PCR was used to diagnose toxoplasmosis in two pairs of Barbari goats infected by oral administration of doses of either 10(4) or 10(5) oocysts of Toxoplasma gondii. Blood and lymph node aspirates were collected from the infected goats and control goat at intervals, and tissues were also collected from a fetus that was aborted and a doe that died during the trial. Both processed and unprocessed samples were used for the PCR, using primers directed to the multicopy B1 gene. None of the blood samples was positive, but a specific signal was obtained from the lymph node aspirates after partial DNA extraction. Direct PCR of the lung, muscle and mesenteric lymph node of the doe and lung tissue of the aborted fetus yielded the target fragment. The simplified PCR protocols, including partial DNA extraction and direct assay of lung tissue, were effective for the diagnosis of toxoplasmosis. 相似文献

Fifteen pregnant does were inoculated orally with 10 to 1,000 oocysts of the GT-1 strain of Toxoplasma gondii. Two does were euthanatized on postinoculation days (PID) 10 and 14, and the remaining 13 does aborted or delivered kids transplacentally infected with T gondii on PID 9 to 65. Tissues of all 34 fetuses or kids from the inoculated does were examined for gross and microscopic lesions. Placental necrosis and encephalomyelitis were the most prominent lesions. Toxoplasma gondii invaded the fetal placenta between PID 9 and 11 and invaded fetal tissues 2 to 3 days later. Necrosis attributed to toxoplasmosis, confined to cotyledons, was found in all placentas examined on PID 18. Encephalomyelitis was found in most fetuses or kids between PID 30 and 65. Lesions in fetal tissues were sparse. Because T gondii is rarely found in lesions, histologic examination of the fetal tissues is not likely to help in diagnosing every case of transplacentally induced toxoplasmosis in goats. 相似文献

Fifteen goat kids were experimentally inoculated with Corynebacterium pseudotuberculosis. Five were given a strain of caprine origin (nitrate-negative biotype) intradermally, 5 were given a strain of equine origin (nitrate-positive biotype) intradermally, and 5 were inoculated intranasally with the caprine-origin strain. Animals were monitored for 127 days. The goats given the inocula intradermally developed abscesses; those given caprine-origin strain had multiple lesions both peripherally and in visceral locations (primarily endothoracic abscesses), whereas those given the equine-origin strain had abscesses only at injection sites and draining nodes. The difference in extent of lesions could be due to biotypic bacterial differences or to the individual strains used. Intranasally inoculated goats did not develop abscesses and were essentially no different from controls. The cranial part of the respiratory tract may not be an important portal of entry for C pseudotuberculosis. Serum samples obtained monthly from all animals were subjected to the synergistic hemolysis-inhibition test, which measures antibodies to the exotoxin of C pseudotuberculosis. Animals with abscesses developed titers within 1 month of inoculation. Animals without abscesses remained seronegative. The synergistic hemolysis-inhibition test may be a reliable diagnostic assay for caseous lymphadenitis in goats. 相似文献

The objective of this study was to evaluate the relationship of parasite-specific serum antibodies with the resistance status of Creole kids. The average breeding values on egg output predicted in a context of natural infection at 11 months of age were distant of 1.07 genetic standard deviation between resistant and susceptible animals. After drenching the animals were maintained worm-free during 1 month until experimental infection with 10,000 Haemonchus contortus infective larvae (L3). Enzyme-linked immunosorbent assay was carried out in serum samples to determine the level of IgG, IgA and IgE anti-H. contortus L3 crude extracts and adult excretion/secretion products (ESP). Parasitological and blood immunological parameters were measured on the 2 extreme groups. Despite the absence of any typical signs of haemonchosis, susceptible kids had more than 11 times higher faecal egg counts (FEC) at 35 days post-infection (d.p.i.) than resistant kids had. Levels of immunoglobulin against H. contortus L3 and ESP increased significantly after infection in both groups. However, no difference in the host immune response mediated by immunoglobulin against H. contortus was evidenced between groups. This finding suggests that, in goats previously infected by H. contortus, a degree of protection occurred and the phenotypic and genetic segregation in resistant and susceptible animals were not related to the humoral immune response. The correlation coefficients between FEC and IgE anti-ESP (r=0.593; P<0.05 was significant in both resistant and susceptible animals. Such correlation suggesting a hypersensitivity reaction dependent on worm prolificacy has never been described. This result needs further studies to understand the mechanisms underlying this observation. 相似文献

Seventeen sows were fed 1,000 Toxoplasma gondii oocysts of isolates GT-1 or PT-1 at 32 to 92 days of gestation, and the products of conception were examined for T gondii antibodies and parasites. Twelve of these sows were euthanatized near term between 21 and 62 days after being fed T gondii; fetal body fluids or fetal sera were examined for agglutinating T gondii antibodies, and tissues were bioassayed in mice for T gondii parasites. Six sows produced pigs that had been transplacentally infected with T gondii; one of them aborted a T gondii-infected fetus 17 days after ingesting oocytes. Agglutinating antibodies were detected in fetuses infected in utero, but transplacental transfer of T gondii antibodies was not observed in noninfected fetuses. Transcolostrally acquired T gondii antibodies disappeared by 3 months of age. Diagnosis of transplacental toxoplasmosis was confirmed on the basis of detection of T gondii organisms in fetal tissues by use of histologic examination and bioassay in mice. In conclusion, finding of T gondii antibodies in body fluids could serve as a rapid screening test for transplacental T gondii infection in pigs. 相似文献

Serum samples of five goats inoculated with Toxoplasma gondii were analyzed using the enzyme linked immunosorbent assay (ELISA), indirect hemagglutination (IHA) and western blotting (WB). Antibodies detected by ELISA peaked between 19 and 62 days after inoculation and persisted throughout the experiment with no association to parasitaemia. Using WB, the main antigens detected had molecular weights of approximately 68, 62, 50, 48, 42, 34, 28, 26, 22 and 19 kDa. Antibody titers of between 1:256 and 1:32000 were observed using IHA, with a significant drop in activity after treatment with 2-mercapto-ethanol between days 12 and 48. This coincided with the parasitaemic period that occurs between 5 and 64 days after inoculation. 相似文献

Three goats, experimentally infected with rinderpest virus were examined for the development and distribution of precipitating antigens in various tissues and secretions using the agar gel immunodiffusion test. Virus antigens were detected in ocular secretions and lymph node biopsies from the second to the fourth and fifth days of pyrexia, respectively, but were not detected in nasal secretions. Precipitating antigens were demonstrated in various lymphoid organs, the lung and abomasum of a goat killed on the fourth day of pyrexia. These findings are discussed in relation to the epidemiology of rinderpest in goats in Africa. 相似文献

Ocular lesions associated with Trypanosoma spp. infection have been described in man and many animal species. However, loss of vision has not been demonstrated in humans presenting Chagas disease or in animals affected by different trypanosome species. In order to assess the possible ocular disorders caused by Trypanosoma evansi infection, six goats were inoculated with 1 x 10(5) T. evansi and maintained for 12 months and four goats were used as control. The inoculated animals became positive at serological and parasitological tests at 1-month post-inoculation and showed a subclinical course of the disease. Unilateral superficial corneal ulceration and retinochoroiditis were observed in two inoculated animals. Data from ocular neurologic examination and electroretinography showed no significant differences between inoculated and non-inoculated goats. It could be concluded that Trypanosoma evansi can produce ocular lesion but without apparent loss of vision in goats. 相似文献

Enzyme-linked immunosorbent assays (ELISAs) were developed to detect caprine herpervirus-1 (CpHV-1)-specific IgG1 and IgG2 in sera from 43 naturally infected goats. The analysis of the IgG subclasses showed a dual pattern of distribution in seropositive goats with a major group of animals (36 out of 43) exhibiting significantly higher levels of IgG2 over IgG1 and a minor group (7 out of 43) possessing equal levels of IgG1 and IgG2. Four goats were experimentally infected with a virulent CpHV-1 Ba.1 strain by the intranasal or the intravaginal route and the kinetics of appearance of CpHV-1-specific IgG, IgG1 and IgG2 in the serum were studied. Two weeks following infection, both IgG1 and IgG2 levels increased although convalescent sera (i.e., collected 5–8 weeks post-infection) showed a clear prevalence of the IgG2 subclass. To determine the contribution of the different IgG subclasses to herpesvirus immunity, serum neutralization (SN) assays were performed in both naturally and experimentally infected goats. The kinetics of SN showed that neutralization activity was mainly associated to the IgG1 subclass and this was also confirmed in naturally infected goats. The results are discussed from the standpoint that the profile of the IgG subclasses is instrumental to study immune responses to CpHV-1 and that vaccination strategies may benefit from this information. 相似文献

The effects in goats of the subcutaneous injection of varying doses of Pseudomonas pseudomallei (90 to 500,000 bacilli) suspended in normal saline are described. High doses (greater than or equal to 500 bacilli) caused acute, fatal infections. Lower doses (90 to 225 bacilli) caused acute or chronic disease when infection became established. However, 11 of 18 goats injected with the lower doses of bacilli showed no sign of infection on clinical or bacteriological examination. Response to antibiotic therapy with long acting tetracycline and chloramphenicol was minimal. Goats surviving the initial phase of infection tended to overcome the disease with a corresponding increase in the number of abscesses that were sterile at necropsy. In infected goats, clinical signs included undulating fever, wasting, anorexia, paresis of the hind legs, severe mastitis and abortion. At necropsy, abscesses were found predominantly in the spleen, lungs, subcutaneous injection site and its draining lymph node. 相似文献

Adult sheep and goats and new-born lambs and kids were experimentally infected with a Wesselsbron disease virus. The viraemia in lambs commenced approximately 27 h after infection and lasted on the average for 50 h. A febrile reaction, which was mostly biphasic, commenced several hours after the viraemia and outlasted it by 50 h. The viraemia in adult animals began about 50 h after infection and lasted for 30 h. The fever usually commenced several hours after the viraemia and, as in 3 cases out of 4 in lambs, it outlasted the viraemia by at least 30 h. The virus could be reisolated in mice from every tissue examined in lambs, although it has previously been shown that pathological lesions are restricted to the liver and lymphatic tissues. 相似文献

A total of 25 pregnant goats without neutralizing antibodies against BVD virus were inoculated with two different pestivirus isolates at eight different stages of gestation. In both infection groups, various malformations were observed in fetuses and neonates. In three twins with neutralizing antibodies against BVD virus leukoencephalomalacia occurred, characterized by gelatinous transformation in the cerebral hemispheres. These lesions were comparable to alterations described in alternative pathology of Border disease in sheep. Although the immunohistological findings are characteristic for immunological tolerance and viral persistence, viable offspring persistently infected with pestivirus was not observed. 相似文献

Histochemical alterations in the duodenum of goats experimentally infected with Paramphistomum cervi
Certain histochemical alterations in the different tunics of duodenum in kids were studied 20, 40, 60 and 80 days post-infection (DPI) with Paramphistomum cervi and the results compared with those of uninfected kids. There was a general reduction of polysaccharide complex substances and glycogen at 20 DPI. A marked decrease in polysaccharide complex substances and glycogen at 20 DPI. A marked decrease in polysaccharide complex substances and glycogen was especially discernible in the Brunner's gland and muscularis mucosa 20 DPI. Thereafter, these substances gradually increased and at 80 DPI this decrease was fully replenished. A slight reduction in mucin and protein content of the infected duodenal goblet cells was noticed at 20 DPI. It is suggested that juvenile P. cervi utilize host-tissue polysaccharide complex substances and glycogen for their growth and development during duodenal migration. 相似文献

Although the use of tanniferous plants or condensed tannins as an alternative to anthelmintics to control gastrointestinal nematodes has been largely documented in sheep, studies remain scarce in goats. The objective of this study was therefore to assess the possible impact of condensed tannins in goats infected with adult Haemonchus contortus. Two groups of cull goats were experimentally infected with 10.000 L3 of H. contortus. After 4 weeks, quebracho extracts, representing 5% of the diet DM, were administered for 8 days to one of the two groups. Goats of the second group remained as controls. One week after the end of quebracho administration, the goats were euthanised. Individual egg excretion and pathophysiological parameters were measured weekly during the study. At the end of the study, worm counts were assessed and histological samples from the abomasa were taken to count the numbers of mucosal mast cells, globule leukocytes and eosinophils. The administration of tannins was associated with a significant decrease in egg excretion, which persisted until the end of experiment. This reduction was not associated with any difference in worm number but with a significant decrease in female fecundity. No significant changes in the mucosal density of the three inflammatory cell types were detected between the two groups. These results indicate that the major consequence of tannin consumption in goats is a reduction in worm fecundity and egg output, which does not seem related to significant changes in the local mucosal response. 相似文献

The ability of alphaherpesviruses to infect different ruminant species may have important implications for control/eradication efforts. Serological data indicate that goats may be naturally infected with bovine herpesviruses. To investigate the susceptibility of goats to bovine herpesvirus-5 (BoHV-5), 3-4-month-old kids were inoculated intranasally with each of three Brazilian BoHV-5 isolates (G1, n=8; G2, n=5; G3, n=5). The acute infection was characterized by virus shedding in nasal secretions for up to 14 days (titers up to 10(5.97)TCID(50)/mL), mild respiratory signs and conjunctivitis. All animals seroconverted to BoHV-5, developing virus neutralizing (VN) titers from 4 to 32 to the homologous viruses. At day 60 post inoculation (pi), two animals from each group were euthanized for tissue collection and the remaining goats were submitted to dexamethasone administration (0.4 mg kg(-1) for 5 days). Dexamethasone treatment resulted in virus reactivation in 9 out of 12 animals, as ascertained by virus shedding and/or by increase in VN titers. Virus shedding was detected in 8/12 animals and lasted from 1 to 9 days. Latent viral DNA was detected by PCR in the olfactory bulb and/or trigeminal ganglia of 6/6 goats euthanized at day 60 pi and in 12/12 animals euthanized 40 days post-dexamethasone. These results show that young goats are susceptible to BoHV-5 and may shed virus upon reactivation of latent infection. Thus, it is reasonable to expect that goats raised in close contact with cattle in areas where BoHV-5 is endemic may be infected and therefore should be considered potential reservoirs of the virus. 相似文献
