Xanthomonas axonopodis pv. glycines causes bacterial pustule of soybean, which is a common disease in many soybean-growing areas of the world and is controlled by a single recessive gene (rxp gene) commonly found in many conventional glyphosate-sensitive soybean cultivars. Since glyphosate-tolerant cultivars are commonly planted today, there has been no information about whether these new cultivars have bacterial pustule resistance. The goal of this study was to screen glyphosate-tolerant soybean cultivars for resistance to X. axonopodis pv. glycines. Three experiments were completed to evaluate resistance. In experiment 1, 525 commercial glyphosate-tolerant cultivars from 2001 were inoculated with X. axonopodis pv. glycines strain UIUC-1. Following inoculation, many of the cultivars were resistant (developed no detectable pustule symptoms) although 152 (~29%) developed bacterial pustule. In experiment 2, the aggressiveness of three strains (UIUC-1, UIUC-2, and ATCC 17915) of X. axonopodis pv. glycines were compared on three bacterial pustule-susceptible, glyphosate-tolerant cultivars. One strain (UIUC-1) was less aggressive than the other two (UIUC-2 and ATCC 17915) on all three cultivars examined. In experiment 3, 45 cultivars from 2005 (all different from 2001) were inoculated with X. axonopodis pv. glycines ATCC 17915. A range of disease severities developed with five cultivars (11%) having disease severity ratings as high as or higher than those on a susceptible check cultivar. Overall, these results suggested that resistance to bacterial pustule occurs in glyphosate-tolerant soybean cultivars, but not at 100% frequency, which means bacterial pustule outbreaks could occur when a susceptible cultivar is planted and conditions are conducive for bacterial pustule development.  相似文献

Pathogen development and host responses in wheat spikes of resistant and susceptible cultivars infected by Fusarium culmorum causing Fusarium head blight (FHB), were investigated by means of electron microscopy as well as immunogold labelling techniques. The studies revealed similarities in the infection process and the initial spreading of the pathogen in wheat spikes between resistant and susceptible cultivars. However, the pathogen’s development was obviously more slow in the resistant cultivars as in comparison to a susceptible one. The structural defence reactions such as the formation of thick layered appositions and large papillae were essentially more pronounced in the infected host tissues of the resistant cultivars, than in the susceptible one. β -1,3-glucan was detected in the appositions and papillae. Furthermore, immunogold labelling of lignin demonstrated that there were no differences in the lignin contents of the wheat spikes between susceptible and resistant cultivars regarding the uninoculated healthy tissue, but densities of lignin in host cell walls of the infected wheat spikes differed distinctly between resistant and susceptible cultivars. The lignin content in the cell walls of the infected tissues of the susceptible wheat cultivar increased slightly, while the lignin accumulated intensely in the host cell walls of the infected wheat spikes of the resistant cultivars. These findings indicate that lignin accumulation in the infected wheat spikes may play an important role in resistance to the spreading of the pathogen in the host tissues. Immunogold labelling of the Fusarium toxin DON in the infected lemma showed the same labelling patterns in the host tissues of resistant and susceptible cultivars. However, there were distinct differences in the toxin concentration between the tissues of the susceptible and resistant cultivars. At the early stage of infection, the labelling densities for DON in resistant cultivars were significantly lower than those in the susceptible one. The present study indicates that the FHB resistant cultivars are able to develop active defence reactions during infection and spreading of the pathogen in the host tissues. The lower accumulation of the toxin DON in the tissues of the infected spikes of resistant cultivars which results from the host’s defence mechanisms may allow more intensive defence responses to the pathogen by the host.  相似文献

Common bacterial blight (CBB) in edible beans (Phaseolus vulgaris), incited Xanthomonas campestris pv. phaseoli, reduces bean yields and seed quality. The main objective of this study was to determine resistance to common bacterial blight in bean genotypes. Twenty-two bean genotypes grown in Turkey including common and snap bean cultivars/lines were collected from different parts of Turkey and tested for resistance against to Xanthomonas campestris pv. phaseoli strain MFD-11. All the common and snap bean lines/cultivars tested were moderately susceptible, susceptible or highly susceptible, except AG-7117 which was found resistant to Xanthomonas campestris pv. phaseoli. This is the first report of a resistance source in a common bean line (AG-7117) against Xanthomonas campestris pv. phaseoli.  相似文献

Tomato early blight occurs worldwide and it is prevalent wherever tomatoes are grown. Alternaria solani Sorauer, the causal agent, has been recognized as a serious foliar pathogen of tomato and there are very few cultivars which possess resistance against early blight. Alternaric acid is the major toxin of A. solani. In this study, alternaric acid and fungal culture filtrate were used as an elicitor in NDT-96 (tolerant) and GP-5 (susceptible) tomato varieties in order to study and compare their abilities to induce defense-related enzymes, viz., catalase, peroxidase, β-1,3 glucanase, phenylalanine-ammonia-lyase (PAL), chitinase and polyphenol-oxidase (PPO) along with total phenols, and total soluble proteins. NDT-96 showed a rapid induction of all these pathogenesis-related enzymes except catalase along with total phenols as compared to GP-5 with both the treatments. Differential expression of total soluble proteins revealed higher protein content in NDT-96 as compared with GP-5. A 49.48 kDa protein was observed to be absent in GP-5. In addition, 25 microsatellite markers (SSR) were screened for polymorphisms among the above mentioned two tomato varieties. Of these, SSR 286 revealed a significant polymorphic band of 108 bp in NDT-96.  相似文献

When lower leaves of rice plants were inoculated with powder formulation of a saprophytic strain ofPseudomonas fluorescens, Pfl, upper leaves, in addition to the inoculated lower leaves, showed resistance to the rice bacterial blight pathogenXanthomonas oryzae pv.oryzae. When the leaves were challenge-inoculated withX. oryzae pv.oryzae 4 days afterP. fluorescens application on lower leaves, the disease intensity in upper leaves decreased from 6.7 to 1.1. When rice seeds were treated with the formulation ofP. fluorescens Pfl and sown, 30-day-old seedlings showed resistance toX. oryzae pv.oryzae and the disease intensity decreased from 6.8 to 1.2. The induced resistance was transient; leaves sprayed withP. fluorescens Pfl at 30 days after treatment and leaves of 60-day-old seedlings fromP. fluorescens-treated seeds did not show resistance to the pathogen. In field trials, seed treatment followed by foliar application of the powder formulation ofP. fluorescens Pfl effectively controlled rice bacterial blight and increased the yield. In the induced resistant leaves a sharp increase in lignification and activities of peroxidase, phenylalanine ammonia-lyase and 4-coumarate: CoA ligase was observed when the leaves were challenge-inoculated withX. oryzae pv.oryzae. An approximately threefold increase in lignin content, peroxidase activity and phenylalanine ammonia-lyase activity and a fivefold increase in 4-coumarate: CoA ligase activity were observed 5 days after challenge inoculation withX. oryzae pv.oryzae in rice leaves pretreated withP. fluorescens for 5 days. A similar increase in defense-related activities was not observed in susceptible interactions or inP. fluorescens-treated plants at later stages of interactions when no resistance to the pathogen was observed.  相似文献

The banana Xanthomonas wilt disease (BXW) has threatened the livelihood of millions of farmers in East Africa. Use of resistant varieties is the most cost-effective method of managing this bacterial disease. A reliable and rapid screening method is needed to select resistant banana varieties. An in vitro screening method was developed for early evaluation of Xanthomonas wilt resistance using small tissue culture-grown plantlets. Eight cultivars of banana were screened with sixteen isolates of Xanthomonas campestris pv. musacearum using this method. There were significant differences (P < 0.0001) in susceptibility among the various banana cultivars tested, whereas no significant difference (P = 0.92) in pathogenicity was observed between the pathogen isolates. The cv. Pisang Awak (Kayinja) was found to be highly susceptible and Musa balbisiana resistant. Nakitembe was found to be moderately resistant while cvs Mpologoma, Mbwazirume, Sukali Ndiizi, FHIA-17 and FHIA-25 were susceptible. The susceptibility of these cultivars was further tested in vivo by artificial inoculation of potted plants with similar results. This study shows that an in vitro screening test can serve as a convenient, cheap and rapid screening technique to discriminate BXW-resistant from BXW-susceptible banana cultivars.  相似文献

Role of stilbenes in the resistance of grapevine to powdery mildew   总被引:1,自引:0,他引:1  
Stilbene phytoalexins are identified as defence response in pathogen–grapevine interactions, but little information is available on the role of stilbenes on Erysiphe necator, causal agent of grapevine powdery mildew. Analysis of stilbenes in artificially infected leaf discs from susceptible to highly resistant cultivars was performed and compared to the development of the pathogen. Results indicate that stilbene synthesis is confined in infected cells, penetrated by an appressorium–peg. Stilbene amounts expressed by infection site allow discriminating susceptible and resistant cultivars. Highest viniferins concentrations on resistant cultivars are in correspondence with the observed inhibition of the pathogen growth. The analysis of stilbenes at the infection site and viniferins accumulation in grapevine defence reaction is discussed.  相似文献

During the last two decades bacterial strains associated with necrotic leaf spots of pepper and tomato fruit spots were collected in Serbia. Twenty-eight strains isolated from pepper and six from tomato were characterized. A study of their physiological and pathological characteristics, and fatty acid composition analysis revealed that all of the strains belong to Xanthomonas campestris pv. vesicatoria. Being non-amylolytic and non-pectolytic, pathogenic on pepper but not on tomato, containing lower amounts of fatty acid 15 : 0 ante–iso, the pepper strains were designated as members of the A group of X. campestris pv. vesicatoria. However, the tomato strains hydrolyzed starch and pectate, caused compatible reactions on tomato but not on pepper, had higher percent of 15 : 0 ante–iso fatty acid, and were classified into B phenotypic group and identified as X. vesicatoria. PCR primers were developed which amplified conserved DNA regions related to the hrp genes of different strains of X. campestris pv. vesicatoria associated with pepper and tomato. Restriction analysis of the PCR product resulted in different patterns and enabled grouping of the strains into four groups. When xanthomonads isolated from pepper and tomato in Serbia were analyzed, they clustered into two groups corresponding to the grouping based on their physiological and pathological characteristics. According to the reaction of pepper and tomato differential varieties, the strains from pepper belong to races P7 and P8 and tomato strains belong to the race T2. All strains were sensitive to copper and streptomycin. Advantages and disadvantages of various bacterial spot management practices are discussed.  相似文献

Polyamine and phenolic acid levels as well as activities of some enzymes of their biosynthetic metabolism were examined in two sugarcane (Saccharum officinarum) cultivars differing in susceptibility to leaf scald, a disease caused by the bacterium, Xanthomonas albilineans. Juice obtained from both infected cultivars showed significantly increased levels of free putrescine and ornithine decarboxylase activity. However, the pathogen induced different changes in the two cultivars in subsequent metabolic steps. Whereas acid insoluble conjugated spermidine completely disappeared from the highly susceptible cv. C 439-52, an increase in acid insoluble conjugated polyamines was observed in the moderately susceptible cv. L 55-5. Phenolic acid metabolism also differed in the two cultivars. Since total phenolic acid content and phenylalanine ammonium lyase activity was greater in both cultivars after infection, distribution of phenolic acids between free or conjugated forms diverted into different pathways. The level of susceptibility of the two cultivars is discussed in terms of changes in these compounds.  相似文献

In 1991 serious losses caused byVerticillium wilt were found on two holdings in the Westland glasshouse district in the Netherlands in which theVerticillium resistant tomato cultivars Calypso and Criterium were grown in soilless systems. Isolates from diseased plants were identified asVerticillium albo-atrum.In inoculation experimentsVerticillium resistant tomato cultivars were seriously affected by the new isolates but not by a control isolate. Moneydor, a susceptible cultivar without the Ve gene, was the most seriously diseased by all isolates. A semi-selective medium for isolation of Xanthomonas axonopodis pv. vignicola from cowpea (Vigna unguiculata) plant and soil samples was developed. Twelve carbon and five nitrogen sources were tested with four strains of X. axonopodispv.vignicola, and 25 antibiotics were screened against saprophytes. -cellobiose (10g) was selected as the optimal carbon source. Among the antibiotics, cefazoline inhibited growth of most of the saprophytes with little effect on strains of the pathogen. ,-methionine enhanced growth of X. axonopodispv.vignicola. Boric acid along with ammonium chloride suppressed growth of Pseudomonas fluorescens. The semi-selective medium designated as cefazoline-cellobiose-methionine (CCM) medium contained K2HPO4 1.34g, KH2PO4 0.4g, MgSO4 0.3g, H3BO3 0.2g, NH4Cl 1.0g, -cellobiose 10g, cycloheximide 0.2g, ,-methionine 1.0g, cefazoline 10mg and agar 14g per l of water (pH 7.2). Colonies of X. axonopodispv.vignicola on CCM medium were whitish, round, raised and 0.2–1.8mm in diameter 96h after incubation. CCM medium generally inhibited growth of Pantoea agglomerans, Bacillus subtilis and saprophytes isolated from cowpea leaves. Colonies of Pseudomonas fluorescens and a saprophytic bacterium, which were not completely suppressed by CCM, could be differentiated from X. axonopodispv.vignicola by their smaller size and different color. The CCM medium proved useful for isolation of X. axonopodispv.vignicola from cowpea plant and soil samples. This is the first report of a semi-selective medium developed for detection of X. axonopodispv.vignicola.  相似文献

 研究了玉米灰斑病菌(Cercospora zeae-maydis)侵染前后的4个玉米品种苯丙氨酸解氨酶、过氧化物酶、多酚氧化酶及木质素含量的变化。3种酶比活性在病菌侵染后都发生明显的先增后降的变化,而抗病品种的变化要明显大于感病品种的变化,尤其是苯丙氨酸解氨酶最大增加活性与品种抗病性呈现极显著的相关性。4个品种的木质素含量在病菌侵染的第9 d时增加到最大值,以后略下降,而且抗病品种的木质素含量峰值高于感病品种的峰值。  相似文献

Large-scale cDNA-AFLP profiling identified numerous genes with increased expression during the resistance response of wheat to the Septoria tritici blotch fungus, Mycosphaerella graminicola. To test whether these genes were associated with resistance responses, primers were designed for the 14 that were most strongly up-regulated, and their levels of expression were measured at 12 time points from 0 to 27 days after inoculation (DAI) in two resistant and two susceptible cultivars of wheat by real-time quantitative polymerase chain reaction. None of these genes was expressed constitutively in the resistant wheat cultivars. Instead, infection of wheat by M. graminicola induced changes in expression of each gene in both resistant and susceptible cultivars over time. The four genes chitinase, phenylalanine ammonia lyase, pathogenesis-related protein PR-1, and peroxidase were induced from about 10- to 60-fold at early stages (3 h–1 DAI) during the incompatible interactions but were not expressed at later time points. Nine other genes (ATPase, brassinosteroid-6-oxidase, peptidylprolyl isomerase, peroxidase 2, 40S ribosomal protein, ADP-glucose pyrophosphorylase, putative protease inhibitor, methionine sulfoxide reductase, and an RNase S-like protein precursor) had bimodal patterns with both early (1–3 DAI) and late (12–24 DAI) peaks of expression in at least one of the resistant cultivars, but low if any induction in the two susceptible cultivars. The remaining gene (a serine carboxypeptidase) had a trimodal pattern of expression in the resistant cultivar Tadinia. These results indicate that the resistance response of wheat to M. graminicola is not completed during the first 24 h after contact with the pathogen, as thought previously, but instead can extend into the period from 18 to 24 DAI when fungal growth increases dramatically in compatible interactions. Many of these genes have a possible function in signal transduction or possibly as regulatory elements. Expression of the PR-1 gene at 12 h after inoculation was much higher in resistant compared to susceptible recombinant-inbred lines (RILs) segregating for the Stb4 and Stb8 genes for resistance. Therefore, analysis of gene expression could provide a faster method for separating resistant from susceptible lines in research programs. Significant differential expression patterns of the defense-related genes between the resistant and susceptible wheat cultivars and RILs after inoculation with M. graminicola suggest that these genes may play a major role in the resistance mechanisms of wheat.  相似文献

Bayoud, caused by Fusarium oxysporum f. sp. albedinis (Foa), is the most damaging disease of date palm in Morocco. In the present study we have investigated the effect of jasmonic acid (JA) on two defence-related enzymes, namely peroxidases (POX) and polyphenoloxidases (PPO) in date palm seedlings root. Our data show that exogenous application of JA at a concentration of 50 μM increased the activity of both enzymes. The increase of POX activity in the presence of JA was much more important than that observed following infection with the pathogen. As compared to untreated plants, PPO activity was 2.2 and 1.3 times higher in BSTN and JHL cultivars respectively. In addition, PAGE analysis revealed increased band intensity of the major constitutive isoforms of POX and PPO in both JA-treated and Foa-treated seedlings. Close examination of symptomatic and asymptomatic plants showed that root tissues of symptomatic plants were massively colonized by Foa. Also, disease development in these plants appeared to involve a marked degradation of the host cell walls early during the process of pathogen invasion. In contrast, the presence of Foa in asymptomatic plants induced limited necrotic lesions (hypersensitive-reaction like lesions) that were probably involved in reducing the progression of the pathogen. Together, our findings indicate that JA is capable of enhancing date palm root resistance to infection by Foa via the activation of defence-related enzymes such as PPO and POX.  相似文献

Fifty-five strains of Xanthomonas axonopodis pv. vignicola, isolated from blight and pustule symptoms of cowpea leaves, originating from 11 countries, were characterized for their carbon-source metabolization pattern using the Biolog GN microplate system. Great variation was found between strains according to origin. Dextrin, glycogen and succinamic acid were not used by strains from Benin, Uganda or Thailand, but by all the other strains (excluding two strains from Mozambique), whereas N-acetyl-D-glucosamine and malonic acid were used by the strains from Benin, Uganda and Thailand, but generally not by the other strains. The strains from Benin, Uganda and Thailand, as well as strains from Venezuela, Brazil and Mozambique, clustered separately from the others in multivariate analysis. Nineteen substrates were used by all the strains, 47 not by any strain and 29 only by some strains. No considerable differences were found between strains isolated from blight symptoms and from pustules. Virulence of strains was not related to the metabolic pattern. The Biolog database was not representative of the diversity of X. axonopodis pv. vignicola, since all strains were identified as Xanthomonas campestris, although belonging to eight pathovars, while only eight of nine strains from Benin and both strains from Thailand were identified as X. campestris pv. vignicola. The Biolog system appeared to be useful for characterizing the diversity of X. axonopodis pv. vignicola strains. A set of representative strains based on metabolic and molecular diversity, virulence and geographic origin is suggested for screening for resistant cowpea cultivars.  相似文献

Colletotrichum lagenarium is the casual agent of anthracnose disease of melons. Light and scanning electron microscopy were used to observe the infection process of C. lagenarium on the leaves of two melon cultivars differing in susceptibility. On both cultivars conidia began germinating 12 h after inoculation (hai), forming appressoria directly or at the tips of germ-tubes. By 48 hai appressoria had melanised and direct penetration of host tissue had begun. On the susceptible cultivar, infection vesicles formed within 72 hai and developed thick, knotted primary hyphae within epidermal cells. By 96 hai C. lagenarium produced highly branched secondary hyphae that invaded underlying mesophyll cells. After 96 hai, light brown lesions appeared on the leaves, coincident with cell necrosis and invasion by secondary hyphae. While appressoria formed more quickly on the resistant cultivar, fewer germinated to develop biotrophic primary or invasive necrotrophic secondary hyphae than on the susceptible cultivar. These results confirm that C. lagenarium is a hemibiotrophic pathogen, and that resistance in melons restricts colonisation by inhibiting the development of necrotrophic secondary hyphae.  相似文献

Twenty isolates of fluorescent pseudomonads were evaluated for their ability to control damping-off in tomato (Lycopersicon esculentum) and hot pepper (Capsicum annuum). These isolates were characterized as Pseudomonas fluorescens and Pseudomonas putida. Two isolates, PFATR and KKM 1 belonged to P. putida and the remaining 18 isolates belonged to P. fluorescens. Among these isolates, P. fluorescens isolate Pf1 showed the maximum inhibition of mycelial growth of Pythium aphanidermatum and increased plant growth promotion in tomato and hot pepper. P. fluorescens isolate Pf1 was effective in reducing the damping-off incidence in tomato and hot pepper in greenhouse and field conditions. Isolate Pf1 was further tested for its ability to induce production of defense-related enzymes and chemicals in plants. Earlier and increased activities of phenylalanine ammonia lyase (PAL), peroxidase (PO) and polyphenol oxidase (PPO) were observed in P. fluorescens Pf1 pretreated tomato and hot pepper plants challenged with Pythium aphanidermatum. Moreover, higher accumulation of phenolics was noticed in plants pretreated with P. fluorescens isolate Pf1 challenged with Pythium aphanidermatum. Thus, the present study shows that in addition to direct antagonism and plant growth-promotion, induction of defense-related enzymes involved in the phenyl propanoid pathway collectively contributed to enhance resistance against invasion of Pythium in tomato and hot pepper.  相似文献

The aim of this study was to investigate changes in defence compounds of common bean cultivars with different levels of resistance to the fungus Sclerotinia sclerotiorum and determine the relation of the compounds to pathogen tolerance. The lines were inoculated with the pathogen and assessed for enzymatic and non-enzymatic parameters related to plant defence: peroxidases (POX), polyphenol oxidases (PPO), catalase (CAT), superoxide dismutase (SOD) and ascorbate peroxidase (APX), total soluble phenol and lignin contents. Stem tissue samples were collected from two regions of the plant for biochemical analyses. Stem tissue samples were collected from two regions of the plant for biochemical analyses. In the position one, 5 cm of the stem was collected from the region with necrosis caused by the pathogen, and in the position two, 5 cm of the stem was collected from the end of the position one at the times of 12, 24, 48, 72, 96 and 120 h after inoculation (HAI). Greater lignin and total soluble phenol contents and greater induction of POX and SOD activity in inoculated plants in the region near the inoculation (position one) indicate local activation with later signalling for activation of defence mechanisms in other regions of the plant. The genotype with a greater level of resistance was superior to the susceptible one in regard to lignin production and the activities of POX, APX and SOD defence enzymes. These results suggest that a combination of these defence responses in common bean may contribute to greater plant resistance to the pathogen and that these enzymes have potential use in selection of common bean genotypes.  相似文献

Initial experiment on the reactions of five Japanese cultivars of cucumber toColletotrichum orbiculare infection in the greenhouse revealed that cv Suyo and Gibai were susceptible and moderately susceptible, respectively, while cv Shogoin fushinari and Sagami hanjiro were resistant to infection byC. orbiculare; cv Ochiai fushinari was moderately resistant. The ability of 16 plant growth promoting fungi (some isolates belonged to species ofPhoma and some non-sporulating isolates) isolated from zoysiagrass rhizospheres to induce systemic resistance in the above five cucumber cultivars was tested by growing plants in potting medium infested with barley grain inocula of PGPF in the greenhouse. The second true leaves of 21-day-old plants were challenge inoculated withC. orbiculare and disease assessed. Nine, out of 16 isolates, caused significant reduction of disease caused byC. orbiculare in at least two cultivars.Phoma isolates (GS8-1 and GS8-2) and non-sporulating isolates (GU21-2, GU23-3, and GU24-3) significantly reduced the disease in all the five cultivars. The disease suppression in cucumber was due to the induction of systemic resistance, since the inducer(s) and the pathogen were separated spatially and that the inducer did not colonize aerial portions. The resistance induced by certain isolates in a susceptible cultivar was less than that in a resistant cultivar. Disease suppression caused by isolate GU21-2 was similar to theC. orbiculare induced control in certain cultivars. The average rate of expansion of lesion diameter on leaves due toC. orbiculare was slower due to induction with the selected plant growth promoting fungi compared to the uninduced control plants.
