seeds in the laboratory conditions(28 ± 2℃) for 8 days.We also monitored biochemical changes during germination by following standard procedures.Seeds lost their viability in 8 days by decreasing to 18% of moisture content from its original value of 31%.The total sugar,protein and lipid contents decreased during seed ageing.An increase was noticed in the protease activity during desiccation.The phenolic content in seed exudates was increased during ageing.The result confirms the degradation of stored biomolecules in seeds during ageing.Desiccation sensitivity and biochemical changes during dehydration confirm its recalcitrant nature.  相似文献

Seeds that lose viability when dried to a water content of less than 12% are said to be recalcitrant. We subjected acorns of Quercus alba L., a species with recalcitrant seeds, to desiccation to determine the effects of drying on lipids, proteins and carbohydrates of the embryonic axis and cotyledon tissues. Samples of fresh seed and seed dried for selected intervals were analyzed for water content and germination, and for lipids, proteins and carbohydrates by Fourier transform-infrared (FT-IR) spectroscopy. Carbohydrates were further analyzed by gas chromatography (GC). The FT-IR analysis revealed that membrane lipid structure initially exhibited reversible shifts between gel and liquid crystalline phases in response to drying and rehydration; however, reversibility declined as viability was lost. Changes in carbohydrate concentration were observed based on peak height comparisons; sucrose concentration in the embryonic axis increased dramatically after 5 days of drying. The most sensitive indicator of desiccation damage was the irreversible change in protein secondary structure in embryonic axes and cotyledon tissue. These changes were illustrated by shifts in amide absorbance near 1650 cm(-1). Gas chromatography indicated an abundance of sucrose in both the embryonic axes and the cotyledon tissue. Although sucrose concentrations in these tissues were initially similar, sucrose concentration in the embryonic axes became significantly greater than in the cotyledons as the acorns dried. We hypothesize that, in drying acorns, increased concentration of sucrose does not prevent loss of viability, but acts as a glycoprotectant against cell collapse and cell wall membrane damage as water stress increases.  相似文献

对白梭梭同化枝自然水分胁迫下的季节抗旱特征进行研究。结果表明:自然干旱胁迫条件下,不同生长发育时期同化枝的主导抗旱生理因子不同。5月31日至6月29日盛花期间,同化枝内源ABA含量甚微,气孔导度值与光合强度处于生长发育过程的最高水平,可溶性糖含量呈现下降趋势,而脯氨酸含量略有增加,植物通过渗透调节作用适应此阶段轻度干旱胁迫;6月29日至7月26日盛夏期间,ABA迅速积累,气孔导度值降为生长发育过程的最低值,叶绿素分解,可溶性糖与脯氨酸均呈现快速积累趋势,且可溶性糖积累强度大于脯氨酸;8月9日至8月22日同化枝生长发育后期,ABA急剧积累为生长发育过程的最高浓度,气孔导度值有所增大,脯氨酸和可溶性糖保持在高水平平稳变化。此阶段高浓度ABA调节植物生理过程适应干旱的效应受CTK、IAA两种内源激素的抑制,进而抑制脯氨酸和可溶性糖的继续积累。  相似文献

采用正交设计从种子含水量、包装方法、保存温度和预回湿方法4个因素来探索山合欢种子超干燥保存的可行性。用硅胶将种子脱水至1.42%~6.35%6个含水量梯度,结合不同处理密封保存1年,随后测定发芽率和一些生理生化指标,包括相对电导率(RC)、脯氨酸(Pro)、丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、可溶性总糖含量和脂肪酸组分。结果表明:种子含水量和预回湿方法对发芽率有显著影响,超干种子与未经超干处理的种子相比,发芽率有明显提高;超干种子发芽率与相对电导率和丙二醛含量呈显著负相关,与脯氨酸含量和3种抗氧化酶活性呈显著正相关;超干种子的油酸和α-亚麻酸含量比对照提高。适宜的超干处理能使抗氧化酶活性增强,使不饱和脂肪酸和可溶性总糖的含量增加,从而减缓种子生活力下降。  相似文献

Seeds of Strychnos nux-vomica have slow and erratic germination; thus different presowing treatments were applied to enhance the germination of its seeds collected from Tamaraikulam, Tamil Nadu, India. In addition, the effects of desiccation and different storage conditions on the germination of S. nux-vomica seeds were investigated. The results show that soaking in 500 ppm gibberellic acid (GA3) for 24 h, incubation of seeds at 40 °C for 3 days and alternate water soaking (16 h) and drying (8 h) for 14 days significantly increased the percentage germination compared to the control. Desiccation of seeds down to 10% moisture content resulted in better germination. Germination of S. nux-vomica seeds differed significantly between different storage periods, moisture contents of the seed and for first and second order interactions (p<0.001). The highest germination (92%) was achieved when seeds with 10% moisture content were stored at ambient temperature for 30 weeks. Evidence from the present study indicates that S. nux-vomica seeds possess physiological dormancy that can be broken effectively by after-ripening. As seeds of S. nux-vomica are found to be desiccation tolerant, dry seed (10% moisture content) can be hermitically stored at ambient temperature for 30 weeks without losing their viability.  相似文献

Knowledge of seed desiccation tolerance is fundamental for conservation and use of forest species. The protocol used for classification of seed desiccation tolerance and storage is time consuming and many times limited by the lack of information about optimum conditions for seed germination and treatments to overcome seed dormancy. This study evaluated 66 Brazilian tree species aiming to correlate seed characteristics with desiccation tolerance. For this purpose, a model was established to explain the relationship of tegument/seed mass ratio (SCR), seed mass, and water content of embryo + endosperm with desiccation tolerance. The principal component analysis showed the establishment of two groups, indicating the interaction between desiccation tolerance and seed characteristics. Recalcitrant seeds are more often associated with the water content of embryo + endosperm and water content of tegument + endocarp, while orthodox seeds are more associated with SCR and number of seeds per kilogram. The classification found using the model proposed was significantly correlated with desiccation tolerance and storage, with 92% confidence for the analyzed species. Seeds morphological characteristics can be used for prediction of desiccation tolerance and storage behavior; however, the use of a model that combines more variables increases the chance of accurate classification.  相似文献

The effect of harvest time on seed germination, desiccation tolerance and level of dormancy is not known in Sorbus mougeotii (Soyer-Will. & Godron). Dry weight (DW), moisture content (MC) and leakage conductivity (LC) were tested on seeds from six harvest times. Chilling requirement and germinability were tested on seeds without drying and after drying and rehydration. The effect of postharvest ripening of seeds with or without drying was also tested. The DW was stable during the last 6 weeks of maturation, whereas MC initially decreased but later increased simultaneously with an increase in LC. Seeds were desiccation tolerant, deeply dormant and after a chilling treatment germinated to a high percentage at all harvest times. The chilling requirement decreased significantly with later harvest, suggesting a decrease in level of dormancy. Drying and postharvest ripening reduced and synchronized the chilling requirement.  相似文献

Mycorrhizas of six spruces of a stand were collected over two years and were analysed for soluble carbohydrates (sucrose, raffinose, glucose, trehalose and mannitol). The level of these carbohydrates was high during winter and low during summer. Upon exposure of excised mycorrhizas to frost or desiccation, the content of trehalose roughly doubled. A positive correlation was found between the trehalose content of mycorrhizas and their ability to survive frost or desiccation as determined by the electrolyte leaching method.  相似文献

The combination of gibberellin acid (GA) soaking with moist chilling plays a pivotal role in seed dormancy breaking. However, knowledge involving physiological and biochemical mechanisms for such a response is limited. This work focused on the effects of GA3 and moist chilling on overcoming seed dormancy as well as the response of endogenous hormones and nutrient reserves to dormancy release in Chinese dogwood (Cornus kousa var. chinensis). Seeds subjected to GA3 soaking and moist chilling exhibited a high germination percentage (average 97%), and the combination of 500 mg·L?1 GA3 soaking for 3 days followed with moist chilling for 50 days was more effective than other treatments. For GA3 treatments, lipids were initially hydrolyzed during the first stage of moist chilling (0–20 days), resulting in the increase of starch and soluble sugars, while proteins were kept relatively stable. During the second stage (20–50 days), contents of lipids, starch, soluble sugars, and proteins decreased by 46%, 82%, 61% and 59%, respectively. Abscisic acid (ABA) contents and ratios of ABA to GA3 in GA3 treatments decreased by nearly 70% and over 90%, respectively, when dormancy was terminated. However, endogenous GA3 contents in all treatments significantly increased with prolonged moist chilling. The present results provide insight into the metabolic mechanism involving hormonal regulation and mobilization of reserves during the release of seed dormancy in Chinese dogwood.  相似文献

以28个栓皮栎种源种子为试验材料,对不同种源间栓皮栎种子的形态特征和营养成分含量差异进行比较分析。结果表明,不同种源间栓皮栎种子的长度、宽度和种子千粒质量以及可溶性糖、淀粉和蛋白质含量差异极显著。栓皮栎种子的长度和宽度变化幅度分别为1.56~2.38 cm和1.22~1.96 cm;种子千粒质量的变化幅度为1.45~5.45 kg;种子可溶性糖、淀粉和蛋白质含量的变幅分别为1.817%~6.325%、42.461%~80.274%和2.699%~9.694%。栓皮栎种子千粒质量与种子长度、宽度间呈极显著正相关,种子长度与宽度也极显著正相关;主成分分析表明,不同种源栓皮栎的差异主要是由种子千粒质量、宽度和长度决定。初步选出云南勐海、湖南城步1、安徽金寨1、重庆酉阳、山东蒙阴1、安徽旌德、安徽绩溪和安徽金寨2种源为优异种质资源。  相似文献

Mass maturity (end of the seed-filling phase) occurred at about 72 days after flowering (DAF) in developing seeds of Mimusops elengi, at which time seed moisture content had declined to about 55%. The onset of ability to germinate was detected at 56 DAF and seeds showed 98% germination by 84 DAF. Tolerance of desiccation to 10% moisture content was first detected at 70 DAF and was maximal by 84 DAF. Delaying collection by a further 14 days to 98 DAF, when fruits began to be shed, reduced seed viability, particularly for seeds first dried to 10% moisture content. Hence the best time for seed collection appears to be about 14 days before fruits shed. In a separate investigation with six different seed lots, desiccation below about 8–12% moisture content reduced viability (considerably in some lots). The viability of dry seeds (below about 10% moisture content) stored hermetically was reduced at cool temperatures (5 °C and below), and none survived storage at sub-zero temperatures. The results suggest that Mimusops elengi shows intermediate seed storage behaviour and that the optimal hermetic seed storage environment is about 10% moisture content at 10 °C, while short-term, moist, aerated storage at high (40%) moisture content is also feasible.  相似文献

Gleditsia triacanthos L. pods were harvested at San Lorenzo del Escorial, Madrid, Spain, before and after seed desiccation. Maturing green pods, harvested from trees, contained soft light-green seeds, whereas fully mature brown pods, collected on the ground, contained hard dark-green seeds. Based on visual inspection of seeds, it was determined that the green color is located in the cotyledons and embryo of soft seeds but mainly in the seed coat of hard seeds. High performance liquid chromatographic analysis indicated that both hard and soft seeds contained the same set of photosynthetic pigments as fully developed leaves, but in different proportions. The hard and soft seeds mainly differed in their chlorophyll a to chlorophyll b ratio and in the composition of the xanthophyll cycle pool of pigments. Fluorescence at -196 degrees C revealed that the molecular organization of the pigment molecules in the seed coat of hard seeds differed from that in intact cotyledons of soft seeds and intact green leaves. The -196 degrees C fluorescence spectra also revealed the presence of a small heterogenous pool of non-photoactive protochlorophyll(ide)s, similar to those found in dark-grown tissues of gymnosperms and angiosperms.  相似文献

  • ? The sensitivity of dormant seeds of mazzard cherry (Prunus avium L.) of Polish provenances to extreme desiccation and/or the ultra-low temperature of liquid nitrogen, LN (?196 °C), was investigated to identify the optimum seed water content (w.c.) at this temperature.
  • ? Germination and seedling emergence tests in this study showed that there is no critical seed w.c. for deeply desiccated seeds (stone w.c. 1.6%), although such desiccated seeds were sensitive to the temperature of LN in the seedling emergence test. For seeds frozen in LN, the highest germinability was observed at w.c. of 9.0–16.9%, but seedling emergence was then significantly lower than in nonfrozen seeds. The 2-year of storage in LN of seeds desiccated to 7.8% w.c. did not decrease germinability after thawing, in comparison with 2-year of storage at ?3 °C. Storage in LN showed that if seeds were stored after breaking of their dormancy, germinability after storage was lower because of the necessity of seed desiccation to the lower level (~ 8%) of w.c. after stratification (before storage). Secondary dormancy was induced in seeds desiccated after stratification.
