An intramuscular injection of 500 I.U. of human chorionic gonadotrophin resulted in an increase of plasma testosterone and progesterone concentrations in the intact male dog, but had no effect on plasma 17B-estradiol concentration. Castration caused a rapid decrease in concentration of testosterone, progesterone, and 17B-estradiol, indicating that the tests were the major organs producing these hormones in the male dog.  相似文献

The glycoprotein hormones of the chicken pituitary gland, follicle‐stimulating hormone (FSH), luteinising hormone (LH) and thyroid stimulating hormone (TSH), have been fractionated and partially purified. Fractions were assayed for gonadotrophins using the testicular uptake of 32P in 1‐d‐old chicks and for thyro‐trophin by chick thyroidal 32P uptake.  相似文献

Ovine growth hormone ( oGH ) was tested for its effects on lipolysis of rat and ovine adipose tissue in vitro. Ovine growth hormone at 1, 5 and 25 micrograms/ml stimulated lipolysis (P less than .05) of chopped rat adipose tissue and isolated rat adipocytes incubated in the presence of 100 mU/ml adenosine deaminase and .2 micrograms/ml dexamethasone, but had no effect on lipolysis of chopped ovine adipose tissue or isolated ovine adipocytes. Isoproterenol, a beta-adrenergic agonist, stimulated lipolysis (P less than .05) of both rat and ovine adipose tissue. Contaminants of the oGH preparation used were examined for lipolytic effects. Thyroid-stimulating hormone (TSH), luteinizing hormone (LH) and adrenocorticotropic hormone (ACTH) content in oGH were measured by radioimmunoassay. When quantities of these hormones contaminating 5 and 25 micrograms oGH were tested for lipolysis in rat adipose tissue, the TSH contamination could account for some (30%) of the lipolysis observed with oGH , while the other hormones had no effect. Also, preincubation of oGH with anti-GH, but not with anti-TSH or anti-LH, removed the principle in oGH responsible for the lipolytic effect on rat adipose tissue.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The aims of the present study were to clarify the effect of kisspeptin10 (Kp10) on the secretion of growth hormone (GH) from bovine anterior pituitary (AP) cells, and evaluate the ability of sex steroid hormones to enhance the sensitivity of somatotrophic cells to Kp10. AP cells prepared from 8–11‐month‐old castrated calves were incubated for 12 h with estradiol (E2, 10?8 mol/L),progesterone (P4, 10?8 mol/L), testosterone (T, 10?8 mol/L), or vehicle only (control), and then for 2 h with Kp10. The amount of GH released in the medium was measured by a time‐resolved fluoroimmunoassay. Kp10 (10?6 or 10?5 mol/L) significantly stimulated the secretion of GH from the AP cells regardless of steroid treatments (P < 0.05), and E2, P4, and T had no effect on this response. The GH‐releasing response to growth hormone‐releasing hormone (GHRH, 10?8 mol/L) was significantly greater than that to Kp10 (P < 0.05). The present results suggest that Kp10 directly stimulates the release of GH from somatotrophic cells and sex steroid hormones do not enhance the sensitivity of these cells to Kp10. Furthermore, they suggest that the GH‐releasing effect of Kp10 is less potent than that of GHRH.  相似文献

Pit-1 is a pituitary-specific POU-domain DNA binding factor, which binds to and trans-activates promoters of growth hormone- (GH), prolactin- (PRL) and thyroid stimulating hormone-beta- (TSHbeta) encoding genes. Thyrotropin-releasing hormone (TRH) is located in the hypothalamus and stimulates TSH, GH and PRL release from the pituitary gland. In the present study, we successfully used the cell aggregate culture system for chicken pituitary cells to study the effect of TRH administration on the ggPit-l* (chicken Pit-1), GH and TSHbeta mRNA expression in vitro. In pituitary cell aggregates of 11-day-old male broiler chicks the ggPit-l * mRNA expression was significantly increased following TRH administration, indicating that the stimulatory effects of TRH on several pituitary hormones are mediated via its effect on the ggPit-l* gene expression. Therefore, a semiquantitative RT-PCR method was used to detect possible changes in GH and TSHbeta mRNA levels. TRH affected both the GH and TSHbeta mRNA levels. The results of this in vitro study reveal that ggPit-1 * has a role in mediating the stimulatory effects of TRH on pituitary hormones like GH and TSHbeta in the chicken pituitary.  相似文献

In young chickens plasma concentrations of growth hormone (GH) are depressed by prostaglandins (PG) E1 and E2, epinephrine, norepinephrine, alpha 2 and beta agonists or thyroid hormones. A primary culture of chicken adenohypophyseal cells was used to examine the direct effects of these agents at the level of the pituitary as evaluated by GH release in the presence and absence of growth hormone releasing factor (GRF). Following collagenase dispersion and culture (preincubation, 48 hr) cells were exposed (incubation, 2 hr) to test agents, except for thyroid hormones which were added during the preincubation, and incubation period. Growth hormone release was increased (P less than .05) in the presence of PGE1 (10(-8)M by 34%; 10(-7)M by 54%), PGE2 (10(-8)M by 29%; 10(-7)M by 29%), PGF2 alpha (10(-8)M by 28%), and the beta agonist isoproterenol (10(-7)M by 46%). Basal GH release from chicken pituitary cells was not affected by dopamine, norepinephrine, epinephrine, thyroxine (T4), triiodothyronine (T3), or alpha adrenergic agonists. Growth hormone releasing factor stimulated GH release was not affected by the presence of prostaglandins E1, E2 or F2 alpha in the incubation media. However, GRF stimulated GH release was reduced by high doses of catecholamines: dopamine (10(-6)M by 34%), norepinephrine (10(-6)M by 74%), epinephrine (10(-8)M by 47%; 10(-7)M by 41%; 10(-6)M by 89%), and by the alpha 1 adrenergic agonist, phenylephrine (10(-7)M by 52%), the alpha 2 agonist, clonidine (10(-8)M by 34%; 10(-7)M by 83%) and the beta agonist, isoproterenol (10(-7)M by 64%).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Feces were collected from two female and one male Siberian tigers, Panthera tigris altaica. Steroid hormones were extracted from lyophilized feces and quantified by enzyme immunoassay. The fecal contents of estradiol-17beta (E(2)) and testosterone in the females and male, respectively, changed markedly throughout the year. The fecal E(2) contents of females Nos. 179 and 238 increased at 26.4 +/- 8.0 and 28.0 +/- 14.2 day intervals, respectively. However, the fecal contents of progesterone (P(4)) in the female kept alone did not change. In contrast, the other female, which was kept with a male, had increased fecal P(4) contents after copulation. The fecal progesterone levels of the pregnant female remained high during her 106-day pregnancy.  相似文献

Two experiments were carried out to investigate the effect of sampling techniques on the plasma concentrations of pituitary and adrenocortical hormones in ferrets (Mustela putorius furo). In the first experiment blood was collected on two occasions from 29 ferrets which were either manually restrained or anaesthetised with isoflurane. In the second experiment eight intact ferrets were fitted with jugular catheters and blood was collected on four occasions, just before and as soon as possible after they had been manually restrained or anaesthetised with medetomidine or isoflurane; blood was also collected 10 and 30 minutes after the induction of anaesthesia. Medetomidine anaesthesia had no effect on the plasma concentrations of pituitary and adrenocortical hormones. Isoflurane anaesthesia resulted in a significant increase in the plasma concentration of alpha-melanocyte-stimulating hormone (alpha-MSH) directly after the induction of anaesthesia. Manual restraint resulted in a significant increase in the plasma concentrations of cortisol and adrenocorticotrophic hormone (ACTH) and a decrease in the plasma concentration of alpha-MSH.  相似文献

The present study examined sex differences and the involvement of gonadal hormones in stress responses caused by road transportation in Shiba goats. In experiment 1, we investigated the stress responses of males and females to transportation. Plasma levels of cortisol (Cor) significantly increased during 1 h of transportation, and those in females were significantly higher than those in males. Plasma glucose (Glu) and free fatty acid (FFA) levels also increased similarly in both females and males by transportation, and there were no sex differences. Food intake following transportation decreased only in males compared with that in the basal session, in which the animals were not transported. Experiment 2 examined the involvement of gonadal hormones in stress responses to transportation using castrated males. Goats were given cholesterol (Cho), 5α‐dihydrotestosterone (DHT) or 17β‐estradiol (Es). The plasma Cor levels increased during transportation regardless of hormone treatment, and those in DHT treated goats were significantly lower than those in Cho or Es treated animals. Plasma Glu and FFA levels also increased during transportation, regardless of hormone treatment, and there were no differences between treatments. Food intake following transportation was significantly lower than that in the basal session only in goats given DHT. In conclusion, gender affects Cor secretion that is increased by transportation and the decrease of food intake following transportation in Shiba goats, and the major cause of these differences is androgen.  相似文献

Calomys callosus is a wild rodent found naturally infected with different Trypanosoma cruzi strains. In the work described here, groups of male and female C. callosus were subjected to orchiectomy, ovariectomy and sham operation. One month after surgery, animals were inoculated intraperitoneally (i.p.) with 4x10(4) blood trypomastigotes of the "Y" strain of T. cruzi. Parasitemia, triglycerides, nitric oxide (NO) and concanavalin A (ConA)-induced proliferation were evaluated. Parasitemia during the course of infection was significantly higher in infected and sham operated animals as compared to infected orchiectomized animals. The opposite was observed in the ovariectomized and infected group. Orchiectomized and infected animals displayed elevated triglyceride levels, as well as a more vigorous immune response, with higher splenocyte proliferation and elevated concentrations of NO. Ovariectomy resulted in an impaired immune response, as observed by a reduction of splenocyte proliferation and NO concentration. The results suggest a pivotal role for gonadal hormones in the modulation of triglyceride levels and the magnitude of the immune response during the acute phase of T. cruzi infection.  相似文献

In order to clarify the functional relationship between thyroid, adrenal and gonadal hormones, hypothyroidism was induced by administration of thiuoracil in adult male and female rats, and the effects of hypothyroidism on the adrenal and the gonadal axes were investigated in the present study. 1. The functional relationship between thyroid and adrenal hormones: Adrenal weights and corticosterone were lowered, whereas the secretion of ACTH, corticotrophin-releasing hormone (CRH) and arginine vasopressin (AVP) increased in hypothyroid rats compared to euthyroid rats. These results indicate that hypothyroidism causes adrenal dysfunction directly and results in hypersecretion of CRH and AVP from the hypothalamus. 2. The functional relationship between thyroid and gonadal hormones: The pituitary response to LHRH was lowered, whereas the testicular response to hCG was not changed in hypothyroid rats. Hypothyroidism suppressed copulatory behavior in male rats. These results suggest that hypothyroidism probably causes dysfunction in gonadal axis at the hypothalamic-pituitary level in male rats. In adult female rats, hypothyroidism inhibited the follicular development accompanied estradiol secretion, whereas plasma concentrations of progesterone and prolactin (PRL) increased in hypothyroid female rats. Hypothyroidism significantly increased the pituitary content of vasoactive intestinal peptide (VIP) though it did not affect dopamine synthesis. These results suggest that hypothyroidism increases pituitary content of VIP and this increased level of VIP likely affects PRL secretion in a paracrine or autocrine manner. In female rats, inhibition of gonadal function in hypothyroid rats mediated by hyperprolactinemia in addition to hypersecretion of endogenous CRH.  相似文献

Gonadal hormones were implanted in pullets aged 12 weeks and the development of feather pecking was compared with that of unimplanted controls. Up to 18 weeks, progesterone produced a moderate but significant increase in pecking, oestrogen and progesterone together a much greater increase. From 18 to 24 weeks the normal onset‐of‐lay rise in pecking in the controls was prevented by testosterone. The findings suggest that the normal onset‐of‐lay rise is hormonal in origin and can be simulated by administering a combination of oestrogen and progesterone. The damaging forms of pecking can be blocked by giving testosterone, at least in pullets.  相似文献

Cytosolic assay was used to detect gonadal steroid receptors in brain tumor tissue from 6 dogs and 2 cats. For 4 samples, the maximal number of binding sites and the equilibrium dissociation constant were calculated, using Scatchard analysis. The goal of this study was to localize antigen presenting cells (APC), which may play roles in defense against pathogens and fertility, and examine the effects of age and gonadal steroids on their population in the rooster epididymis. Healthy White Leghorn male birds (immature 60-day-old birds; matured 150-, 330-, and 550-day-old), and immature birds treated with testosterone propionate (TP) or estradiol benzoate (EB) for 3 or 6 days were used. Cryostat sections of the epididymis and ductus deference were immunostained for Ia to identify APC. RT-PCR was performed to confirm the expression of major histocompatibility complex class II (MHC class II) mRNA in the epididymis. Ia+ cells were localized in the surface epithelium and subepithelial layer of the ductules and occasionally in the luminal content of the epididymis and ductus deference. RT-PCR analysis confirmed expression of MHC class II mRNA in the epididymis, ductus deferens, testis, and spleen. The frequency of Ia+ cells in the subepithelial layer was significantly greater in the proximal efferent ductules than in the other two types of ductules in the epididymis of 550-day-old birds. Although there were no significant differences in the frequencies in the subepithelial layer of the proximal efferent ductules between 60- and 150-day-old birds, the frequencies were significantly greater in 330- and 550-day-old birds than in 60-day-old birds. The frequencies of Ia+ cells in the ductus deferences was increased in the 150-day-old birds compared with the 60-day-old birds, with a larger increase in 330- and 550-day-old birds. The Ia+ cell frequency was significantly increased by EB-injection, but not by TP-injection, on Days 3 and 6 of injection compared with Day 0. These results suggest that the population of APC in the epididymis increases with age after sexual maturation, and estrogen may be one of the factors involved in induction of Ia+ cells.  相似文献

The effect of gonadal hormones on the plasma elimination and urinary metabolite profile of antipyrine was studied in dwarf goats. Female goats were treated with testosterone and male goats were treated with 17β-oestradiol. Castrated males were treated with either testosterone or 17β-oestradiol. Antipyrine (25 mg/kg, i.v.) was given both before and after the hormonal treatments. The effects of the hormonal status on the plasma elimination of the parent compound were not consistent. This was possibly due to the fact that formation of the main metabolite of antipyrine in the goat, 4-hydroxy antipyrine (OHA), was not affected by sex or hormonal treatment. On the other hand, there were clear effects of hormonal status on urinary excretion of the three other metabolites. In females and castrated males testosterone suppressed the formation of norantipyrine (NORA), 3-hydroxymethylantipyrine (HMA) and 4,4'-dihydroxyantipyrine (DOHA). Intact males produced smaller amounts of these metabolites than females. It is concluded that distinct xenobiotic metabolizing pathways exist in the dwarf goat, which are influenced in their activity by gonadal hormones. This confirms previous findings in rats and mice. The possibility that sex hormones influence drug metabolism in food-producing animals could have consequences for veterinary therapeutics and public health. This study also demonstrates that, when using the antipyrine test for the assessment of hepatic drug metabolism, it is very important to include the determination of metabolites.  相似文献

为进一步探讨黄芪多糖(APS)的免疫调节机制,建立了特异性荧光探针Fura-2/AM测定鸡脾脏淋巴细胞游离钙离子浓度的方法,观察了APS对体外培养的鸡脾脏淋巴细胞内游离钙离子水平的影响。结果表明:APS以200μg/ml的终浓度加入体外培养的鸡脾脏淋巴细胞培养液中,可极显著升高细胞内游离钙离子的水平(P<0.01)。提示黄芪多糖通过调节细胞内游离钙离子的浓度而影响机体免疫细胞的信号转导。  相似文献
