This study sought to establish whether temperature gradients between the cervix, vagina, and rectum at and 7 days post-artificial insemination (AI) were associated with the incidence of pregnancy in lactating dairy cows (Experiment I; n = 90 ovulating cows) and to evaluate temperature gradient dynamics from the time of insemination to 7 days post-AI under heat stress conditions (Experiment II; n = 16 ovulating and 4 non-ovulating cows). In Experiment I, 39 cows (43.3%) became pregnant. The odds ratio for pregnancy was 2.5 for each one-tenth of a degree drop in cervical temperature with reference to the control rectal temperature at the time of AI (P = 0.01), whereas the same decrease in the cervix–rectum temperature differential 7 days post-AI resulted in an odds ratio of 0.44 (P = 0.02). In Experiment II, 5 of the ovulating cows (31.3%) became pregnant. The mean values of the vagina–rectum, vagina–cervix, and cervix–rectum temperature differentials at AI (day 0), 8 h, 24 h, and 7 days post-AI changed significantly from day 0 to day 7 (within-subject effect; P < 0.02) in ovulating cows but not in non-ovulating cows. Temperature differentials on days 0 and 7 were similar between ovulating cows and cows of Experiment I. Overall, our findings support the notion that a temperature differential between the caudal cervical canal and rectum at AI may be an indicator of the likelihood of pregnancy. Possible prospects of confirming estrus at the herd-level are also suggested.  相似文献

The objectives of this study were to determine the effects of incorporating a progesterone intravaginal insert (CIDR) between the day of GnRH and PGF2alpha treatments of a timed AI protocol using estradiol cypionate (ECP) to synchronize ovulation on display of estrus, ovulation rate, pregnancy rate, and late embryonic loss in lactating cows. Holstein cows, 227 from Site 1 and 458 from Site 2, were presynchronized with two injections of PGF2alpha on study d 0 and 14, and subjected to a timed AI protocol (100 mixrog of GnRH on study d 28, 25 mg of PGF2alpha on study d 35, 1 mg of ECP on study d 36, and timed AI on study d 38) with or without a CIDR insert. Blood was collected on study d 14 and 28 for progesterone measurements to determine cyclicity. Ovaries were scanned on d 35, 37, and 42, and pregnancy diagnosed on d 65 and 79, which corresponded to 27 and 41 d after AI. Cows receiving a CIDR had similar rates of detected estrus (77.2 vs. 73.8%), ovulation (85.6 vs. 86.6%), and pregnancy at 27 (35.8 vs. 38.8%) and 41 d (29.3 vs. 32.3%) after AI, and late embryonic loss between 27 and 41 d after AI (18.3 vs. 16.8%) compared with control cows. The CIDR eliminated cows in estrus before the last PGF2alpha injection and decreased (P < 0.001) the proportion of cows bearing a corpus luteum (CL) at the last PGF2alpha injection because of less ovulation in response to the GnRH and greater spontaneous CL regression. Cyclic cows had greater (P = 0.03) pregnancy rates than anovulatory cows at 41 d after AI (33.8 vs. 20.4%) because of decreased (P = 0.06) late embryonic loss (16.0 vs. 30.3%). The ovulatory follicle was larger (P < 0.001) in cows in estrus, and a greater proportion of cows with follicles > or = 15 mm displayed estrus (P < 0.001) and ovulated (P = 0.05) compared with cows with follicles <15 mm. Pregnancy rates were greater (P < 0.001) for cows displaying estrus, which were related to the greater (P < 0.001) ovulation rate and decreased (P = 0.08) late embryonic loss for cows in estrus at AI. Cows that were cyclic and responded to the presynchronization protocol (high progesterone at GnRH and CL at PGF2alpha) had the highest pregnancy rates. Incorporation of a CIDR insert into a presynchronized timed AI protocol using ECP to induce estrus and ovulation did not improve pregnancy rates in lactating dairy cows. Improvements in pregnancy rates in cows treated with ECP to induce ovulation in a timed AI protocol are expected when more cows display estrus, thereby increasing ovulation rate.  相似文献

This experiment was conducted to investigate the effects of different exogenous progesterone sources administrated after artificial insemination (AI) on serum progesterone (P(4) ) concentration and pregnancy rates in Holstein lactating cows. Sixty-four lactating Holstein dairy cows were allocated to four different treatments (n = 16 per treatment): the cows 1) were injected with physiological saline on days 5 and 13 after AI (control group); 2) were injected with progesterone on days 5 and 13 after AI (P group); 3) received controlled internal drug releasing device (CIDR) for a period from day 5 to 19 after AI (CIDR group); and 4) were injected with gonadotropin-releasing hormone (GnRH) agonist on days 5 and 13 after AI (GnRH group). Blood samples were collected on days 0 (AI day), 5, 13, 16 and 19 after AI to determine serum P(4) concentration. The results revealed a significant difference among treatment groups for serum P(4) concentration on days 13, 16 and 19 with the lowest concentration of serum P(4) for the control group. The pregnancy rate was also positively affected by all the treatments with CIDR having the greatest effect on pregnancy rate. Overall, the results indicated that CIDR has the greatest effect on serum P(4) concentration and pregnancy rate, although the administration of P and GnRH during days after AI increased serum P(4) concentration in lactating dairy cows as well.  相似文献

This study examined the effect of acute bull exposure around the time of artificial insemination (AI) on oxytocin and progesterone concentrations, and pregnancy rates in dairy cows. Ninety six dairy cows, stratified according to parity into primiparous and pluriparous, were divided into three groups; short bull exposure (SBE; 10 min, n = 32), long bull exposure (LBE; 4 h, n = 32) or no bull exposure (NBE; n = 32). On day 45 post‐partum, all cows were treated with PGF2α on three occasions 11–14 days apart to synchronize oestrus. They were submitted to fixed time AI 80 h after the third PGF2α injection. Cows in the SBE and LBE groups were artificially inseminated 5 min after the introduction of the bull. From a subset of cows (n = 6 per group; three primiparous and three pluriparous), blood samples were collected once every 5 min starting 15 min before AI until 15 min after AI and analysed for oxytocin concentrations. Additional blood samples were collected for measurements of progesterone (P4) concentrations once daily for 4 days starting on the day of AI and once every 3 days thereafter until day 22. The effects of bull exposure, time, parity, difficulty of AI, and pregnancy on oxytocin and P4 concentrations were analysed using the mixed linear model procedure. Mean oxytocin concentrations or change in oxytocin concentrations after bull exposure or AI were not different among groups. Pregnancy rates for the NBE, SBE and LBE groups were 55.5%, 33.3% and 44.4%, respectively, and were not different among groups. In conclusion, acute bull exposure around the time of AI did not affect oxytocin and progesterone concentrations and did not improve pregnancy rates in dairy cattle under these farms conditions.  相似文献

This study was designed to evaluate whether the outcome of artificial insemination (AI) was affected by the metabolic and oxidative status of dairy cows. Seventy-nine inseminations in 40 cows, were classified, on the basis of blood progesterone (P4) and pregnancy-associated glycoprotein (PAG) concentrations and clinical confirmation of pregnancy into, three categories: (1) positive (AI+, resulted in pregnancy, n=26; 33%), (2) negative (AI-, did not result in pregnancy, n=49; 62%), and (3) embryonic mortality (EM, n=4; 5%). Reactive oxygen metabolites, biological antioxidant potential, oxidative stress index, body condition score, glucose, total proteins, albumin, urea, non-esterified fatty acids (NEFAs), cholesterol, triglycerides, haptoglobin and advanced oxidative protein products (AOPPs) were measured on the day of AI (day 0), and 30 and 42days later. Cows with EM had lower BCS scores (2.5) than AI+ (2.8) and AI- (2.9) cows (P<0.05). During the post-partum period, body condition score (BCS) increased and NEFAs decreased (P<0.05) suggesting a recovery from the negative energy balance (NEB). The only significant differences found were that the mean concentration of AOPPs was higher and that of albumin lower in EM cows than in AI+ and AI- (P<0.05) animals. Plasma concentration of reactive oxygen metabolites and biological antioxidant potential were not related to AI outcome. Further studies are required to confirm this finding and to clarify the role of oxidative status on cows' fertility.  相似文献

This study compared the responses shown by lactating dairy cows to four different P4-based protocols for AI at estrus. Cows with no estrous signs 96 h after progesterone intravaginal device (PRID) removal were subjected to fixed-time AI (FTAI), and their data were also included in the study. In Experiment I, follicular/luteal and endometrial dynamics were assessed every 12 h from the beginning of treatment until AI. The estrous response was examined in Experiment II, and fertility was assessed in both experiments. The protocols consisted of a PRID fitted for five days, along with the administration of different combinations of gonadotropin releasing hormone (GnRH), equine chorionic gonadotropin and a single or double dose (24 h apart) of prostaglandin F. In Experiment I (40 cows), animals receiving GnRH at the start of treatment showed a significantly higher ovulation rate during the PRID insertion period while estrus was delayed. In Experiment II (351 cows), according to the odds ratios, cows showing luteal activity at the time of treatment were less likely to show estrus than cows with no signs of luteal activity. Treatment affected the estrous response and the interval from PRID removal to estrus but did not affect conception rates 28–34 days post AI. Primiparous cows displayed a better estrous response than multiparous cows. Our findings reveal acceptable results of 5-day P4-based protocols for AI at estrus in high-producing dairy cows. Time from treatment to estrus emerged as a good guide for FTAI after a 5-day P4-based synchronization protocol.  相似文献

Two experiments were conducted to evaluate whether hCG administered 7 d before initiating the CO-Synch + controlled internal drug release (CIDR) ovulation synchronization protocol (Exp. 1 and 2), or replacing GnRH with hCG at the time of AI (Exp. 1), would improve fertility to a fixed-time AI (TAI) in suckled beef cows. In addition, the effects of hCG on follicle dynamics, corpus luteum development, and concentrations of progesterone (P4) were evaluated. In Exp. 1, cows were stratified by days postpartum, age, and parity and assigned randomly to a 2 × 2 factorial arrangement of 4 treatments: 1) cows received 100 μg of GnRH at CIDR insertion (d -7) and 25 mg of PGF(2α) at CIDR removal (d 0), followed in 64 to 68 h by a TAI plus a second injection of GnRH at TAI (CG; n = 29); 2) same as CG but the second injection of GnRH at the time of insemination was replaced by hCG (CH; n = 28); 3) same as CG, but cows received hCG 7 d (d -14) before CIDR insertion (HG; n = 28); and 4) same as HG, but cows received hCG 7 d (d -14) before CIDR insertion (HH; n = 29). Pregnancy rates were 52, 41, 59, and 38% for GG, GH, HG, and HH, respectively. Cows receiving hCG (39%) in place of GnRH at TAI tended (P = 0.06) to have poorer pregnancy rates than those receiving GnRH (56%). Pre-CO-Synch hCG treatment increased (P < 0.05) the percentage of cows with concentrations of P4 >1 ng/mL at d -7, increased (P < 0.02) concentration of P4 on d -7, and decreased (P < 0.001) the size of the dominant follicle on d 0 and 3, compared with cows not treated with hCG on d -14. In Exp. 2, cows were stratified based on days postpartum, BCS, breed type, and calf sex and then assigned to the CG (n = 102) or HG (n = 103) treatments. Overall pregnancy rates were 51%, but no differences in pregnancy rates were detected between treatments. Pre-CO-Synch hCG treatment increased (P < 0.05) the percentage of cows cycling on d -7 and increased (P < 0.05) concentrations of P4 on d -7 compared with pre-CO-Synch controls. Therefore, pretreatment induction of ovulation after hCG injection 7 d before initiation of CO-Synch + CIDR protocol failed to enhance pregnancy rates, but replacing GnRH with hCG at the time of AI may reduce pregnancy rates.  相似文献

The study compared response to prostaglandin F2α (PG), synchrony of ovulation and pregnancy per AI (P/AI) in a 5‐ vs a 7‐day Ovsynch + PRID protocol and investigated whether the initial GnRH affects P/AI in lactating dairy cows. Two hundred and seventy‐six cows (500 inseminations) were assigned to one of four timed‐AI (TAI) protocols: (i) PRID‐7G; 100 μg GnRH im, and a progesterone‐releasing intravaginal device (PRID) for 7 days. At PRID removal, PG (500 μg of cloprostenol) was given im. Cows received the second GnRH treatment at 60 h after PRID removal and TAI 12 h later. (ii) PRID‐5G; as PRID‐7G except the duration of PRID, treatment was 5 days and PG was given twice (12 h apart). (iii) PRID‐7NoG; as PRID‐7G except the initial GnRH, treatment was omitted. (iv) PRID‐5NoG; as PRID‐7NoG except the duration of PRID, treatment was 5 days. Response to treatments and pregnancy status at 32 and 60 days after TAI was determined by ultrasonography. The percentage of cows ovulating before TAI was greatest in PRID‐7G (17.1%), and the percentage of cows that did not have luteal regression was greatest in PRID‐5G (9.5%). The overall P/AI at 32 and 60 days did not differ among TAI protocols. However, during resynchronization, cows subjected to the 5‐day protocols had greater (p < 0.05) P/AI (45.3% vs 33.6%) than cows subjected to the 7‐day protocols. Pregnancy loss between 32 and 60 days tended (p = 0.10) to be greater in cows that did not receive initial GnRH (14.8%) compared to those that received GnRH (8.2%). In conclusion, the PRID‐5G protocol resulted in fewer cows responding to PG, but P/AI did not differ among TAI protocols. A 5‐day protocol resulted in more P/AI in resynchronized cows, and cows that did not receive initial GnRH tended to experience more pregnancy losses.  相似文献

OBJECTIVE: To identify potential risk factors for embryonic loss before 35 to 42 days of gestation in dairy cattle. DESIGN: Prospective observational study. ANIMALS: 381 cows. PROCEDURE: Body condition score was determined at the time of artificial insemination (AI; day 0) and on days 20, 23, and 27 and between days 35 and 41; serum progesterone concentration was measured on days 0; 20 or 21; and 23, 24, or 25. Cows were excluded from analyses if day 0 serum progesterone concentration was > or = 1.0 ng/mL and classified as pregnant on day 23 if serum progesterone was > 1.5 ng/mL on day 20 or 21 and day 23, 24, or 25. Cows were examined via transrectal ultrasonography on day 27 or 28 and rectally palpated for pregnancy on days 35 to 41. RESULTS: 39% of cows that were pregnant on day 23 lost their embryo by day 27, and 18% of cows that were pregnant on day 27 or 28 were not pregnant on days 35 to 41. Breeding a pregnant cow posed the greatest risk for embryonic loss at both time periods. Mean serum progesterone concentrations on day 21 or 22 and day 23, 24, or 25 were lowest for cows that lost an embryo between days 24 and 28. Cows with a linear somatic cell count score > 4.5 before AI were twice as likely to lose the embryo by 35 to 41 days, compared with cows with a score < 4.5. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that embryonic loss could be reduced by more accurate estrus detection, reducing mastitis, and strategies to improve progesterone concentration after breeding.  相似文献

An experiment was conducted to examine the effect of progesterone (P(4)) and oestradiol benzoate (ODB) on fertility of repeat-breeder lactating dairy cows during summer. One hundred repeat-breeder lactating dairy cows were randomly allocated to four groups (Tr1, Tr2, Tr3 and C) in a study conducted at a private dairy farm. All cows were injected with 2 mg ODB (day 0), which were at random stages of their oestrous cycles. Cows in Tr1, Tr2 and Tr3 were administered with intravaginal progesterone-releasing devices (controlled internal drug-releasing, CIDR) at the time of ODB injection for 7 days and those in group C were untreated and served as controls. Following CIDR removal, all cows were given an intramuscular injection of 25 mg Prostaglandin (PGF(2 alpha)). Twenty-four hours after the PGF(2 alpha) injection, cows in Tr1, Tr2 and C groups were injected with 1 mg ODB. Cows in Tr3 group were injected with 10 microg gonadotropin-releasing hormone (GnRH) agonist 48 h after CIDR removal. Artificial insemination was performed between 24 and 30 h following the second ODB injection for cows in Tr1 group and at the time of GnRH injection for cows in Tr3 group. Cows in Tr2 and C groups were inseminated at detected oestrus. Plasma P(4) and oestradiol 17beta (E(2)) concentrations were determined for all cows daily from day 0 to day 9. Plasma concentrations of P(4) and E(2) among cows of groups Tr1, Tr2 and Tr3 were increased and reached maximum values within 48 h following administration and were greater (p < 0.001) than those of group C cows. The proportion of cows detected in oestrus based on P4 concentration on day 9 was 88%, 72%, 88% and 60% in groups Tr1, Tr2, Tr3 and C, respectively. Oestrous detection rate differed (p < 0.01) significantly between time-inseminated groups (Tr1 and Tr3) and those inseminated at observed oestrous (Tr2 and C) groups. Pregnancy rates based on ultrasonography performed on day 28 were 52%, 56%, 60% and 40%, and those based on rectal palpation on day 45 were 32%, 44%, 36% and 28% for Tr1, Tr2, Tr3 and C cows (p > 0.1), respectively. Whereas pregnancy rates for cows with four or more previous services in all groups (54.55%) were higher (p < 0.03) than those for cows with three previous services (29.49%). In pregnant cows, mean days from calving to the day of insemination were higher (p < 0.01) among cows with four or more previous services (204 +/- 8.0 days) than those with three previous services (157 +/- 6.0 days). Results indicate that treatment with a combination of ODB and CIDR in repeat-breeder dairy cows causes elevation in plasma concentrations of E(2) and P(4). Oestrous detection rate was better in cows that were primed with P(4) than those without P(4) priming. Cows with four or more previous services had significantly higher pregnancy rates than those with three previous services.  相似文献

The objectives were to evaluate the reproductive indices and survival analysis of pregnancy outcomes in multiparous repeat breeder Holstein cows (n = 557). The cows were synchronized to ovulate by Ovsynch, new controlled internal drug release device (N‐CIDRsynch), and once‐used CIDR device (U‐CIDRsynch). The pregnancy per AI at 28 days post‐insemination (P/AI 28) in the N‐CIDRsynch group (28.75%) was significantly (COR = 1.49; p = 0.011) greater than that reported in the Ovsynch (23.46%) and U‐CIDRsynch (21.73%) groups. Furthermore, the pregnancy per AI at day 75 post‐insemination (P/AI 75) in the N‐CIDRsynch group was significantly greater than the Ovysync group (COR = 1.35; p = 0.050). The repeat breeder cows received a N‐CIDR device had a significantly higher progesterone level on day 2 and day 4 of CIDR insertion (1.38 and 1.67 ng/ml, respectively) than those received a U‐CIDR device or the control group (p = 0.012 and 0.001, respectively). The Cox regression model recorded significant associations for synchronization protocols, THI at the TAI and season of calving with the hazard of P/AI 28 and P/AI 75 (p = 0.044 and 0.046; 0.001 and 0.005; 0.003 and 0.001, respectively). Multiparous repeat breeder cows (>3) had a lower hazard ratio (HR) of P/AI 28 than that reported in the reference (2nd parity) group (HR = 0.74, p = 0.050). The repeat breeder cows inseminated at 76–80 and >80 temperature‐humidity index (THI) had significantly lower HR of P/AI 28 than those inseminated at the baseline (<70) THI value (HR = 0.73 and 0.30, p = 0.036 and 0.001, respectively). The current results indicate that the use of N‐CIDR synch protocol may achieve satisfactory pregnancy outcomes in repeat breeder cows.  相似文献

Three different treatments were compared to improve pregnancy per artificial insemination (P/AI) in repeat-breeder (RB) dairy cows. All cows (n = 103) were assigned to one of four groups: (1) gonadotropin-releasing hormone (GnRH); (2) human chorionic gonadotropin (hCG); (3) once-used controlled internal drug release (CIDR) device; and (4) control. All treatments performed 5-6 days after artificial insemination (AI) and milk samples were collected just before treatment for progesterone assays. There were no significant differences in milk fat progesterone concentration among trial groups. Cows were observed for estrus signs thrice daily. Pregnancy per AI on day 45 in hCG and CIDR groups were significantly higher than GnRH and control groups (60.0% and 56.0% vs. 26.9% and 29.6%, respectively), but there were no differences in P/AI between GnRH and control groups. There were also no significant differences between hCG and CIDR groups. Milk fat progesterone concentrations were compared between pregnant and non-pregnant cows in each group and only in the hCG group it was significantly lower in pregnant cows. In conclusion, treating repeat-breeder cows with hCG or once-used CIDR 5-6 days after AI improved P/AI.  相似文献

A system for the artificial insemination fo beef cattle was described. The system involved preparation of the cattle and facilities, visual oestrous detection twice a day, drafting each morning and insemination of oestrous cows twice a day. The AI program was evaluated during the course of the program and at pregnancy test 35 to 90 days after the end of the insemination period. The insemination period was from 25 to 60 days depending on efficiency, with programs from 25 to 35 days being favoured. Results for 12218 first inseminations (65.9%) and 2222 second inseminations (57.8%) in 56 AI programs compared favourably with results in dairy AI. Pregnancy rates in lactating cows (70.5%) were significantly higher than in nonlactating cows and heifers (66.1%). Overall efficiency of well-managed AI programs were represented by 63% of cows put into a program becoming pregnant in a 29-day insemination period. Seventy-two per cent of the cows put into the program were inseminated yielding an 88% pregnancy rate. The efficiency of visual oestrous detection was 95% but this estimate did not include false positives. Efficiency of AI programs were effected by management, nutrition, infection, inseminators, facilities and the origin and type of cattle put into a program. Environmental factors such as handling, stress and thunderstorms did not effect efficiency except via a nutritional effect.  相似文献

AIMS: To quantify the effect of treating lactating dairy cows that had histories of periparturient disorders or disease with a single intrauterine infusion of 0.5 g cephapirin, 3-6 weeks prior to the start of the seasonal breeding period. METHODS: Cows (n=690) from spring-calving dairy herds (n=22) were enrolled in a prospective case-control study if they had a history of dystocia, a dead calf at calving or within 24 h of calving, retained foetal membranes (RFM), metabolic disease, twins or a vulval discharge 13 days postpartum. Cows were blocked by age, periparturient condition and calving date, and randomly assigned to be either treated with 0.5 g of cephapirin by intrauterine infusion, or left as untreated controls. Treatment occurred 24-42 days before the planned start of mating (PSM) within each herd. In addition, the reproductive tracts of 91 cows from 4 herds were examined using a vaginal speculum and rectal palpation and cervical-os discharge was scored on a scale from 0 (nil) to 3 (purulent). RESULTS: Overall, more treated cows were retained in the trial herds until pregnancy testing than control cows (p0.05). Average 28-day submission rate was higher (93.1% vs 87.1%; p0.05) and PSM-to-first-service interval was shorter (9+/-1 vs 11+/-1 days; p0.05) in treated than control cows. The percentage of cows not pregnant at the end of the mating period was not affected by treatment overall. However, amongst cows diagnosed with RFM, a dead calf, or vulval discharge, pregnancy rates 28 and 56 days after PSM were higher for treated than control cows (p0.05). Cows with a purulent cervical-os discharge had lower 28-day submission rates (p0.01), lower 28-day pregnancy rates (p0.05) and higher non-pregnant rates at the end of mating than cows with nil or mucoid cervical-os discharges. CONCLUSIONS: Intrauterine treatment with 0.5 g cephapirin improved reproductive performance of dairy cattle, especially those that had a history of RFM, a calf dead at calving or within 24 h of calving, or a vulval discharge.  相似文献

This study aimed to (i) assess the prevalence of cytological endometritis (CYTO) diagnosed at artificial insemination (AI); (ii) evaluate the effect of CYTO on the pregnancy outcome of the same AI sample; and (iii) determine the risk factors associated with CYTO diagnosed at AI in repeat breeder (RB) dairy cows. We analysed the productive and reproductive performances of 146 RB Holstein‐Friesian cows. To obtain a CYTO sample at AI, we used the cytotape technique. Generalized mixed effect models were computed to find the risk factors associated with the pregnancy and CYTO outcome. Based on ≥1% PMN cut‐off point, the CYTO prevalence at AI in RB cows was 25.3%. The overall pregnancy at AI was 44.2%. The conception rate in CYTO‐positive (n = 37) RB cows was 29.7% versus 49.5% for CYTO‐negative (n = 109) cows. A RB cow diagnosed CYTO positive at AI had 0.47 [odds ratio (OR)] odds to become pregnant in comparison with a CYTO‐negative cow. Cows that produced more milk than their counterparts in this study had increased odds (OR = 1.01) to be CYTO positive at AI. A novel risk factor positively associated with CYTO diagnosed at AI in RB cows was the level of daily milk urea (OR = 1.11). To conclude, CYTO at the moment of AI had a significantly negative effect on the pregnancy outcome in RB dairy cows. The aim of this study was to investigate whether the skim milk progesterone concentrations at artificial insemination (AI) and day of rise of post-ovulatory progesterone concentration thereafter affect the conception and embryonic death rates in repeat-breeding cows. Milk samples were obtained from 96 repeat-breeding cows that failed to conceive to three or more AIs. The samples were taken from the cows at the day of AI and three times/week until day 45 post-AI. Skim milk was obtained after centrifugation and used for progesterone assay. The cows with a progesterone concentration more than 0.5 ng/ml at AI showed a significantly higher incidence of late embryonic death than those having a progesterone concentration<0.5 ng/ml at AI (p<0.01). As the progesterone level at insemination rose, conception rate declined. A negative correlation was shown between conception rate and skim milk progesterone level at AI. Of 56 cows showing a rise of progesterone to 1 ng/ml or more within 6 days after AI, 28 cows (50%) conceived. On the contrary, only eight of 39 cows (20.5%) conceived when the progesterone rose up to 1 ng/ml after day 6 post-AI. We concluded that increased progesterone concentration at the time of AI and delayed rise of progesterone post-AI might lead to decrease in fertility in repeat-breeding cows.  相似文献

This study compares in two experiments the responses of lactating dairy cows to four different progesterone-based protocols for fixed-time artificial insemination (FTAI) in terms of their effects on follicular/luteal dynamics and fertility. The protocols consisted of a progesterone intravaginal device fitted for five days, along with the administration of different combinations of gonadotropin releasing hormone, equine chorionic gonadotropin and a single or double dose (24 h apart) of prostaglandin F. In Experiment I, the data were derived from 232 lactating cows. Binary logistic regression identified no effects of treatment on ovulation failure or multiple ovulation 10 days post artificial insemination (AI). Based on the odds ratio, the likelihood of ovulation failure was lower (by a factor of 0.1) in cows showing at least one corpus luteum (CL) upon treatment than in cows lacking a CL; repeat breeders (> 3 AI) and cows with multiple CLs at treatment showed lower (by a factor of 0.44) and higher (by a factor of 9.0) risks of multiple ovulation, respectively, than the remaining animals. In Experiment II, the data were derived from 5173 AIs. The independent variable treatment failed to affect the conception rate 28–34 days post AI, twin pregnancy or early fetal loss 58–64 days post AI. The results of this study demonstrate the efficacy of 5-day progesterone-based protocols for FTAI. All four protocols examined were able to induce ovulation in both cyclic and non-cyclic animals so that FTAI returned a similar pregnancy rate to spontaneous estrus. Our results suggest that the ovarian response and fertility resulting from each treatment are due more to the effect of ovarian structures at treatment than to the different combinations of hormones investigated.  相似文献

The percentage of dairy cows that were mated and became pregnant during a 42 day breeding period was not significantly different when animals were routinely observed for estrus and mated when detected (71%, n = 56), inseminated at estrus following a single injection of cloprostenol (72%, n = 29), or inseminated at a fixed time after two cloprostenol treatments 11 days apart (69%, n = 28). However, 11 cows in the control group were not detected and mated during the breeding period and inclusion of these animals reduced the actual pregnancy rate to 60%. Results indicated that a controlled breeding program could have practical application in dairy herds but should be used with caution. Practitioners must evaluate management programs and decide, in consultation with the herdsman, if the introduction of controlled breeding technology might be advantageous.  相似文献

The objective of this study was to use luteinizing hormone-releasing hormone A3 (LRH-A3) and human chorionic gonadotrophin (HCG) to improve pregnancy rate of dairy cows during timed artificial insemination (TAI). In experiment 1, the TAI process (0 d, GnRH, 100 μg; 7 d, PGF2α, 0.4 mg; 56 hr, GnRH, 100 μg; 16 hr, AI) was applied to 160 dairy cows on 50th and 60th days after parturition respectively. In experiment 2, 320 postpartum dairy cows were treated with TAI (Group A), TAI + 25 μg LRH-A3 (Group B), TAI + 1,500 IU HCG 5 days after AI (Group C), and TAI + 25 μg LRH-A3 + 1,500 IU HCG 5 days after AI (Group D). In experiment 3, endometrial cells were treated with HCG. The results showed that TAI did not affect the pregnancy rate, while LRH-A3 and HCG increased the pregnancy rate of the cow. HCG of 5 IU/ml and 10 IU/ml increased the expressions of leukemia inhibitory factor but decreased those of interleukin-6, epidermal growth factor and vascular endothelial growth factor in endometrial cells. This study provided a plan for the use of LRH-A3 and HCG to increase pregnancy rate during TAI in dairy cows.  相似文献

The relationship between the concentration of plasma progesterone (P4) during embryo attachment or at recognition of pregnancy, and that of pregnancy-associated glycoprotein (PAG) was assessed in dairy cows. The outcome of artificial insemination (AI) was classified as positive (AI+), negative (AI?), or late embryonic mortality (EM) by measuring circulating PAG concentrations and by ultrasonography. Based on P4 concentrations at either day 21 or day 15, AI+ and EM cows were classified into ‘low’ (P4 concentrations < mean) and ‘high’ (P4 concentrations > mean) P4 groups. In both experiments, the threshold of P4 concentration between the ‘low’ and ‘high’ groups was approximately 6 ng/mL. PAG concentrations were lower in the ‘low’ group only when P4 concentrations were below the threshold. The study findings suggest that a possible P4 threshold exists below which PAG secretion may be impaired.  相似文献
