Disks from different tissues were obtained from "Redchief Delicious" apple fruit (Malus domestica Borkh.) and analyzed for the ability to metabolize 1-pentanol as well as synthesize constitutive esters and alcohols under anoxic and aerobic conditions. The skin tissue displayed a greater capacity to synthesize pentanal, pentyl acetate, pentyl propionate, pentyl butyrate, and pentyl hexanoate than the hypanthial and carpellary tissues during incubation with 1-pentanol. With the exception of pentyl acetate and pentyl propionate biosynthesis, the hypanthial tissue synthesized these compounds at a higher rate than the carpellary tissue. Anoxia inhibited both constituent and 1-pentanol-derived ester biosynthesis. While anoxia inhibited ester biosynthesis, ethanol biosynthesis increased at a greater rate in tissue disks held under these conditions. Biosynthesis of 1-butanol, 2-methyl-1-butanol, and 1-hexanol was greater in tissue disks held in air during the first part of the measurement period and dropped off more rapidly than those transpiring in tissue disks held under anoxic conditions. The biosynthetic rates of all esters, both constituent and 1-pentanol-derived, increased as a result of air exposure. While hypoxic or anoxic conditions may promote ethanol synthesis, these conditions also appear to inhibit the formation of the ethanol-derived esters partially responsible for the off-flavor in apples attributed to ultralow O(2) controlled atmosphere storage.  相似文献

The effect of several cycles of varying length of alternate aerobic and anaerobic conditions on redox potential, organic matter decomposition and loss of added and native nitrogen was investigated under laboratory conditions in flooded soil incubated for 128 days. Redox potential decreased rapidly when air was replaced with argon for the short-time cycles, but decreased more slowly where the aerobic period was long enough to permit build-up of nitrate. The minimum redox potential reached during the anaerobic period was generally lower for the longer cycles, but in all cases was low enough for denitrification to occur. Rate of decomposition of organic matter was faster in the treatments with a greater number of alternate aerobic and anaerobic periods. Total N (native and applied) losses as high as 24.3 per cent occurred in the treatment with the maximum number of cycles and with alternate aerobic and anaerobic periods of 2 and 2 days. Increasing the durations of the aerobic-anaerobic periods decreased the loss of N. A maximum loss of 63.0 per cent of applied 15NH4-N resulted from the shortest (2 and 2 day) aerobic and anaerobic incubation. For soil undergoing frequent changes in aeration status the only labelled N that remained at the end of incubation was found in the organic fraction. Loss of N may have been even greater if labelled inorganic N had not been immobilized by microorganisms decomposing the added rice straw. The greater loss of N resulting from the 2 and 2 day aerobic-anaerobic incubation shows that, in soils where the redox potential falls low enough for denitrification to occur, increasing the frequency of changing from aerobic to anaerobic conditions will increase the loss of N.  相似文献

Mondial Gala apples were harvested at commercial maturity and stored at 1 degrees C under either air or controlled atmosphere (CA) conditions (2 kPa O2/2 kPa CO2 and 1 kPa O2/1 kPa CO2), where they remained for 3 or 6 months. Data on emission of selected volatile esters, alcohol precursors, and activity of some aroma-related enzymes in both peel and pulp tissues were obtained during subsequent shelf life of fruit and submitted to multivariate analysis procedures. CA storage caused a decrease in the emission of volatile esters in comparison to storage in air. Results suggest that lessened ester production was the consequence of modifications in activities of alcohol o-acyltransferase (AAT) and lipoxygenase (LOX) activities. For short-term storage, inhibition of lipoxygenase activity in CA stored fruit possibly led to a shortage of lipid-derived substrates, resulting in decreased production of volatile esters in spite of substantial ester-forming capacity that allowed for some recovery of fruit capacity for ester emission during the shelf life. For long-term storage, strong inhibition of AAT activity in CA stored fruit in combination with low LOX activities resulted in unrecoverable diminution of biosynthesis of volatile esters.  相似文献

To study the formation of fumonisin artifacts and the binding of fumonisins to matrix components (e.g., saccharides and proteins) in thermal-treated food, model experiments were performed. Fumonisin B(1) and hydrolyzed fumonisin B(1) were incubated with alpha-d-glucose and sucrose (mono- and disaccharide models), with methyl alpha-d-glucopyranoside (starch model), and with the amino acid derivatives N-alpha-acetyl-l-lysine methyl ester and BOC-l-cysteine methyl ester (protein models). The reaction products formed were analyzed by liquid chromatography-electrospray ionization-tandem mass spectrometry. The incubation of d-glucose with fumonisin B(1) or hydrolyzed fumonisin B(1) resulted in the formation of Amadori rearrangement products. Whereas conjugates were found following the reaction of sucrose, methyl alpha-d-glucopyranoside, and the amino acid derivatives with fumonisin B(1), the heating with hydrolyzed fumonisin B(1) yielded no artifacts. For structural determination, the stable reaction product formed by heating of methyl alpha-d-glucopyranoside (as starch model) with fumonisin B(1) was purified and identified by nuclear magnetic resonance spectroscopy as the diester of the fumonisin tricarballylic acid side chains with methyl alpha-d-glucopyranoside. These model experiments demonstrate that fumonisins are able to bind to polysaccharides and proteins via their two tricarballylic acid side chains.  相似文献

为了模拟水稻土淹水过程,探讨不同淹水培养时间水稻土中铁还原微生物群落利用不同碳源的活性变化特征,以接种不同淹水时期的浙江水稻土浸提液作为微生物群落来源,以人工合成的Fe(OH)3为惟一的电子受体,不同碳源作为惟一电子供体,在30℃恒温条件下厌氧培养,定期测定Fe(Ⅱ)含量和pH值变化,采用Logistic模型进行动力学分析。研究结果表明,葡萄糖作为电子供体时,不同淹水时期的微生物群落总体上对Fe(OH)3还原反应有较快的响应;丙酮酸盐作为碳源时,铁还原反应启动的时间整体迟于葡萄糖,Fe(Ⅱ)累积量在反应30d才表现出显著累积并逐渐趋于稳定;淹水20d的微生物群落能最先利用乳酸盐还原Fe(Ⅲ),反应15 d的Fe(Ⅱ)累积量达到601.60 mg.L^-1;淹水30 d的铁还原微生物群落对乙酸盐的利用能力增强,最大Fe(Ⅱ)累计量升高到538.47 mg.L^-1,Fe(Ⅲ)还原率达到75.81%。不同淹水时期利用各种碳源的体系pH表现为葡萄糖从中性下降至酸性,丙酮酸盐和乳酸盐中性偏酸,乙酸盐的pH略微偏碱,不同淹水时期出现的水稻土微生物群落结构不同是导致Fe(OH)3还原能力不同的主要原因。不同的碳源利用可以指示不同的铁还原微生物群落变化:淹水培养早期的铁还原微生物群落对葡萄糖和丙酮酸盐的利用较为迅速和显著,同一时期出现的微生物群落不能以乙酸盐作为电子供体;淹水培养后期的铁还原微生物群落以乳酸盐和乙酸盐为优势碳源来还原Fe(OH)3。  相似文献

The transformation of MeHg under anaerobic conditions in axenic cultures ofDesulfovibrio desulfuricans strain LS is compared to that in anoxic marine sediments contaminated by Hg of industrial origin. MeHg was added to cultures ofD. desulfuricans strain LS and incubated at 28°C for two weeks. Significant amounts of dimethylmercury and metacinnabar were produced. These two Hg compounds were formed from the slow decomposition of the intermediate dimethylmercury-sulfide. Other collateral compounds, such as methane and ionic Hg, were also detected during the MeHg degradation process. On the other hand a sample of fresh sediment (1.5 g d.w.) was spiked with 10 μg of MeHg and 2 mmoles.ml?1 of pyruvate, as carbon source for sulfate-reducing bacteria. After 9 days of incubation at 28°C, significant amounts of dimethylmercury were produced. A lower content of this volatile species was found in a subsample of sediment supplemented with sodium molybdate, which is a strong inhibitor of sulfatereducing activity. A kinetic study showed the disappearance of monomethylmercury from the sediment and the formation of dimethylmercury over the incubation period. The environmental significance of dimethylmercury and dimethylmercury-sulfide in the natural biogeochemical cycle of Hg is discussed.  相似文献

Abstract

The enzyme arylamidase [EC 3.4.11.2] catalyzes the hydrolysis of N‐terminal amino acids from arylamides. Because it has been proposed that this enzyme may play a major role in nitrogen (N) mineralization in soils, studies were carried out using short‐term laboratory incubations under aerobic and anaerobic conditions and chemical hydrolysis of soil organic N to assess the N mineralization in a range of 51 soils from six agroecological zones of the North Central region of the United States. The enzyme activity was assayed at its optimal pH value. With the exception of the values obtained for field‐moist soils incubated under anaerobic conditions, the amounts of N mineralized by all the biological and chemical methods studied were significantly correlated with arylamidase activity, with r values of 0.54*** for the amounts of inorganic N produced under aerobic incubation, of 0.44** for anaerobic incubation of air‐dried soils, of 0.53*** and 0.55*** for the amounts of ammonium (NH4 +)‐N released by steam distillation with PO4‐B4O7 for 4 and 8 min, respectively; and of 0.49*** and 0.53*** for the amounts of NH4 +‐N released by steam distillation with disodium tetraborate (Na2B4O7) for 4 min or 8 min, respectively. The amounts of N extractable with hot potassium chloride (KCl) were most significantly correlated with arylamidase activity (r=0.56***). Arylamidase activity was significantly correlated with organic carbon (C) (r=0.49***), organic N (r=0.55***), and fixed ammonium (NH4 +)‐N (r =0.42**).  相似文献

Riparian wetlands are subject to nitrogen enrichment from upgradient agricultural and urban land uses and also from flooding by nitrogen-enriched surface waters. The effects of this N enrichment on wetland soil biogeochemistry may be mediated by both the presence of plants and the presence of redox-active compounds, specifically iron oxides in the soil. Despite the extensive research on wetland N cycling, the relative importance of these two factors on nitrogen is poorly known, especially for forested wetlands. This study evaluates the responses of the N and the Fe cycles to N enrichment in a riparian forested wetland, contrasting vegetated field plots with plots where the vegetation was removed to test the role of plants. Furthermore, in vitro anaerobic incubations of the experimental soils were performed to track Fe chemical changes over time under anoxic or flooded conditions. Wetland soils treated with N in form of urea, as expected, had significantly higher amounts inorganic nitrogen. In the soils where vegetation was also removed, in addition to inorganic nitrogen pool, increase in organic nitrogen pool was also observed. The results demonstrate the role of vegetation in limiting the effects excess urea has on different soil nitrogen pools. Results from anaerobic incubation of the experimental soils demonstrated the effects of N enrichment on the wetland Fe cycle. The effects of excess nitrogen and the role of vegetation on the Fe cycle in riparian wetland soil became more evident during anaerobic incubation experiments. At the end of the field experiment, Fe concentrations in the soils under the treatments were not significantly different from the control soils at the 5% confidence level. However, during the anaerobic incubation experiment of soils collected at the end of the experiment from these plots, the N-enriched soils and the unvegetated soils maintained significantly elevated concentrations of reducible Fe(III) for the initial 2-week period of incubation, and the soils collected from the plots with both the treatments had the highest Fe(III) concentrations. After 20 days of incubation, however, the Fe(III) concentrations decreased to the similar concentrations in all the incubated soils. The study clarifies the roles vegetation play in mediating the effects of N enrichment and also demonstrates that N enrichment does affect wetland redox cycle, which has strong implications on ecosystem services such as water quality improvement.  相似文献

Fertilization rates of nitrogen (N) in rice crop are based on crop needs and the capacity of the soil to supply N, which is determined by several methods. This study evaluated a method of anaerobic incubation without shaking for extended periods of time. Three rice soils of the Inceptisol, Alfisol, and Vertisol orders were fertilized with N rates of 0, 80, and 160 kg ha?1 and incubated for 0, 7, 14, 21, and 28 d at 20 and 40 °C. The methodology also used short-term anaerobic incubation and constant shaking for 7 d at 40 °C. Field experiments were conducted with rice in the same soils with equal N rates, and extracted plant N was compared with mineralized N. Results indicated that the methodology without shaking is comparable to the constant shaking procedure for short-term incubations. However, incubations without shaking for longer periods of time had the greatest relation to crop extracted N.  相似文献

Apple (Malus × domestica Borkh.) fruit intended for long-term storage are frequently harvested commercially before becoming fully ripe, often resulting in poor aroma development. Since postharvest calcium dips have proved effective for the enhancement of flavor-related volatile esters after cold storage of apples, this study was undertaken in order to assess whether preharvest calcium sprays (7 weekly applications at 1.6%, w/v, 81-123 days after full bloom) could also aid in improving this important attribute at harvest. This procedure significantly increased calcium content in treated fruit. The emission of aroma-related volatile esters by untreated and calcium-treated 'Fuji' apples was then monitored during maturation and ripening over two months prior to commercial harvest. Results indicate that most of the compounds contributing to overall flavor in ripe fruit were enhanced in response to preharvest calcium applications, suggesting that this procedure may be suitable for the improvement of fruit aroma at harvest. The emission of acetate esters was particularly favored, consistent with higher acetaldehyde contents in treated fruit. These effects arose apparently from increased pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH) activities, possibly leading to a better supply of alcohols and acyl CoAs for ester biosynthesis.  相似文献

Phytotoxicity was demonstrated in the aqueous extract of wheat (Triticum aestivum, L.) straw suspensions. When extracts were incubated under anaerobic conditions, the development of phytotoxicity was greater at 20°C than at 10°C. The toxic products formed during incubation depended upon the incubation medium. Acetic and butyric acids were the major toxins produced in liquid straw fermentations during the first 2 weeks. However, after that time the acids did not account for the total toxicity, suggesting formation of unidentified phytotoxins. Acetic, propionic and butyric acids were the toxins formed in sand culture. The development and accumulation of phytotoxins were favored where the sand-straw mixtures were water-saturated. The implications of these findings to wheat cultural practice is discussed.  相似文献

【目的】动态连续监测添加碳氮底物后各气体产物—O2、 NO、 N2O、 CH4和N2的排放,对土壤碳氮转化过程和气体产生过程做更深入的理解,揭示不同土地利用方式典型红壤的温室气体产生机制。【方法】采集长江中游金井小流域不同土地利用方式稻田和菜地土壤为研究对象,利用全自动连续在线培养检测体系(Robot系统),通过两组试验分别研究土壤碳氮转化过程中各气体产物的动态变化。试验1采用菜地和稻田土壤进行好气培养,设置不施氮对照、 添加40 mg/kg铵态氮、 添加40 mg/kg铵态氮+1%硝化抑制剂、 添加40 mg/kg硝态氮、 添加40 mg/kg硝态氮+1%葡萄糖、 缺氧条件下添加40 mg/kg硝态氮+1%葡萄糖6个处理。试验2采用稻田土壤进行淹水培养,设不施氮对照、 添加40 mg/kg铵态氮、 添加40 mg/kg铵态氮+1%硝化抑制剂、 添加40 mg/kg铵态氮+1%秸秆、 缺氧条件下添加40 mg/kg铵态氮+1%的葡萄糖、 添加40 mg/kg硝态氮、 添加40 mg/kg硝态氮+1%葡萄糖、 缺氧条件下添加40 mg/kg硝态氮+1%葡萄糖8个处理。培养温度均为20℃,土壤水分含量为70% WFPS (土壤孔隙含水量),培养周期为15天。【结果】从菜地和稻田土壤不同碳氮添加处理气态产物及无机氮的动态变化可看出: 1)菜地土壤好气培养初期硝化作用产生了大量N2O; 受低碳和低含水量的限制,反硝化作用较弱。当提供充足碳源和厌氧条件,出现N2O和NO的大量排放。2)在好气稻田和淹水稻田培养过程中,反硝化作用是N2O产生的主要途径。3)稻田土壤中,提供充足碳源和厌氧条件,各气态产物出现的顺序依次是NO、 N2O和N2,与三种气体在反硝化链式反应过程中的生成顺序一致。淹水稻田加铵态氮和碳源处理N2为主要产物,添加硝态氮处理后,N2O成为主要气态产物。当土壤碳源充足时,反硝化过程进行彻底,反硝化产物以终产物(N2)为主。4)在稻田土壤出现厌氧或添加碳源条件下,均检测到大量CH4产生; 且在甲烷产生的同时,NO-3几乎消耗殆尽。【结论】金井小流域典型红壤菜地N2O主要来自于硝化作用,好气和淹水稻田N2O主要来源于反硝化作用; 当碳源充足和厌氧时,菜地及稻田反硝化作用增强; 反硝化产物组成、 产物累积量及出峰顺序与碳源和氧气浓度有关。  相似文献

The effect of soil aeration status on carbon partitioning of a labelled organic substrate (14C-[U]-glucose) into CO2, microbial biomass, and extra-cellular metabolites is described. The soil was incubated in a continuous flow incubation apparatus under four different aeration conditions: (1) permanently aerobic, (2) permanently anaerobic, (3) shifted from anaerobic to aerobic, and (4) shifted from aerobic to anaerobic. The soil was pre-incubated for 10 days either under aerobic or under anaerobic conditions. Afterwards, glucose was added (315 g C g–1) and the soils were incubated for 72 h according to four treatments: aerobic or anaerobic conditions maintained, aerobic conditions shifted to anaerobic conditions and anaerobic conditions shifted to aerobic conditions. Carbon partitioning was measured 0, 8, 16, 24, 48 and 72 h after the glucose addition. In permanently aerobic conditions, the largest part of the consumed glucose was built into microbial biomass (72%), much less was mineralised to CO2 (27%), and only a negligible portion was transformed to soluble extra-cellular metabolites. Microbial metabolism was strongly inhibited when aeration conditions were changed from aerobic to anaerobic, with only about 35% of the added glucose consumed during the incubation. The consumed glucose was transformed proportionally to microbial biomass and CO2. In permanently anaerobic conditions, 42% of the consumed glucose was transformed into microbial biomass, 30% to CO2, and 28% to extra-cellular metabolites. After a shift of anaerobic to aerobic conditions, microbial metabolism was not suppressed and the consumed glucose was transformed mainly to microbial biomass (75%) and CO2 (23%). Concomitant mineralisation of soil organic carbon was always lower in anaerobic than in aerobic conditions.  相似文献

The inhibitor of tyrosinase activity in black rice bran was investigated. The methanol extract from black rice bran was re-extracted with hexane, chloroform, ethyl acetate, or water. The ethyl acetate extract had the most potent inhibition against tyrosinase activity by 80.5% at a concentration of 0.4 mg/mL. Inhibitory compound in the ethyl acetate fraction was isolated by silica gel column chromatography, and identified as protocatechuic acid methyl ester (compound 1) by GC, GC-MS, IR, and 1H and 13C NMR spectroscopy. Compound 1 inhibited 75.4% of tyrosinase activity at a concentration of 0.50 micromol/mL. ID(50) (50% inhibition dose) value of compound 1 was 0.28 micromol/mL. To study the structure-activity relationship, protocatechuic acid (2), vanillic acid (3), vanillic acid methyl ester (4), isovanillic acid (5), isovanillic acid methyl ester (6), veratric acid (7), and veratric acid methyl ester (8) were also assayed.  相似文献

A method for the determination of manganese in blood and animal tissue has been developed involving wet digestion of the sample by perchloric and nitric acids, complexing with sodium diethyldithiocarbamate, and extracting with methyl isobutyl ketone at pH 6.7. The methyl isobutyl ketone is removed, and the residue is dissolved in 0.05N HCl in acetone-water (9 + 1) and is aspirated into an air-acetylene flame of an atomic absorption spectrometer. The limit of detectability is about 10 ng Mn/ml in the solution aspirated or 2 ng Mn/ml in bovine blood for a 20 ml sample.  相似文献

Abstract

Soil nitrogen (N) mineralization indicators are useful only if they are sensitive to management practices. The precision of measurement and the sensitivity of the following indicators to crop sequence, tillage, and liming effects were compared: (i) mineral N production during a 24‐day incubation under aerobic conditions, (ii) ammonium (NH4)‐N production under waterlogged conditions, (iii and iv) hot potassium chloride (KCl)–extractable and hydrolyzable NH4‐N (the latter obtained by subtracting initial NH4‐N from extracted NH4‐N), and (v) protease activity. The coefficients of variation decreased in this order: protease activity>KCl‐hydrolyzable NH4‐N>aerobic incubation>KCl‐extractable NH4‐N=anaerobic incubation. Most of the test results obtained using the indicators were correlated with each other. Mineralizable N measured by aerobic and anaerobic incubation was sensitive to tillage, liming, and crop sequences, especially when using soil 5‐cm deep. Hot KCl‐extractable NH4‐N was influenced by tillage but not liming, and less sensitive than the incubation procedures to crop sequence. The protease assay produced no significant test. It was concluded that anaerobic incubation can provide a relatively sensitive assessment of management effects on soil mineralizable N.  相似文献

A novel system for low-temperature alcoholic fermentation of glucose is described. This system consists of kefir yeast immobilized on delignified cellulosic materials. Batch fermentations were carried out at various pH values, and the effect of temperature on kinetic parameters, in the range of 5-30 degrees C, was examined. At pH 4.7 the shortest fermentation time was obtained. The formation of volatiles indicates that the concentration of amyl alcohols (total content of 2-methylbutanol-1 and 3-methylbutanol-1) is reduced as the temperature becomes lower. Propanol-1 and isobutyl alcohol formation drops significantly below 15 degrees C. The percentage of ethyl acetate increases as the temperature is diminished. At 5 degrees C the content of total volatiles in the product was only 38% of the volatiles formed during fermentation at 30 degrees C.  相似文献

Fatty acid (FA) selectivity of immobilized Candida antarctica B lipase was assessed as influenced by various cosubstrate systems for ester synthesis. Reaction mixtures contained a homologous series of even-chain n-acyl donor (C(4)(-)(16)) substrates (FA or their methyl esters, FAME) and a single alcohol cosubstrate (propanol, 2-propanol, or their acetate derivatives) in hexane. Multiple FA optima were often observed, with preferences for C(6) (or C(4)) followed by C(14) and sometimes C(10). The degree of selectivity among acyl donors was modest (up to 1.28-2.60, based on ratios of selectivity constants) and was dependent on the choice of cosubstrate system. Acyl group selectivity ranged up to 1.31-1.36 for [FA + alcohol], 1. 48-2.60 for [FAME + alcohol], 1.30-1.72 for [FA + alcohol acetate], and 1.28-1.88 [FAME + alcohol acetate] reaction systems. General shifts in selectivity were observed between short-chain (C(4)(-)(8)) and long-chain (C(10)(-)(16)) FA as groups with propanol cosubstrate, whereas shifts in reaction selectivity were observed toward specific FA(s) for 2-propanol cosubstrate. Selectivity among a series of alcohol cosubstrates ranged up to 13-fold in esterification reactions with C(6) FA.  相似文献
