Levels of bovine IgA, IgG1 and total protein (TP) were determined in serum, saliva, tears and individual quarter lacteal secretions of six Holstein-Friesian cows sampled from six weeks before to four weeks after parturition. Hierarchal analyses of variance indicated significant variations among weeks, cows and quarters of the udder. A precipitous but non proportional drop in the levels of IgA and IgA1 in lacteal secretions occurred at calving. There was a concomitant increase in IgG1, and decrease in IgA, in serum. Correlation studies supported the concept of selective transport of IgG1 from serum to lacteal secretions in regulated amounts independent of serum IgG1 levels. Changes in the IgG1/TP ratio of serum and lacteal secretions supported the idea of a decrease in the selective transport mechanism. Correlation studies and estimations of secretory IgA (SIgA) in serum suggest that serum IgA is derived from IgA synthesized in secretory tissues. Highly significant correlations between IgA and IgG1 levels in all secretions postpartum suggest that local IgA synthesis and either IgG1 transport or local IgG1 synthesis are initiated by the same stimuli. Although some of the variation in the level reported for IgA and IgG1 in secretions resulted from protein dilution, much of the variation represents physiological differences between individual animals and tissues in the same animal. An IgG2/IgG1 ratio approaching that of serum occurred in a mastitic quarter of one cow. IgA was the principal immunoglobulin in saliva and tears, comprised a greater proportion of the immunoglobulin in milk whey than in prepartum lacteal secretions and was a minor immunoglobulin in bovine serum.  相似文献

In the first-litter sows lower serum levels were found for all three Ig classes as compared to multiparous sows. The same was true for IgA in lacteal secretions and in piglet serum during the first days of life, while no differences were found for IgG levels. In contrast to these findings, IgM levels were found to be higher in lacteal secretions of first-litter sows and in piglet serum during the first days of life as compared to their counterparts. From three weeks after birth Igs found in piglet serum mainly originate from de novo synthesis. In this period piglets of first litter sows showed a higher IgA level up to the 6th week of life and higher IgG and IgM levels up to the end of the investigation period. Results are discussed in terms of maternal-neonatal immune regulation, focussing on the apparent suppressive role of maternally-derived IgG on total de novo Ig synthesis by suckling piglets.  相似文献

The concentrations of IgG, IgM and IgA in sera collected from 3855 sows (3208 pregnant and 647 lactating) at a single time point were determined. This experimental design allowed changes in serum immunoglobulin over the reproductive cycle to be studied without bias from seasonal influence. The concentrations of the three immunoglobulins changed independently during the reproductive cycle. Serum levels of IgM and IgG began a progressive postpartal decline during the 14th–17th week of gestation. At the onset of lactation serum IgG levels progressively increased while IgM levels continued to decline, the latter reaching their lowest level during the third week of lactation. In contrast to IgM and IgG, serum IgA levels increased 35% during weeks 14–17 of gestation and continued to increase throughout lactation, reaching their highest serum levels in the third week of lactation; the serum IgA concentration at this time was twice that observed during the first 13 weeks of gestation. Results of these studies allowed the reproductive cycle to be classified into four phases on the basis of serum immunoglobulin concentrations: (1) weeks 1–4 of gestation; (2) weeks 5–13 of gestation; (3) weeks 14–17 of gestation and (4) lactation.  相似文献

A total of 103 ewes from the breeds Black mutt., Texel, Finn. L., Heidschnucke and the crossbreeds Texel x Finn. L. and Black. mutt. x Finn. L. was studied. Blood samples were drawn at days 1, 7, 21 and 42 and milk samples at the 4th, 12th, 24th and 72nd hour after the onset of lactation and subsequently on days 7, 21 and 42. The concentrations of the immunoglobulins IgG1, IgG2, IgM and IgA were assayed in serum and milk. The following results were obtained: 1. The total immunoglobulin contents in the serum was not significantly different between breeds. 2. All ewes showed a rise in serum immunoglobulin concentrations by about one third over the first six weeks of lactation. Between 50-60% of this increase were on the account of IgG1. 3. The serum concentration of IgG1 and IgG1 rose as of the third day, those of IgM as of day 21 after lambing. 4. The rise in serum immunoglobulin concentration continued after the weaning of the lambs. 5. The ratio of IgG1 to IgG1 in ewe serum was 2:1. 6. The immunoglobulin concentration in milk dropped sharply on the first day of lactation, followed by a continuous, more gradual decrease over the entire course of lactation. A terminal rise, as observed in sows, could not be detected. 7. The ratio of IgG1:IgG2 : IgM : IgA in the whey changed from 85 : 1 : 12 : 2 on day one to 70 : 7 : 12 : 11 on the last day of lactation. 8. While characteristic trends in immunoglobulin patterns in the sera of ewes over the course of lactation are clearly discernible, it is not possible to denote "normal" values.  相似文献

Rotavirus antibodies were demonstrated in lacteal secretions and sera of 20 parturient sows and in sera of their newborn by an enzyme-linked immunosorbent assay blocking technique, using bovine rotavirus cell culture antigen and monospecific antibody to bovine rotavirus. Antibodies to rotavirus occur in the 3 immunoglobulin (Ig) classes IgM, IgA, and IgG in lacteal secretions. High and long-persisting antibody activity was mainly associated with the IgA class. The IgM and IgG decreased to undetectable concentrations in most sows during the 14-day investigation period. Serum antibodies of newborn pigs nursing their dams also decreased rapidly during this time. The heterologous enzyme-linked immunosorbent assay blocking technique was a reliable and rapid procedure for the demonstration of rotavirus antibodies.  相似文献

Lavage techniques were used to obtain secretions from the nasal cavity, trachea and bronchi of conscious horses. The techniques, which utilised fibreoptic endoscopy for recovery of tracheal and bronchial secretions, were well tolerated by the horses. The recovery rates of the lavaged fluids were acceptable, but were lowest for bronchial secretions, and there was minimal contamination by blood. The fluids were analysed for IgG and IgM by single radial immunodiffusion, and for IgA and albumin by rocket immunoelectrophoresis. Relative to albumin there was significantly more IgA and IgM, and significantly less IgG, in the nasal cavity than the trachea. IgA and IgM levels were greatest in the nasal cavity and decreased progressively to the bronchi, whereas IgG levels showed the reverse trend. The immunoglobulin: albumin ratios of secretions taken from many levels of the tract were significantly higher than those of serum, suggesting local production of immunoglobulin in the respiratory tract.  相似文献

IgG, IgA, IgM and albumin concentrations were measured in serum, follicular fluid and oviductal, uterine and intestinal secretions of the horse. Follicular protein concentrations were found to be dependent on serum concentration and molecular size. Of the immunoglobulins only IgG was detectable in oviductal secretions, but IgG:albumin ratios did not differ significantly from those in serum. IgG, IgA and IgM were measured in uterine secretions, with IgG predominant. Serum transudation into uterine secretions was minimal. In intestinal secretions, IgA levels were slightly higher than IgG, with albumin and IgM at low levels. In five mares with histories of chronic metritis, IgG, IgA and albumin concentrations were significantly elevated in uterine secretions.  相似文献

Methods of collecting sputum and tracheobronchial washings are described.Proteins in normal porcine tracheobronchial secretions are studied by immunochemical qualitative and quantitative methods. Among the immunoglobulins, IgA, IgG and IgM are identified. Immunoelectrophoretic analysis of sputum shows albumin, transferrin and in the α-region 2 components not found in serum. The predominant immunoglobulin in tracheobronchial secretions is IgA, which accounted for about 85 % of the total immunoglobulin concentration in sputum and 61 % of the immunoglobulin concentration in tracheobronchial washings.  相似文献

Samples of pharyngeal fluid and serum were collected from cattle after exposure to live foot-and-mouth disease (FMD) virus (with or without prior vaccination) or after subcutaneous vaccination with inactivated virus. The pharyngeal fluid samples were examined for FMD neutralising activity and specific anti-FMD IgG, IgM and IgA antibodies. The neutralising activity of the serum was also monitored. A peak of neutralising activity which occurred in the pharyngeal fluid of unvaccinated cattle seven days after virus exposure corresponded to a rise in specific IgM and IgA antibodies. This peak appeared to be due to serum and tissue fluid escaping from the damaged mucosa during the acute inflammatory phase of infection. At later stages (20 to 60 days after virus exposure) the pharyngeal fluid neutralising activity corresponded to a rise in specific IgA antibodies, suggesting that active local antibody production was taking place. The pharyngeal fluid neutralising activity detected after revaccination with oil emulsion or aqueous vaccines, without exposure to live virus, corresponded to a rise in specific IgG and IgM antibody levels and this may have been due to serum transudation.  相似文献

Fifty-four neonatal pigs were allotted to 4 groups and reared in an electrically controlled automatic feeding device (autosow). Each group was reared on a different pool of bovine colostrum: fresh, stored 1 month, stored 6 months, and stored 8 years. Bovine and porcine immunoglobulins in the sera of these pigs, and in a group of conventionally reared pigs, were measured periodically during the first 42 days after birth. The maximal concentration of absorbed bovine immunoglobulin was reached between 12 and 18 hours and equaled or exceeded the amount of porcine immunoglobulin absorbed by the conventionally reared pigs. Large differences in the concentrations of the bovine immunoglobulin isotypes among the various pools of colostrum were positively correlated with concentration of these isotypes in the sera of the neonatal pigs fed these pools. Relative to their concentrations in colostrum, approximately 41% of the IgG1, 55% of the IgG2, 29% of the IgM, and 67% of the IgA was absorbed. The IgA was absorbed the best and IgM was least absorbed. Significant trends or differences in absorption were not observed among groups. Neonatal pigs given fresh colostrum, which had a higher fat content, had significantly more weight gain (P less than 0.05). This occurred, despite the fact that the fresh colostrum had the lowest concentration of bovine immunoglobulin. Serum half-lives for bovine IgG1 and IgG2 were significantly less than for porcine IgG (P less than 0.05), whereas the half-lives for bovine and porcine IgM and IgA were similar. De novo-synthesized immunoglobulins were detectable in serum after 6 days; IgM concentrations reached a maximum at 15 days in neonatal pigs given stored, but not fresh, colostrum.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The influence of age and breed on the concentrations of IgG, IgA and IgM in the sera of sows throughout the reproductive cycle was investigated in 4137 sows which had had 0 – 20 gestations and representing three breeds: Swedish Landrace, German Landrace and L-12 (=Swedish Landrace × Large White). Data revealed an increase in total immunoglobulins (Ig), IgM and IgG serum levels with increasing gestation number; the latter contributed > 80% to total Ig levels. IgG was significantly increased up to the fourth gestation, whereas IgM showed a significant increase only to the third gestation. IgA showed only minor differences. Age-dependent increases in serum IgM were ascribed to the incraased probability of antigenic exposure during suckling, while failure to observe this change in serum IgA was ascribed to its role as a local Ig. Breed differences were observed to be significant for all three Ig's. It is concluded that establishment of group norms for serum Ig's should consider age and breed difference as well as stage of gestation or lactation.  相似文献

Immunoglobulin concentrations (IgG, IgM, and IgA) in bovine serum, follicular fluid, and uterine and vaginal secretions were determined. The specificities of IgG, IgM, and IgA for virus-neutralizing antibody against bovine viral diarrhea (BVD) and infectious bovine rhinotracheitis (IBR) viruses were also examined. High concentrations of IgG were present in both serum and follicular fluid. The IgG, IgM, and IgA concentrations were low in uterine and vaginal secretions. There was more IgG in the uterus during estrus than at any other time. Virus-neutralizing antibodies against BVD and IBR in serum of cows were mainly the IgG class. There was positive correlation between serum and follicular fluid virus-neutralizing antibody titers fro BVD and IBR. These antibodies may provide some protection for recently ovulated ova.  相似文献

Immunoglobulin (Ig) concentrations in serum and in nasal secretions were correlated with pneumonia and diarrhea during the first 12 weeks of life in 56 calves. The peak onset of pneumonia occurred between 2 and 4 weeks of age when the calves' serum IgG1, IgG2, and IgA concentrations were lowest. As IgG2 concentrations increased, fewer calves developed pneumonia. Peak onset of pneumonia was also correlated with the lowest IgG and IgA concentrations in the calves' nasal secretions. Most calves developed pneumonia when serum concentrations of IgG1 were less than 1.5 g/dl, IgG2 less than 0.3 g/dl, IgA less than 0.1 g/dl, and IgM less than 0.2 g/dl and when the combined IgG and IgA values in nasal secretions were less than 0.2 mg of Ig/mg of protein. In study A, diarrhea preceded pneumonia in 63% of 56 calves. In study B, 38% of 23 calves had diarrhea and/or hemorrhagic feces before pneumonia. Seemingly, there was a relationship between diarrhea and pneumonia. Furthermore, pneumonia occurred at or just after the time when IgG1, IgG2, and IgA concentrations in serum and the combined IgG and IgA concentrations in nasal secretions were lowest. Pneumonia is a common disease of calves between 1 and 5 months of age, a period coinciding with the usual low point in serum immunoglobulin (Ig) concentrations due to catabolism of passively acquired antibodies. Calves that absorb less than adequate amounts of Ig may be susceptible to pneumonia at approximately 2 months of age, when serum Ig concentrations would be lowest.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Turkey immunoglobulin-A (IgA) was isolated from bile, intestinal secretions, and serum by affinity chromatography using monospecific anti-turkey IgA coupled to CNBr-activated Sepharose. The isolated immunoglobulin was antigenically distinct from IgM and IgG. The purity of IgA was demonstrated by immunodiffusion, immunoelectrophoresis, and analytical ultracentrifugation. The predominant forms of polymeric IgA in bile and intestinal secretions had respective So20w values of 16.1 and 15.2. Larger polymers (25-26S) were also present. Two molecular forms (8.5S and 17S) were found in serum. The 8.5S peak was higher than the 17S, indicating a greater concentration of 8.5 S.  相似文献

Responses to prepartum injection of sows with Se and vitamin E (E) were evaluated by determining immunoglobulin (IgA, IgM, IgG) levels in the colostrum and serum of the sows and the serum of their offspring. Fifty-four sows (40 multiparous, 14 primiparous) receiving diets adequate in E and Se according to current NRC (1988) standards were randomly allotted to four treatment groups in which a single i.m. injection of saline (controls), 5 mg of Se, 1,000 IU of E, or both Se and E were given on d 100 of gestation. Sows were bled prior to and 7 d after injection, at farrowing and on d 14 and 28 of lactation. Colostral samples were collected at the initiation of farrowing. Pigs were bled 20 h postpartum and at 14 and 28 d of age. Major immunoglobulin changes in the serum of the sows due to treatment were not seen prior to parturition. Injections of Se and(or) E resulted in higher colostral IgM levels (8.4, 10.7, 9.8 and 9.6 mg/ml, respectively), but only the response from Se was significant (P less than .05). Concentrations of colostral IgA or IgG were not affected by treatment (P greater than .30). Compared with controls, all three treatments increased (P less than .10) IgM concentrations in serum from pigs at birth (28.3, 33.3, 36.0 and 33.5 mg/ml, respectively), whereas IgA and IgG concentrations were not affected (P greater than .30).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Enteric cryptosporidiosis was studied in colostrum-deprived lambs each infected at five days old with 10(6) oocysts. The prepatent period was three to five days and faecal oocyst concentration fell below detectable levels by day 16 after infection. Specific IgA, the only isotype detected by immunofluorescent assay in faecal extracts from infected lambs, was first evident on day 10 and titres continued to rise until day 16 of infection in association with declining oocyst output. Specific IgM and IgG antibodies were first detected in serum seven days after infection. No specific antibody was detected in uninfected control lambs. Immunoblotting methods showed that serum antibody and faecal IgA had similar profiles of antigen recognition. Antigens with approximate molecular weights of 180,000, 23,000 and 15,000 were consistent features on immunoblots performed with convalescent sera and faecal extracts. The results suggest that specific IgA in intestinal secretions has an important role in immunity to cryptosporidiosis.  相似文献

The sera of 188 lambs from seven breed groups were analyzed for the concentrations of IgG1, IgG2, IgM and IgA by radial immunodiffusion using monospecific antibodies. From each lamb, 14 blood samples were drawn before and 5 samples after weaning. The following results were obtained: 1. Immunoglobulins could not be detected in sera drawn before the first intake of colostrum. 2. In normally suckling lambs, the peak concentrations of maternal immunoglobulins are attained at 0-18 hrs after birth. They can be assessed in a single blood sample drawn between 18 and 24 hrs. 3. The half-life times of maternal immunoglobulin in lamb sera are 11 days for IgG1, 7 days for IgG2, 6 days for IgM and 18 hours for IgA. 4. The absolute peak heights relate to the amounts of colostrum ingested before 12-18 hrs after birth. 5. The decline of maternal immunoglobulins in lamb sera over-laps with the onset of lamb immunoglobulin synthesis. Renewed rises of concentrations are observed for IgG2 after week 2, for IgM after week 3 and for IgG1 after week 7. The concentrations of IgA remain at the low levels characteristic for the serum of grown sheep. 6. The role of immunoglobulin synthesis in suckling lambs is only briefly and to a small extent reduced after weaning.  相似文献

Samples of urine and serum from 45 newborn rottweiler puppies from six litters, and milk from their mothers, were taken 24, 48 and 72 hours and seven and 14 days after birth. Urine total protein and creatinine concentrations were determined and the ratios calculated. The immunoglobulin (Ig) concentrations of IgG, IgM and IgA in urine, serum and milk were determined with a commercially available elisa kit. The concentration of total protein in urine decreased from 1.64 to 0.29 mg/ml, and it and the ratio of total protein to creatinine in the urine of the neonatal puppies exceeded the normal values for adult dogs, but all the puppies developed normally. The average concentration of IgG in urine decreased from 0.0035 to 0.0003 mg/ml, that of IgA from 0.0035 to 0.0002 mg/ml and that of IgM from 0.0006 mg/ml to undetectable levels after two weeks. After two weeks, 47 per cent of the puppies had measurable levels of IgA and 70.6 per cent had measurable levels of IgG, but none of them had measurable levels of IgM.  相似文献
