BACKGROUND: The B‐type Bemisia tabaci (Gennadius) has become established in many regions in China, and neonicotinoids are extensively used to control this pest. Imidacloprid resistance in a laboratory‐selected strain of B‐type B. tabaci was characterised in order to provide the basis for recommending resistance management tactics. RESULTS: The NJ‐Imi strain of B‐type B. tabaci was selected from the NJ strain with imidacloprid for 30 generations. The NJ‐Imi strain exhibited 490‐fold resistance to imidacloprid, high levels of cross‐resistance to three other neonicotinoids, low levels of cross‐resistance to monosultap, cartap and spinosad, but no cross‐resistance to abamectin and cypermethrin. Imidacloprid resistance in the NJ‐Imi strain was autosomal and semi‐dominant. It is shown that enhanced detoxification mediated by cytochrome‐P450‐dependent monooxygenases contributes to imidacloprid resistance to some extent in the NJ‐Imi strain. Results from synergist bioassays and cross‐resistance patterns indicated that target‐site insensitivity may be involved in imidacloprid resistance in the NJ‐Imi strain of B. tabaci. CONCLUSION: Although oxidative detoxification mediated by P450 monooxygenases is involved in imidacloprid resistance in the NJ‐Imi strain of B‐type B. tabaci, target‐site modification as an additional resistance mechanism cannot be ruled out. Considering the high risk of cross‐resistance, neonicotinoids should be regarded as a single group when implementing an insecticide rotation scheme in B. tabaci control. BACKGROUND: Laodelphax striatellus (Fallén) is a major pest of cultivated rice and is commonly controlled in China with the organophosphate insecticides. To develop a better resistance management strategy, a chlorpyrifos‐resistant strain of L. striatellus was selected in the laboratory, and its cross‐resistance to other insecticides and possible mechanisms of the chlorpyrifos resistance were investigated. RESULTS: After 25 generations of selection with chlorpyrifos, the selected strain of L. striatellus developed 188‐fold resistance to chlorpyrifos in comparison with the susceptible strain, and showed 14‐ and 1.6‐fold cross‐resistance to dichlorvos and thiamethoxam respectively. There was no apparent cross‐resistance to abamectin. Chlorpyrifos was synergised by the inhibitor triphenyl phosphate; the carboxylesterase synergistic ratio was 3.8 for the selected strain, but only 0.92 for the susceptible strain. The carboxylesterase activity of the selected strain was approximately 4 times that of the susceptible strain, whereas there was no significant change in the activities of alkaline phosphatase, acid phosphatase, glutathione S‐transferase and cytochrome P450 monooxygenase between the strains. The Michaelis constant of acetylcholinesterase, maximum velocity of acetylcholinesterase and median inhibitory concentration of chlorpyrifos‐oxon on acetylcholinesterase were 1.7, 2.5 and 5 times higher respectively in the selected strain. CONCLUSION: The high cross‐resistance to the organophosphate dichlorvos in the chlorpyrifos‐resistant strain suggests that other non‐organophosphate insecticides would be necessary to counter resistance, should it arise in the field. Enhanced activities of carboxylesterase and the acetylcholinesterase insensitivity appear to be important mechanisms for chlorpyrifos resistance in L. striatellus. BACKGROUND: Methoxyfenozide is a lepidopteran‐specific insecticide that belongs to a new group of insecticides, the non‐steroidal ecdysteroid agonists, also called moulting accelerating compounds (MACs). To investigate the risk of resistance and possible mechanisms conferring resistance to methoxyfenozide, the authors selected in the laboratory for a resistant strain of the cotton leafworm Spodoptera littoralis (Boisd.), which is a representative lepidopteran model and an important pest in cotton and vegetables worldwide, with a high risk for resistance development. RESULTS: After selection with methoxyfenozide during 13 generations, toxicity data showed that the selected strain developed fivefold resistance to methoxyfenozide in comparison with the susceptible strain. Measurement of the detoxification enzymes demonstrated that the monooxygenase (MO) activity was 2.1 times higher in the selected strain, whereas there was no change for esterases and glutathione‐S‐transferases. When the inhibitors piperonyl butoxide (PBO), S,S,S‐tributyl phosphorotrithioate (DEF) and diethyl maleate were tested as synergists, the respective synergistic ratios were 0.97, 0.96 and 1.0 for the susceptible strain, and 2.2, 0.96 and 1.1 for the resistant strain. The significant synergistic effect by PBO concurs with the increased MO activity in the selected strain. CONCLUSION: Taken overall, the present study supports the importance of MO‐mediated metabolism in resistance to methoxyfenozide, directing tactics to fight against resistance development for this novel group of insecticides. In teflubenzuron and teflubenzuron/chlorfluazuron laboratory-selected sub-populations of a Cameron Highlands (1988) strain of Plutella xylostella L., resistance to teflubenzuron reached much greater levels than chlorfluazuron but declined at a correspondingly greater rate to give similar resistance levels after 10–13 generations. There was little evidence of cross-resistance to flufenoxuron and none to hexaflumuron in these populations. Reselection of a Cameron Highlands population with teflubenzuron after 22 generations resulted in a significant (P < 0.05) increase in resistance after three generations. In two more recently collected strains from lowland Malaysia, Sawi (1991) and Serdang (1992) high levels of resistance to teflubenzuron were found, but only the former showed any apparent resistance to chlorfluazuron. There was also evidence of resistance to diflubenzuron in the Sawi strain (Serdang not examined). Resistance to teflubenzuron appeared to be unstable in unselected sub-populations of both lowland strains. Selection of the Sawi strain with teflubenzuron did not effect cross-resistance to chlorfluazuron, while attempted selection with chlorfluazuron had no effect on resistance to either compound. Selection of the Sawi strain with diflubenzuron increased resistance to teflubenzuron but had no effect on resistance to chlorfluazuron. In contrast to the Sawi strain, selection of the Serdang strain with teflubenzuron resulted in cross-resistance to chlorfluazuron. Synergist experiments with piperonyl butoxide (PB) and S,S,S-tributyl phosphorotrithioate (DEF) suggested that resistance to teflubenzuron in the lowland strains may involve both microsomal monooxygenases and esterases. The ovicidal activity of teflubenzuron, chlorfluazuron, hexaflumuron and flufenoxuron was consistently less than their activity against second-instar larvae, hexaflumuron, followed by teflubenzuron, showing the greatest relative ovicidal activity. Hexaflumuron was markedly more active than teflubenzuron against eggs from acylurea-selected Cameron Highlands populations and there was some evidence that resistance to teflubenzuron may be expressed at a lower level in eggs than in larvae in one such strain.  相似文献

Enhanced oxidative metabolism appeared to be a major factor involved in resistance to permethrin in a field strain of house flies, selected with permethrin over 4 years. This was shown in the 7.8-fold synergism by piperonyl butoxide which reduced the resistance ratio from 97 to 15. The rate of permethrin detoxication was significantly higher (P=0.05) in the resistant flies compared with a susceptible strain or resistant flies pretreated with piperonyl butoxide. The esterase inhibitor S,S,S-tributyl phosphorotrithioate did not reduce the level of resistance to permethrin in the resistant strain, although some hydrolytic metabolism was apparent. Rates of penetration were similar in susceptible and resistant flies and in resistant flies pre-treated with piperonyl butoxide. A minor unidentified resistance factor, possibly reduced sensitivity of the nervous system, may also have been present in the resistant strain.  相似文献

BACKGROUND

Tebufenozide is widely used to control populations of the smaller tea tortrix, Adoxophyes honmai. However, A. honmai has evolved resistance such that straightforward pesticide application is an untenable long-term approach for population control. Evaluating the fitness cost of resistance is key to devising a management strategy that slows the evolution of resistance.

RESULTS

We used three approaches to assess the life-history cost of tebufenozide resistance with two strains of A. honmai: a tebufenozide-resistant strain recently collected from the field in Japan and a susceptible strain that has been maintained in the laboratory for decades. First, we found that the resistant strain with standing genetic variation did not decline in resistance in the absence of insecticide over four generations. Second, we found that genetic lines that spanned a range of resistance profiles did not show a negative correlation between their LD50, the dosage at which 50 % of individuals died, and life-history traits that are correlates of fitness. Third, we found that the resistant strain did not manifest life-history costs under food limitation. Our crossing experiments indicate that the allele at an ecdysone receptor locus known to confer resistance explained much of the variance in resistance profiles across genetic lines.

CONCLUSION

Our results indicate that the point mutation in the ecdysone receptor, which is widespread in tea plantations in Japan, does not carry a fitness cost in the tested laboratory conditions. The absence of a cost of resistance and the mode of inheritance have implications for which strategies may be effective in future resistance management efforts. © 2023 The Authors. The desethylation (gene a) and penetration delaying (pen) factors of resistance to organophosphorus insecticides isolated by genetical methods from the diazinon-selected SKA strain of houseflies (Musca domestica L.) were inbred into a strain whose resistance was compared with that of flies with single factors of resistance and the SKA strain. Pen alone decreases kill marginally, and gene a alone raises LD50 to 1·4–48 times that of susceptible flies, depending on the insecticide. The two factors together increase resistance greatly to many organophosphorus insecticides (up to 5–10 times or more to diazinon, malathion-ethyl and chlorthion-ethyl than of flies with gene a only) indicating that the two factors interact. Interaction is greater against the thionates than the corresponding phosphates, probably because pen delays entry of thionates more than of the corresponding phosphates. The role in resistance of each factor when hetero-and homo-zygous, and the reasons for interaction, are discussed.  相似文献

Abamectin resistance was selected in the western flower thrips [Frankliniella occidentalis (Pergande)] under the laboratory conditions, and cross-resistance patterns and possible resistance mechanisms in the abamectin-resistant strain (ABA-R) were investigated. Compared with the susceptible strain (ABA-S), the ABA-R strain displayed 45.5-fold resistance to abamectin after 15 selection cycles during 18 generations. Rapid reversion of abamectin resistance was observed in the ABA-R strain in the absence of the insecticide selection pressure. Moderate and low levels of cross-resistance to chlorpyrifos (RR 11.4) and lambda-cyhalothrin (3.98) were observed in the ABA-R strain, but no significant cross-resistance was found to spinosad (2.00), acetamiprid (1.47) and chlorfenapyr (0.26). Our studies also showed that the esterase inhibitor S,S,S-tributyl phosphorotrithioate (DEF) and glutathione S-transferase inhibitor diethyl maleate (DEM) were not able to synergize the toxicity of abamectin, whereas the oxidase inhibitor piperonyl butoxide (PBO) conferred a significant synergism on abamectin in the ABA-R strain (SR 3.00). Biochemical analysis showed that cytochrome P450 monooxygenase activity of the ABA-R strain was 6.66-fold higher than that of the ABA-S strain. It appears that enhanced oxidative metabolism mediated by cytochrome P450 monooxygenases was a major mechanism for abamectin resistance in the western flower thrips.  相似文献

B-biotype Bemisia tabaci has developed high levels of resistance to many insecticides. To investigate the risks and explore possible mechanisms of resistance to diafenthiuron in B. tabaci, a 32.8-fold diafenthiuron-resistant strain (R-DfWf) was established after selection for 36 generations compared with the susceptible strain (S-Lab). Biochemical assays showed that the activity of cytochrome P450 towards p-NA was significantly higher (4.37-fold higher) in the R-DfWf strain than in the S-Lab strain. Similarly, the carboxylesterase (COE) activity and glutathione S-transferase (GST) activity were also significantly higher (3.12- and 1.83-fold higher, respectively) in the R-DfWf strain than in the S-Lab strain. The expression of five of seven P450 genes was significantly higher (>3-fold) in the R-DfWf strain than in the S-Lab strain. The expression of COE2 was significantly higher (>2.5-fold) in the R-DfWf than in the S-Lab strain. The expression of GST and GST2 was significantly higher (>2.3-fold) in the R-DfWf than in the S-Lab. Thus, cytochrome P450, COE and GST may appear to be responsible for the resistance to diafenthiuron in B. tabaci. It is also valuable for usage of insecticides for resistance management and control of this species.  相似文献

烟粉虱对螺虫乙酯的抗性监测及交互抗性测定   总被引:1,自引:1,他引:0  
为明确螺虫乙酯在湖北地区的抗性水平以及与其存在潜在交互抗性风险的常用药剂类型,采用着卵叶片浸渍法,于2013年对湖北省13个烟粉虱MEAM1和MED隐种地理种群对螺虫乙酯的抗药性进行了监测,并以室内敏感品系SUD-S为参照,对经螺虫乙酯连续17代抗性筛选的WH-2种群进行了交互抗性分析。结果表明:湖北省13个烟粉虱地理种群对螺虫乙酯均表现出极低的抗性,LC50介于0.015~0.081 mg/L之间;其中MED隐种WH-2种群对螺虫乙酯的抗性最高,LC50为0.081 mg/L。在室内用螺虫乙酯对MED隐种WH-2种群连续17代抗性筛选后,其对螺虫乙酯的抗性倍数上升约7倍。经螺虫乙酯筛选的WH-2种群对吡虫啉、乙酰甲胺磷、灭多威、氟氯氰菊酯、氰戊菊酯和氟啶虫胺腈均产生了低水平的交互抗性,抗性倍数分别为8.699、7.165、5.317、6.681、2.958、5.662倍,而对毒死蜱和阿维菌素无交互抗性。表明烟粉虱在湖北省部分地区对螺虫乙酯存在低水平抗性,且螺虫乙酯与部分常用杀虫剂存在交互抗性风险。  相似文献

BACKGROUND: The insect growth regulator pyriproxyfen has provided effective control of the whitefly Bemisia tabaci Gennadius in many countries. Here, whether or not fitness costs were associated with pyriproxyfen resistance in a laboratory‐selected resistant strain (QC02‐R) of the B biotype was determined. RESULTS: Mortality caused by pyriproxyfen and fitness traits over time were measured in unselected and selected hybrid strains, which were created by crossing individuals of the resistant strain with individuals of a susceptible strain. Fitness costs were not associated with resistance in QC02‐R, as mortality caused by pyriproxyfen did not increase over time in unselected hybrid strains and fitness traits were similar in unselected and selected hybrid strains. Using a new method to examine the inheritance of resistance, based on data from fitness cost experiments, it was estimated that pyriproxyfen resistance is controlled by two loci in the QC02‐R strain. CONCLUSION: The lack of fitness costs associated with pyriproxyfen resistance could promote the evolution of resistance in field populations with similar traits to QC02‐R. BACKGROUND: Plutella xylostella (L.) has evolved resistance to various kinds of insecticide in the field. Reversion and selection, cross‐resistance, inheritance and mechanisms of abamectin resistance were characterised in a field‐derived multiresistant population of P. xylostella from China. RESULTS: Compared with a susceptible Roth strain, the field‐derived TH population showed ~5000‐fold resistance to abamectin. Rapid reversion of abamectin resistance was observed in the TH population when kept without insecticide selection. The TH‐Abm strain, selected from the TH population with abamectin, developed 23 670‐fold resistance to abamectin, a high level of cross‐resistance to emamectin benzoate and low levels of cross‐resistance to spinosad and fipronil. Genetic analyses indicated that abamectin resistance in the TH‐Abm strain was autosomal, incompletely dominant and polygenic. P450 monooxygenase activities in the TH‐Abm strain were significantly elevated compared with the TH strain. Piperonyl butoxide (PBO) inhibited a small part of abamectin resistance in the TH‐Abm strain. CONCLUSION: Field‐evolved high‐level resistance to abamectin in the TH population was not stable. Selection of the TH population with abamectin resulted in an extremely high level of cross‐resistance to emamectin benzoate and low levels of cross‐resistance to spinosad and fipronil. BACKGROUND: The diamondback moth (DBM), Plutella xylostella (L.), is a cosmopolitan pest of cruciferous crops. Fufenozide, a novel non‐steroidal ecdysone agonist, exhibits good efficacy and plays an increasingly important role in the control of Lepidopterous pests in China. A laboratory strain of DBM was selected for resistance to fufenozide, and the genetic basis of resistance was studied. RESULTS: The resistant strain, selected under laboratory conditions, exhibited a higher level of resistance to fufenozide (302.8‐fold based on LC50s) than the laboratory susceptible strain. Mortality data from the testing of F1 progeny of reciprocal crosses of resistant and susceptible DBM indicated that resistance was autosomal and incompletely recessive with a degree of dominance of ?0.664. Chi‐square analysis from responses of a backcross of crossed F1 progeny and the resistant strain and F2 progeny were highly significant, suggesting that the resistance was probably controlled by more than one gene. The estimated realised heritability (h2) of fufenozide resistance was 0.08, indicating that diamondback moth may have a lower chance of developing resistance to fufenozide than other kinds of insecticide. CONCLUSION: The resistance of DBM to fufenozide might be autosomal and incompletely recessive, and the resistance is probably controlled by more than one gene. A French strain of the tobacco aphid Myzus nicotianae Blackman (Homoptera: Aphididae), strain FR, showed high tolerance to imidacloprid in short-term (48-h) oral ingestion bioassays when compared to a susceptible reference strain of Myzus persicae, strain NS. The resulting tolerance factors were >50. Measures of the contact activity of imidacloprid by the FAO dip method failed to detect these high factors of tolerance. The tolerance factor was in general <10 when using the dip method. The resulting difference between tolerance factors could be attributed to a behavioural component to fitness between strain FR and strain NS as further experiments revealed. When measuring the effect of systemically applied imidacloprid on honeydew excretion, a 50% reduction occurred in both strain FR and strain NS at nearly the same concentration of imidacloprid, providing evidence for a similar antifeedant response in both strains. Starvation experiments revealed that the French strain was able to survive approximately 24 h longer than a reference laboratory strain of M. persicae. This result coincided with the fact that systemically applied imidacloprid showed the same aphicidal potential against strain FR after three days as against strain NS after two days, i.e. 24 h later. After rearing in the laboratory for six months the French strain of M. nicotianae lost its hardiness and also its apparent ability to tolerate imidacloprid. However, strain FR was a heterogeneous field strain and it is possible that a susceptible variant out-reproduced a more hardy variant. These findings indicate that the type of bioassay is very important when assessing aphid populations for resistance against the chloronicotinyl insecticide imidacloprid, because of its distinct mode of action. It is obvious that an aphid dip test, i.e. FAO dip test, produces more reliable results than the different kinds of short-term oral ingestion bioassays, because of the reversible behavioural changes induced by imidacloprid after oral uptake. Thus a short-term oral ingestion bioassay (≤48 h) is not recommended for precise detection of possible resistance of Myzus sp. to imidacloprid, although this mode of uptake for imidacloprid might be sometimes more realistic in terms of field behaviour. The ideal test to generate most accurate data would be a slightly longer (72-h) feeding bioassay, perhaps used in conjunction with a dip test. A pyrethrins-resistant strain of houseflies, 213ab, previously selected with a 1:10 (by wt.) mixture of natural pyrethrins and piperonyl butoxide, was further selected either with natural pyrethrins alone (strain NPR) or with resmethrin (strain 104). After 50 generations the two populations differed in their resistance to the natural and synthetic esters. Both were resistant to all pyrethroids. Part of strain NPR was immune and very much more resistant than strain 104 to the natural pyrethrins and allethrin, but it was only 2–3 times more resistant than strain 104 against the new synthetic esters resmethrin (5-benzyl-3-furylmethyl (±)-cis-trans-chrysanthemate), bio-resmethrin (5-benzyl-3-furylmethyl (+)-trans-chrysanthemate), pyresmethrin (5-benzyl-3-furylmethyl pyrethrate) and 5B2Me3FC (5-benzyl-2-methyl-3-furylmethyl (±)-cis-trans-chrysanthemate). Pretreatment of both strains with sesamex diminished but did not eliminate resistance. Synergism was greater in strain NPR, especially with natural pyrethrins and allethrin. Both strains had great resistance to DDT indicating that resistance to DDT and pyrethroids is linked. Differences in resistance to different compounds suggest that at least three factors can confer resistance, one of which, pen, delays penetration and two others involve detoxication, one py a on the acid side of the ester linkage and the other, py b, on the alcohol side. Natural pyrethrins and resmethrin select for different groupings of these factors. Treatment with resmethrin does not select for py b presumably because this mechanism cannot attack the resmethrin molecule. Similarly when piperonyl butoxide is added to the natural pyrethrins py b is inhibited and so removed from selection pressure. Under these conditions, the strain produced contains the same factors as one selected by resmethrin and so shows the same small resistance to natural pyrethrins alone.  相似文献

Rhizomania, one of the most devastating diseases in sugar beet production, is caused by Beet necrotic yellow vein virus (BNYVV) and transmitted by Polymyxa betae. Previously, disease control was possible by cultivation of sugar beet hybrids carrying a major resistance gene Rz1, which restricts virus accumulation in taproots and suppresses symptom development. Over the last few years, BNYVV strains with four RNA components have arisen, which are able to overcome Rz1‐mediated resistance. All strains described so far possess an A67V amino acid exchange within the RNA3‐encoded P25 pathogenicity factor. In this study, BNYVV was isolated from Rz1 plants, collected in the United Kingdom, the Netherlands and Germany, displaying patches of strong rhizomania symptoms. Sequencing of the coat protein and P25 gene of three isolates showed 100% nucleotide sequence identity and detected AYPR as the P25 tetrad amino acid composition. The ability of this strain to accumulate to higher levels in young plants of Rz1 resistant but not in Rz1 + Rz2 resistant genotypes was initially demonstrated in a greenhouse assay in natural field soil from the Netherlands. This strain was loaded into a virus‐free P. betae population and compared to reference strains. The AYPR strain retained its resistance‐breaking ability in the Rz1 genotypes and displayed replication at a higher rate compared to the Rz1‐resistance‐breaking P type. The strain origin is unclear and it remains speculative whether the occurrence at different geographic locations is the result of independent selection or displacement of infested soil.  相似文献

为明确室内筛选的红铃虫Pectinophora gossypiella抗性品系AQ-R对Cry1Ac的抗性机制,采用室内生物测定法明确该品系对Cry1Ac和Cry2Ab的敏感性,通过遗传杂交和基因克隆分析抗性基因的显隐性及突变位点,并进行细胞学试验分析突变蛋白的亚细胞定位。结果显示:红铃虫AQ-R抗性品系对Cry1Ac的抗性倍数为181.67倍,对Cry2Ab没有交互抗性;该品系携带了一种新型的隐性钙粘蛋白抗性等位基因PgCad1,其编码蛋白的钙粘蛋白重复区、前蛋白区和近膜区共发生了17个氨基酸替换。表达野生型PgCad1-s基因的Hi5细胞对Cry1Ac敏感,且钙粘蛋白定位于细胞膜;而表达抗性PgCad1-r基因的Hi5细胞则对Cry1Ac不敏感,且钙粘蛋白错误定位到内质网。表明钙粘蛋白氨基酸点突变能导致其定位错误,从而促成红铃虫AQ-R品系对Cry1Ac产生抗性。  相似文献

Disease resistance mediated by the resistance gene Xa21 is developmentally controlled in rice. We examined the relationship between Pathogenesis Related (PR) defense gene expression and Xa21-mediated developmental disease resistance induced by Xanthomonas oryzae pv. oryzae (Xoo). OsPR1a, OsPR1b, and OsPR1c genes were cloned and their induction was analyzed, in addition to the OsPR10a gene, at the juvenile and adult stages in response to a wildtype Xoo strain that induces a resistance response (incompatible interaction) and an isogenic mutant Xoo strain that does not (compatible interaction). We found that the adult stage leaves are more competent to express these OsPR1 genes and that the Xa21 locus is required for the highest levels of induction.  相似文献

Activities of a range of pyrethroidal esters, incorporating structural variations in all regions of the acid and alcohol components, have been measured against two fully characterised and homozygous resistant strains of Musca domestica (kdr and super-kdr). The results, limited in this paper to esters of alcohols with acyclic side chains, show the same level of uniform resistance as the kdr strain observed previously, across a range of acid and alcohol variations. Against the super-kdr strain, in contrast to the cyclic side chain set, resistance factors are barely higher than for the kdr strain. Against super-kdr flies, some dependence of resistance factor on the position of the side chain substituent was detected. Polyfluorobenzyl esters confer particularly low resistance factors.  相似文献

A resistant laboratory strain of the German cockroach, Blattella germanica, was developed from a normal laboratory strain by selection with propoxur. Resistance to all insecticides except chlordane began increasing after 15 generations of selection and reached a plateau for most insecticides by generation 27. The resistant colony, designated B-strain, developed significant resistance to carbamates, organophosphorus compounds, pyrethrins and DDT, developed low resistance to gamma-BHC and no resistance to chlordane. The resistance spectrum, effect of synergists and inheritance of resistance of this propoxur resistant strain are similar to a diazinon resistant strain. Therefore, diazinon and propoxur may select for the same resistance mechanism(s) in this species. The practical implications of this research are discussed.  相似文献
