Introgression libraries can be used to localize genomic regions carrying quantitative trait loci (QTL). We used this approach to detect QTL regions affecting the per se performance of agronomic and quality traits with two rye (Secale cereale L.) introgression libraries. Our objectives were to detect candidate introgression lines (pre‐ILs) that have a different per se performance than the recurrent parent and to identify the underlying QTL regions. The introgression libraries containing 40 BC2S3 lines each were established with marker‐assisted backcrossing from crosses of the heterozygous Iranian primitive rye accession Altevogt 14160 and the elite inbred line L2053‐N. To assess the phenotypic effect of the donor chromosome segments (DCS) the pre‐ILs were evaluated for grain yield, plant height, thousand‐kernel weight, test weight, falling number and protein content in replicated field trials at five locations in Germany over 2 years. In total, 58 significant (P < 0.05) differences between pre‐ILs and L2053‐N were observed in each introgression library. The DCS in pre‐ILs differing from the recurrent parent possess most likely the responsible QTL. Genomic regions carrying favourable QTL alleles were detected for test weight, thousand‐kernel weight and protein content. We conclude that Altevogt 14160 can not only be used to enrich the genetic variation of the restricted hybrid rye gene pools but will also allow the breeder to efficiently detect favourable QTL for marker‐assisted selection.  相似文献

Introgression populations consist of a set of introgression lines or families, constructed by continuous backcrossing to the recurrent parent, while carrying a limited number of chromosome segments from a donor parent in their genomes. Increasing the genome coverage is an important aim when constructing introgression population. In this study, we proposed bulk pollen pollination (BPP) method and used it to increase the genome coverage of a maize introgression population. The results showed that the genome coverage of the introgression population constructed using BPP method reached 100% at BC3 generation, which accorded with the simulation result. The BPP‐based BC3F1:2 population could identify most quantitative trait loci (QTL) detected using the F2:3 population, especially major QTL. Simulation analysis showed that the genome coverage of introgression population increased with the increase of population size and the number of bulked plants, and decreased with the increase of backcross generation. Our results proved the reliability of the BPP‐based introgression population in increasing genome coverage and detecting QTL, and provided references for constructing high‐coverage introgression populations.  相似文献

Backcrossing is a trait introgression method of renewed importance in crops. The evolution of introgressed or substituted segments through backcross generations has been analyzed theoretically using simulations. In this study, the content of donor genomes, including donor segment number (DSN), donor segment length (DSL), and donor genome size (DGS), were directly analyzed in six crosses over three successive backcrosses using a set of single sequence repeat (SSR) markers covering the entire rice genome. The results of this analysis demonstrated that the average DSN in each genome was 8.39 in BC2F1, 4.13 in BC3F1, and 2.41 in BC4F1, decreasing nearly by half with each backcrossing. The average DSL was 33.43 cM (centiMorgans) in BC2F1, 29.04 cM in BC3F1, and 25.07 cM in BC4F1, display a progressive decrease slightly greater than 10% in each additional backcross generation. Meanwhile the average DGS was 280.51 cM in BC2F1, 119.97 cM in BC3F1, and 60.53 cM in BC4F1, decreasing 57.2% from BC2F1 to BC3F1 and 50.4% from BC3F1 to BC4F1. This revealed that the reduction in DGS was approximately 50% with each backcrossing. These results provide a guide for introgression or substitution of target chromosome segments from donors into recipients in backcross programs. Marker assisted backcrossing has been used effectively to transfer the submergence tolerance gene SUB1 into popular rice varieties, but the approach can be costly. The selection strategy comprising foreground marker and phenotypic selection was investigated as an alternative. The non-significant correlation coefficients between ranking of phenotypic selection and ranking of background marker selection in BC2F1, BC3F1 and BC3F2 generations indicated inefficiency of phenotypic selection compared to marker-assisted background selection with respect to recovery of the recipient genome. In addition, the introgression size of the chromosome fragment containing SUB1 was approximately 17 Mb, showing the effects of linkage drag. The significant correlation coefficient between rankings of phenotypic selection with the percentage of recipient alleles in the BC1F1 generation suggested that background selection could be avoided in this generation to minimize the genotyping cost. The phenotypically selected best plant of the BC3F1 generation was selfed and backcross recombinant lines were selected in the resulting BC3F4 generation. The selection strategy could be appropriate for the introgression of SUB1 QTL in countries that lack access to high-throughput genotyping facilities.  相似文献

The allelic diversity (AD) project of the Germplasm Enhancement of Maize (GEM) programme utilized the double haploid (DH) breeding method to expedite development and release of lines derived from 300 exotic maize races. Using 18 races in this study, differential effects on haploid induction rates (HIRs) and doubling rates (DRs) by the recurrent parents PHB47 and PHZ51, the elevation that the race is traditionally grown at, and by the race itself were addressed in this study. Races from the AD project were grouped by elevation of their origin, high, middle or low altitude. Six races per elevation were randomly selected and backcrossed using both recurrent parents to generate 36 populations. Ten replications were randomized in a complete randomized design for two growing seasons. The recurrent parent effect was significant, with PHB47 having a higher HIR than PHZ51. Effect of elevation was significant with higher HIR associated with low‐elevation origin, and race also proved to be significant. Effects of elevation, recurrent parent and race were not significant for DR.  相似文献

Fusarium wilt, caused by Fusarium oxysporum f. sp. melonis is a common vascular wilt fungal disease in melon across the world. The resistance gene to race 1 of this causal agent, Fom-2, has been previously cloned and its sequence is available. The objective of this research was the introgression of Fom-2 from one resistant (Isabelle) genotype into two susceptible cultivars (Garmak and Tile-torogh) via marker assisted backcrossing. First, the leucine-rich repeats (LRR) domain of Fom-2 from resistant and susceptible genotypes was sequenced to develop functional markers. A length of 1274 bp of the 3′ end of this gene was isolated, cloned and sequenced. The difference between resistant and susceptible genotypes in this region was 28 nucleotide substitutions. Two allele specific primer pairs, Fom2-R409 and Fom2-S253, were designed based on nucleotide substitutions to amplify resistant and susceptible alleles, respectively. For introgression of the gene, donor (Isabelle) and recurrent (Garmak and Tile-torogh) parents were crossed. Resistant plants in BC1F1 and BC2F1 generations were first detected using artificial pathogen inoculation and later the plants were genotyped by functional markers to validate their resistance. The resistant plants were also selected phenotypically in each generation for background genome recovery, which conduced to high similarity of BC3 generation with the recurrent parents. It was proved the developed markers are more precise and efficient than inoculation trial and could be used as confident tools for screening of resistant melon genotypes to Fusarium wilt.  相似文献

The genetic stability and maintenance of Raphanus sativus‐Brassica rapa monosomic chromosome addition lines (a‐h‐types MALs, 2n = 19, BC2), developed by backcrossing the synthesized amphidiploid Raphanobrassica (Raphanus sativus × Brassica rapa, 2n = 38, RRAA) with R. sativus cv. ‘Shogoin’ (2n = 18, RR), was investigated. Transmission of the added alien chromosome through selected smaller seeds (SSS) and the inheritance of morphological traits and random amplified polymorphic DNA (RAPD)‐specific markers together with meiotic chromosome configuration and seed fertility were also investigated for three successive generations (BC3 to BC5). The distinctive traits and the RAPD‐specific markers of the eight types of MAL were substantially inherited and stably maintained throughout three generations, although a few variant plants (2n =18) resembling MALs (2n = 19) and hyperploidal plants (2n = 26 and 2n = 37) were generated in the earlier generations of BC3 and BC4 in comparison with BC5. The average transmission rates for three generations ranged from 26% for both the b‐type and the d‐type to 44% for the e‐type through SSS. On the other hand, the transmission rates through randomly selected seeds (RSS) were lower, ranging from 6.5% for the f‐type to 23.5% for the b‐type. In meiosis, more than 90% of PMCs showed the 9II +1I pairing configuration at metaphase I throughout three generations. For seed fertility, when backcrossed with the radish cv. ‘Shogoin’, the values were approximately 180% to 500% with the mode around 300% with the seed harvested from a pod increasing with the advancing generations. Genetic recombination between the radish chromosomes and the added chromosome is probably rare, suggesting that the added chromosome is mostly maintained unaltered in the background of the radish genome.  相似文献

The development of soybean varieties that lack the β‐conglycinin α‐subunit is an attractive goal because the β‐conglycinin α‐subunit negatively influences the nutrition and gelation of tofu and is a major allergen. To remove this undesirable allergen and simultaneously improve the seed nutritional value and food‐processing quality, marker‐assisted background selection (MABS) was used in backcross breeding to incorporate cgy‐2, a null phenotype version of the gene encoding the β‐conglycinin α‐subunit, from the donor line ‘RiB’ into the genetic background of the Chinese cultivar ‘Dongnong47’ (DN47), a popular high‐oil superfine seed soybean cultivar from Heilongjiang Province, China. In each F2 (F2, BCnF2) generation of the breeding programme, the offspring that carried the introgressed cgy‐2 were identified by sodium dodecyl sulphate–polyacrylamide gel electrophoresis and rescreened by MABS using simple sequence repeat markers to accelerate recurrent parent genome recovery. Of the 49 advanced backcrossing breeding lines (ABLs), the three best lines, ABL1, ABL2 and ABL3, were selected from the BC1, BC2 and BC3 populations, respectively. The ABLs were evaluated for desirable agronomic characteristics, yield‐related traits, amino acid composition, free amino acid composition and tofu‐processing quality in the mature seeds. All of the ABLs lacked the α‐subunit but grew and reproduced normally without deleterious effects on physiological processes such as seed development and germination. The free amino acid content of ABL1 was significantly higher than that of ‘DN47’, with arginine (Arg) being particularly enriched. Compared to the recurrent parent ‘DN47’, the total protein content of the three ABLs was higher, the amino acid composition of the seed proteins was markedly modified and the yield and hardness of the tofu that was made from the ABLs were significantly increased. MABS combined with stringent phenotypic selection in a backcross breeding programme is a feasible strategy for the genetic engineering of seed protein components to produce allergenic subunit‐deficient variant alleles.  相似文献

Vivek Maize Hybrid 9‐ a popular single‐cross hybrid developed by crossing CM 212 and CM 145 was released for commercial cultivation in India. The parental lines, being deficient in lysine and tryptophan, were selected for introgression of opaque‐2 allele using CML 180 and CML 170 as donor lines through marker‐assisted backcross breeding. The opaque‐2 homozygous recessive genotypes with >90% recovery of the recurrent parent genome were selected in BC2F2, and the seeds with <25% opaqueness in BC2F3 were forwarded for seed multiplication. Vivek Quality Protein Maize (QPM) 9, the improved QPM hybrid, showed 41% increase in tryptophan and 30% increase in lysine over the original hybrid. The grain yield of the improved hybrid was on par with the original hybrid. The newly improved QPM maize hybrid released in 2008 will help in reducing the protein malnutrition because its biological value is superior over the normal maize hybrids. This short duration QPM maize hybrid has been adopted in several hill states of North Western and North Eastern Himalayan regions.  相似文献

Fusarium head blight (FHB) has become an important disease of wheat. We introgressed three resistance quantitative trait loci (QTL) alleles on chromosomes 3B, 5A (from CM82036) and 3A (from ‘Frontana’) into European elite spring wheat and performed phenotypic selection among double‐cross (DC) derived progeny in generations DCF2 and DCF3. After recombination and selfing, we analysed 135 phenotypically selected progeny by simple sequence repeat (SSR) markers linked to the QTL. In a second experiment, we forwarded the best 20 progeny for a further two generations by pedigree selection. Progeny were inoculated at two to four locations with Fusarium culmorum and the percentage of infected spikelets per plot was estimated. Both experiments show that phenotypic selection was highly effective. One‐hundred out of 135 phenotypically selected DCF1:3 progeny had the combination of donor‐QTL alleles (3B + 5A + 3A, 3B + 5A) with the highest effects on FHB resistance. In the subsequent generations, sufficient genotypic variance was detected. The best F5:7 bulks had similar resistance to the donor CM82036. The FHB rating was reduced in total by 45% points compared to the parental mean. QTL with high effects can be detected solely by phenotypic selection after targeted introgression.  相似文献

Overuse of a small number of Upland cotton cultivars has narrowed cotton's genetic base, leading to major difficulties in developing cotton cultivars with diverse genetic backgrounds that are able to adapt to adverse conditions. To effectively broaden the genetic base, chromosome introgression lines (ILs) were developed, where TM‐1, the genetic standard of Upland cotton, was used as the common recipient and its two feral landraces, TX‐256 and TX‐1046, were used as the donors. A total of 115 ILs, with an average segment length of 11.15 cM, were first developed via intraspecific hybridization by marker‐assisted selection (MAS) in BC3F2 generations, spanning 3887.75 cM of the cotton genome. Association analysis showed that 63 markers were found to be associated with boll weight (BW), lint percentage (LP) and seed index. The percent of phenotypic variance explained by 148 QTLs detected was 4.12% on average. Eleven and five new QTLs for BW and LP (one stable QTL identified for LP in all environments) were detected, respectively, which can be used for efficiently pyramiding favourable alleles into one cultivar by MAS.  相似文献

Restoration of pollen-fertility is required to guarantee successful seed production of hybrid varieties based on cytoplasmatic male-sterility (CMS) induced by Pampa cytoplasm. As most European restorer lines have still numerous disadvantages, using genetic resources via introgression libraries might be promising to identify and localize new and more effective restorer genes. The objectives of this study were to (1) evaluate in field trials the performance of 39 candidate introgression lines (pre-ILs) for pollen-fertility restoration and (2) to identify donor chromosome segments (DCS) underlying pollen-fertility restoration. Exotic alleles from the Iranian primitive rye accession Altevogt 14160 were introduced into the elite non-restorer inbred line L2053-N by marker-assisted backcrossing. BC2S3 pre-ILs were genotyped using amplified fragment length polymorphisms and simple sequence repeats. Pollen-fertility restoration was evaluated in replicated field trials across two testers at two locations in 2006 for analysing the phenotypic effects of the exotic DCS. Out of 39 pre-ILs, we determined three showing full pollen-fertility across both testers and locations carrying a DCS on chromosome 4R. Moreover, one pre-IL with partial pollen-fertility was detected showing a DCS on chromosome 1R. We conclude that our introgression library can serve as a valuable resource for mining favourable genes from exotic primitive rye, which can greatly promote hybrid rye breeding.  相似文献

To improve salt tolerance of two elite rice varieties, Ce258 and Zhongguangxiang1 (ZGX1), two sets of introgression lines (ILs) each comprising 200 BC1F10 lines derived from a common donor, IR75862, and two recipient parents, Ce258 and ZGX1, were used for mapping of QTLs for four salt tolerance‐related traits at the seedling stage. Although the three parents were susceptible to salt, the two IL populations showed transgressive segregations for salt tolerance with 12 and 8 salt tolerance ILs in the Ce258‐ILs and ZGX1‐ILs. Eighteen main‐effect QTLs were identified for the four traits in the two IL populations, and the IR75862 alleles at most loci showed increased and decreased salt tolerance in the ZGX1 and Ce258 backgrounds, suggesting overwhelming genetic background effects on QTL detection for salt tolerance. The qDSS11 simultaneously detected in the two backgrounds was validated in a F2 population derived from a salt tolerance line and ZGX1. Our results indicated that salt tolerance‐enhancing allele could be identified in the elite susceptible breeding lines and that introgression of the favourable alleles could facilitate the development of superior lines with greater salt tolerance levels.  相似文献

The tepary bean (Phaseolus acutifolius A. Gray) is a desirable genetic resource for incorporation of improved disease, pest, and stress resistance into common bean (P. vulgaris L.). Reproductive barriers separate the two species and the degree to which tepary genes may be introgressed into the common bean genome has not been well described. Greenhouse studies of gene introgression through recurrent back-crossing to common bean were performed using two first backcross (BC1) and nine second backcross (BC2) populations. The truncate primary leaf characteristic of tepary bean was readily observed but the short primary leaf petiole and narrow bract tepary traits were infrequently observed in both BC1 and BC2 populations. In one BC, population high frequencies of adaxial stomata (characteristic of P. acutifolius) and the presence of a 30 kD cotyledon polypeptide from P. acutifolius were also observed; however, a diapho-rase isozyme from P. acutifolius appeared to be eliminated from BC1 progeny at a high rate whereas 6-phosphogluconate dehydrogenase alleles appeared to be transmitted normally. The expression of tepary primary leaf truncate morphology and bract width were correlated with decreased fertility in one of the two BC1 populations. Given suitable parental genotypes and population sizes it should be possible to transfer genes from P. acutifolius to P. vulgaris, especially in regions of the genome which are not associated with inviability or sterility. It will be difficult to transfer factors from certain regions of the tepary genome which are preferentially eliminated during introgression.  相似文献

Historically, conventional breeding has been the primary strategy used to develop a number of Striga‐resistant varieties currently grown in the Sahel of Western Africa. In this study, we have successfully developed and applied a marker‐assisted selection strategy that employs a single backcross programme to introgress Striga resistance into farmer preferred varieties of cowpea for the Nigeria savannas. In this strategy, we have introduced the Striga resistance gene from the donor parent IT97K‐499‐35 into an elite farmer preferred cowpea cultivar ‘Borno Brown’. The selected 47 BC1F2 populations confirmed the recombinants with desirable progeny having Striga resistance gene(s). The 28 lines selected in the BC1F2:4 generation with large seed size, brown seed coat colour and carrying marker alleles were evaluated in the field for resistance to Striga resistance. This led to the selection of a number of desirable improved lines that were immune to Striga having local genetic background with higher yield than those of their parents and standard varieties.  相似文献

Introgression of yellow mosaic disease (YMD) resistance and effect of recurrent parent genome (RPG) on grain yield was studied in 84 soybean genotypes from four populations namely, F2:7, BC1F6, BC2F5 and BC3F4 derived from cross JS335 x SL525. It was observed that in F2:7, BC1F6, BC2F5 and BC3F4 derived lines, RPG contribution was 42.5%, 54.9%, 66.4% and 77.6%, respectively, which is significantly less than expected values. Linkage drag from donor parent with YMD resistance gene may be a possible reason for such deviations. Average grain yield per plant in F2:7, BC1F6, BC2F5 and BC3F4 generations was observed as 13.0, 14.3, 14.9 and 16.1 g, respectively. It was observed that genotypes with more than 80% RPG observed to have both YMD resistance and good yield potential. Graphical genotyping (GGT) analysis revealed that maximum RPG was recovered in chromosomes 8 and 10 and maximum introgression occurred in chromosomes 6 and 19. Our results demonstrated that RPG was positively associated with yield as evident from yield increase with increase in RPG.  相似文献

In a series of three experiments during 1998‐99 and 1999‐2000 at Gazipur, Bangladesh, the causes of segregation of Ogura cytoplasmic genetic male sterility in local cultivars of radish were studied. Male‐sterile populations at the BC5 and BC6 generations were grown under a range of field temperatures for 2 years and the results on pollen fertility tests revealed that the expression of male sterility was not affected by temperature. Neither was a genotype‐year interaction found. The unexpected segregation observed in the male‐sterile backcross generations might be due to the presence of restorer alleles in the maintainer parents.  相似文献

Flooding is one of the major hazards of rice production for the rainfed lowland rice ecosystem, and tolerant cultivars are urgently needed to help protect farmers from submergence damage. A quick and efficient strategy was implemented to introgress SUB1, a major QTL for submergence tolerance, into a rainfed lowland mega variety BR11 of Bangladesh by only two backcrosses and one selfing generation. In marker-assisted backcrossing (MABC), one tightly-linked simple sequence repeat (SSR) and two gene-based markers, four flanking SSR and 116 background SSR markers were used for foreground, recombinant and background selection, respectively, in backcrosses between a SUB1 donor IR40931-33-1-3-2 and BR11. BR11-Sub1, identified in a BC2F2 plant, possessed BR11 type SSR alleles on all fragments analyzed except the SUB1 QTL. The introgression size in BR11-Sub1 was 800 Kb indicating approximately 99.8% identity to BR11. BR11-Sub1 along with other introgression lines showed submergence tolerance similar to the tolerant parent. Yield, yield-component parameters and grain physico-chemical properties showed successful recovery of the BR11 traits in BR11-Sub1, with yield potential ranging from 5.2 to 5.6 t/ha, not significantly different from the recurrent parent mega variety BR11. Producing a large number (~1000) of backcross F1 plants was considered essential to achieve recombination on both sides of the gene, limiting linkage drag with only two backcrosses. A large number of background markers ensured proper recovery of the recurrent parent genome in the BC2F2 generation. The study demonstrates a rapid and highly precise strategy to introgress a major QTL by BC2F2 generation into a modern rice variety using an unadapted donor. The variety can replace BR11 on more than 2 million of ha in Bangladesh and provide major increases in rice production.  相似文献

To combine high yield and improved cold tolerance (CT) in a japonica rice variety, ‘Chaoyou1’ (CY1), 324 BC2F5 introgression lines (ILs) selected for CT from 11 CY1 BC2F4 populations were evaluated in replicated experiments for their CT at the reproductive and seedling stages. A mean realized heritability of 0.747 was achieved in this study for CT. Evaluation of 116 ILs from five BC populations in replicated experiments under stress and normal conditions identified 18 promising ILs that had greatly improved CT and yield compared with CY1. Detailed comparisons between the ILs and CY1 for CT and yield‐related traits under stress and non‐stress conditions provided useful information and better understanding of important issues such as donor selection, selection efficiency and associated changes in non‐target traits in the BC breeding process. The large numbers of CT ILs developed provide useful materials for genetic, physiological and molecular dissection of CT and yield traits using DNA markers and ‐omic tools, and as parents for further improving these traits by designed QTL pyramiding.  相似文献

Common sunflower (i.e., Helianthus annuus) is occasionally crossed with other Helianthus species to gain favourable agronomic traits. An interspecific cross between H. annuus and Helianthus mollis was used to examine events that occurred during the five first generations. First generation hybrid plants were verified using GISH, and F1 hybrid (#17) was back-crossed to common sunflower to obtain BC1 (#4) and BC2 (#21). Some BC2 progeny possessed new traits, i.e., compact (Compact plant architecture is due to short inter-node length, short petiole length and short leaf lamina length) architecture without and with corrugated leaves, which were not observed in the parents. These traits were selected for and fixed after the BC2-S3 (#18) to produce a H. mollis introgression line (namely HM374). Although the size of the population was small, we tentatively construct a genetic map using data from the BC2 generation. Two linkage groups were identified carrying H. mollis specific AFLP markers that were associated with compact plant architecture, and one unlinked marker was shown to explain variation for the corrugated leaves. Introgression line HM374 was used to test the inheritance of compact plant architecture in a cross with an elite high oleic line, OL75HO. Microsatellite (SSR) markers were used to genotype F1 and F2 offspring’s. Our data suggested that the line HM374 was still heterozygous for introgression after eight generations. HM374 displays new architecture traits (i.e. compact architecture with or without corrugated leaves) and carries H. mollis fragments anchored on LG1 and LG11 of sunflower public map.  相似文献
