Stress levels in organisms provide a rapid measure for assessing population health. Handling and capture stress, however, cause error in blood measures, so this method is rapidly being replaced by assessing levels of stress metabolites in faeces. This eliminates the source of error because there is a lag period between stress perception and the resultant stress metabolite accumulation within faeces. This lag period is correlated with specific intestinal passage time, a measure that can vary greatly between taxa, particularly amongst ectotherms. Due to two deleterious consequences associated with extended exposure of the metabolites to the intestinal environment, species that exhibit long and variable intestinal passage times are not good candidates for metabolite studies. We measured gut and intestinal passage times in Trachylepis margaritifer to ascertain whether it would be an appropriate candidate for stress metabolite studies. We first tested if barium sulphate in the meal had an effect on gut passage time at three ambient temperatures (25, 27 and 32 °C). Barium sulphate had no effect; however, temperature had a significant effect with an unexpected pattern: gut passage time was fastest at 32 °C but was slower at 27 °C than at 25 °C. We then used X‐ray technology and barium sulphate‐loaded meals to measure gut and intestinal passage times at 25 and 27 °C. This allowed us to observe which parts of the digestive process were responsible for increased passage times at 27 °C: the faster passage time at 25 °C was due to faster intestinal passage time; there was no difference in gastric emptying time. We assess the species to be a suitable candidate for studies using faeces to measure stress. It is imperative however, that the effect of temperature on passage rates is known and taken into account in such studies.  相似文献

The specific oxygen consumption rate (VO 2 IMb) of Labeo capensis, the freshwater mudsucker, was determined for small and large fish at winter (8°C) and summer (23°C) temperatures. VO JMb was also determined during hypoxic conditions of the experimental water. It was found that VO JMb does not differ substantially compared with other active fish. The VO 4Mb for a fish with a body mass of 250 g is 0,053 ml 0 2 per gram wet body mass per hour for fish acclimated at 8°C and 0,127 ml 0 2 per gram per hour for fish acclimated at 23°C. The calculated mass exponent, b, is 0,71 at 23°C and 0,68 at 8°C. Hypoxic conditions are relatively well tolerated especially by 23°C- adapted fish.  相似文献

Digestion rates were determined for natural prey items offered to each of seven species of sea anemones found at Wooley’s Pool in False Bay and to the single species (Bunodactis reynaudi) recorded at Biouberg on the Atlantic coast of the south-western Cape, South Africa.This was done by examining prey items removed from the coelenterons at regular intervals after feeding and assessing their degree of digestion according to a predetermined scale. There was considerable variation in the gut retention times between different anemone species offered the same prey types, with Actinia equina consistently showing the shortest gut retention times (12 h for amphipods, 15 h for pelecypods and 23 h for isopods). Of the various prey categories tested, amphipods tended to be the most rapidly digested group, while molluscs and echinoderms usually remained in the coelenteron the longest. Mean gut retention times in B. reynaudi, the only species found both in False Bay (17°C) and on the cold west coast (12°C) were markedly longer (72 vs 60 h and 43 vs 30 h for pelecypods and gastropods respectively) at the lower temperature.  相似文献

The influence of specific thermal energy (STE) applications on extruder preconditioner was evaluated in a dry food for cats. In the first study, six STE applications were tested with mass temperatures of 45°C, 55°C, 65°C, 75°C, 85°C and 95°C. The extrusion parameters, starch gelatinization and kibble formation were evaluated. Diets were given to cats to evaluate digestibility, faecal characteristics and palatability. In the second experiment, three treatments were compared: low STE—a preconditioner temperature of 45°C (LSTE); high STE—a preconditioner temperature of 95°C (HSTE); high STE (preconditioner temperature of 95°C) combined with an increase in the mass flow rate to obtain a motor amperage similar to that of the LSTE (HSTEflow). Data were analysed by polynomial contrasts (Experiment 1) or Tukey's test (Experiment 2; p < 0.05). An increase in STE reduced motor amperage, mass pressure and specific mechanical energy (SME) implementation (p < 0.001) and increased total specific energy (TSE) and mass temperature (p < 0.01). The increase in STE induced greater kibble expansion and starch gelatinization (p < 0.001). No changes in apparent nutrient digestibility or faeces characteristics were observed (p > 0.05). Lower STE and starch gelatinization induced higher butyrate and total volatile fatty acid (VFA) contents in faeces (p < 0.01). Cats showed greatest preference for the formulation with the highest STE (p < 0.01). In the second experiment, when the motor amperage was increased in the HSTEflow treatment to a value similar to that of the LSTE, the mass flow rate increased 40%, and the electric energy consumption remained unchanged (p < 0.001), with gains observed for efficiency and cost. In conclusion, STE application is important for sufficient TSE implementation, enhancing kibble expansion, starch gelatinization, cat preferences for food, extruder productivity and reducing SME application. Foods with lower starch gelatinization lead to increased VFA in faeces, with possible implications for gut health.  相似文献

Abstract

The effect of water temperature on the progress of experimentally induced Cytophaga psychrophila infection was investigated in juveniles of coho salmon Oncorhynchus kisutch, chinook salmon O. tshawytscha, and rainbow trout O. mykiss (formerly Salmo gairdneri). A virulent strain of C. psychrophila was administered to fish by subcutaneous injection. Infected fish were held in tanks containing pathogen-free well water at temperatures ranging from 3 to 23°C. Mean times from infection to death of the fish were shortest at 12–15°C, which were the temperatures associated with the shortest time for doubling the population of this bacterium in vitro. Juvenile steelhead (anadromous rainbow trout) injected with viable C. psychrophila cells and held in 22°C water did not become diseased.  相似文献

Studies were conducted to examine the possibility of preserving slaughterhouse‐derived buffalo ovaries at 4°C for 0 (control), 12 and 24 h to maintain the developmental competence of the oocytes (experiment 1), to assess the effect of incubation temperature during oocyte maturation on rates of in vitro maturation (IVM) and in vitro fertilization (IVF) of buffalo oocytes and embryo development (experiment 2), and to examine the effect of storage at 25°C for 0 (control), 4 and 8 h of frozen–thawed buffalo sperm and BO and H‐TALP as sperm processing and fertilization media on cleavage and embryo development in vitro of buffalo oocytes (experiment 3) in order to optimize the IVF technology in buffalo. Results suggested that storage of ovaries at 4°C for 12 or 24 h significantly (p < 0.05) reduced the developmental potential of oocytes. Incubation temperatures during the IVM influenced the fertilization rate but had no significant effect on maturation and subsequent embryo development. The incubation temperature of 38.5°C during IVM was found to be optimum for embryo production in vitro. Storage of frozen–thawed sperm at 25°C for 8 h significantly (p < 0.05) decreased its ability to cleave the oocytes. Sperm processed in BO medium had significantly (p < 0.05) higher ability to cleave the oocytes than the H‐TALP medium.  相似文献

Mousebirds (Coliiformes) exhibit well-developed communal roosting behaviour as well as a pronounced capacity for facultative hypothermic responses. We recorded body temperature (Tb)) in speckled mousebirds (Colius striatus) under semi-natural conditions in outdoor aviaries, and examined interactions between behavioural and metabolic thermoregulation by experimentally manipulating food availability and communal roosting behaviour. When food was available ad libitum, mousebirds roosting in a cluster maintained approximately constant rest-phase Tb, with 32°C < Tb < 42°C. By contrast, rest-phase Tb in single mousebirds decreased at 0.5°C/hr and minimum rest-phase Tb was significantly lower than when clustering. When food availability was restricted, the mousebirds exhibited facultative hypothermic responses that were less pronounced in clustering groups (minimum rest-phase Tb = 33.3°C, circadian amplitude of Tb = 9.5°C) compared to single birds (minimum rest-phase Tb = 30.7°C, circadian amplitude of Tb = 11.8°C). When clustering, rest-phase Tb was highly synchronized among individuals. Our data reveal that communal roosting has profound consequences for rest-phase thermoregulation in C. striatus, and provide further insights into the potential role of physiological constraints in the evolution of avian sociality.  相似文献

To study reproduction and embryogenesis, Pimelodus maculatus specimens were kept in captivity and captured bimonthly during 1 year. Gonads samples (211 specimens) were collected and submitted to routine histological techniques. Pimelodus maculatus prepared to reproduce when water temperature was high, and even reached advanced maturation but did not spawn in captivity. Spent fish gonads were not documented, and atretic follicles were frequent (60%) in late maturation females. When then submitted to hypophysation, 70% of the females responded positively to hormonal treatment. Oocyte extrusion occurred 8 h after a second hormonal injection at 26°C. The fertilisation rate was 65.1 ± 9.2% at 24°C. Recently spawned oocytes of P. maculatus were spherical, non‐adhesive, yellow in colour, with an average diameter of 1113.92 ± 37.02 μm and covered by a thick gelatinous layer. Blastopore closure occurred 7 h and 30 min after fertilisation. Embryonic development was completed within 18 h after fertilisation. The results of this work provide important knowledge for the handling and cultivation of not only P. maculatus, but other species of potential value for fish culture.  相似文献

Pelibuey and Suffolk sheep were compared as to their capacity to regulate body temperature under environmental hyperthermia by measuring their differences in cellular response to heat stress (HS). In a first experiment, seven Pelibuey and seven Suffolk ewes were kept in a climatic chamber for 6 h daily during 10 days (temperatures within the 18 to 39.5 °C range). As chamber temperature rose, sheep rectal temperature increased in both groups, but to a lesser extent in Pelibuey (0.3 °C) than in Suffolk sheep (0.7 °C) (P?<?0.05). In a second experiment, cellular viability was assessed using cultured blood mononuclear cells from 15 Pelibuey and 15 Suffolk sheep. They were incubated at 37 °C for 24 h (control) or 43 °C for 6 h followed by 18 h at 37 °C (HS). In a third experiment, another blood mononuclear cells culture from eight Pelibuey and eight Suffolk sheep was kept at 37 °C for 15 h; these were subsequently cultured for 6 h at 37 °C (controls) or 43 °C (HS). Next, HSP-70 concentration was determined. HS reduced the percentage of viable cells to a greater extent in Suffolk [37 °C (73.7 %) vs. 43 °C (61.9 %); P?<?0.05] than in Pelibuey sheep [37 °C (74.9 %) vs. 43 °C (66.7 %); P?>?0.05]. HS significantly increased HSP-70 average concentrations for both breeds at 43 °C. A significant effect was observed for the breed by temperature interaction (P?<?0.05) caused by a greater difference between Pelibuey and Suffolk at 43 °C (2.85 vs. 0.53 ng/mL, respectively; P?<?0.05) than at 37 °C (0.05 vs. 0.03 ng/mL, respectively; P?>?0.05). In conclusion, Pelibuey sheep show more effective body temperature regulation under conditions of environmental hyperthermia. Also, cell viability after HS was higher in Pelibuey than in Suffolk, an effect that could be mediated by an HSP-70-related mechanism.  相似文献

Fourteen strains of Bdellovibrio‐like organisms were isolated from cultured fish ponds using Aeromonas hydrophila J‐1 as host, one of them formed large plaques after 48 h of incubation at 28°C on a double layer plate, designated as Bdellovibrio C‐1. The Bdellovibrio was confirmed by electron microscopy and PCR amplification of Bdellovibrio‐specific 16S rDNA. The optimum temperature for the growth of BdC‐1 was between 15–37°C and with optimal activity at temperatures of 25–30°C. The ability of BdC‐1 to lyse A. hydrophila was similar in the pH range of 6.5 to 8.5. It can lyse 23 Gram‐negative bacterial strains comprising three genera of fish pathogens and one strain of Escherichia coli but cannot lyse Gram‐positive bacteria such as Bacillus subtillis and Staphylococcus aureus. Immersion of fish in water containing different concentrations of BdC‐1 was used in protection against an experimental infection of A. hydrophila J‐1. Results show that the mortality of groups immersed with BdC‐1 was lower than the group without BdC‐1. These results suggest that it may be possible to use Bdellovibrio to control the disease caused by A. hydrophila.  相似文献

Abstract

The effects of temperature on experimental transmission of dermal sarcoma, a spontaneous tumor of adult walleyes Stizostedion vitreum to fingerling walleyes was determined. Test temperatures were 10, 15, and 20°C. Three-month-old walleyes were inoculated intramuscularly with a cell-free filtrate from a dermal sarcoma collected from an adult walleye in the spring of the year. Tumors were first grossly visible in fish held at 15°C at 8 weeks postinoculation. At 12 weeks postinoculation, all fish were euthanized and examined for presence of tumors. Tumor transmission was most successful at 15°C, followed by that at 20°C; many fish maintained at these temperatures had grossly visible tumors. Although the majority offish held at 10°C also had tumors, tumors in these fish developed to a lesser degree than those observed in fish held at the higher temperatures.  相似文献

In order to conserve and culture the cichlid fish Archocentrus nigrofasciatus, more information about its reproductive biology and its larval behavior and morphogenesis is necessary. Currently, temperatures ranging from 21 to 27 °C are used in ornamental aquaculture hatcheries. Lower temperatures are preferred to reduce the costs of water heating, and 23 °C is usually the selected temperature. However, there is limited information on culturing protocols for ornamental species and most of the information generated on this topic remains scarce. Thus, the present study examines the morphological development of Archocentrus nigrofasciatus during the yolk‐sac period up to the age of 100 h post‐hatching in relation to 2 temperature regimes used in ornamental aquaculture: a temperature of 27 °C (thermal optimum) and a decreased temperature of 23 °C (thermal tolerance). The results of this study suggest that the 27 °C temperature generates intense morphological changes in yolk‐sac development in a shorter period. This has advantages as it reduces the time of yolk‐sac larval development, and, thus, minimizes the transition phase to exogenous feeding and maximizes the efficiency at which yolk is converted into body tissues. The present paper provides necessary information to produce freshwater ornamental fish with better practices so as to increase larval survival and capitalize on time for growth.  相似文献

The gut contents of 313 specimens of Hyporhamphus knysnaensis from Rondevlei were analysed. The total lengths of the collected fish varied between 5,1 and 21,8 cm. It was found that the annual cycle in the occurrence of submerged macrophytes in Rondevlei appears to directly influence the feeding of H. knysnaensis of 9 to 17 cm long. This length- group fed mainly on animal material (mostly amphipods and isopods) in winter and the first half of spring while submerged macrophytes were scarce. After the submerged macrophytes became more abundant, however, the 9 to 17-cm length-group changed to a predominantly plant diet (consisting of Ruppia spiralis and Potamogeton pectinatus). H. knysnaensis between approximately 5 and 9 cm long appeared in summer and fed mainly on animal material (mostly amphipods and isopods). Of the 47 specimens collected in this length-group between December 1978 and March 1979, all had animal material in their gut contents. It was the dominant food in 94% of the specimens and formed 90% of the gut contents. In general, therefore, H. knysnaensis can be considered an omnivore.  相似文献

Oxygen consumption of individual long-fingered bats, Miniopterus schreibersii, was measured at air temperatures (Tr) between 2 and 42°C and that of clusters of four and six bats between 5 and 30°C. BMR of individuals was estimated to be 2.29 ml O2 g 1 h-1 between 34 and about 38°C. M. schreibersii showed two different responses to Ta, either maintaining body temperature (Tb) by increasing metabolic rate, or allowing Tb to fall close to ambient temperature and conserving energy (torpor). Euthermic clusters of four and six bats had lower rates of mass-specific oxygen consumption and lower thermal conductances than individuals at equivalent temperatures and torpid clusters maintained a greater temperature differential between Tb and Ta. Lowest rates of metabolism were measured for euthermic bats at air temperatures higher than those available to them in their natural roost in summer, so typical roost temperatures result in metabolic rates that are about 2 4–3.1 times estimated BMR. Roost temperatures of M. schreibersii in South Africa are substantially higher in winter than are generally accepted as being suitable for hibernation but these do not substantially affect torpid metabolic rates, which are low and independent of ambient temperature below 22°C. Clearly at least some species of insectivorous bats are capable of hibernating at temperatures generally considered to be too warm for this activity.  相似文献

The pharmacokinetics and tissue residues of moroxydine hydrochloride were studied in gibel carp at water temperature of 15 and 25 °C. Samples (blood, skin, muscle, liver, and kidney) were collected over 10 days after the treatment and analyzed by high‐performance liquid chromatography with an ultraviolet detector. The results indicated that the influence of water temperature on the metabolism of the drug was significant. The plasma concentration–time data of moroxydine hydrochloride conformed to single‐compartment open model at the two water temperatures. There were higher absorption rate (t1/2ka) and longer elimination half‐lives (t1/2ke) at 15 °C (4.29 and 15.87 h, respectively) compared with those at 25 °C (3.02 and 4.22 h, respectively). The maximum plasma concentration (Cmax) and the time‐point of maximum plasma concentration (Tp) were 2.98 μg/mL and 10.35 h at 15 °C and 3.12 μg/mL and 4.03 h at 25 °C, respectively. The distribution volume (Vd/F) of moroxydine hydrochloride was estimated to be 4.55 L/kg at 15 °C and 2.89 L/kg at 25 °C. The total body clearance (CLb) of moroxydine hydrochloride was determined to be 0.25 and 0.49 L/(h·kg) at 15 °C and 25 °C, respectively; the areas under the concentration–time curve were 75.89 μg·h/mL at 15 °C and 42.33 μg·h/mL at 25 °C. The depletion of moroxydine hydrochloride in gibel carp was slower with a longer half‐life period, especially at lower water temperature that was tested.  相似文献

1. Four rearing temperature regimes (15, 20, 25, 30 °C) and three feeding schedules (ad libitum, restricting to the ad libitum intake of the 12th week and feeding 70% of this rate) were carried out with layer replacement pullets to 170 d of age. From this age, during lay, birds were kept at either 21 °C or on a 24‐h cycle of 21 for 18 h and 28 °C for the 6 h before lights out. Both a white and a brown egg‐laying strain were used.

2. Body weight at 169 d of age varied from, on average, 1409 g (15 °C, ad libitum) to 943 g (30 °C, 70% schedule) for the white strain and 1947 to 1250 g for the same treatments respectively for the brown strain. Sexual maturity was considerably delayed by the 70% feeding schedule, only slightly by rearing at 30 °C.

3. Rearing at 30 °C tended to depress subsequent egg output. The 70% feeding schedule at least maintained egg output compared with birds fed ad libitum in rearing.

4. There was a highly significant effect of temperature treatment during lay on food intake. The reduction in food intake due to the 21–28 °C cycle, however, appeared small.  相似文献

The activity of the ant Camponotus detritus was studied in the dunes of the central Namib Desert. Activity was divided into two components: transit activity and honeydew collection. Temperature governed both forms, but light controlled the initiation and termination of transit activity, which was bimodal in warm conditions and unimodal in cool conditions. No transit activity occurred at night. Ants were active over a surface temperature range of 10-55°C. Owing to a steep thermal gradient above the sand surface during the day in summer, the temperatures which the ants bodies experienced at 5 mm above the sand were 10-15°C lower than surface temperatures; well within the ants' physiological limits. Winds of about 16 km h-1 inhibited activity which ceased when wind speeds reached about 25 km h-1. In summer, the number of ants collecting honeydew was negatively correlated with air temperature while in winter it was positively correlated.  相似文献

Abstract

The digenean Bolbophorus damnificus infects commercial channel catfish Ictalurus punctatus, causing mortality, lower feed consumption, and reduced growth in surviving fish. The purpose of this study was to determine the length of time for which B. damnificus prodiplostomulum metacercariae (juvenile trematode stage that infects fish) would remain viable (parasite appearing to be intact or exhibiting movement) in channel catfish. Fish (n = 210) were infected with molecularly confirmed B. damnificus cercariae harvested from naturally infected marsh rams-horn snails Planorbella trivolvis. During the first sampling (at 20 d postinfection), 8.3 ± 3.6 metacercariae/fish (mean ± SD) were found in the host muscle and visceral organs. The channel catfish were then acclimated to a water temperature of either 18°C or 28°C. After 11 months, 6.8 ± 3.5 and 5.9 ± 3.0 metacercariae/fish were found in groups held at 18°C and 28°C, respectively. The mean number of parasites per fish did not significantly differ between fish held at the two temperatures and did not significantly decline over time at either temperature. Fish examined from 13 to 30 months postinfection all contained viable metacercariae that were morphologically and molecularly identified as B. damnificus. At 18 months, 12 metacercariae (of which 11 were intact and 10 displayed movement) were found in the one fish sampled; at 30 months, the last fish sampled contained three intact metacercariae (one displayed slight movement). Our results indicate that B. damnificus metacercariae can remain viable in channel catfish for at least an 18–30-month production cycle during which they have the potential to affect fish growth; in addition, infected fish may serve as intermediate hosts for these metacercariae for at least 2.5 years postinfection.

Abstract

Upper and lower temperature tolerance limits, reported as temperatures lethal to 50% of the test fish (LT50s), were determined for juveniles of red drum Sciaenops ocellatus obtained from Texas and South Carolina brood stock. This information on red drum thermal tolerance limits was used to determine whether a more northerly distributed population exhibits greater cold tolerance and could therefore be used to improve success of stocking programs. A 96-h temperature tolerance test was performed on juveniles (71–155 mm total length) acclimated to water temperatures ranging from 12.0 to 28.0°C at a salinity of 20‰. Test fish were transferred directly into a series of high-temperature (27.0–37.0°C) or low-temperature (2.0–16.0°C) water baths. The upper LT50 for Texas red drums acclimated to 28.0°C was within 1.1 °C of that calculated for South Carolina red drums acclimated to the same temperature (upper LT50s were 35.7 and 34.6°C). Upper LT50s for fish acclimated to 20.0 and 12.0°C were also similar between the two stocks. Likewise, lower LT50s were similar between the two stocks of red drums: as determined from fish acclimated to 14.0 and 28.0°C, ranges of lower LT50s were 2.8–9.0°C for the Texas stock and 2.69.3°C for the South Carolina stock. The lower LT50 could not be determined for red drums acclimated to 12.0°C, because only three fish died. No consistent differences in temperature tolerance limits were found between the two hatchery stocks, precluding any recommendation to stock one group instead of the other.  相似文献
