The hypothalamic-pituitary-adrenal (HPA) axis exerts a large range of effects on metabolism, the immune system, inflammatory processes, and brain functions. Together with the sympathetic nervous system, it is also the most important stress-responsive neuroendocrine system. Both systems influence production traits, carcass composition, and meat quality. The HPA axis may be a critical target for genetic selection of more robust animals. Indeed, numerous studies in various species have demonstrated the importance of genetic factors in shaping the individual HPA axis phenotype, and genetic polymorphism can be found at each level of the axis, including hormone production by the adrenal cortices under stimulation by adrenocorticotropic hormone (ACTH), hormone bioavailability, or receptor and postreceptor mechanisms. The aim of the present experiment was to extend these findings to the brain neurochemical systems involved in stress responses. To this end, a number of candidate genes were sequenced for molecular polymorphisms and their association was studied with stress neuroendocrine and production traits in a genetically diverse population consisting of 100 female pigs from an advanced intercross (F10-F12) between 2 highly divergent breeds, Large White (LW) and Meishan (MS). The LW breed has a high production potential for lean meat and a low HPA axis activity, and the MS breed has low growth rate, fat carcasses—but large litters of highly viable piglets—and a high HPA axis activity. Candidate genes were chosen in the catecholaminergic and serotonergic pathways, in the pituitary control of cortisol production, among genes previously demonstrated to be differentially expressed in ACTH-stimulated adrenal glands from LW and MS pigs, and in cortisol receptors. Sixty new polymorphisms were found. The association study with carcass and meat quality traits and with endocrine traits showed a number of significant results, such as monoamine oxidase (MAOA) polymorphisms with growth rate (P = 0.01); lean content and intramuscular fat (P < 0.01), which are the most important traits for carcass value; dopamine receptor D3 (DRD3) with carcass composition (P < 0.05); and vasopressin receptor 1B (AVPR1B) with meat quality traits (P ≤ 0.05). The effect of these polymorphisms on neuroendocrine parameters (eg DRD3 and HPA axis or AVPR1B and catecholamines) indicates information regarding their biological mechanism of action.  相似文献

The possible involvement of the pituitary-adrenocortical axis with severe pre- and post partum udder oedema was studied in a herd of 60 Holstein/Friesian cows. The prevalence of udder oedema in this herd was about 75 per cent. The animals were fed a diet containing high levels of potassium (average daily potassium intake 410 g per cow). The concentrations of sodium, potassium, chloride and total protein in the plasma of the cows were within their normal ranges. In one animal a very low plasma cortisol concentration (2.30 ng/ml) was accompanied by normal plasma glucose and beta-hydroxybutyrate concentrations. The adrenal responsiveness of two heifers with severe udder oedema was impaired one month after parturition in comparison with 11 normal animals (increases from 3.27 to 38.8 and 23.2 to 37.8 ng cortisol/ml compared with 8.02 +/- 1.19 to 146 +/- 16.7 ng cortisol/ml but returned to normal spontaneously after 175 days. One cow was diagnosed as having primary adrenocortical insufficiency.  相似文献

The objective of this study was to estimate breed-specific genetic correlations between lean growth and litter traits for four U.S. swine breeds. Records for lean growth and litter traits on Yorkshire, Duroc, Hampshire, and Landrace pigs collected between 1990 and April 2000 in herds on the National Swine Registry Swine Testing and Genetic Evaluation System were analyzed. A bivariate animal model and restricted maximum likelihood procedures were used to estimate genetic and environmental correlations between lean growth rate, days to 113.5 kg, backfat, and loin muscle area with litter traits of number born alive, litter weight at 21 d, and number weaned. Most genetic correlation estimates between lean growth and litter traits were small in magnitude and consistent across breeds. Backfat had the largest within-breed genetic correlations with number born alive (0.18 to 0.20) and litter weight at 21 d (-0.27 to -0.30). Estimates of genetic correlations between lean growth traits and number weaned were very small. Estimates of the environmental correlations between lean growth and litter traits also were very small for all traits and for all four breeds. Results indicate that selection for lean growth traits could have a long-term effect on litter traits. Including lean growth traits in a maternal-line evaluation using a multiple-trait model could increase the accuracy of the genetic evaluation for litter traits.  相似文献

Interest in selection for improved feed efficiency is increasing, but before any steps are taken toward selecting for feed efficiency, correlations with other economically important traits must first be quantified. The objective of this study was to quantify the genetic associations between feed efficiency measured during performance testing and linear type traits, BW, live animal value, and carcass traits recorded in commercial herds. Feed efficiency data were available on 2,605 bulls from 1 performance test station. There were between 10,384 and 93,442 performance records on type traits, BW, animal value, or carcass traits from 17,225 commercial herds. (Co)variance components were estimated using linear mixed animal models. Genetic correlations between the muscular type traits in commercial animals and feed conversion ratio (-0.33 to -0.25), residual feed intake (RFI; -0.33 to -0.22), and residual BW gain (RG; 0.24 to 0.27) suggest that selection for improved feed efficiency should increase muscling. This is further evidenced by the genetic correlations between carcass conformation of commercial animals and feed conversion ratio (-0.46), RFI (-0.37), and residual BW gain (0.35) measured in performance-tested animals. Furthermore, the genetic correlations between RFI and both ultrasonic fat depth and carcass fat score (0.39 and 0.33, respectively) indicated that selection for improved RFI will result in leaner animals. It can be concluded from the genetic correlations estimated in this study that selection for feed efficiency will have no unfavorable effects on the performance traits measured in this study and will actually lead to an improvement in performance for some traits, such as muscularity, animal price, and carcass conformation. Conversely, this suggests that genetic selection for traits such as carcass quality, muscling traits, and animal value might also be indirectly selecting for more efficient animals.  相似文献

Correlated responses in physiological traits in lines of Texel-Oxford sheep selected for high or lowcarcass lean content were examined. Serum samples were taken at the end of performance test, 20 weeks of age, from 66 rams when fed ad libitum and every 24 h when fasted for 56 h. The lean line had higher serum concentrations of β-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA) and insulin-like growth factor- 1 (IGF-1) with lower concentrations of triglyceride (TRIG), creatinine (CREA) and UREA, but only the differences in UREA were statistically significant. There were substantial changes in serum concentrations of physiological traits in response to fasting as glucose and IGF-1 decreased while BHB, NEFA, TRIG, CREA and UREA increased. The correlated responses suggested that the lean line preferentially synthesises protein rather than deposits fat during normal feeding. When the animals are fasted, there may be relatively greater use of fat as an energy source in the lean line, rather than using products from protein catabolism as glucose precursors. The accuracy of selection was examined when physiological traits were incorporated into a selectionindex, which included the performance test traits: liveweight, ultrasonic backfat and muscle depths, to predict genetic merit for carcass lean content. UREA may be a useful predictor of genetic merit for lean meat production as it was correlated with estimated carcass lean content and there were substantial differences between the selection lines.  相似文献

Associations between production traits and the genes for halothane sensitivity (HAL), S, A and H blood group systems and phosphohexose isomerase (PHI) and 6-phosphogluconate dehydrogenase (6-PGD) enzyme systems were investigated in two lines of pigs selected for an index. The phenotypic variance-covariance matrix of the index included backfat thickness and daily gain, whereas the genetic variance-covariance matrix included daily gain, feed conversion and percentage of lean meat. The experiment was conducted at the experimental station of the Institute of Animal Production and has been underway since 1973. The same index was applied but in two opposite directions to give a superior and inferior line in relation to the production traits. One hundred twenty-nine animals of the superior line in the seventh generation and 88 animals of the inferior line in the sixth generation were studied. Forty-two percent (54/129) of the animals of the superior line were halothane-positive. No animals in the inferior line were halothane reactors. Of the halothane-positive pigs, 70.4% (38/54) in the superior line had the HaHa and 94.4% (51/54) had the SsSs genotype, whereas only 4% (3/75) of the HaHa and 12% (9/75) of the SsSs pigs were halothane-negative. By practicing selection at the H and S loci, it seems possible to efficiently reduce halothane sensitivity in Swiss Landrace pigs. In pigs of the superior line, there were significant differences in percentage of lean meat, carcass length, pH1 (pH value at 45 min to 1 h postmortem, M. longissimus) and reflectance values among genotypes of the HAL, S and H systems and among some genotypes of the 6-PGD system. Poorest meat quality, highest percentage of lean meat and shortest carcass length were observed in pigs homozygous for the alleles HALn, Ss, Ha, PHIB and 6-PGDA. In the inferior line, these associations were absent. As the HAL locus is associated with the above mentioned production traits, linkage disequilibria may explain the observed associations among the H, S, PHI and 6-PGD types and production traits.  相似文献

Long-term selection on production results in increased environmental sensitivity. This often is expressed through decreased fertility and increased health problems. The phenomenon has been described in all common farm animal species. One theory is that potential resource intake is insufficient to express production potential. Additional resources are drawn away from fitness-related traits, such as fertility and health, to further increase observed production. In addition, resources for maintaining fitness depend on the demands by the environment. In a harsh environment, more resources are required for fitness-related traits than in an optimal environment. Literature results show that selection in an optimal environment will increase sensitivity to less optimal environments. The objectives of this paper were to increase understanding of the underlying mechanism behind the development of environmental sensitivity and to gain insight into correlated response(s) when selection is on observed production. A resource allocation model was defined where observed production depended on production potential, resource intake potential, and the allocation of resources to production or fitness, including maintenance, health, and reproduction. Penalties for reproductive performance and probability of survival were included when the proportion of resources assigned to fitness dropped below a certain, environment-related, threshold. Mass selection was practiced on observed production during 40 generations using stochastic simulation. Depending on the heritabilities of the underlying components and on the environment, selection on observed production resulted in a decrease in reproductive rate and in the development of environmental sensitivity when resource intake becomes limiting. Correlations of observed production with underlying components changed across generations, following a nonlinear pattern. The proposed model is simple, but increases the understanding of underlying mechanisms and consequences of selection for production when resources are limiting.  相似文献

Selection is the major tool used by breeders to improve the genetic quality of their livestock. Traditional methods of selection are well proven and useful in improving the economic merit of livestock. The performance of an animal is affected by its genetic quality and by the environment in which it is reared. While environmental improvement is expensive and requires continuous inputs, genetic improvement is cumulative and permanent, provided that selection is maintained. To select an animal on its genetic merit account must be taken of the environmental effects on its performance. Comparisons between the performance of animals on different farms or in different years are not valid unless they have genetic material in common. The speed at which genetic improvement is passed on to the rest of a population is affected by the variation and heritability of the traits being selected, the selection intensity and the generation interval. The deer population in the United Kingdom has a high degree of variation for important traits but the selection intensity is low and the generation intervals are larger than in other farmed species. Central performance testing, group breeding schemes and the use of artificial insemination are tools which will be important in the genetic improvement of farmed deer.  相似文献

Variance components for production traits were estimated using different models to evaluate maternal effects. Data analysed were records from the South African pig performance testing scheme on 22 224 pigs from 18 herds, tested between 1990 and 2008. The traits analysed were backfat thickness (BFAT), test period weight gain (TPG), lifetime weight gain (LTG), test period feed conversion ratio (FCR) and age at slaughter (AGES). Data analyses were performed by REML procedures in ASREML, where random effects were successively fitted into animal and sire models to produce different models. The first animal model had one random effect, the direct genetic effects, while the additional random effects were maternal genetic and maternal permanent environmental effects. In the sire model, the random effects fitted were sire and maternal grand sire effects. The best model considered the covariance between direct and maternal genetic effects or between sire and maternal grand sire effects. Fitting maternal genetic effects into the animal model reduced total additive variance, while the total additive variance increased when maternal grand sire effects were fitted into the sire model. The correlations between direct and maternal genetic effects were all negative, indicating antagonism between these effects, hence the need to consider both effects in selection programmes. Direct genetic correlations were higher than other correlations, except for maternal genetic correlations of FCR with TPG, LTG and AGES. There has been direct genetic improvement and almost constant maternal ability in production traits as shown by trends for estimated (EBVs) and maternal breeding values (MBVs), while phenotypic trends were similar to those for EBVs. These results suggest that maternal genetic effects should be included in selection programmes for these production traits. Therefore, the animal–maternal model may be the most appropriate model to use when estimating genetic parameters for production traits in this population.  相似文献

Effects of social isolation or restraint, applied outside the home pen, on adrenocortical and nociceptive responses were examined in 28 loose-housed dairy cows. Treatments lasted 15 min and consisted of social isolation in novel surroundings or restraint by the head in a test pen. A control treatment was applied in the test pen as well. Each cow was exposed to all treatments in a balanced order, with 3 to 4 d between treatments. Compared with the control treatment, social isolation in novel surroundings led to increased plasma concentration of cortisol (P < 0.001) as well as to indications of hypoalgesia [posttreatment lack of decrease in latency to respond toward nociceptive laser stimulation, a tendency for decreased frequency of kicking in the pauses between laser stimulations (P = 0.06), and an increased proportion of leg moving (least possible active response) after treatment (P = 0.04)]. Indications of hypoalgesia were also observed after restraint (reduced kicking in response to laser stimulation, P = 0.04); however, the indications were to a lesser extent than after social isolation, and restraint treatment did not lead to increased plasma concentration of cortisol. For control and restraint treatment, an initial increase (P < 0.02) in plasma concentration of cortisol was found, suggesting effects of pretreatment factors such as handling. No correlations between adrenocortical and nociceptive responses toward social isolation were found. The results confirm earlier reports stating that nociceptive changes induced by environmental challenges can be shown in dairy cows, even when they are kept in groups and removed from the home pen during the study of stress responses. However, testing outside the home pen seemed to affect the nociceptive and adrenocortical responses, thereby suggesting that care should be taken to avoid effects of pretreatment situational factors.  相似文献

Genetic Improvement of Dairy Cow Reproductive Performance   总被引:1,自引:0,他引:1
The welfare of cow along with profitability in production are important issues in sustainable animal breeding programmes. Along with an intense/intensive selection for increased milk yield, reproductive performance has declined in many countries, in part due to an unfavourable genetic relationship. The largely unchanged genetic trend in female fertility and calving traits for Scandinavian Red breeds shows that it is possible to avoid deterioration in these traits if they are properly considered in the breeding programme. Today's breeding is international with a global selection and extensive use of the best bulls. The Nordic countries have traditionally recorded and performed genetic evaluation for a broad range of functional traits including reproduction. In recent years many other countries have also implemented genetic evaluation for these traits. Thus, the relative emphasis of dairy cattle breeding objectives has gradually shifted from production to functional traits such as reproduction. Improved ways of recording traits, e.g. physiological measures, early indicator traits, assisted reproductive techniques and increased knowledge of genes and their regulation may improve the genetic selection strategies and have large impact on present and future genetic evaluation programmes. Extensive data bases with phenotypic recordings of traits for individuals and their pedigree are a prerequisite. Quantitative trait loci have been associated to the reproductive complex. Most important traits, including reproduction traits are regulated by a multitude of genes and environmental factors in a complex relationship, however. Genomic selection might therefore be important in future breeding programmes. Information on single nucleotide polymorphism has already been introduced in the selection programmes of some countries.  相似文献

A study was conducted to determine whether within-breed differences in adrenocortical response to exogenous adrenocorticotropin (ACTH) might be accounted for by differences in responsiveness of the adrenocortical cells per se. Large White x Landrace male pigs (n = 20) were used; 10 had high adrenocortical response to ACTH administration and 10 had low response. Five high and 5 low responders were euthanatized at 15 weeks of age, and the remaining 5 high and 5 low responders were euthanatized at 21 weeks of age. Adrenal glands were removed and weighed, and adrenocortical cells were dispersed by tryptic digestion and incubated for 2 hours with synthetic ACTH at concentrations ranging from 0.5 to 10,000 pg/ml. Samples were taken at 30-minutes intervals, and cortisol concentration was determined by use of a radioimmunoassay. Results indicate that for pigs of both age groups, high responders had heavier adrenal glands, with higher adrenocortical cell density and higher cell yield than did low responders. Synthetic ACTH had a stimulatory effect on dispersed porcine adrenocortical cells, as indicated by changes in cortisol concentration in vitro. Adrenocortical cells from high responders produced less cortisol, on a per-cell basis, than did those from low responders. However, when corrected for total cell yield, the potential cortisol production by each pair of adrenal glands was significantly (P less than 0.05) higher in the high responders than in the low responders. Thus, high-responding pigs have larger adrenal glands and higher adrenocortical cell density, which may result in higher output of cortisol after ACTH administration or exposure to stressors.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Genetic factors are undoubtedly involved in inter-individual variability of the behaviours that may be important for livestock production, as shown by pedigree studies, comparison of genetic stocks raised in the same environment, and selection experiments. The knowledge of gene polymorphisms responsible for genetic variability would increase the efficiency of selection, as shown for instance by the identification of the ryanodine receptor gene that harbours the mutations responsible for the porcine stress syndrome, that allows the eradication of the susceptibility allele. One strategy is to screen systematically the genes that are known to be involved in regulation of behaviour (functional candidate genes). This strategy is however very difficult for most behavioural traits, since behaviour is an emerging function from the whole brain/body and the molecular pathways involved in genetic variability are very poorly understood. Another strategy is to investigate linkage between trait variation and genetic markers in a segregating population (usually an intercross or backcross between two strains or breeds contrasting for the trait under study). It allows the detection of genomic regions influencing that trait (quantitative trait loci or QTL), and further investigation aims at the identification of the gene(s) located in each of these regions and the molecular polymorphisms involved in phenotypic variation. Although many QTL have been published for behavioural traits in experimental animals, very few examples are available where strong candidate genes have been identified. Further progress will be very much dependent upon the careful definition of behavioural traits to be studied (including their importance for animal production), on the reliability of their measurement in a large number of animals and on the efficient mastering of environmental factors of variability. The fast increase in the knowledge of genome sequence in several species will undoubtedly facilitate the application to farm animal species of the knowledge obtained in model organisms, as well as the use of model organisms to explore candidate genes detected by QTL studies in farm animals.  相似文献

ABSTRACT

Small ruminants are particularly well suited to meet United Nations Sustainable Development Goals surrounding food security, human wellbeing and poverty alleviation in different environmental and climatic settings. However the current efficiency of food production from small ruminants in both developed agricultural regions and in lower and middle income countries is woefully inadequate to meet predicted global needs over the forthcoming decades. Most global research to address this challenge is focussed on the genetics of animal growth, conformation and disease tolerance or resistance traits, albeit the practical consequences of such selection and strategies to use genetically improved animals in the field are uncertain. Any long-term benefits derived from small ruminant genetic selection will only be impactful if steps are first taken to keep animals alive, healthy and productive through iterative planned health management. Parasites are the foremost global infectious disease constraints to efficient small ruminant production. Their genetic adaptability to exploit opportunities afforded by effects of climatic or management changes on free-living stages, or exposure of parasitic stages to drugs, presents specific challenges to their sustainable control. Hence, parasite control provides a relevant means of engagement with livestock keepers and farmers on the topic of planned animal health management. The value of parasitology in this regard is enhanced by the availability of simple to use and accessible diagnostic tools.  相似文献

The aim of the present study was to estimate genetic parameters for flight speed and its association with growth traits in Nellore beef cattle. The flight speed (FS) of 7,402 yearling animals was measured, using a device composed of a pair of photoelectric cells. Time interval data (s) were converted to speed (m/s) and faster animals were regarded as more reactive. The growth traits analyzed were weaning weight (WW), ADG from weaning to yearling age, and yearling scrotal circumference (SC). The (co)variance components were estimated using REML in a multitrait analysis applying an animal model. The model included random direct additive genetic and residual effects, fixed effects of contemporary groups, age of dam (classes), and age of animal as covariable. For WW, the model also included maternal genetic and permanent environmental random effects. The direct heritability estimate for FS was 0.26 ± 0.05 and direct heritability estimates for WW, SC, and ADG were 0.30 ± 0.01, 0.48 ± 0.02, and 0.19 ± 0.01, respectively. Estimates of the genetic correlation between FS and the growth traits were -0.12 ± 0.07 (WW), -0.13 ± 0.08 (ADG), and -0.11 ± 0.07 (SC). Although the values were low, these correlations showed that animals with better temperaments (slower FS) tended to present better performance. It is possible to infer that longterm selection for weight and scrotal circumference can promote a positive genetic response in the temperament of animals. Nevertheless, to obtain faster genetic progress in temperament, it would be necessary to perform direct selection for such trait. Flight speed is an easily measured indicator of temperament and can be included as a selection criterion in breeding programs for Nellore cattle.  相似文献

Evaluation of strategies for selection for lean growth rate in pigs   总被引:1,自引:0,他引:1
Lean growth rate (LGR) in pigs is a nonlinear biological function of growth rate and lean quantity. According to animal breeding theory, genetic progress for LGR is maximized with selection on a linear index of its component traits, but selection on direct EBV for LGR is also common. In this study, the performance of five criteria for selection on estimated LGR in pigs was evaluated through simulation over five generations: linear indexes of multiple-trait EBV of component traits with or without updating index weights in each generation; a nonlinear index of multiple-trait EBV of component traits; and direct selection on EBV for LGR from a single-trait model or a multiple-trait model that included LGR and component traits. The nonlinear index yielded the highest response in LGR in Generation 5, but the linear index with updating performed almost as well. Not updating weights for the linear index reduced response in LGR by 1.1% in Generation 5 (P < 0.05). Direct selection on single-trait EBV for LGR yielded the lowest responses in Generation 5. Direct selection on EBV for LGR from a multiple-trait animal model yielded a 3.1% greater response in LGR in Generation 5 than direct selection on EBV for LGR based on a single-trait animal model (P < 0.05), but yielded a 1.9% lower response than the nonlinear index. Although differences in response in LGR were limited, alternative selection criteria resulted in substantially different responses in component traits. Linear index selection for LGR placed more emphasis on lean quantity, whereas direct selection for LGR emphasized growth rate. Based on the relative changes in the responses in LGR, selection for estimated LGR based on a nonlinear index or a linear index with updating is recommended for use in the swine industry.  相似文献
