云南、福建、湖南烟区烟草花叶病主要病毒种类检测及黄瓜花叶病毒亚组鉴定 总被引:10,自引:0,他引:10
采用抗原直接包被和双抗体夹心酶联免疫吸附测定法(ELISA)对采自云南、福建、湖南烟区烟草花叶病样品进行了病毒种类检测,利用三抗体夹心ELISA对黄瓜花叶病毒(Cucumber mosaic virus,CMV)的亚组类型进行了鉴定。在云南采集的520个花叶病样品中,烟草花叶病毒(Tobacco mosaic virus,TMV)、CMV和马铃薯Y病毒(Potato virus Y,PVY)总检出率分别为71.74%、55.01%和6.35%;在福建采集的150个花叶病样品中,TMV、CMV和PVY的总检出率分别为94%、24.66%和8.00%;在湖南采集的74个花叶病样品中,TMV、CMV和PVY的总检出率分别为58.11%、51.35%和2.70%。部分样品为2种以上病毒复合侵染。云南、福建和湖南采集的64个CMV阳性样品中,属亚组Ⅰ的样品为57个,占89.1%;属亚组Ⅱ的样品为10个,占15.6%;其中3个样品为亚组Ⅰ和亚组Ⅱ的复合侵染。 相似文献

1988年自福建省亚热带植物研究所果园杂交种西番莲(Passif lora edulis XP.edulis var.flavicarpa)表现环斑花叶和果实木质化病状的病状的病株分离到一个黄瓜花叶病毒(Cucumber mosaic cucumovirus,CMV)分离物。该病毒分离物寄主范围广泛,在供试的西番莲、藜科、茄科、葫芦科、豆科、夹竹桃科、苋科、菊科、番杏科、胡麻科、十字花科等11科33种(或品种)植物中,能侵染西番莲科、藜科、茄科、葫芦科、豆科、夹竹桃科、苋科、菊科、番杏科、胡麻科等10科的24种(或品种)植物。 相似文献

中国北方番茄主要病毒种类普查及TMV、CMV株系鉴定 总被引:3,自引:0,他引:3
1990—1994年,对中国北方15省(市)番茄4种主要病毒种类进行了普查,在3761份样本中,烟草花叶病毒(TMV)和黄瓜花叶病毒(CMV)的检出率分别为34.5%—65.7%和11.6%—62.6%,马铃薯X病毒(PVX)和马铃薯Y病毒(PVY)的检出率一般为10%—20%。与此同时,也对番茄上的TMV和CMV进行了与抗病毒育种紧密相关的株系分化研究,在333个TMV分离物中鉴定出0、1、2和1.2株系,以0和1株系为主,在绝大多数省(市)中分别占分离物的50%和30%以上;在232个CMV分离物中鉴定出轻花叶、重花叶、坏死和黄化株系,以轻花叶和重花叶株系为主,在绝大多数省(市)中分别占分离物的30%和50%以上,这一研究结果为番茄病毒病的综合防治和抗病毒育种提供了科学依据。 相似文献

广西局地西番莲病毒病的病原鉴定及优势病毒分析 总被引:3,自引:3,他引:0
在广西采集表现斑驳、花叶症状疑似病毒病的西番莲叶片样品,利用小RNA测序技术,结合生物信息学分析,鉴定侵染西番莲的病毒种类。参考测序结果采用DAS-ELISA和RT-PCR方法对2015~2018年间采集的385份疑似病毒病样品进行检测,分析引发广西西番莲病毒病的优势病毒种类。结果显示:小RNA共获得20 921 061 clean reads,拼接获得560个contigs,其中99个contigs被注释为夜来香花叶病毒(telosma mosaic virus,TeMV),97个contigs被注释为黄瓜花叶病毒(cucumber mosaic virus,CMV),69个contigs被注释为东亚西番莲病毒(East Asian passiflora virus,EAPV),12个contigs被注释为大豆花叶病毒(soybean mosaic virus,SMV)。提取送测序的同批样品叶片的总RNA进行RT-PCR验证,可检测出TeMV、EAPV和CMV 3种病毒,未检出SMV。采用DAS-ELISA和RT-PCR对采集的样品进行检测,结果发现284份样品为阳性样品,检出率为73.76%,其中TeMV的检出率最高为64.16%,其次为EAPV和CMV,检出率分别为41.30%和11.43%;3种病毒存在复合侵染现象,其中TeMV+EAPV的检出率最高为24.94%,TeMV+CMV的检出率为4.16%,EAPV+CMV的检出率为0.26%,3种病毒复合侵染的检出率为4.94%。 相似文献

白术矮化病毒病病原的分子鉴定和部分序列分析 总被引:2,自引:0,他引:2
本试验在生物学接种的基础上,利用非序列依赖性PCR扩增(sequence-independent amplification,SIA)对白术矮化病毒病病原进行了分子鉴定,序列测定及分析。结果发现,具有矮化症状的白术为黄瓜花叶病毒(Cucumber mosaic virus,CMV)所侵染。为明确CMV白术分离物(CMV-Am)的分类地位,本研究进一步克隆了CMV-Am的外壳蛋白基因(CP)和移动蛋白基因(MP)全序列。序列比对分析表明,CMV-Am与CMV亚组ⅠB中株系XJ2序列同源性最高,核苷酸、氨基酸序列同源性分别为98.9%、99.1%。氨基酸序列同源性聚类分析表明,CMV-Am与中国大多数CMV分离物一样,属于CMV亚组ⅠB。 相似文献

我国部分地区常见农作物上黄瓜花叶病毒分离物核酸多样性分析 总被引:2,自引:2,他引:0
为明确我国黄瓜花叶病毒株系分化及系统进化基本情况,从湖南、新疆、青海和海南4省区采集1 367个样品对其进行酶联免疫和RT-PCR检测,并对分离获得的15个黄瓜花叶病毒(Cucumber mosaic virus,CMV)纯化分离物CP、MP、2b核苷酸序列进行相似性和进化树分析及生物学性状比较。结果表明,辣椒、龙葵和黄瓜的CMV阳性检出率较高,分别为54.13%、29.19%和18.46%。进化树分析显示CMV-Q5与CMV亚组II的亲缘性较高;CMV-N7为新发现的重组株系,其CP、2b基因属于CMV亚组IB,MP基因却属于CMV亚组II;其余13个分离物均属于CMV亚组IB。CMV-N7和CMV-Q5在系统寄主心叶烟和枯斑寄主苋色藜上引发的症状相似,但比对照株系CMV-P3613(IB)的发病时间要晚1~2 d,系统花叶较温和,枯斑较小。表明在以上4省区常见农作物上广泛流行的CMV存在分子变异。 相似文献

本文对枯萎病菌侵入黄瓜不同抗性品种的途径,以及黄瓜受侵染根部维管组织的变化进行了观察。分生孢子在幼根表皮上萌发产生菌丝,通过胞间层侵入表皮细胞,之后菌丝继续向内部生长,穿过皮层组织进入导管。受侵染的导管内相继出现壁覆盖物、侵填体及褐色物。抗病品种的壁覆盖物比感病品种的厚:侵填体由受侵染导管旁边的薄壁细胞产生,一个细胞可产生多个侵填体,并在侵填体中观察到细胞核的存在,感病品种中的侵填体发育不充分,抗病品种中的侵填体能完全堵塞导管;谒色物在感病、抗病品种中均能完全堵塞导管,对菌丝的侵入构成了较强的机械障碍。 相似文献

Cucumber mosaic virus (CMV) has resulted in much damage to Danshen (Salvia miltiorrhiza Bunge) crops in China. A 5-year survey was conducted in Hebei, Shandong, Sichuan, Shanxi, Henan and Gansu Provinces, all
major Danshen-growing areas. A total of 156 Danshen plant samples with CMV symptoms were collected and tested for the presence
of CMV by a double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) using polyclonal antibodies against CMV
and a positive control, according to the supplier’s instructions (Agdia). They were confirmed to be CMV by amplification of
complete coat protein gene and analysis of the gene sequence. The results showed that 122 samples were infected by CMV and
all of these CMV isolates belonged to subgroup IB.

Serological and biological variation between and within subgroup I and II strains of cucumber mosaic virus 总被引:1,自引:2,他引:1
Fourteen strains of cucumber mosaic virus (CMV) from Australia have been characterized by their host range and symptomatology. They were classified as subgroup I or II strains by a dot-blot molecular hybridization assay between their total viral RNAs and selected cDNAs. The strains FNY and LNy , both from the USA, were used as the subgroup I- and subgroup II-type strains, respectively. A range of serological tests was used to compare these isolates. Gel immunodiffusion tests, with standard antigens homologous to the antisera prepared against glutaraldehyde-fixed virus of 11 strains, showed that they could be divided into three serogroups on the basis of spur formation in heterologous reactions. Two of the serogroups included either subgroup I or subgroup II isolates, whereas the third serogroup consisted of only one strain (YWA ) which was homologous to all the strains tested. Use of heterologous standard antigens in this test failed to show further subgrouping of the antigens. Double-antibody sandwich (DAS) ELISA using polyclonal antibodies to distinct virus strains also placed the 14 strains in the same three serogroups. When eight different monoclonal antibodies (MAbs) were used in indirect ELISA, one of them distinguished subgroup-I strains and another distinguished subgroup-II strains; the YWA strain fell into subgroup II. Other MAbs showed narrower or broader specificity. Thus both molecular hybridization with total RNA and specific MAbs may be useful for separating isolates of CMV into subgroups I and II. Spur formation using heterologous standard antigens to the antisera, as well as being more difficult to interpret, was not a reliable criterion for classification. 相似文献

Symptoms of Cucumber mosaic virus (CMV) on yellow passion flower ( Passiflora edulis f. flavicarpa ) are characterized by bright yellow mottling on leaves, starting at random points on the vine and diminishing in intensity towards the tip, which becomes symptomless as it grows. To determine whether symptomless portions of vines are CMV-free or represent latent infection, leaves with and without symptoms were collected from infected vines in the field. Biological, serological (plate-trapped antigen enzyme-linked immunosorbent assay, PTA-ELISA), Western blot and dot-blot hybridization assays showed that portions of the vines without symptoms were CMV-free. Vegetatively propagated vines with symptoms showed remission of symptoms on newly developed leaves. One year later, no CMV was detected in the upper leaves of these plants. Mechanically inoculated passion flower seedlings behaved similarly; symptoms were shown by few leaves after inoculation. Afterwards, plants became symptomless and CMV was not detected in the upper leaves or root system, 40 or 85 days after inoculation. The mechanism responsible for remission of symptoms accompanied by CMV disappearance is not known. 相似文献

为调查我国太子参主产区病毒病发生情况,使用RT-PCR法对采自福建省柘荣县,贵州省施秉县、丹寨县和安徽省宣城市的71份具有典型病毒病症状的太子参样品进行检测,并根据CP氨基酸序列对不同地区病毒进行系统进化分析。RT-PCR结果显示,65份样品检出病毒,检出率为91.55%,其中,56份样品检出芜菁花叶病毒Turnip mosaic virus(TuMV),49份样品检出蚕豆萎蔫病毒2号Broad bean wilt virus 2(BBWV2),9份样品检测出黄瓜花叶病毒Cucumber mosaic virus(CMV),检出率分别为78.87%、69.01%和12.68%。未检测到烟草花叶病毒Tobacco mosaic virus(TMV)。序列和系统发育进化分析显示,本研究获得的TuMV、BBWV2和CMV与NCBI数据库中的序列相似度较高,核苷酸相似度分别为98.05%~98.98%、93.61%~94.25%和98.02%~99.17%,氨基酸序列相似度分别为:96.14%~97.47%、97.33%~98.50%和98.00%~100%,分别属于World-B组、Ⅰ-a亚... 相似文献
