A new disease of peppers (Capsicum annuum) is described. The most characteristic symptom is vein yellowing of the youngest leaves. Some streaking occurs on the first-formed fruits. The symptoms were reproduced in pepper plants cv. Bellboy by graft and Olpidium transmission. Most pepper cultivars tested were susceptible but some appeared to he resistant. The disease had no effect on fruit weight.  相似文献

ABSTRACT Apple fruit tissues infected with Botryosphaeria dothidea were examined by transmission electron microscopy using susceptible cv. Fuji and resistant cv. Jonathan. Immature (green) and mature (red) fruits of cv. Fuji with restricted or expanding lesions were also examined to reveal subcellular characteristics related with latent and restricted disease development. In infected susceptible mature fruits, cytoplasmic degeneration and organelle disruption commonly occurred, accompanying cell wall dissolution around invading hyphae. Cell wall dissolution around invading hyphae in subepidermis was rare in immature, red halo-symptomed cv. Fuji and resistant cv. Jonathan fruits. In infected immature fruits of cv. Fuji, presumably at the latent state of disease development, cellular degeneration was less severe, and invading hyphae contained prominent microbody-lipid globule complexes or the deposition of thin electron-dense outer layer around cell wall of intercellular hyphae. Both mature fruits with red halos and resistant apple fruits formed cell wall protuberances at the outside of cell walls. In addition, electron-dense extramural layers were formed in the resistant apple fruits. Aberrant hyphal structures such as intrahyphal hyphae were found only in resistant fruit tissues, indicating the physiologically altered fungal growth. These ultrastructural changes of host tissues and fungal hyphae may reflect the pathogenesis of apple white rot under varying conditions of apple fruits.  相似文献

Disease severity of powdery mildew (causal agentLeveillula taurica (Lév.) Arn.) on pepper (Capsicum annuum), defoliation and fruit sunscald rating were significantly reduced (P=0.05) by foliar applications of sodium or potassium bicarbonate solutions, compared with water or penconazole (Ophir). Preharvest application of bicarbonate solutions significantly reduced (P=0.05) postharvest decay development on sweet red pepper fruits.  相似文献

Microscopical studies of fungal development and host responses during infection of the hop cultivars Northern Brewer (susceptible) and Wye Target (resistant) with a Zenith isolate of Sphaerotheca humuli are described. Resistance to powdery mildew disease in cv. Wye Target is principally determined by the R2 major gene. Fungal growth was typically restricted in cv. Wye Target following formation of a single haustorial initial or haustorium which failed to develop the characteristic lobes found in susceptible cells. The hypersensitive reaction of penetrated epidermal cells was associated with death of haustorial initials but the associated appressorium remained alive during the first 2 days after inoculation. In leaves expressing resistance, histochemical staining revealed deposition of lignin-like material and callose in penetrated cells and to a lesser extent in underlying palisade mesophyll cells. Transmission electron microscopy and enzymic digestion clearly demonstrated extensive paramural deposition of β-1.3 glucans (callose) in reacting mesophyll cells. Plant cell death, lignification and widespread callose deposition were rarely observed at infection sites in cv. Northern Brewer but collars of callose were deposited around the necks of all haustoria formed.  相似文献

The effects of plant age, fungus isolate, zoospore concentration and inoculation method on the resistance of pepper ( Capsicum annuum) to blight ( Phytophthora capsici) were studied under controlled conditions. Each of the factors studied produced differing responses in the pepper lines tested hut line CNPH 286 proved to be a reliable source of resistance and is now being used in a breeding programme. An inoculation method for screening pepper lines for blight resistance is proposed, using a standardized zoospore suspension applied to the base of plants 45 days from sowing.  相似文献

ABSTRACT In the pepper Capsicum annuum CM334, which is used by breeders as a source of resistance to Phytophthora spp. and potyviruses, a resistance gene entirely suppresses reproduction of the root-knot nematode (Meloidogyne spp.). The current study compared the histological responses of this resistant line and a susceptible cultivar to infection with the three most damaging root-knot nematodes: M. arenaria, M. incognita, or M. javanica. Resistance of CM334 to root-knot nematodes was associated with unidentified factors that limited nematode penetration and with post-penetration biochemical responses, including the hypersensitive response, which apparently blocked nematode migration and thereby prevented juvenile development and reproduction. High-performance liquid chromatography analysis suggested that phenolic compounds, especially chlorogenic acid, may be involved in CM334 resistance. The response to infection in the resistant line varied with root-knot nematode species and was correlated with nematode behavior and pathogenicity in the susceptible cultivar: nematode species that quickly reached the vascular cylinder and initiated feeding sites in the susceptible cultivar were quickly recognized in CM334 and stopped in the epidermis or cortex. After comparing our data with those from other resistant pepper lines, we suggest that timing of the resistance response and the mechanism of resistance vary with plant genotype, resistance gene, and root-knot nematode species.  相似文献

Using hydroponics and novel non‐destructive pot culture systems which enable inoculation at specific tuber development stages, the dynamics of common scab infection patterns in potato were studied in order to provide more precise identification of tuber physiological factors associated with susceptibility. At the whole‐tuber level, infection percentages were greatest when Streptomyces scabiei inoculation occurred early; at 2 weeks after tuberization (WAT) 68% of tubers became infected, contrasting with late inoculation (8 WAT), when only 4% infection occurred. The first‐formed internodes were most susceptible to infection, whilst later‐forming and slower‐expanding internodes were less susceptible. Detailed tuber physiological examination of internode 2 showed that pathogen‐induced changes, including increased phellem (periderm) thickness, cell layers and phellem suberization (key physiological features believed critical to S. scabiei infection) were promoted through S. scabiei inoculation. Sequential harvesting showed enhanced phellem suberization (28% greater than the control) within 7 days of pathogen exposure, while phellem thickness and layer responses were also initiated early in the infection process (10–14 days after pathogen exposure) and these responses were independent of symptom expression. Differences in cultivar response were observed, with greater phellem suberization observed 10 days after tuberization (DAT) in the common‐scab‐tolerant cv. Russet Burbank than in the susceptible cv. Desiree. Likewise, Russet Burbank had thicker and more numerous cell layers in the phellem (up to eight cell layers) during early tuber growth (20–30 DAT) than Desiree (up to six cell layers).  相似文献

研究发现叶、茎、茎基部和果实接种4种方法均能用于鉴定辣椒对辣椒疫霉的抗性。叶与茎接种法操作简单,接种部位发病快,可同时检测大量品种,但要求空气湿度大;茎基部接种法模拟田间病害的发生过程,最能反映品种的真实抗性,但需要菌量大,工作量也大;果实接种法不仅操作简单、接种部位发病快、菌需求量小,而且可同时筛选大量品种,可以通过量化达到有效区分抗病和感病品种的目的,更具有完整保存原有植株的优势。  相似文献

A severe rot of postharvest fruits of sweet pepper, a variety of Capsicum annuum, was found in Kagawa Prefecture in southwestern Japan in August 1999. A fungus, isolated repeatedly from the diseased fruits and identified as Stemphylium lycopersici, was demonstrated to be pathogenic to fruits of sweet pepper. The disease was new to Japan, and the fungus was added to the pathogens causing fruit rot of C. annuum.  相似文献

The arrangement of microtubules in soybean ( Glycine max ) cells was examined during compatible and incompatible interactions of hypocotyls of soybean cv. Harosoy (susceptible) and cv. Haro 1272 (resistant) with race 1 of the soybean-specific pathogen Phytophthora sojae . Both reaction types were similar during the first 3 h after zoospore inoculation in terms of the number of cells penetrated, and depth penetrated into the cortex. By 3 h postinoculation, clear differences had developed between the two interaction types: incompatible interactions were characterized by a hypersensitive response that was confined to single penetrated cells; while compatibly responding cells appeared unchanged. Both types of response were characterized by autofluorescence of cell walls or cytoplasm and, at 6 h after inoculation, complete disorganization of cell cytoplasm. Reorientation and loss of microtubules was seen in the early stages of the incompatible interaction in association with cellular hypersensitivity, but not in compatible responses. In cells adjacent to those that reacted hypersensitively, there was little evidence of change in microtubule orientation. Treatment of hypocotyls with the microtubule depolymerizer oryzalin prior to inoculation did not alter the compatible response, but led to breakdown of the incompatible response. Changes in microtubule orientation and state are thus among the first structural changes that are visible within cells during incompatibility in this system.  相似文献

Following inoculation of host and nonhost plants with Plasmopara viticola, the grapevine downy mildew, a histological survey was undertaken to identify the stage where its development is contained in nonhosts and in resistant host plants. Three herbaceous nonhost species, Beta vulgaris, Lactuca sativa, and Capsicum annuum, and three grapevine species displaying different level of resistance (Vitis vinifera [susceptible], Vitis riparia [partially resistant] and Muscadinia rotundifolia [totally resistant]) where inoculated by P. viticola using a controlled leaf disk inoculation bioassay. During the early steps of infection, defined as encystment of zoospores on stomata, penetration of the germ tube, and production of the vesicle with the primary hypha, there was no evidence of a clear-cut preference to grapevine tissues that could attest to host specificity. The main difference between host grapevine species and nonhosts was observed during the haustorium formation stage. In nonhost tissues, the infection was stopped by cell wall-associated defense responses before any mature haustorium could appear. Defense responses in resistant grapevines were triggered when haustoria were fully visible and corresponded to hypersensitive responses. These observations illustrate that, for P. viticola, haustorium formation is not only a key stage for the establishment of biotrophy but also for the host specificity and the recognition by grapevine resistance factors.  相似文献

The reproduction of a Meloidogyne hispanica isolate from Portugal was evaluated in 63 plant species/cultivars, in pot assays at 25?±?2.0°C, on the basis of root gall index (GI) and reproduction factor (Rf?=?final/initial egg density) at 60 days after inoculation. Cultivars of aubergine, bean, beetroot, broccoli, carnation, corn, cucumber, French garlic, lettuce, melon, onion, parsley, pea, potato, spinach, and tobacco and two of cabbage were susceptible (3?≤?GI?≤?5; 1.15?≤?Rf?≤?262.86). Cabbage cv. Bacalan, cauliflower cv. Temporão and pepper cv. Zafiro R2 were hypersusceptible or poor hosts (Rf??2) and pepper cvs. Aurelio and Solero were resistant (0.0?≤?GI?≤?0.4; 0.00?≤?Rf?≤?0.03). The response of the pepper cultivars and the Mi-1 resistant tomato cv. Rossol was also conducted in pots using two inoculum levels and four temperatures, three growth chamber (25?±?2.7°C, 29.3?±?1.8°C and 33.6?±?1.2°C) and one outdoors (24.4?±?8.2°C). At 24.4?±?8.2°C and 25?±?2.7°C, the reproduction on the resistant tomato was significantly lower compared to the susceptible cv. Easypeel. At all temperatures, resistance was evident for the pepper cultivars, despite the fact they were not found to contain any of the Me1, Me3, Me7 and N genes. The eggs obtained on cv. Aurelio at 33.6?±?1.2°C were used to get a selected resistance breaking isolate of M. hispanica that was able to reproduce on the three pepper cultivars. Our results suggest that the initial M. hispanica isolate is a mixture of virulent and avirulent individuals. The pepper cultivars tested, have potential to reduce M. hispanica populations in agro-ecosystems under certain conditions, but they should be used as a part of an integrated management strategy in order to prevent the development of virulent populations.  相似文献

The infection and colonization process of Colletotrichum acutatum on ripe blueberry fruit from two cultivars with different susceptibility to anthracnose were examined using light and confocal laser scanning microscopy. Ripe fruit from susceptible cv. Jersey and resistant cv. Elliott were drop-inoculated with a conidial suspension of C. acutatum, and epidermal peels were evaluated at selected times after inoculation and incubation. Results from pre-penetration studies demonstrated that there were significant differences in the rate of formation of melanized appressoria between the two cultivars, with the rate of formation being faster in the susceptible one. In both cultivars, penetration by the pathogen occurred via appressoria 48 h post-inoculation (hpi). However, in the susceptible cv. Jersey, C. acutatum then adopted an intracellular hemibiotrophic-like infection strategy, whereas in the resistant cv. Elliott subcuticular intramural-like infection occurred. In cv. Jersey by 108 hpi, intracellular growth of the pathogen led to the formation of numerous acervuli, with orange conidial masses. By 120 hpi, the conidial masses had coalesced covering the entire inoculated area. In cv. Elliott, acervuli were not seen until 144 hpi and contained few conidia. These results demonstrate for the first time the ability of C. acutatum to adopt a different infection and colonization strategy depending on the susceptibility of the host tissue being colonized.  相似文献

Real-time PCR was used to detect and quantify Verticillium dahliae and to assess the susceptibility of four Capsicum annuum cultivars (Luesia, Padrón, SCM331 and PI201234) and the Capsicum chinense cv. C118 to this pathogen. The symptoms which developed after infection included stunting and yellowing, and were more acute in the cv. SCM331, which also suffered defoliation in later stages of the disease and in C118, which suffered severe stunting. Quantification of the pathogen DNA in roots 23 and 34 days post-inoculation (dpi) revealed that there were significantly higher amounts of Verticillium dahliae DNA in C118 than in the other cultivars, followed by SCM331, Padrón and PI201234. The lowest amounts of fungal DNA in roots were found in Luesia. In hypocotyls, the highest amounts of fungal DNA were found in SCM331, while Luesia, Padrón and PI201234 had much lower amounts, and C118 had intermediate levels. When a compatible versus an incompatible system was studied, using the near-isogenic tomato lines LA3030 (susceptible) and LA3038 (resistant to V. dahliae), we were able to detect fungal DNA in both lines. As expected, the fungus/plant DNA ratio was lower in LA3038 than in LA3030 and it decreased with time in LA3038. The amount of Verticillium dahliae DNA in the roots of LA3030 remained constant between days 23 and 34 post-inoculation, but increased 10-fold in collars. Finally, when real-time PCR was applied as a diagnostic method to samples from pepper plants, soil and water collected from farms in northwest Spain, we were able to detect V. dahliae DNA in these samples even when symptoms of the disease were not evident.  相似文献

Colletotrichum gloeosporioides (Penz.) Penz. & Sacc. is an important fungal pathogen known to cause glomerella leaf spot (GLS). The objective of this study was to analyze the infection of C. gloeosporioides on leaves of apple cv. Fuji (resistant) and cv. Gala (susceptible) and apply proteomics techniques to study the apple defense responses 48 h after inoculation (h.a.i.). On both of cultivars, C. gloeosporioides started to germinate at 3 h.a.i. on adaxial surface and produced appressoria adhering to epidermal cell juxtapositions. Histological analysis showed more stratified parenchyma in leaves of cv. Fuji than cv. Gala associated with differences in the chemical composition of cell walls. Total and unique proteins expressed by cvs. Fuji and Gala at 3, 12, 24 and 48 h.a.i. were detected by comparative proteomes analysis. A total of 42 unique proteins expressed at 24 and 48 h.a.i. were identified by MALDI/TOF mass spectrometry, and most of these proteins were identified as directly involved in defense responses.  相似文献

A stem and fruit rot of sweet peppers ( Capsicum annuum ) occurred in 1992 in West Sussex affecting a large proportion of plants of cv. Tasty and to a lesser extent cv. Mazurka. Fusarium solani was isolated from the stem lesions and also from a short distance above and below them. The disease was largely nodal. Distinctive fruit symptoms also occurred. Symptoms were reproduced by inoculation of wounds on the stem. On agar and in a small in vivo test the pathogen was sensitise to benomyl at 2 μg ml. The disinfectants, commercial sodium hypochlorite, Purogene and Tego 51 effectively killed macrospores following exposure for 15 min.  相似文献

L型凝集素类受体激酶(LecRKs)广泛参与植物的先天免疫过程。目前未见在辣椒Capsicum annuum中全基因组鉴定LecRKs的报道。本研究对辣椒中的CaLecRK进行了全基因组鉴定, 并在接种辣椒疫霉Phytophthora capsici条件下通过基因表达分析探究其对辣椒疫霉的响应情况, 旨在挖掘参与辣椒抗疫病防御反应的CaLecRK基因。研究结果表明, 辣椒基因组中共鉴定出24个CaLecRK, 以其构建系统发育树发现, 可将24个CaLecRK分为7个分支。基因表达分析结果显示, 有4个CaLecRK基因(CaLecRK2.2、CaLecRK3.2、CaLecRK8.1和CaLecRK10.1)受辣椒疫霉诱导, 和接菌后0 h相比, 接菌处理后12 h 或36 h基因表达差异显著, 推测其在辣椒抗疫病防御反应中发挥了重要作用。  相似文献

The response of a susceptible coffee cultivar (Caturra) to infection by the root-knot nematode Meloidogyne exigua was compared histologically with that of cv. Iapar 59 possessing the recently identified Mex-1 resistance gene. The reproductive behaviour of the nematode was also compared in the two cultivars. Penetration and development in resistant plants were reduced in comparison with susceptible plants. Several cell features, including dark-stained cytoplasm and altered organelle structure, were observed in the resistant cultivar, indicating a hypersensitive-like (HR) response of the infested host cells. Features of giant cells were sometimes found beside necrotic-like areas, but the corresponding feeding sites were frequently associated with nematodes displaying abnormal shape. Six weeks after inoculation, root systems of cv. Caturra contained significantly more nematodes than those of cv. Iapar 59 (mean values 1574 and 41, respectively). The susceptible cultivar presented a minimum of 11 galls per plant, compared with only one or two galls per plant in the resistant cultivar. The findings are discussed in the context of plant–pathogen interactions.  相似文献

Proliferation and collapse of subcuticular hyphae of Venturia nashicola race 1 were studied ultrastructurally, after inoculation of susceptible Japanese pear cv. Kousui, resistant Japanese pear cv. Kinchaku, resistant Asian pear strain Mamenashi 12 and nonhost European pear cv. Flemish Beauty leaves, to understand the nature of the resistance mechanism. After cuticle penetration by the pathogen, the hyphae were observed at lower frequency in epidermal pectin layers and middle lamellae of leaves of the three resistant plants than in those of susceptible ones. This result suggested that fungal growth was suppressed in the incompatible interaction between pear and V. nashicola race 1. In the pectin layers of all inoculated plants, some hyphae had modifications such as breaks in the plasmalemma with plasmolysis, necrotic cytoplasm and degraded cell walls. More hyphae had collapsed in the leaves of the three resistant plants than in those of the susceptible cv. Kousui. In collapsed hyphae, the polymerized cell walls broke into numerous fibrous and amorphous pieces, showing that the scab resistance might be associated with cell wall-degrading enzymes from pear plants.  相似文献
