The relative and absolute values of the electrophoretic fractions of serum proteins of 50 clinically normal cats were determined, using agarose as the supporting matrix. Six protein fractions were clearly and consistently resolved: albumin and alpha1-, alpha2-, beta1-, beta2-, and gamma-globulins. In many cats, the alpha1-, alpha2-, and beta2-fractions were each divided into 2 subfractions. Cats which lived in a research colony environment were found to have significantly increased levels of gamma-globulins as compared with the values in cats kept as house pets. The results of serum protein fractionation using this technique have been compared with the normal feline values reported in the literature.  相似文献

A five year investigation of Brucella antibody prevalence in camel sera was conducted in 1502 one-humped camels of both sexes and different ages. The average (mean +/- SD) incidence rate of positive results was 6.95 +/- 1.55%. Among adult one-humped camels, the rate was 4.94 +/- 2.51% in males and 13.76 +/- 4.41% in females. Juvenile one-humped camel calves showed a 0% incidence rate in males and a 1.82 +/- 3.64% in females. Antibodies against Brucella abortus were prevalent in one-humped camel sera throughout the five years of the survey with incidence rates of 6.54, 5.79, 9.32, 5.03 and 8.06%, respectively from 1985 to 1989.  相似文献

In this preliminary investigation, various hematologic variables potentially influential in determining the degree of blood viscosity were evaluated in 10 Thoroughbred horses subjected to competitive acute running exercise. Following completion of sprints over a distance of 1.25 miles, mean percent (+/- SD) increases in PCV (38.3 +/- 12.9%), RBC (47.8 +/- 15.3%), and rouleaux index (232.7 +/- 176.8%) were recognized. Simultaneous increases in total plasma protein (28.3 +/- 5.31%), serum albumin (26.7 +/- 6.80%), alpha 1-globulin (60.0 +/- 49.0%), alpha 2-globulin (25.5 +/- 27.9%), beta 1-globulin (46.7 +/- 21.1%), beta 2-globulin (35.0 +/- 50.6%), gamma 1- and 2-globulins (38.7 +/- 29.6%), and plasma fibrinogen (12.5 +/- 10.4%) concentrations increased simultaneously. Horses also had consistent decreases in albumin:globulin ratio (-10.0 +/- 7.43%). Alterations in these hematologic values after acute running exercise in Thoroughbred horses accompanied increases in serum (69.3 +/- 39.7%), plasma (39.7 +/- 11.9%), and blood (134.7 +/- 55.3%) viscosity.  相似文献

Serum protein values were determined in 26 healthy dogs using agarose gel electrophoresis (SPE), splitting the electrophoretic separation into six regions: albumin, alpha(1), alpha(2), beta(1), beta(2)and gamma globulins. High-resolution electrophoresis (HRE) was used to separate single proteins. Serum proteins from dogs (26 healthy and 20 affected by various diseases) were then characterized by electrophoretic immunofixation (IFE) and Sudan black staining on HRE film. Haemoglobin and normal canine plasma and serum were used to identify haptoglobin and fibrinogen, respectively.In the standard pattern, determined by HRE, the following proteins were identified: albumin, alpha(1)-lipoprotein (alpha(1)-region), haptoglobin and alpha(2)-macroglobulin (alpha(2)-region), beta -lipoprotein and C3 (beta(1)-region), transferrin and IgM (beta(2)-region), IgG (mostly in gamma -region and partly in beta(2)-region). The HRE pattern shown by healthy dogs could be compared with those of dogs affected by various diseases to obtain clinical information.  相似文献

The aim of this study was to evaluate total serum protein concentration measured using biuret reaction and the protein fractions determined using acetate cellulose electrophoresis in Ragusana donkeys (Equus asinus). Blood samples were collected from 68 clinically healthy female donkeys by jugular venipuncture. The serum levels of total proteins were determined using biuret method, and the separation of proteins was performed using acetate cellulose electrophoresis. Coefficients of variation were also calculated for within-assay precision, and were found to be less than 5% for α- and β1-globulins and 8% or less for albumin, β2-, and γ-globulins. A total of five protein fractions were separated and quantified: albumin, α-, β1-, β2-, and γ-globulins. Data obtained from young and adult subjects were compared using the Mann–Whitney U test. Reference intervals (2.5%-97.5% quantiles) were determined for total proteins (50.0-84.0 g/L), albumin (16.2-36.6 g/L), α-globulins (4.85-19.5 g/L), β1-globulins (2.25-10.35 g/L), β2-globulins (3.30-14.85 g/L), γ-globulins (10.0-30.5 g/L), and albumin/globulin ratio (0.41-1.13). In relation to age, statistically significant differences were found in total protein concentration and γ-globulins. The results obtained in the present study contributed to establish reference intervals of serum protein fractions obtained using acetate cellulose electrophoresis in female Ragusana donkeys to be used by practitioners for health control.  相似文献

The aim of this study was to evaluate the influence of different physiological phases on serum total proteins and their fractions of ten Comisana ewes housed in Mediterranean area. From each animal, blood samples were collected at different physiological phases: late pregnancy, post-partum, early, mid-, end lactation and dry period. On all samples serum total proteins were determined by the biuret method, and albumin, α-globulins, β(1) -globulins, β(2) -globulins and γ-globulins concentrations were assessed using an automated system. One-way repeated measures analysis of variance was applied to determine the significant effect of different physiological phases on the parameters studied. During the late pregnancy and post-partum, total proteins, β1- and β2-globulins and γ-globulins showed the highest values. Starting from post-partum, α-globulins increased to reach their peaks in mid-lactation. Early lactation was characterized by low γ-globulins values. The increase in serum albumin concentration and the drop in some globulin fractions determined the significant increase in albumin/globulin ratio. The obtained results contributed to improve the knowledge on electrophoretic profile during the different physiological phases in ewes, confirming that pregnancy and lactation periods affect the protein metabolism. Particularly, serum protein fractions pattern could give information about dehydration, plasma volume expansion and hepatic function, which occur during the different physiological phases. Dynamics of the protein profile - from pregnancy to dry period - which are provided by our results, could be considered as guidelines for the management strategies to guarantee the nutritional needs of these animals during the different physiological phases and to avoid a decline of productive performance and consequently an economic loss.  相似文献

Copper was determined in the blood of breeding camels, camel calves and racing camels to evaluate copper status in these animals in UAE. Low blood copper concentrations were reported in newly born camel calves (100%) and calves 2–4 months old (68%), breeding camels at early (55.6%) and at mid lactation (48%) and at late pregnancy (69%). This is attributed to the low copper and high sulfate in the Rhodes grass which is the only diet offered to the breeding camels. On the other hand only 9.7% of racing camels showed low copper levels. This is because copper is routinely offered to racing camels when their blood copper is low. Cupric oxide needle capsules orally administered at the rate of 8 g per adult camel was effective in elevating blood copper from 7.083 μmol/L at day zero to 10.074 μmol/L at day 28 after dosing.  相似文献

Blood samples were taken from calves with respiratory disease the first day of examination for determination of the serum concentration of haptoglobin, fibrinogen, alpha-2- and gamma-globulins, and albumin. A clinical examination was performed daily for the duration of the disease. The animals were retrospectively classified in two categories: those animals requiring no treatment or antibiotics alone (group A), and antibiotics associated to anti-inflammatory drugs (group B). The serum proteins were tested in order to check whether they were able to distinguish, on the first day of clinical examination, between calves requiring anti-inflammatory treatment (group B) or not (group A). About 80% of calves were properly classified in both groups by the combined use of the two serum proteins haptoglobin and fibrinogen: these two proteins, and especially haptoglobin, were useful for the identification of calves requiring an anti-inflammatory treatment.  相似文献

The serum proteins in 1 group of healthy breeders, 1 group of healthy pups and 4 groups of foxes suffering from various diseases were separated electrophoretically on cellulose acetate membranes. In most of the sera, the proteins were separated into 1 albumin fraction and 5 globulin fractions designated alpha1, alpha2, alpha3, beta and gamma. The mean concentrations of the total proteins and the various serum fractions in the disease groups were compared statistically with the mean values of the normal groups.A nosematosis group was characterized by a distinct hypergammaglobulinaemia together with an increase in the total protein — and a decrease in the albumin concentrations. However, in 2 cubs recovering from nosematosis the hypergammaglobulinaemia was shown to be reversible.In a feed intoxication group the concentration of albumin was found to be lower and the alpha3-globulin higher than the corresponding values in the healthy group. A virus hepatitis group was characterized by a decrease in the concentrations of albumin and beta-globulin and an increase in the alpha1- and alpha3-globulins. In the toxoplasmosis group the total protein and alpha1- and alpha3-globulins showed concentrations below the normal values.  相似文献

Total serum protein levels, serum protein fraction levels, and specific serum immunoglobulin class or subclass levels were measured in colostrum-fed (CF) and colostrum-deprived (CD) calves during the first 144 hours after birth. Total serum protein values increased at 24 hours in the CF group and then decreased slightly at 144 hours. The increase in total serum protei5) in beta1-, beta2-, and gamma-globulins. The beta2- and gamma-globulin levels decreased by 144 hours, while the serum level of beta1-globulin continued to increase. The CD calves exhibited a significant decrease (P less than 0.05) in total serum protein at 24 hours, folhours, the level of beta1-globulin inlowed by a significant increase (P less than 0.05) at 144 hours. At 24 hours, the level of beta1-globulin decreased slightly, and the level of beta2- and gamma-globulins increased slightly. At 144 creased, and the level of gamma-globulin decreased. The beta2-globulin level did not change. At birth, immunoglobulin (Ig) M was detected in 5 of the 10 calves, IgG1 in 6 of the 10 calves, and IgG2 in 3 of the 10 calves. By 24 hours after birth, all CF calves had detectable levels of IgM, IgG1, and IgG2, and there were significant increases (P less than 0.01) in the mean serum levels of all 3 immunoglobulins. By 144 hours after birth, the serum levels of IgM, IgG1, and IgG2 decreased to various degrees. At 24 hours, the IgM level had not increased in CD calves; however, the level of IgG2 appeared to increase slightly, and the mean IgG1 level increased by approximately 50%. By 144 hours after birth, there was a significant increase (P less than 0.01) in the mean level of serum IgM. The level of IgG, also appeared to increase substantially, while the level of IgG2 appeared to increase slightly.  相似文献

A method is described for production of an equine anti-bovine leukocyte serum (EABLS). Leukocytes were harvested from the milk of cow's udders which had been irritated with endotoxin. The washed leukocytes as antigens were administered to 2 horses in a series of subcutaneous and intravenous injections. There was a variably progressive increase in total serum proteins and a decrease in albumin/blobulin ratios, but the most pronounced change was an increase in beta2-globulins. Accompanying these changes was an increase in the number of precipitin lines as shown by Ouchterlony analysis. Four old cows and 2 calves were intravenously given a 50- and 20-ml dose, respectively, of the EABLS. Two of the cows and 1 of the calves were given the EABLS which had been heated to 56 C for 30 minutes. Both calves and the 2 cows given heated EABLS and 1 cow given unheated EABLS developed marked neutropenia within the 1st hour. The 4th cow given unheated EABLS developed only slight neutropenia. It was concluded that there may be variation in the individual animal's susceptibility to the neutropenia-inducing activity of EABLS.  相似文献

Values for total serum proteins and relative percentages of albumin, alpha 1-globulin, alpha 2-globulin, beta-globulin, and gamma-globulin were determined for the goat. These normal values were compared with those obtained for goats infected with Corynebacterium pseudotuberculosis. Goats chronically infected with C pseudotuberculosis show significantly higher total serum protein values than normal goats, apparently due to increased gamma-globulins. This higher protein value is also associated with a decrease in serum alpha 2- and beta-globulins.  相似文献

It has been postulated that the one‐humped (Arabian) dromedary and the two‐humped (Bactrian) camel originated from a single ancestor. Consequently, the dromedary was considered a breed of the two‐humped camel, based on an anatomical study by Lombardini L, 1879: Ann. Del. Universita Toscane, 259 , 147, who described a reduced second hump like structure in foetal and adult dromedaries. To resolve this lingering issue, we analysed dromedary foetuses and calves. In contrast to the situation in two‐humped camels, we never observed any rudimentary second hump in the dromedary foetuses or calves.  相似文献

Status of certain oxidative stress indices and zinc, copper and iron concentrations in blood were estimated in camel with liver cystic echinococcosis. In comparison to healthy control, the index of serum lipid peroxidation assessed by the malondialdehyde (MDA) level was significantly higher in the parasitized group. However, mean serum total antioxidant status (TAS) and erythrocyte glutathione peroxidase (GPx) were significantly lower in the parasitized group. Serum zinc concentration in camels with liver cystic echinococcosis was significantly lower than healthy control. In parasitized camels, a significant positive correlation of MDA with gamma-glutamyl transferase (GGT), aspartate aminotransferase (AST) and bilirubin was observed. By contrast, MDA was inversely correlated with the values of packed cell volume (PCV), serum albumin and zinc. From the present study, it was concluded that cystic echinococcosis in camel is associated with oxidative stress. The resulting oxidative stress seems to have a role in the injury of hepatocytes, changes of trace elements and destruction of erythrocytes.  相似文献

This experiment was conducted to determine the effects of castration on some metabolic alterations concerning proteins, lipids and minerals metabolism in both immature and mature rabbits. Twenty male New Zealand White rabbits (10 immature and 10 mature) were used. Blood samples were collected from the immature and mature intact animals (control non castrated). Moreover, the castrated animal groups (immature and mature) were sampled at 2, 4, 6 and 8 weeks after castration. The obtained results revealed that, castration caused a significant decrease in serum total protein, albumin, alpha 1-globulins and alpha 2-globulins concentrations. Meanwhile, gamma-globulins level was decreased temporarily at 2 weeks after castration in mature rabbits. beta-globulins level revealed non significant decrease at 2 weeks after castration in immature rabbits followed by a non significant increase. In mature rabbits beta-globulins level non significantly increased. A significant decrease in serum total nucleic acid concentrations was observed after castration throughout the experimental periods, whereas the value of serum uric acid level markedly increased after castration. Serum lipids concentrations (total lipids, total cholesterol, phospholipids and non esterified fatty acids) were increased after castration. The value of serum trace elements including copper, iron, zinc and manganese concentrations showed a significant decrease after castration. The decrease in serum copper and zinc levels were markedly observed in mature castrated rabbits. Also, serum sodium and potassium concentrations were decreased after castration. Testosterone propionate administration in mature castrated rabbits restored most of the investigated serum blood parameters to the levels of intact non castrated mature rabbit. It could be concluded that castration (deficiency of testosterone) induced marked decrease in protein synthesis and increased protein degradation, but serum lipids markedly elevated after castration in both immature and mature animals. Also, castration caused a marked decrease in some serum minerals and trace elements.  相似文献

OBJECTIVE: To determine the effect of transportation stress on serum concentrations of oxidative stress biomarkers of calves. ANIMALS: 105 crossbred beef steer calves (mean [+/-SD] body weight, 207 +/- 21.2 kg). PROCEDURE: Calves were assembled at 1 location in Tennessee, and pretransit (day -3) blood samples were collected. Calves were allotted randomly by body weight into 2 groups. Calves were transported 1,930 miles to a feedlot in Texas, and 1 group received tilmicosin phosphate (33 microg/kg, s.c.) upon arrival. Calves were weighed and blood samples collected on the day of arrival (day 1) and on days 15, 22, and 28. Calves were scored daily for signs of bovine respiratory disease (BRD). Serum total antioxidant capacity (TACA) and serum malondialdehyde (MDA) concentrations were determined. RESULTS: Transportation stress significantly decreased mean serum TACA concentrations (from 147 +/- 31.2 U/mL to 133 +/- 20.1 U/mL) and significantly increased serum MDA concentrations (from 10.9 +/- 18.3 microg/mL to 30.2 +/- 50.5 microg/mL). Calves that died had a 43% increase in serum MDA concentration on day 1, compared with calves that lived (42.2 +/- 67.0 microg/mL vs 29.4 +/- 49.4 microg/mL, respectively). Calves that had > or =3 episodes of BRD had 2-fold higher serum MDA concentrations on day 1 than healthy calves. Tilmicosin-treated calves had a 20.8% significantly greater average daily gain and significantly greater serum TACA concentration than nontreated calves on day 28. CONCLUSIONS AND CLINICAL RELEVANCE: Transportation stress increases serum concentrations of oxidative stress biomarkers that are related to episodes of BRD and mortality in calves.  相似文献

In the present study, we determined the levels of cytokines produced by camel (Camelus bactrianus) peripheral blood mononuclear cells (PBMCs) in response to live attenuated Brucella abortus (B. abortus) S19 vaccine. Seven camels were vaccinated with commercial B. abortus S19 vaccine, and their cytokine responses were determined using a real-time PCR assay. Cytokine responses to B. abortus S19 were examined at 6 hr, 48 hr and 1, 2 and 3 weeks post-vaccination. Serological tests were performed to further confirm these immune responses. The results revealed that IFN-gamma and IL-6 were upregulated during the first week post-vaccination. Low level expressions of IL-1alpha, IL-1beta, TNFalpha and IL-10 and no expression of IL-2 and IL-4 were observed compared with the control camels. The findings showed that B. abortus stimulates cell-mediated immunity by directly activating camel Th1 cells to secrete IFN-gamma. This quantification of cytokine expression in camels is essential for understanding of Camelidae disease development and protective immune responses. This is the first report of in vivo camel cytokine quantification after vaccination.  相似文献

Neomycin sulfate was administered to calves by the intravenous and intramuscular routes. Serum drug levels were determined and the intravenous pharmacokinetic parameters derived using the Gauss-Newton nonlinear fitting algorithm and the two compartment open model. The ultrasonographic appearance and measurements of the normal buffalo and camel eye globes were described in 60 buffaloes (Bos bubalis) aged 1?year (28 eyes) and 10?years (32 eyes), and in 51 humped camels (Camelus dromedarius) aged 1?year (26 eyes) and 10?years (24 eyes). Ocular measurements were recorded by A- and B-scan ultrasonographic examination of 40 buffalo eyes (18 young and 22 adult eyes) and 34 camel eyes (14 young and 20 adult eyes) using a KANGH ultrasound scanner equipped with 10?MHz probe. For gross measurements, 20 buffalo and 16 camel eye globes were frozen and dissected and the same measurements were made using fine callipers macroscopically. The aqueous and vitreous humour of the buffalo and camel eyes appeared anechoic. The cornea, anterior and posterior lens capsule and iris appeared hyperechoic. The ocular measurements for the axial length, vitreous chamber depth (VCD), corneal thickness, lens thickness and scleroretinal rim thickness increase with the advance of age in both buffaloes and camels. Except for the anterior chamber depth, VCD and lens thickness, which were larger in adult camels than in adult buffaloes, no other differences between ocular dimensions were observed in both species. The results of this study are valuable for comparative ocular anatomy and will be useful for ultrasonographic evaluation of ocular diseases in buffaloes and camels.  相似文献

Bactrian camel is an ancient and precious species of livestock; that is, unique resources exist in the desert and have important economic and scientific value. In recent years, the number of Bactrian camels has declined sharply. Due to its long reproductive cycle and seasonal oestrus, the mechanism of oestrus is unknown. To identify candidate biomarkers of reproduction, we performed a comprehensive proteomic analysis of serum from Bactrian camel in oestrus and non-oestrus, using isobaric tags for relative and absolute quantitation (iTRAQ) coupled with tandem mass spectrometry. We identified 359 proteins, of which 32 were differentially expressed: 11 were up-regulated and 21 were down-regulated in samples from camels in oestrus. We validated the differential expression of a subset of these proteins using qPCR and Western blot. Gene ontology annotation identified that the differentially expressed proteins function in cellular processes, metabolic processes and immune system processes. Notably, five of the differentially expressed proteins, PCGF5, histone H1.2, RBP4, FOLR1 and ANTXR2, are involved in reproductive regulatory processes in other animals. KEGG enrichment analysis demonstrated significant enrichment in several cardiac-related pathways, such as ‘dilated cardiomyopathy’, ‘hypertrophic cardiomyopathy’, ‘cardiac muscle contraction’ and ‘adrenergic signalling in cardiomyopathy’. Our results suggest that candidate biomarker (PCGF5, histone H1.2, RBP4, FOLR1 and ANTXR2) discovery can aid in understanding reproduction in Bactrian camels. We conclude that the profiling of serum proteomes, followed by the measurement of selected proteins using more targeted methods, offers a promising approach for studying mechanisms of oestrus.  相似文献
