‘Swingle’ citrumelo [Citrus paradisi MacFaden × Poncirus trifoliata (L.) Raf.] has been extensively used as a rootstock in several citrus growing regions of the World, including Southern Brazil where ‘Rangpur’ lime (Citrus limonia Osbeck) is still the predominant variety despite being affected by several important pathogens. In this case, ‘Swingle’ citrumelo is used to produce nursery trees to establish new orchards or to be inarched in adult and healthy groves in order to change the rootstock. We report herein a system to produce trees on ‘Swingle’ citrumelo more rapidly by budding onto non-rooted cuttings, as well as assessing potential to rapidly multiply ‘Swingle’ through rooting of non-budded cuttings. Therefore, two potential products are described: budded trees and rooted rootstock cuttings. ‘Valencia’ sweet orange [Citrus sinensis (L.) Osbeck] was budded at different heights on cuttings derived from eight-month old rootstocks. Grafted and additional non-budded cuttings were then treated with indole-3-butyric acid (500 mg L−1) or left untreated before rooting. Three types of cuttings were evaluated: softwood, semi-hardwood and hardwood. The use of nursery trees derived from pre-budded hardwood cuttings of ‘Swingle’ citrumelo is an alternative grafting method on this cultivar. Softwood cuttings with one leaf pair were considered the most adequate material for rapid multiplication of ‘Swingle’ citrumelo by cutting. This could be particularly useful for inarching production or conventional budding after transplant of cutting-derived rootstocks.  相似文献

Summary

Annatto (Bixa orellana L.) is a tree indigenous to tropical America that has been naturalised throughout the Indian sub-continent. The plant is chiefly valued for its carotenoid pigments, annatto. Methods for the clonal propagation of hardwood and softwood cuttings, air-layering, budding, and grafting of annatto were investigated. Hardwood cuttings collected during the wet Summer months (June-July) were treated with indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), or naphthaleneacetic acid (NAA) at varying concentrations. The highest percentage rooting (63.4%), along with highest number of roots (8.33 per cutting) were achieved by hardwood cuttings after 12 h of 2.5 mM IAA treatment. Among the various auxin treatments of softwood cuttings, 5.0 mM IBA in combination with 5.0 mM NAA for 5 min resulted in a significant (P < 0.05) rooting percentage (56.7%). The responses of softwood cuttings to an auxin analogue (boric acid) and/or to thiamine hydrochloride (vitamin B) were inferior to synthetic auxins. When subjected to air-layering, hardwood branches gave 100% rooting without any application of hormone. Among the different types of budding methods attempted (patch, T, or I), patch budding produced the highest efficiency (78%) of propagation. Splice-grafting could also be used for conventional propagation of annatto, with a 50% survival rate. Annatto can therefore be cloned by adopting these methods. Propagation based on softwood cuttings facilitated moderate-scale cloning of this valuable, elite germplasm.  相似文献

Seven F1 hybrids of Antirrhinum majus were grown in a range of environments and measurements were made of the time from sowing to budding and of the time from budding to flowering. The time to budding of all cultivars varied with environment, but was always longer for late-flowering types. The time from budding to flowering changed with environment in the early-flowering cultivars. This period was shorter and apparently independent of environment in the late-flowering types. The possibility of selecting types which combine the short vegetative phase of the early-flowering cultivars with the lower time for inflorescence development of the late ones is discussed.  相似文献

Simple techniques and growth regulator treatments to improve micro-grafting success of ‘Mexican’ lime, ‘Valencia’ orange and ‘Star Ruby’ grapefruit were studied. Most scion cultivars grafted best on ‘Carrizo’ citrange [Poncirus trifoliata (L.) Rat. x Citrus sinensis (L.) Osbeck]. Dipping the shoot-tip in 2,4-D or kinetin before grafting doubled the percentage of successful grafts. Highest grafting success occurred when the scion shoot-tip was placed in an inverted-T incision on the epicotyl of the rootstock.  相似文献

Softwood and semi-hardwood cuttings of 4 Tilia taxa were taken at different times and treated with high concentrations of IBA. Rooting-response varied with taxa and sampling-date. Rooting in Tilia americana L. cuttings was maximum when taken on 19 May and 1 June. Cuttings of Tilia cordata Mill. did not differ significantly in rootability when taken on 19 May, 22 June or 13 July, but rooting was significantly less with 3 August cuttings. Tilia × euchlora Koch cuttings did not root. All Tilla × euchlora Koch ‘Redmond’ cuttings taken on 19 May or 22 June rooted. A 5-second basal dip of IBA significantly stimulated rooting, especially in the range of 20 000–35 000 mg 1?1.  相似文献

There has been recent market interest in “baby” salad greens. However, little information exists on the nutritional differences between immature “baby” greens and produce traditionally sold at the fully mature stage. Kale (Brassica oleracea L. var. acephala D.C.) contains high levels of lutein and β-carotene, which possess important human health properties. Kale was grown in a controlled environment and pigments were measured in young (<1 week), immature (1–2 weeks), mature (2–3 weeks), fully developed (3–4 weeks) and senescing (>4 weeks) leaves using high-performance liquid chromatography (HPLC). Significant differences were observed for all pigments during leaf development. Pigment accumulation followed a quadratic trend, with maximums occurring between the 1st and 3rd week of leaf age. The highest concentrations of lutein measured 15.1 mg/100 g fresh mass and occurred in 1–2 week old leaves. The remaining pigments reached maximum levels at 2–3 weeks (β-carotene at 11.6 mg/100 g; chlorophyll a at 251.4 mg/100 g; and chlorophyll b at 56.9 mg/100 g fresh mass). Mature fully expanded kale leaves accumulated higher carotenoid concentrations than immature or “baby” leaves, with senescent leaves having the lowest carotenoid concentrations. Harvesting kale leaves at a mature stage of development resulted in maximum carotenoid values. Cultural management practices that increase carotenoid concentrations would be expected to improve nutritional quality for fresh markets.  相似文献

Plants of two Begonia × hiemalis cultivars (‘Schwabenland’ and ‘Nixe’) were grown in a greenhouse and continuously illuminated (24 h a day) from late October to early March, with light from warm white fluorescent lamps (L“WW”), high pressure mercury lamps (HPI/T) or high pressure sodium lamps (SON/T), each at three irradiance levels (5, 10 or 15 W m?2). The control received light continuously (0.8 W m?2) from incandescent lamps (INC). Every third week, plant material (flower peduncles) was collected for in vitro culture. The adventitious shoot formation was strongly dependent on cultivar, length of mother plant illumination and light source. The use of L“WW”, HPI/T or SON/T improved the regeneration ability of flower peduncle segments compared to control. Prolonged irradiation lowered the regeneration in ‘Schwabenland’, especially when HPI/T lamps were used.  相似文献

Tissue-culture methods are described for the vegetative propagation of several palm species either through shoot tip culture or plantlet differentiation via embryogenic callus. The influence of explant size, medium composition and physical environment required for the establishment of palm shoot tips in vitro was determined. Date palm (Phoenix dactylifera L.) seedling shoot tips of various sizes were cultured in either liquid or agar modified Murashige and Skoog (MS) medium containing 0.0–1.0 mg 1?1 α-naphthaleneacetic acid (NAA) and 0.0–15.0 mg 1?1 benzyladenine or N6-(Δ2-isopentenyl) adenine (2iP) in order to enhance shoot growth and induce axillary budding. Satisfactory date palm shoot tip growth and proliferation was obtained from explants that were 3 mm in length, consisting of the apical meristem region and 2–5 adjacent leaf primordia. Optimum shoot tip development and axillary budding was obtained by initially establishing explants on an agar medium for 2 weeks, then transferring to a liquid medium. Shoot tips from several palm species were cultured on MS media containing 100 mg 1?1 2,4-dichlorophenoxyacetic acid (2,4-D), 3 mg 1?1 2iP and 3 g 1?1 activated charcoal, or on MS medium containing 1 mg 1?1 NAA and charcoal, to determine their morphogenetic responses in vitro. Shoot tips of Metroxylon sp., Phoenix canariensis Hort. ex. Chabaud., P. dactylifera ‘Khalasa’, ‘Thoory’ and ‘Zahidi’, and P. roebelenii O'Brien planted on medium with 2,4-D and 2iP initiated callus, asexual embryos and free-living plantlets after 4–8 months in culture. Shoot tips from Erythea edulis S. Wats., P. canariensis, P. dactylifera ‘Khalasa’, Thoory' and ‘Zahidi’, Washingtonia filifera Wendl. and W. robusta Wendl. cultured on medium containing NAA developed into plantlets with well-developed leaves and adventitious roots within 2–6 months from the time of planting. In some cases, cultured date palm shoot tips gave rise to axillary buds.  相似文献

Viburnum plicatum Thunb. f. tomentosum Rehd. and Viburnum dentatum L. were fertilized with N, P, and K at a high or low rate during late spring and late summer. V. plicatum f. tomentosum absorbed more K than N and P. Potassium applications to soil increased foliar K significantly, but increases were much greater following spring applications than following late summer applications. There was no luxury consumption of K by V. dentatum. N and K applications decreased cold hardiness of V. plicatum f. tomentosum in late October. Nutrient applications had no effect on cold hardiness of V. dentatum regardless of date of application.

Eggplant (Solanum melongena L.) seedlings (cv. Tsakoniki) were cultivated in soil artificially infested with V. dahliae Kleb. and then sterilized by either methyl bromide (MB) or calcium cyanamide. Grafted seedlings on the wild species Solanum torvum Sw. and the control seedlings (auto-rooted) were cultivated in soil sterilized by MB and then artificially infested with V. dahliae. The plants grafted on S. torvum and the ones grown in soil treated with calcium cyanamide (2001) exhibited significantly lower leaf symptom index (average value LSI = 1.55 and LSI = 1.00) and disease index (average value DI = 2.05 and DI = 1.20), respectively, as compared to the controls (average value LSI = 3.80 and DI = 5.50). Grafted plants on S. torvum and plants grown in soil treated with calcium cyanamide (2001) were more vigorous, as measured by plant height, stem diameter and root biomass than the controls. This resulted in an increased (over years) early (487.8% and 416.2%, 2001) and late production (277.0% and 241.3%, 2001) and mean fruit weight (over years) in early (93.7% and 49.6%, 2001) and late production (38.3% and 22.8%, 2001) as compared to the controls. In conclusion, grafting of eggplant and soil sterilized by calcium cyanamide had positive effects on growth, production and Verticillium wilt control.  相似文献

The time from sowing to 50% radicle protrusion of parsley seeds (Petroselinum crispum L.) was about 1 month at 5°C and about 1 week at 15°C. Osmotic pre-treatment (“priming”) with a polyethylene glycol “6000” solution of ?12 bars water potential (302 g PEG “6000” per 1 kg distilled water) at 15°C for 3 weeks under aerobic conditions reduced this median radicle protrusion time at 15°C to 1 or 2 days for seeds which had been surface-dried after treatment, 2 or 3 days when they had been air-dried and stored for 1 week, and 3 or 4 days after 10 weeks' dry storage. The uniformity in germination time of an untreated seed population was usually good, and care was needed not to make this worse by “over-priming”. An appropriate pre-treatment could, however, slightly improve the uniformity. Germination percentage remained unaffected but the pre-treatment could appreciably improve the emergence percentage of seeds sown in wet soil. In a small-scale experiment out of doors, the weight of plants from certain seed treatments was significantly greater after 14 weeks' growth than that from untreated seeds.  相似文献

A technique is described by which mature fruit has been produced on Sultana vines in the first season. The vines were propagated by aerial layering. The best development of inflorescences and fruit occurred when layers were removed from the parent at budburst.

Other techniques failed to produce mature fruit in the first season. These included propagation from dormant cuttings, grafting or budding dormant fertile buds to rooted cuttings, and application of growth substances to inflorescences and/or shoots of rooted cuttings.  相似文献

To evaluate genetic damage, parameters including root growth, chromosomal aberrations, and interphase silver-stained nucleolar organizer regions (AgNORs) in root tip cells of Allium cepa L. were investigated after treatment with NaCl (40–160 mM). The results showed that NaCl caused a decrease in root growth. All concentrations of NaCl showed an inhibitory effect on dividing cells in root tips of A. cepa L. and caused a reduction in mitotic index values. Upon exposure to NaCl, roots exhibited various mitotic abnormalities, including c-mitosis, anaphase bridge, and chromosome stickiness. In addition, interphase cells with micronuclei, budding nuclei, and unequal-sized nuclei were observed. Moreover, total cell aberration increased with increasing NaCl concentration. For AgNOR parameters, the average number of AgNORs per nucleus decreased in roots treated at all NaCl concentrations. The singular AgNOR area and whole AgNOR area in the nucleus containing 1–3 AgNORs were inversely proportional to NaCl concentrations.  相似文献

In addition to managing soil-borne diseases in muskmelon (Cucumis melo L.) production, grafting with resistant rootstocks may impact fruit quality. The ethylene antagonist 1-methylcyclopropene (1-MCP) has been shown to extend shelf life of fresh muskmelon fruit. Postharvest characteristics of 1-MCP-treated melon fruit as affected by grafting, however, have not been well examined. This study was conducted to explore the influence of grafting with different rootstocks on ripening and quality attributes of 1-MCP-treated muskmelon fruit during postharvest storage. Grafted ‘Athena’ muskmelon with two commercial squash interspecific hybrid rootstocks including ‘Strong Tosa’ and ‘Tetsukabuto’ as well as non-grafted and self-grafted ‘Athena’ were grown in replicated field plots at the University of Florida Plant Science Research and Education Unit (Citra, FL, USA) during April–June 2010. Half-slip fruit from two harvests were treated with 1.0 μL L−1 1-MCP (18 h, 20 °C) and analyzed during storage at 13 °C. For fruit from the 27 May harvest, whole fruit and mesocarp firmness, titratable acidity, soluble solids, and ascorbic acid content were measured, while production of ethylene and CO2 was determined on fruit from the 29 June harvest. Grafting did not show a significant impact on fruit yield but affected the fruit shelf life significantly. Fruit from non-grafted ‘Athena’ and ‘Athena’ grafted onto ‘Strong Tosa’ demonstrated a shelf life of 31 d for the first harvest and 22 d for the second harvest. Shelf life of fruit from self-grafted ‘Athena’ and ‘Athena’ grafted onto ‘Tetsukabuto’ declined by 6 d and 3 d for the first and second harvest, respectively. Whole fruit firmness decreased by approximately 15.5% on average from 13 to 31 d except day 19 as a result of grafting, but to a lesser extent with ‘Strong Tosa’ rootstock. Mesocarp firmness of grafted melon was reduced by about 30.2% at days 13 and 19 compared to non-grafted ‘Athena’ fruit. In contrast, titratable acidity, soluble solid content, and ascorbic acid concentration were less affected by grafting. All the measurements except for ethylene and CO2 production declined during storage regardless of the grafting treatment. Compared with ‘Strong Tosa’ rootstock, ‘Tetsukabuto’ resulted in a more rapid ripening under 1-MCP application, as reflected by earlier increase in ethylene production and higher respiratory rate. The study demonstrates that grafting effects on postharvest ripening and quality of ‘Athena’ muskmelon can vary markedly with rootstocks used.  相似文献

The Spanish type cultivar ‘Piel de Sapo’ (Cucumis melo L. var. saccharinus), has a limited compatibility with the Cucurbita maxima × Cucurbita moschata hybrids currently used as rootstocks. Double grafting can be used to improve compatibility between rootstock and scion by means of an intermediate rootstock compatible with both. Non-grafted, single, and double grafted melon plants of the cultivar Piel de Sapo were evaluated for water, nutrient absorption, photosynthesis activity, biomass production in early phases, as well as for yields and fruit quality in a long term trial. The hybrid ‘Shintoza’ (C. maxima × C. moschata) was used as rootstock, and the cantaloupe type melon cultivar ‘Sienne’ as an intermediate scion. Grafting did not affect net photosynthetic values, yet increased water use efficiency (35%). Double grafted plants increased aerial dry weights (66% and 31% with respect to non-grafted and simple graft plants, respectively), and also increased capacity for uptaking beneficial minerals (between 61% and 13% and particularly for NO3, P, K, Ca, Mn, and Zn) with respect to non-grafted and single grafted plants. The quantum efficiency PSII photochemistry values increased in double grafted plants (12%) with respect to the control plants. Consequently, double grafting on a vigorous rootstock such as ‘Shintoza’, with an intermediate scion, results in improved mineral and water absorption and achieves an increase in ion influx to the scion – so enabling an increase in light photosynthetic reaction and biomass. Double grafted plants increased fruit yield when compared to simple grafted and non-grafted plants (12% and 56%, respectively) and did not affect fruit quality in terms of °Brix and colour. In conclusion, double grafting presents several beneficial aspects that are counter-balanced by the extra cost of the technique. The difference in yields reflects compatibility problems.  相似文献

Aqueous slurries of commercial preparations of Trichoderma harzianum Rifai strain KRL-AG2 G41 (Th), T. virens Strain G-41 (Tv), or their combination (ThTv, at half rates each of the single application rate) were applied to ‘Marktetmore 76’ cucumber seeds (Cucumis sativus L.) that were non-primed or primed for 3 days at 25 °C either osmotically (−2.5 MPa from 0.337 molal Ca(NO3)2) or osmomatrically (−1.0 MPa from 0.135 molal Ca(NO3)2 plus −1.5 MPa from 50% water in exfoliated grade 5 vermiculite). Slurries were applied to seeds (1 mg per seed) either before or after priming. Seeds were sown in soilless, peat-based media with or without inoculation with Pythium aphanidermatum (Pa). Protection against damping-off caused by high pressures of Pa (16% emergence in non-coated, non-primed seeds) was increased by slurry coating Th on non-primed (76.4% emergence) or on osmotically primed seeds, with coating either before or after priming having no effect on efficacy (average 62.6% emergence). Slurry coating Th on osmomatrically primed seeds failed to increase final emergence percentage (FEP). Colony forming units per three seeds (CFU, all 103) was 2.8 for non-primed seeds, and 3.2 and 2.6, respectively, when osmotically and osmomatrically primed seeds were coated after priming. In a second study with lower disease pressure (58.1 FEP from non-coated, non-primed seeds), slurry coating of non-primed or osmotically primed seeds with Th, Tv or ThTv reduced percentage damping-off and increased FEP. The combination coating eliminated damping-off only in non-primed seeds, and tended to reduce percentage damping-off in osmotically or osmomatrically primed seeds compared to coating with Th or Tv alone. In a third study using only non-primed seeds, slurry coatings with mefenoxam fungicide, Th, Tv, or ThTv decreased total damping-off to 2.6%, 7.4%, 2.0%, and 0%, respectively, from the 30.1% occurring in non-coated seeds. Th, Tv or ThTv applied to growth media at the same rate as the seed coating (1 mg per seed) were generally as effective as the seed coatings, and only the ThTv growth medium application eliminated damping-off. A fourth experiment revealed that Th, Tv or ThTv remained viable on non-primed seeds for up to 4 weeks (the longest storage duration) at 21 or 4 °C, but 21 °C storage resulted in faster seed germination by week 3 and higher CFU per three seeds by week 4. In summary, coating of non-primed seeds with Th, Tv or ThTv was more effective than coating primed seeds in reducing percentage damping-off. While priming treatments generally led to faster seedling emergence and greater seedling shoot fresh weight than was achieved with non-primed seeds, only for non-primed seeds was damping-off eliminated by the ThTv seed coating or growth medium application.  相似文献

Causes of variable and inconsistent bud-grafting success in chip-budded Norway maple (Acer platanoides) were investigated using the red-leaved scion ‘Crimson King’. Large differences in bud-take were associated with the year of budding, which over a 14 year period ranged from 22 to 94%. The ‘year effect’ could not be explained by weather, or by differences in budding success found between commercial sources of red-leaved scions. Neither was incompatibility between the ‘Crimson King’ scion and individual genotypes among the seedling rootstocks implicated, although there were reasons why at first sight incompatibility was thought to be present. The size of rootstock when planted was not correlated with subsequent budding success, although large differences in the quality of rootstock growth at budding were associated with large differences in bud-take, but these were confounded with year and with the fields used for the experiments. The ‘year effect’ was best explained by the field in which budding took place. One field gave consistently poor results, associated with a readily compacting soil, pointing to the strong possibility that bud-take is determined by conditions which affect shoot and/or root growth during the budding season or specifically while the union is forming in late summer. It was shown also that the presence of the scion bud on the chip-graft contributed to union formation, and that infection of the rootstock with the soil-borne verticillium wilt disease depressed bud-take, but this did not explain the ‘field effect’.  相似文献
