Epicoccum purpurascens, a component of the bark micromycota, was evaluated as a potential biocontrol agent for three decay‐causing hymenomycetes frequently found in Buenos Aires city trees, Ganoderma platense, Inonotus rickii and Rigidoporus ulmarius. Treatments consisted of dual cultures of E. purpurascens and xylophagous fungi on box elder (Acer negundo) wood blocks. Two inoculation sequences were tested: (i) the biocontrol agent as the first microorganism inoculated followed by the pathogen; and (ii) the xylophagous fungus as the first microorganism inoculated followed by the biocontrol agent. After 3 months of incubation, percentage dry weight loss was calculated. When E. purpurascens was allowed to colonize wood blocks before the introduction of the xylophagous fungus, dry weight loss was significantly reduced in all instances. However, as a secondary colonizer, E. purpurascens mildly reduced the weight loss caused by G. platense, had no effect on degradation by I. rickii and increased wood weight loss by R. ulmarius.  相似文献

Teratosphaeria gauchensis (Capnodiales) causes a serious stem canker disease on commercially propagated Eucalyptus species in South America. Recently, this pathogen was detected for the first time in Africa. Very little is known regarding the biology or origin of Tgauchensis, but it has been suggested that it is native to South America. The aim of this study was to compare isolates from Africa and South America using microsatellite markers. Bayesian analysis conducted in STRUCTURE, principal coordinates analysis and a UPGMA dendrogram revealed two distinct genetic groups for these isolates. The South American isolates were more genetically diverse than those from Africa. Patterns of genetic diversity in Africa suggest that T. gauchensis could have been introduced into Zimbabwe before spreading north‐eastwards. The existence of the two genetic groups and high haplotype richness associated with the South American and Zimbabwean populations suggest that it will be more difficult to reduce the impact of disease caused by T. gauchensis in these regions than in those areas where there is limited genetic diversity.  相似文献

Inonotus rickii was detected for the first time causing cankers and decay in Acer negundo and Celtis australis in Italy. In a boxelder boulevard, declining trees showed sparse foliage, exudations and cracks in the bark; in some cases, chlamydospore masses were present. Five isolates were collected and compared by growth tests in vitro and electrophoretic analyses; three isolates from the same boulevard showed very similar physiological characters. The increasing importance of the pathogen in urban areas is underlined and discussed.  相似文献

Chrysoporthe austroafricana is one of the most damaging pathogens of Eucalyptus trees in southern Africa. It also occurs on non‐native Tibouchina granulosa trees and native Syzygium species. Additional isolates of the pathogen from previously unstudied countries in the region have become available from survey studies. The aim of this study was to use VCGs to consider the diversity in populations of isolates collected in various countries in southern Africa (Malawi, Mozambique, Namibia, South Africa and Zambia) and from different hosts. We also wanted to determine whether there are shared VCGs among these countries and hosts in southern Africa and establish a VCG tester strain database. Results showed a high diversity amongst isolates from different countries and hosts, but suggested little movement of VCGs among countries or hosts based on the available isolates. A total of 108 VCG tester strains were identified for southern Africa.  相似文献

Diplodia scrobiculata, a latent pathogen of Pinus spp. and other conifers with a limited distribution in the United States, Mexico and southern Europe, has not been reported previously in the southern hemisphere. This is unlike its close relative Diplodia pinea that is found in most parts of the world where pines are native or have been introduced. During an intensive D. pinea survey conducted in Pinus patula plantations in eastern parts of South Africa, a small number of isolates atypical of D. pinea were found. Morphological studies and DNA sequence comparisons showed that these isolates represent D. scrobiculata. Microsatellite analyses suggest that the South African isolates of D. scrobiculata might have originated from California. Pathogenicity tests showed that some of the D. scrobiculata isolates were as pathogenic as those of D. pinea on Pinus radiata and Pinus elliottii.  相似文献

Raffaelea quercus‐mongolicae is a fungus associated with oak wilt and deemed to cause extensive oak mortality in South Korea. Since the discovery of this fungus on a dead Mongolian oak (Quercus mongolica) in 2004, the mortality continued to spread southwards in South Korea. Despite continued expansion of the disease and associated significant impacts on forest ecosystems, information is lacking about the origin and genetic diversity of Rquercus‐mongolicae. Restriction‐site‐associated DNA (RAD) sequencing was used to assess genetic diversity and population structure among five populations (provinces) of Rquercus‐mongolicae in South Korea. In total, 179 single nucleotide polymorphisms (SNPs) were identified among 2,639 RAD loci across the nuclear genome of the 54 R. quercus‐mongolicae isolates (0.0012 SNPs per bp), which displayed an overall low expected heterozygosity and no apparent population structure. The low genetic diversity and no apparent population structure among South Korean populations of this ambrosia beetle‐vectored fungus support the hypothesis that this fungus was introduced to South Korea.  相似文献

The F intersterility group of Heterobasidion annosum, specialized to Abies species and earlier reported from southern parts of Europe, was identified from Abies alba and Picea abies in the Carpathian Mountains in southern Poland, close to northern limit of the natural distribution of A. alba. However, the fungus is not common in this area and seems to be primarily a saprophyte, growing on stumps and dead trees.  相似文献

The blue‐stain fungus Ceratocystis resinifera colonizes wounds on living Picea spp. and other conifers in Europe and North America. Little is known regarding the pathogenicity of this fungus and consequently, four Norwegian C. resinifera isolates were inoculated on to Norway spruce (Picea abies) using two different techniques. These included single‐point inoculations on young trees (two inoculations per tree on 14‐year‐old trees) and mass‐inoculations on older trees (~200 inoculations per tree on 34‐year‐old trees). In both experiments, C. resinifera induced minor symptoms that in most cases did not differ significantly from inoculation with sterile agar. The virulent blue‐stain fungus C. polonica, which was inoculated for comparative purposes, induced extensive symptoms, causing 83% dead cambium circumference and 82% blue‐stained sapwood, and long necrotic lesions in the phloem. The results suggest that C. resinifera is non‐pathogenic or only mildly pathogenic to Norway spruce and does not present a threat to these trees.  相似文献

Ceratocystis albifundus causes the disease known as wattle wilt of non‐native Acacia mearnsii trees in South Africa, Uganda and Kenya. Infection results in rapid wilt and death of susceptible trees and stem cankers on more tolerant trees. It has been suggested that C. albifundus is indigenous to southern Africa, possibly having spread from native Protea spp. to non‐native A. mearnsii and A. decurrens trees. Although C. albifundus has been collected from Protea spp., these reports are based on limited records for which only aged herbarium specimens exist. During surveys of wound‐infecting fungi on native tree species in South Africa, a fungus resembling C. albifundus was collected from Protea gaguedi, Acacia caffra, Burkea africana, Combretum molle, C. zeyheri, Faurea saligna, Ochna pulchra, Ozoroa paniculosa and Terminalia sericea. The identity of the fungus was confirmed as C. albifundus, using comparisons of DNA sequence data for the ITS and 5.8S gene of the rRNA operon. In pathogenicity trials, lesions were produced on C. molle and A. caffra, with some trees beginning to die at the termination of the experiment. This study represents the first report of C. albifundus from native tree species in South Africa and provides unequivocal evidence that the fungus occurs naturally on native Protea spp. The wide host range of C. albifundus, as well as its abundance on these indigenous hosts lends further support to the view that it is a native African pathogen.  相似文献

Armillaria root rot is a well‐known disease on a wide range of plants, world‐wide. In Ethiopia, the disease has previously been reported on Pinus spp., Coffea arabica and on various native hardwoods. The causal agent of the disease has been attributed to Armillaria mellea, a species now known to represent a complex of many different taxa. The aim of this study was to determine the extent of Armillaria root rot and the identity of the Armillaria sp. in Ethiopian plantations. As part of a plantation disease survey in 2000 and 2001, samples were collected in plantations at and around Munessa Shashemene, Wondo Genet, Jima, Mizan and Bedele, in south and south‐western Ethiopia. Basidiocarps were collected and their morphology studied. Morphological identification was confirmed by sequencing the intergenic spacer (IGS‐1) region of the ribosomal rRNA operon and comparing data with published sequences of Armillaria spp. Armillaria isolates were collected from Acacia abyssinica, Pinus patula, Cedrela odorata and Cordia alliodora trees. Sporocarps were found on stumps of native Juniperus excelsa. Basidiocarp morphology and sequence data suggested that the fungus in Ethiopia is similar to that causing disease of Pinus spp. in South Africa and previously identified as A. fuscipes. This identification was confirmed for all isolates, based on sequence data. Armillaria fuscipes is known to be common in southern Africa. Its widespread occurrence in Ethiopia suggests that it is also the major cause of Armillaria root rot in that country.  相似文献

To investigate the taxonomy of Heterobasidion in Eurasia, 49 specimens belonging to H. annosum sensu lato from Asia were identified with the aid of mating tests. Most of the specimens originated from north‐eastern and south‐western China and from the Altai region in southern Siberia, but a few isolates from Kirghizia, Japan and India were also tested. In addition to mating tests, the material from China was investigated with DNA fingerprinting. Heterobasidion annosum sensu stricto was identified only from the Altai region. Homokaryotic isolates from other specimens, except the Indian ones, were sexually compatible with H. parviporum, but they also showed a high degree of compatibility with H. abietinum and with the North American S group. The isolates from SW China (eastern Himalayas) mated with about equal frequency with the European strains of H. parviporum and H. abietinum. However, the DNA fingerprinting showed that these isolates were more closely related to H. parviporum, and hence they were tentatively included in this species. The North American S group was more distant from these Eurasian taxa. Four old isolates from India mated only weakly with the members of the H. annosum s.lat. According to the species concept presented, the distribution of H. parviporum extends from western Europe through southern Siberia to northern China, Japan and the eastern Himalayas. H. annosum s.str. is so far identified only from the Altai region outside Europe, and H. abietinum only from Europe.  相似文献

The basidiomycete Laetiporus sulphureus (Bull) Murrill is a forest pathogen causing brown cubical heart rot in a broad range of host trees. Despite its wide distribution and importance, studies aimed at understanding the epidemiology of the fungus in specific areas or hosts are lacking. In this study, an incidence of L. sulphureus as high as 34% was determined through molecular analysis of wood samples collected from 70 carob (Ceratonia siliqua L.) trees in the South‐west of Sicily, Italy. A phylogenetic analysis of Internal Transcribed Spacer (ITS) sequences indicated that all carob isolates belonged to the cluster E of Laetiporus taxonomy. Ten molecular markers based on Single Sequence Repeats (SSRs) designed on the L. sulphureus genome were developed, and isolates were genotyped through High‐resolution Melting (HRM) analysis. High gene diversity (0.581), no correlation between fungal genotype and host tree species and significant correlation between spatial and genetic distance were observed, suggesting an important role of basidiospores in the epidemiology of the fungus and a risk of transmission from a host tree species to the others. Finally, from a prognostic perspective, significant differences among isolates in terms of mycelial growth suggest that in addition to an accurate identification, a phenotypic characterization of isolates affecting trees may also be important.  相似文献

Gremmeniella abietina isolates from Pinus contorta in northern Sweden produced, in vitro, shorter conidia with fewer septa compared with isolates from Pinus sylvestris in the southern part of the country. After mycelial inoculation of shoots with G. abietina isolates from both host species, the resulting necroses were longer in P. sylvestris than in P. contorta. Keeping seedlings in artificial mild winter climate or detaching shoots from the seedling before inoculation caused longer necroses. No host specificity in colonization was found. Isolates from P. sylvestris caused longer necroses than did isolates from P. contorta, and both types of isolates caused longer necroses in P. sylvestris than in P. contorta. The differences found between the two G. abietina populations probably reflect regional variation in the fungus.  相似文献

In a recent survey of Eucalyptus clones in the Republic of Congo, Central Africa, a serious wilt and die-back disease of two different hybrid clones was observed. Affected trees ranged in age from approximately 6 months to 4 years. Isolations from symptomatic plant material consistently yielded a Ceratocystis species. On the basis of morphology and sequence data this fungus was identified as Ceratocystis fimbriata, a well-known wilt and canker pathogen of many economically important plants. The Eucalyptus isolates were compared with other Ceratocystis spp. based on sequence data generated from the ITS and 5.8S region of the rRNA operon. The results confirmed the identity of the Ceratocystis isolates from Eucalyptus as C. fimbriata and showed that they group with other C. fimbriata isolates from Brazil, South Africa and Europe. Inoculations on young Eucalyptus plants were conducted in the greenhouse and all three of the Congolese isolates tested, produced typical lesions in the bark and xylem. This study represents the first report of C. fimbriata as a pathogen of Eucalyptus in Africa. This is a serious new disease that will require considerable study in order to ensure that losses, caused by C. fimbriata, do not continue.  相似文献

The virulence of Armillaria ostoyae isolates from coastal (16) and interior (33) British Columbia, elsewhere in North America (eight) and Europe (six) was assessed on 2‐year‐old Douglas‐fir seedlings in pots during a 3‐year trial. Isolates from most geographical locations infected similar proportions of seedlings, had similar average damage scores and killed a similar percentage of diseased seedlings. Isolates from the coastal region had a significantly higher probability than interior isolates that a diseased seedling received a damage score > 3 on a 1–5 scale, and coastal isolates killed a higher proportion of diseased seedlings than interior isolates. The mean damage score for isolates that had been in culture for 20–25 years was about 25% lower than that for recently collected isolates. The results indicate that the higher incidence and longer duration of mortality in the southern interior of British Columbia compared to the coast can not be attributed to greater virulence of interior isolates of A. ostoyae.  相似文献

A case of variability in Sphaero sis sapinea . Two isolates (type I and type II) of Sphaeropsis sqinea were isolated from the co lar of a declining Cedrus deo daa in the South of France. Cultural characteristics on PDA and MEA as well as conidial dimensions were compared with those of types A and B of S. sapinea described to occur in North America. Type II was similar to type A in respect to cultural aspects and growth but distinctly different in conidial dimensions. Slower growing type I was similar to type B. However, type I grew faster on PDA, had smaller conidia and lacked microconidia. Single-conidia isolates obtained from the four types showed stability of these characteristics.  相似文献

The efficiency of RAPD analysis in the evaluation of genetic variability in Hypoxylon mediterraneum has been tested on 49 monoascospore isolates of the fungus from different localities. A large genetic variability was detected within single stromata and among monoascospore isolates in a restricted area. The large variability in restricted populations can be explained by the heterothallic mating system and the life cycle of the fungus and its ecological adaptations. RAPD markers can discriminate monoascospore isolates both in the single stromata and among stromata. The application of RAPD markers for epidemiological studies is discussed.  相似文献

Pinus leiophylla is a relatively common pine that occurs in the mountains of western and central Mexico. Between 1987 and 1990, Camcore, North Carolina State University, sampled 11 populations and 309 mother trees of the species to determine patterns of genetic variation in survival, growth and stem form. Fifteen provenance/progeny trials were established in southern Brazil and South Africa across a wide range of sites and assessed at 3, 5 and 8 years of age for height, diameter and stem form. Results indicated that average productivity ranged from 5 to 19m3 ha?1 y?1 depending on the site and that performance was not as competitive as P. patula (South Africa) and P. taeda (Brazil) controls. Provenances from central Mexico (Michoacán) were statistically better in productivity than those from the southern part of the country (Oaxaca), which in turn were superior to those from the northern part of the country (Durango). However, provenances from northern Mexico exhibited superior survival to seed sources from other locations in Mexico when grown on cold sites (winter minimums –10 to 5°C) in South Africa and superior stem form across all locations. Seedlings from the original 11 provenances were screened for resistance to the pitch canker fungus (PCF; Fusarium circinatum). Results indicated that generally P. leiophylla is as susceptible to PCF as P. patula (stemkill = 85%) with the exception of two fast-growing, moderately resistant populations from Michoacán, La Pinalosa (stemkill = 44%) and Ario de Rosales (stemkill = 73%). The future potential of P. leiophylla might be in hybrid combination with P. patula in the seasonally dry areas of southern Africa where fires are common and pitch canker is a problem.  相似文献
