The influence of a low and a high prepartum calcium (Ca) intake on Ca mobilization rate around parturition was studied in 44 dairy cows fed a ration sufficient for 1.90 times maintenance requirements during the dry period. The plasma Ca level declined on the day of parturition in the group fed the low Ca intake (LCa: 26.4 g/d) as well as in the group fed the high Ca intake (HCa: 87.2 g/d). Plasma Ca levels of the HCa group were lower at parturition, and in this group 1 cow had milk fever after parturition. In the HCa group 6 cows had a plasma Ca level less than or equal to 2.0 mmol/l at parturition and/or 10 h post-partum (pp), versus 1 cow in the LCa group. Na2EDTA was intravenously infused at 10 h pp to induce hypocalcaemia to a level of plasma Ca not bound to EDTA of approximately 1.0 mmol/l. The LCa groups tended to require more Na2EDTA than the HCa groups, however the difference was significant only in the younger cows. After the Na2EDTA infusion 7 cows of the HCa group and 1 of the LCa group did not recover spontaneously and needed to be treated. The mean plasma PTH levels of the LCa group ante-partum were slightly higher than those of the HCa group. The Ca level of the prepartum ration did not influence urinary hydroxyproline excretion, which suggests that the Ca intake of 26.4 g/d was too high to stimulate bone turnover. Comparison of the present results with those of an earlier experiment in which the prepartum Ca intake at a low feeding level (1.12 times maintenance) was studied, led to the conclusion that higher prepartum feed intake has a clear negative influence on Ca homeostasis around parturition.  相似文献

Six aged Merino ewes were used in an experiment in which five were infused with 4.7% Na2EDTA solution intravenously for 18 h at a rate designed to produce hypocalcaemia and maintain recumbency, and five with 0.9% sodium chloride solution at the same rate for the same period (four were infused at different times with both solutions). Blood samples were collected every 3 h and determinations made of plasma Ca, Na, K, Mg, and inorganic P (PiP), erythrocyte Na, K and Mg, and PCV. Three of the hypocalcaemic sheep took 36-64 h to regain their feet. Plasma Ca and K, and erythrocyte Na showed significant (all P less than 0.01) decreases in the group infused with Na2EDTA compared with the group infused with saline while PCVs were significantly (P less than 0.01) greater in the former group. The sheep model used could be suitable for the study of the effects of prolonged hypocalcaemia and recumbency in cows.  相似文献

The intravenous Na2EDTA infusion technique allows effective specific chelation of circulating Ca2+ leading to a progressive hypocalcaemia. Methods previously used were not described in detail and results obtained by monitoring total and free ionic calcium were not comparable due to differences in sampling and analysis. This paper describes a standardized EDTA infusion technique that allowed comparison of the response of calcium, phosphorus and magnesium between 2 groups of experimental cows. The concentration of the Na2EDTA solution was 0.134 mol/l and the flow rate was standardized at 1.2 ml/kg per hour. Involuntary recumbency occurred when ionised calcium dropped to 0.39-0.52 mmol/l due to chelation. An initial fast drop of ionized calcium was observed during the first 20 min of infusion followed by a fluctuation leading to a further drop until recumbency. Pre-infusion [Ca2+] between tests does not correlate with the amount of EDTA required to induce involuntary recumbence. Total calcium concentration measured by atomic absorption remained almost constant during the first 100 min of infusion but declined gradually when the infusion was prolonged. The concentration of inorganic phosphate declined gradually in a fluctuating manner until recumbency. Magnesium concentration remained constant during infusion. Such electrolyte responses during infusion were comparable to those in spontaneous milk fever. The standardized infusion technique might be useful in future experimental studies.  相似文献

Eight pregnant heifers (primiparous cows) and seven pregnant cows in their second, third and forth pregnancies (multiparous cows) were assigned to two groups and fed either a low calcium (Ca) diet (Ca, 0.46%) or a high Ca diet (Ca, 0.86%) ad libitum from 3 weeks before the expected calving date to 3 days after parturition. All cows were examined for a change in dry matter intake (DMI), plasma minerals and bone turnover around parturition. The dietary Ca level did not affect the DMI in both primiparous and multiparous cows. The DMI of primiparous cows was significantly lower than that of multiparous cows (P < 0.05) in both the low and high dietary Ca groups. The dietary Ca level did not affect the concentrations of plasma Ca, phosphorus, magnesium and parathyroid hormone throughout the experimental period. Plasma phosphorus in primiparous cows was significantly higher (P < 0.05) than that of multiparous cows around parturition. Plasma Ca and magnesium tended to be higher (P < 0.10) in primiparous cows. The dietary Ca level did not affect the plasma osteocalcin (OC) level measured as bone formation or the urinary deoxypyridinoline (DPD) excretion measured as bone resorption before parturition in both primiparous and multiparous cows. After parturition, the plasma OC level was lower than it was before parturition in multiparous cows fed a low Ca diet, and in primiparous cows. There were no significant differences in urinary DPD excretion between each group before and after parturition. Both the plasma OC level and urinary DPD excretion of primiparous cows were significantly higher than those of multiparous cows in both the low and high dietary Ca groups.  相似文献

The bone activity and bone mineral content in rib bones resected from 33 dairy cows between 3 and 8 h after parturition were measured, and the effects upon them of a deficient supply of dietary magnesium (Mg) during the last seven weeks of pregnancy were studied. The cows were fed a diet containing either 0.22% magnesium (low Mg) or 0.82% magnesium (high Mg) in the dry matter (DM), and the potassium content of both rations was increased to approximately 4.1% in the DM to reduce the absorption of magnesium. In the cows fed the low-Mg diet a fall in plasma Mg concentration was observed. In the low-Mg, low-parity cows the plasma Mg concentrations at parturition were higher than in the low-Mg, high-parity cows, i.e. 0.83 mmol/l and 0.54 mmol/l, respectively. After parturition four cows in the low-Mg, high-parity group showed clinical signs of hypocalcaemia but none of the other groups did so. The bone formation in low-parity cows was significantly (P less than 0.05) affected by Mg supply, with higher percentages of both trabecular surface covered by osteoid and osteoid volume in the low-Mg group. In the high-parity cows no significant differences in bone formation were found between the low- and high-Mg groups. An inadequate Mg supply resulted in a significantly (P less than 0.05) higher Ca content in the bone ash of low-parity cows and a significantly (P less than 0.05) higher bone ash percentage in the bone of high-parity cows.  相似文献

The hypothesis that hypocalcaemia may play a role in dystocia in captive Asian elephants (Elephas maximus) was investigated. The objectives of the study were to measure the total calcium concentration in elephant plasma; assess the changes in parameters of calcium metabolism during a feeding trial; investigate a possible relationship between calcium metabolism and dystocia; and assess bone mineralisation in captive Asian elephants in vivo. The following parameters were measured: total and ionised calcium, inorganic phosphorous and magnesium, the fractional excretions of these minerals, intact parathyroid hormone, 25-OH-D(3) and 1,25-OH-D(3). Radiographs were taken from tail vertebrae for assessment of bone mineralisation. The mean (sd) heparinised plasma total calcium concentration was 2.7 (0.33) mmol/l (n=43) ranging from 0.84 to 3.08 mmol/l in 11 Asian elephants. There was no significant correlation between plasma total calcium concentration and age. Following feeding of a calcium rich ration to four captive Asian elephant cows, plasma total and ionised calcium peaked at 3.6 (0.24) mmol/l (range 3.4 to 3.9 mmol/l) and 1.25 (0.07) mmol/l (range 1.17 to 1.32 mmol/l), respectively. Plasma ionised calcium concentrations around parturition in four Asian elephant cows ranged from 0.37 to 1.1 mmol/l only. The present study indicates that captive Asian elephants might be hypocalcaemic, and that, in captive Asian elephants, the normal plasma concentration of total calcium should actually be around 3.6 mmol/l and normal plasma concentration of ionised calcium around 1.25 mmol/l. Given the fact that elephants absorb dietary calcium mainly from the intestine, it could be concluded that elephants should be fed calcium-rich diets at all times, and particularly around parturition. In addition, normal values for ionised calcium in captive Asian elephants should be reassessed.

The hypocalcaemia caused by parturition and onset of lactation in high-production dairy cows was mimicked by subjecting cows to starvation periods before and after partus. The changes in plasma calcium, phosphate and magnesium were followed and compared with immunoreactive parathyroid hormone (iPTH) in 2 cows. During the starvation periods before partus, the cows developed hypocalcaemia with no or only small changes in the plasma concentration of magnesium. After the onset of hypocalcaemia, the concentration of iPTH increased on the average 3–4-fold and the raised hormone levels lasted about 24 h after start of refeeding. An increase in plasma phosphate occurred somewhat later than the rise in iPTH and lasted longer. After partus hypocalcaemia developed, together with smaller increases in iPTH concentration (about 2-fold). The post-partum starvation period again resulted in hypocalcaemia and raised iPTH concentrations. In conclusion, starvation and parturition induced inverse changes in plasma calcium and iPTH in dairy cows. The increases in plasma iPTH were reversible and considered secondary to the hypocalcaemia. Through the effect of paratyroid hormone, plasma calcium was normalized and phosphate concentration increased. Therefore, fatal hypocalcaemia which may occur following the course of parturition and onset of lactation is not due to impaired PTH secretion.  相似文献

The objective of this experiment was to induce acidification by anion salt supplementation for 2 days or 10 days and to study the prophylactic effects of such supplementation in preventing hypocalcaemia in cows. It was further attempted to monitor the extent to which any effect on the calcium-regulating mechanisms would persist following a 10-day period of acidification with anion salts. Study animals were three untreated control cows and three cows supplemented with ammonium chloride and ammonium sulphate in their ration for 2 days or 10 days through the rumen cannula. The basic ration of hay was dominated by Urochloa spp. The pH of the urine of the control cows was around 8.00 throughout the experiment and was considered normal. Anion-supplemented cows produced urine with a daily mean pH between 5.5 and 7.0, possibly due to anion salt exposure. The ability to withstand hypocalcaemic challenges was tested by a standardized intravenous infusion of disodium ethylenediaminetetraacetate (Na2EDTA). The calcium regaining time (CRT), expressed as time spent to reach 1.00 mmol/l of ionized calcium during recovery from the EDTA-induced hypocalcaemia, was used to compare cow responses. In the control cows the unexpectedly short CRT, especially during the weekly EDTA tests, could be a result of the repeated induced episodes of hypocalcaemia caused by the EDTA infusions. The improved CRT in the anion-supplemented cows may thus be interpreted as the combined effect of the repeated hypocalcaemic episodes due to EDTA infusions and probably the effect of anion-induced metabolic acidosis on endocrine-regulated calcium homeostatic mechanisms. The effect of anion salt exposure for 10 days on the improvement of calcium-regulating mechanisms was not clear due to the unexpected improvement of CRT that was exhibited by the untreated control cows as well. An on-farm trial of the effect of a 2-day or 10-day anion exposure of dry cows on calcium-regulating mechanisms is suggested.  相似文献

Six HK (high erythrocyte potassium) and 7 LK (low erythrocyte potassium) dairy cows were subjected to a 4-h intravenous infusion of 4.7% Na2EDTA solution to induce and maintain hypocalcaemia. Blood samples taken immediately before infusion, hourly for 7 h, and at 24 h after commencement of infusion were subjected to determination of concentration (or count) of 16 analytes. The mean changes in concentrations (or counts) of the various blood analytes were calculated for the periods 0-4, 4-7, 7-24, and 0-24 h after commencement of the infusion for all cows combined, and then separately for the HK and LK groups of cows. Plasma Ca(PCa), plasma inorganic phosphorus (PiP) and plasma potassium (PK) showed significant decreases during the 4-h infusion period and were still below pretreatment levels 24 h later. AST, CPK, PCVs and white cell-counts (WCCs) showed significant early increases which were still significantly elevated 24 h later. Plasma magnesium (PMg) and erythrocyte Na(ENa) and K(EK) all showed delayed changes which still persisted 24 h later. Significant between-group differences were present for PCVs which increased significantly more in the LK than the HK group during the infusion period, for PCa which showed a greater increase in the HK cows than the LK cows during the 4-7 h early clinical recovery period, and for plasma bilirubin (PBil) which showed a greater increase from 0 to 24 h in the HK group than in the LK group. Urine samples, collected before infusion, 4-7 h and 24 h after commencement of the infusion, were subjected to analysis for glucose, protein, pH, 'blood' and ketones. Most cows showed increases in urinary glucose, protein and 'blood'.  相似文献

The effects of induced subclinical hypocalcaemia (SCHC) on feed intake and chewing activity during eating and rumination were studied in dairy cows. Two non-lactating and non-pregnant cows were subjected to three different treatments, with one test per day, such that the plasma free (ionized) calcium (FCa) concentration was maintained at the eucalcaemic level or at one of two constant SCHC levels. The cows and test days followed a 2×3 crossover design. SCHC was maintained for 7 h by repeatedly infusing 5% Na2EDTA so that constant FCa concentrations of 0.8 mmol/L or 0.6 mmol/L in plasma were achieved. Control conditions were achieved by infusing isotonic saline. Feed intake and the number of the rumination periods were recorded during test days. The proportion of feed eaten during each test meal (EatPro) was related to the mean plasma FCa. An almost linear decrease in EatPro was observed when the plasma FCa was 0.6–0.9 mmol/L. The cows showed no other clinical signs of hypocalcaemia during Na2EDTA-infusion. The time spent chewing during eating and rumination, and the number of rumination periods during a test day, decreased with a decline in plasma FCa concentration. It was concluded that induced SCHC depresses the feed intake and ruminative activity of dairy cows.  相似文献

The influence of a deficient magnesium supply during the dry period on the ability of dairy cows to mobilise calcium at parturition was investigated. The daily rations pre-partum consisted of 6 kg grass hay, and 4 kg concentrates containing either 1.58 per cent magnesium (HMg) or 0.22 per cent magnesium (LMg). Nine cows of parity one to six received the LMg diet and 10 cows of parity one to seven the HMg diet. Both rations provided 1.40 times maintenance on an energy basis and contained 4 per cent potassium in the dry matter. Throughout the dry period the mean concentration of magnesium in the plasma of the LMg group was significantly lower than in the HMg group and at parturition the mean plasma concentrations were 0.65 mmol litre-1 and 1.16 mmol litre-1, respectively. After parturition two cows in the LMg group showed clinical signs of hypocalcaemia but none of the HMg group did so. Cows of the LMg group had a lower mean rate of calcium mobilisation (0.27 mmol min-1) than those of the HMg group (0.34 mmol min-1) but the difference could not be explained by differences in bone turnover as reflected by urinary hydroxyproline excretion. Such a reduced rate of calcium mobilisation may be a contributory factor in the high incidence of milk fever in some herds fed inadequate magnesium in the dry period.  相似文献

The objective of this experiment was to determine the effect of dietary grain on calcium homeostasis. Six rumen-fistulated dairy cows with 3 or more previous lactations and no history of parturient paresis were randomly assigned to a sequence of diets in a crossover study with 4 periods of 10 days each. Dietary treatments were: A control ration consisting of wrap grass silage alone (1), the control ration supplemented with ammonium chloride and ammonium sulphate salt solution (2), control ration following a period with supplementation (3) and control ration supplemented with increasing amounts of barley from 4 to 10 kg/cow per day, expected to produce subclinical rumen acidosis (4). Daily intake of the diets was adjusted to 14 kg DM/cow per day. On day 11, the calcium-regulating mechanisms in cows were challenged until recumbency by a standardized intravenous EDTA infusion and cows were left to recover spontaneously. Anion supplementation and the feeding of highly fermentable carbohydrate lowered urine pH below 7.0 due to subclinical acidosis. During spontaneous recovery from EDTA induced hypocalcaemia, the cows more quickly regained a whole blood free calcium concentration of 1.00 mmol/L if they had most recently been supplemented with either anionic salts or with increasing amounts of barley, as compared to the basic ration. It is concluded that so-called slug-feeding or 'steaming up' with highly fermentable carbohydrates before parturition in milk fever susceptible cows enhanced calcium homeostasis similar to the effect seen in cows on anionic diets.  相似文献

At the initiation of lactation, Ca homeostatic mechanisms have to react to a tremendous increase in demand for Ca. Mobilization of Ca from bone and increased absorption from the gastrointestinal tract are required to re-establish homeostasis. It has been shown that dietary anions play an important role in the prevention of milk fever by mobilizing Ca from bone and by increasing Ca absorption in the GI tract. The purpose of the present study was to investigate the influence of different Ca contents in diets supplemented with anionic salts on bone metabolism of dairy cows. Twenty-four holstein cows (housed inside, second to fourth lactation) without a milk fever history were divided into four groups (A, B, C, D). Each group was fed a different diet which was given from day 263 of gestation till the day of parturition. Group A and B received a low calcium diet (4 g/kg DM) whereas group C and D received a high Ca diet (8 g/kg DM). In addition group B and D received anionic salts. The DCAD was calculated with the formula: DCAD (mEq/kg DM)=(0.2 Ca2++0.16 Mg2++Na++K+)-(Cl-+0.6 S2-+0.65 P3-). Blood and urine samples were collected on days 256, 270 and 277 of gestation, on the day of parturition as well as the following 5 days and on days 9, 14 and 19 after parturition. Serum Ca, P, Mg, ICTP, OC, VITD, PTH and urinary pH were analysed. The bone resorption marker ICTP showed a significant increase after parturition in all the groups. On the contrary, the bone formation marker OC decreased after parturition in all the groups. The VITD concentrations in group D and the urinary pH in group B were significantly lower compared to the other groups (p<0.05). The Ca concentrations tended to be higher in group B around parturition than in all the other groups. No significant influence of the four different diets on all the other parameters could be shown. In conclusion, this data showed that the addition of anions and the different Ca contents had no significant influence on bone resorption and bone formation markers. This may be because of the fact that the dietary cation-anion balance was not low enough (DCAD-group A: 181 mEq/kg DM, group B: -48 mEq/kg DM, group C: 210 mEq/kg DM and group D: 28 mEq/kg DM) to induce a metabolic acidosis with all its positive effects on calcium metabolism.  相似文献

The rate of osteogenesis was studied in 8 non-pregnant, non-lactating Friesian dairy sheep, 3–6 years old, by means of a treatment with 3 different bone seeking agents. Four sheep were fed a low calcium ration (LCa : 1.8 g Ca/d) and four other sheep a high calcium ration (HCa : 12.7 g Ca/d). The bone markers, oxytetracycline-HCl, alizarine-complexon and demeclocycline-HCl, were administred at intervals of 6 weeks, and the sheep were killed 1 week after administration of the last marker. In undecalcified cross sections from the middle of ribs 2, 10 and 12, and from the proximal and distal parts of rib 10, the numbers of labelled osteons and the number of osteons with 1, 2 or 3 markers were counted under fluorescent microscopy. In the ribs of sheep from the LCa group, the number of labelled osteons and the quantity of labels per osteon tended to be higher than those of sheep from the HCa group. When osteogenic activity was compared in the different sites of ribs analysed, lowest osteogenic activity was observed in the proximal part of the 10th rib.The use of fluorescing markers offers the possibility of studying osteogenic activity over a certain period of time in adult sheep.  相似文献

To investigate the roles of mammary PTHrP in calcium uptake and/or release in the mammary gland of cows, plasma PTHrP and Ca levels, and their arterial-venous differences were examined in a Jersey cow during the periparturient period. Levels of Ca in both abdominal aorta and abdominal subcutaneous vein blood slightly decreased around the parturition and at 24 days after the parturition, however, no remarkable arterial-venous differences were observed. Plasma PTHrP levels in both arterial and venous samples were below the detection limit (0.57 pmol/l) during the experimental period. Milk PTHrP and Ca levels were measured in 9 Holstein dairy cows. Although plasma PTHrP levels in all arterial and venous samples were also below the detection limit, milk PTHrP and Ca levels were remarkably high, ranging from 14,900 pmol/l to 41,200 pmol/l and from 772 mg/l to 1,200 mg/l, respectively. In addition, a significant positive correlation (P<0.01) was observed between milk PTHrP and Ca levels. These results suggested that mammary PTHrP is closely related to Ca concentration in the milk.  相似文献

Forty-eight Friesian or Friesian-X factory supply dairy cows were divided into two groups. Group 1 received a supplement of sodiumtripolyphosphate (TPP, 25g P, 25g Na/cow/day), and group 2 a supplement of sodium chloride (25g Na/cow/day). Supplementation began at peak lactation, when the mean serum inorganic phosphorus (Pi) of all cows was 1.13 mmol/l. After four weeks, group 2 changed from NaCl to dicalcium phosphate supplementation (25g P/cow/day). Serum Pi and yields of milk, butterfat and protein were measured before, during and after supplementation. Pasture availability was assessed and P and Ca contents in pasture and the Pi content in milk were also determined. Supplementation raised serum Pi from 1.30 mmol/l (NaCI) to 1.42 mmol/l (TPP, P<0.05) but when dicalcium phosphate replaced NaCl the difference between groups disappeared (P>0.05). P supplementation had no effect on any milk parameter. Pre-grazing pasture mass above estimated grazing height averaged 2260 kg DM and contained >or=0.39 per cent P. It is concluded that a herd mean serum Pi concentration of around 1.2-1.3 mmol/l imposes no limitation to dairy production around the period of peak lactation of grazing dairy cattle.  相似文献

AIM: To determine differences in plasma proteomic profiles between healthy cows and those with subclinical hypocalcaemia within 12 hours after calving, and thereby explore the underlying biological mechanism of subclinical hypocalcaemia in dairy cows.

METHODS: Plasma samples were collected within 6 hours of calving from Holstein cows on a farm in Heilongjiang, China; 32 with subclinical hypocalcaemia (plasma calcium concentration 1.38–2.00?mmol/L and no clinical signs) and 59 control cows (plasma calcium concentration 2.10–2.8?mmol/L). Plasma samples were applied to weak cationic exchange protein chips for protein profiling by surface-enhanced laser desorption/ionisation time-of-flight mass spectrometry (SELDI-TOF-MS), and the data were analysed using the PBS-IIC system. The amplitude of peaks for the two groups were compared using the Wilcoxon sum-rank test, and the mass-to-charge ratio of the peaks that differed was used to identify peptide fragments using the Swiss-Prot protein database.

RESULTS: Seven peaks were identified in the subclinical hypocalcaemia group that differed from those of the control group (p<0.001), that represented six unique proteins. Expression of serum albumin, fibrinogen alpha chain, amyloid beta A4 proteins and neurosecretory protein VGF were increased, and expression of apolipoprotein A-II and serum amyloid A proteins were decreased in the subclinical hypocalcaemic cows compared with control cows.

CONCLUSION: Use of SELDI-TOF-MS technology can effectively identify differences in plasma protein expression patterns in cows with subclinical hypocalcaemia. Neurosecretory protein VGF and amyloid beta A4 protein might represent useful biomarkers for diagnosis of subclinical hypocalcaemia.  相似文献

Abstract

Pre-grazing pasture mass above estimated grazing height averaged 2260 kg DM and contained ?0.39 per cent P. It is concluded that a herd mean serum Pi concentration of around 1.2–1.3 mmol/l imposes no limitation to dairy production around the period of peak lactation of grazing dairy cattle.  相似文献

Serum inorganic phosphorus (Pi) and calcium (Ca) concentrations were assessed in 20 cows on each of four Manawatu factory supply dairy farms. Blood was taken from each cow before calving and at six-week intervals during lactation. Bleeding coincided with herd testing. Herds of Friesian or Friesian X cows and Jersey or Jersey X cows were compared on adjacent farms on a Central Yellow-brown Sand and on adjacent farms on a Peat Loam overlying a Central Yellow-brown Earth soil. Pasture mass and composition were estimated to grazing height in the next two paddocks to be grazed in the rotation. Mean serum Pi concentration was higher in cows on sandy soils (1.55 mmol Pi/l than in cows on the peat loam (1.34 mmol Pi/l (P<0.001). Concentrations were highest before calving (1.69 mmol Pi/l) but fell to low levels at peak lactation (1.17mmol Pi/l when 70% of cows were below the minima of the 'normal range', and during the drought (1.29 mmol Pi/l. Pasture phosphorus (P) concentrations were adequate to support cow nutrition for lactation (>0.33% DM, ad lib. feeding) until the summer drought when low herbage mass would have restricted milk production. Serum Ca was adequate for lactating cows and changed little between months or between cows (mean 2.12 mmol Ca/l). No metabolic disorders relating to mineral deficiencies were observed. It appears that serum Pi in a high proportion of cows falls below the normal range during peak lactation without cows displaying clinical deficiency symptoms or a depression in butterfat production.  相似文献
