Precision parameters of miscellaneous methods for the analysis of drug dosage forms approved by AOAC since 1972, and not previously reviewed in this series, were recalculated on a consistent statistical basis by using the computer program FDACHEMIST. Seventeen published collaborative studies were reviewed; the studies encompassed 19 analytes in 80 different materials (dosage forms), 102 collaborative assays, approximately 10 laboratories per study, and principally direct spectrophotometric, polarographic, and spectroscopic methods, for a total of 1451 determinations. The average repeatability relative standard deviation (within-laboratories, RSDo) for the instrumental methods was 1.5%; the reproducibility relative standard deviation (among-laboratories, including within-, RSDx) was 2.6%; the ratio RSDo/RSDx of the averages was 0.57, with an average outlier rate of 2.7% of the reported determinations. The line of best fit of RSDx for the instrumental methods plotted against the negative logarithm of the concentration increases slightly with decreasing concentration, extending from an RSDx of approximately 2.0% at 100% concentration to an RSDx of 3.4% at 0.001% (10 ppm) concentration; this represents an RSDx change of approximately 0.3% (absolute) for each 10-fold decrease in concentration, independent of analyte, matrix, and method. A method for determining precipitated allergenic protein by the micro-Kjeldahl technique appeared to be outside this general relation, showing an RSDx of about 13% at a concentration of 0.015% (150 ppm) nitrogen.  相似文献

A selective and sensitive method utilizing liquid chromatography-mass spectrometry (LC-MS) has been developed for determining residues of the natural insect control agent spinosad in several crop matrices that are difficult to analyze by HPLC with UV detection. The method determines the active ingredients (spinosyns A and D) and three minor metabolites (spinosyns B and K and N-demethylspinosyn D) in alfalfa hay, wheat hay, wheat straw, sorghum fodder, and corn stover. The analytes are extracted from the samples with an acetonitrile/water solution, and the extracts are purified by solid phase extraction with a C(18) disk and a silica cartridge. All five analytes are determined simultaneously in a single injection using positive atmospheric pressure chemical ionization LC-MS with selected ion monitoring. The average recoveries ranged from 69 to 96% with standard deviations ranging from 4 to 15%. The method has a validated limit of quantitation of 0. 01 microgram/g and a limit of detection of 0.003 microgram/g. The LC-MS method can also provide residue confirmation in addition to quantitation.  相似文献

In 1971, a chemical method for the assay of vitamin D in concentrates containing only vitamin D was collaboratively studied by 14 laboratories, using 6 different samples from 2 European manufacturers. On the basis of these results, the laboratories were divided into 2 groups: 5 with significant laboratory biases of greater than or equal to 2%, and 9 laboratories with nonsignificant bias. The 9 laboratories were subdivided into 2 groups which differed significantly as to reproducibility within laboratories. The reproducibility between laboratories, expressed as a standard deviation in per cent with 95% confidence limits, was 1.2% (confidence range 0.6-7.3) and 4.7% (confidence range 2.4-29.3) for 3 and 6 laboratories, respectively. A second collaborative test was performed in 1974, using 12 vitamin D resin samples in oil from 3 United States manufacturers, to compare 2 chemical vitamin D assay methods (with and without maleic anhydride) and to compare results from the chemical and biological methods; 9 laboratories participated in the chemical method study and 3 in the rat bioassay study. The correlation of results of the chemical method including maleic anhydride treatment and the rat bioassays was satisfactory. The reproducibility of the chemical method was about the same as that in the first collaborative test.  相似文献

Determinations of the active ingredient in technical and formulated metribuzin were compared by gas-liquid chromatography (GLC) using various solvents, infrared (IR) spectrophotometry, and high-pressure liquid chromatography (HPLC). Ketones could not be used as a solvent for GLC analysis because of Schiff base formation. The GLC method using ethylene glycol monomethyl ether gave results about 1% higher for technical metribuzin than the other methods. GLC using methylene chloride solvent agreed with IR and HPLC results to within 0.4%. GLC using methylene chloride is the preferred method.  相似文献

A sensitive, reproducible method that uses liquid chromatography in the reverse phase mode has been developed for the determination of phthalic acid, resorcinol, and 2-(2',4'-dihydroxybenzoyl)benzoic acid in D&C Yellow No. 7 and D&C Yellow No. 8. The method uses a 10 micron, C-8 column, a 1% acetic acid-methanol gradient, and UV absorption detection at 280 nm. Average recoveries of phthalic acid, resorcinol, and 2-(2',4'-dihydroxybenzoyl)benzoic acid were 100, 98, and 102%, respectively, from fluorescein standard (certifiable as D&C Yellow No. 7) spike with each compound at levels ranging from 0.13 to 1.3%.  相似文献

A method is given for determining monosodium glutamate (MSG) in soups and soup bases by gas-liquid chromatography (GLC) of the trimethylsilyl ether derivative of glutamic acid. This method compared favorably with existing methods including GLC of the trifluoroacetate (TFA)/butyl ester derivative, and analysis on an amino acid analyzer. In addition, some samples were analyzed by GLC/mass spectrometry. The MSG content of various products ranged from approximately 0.2% in some condensed soups to 13.1% on bouillon cubes. The method, which can detect as little as 0.05%, requires only a 10 min single step derivatization at room temperature and is preferred to the TFA/butyl ester technique.  相似文献

Two automated fluorometric methods were compared with the official AOAC methods for determining vitamin C in fortified ready-to-eat cereals, fruits, vegetables, baby foods, flour products, pet foods, meats, frozen dinners, juices, and nutritional health bars. Each sample was analyzed in duplicate on 2 days and included a recovery study on all products. Egberg's semiautomated method (Method 1) and Roy's automated method (Method 2) were compared with the manual AOAC tritrimetric and fluorometric methods. The correlation factor for Methods 1 and 2 were 0.999 and 0.979, respectively, when compared with the AOAC methods. The recovery study showed average recoveries of 97.8% for Method 1, 99.3% for Method 2, and 100.6% for the AOAC methods. The results suggest that Method 1 is the method of choice for the majority of the products analyzed.  相似文献

Current tools used to assess the safety of food and feed derived from modern biotechnology emphasize the investigation of possible unintended effects caused directly by the expression of transgenes or indirectly by pleiotropy. These tools include extensive multisite and multiyear agronomic evaluations, compositional analyses, animal nutrition, and classical toxicology evaluations. Because analytical technologies are rapidly developing, proteome analysis based on two-dimensional gel electrophoresis (2DE) was investigated as a complementary tool to the existing technologies. A 2DE method was established for the qualitative and quantitative analysis of the seed proteome of Arabidopsis thaliana with the following validation parameters examined: (1) source and scope of variation; (2) repeatability; (3) sensitivity; and (4) linearity of the method. The 2DE method resolves proteins with isoelectric points between 4 and 9 and molecular masses (MM) of 6-120 kDa and is sensitive enough to detect protein levels in the low nanogram range. The separation of the proteins was demonstrated to be very reliable with relative position variations of 1.7 and 1.1% for the pI and MM directions, respectively. The mean coefficient of variation of 254 matched spot qualities was found to be 24.8% for the gel-to-gel and 26% for the overall variability. A linear relationship (R2 > 0.9) between protein amount and spot volume was demonstrated over a 100-fold range for the majority of selected proteins. Therefore, this method could be used to interrogate proteome alterations such as a novel protein, fusion protein, or any other change that affects molecular mass, isoelectric point, and/or quantity of a protein.  相似文献

A liquid chromatographic (LC) multiresidue method for determining residues of N-methylcarbamate insecticides in crops was collaboratively studied in 6 laboratories. Methanol and a mechanical ultrasonic homogenizer are used to extract the carbamates. Water-soluble plant coextractives and nonpolar plant lipid materials are removed from the carbamate residues by liquid-liquid partitioning. Additional crop coextractives (e.g., carotenes, chlorophylls) are removed with a Nuchar SN-silanized Celite column. The carbamate residues are then separated on a reverse phase LC column, using an acetonitrile-water gradient mobile phase. Eluted residues are detected by an in-line post-column fluorometric detection technique. Seven carbamates and 2 carbamate metabolites were included in the collaborative study. Each collaborator determined all the carbamates at 2 levels (approximately 0.05 ppm and United States tolerance) in blind duplicate samples of grapes and potatoes. Fortified and control samples were analyzed. Repeatability coefficients of variation for all the carbamates on the 2 crops averaged 4.7% and ranged from 2.4 to 7.1%. Reproducibility coefficients of variation for all the carbamates on the 2 crops averaged 8.7% and ranged from 5.3 to 12.4%. Accuracy, measured by comparison with fortification values, averaged 95% and ranged from 79 to 103%. The estimated limit of quantitation is 0.01 ppm. A simple and rapid method for determining riboflavin content in cooked sausages by ion-pair reversed-phase liquid chromatography has been set up. Samples were subjected to acid and enzymatic hydrolysis. Sample extracts were directly chromatographed, avoiding purification and concentration treatment. Final determination was performed by high-performance liquid chromatography with fluorescence detector (excitation, 227 nm; emission, 520 nm), on a 25 cm x 4 mm i.d. Spherisorb ODS-2 cartridge using a mixture of 5 mM heptanesulfonic acid adjusted to pH 2.7 with phosphoric acid and acetonitrile (75:25, v/v) as mobile phase. Precision of the method was 1.3% (within a day) and 2.6% (between days). The detection limit was 0.015 mg/100 g. The recovery was >95%.  相似文献

The determination of total vitamin C in the form of both l-ascorbic acid (AA) and dehydroascorbic acid (DHAA) present in pharmaceutical preparations has been automated. Total vitamin C (completely oxidized to DHAA) was determined by reaction with 2,4-dinitrophenylhydrazine while blanks utilized the same reagent after reducing all DHAA to AA. The automated method was applicable to a variety of multivitamin preparations including those containing iron and copper. The mean recovery of L-ascorbic acid added to 11 multivitamin preparations was 99.4% with a coefficient of variation of 2.5%. In the analysis of these products, results obtained by the automated method were essentially the same as those obtained by the original manual method. For preparations containing no copper salts, the results were also comparable to those obtainable by titration with 2,6-dichloroindophenol except in 1 product which contained some DHAA.  相似文献

A collaborative study was carried out which compared the official chemical method, 43.B14-43.B24, the official rat bioassay, 43.165, and the high pressure liquid chromatographic method for vitamin D3 resin, vitamin D3 resin in oil, and dry concentrate. A total of 340 samples were distributed to 17 collaborators for analysis. Five laboratories performed both the chemical and HPLC methods on 5 sets of blind duplicates. A 2-way analysis of variance comparing both methods for each sample showed a significant (P less than 0.01) difference between methods only for Sample 5. When the 2 methods were compared over all the samples, no significant (P less than 0.05) difference was found. Except for Sample 5, there were no differences in the repeatability of the methods. Per cent recoveries on Sample 3, which contained exactly 0.200 X 10(6) IU/g, showed 98.2% for the chemical method and 100.6% for the HPLC method for the 5 laboratories that performed both methods. The assay results of the HPLC and chemical methods are in good agreement with those found by the biological assay on Samples 1-4, but not for Sample 5. Evidence indicates that Sample 5 degraded partially to isotachysterol, and while the HPLC method yielded a reasonable value on this material, the chemical method erroneously showed full potency. An amendment is included for the collaboratively studied HPLC method which detects and eliminates 5,6-trans-vitamin D3, a possible interferant.  相似文献

A simplified liquid chromatographic (LC) method for determining vitamin D in vitamin AD concentrates (greater than or equal to 5000 IU vitamin D/g) was collaboratively studied. In the simplified method, the 2 columns specified in AOAC LC method 43.101-43.109 are replaced by a single column, which separates the vitamin D isomers and the vitamin A esters. The procedure for oils includes dissolution and quantitation by normal phase LC. Dry multivitamin concentrates and aqueous dispersions are treated with an enzyme system and the vitamins are extracted with n-pentane. Six coded samples were distributed to 16 laboratories; 15 collaborators returned their results. Estimates of repeatability and reproducibility for the oil samples were 1.1 and 3.1%, respectively; for the high-level concentrated dry preparation 1.4 and 3.9% and for the low-level concentrated dry preparation 1.3 and 11.4%, respectively. These values are a considerable improvement over the results obtained in the 1979 multivitamin collaborative study. The method has been adopted official first action for determination of vitamin D in vitamin AD concentrates containing greater than or equal to 5000 IU vitamin D/g.  相似文献

Vitamin D is determined in preparations containing other fat-soluble vitamins by high performance liquid chromatography (HPLC). The unsaponifiable residue is extracted and separated from interferences by reverse phase chromatography; the fraction corresponding to vitamin D3 is collected and quantitated using normal phase chromatography (amylalcohol-n-hexane as mobile phase) by measuring the vitamin D3 and pre-vitamin D3 peaks at 254 nm. Previtamin D3 content is calculated as vitamin D3 with a conversion factor (determined on the equipment used). Application of the method to vitamin AD3 mixtures in oils gives 98-102% recovery. The reproducibility, using an external standard, is 2-3%, calculated as the coefficient of variation; with an internal standard, the coefficient of variation is 1-1.5%. The method measures potential vitamin D3 content in preparations containing greater than or equal to 200 IU/g in the presence of all known vitamin D3 isomers, vitamin A, and vitamin E.  相似文献

Research has shown that dietary saponins may have health benefits. A simple, rapid method for the determination of saponins in legumes, using densitometry, is described. Saponin preparations, after pretreatment to remove nonsaponin components, are spotted in rows on a thin-layer chromatography plate, along with soyasaponin standards. The plate, without solvent development, is directly treated with sulfuric acid and heated. Violet spots develop which have a density proportional to the amount of saponin present. The standard curve has a correlation coefficient of 0.99 and is linear over the range of 1.25 to 10 microg of soyasaponins applied. The method has a coefficient of variation of less than 3% and compares favorably with quantitative thin-layer chromatography. Using this method the saponin contents of defatted soy flour (0.58%), dried navy beans (0.32%), and dried kidney beans (0.29%) were determined, and these results were found to be consistent with previous reports in the literature.  相似文献

A thin layer chromatographic (TLC) method is described for the determination of citrinin in feeds. Citrinin is extracted from feed with methanol and water, the mixture is made alkaline with 10% sodium carbonate, and the aqueous solution is filtered and extracted with chloroform to remove most of the interfering materials. The aqueous layer is acidified with 2N HCl and extracted with chloroform. The chloroform extract is concentrated and spotted on a thin layer chromatographic (TLC) plate which is developed in chloroform-acetone-ethanol-water (60 + 40 + 10 + 1). The citrinin is viewed under ultraviolet light after TLC. Either visual or fluorodensitometric quantitation is used. Recoveries of citrinin from various feed samples spiked at levels of 2.0--5 micrograms/g were 75--92%. The proposed method can detect 0.5 micrograms/g feed, including corn, silage, ready mixed feeds, and feed pellets.  相似文献

The official first action method for determining vitamin D in multivitamin preparations was modified. The method was collaboratively studied by 7 laboratories, using 6 preparations in oil. The preparations consisted of vitamin D at various levels and at various ratios (in w/w) in vitamin A. Three samples contained cholecalciferol and 3 samples contained vitamin D3 from vitamin D3 resin. After outliers were eliminated by the Dixon test, data were analyzed and averages were compared with amounts of vitamin D known to be in each sample. For samples with vitamin D: vitamin A ratios of 1:0.5, 1:5, and 1:10, the mean vitamin D recoveries were 98.8, 94.6, and 90.7%, respectively. A high pressure liquid chromatographic (HPLC) method is proposed for determining aflatoxins in corn. The sample is extracted with methanol-10% NaCl (4 + 1), pigments are precipitated with zinc acetate, and the extract is cleaned up on a small (2 g) silica gel column. Aflatoxins in the purified extract are resolved by normal phase HPLC on a microparticulate (10 micrometer) silica gel column with water-saturated chloroform-cyclohexane, acetonitrile solvent, and detected by fluorescence on a silica gel-packed flowcell. The method was compared with chloroform-water extraction of the official CB method on 15 samples of contaminated corn. In 5 of the 6 samples containing aflatoxins B1, B2, G1, and G2, methanol-10% NaCl extracted more aflatoxin than did cloroform-water, as measured both by HPLC and by thin layer chromatography. In samples containing only B1 and B2, the 2 extraction solvents were virtually equivalent. Agreement was good between HPLC and TLC for each extraction solvent. Average recovery of aflatoxins B1, B2, G1, and G2 added to yellow cornmeal at 3 levels was greater than 90%.  相似文献

A simple and rapid HPLC method to evaluate residues of the major ryanoids (ryanodine and dehydroryanodine) on three fruits (olives, apples, and pears) has been developed. The pesticides were extracted from the fruits with hexane and acetone solution (1:1, v/v). Cleanup was carried out with aminopropyl-bonded silica cartridges. This method is characterized by recovery >75%, precision <11% RSD, and sensitivity of 0.020 mg/kg. The method can also be used to determine the level of active ingredients in ryania powdery wood.  相似文献

Six laboratories collaboratively studied a method for the quantitative gas-liquid chromatographic (GLC) determination of phencyclidine.HCl in powders. The phencyclidine.HCl and other water-soluble compounds are dissolved in dilute HCl. A portion of the aqueous solution is made weakly basic with K2HPO4, and the organic-soluble compounds are extracted in CHCl3 for the GLC determination of the phencyclidine.HCl. Eicosane (n-C20) is incorporated in the extracting CHCl3 as an internal standard. The samples collaboratively studied included samples of known phencyclidine.HCl concentration and one sample of unknown purity. Recoveries ranged from 92.1 to 104% and per cent standard deviations from 1.05 to 3.39. The method was adopted as official first action.  相似文献
