The path of upward movement of acid fuchsin dye was traced inpole-size stems of Japanese larch (Larix leptolepis Murr.),European larch (Larix decidua Mill.), Lawson cypress (Chamaecyparislawsoniana (A. Murr. Parl.)), Sitka spruce (Picea sitchensis(Bong. Carr.)), Norway spruce (Picea abies (L.) Karst.), andScots pine (Pinus sylvestris L.). In all of these species amarked, preponderantly clockwise spiral pattern of dye ascentwas observed. In general the upward movement of the dye followedtracheid alignment, except at the sapwood-heartwood boundary.The characteristic patterns of dye movement indicated that spiralgrain was much commoner than was straight grain among the pole-sizegymnosperms investigated. Large differences in spiral grainwere found among species, among trees within species, in differentannual rings within trees, and at different stem heights. Patternsof dye uptake were influenced somewhat by the method of injection.Dye injections through tubes driven into the stem were foundmore suitable for determining tracheid arrangement than wereinjections through individual roots immersed in dye. Factorsinfluencing spiral grain development and applications of thedye injection technique to spiral grain determination are discussed.  相似文献

To visualize water-conduction pathways in living trees, we introduced aqueous solutions of safranin and acid fuchsin into stems of Populus sieboldii Miquel. To examine the spread of each dye in the trees, we compared several techniques for preparing tissue for light microscopy. Acid fuchsin was distributed more rapidly and more widely than safranin, reflecting differences between the dye molecules in state of ionization. We prepared some sections without allowing the dye to redissolve after it had been stabilized by freeze-drying. In these sections, the dye was observed in vessels and in some of the adjacent ray parenchyma cells. Other sections were prepared without stabilizing the dye. In these sections, acid fuchsin in the sap stream left cell walls unstained, whereas safranin stained wood fibers in the vicinity of vessels, as well as the vessels themselves, provided that the sections were mounted in glycerin, which dissolves safranin. Although stained with safranin, the wood fibers contained no water. The results indicate that stabilization of the introduced dye and subsequent preparation of tissues under conditions that avoid dye resolublilization allow accurate visualization of water-conduction pathways at the cellular level.  相似文献

The sap flow method of wood impregnation was conducted to aid the movement of bacteria through the living tree, thereby accelerating their distribution through wood within a short time. When log-pond water containing mixed species of bacteria were introduced in the living trees by butt-end dipping and then laid horizontally for 6 months, bacteria could be delivered by sap flow vertically through the sapwood tracheids up to the high portions from the butt-end of trees; they could be detected in the ray parenchymal cells. The sap-flow method was assumed to deliver the bacteria to sapwood and heartwood at high levels of standing sugi (Cryptomeria japonica D. Don) trees. Degradation of the pit membranes was observed even at more than 3 m upward from the butt-end after the treatment in sapwood, as well as around the butt-end of the trees. Sap flow rates were measured simultaneously by the heat pulse and deuterium tracing techniques in nine Eucalyptus grandis W. Hill ex Maiden. trees at two sites (1) to compare results from the two techniques and (2) to assess the impact of the assumptions underlying the deuterium tracing method on the calculation of sap flow for a range of tree sizes. The trees ranged in height from 4 to 14 m with leaf areas of 5 to 35 m(2). In all trees, sap flow estimated by the deuterium tracing technique was higher than sap flow estimated by the heat pulse method, with differences of 11 to 43% in eight of the trees and 113% in one tree. The largest difference was attributed to errors in the heat pulse method, as indicated by aberrant relationships between sap flow measured by the heat pulse method and tree size characteristics (i.e., diameter, sap wood area, leaf area) for that tree compared with the other experimental trees. Drilling holes in the trees to allow injection of deuterium had no significant effect on sap flow, even when 32 holes were drilled. Sap flow measured by the heat pulse method was only lower after drilling than before drilling in three trees, and the difference only persisted for about 1 h. Deuterium concentrations of water collected from the tree canopies had not returned to background values 17 days after injection. Twenty-one days after injection, sapwood and heartwood samples taken from trunks near the injection sites contained considerable concentrations of deuterium, indicating that some of the deuterium injected into the trees was still present. An experiment performed on two trees showed that deuterium was stored in the heartwood and sapwood throughout the trees, and its distribution within the trees four days after injection was similar whether it was injected into only the sapwood (where it should mix with sap and be transported from the tree most readily) or into both the sapwood and heartwood, indicating that there was considerable movement of deuterium between the heartwood and sapwood. Deuterium storage was accounted for by an approximate means in the sap flow calculations, and may have resulted in an error of about 10% in sap flow estimated by this method. We conclude that the heat pulse and deuterium tracing techniques can be used simultaneously to increase the number of sap flow measurements obtained from a forest, thereby increasing the precision of forest water use estimates. Their combination would be most effective in stands with a wide range of tree sizes and sap flow rates, where the relative differences in sap flux estimates between the methods is small compared with differences in sap flow between trees.  相似文献

The blue‐stain fungus Ceratocystis polonica is pathogenic to Norway spruce (Picea abies) in Europe, as well as to Yezo spruce (Picea jezoensis) and Sachalin spruce (Picea glehnii) in Japan. The wilting mechanism in P. jezoensis saplings after inoculation with C. polonica was examined based on anatomical studies of the phloem and xylem of periodically harvested trees. In addition, the course of sap ascent in the trunks was traced by injection of acid fuchsin solution at harvest. As an initial external symptom, needle discolouration was observed. In dye conduction tests, xylem dysfunction in the xylem of inoculated trees became obvious. The dehydrated xylem area (dry zone) had extended more than 20 cm above the inoculation wounds, within 1 month after inoculation. When the sap flow to the branches had nearly stopped, the leaves began to discolour. Hyphae of C. polonica colonized the ray tissue around the inoculation wounds, but were absent at the front of the dry zones. Defence reactions occurred in ray parenchyma cells adjacent to the penetrating hyphae. It is suggested that secondary metabolites, which are formed by the ray cells and epithelial cells of resin canals, are involved in the obstruction of sap flow. Limited necrotic lesions of the phloem and cambium were not associated with foliar symptoms. It is proposed that the dry zone formation caused by C. polonica is the main mechanism leading to tree death.  相似文献

The natural abundance of stable carbon isotopes in the annual rings of forest trees is used as a tracer of environmental changes such as climate and atmospheric pollution. Although tree-ring delta(13)C varies by about 2 per thousand from year to year, variability within the foliage can be as high as 6 per thousand. Recent studies have shown that branch length affects stomatal response, which influences the integrated foliar delta(13)C signal. To improve the ability of delta(13)C to predict climate differences, we examined the relationship between branch length and foliar delta(13)C in Pseudotsuga menziesii (Mirb.) Franco from four sites across a steep climate gradient in Oregon. The transect spanned the boundary between the ranges of the coastal variety, P. menziesii var. menziesii (three sites), and the Rocky Mountain variety, P. menziesii var. glauca (one site). At the most maritime site, branch length explained 76% of within-site variation of 5 per thousand, whereas at the harshest site, branch length accounted for only 15% of this variation. We considered the possibility that cavitation in the water-conducting xylem obscures the branch length effect in the harsher climates. Cavitation, as measured by dye perfusion, was most extensive at sites where the branch length effect in the coastal variety was weakest. Trees at the site with the most substantial cavitation displayed seasonal xylem refilling. Branch length standardization significantly improved the relationship between delta(13)C and climate. With standardization to constant length, delta(13)C values were significantly related to the degree that climatic variables, as modeled with a forest growth simulation model, constrain transpiration (R(2) = 0.69, P < 0.0001). Without standardization, the R(2) was 0.27. We conclude that sampling standard length branches or tree rings from trees of similar shape and size is desirable when seeking correlations between isotopic composition and climate.  相似文献

The adsorptive properties and selectivity of dyes and water molecules to cellulosic sheet, dependence of adsorptive properties of dyes on the concentration of dye solution, and discoloration of the dye solution due to the UV-ray irradiation were determined for 18 kinds of commercial dyes. The results are as follows: 1) the adsorptive properties of dyes to cellulose sheet differed greatly, but did not depend on the dye types such as acidic, basic and so on; 2) adsorptive properties of dyes to cellulosic sheet de- pended on the concentration of dye solution and were classified into 4 types: concentration-independent, increasing or decreasing with dye concentration, and having a maximum. This classification was irrelevant to the dye types; 3) the irradiation of UV-ray did not cause significant discoloration of dye solution itself, which suggested that wood components as well as dye molecules influence the discoloration of wood.  相似文献

Summary Water in contact with wood surfaces is able to penetrate into the cellular structure by three routes: 1. As liquid water into cell lumena, by capillarity. — 2. As water vapour, by diffusion into cell lumena. — 3. As bound water, by diffusion within the cell wall.Transport from cell lumena into adjacent cell walls occurs rapidly by diffusion. In Scots pine (Pinus sylvestris) sapwood, it is shown that transport over short distances occurs much more rapidly by capillarity than by either of the diffusion processes. Treatment of wood with resin/wax water repellent formulations greatly reduces the rate of water flow due to capillarity and hence significantly cuts down the rate of dimensional change of specimens exposed to wet conditions.Swelling rates due to vapour phase and bound water diffusion were measured experimentally, and these data were used to predict the water sorption rates for specimens treated with a theoretically perfect water repellent, viz. that which excludes all liquid water.It was found experimentally that specimens immersed in water, after treatment with resin/wax water repellents, swelled more rapidly than predicted by the above procedure. This more rapid swelling is probably due to a certain amount of liquid water flow made possible by displacement of the hydrophobic film from cell wall surfaces (preferential wetting). It is suggested that the use of hydrophobic agents bonded chemically to the cell wall may be necessary to attain optimum water repellent effectiveness.  相似文献

Patterns of gene dispersal by seeds and pollen greatly influence the genetic structure of plant populations and their effective size. This paper reviews methods of measuring gene dispersal and current information on patterns of dispersal within local populations of forest trees. Recently, a number of statistical procedures for investigating gene movement based on the use of large numbers of isozyme loci have been described. These procedures include various forms of parentage analysis and the fitting of mating models to genotypic arrays of offspring from individual maternal plants. With the levels of genetic discrimination currently possible in forest trees, the model approach appears to be the most reliable means of estimating gene dispersal parameters. Too little data are available to draw general conclusions about patterns of gene movement within natural populations. Nevertheless, reports to date indicate that dispersal by both pollen and seed can be considerable. For any one mother tree, the bulk of effective pollen in conifers may come from distant males in the same stand or from surrounding stands (gene flow). In insect-pollinated angiosperms, gene flow may also be substantial, but cross-fertilization within stands may primarily be between nearest flowering trees.  相似文献

The use of a stress-wave timer as a minimally destructive device for detecting decay or defect in living trees was evaluated. Measurements were conducted on five tree species (Picea jezoensis, P.glehnii, Betula platyphylla var.japonica, Abies sachalinensis, andLarix kaempferi) with or without decay. Except in sap-rottedA. sachalinensis, the apparent stress-wave velocity in most decayed trees was considerably lower than the value obtained from healthy trees. Our results showed that defect or decay in the trees was detectable more effectively by the method used in the field survey, although the device occasionally failed to detect decay that was incipient, of small extent or confined to sapwood. Other disadvantages of this method are briefly discussed in this paper.  相似文献

To determine effects of stem nitrogen concentration ([N]) on the seasonal course of respiration, rates of stem respiration of ten control and ten irrigated-fertilized (IL), 30-year-old Norway spruce trees (Picea abies (L.) Karst.), growing in northern Sweden, were measured on seven occasions from June 1993 to April 1994. To explore sources of seasonal variation and mechanisms of fertilization effects on respiration, we separated total respiration into growth and maintenance respiration for both xylem and phloem bark. Stem respiration increased in response to the IL treatment and was positively correlated with growth rate, volume of living cells and stem nitrogen content. However, no significant effect of IL treatment or [N] in the living cells was found for respiration per unit volume of live cells. Total stem respiration during the growing season (June to September) was estimated to be 16.7 and 29.7 mol CO(2) m(-2) for control and IL-treated trees, respectively. Respiration during the growing season accounted for approximately 64% of total annual respiration. Depending on the method, estimated growth respiration varied between 40 and 60% of total respiration during the growing season. Between 75 and 80% of the live cell volume in the stems was in the phloem, and phloem maintenance accounted for about 70% of maintenance respiration. Because most of the living cells were found in the phloem, and the living xylem cells were concentrated in the outer growth rings, we concluded that the best base for expressing rates of stem growth and maintenance respiration in young Norway spruce trees is stem surface area.  相似文献

Two activable tracers, Rb and Eu, were injected into the sapwood of Japanese cedars (Cryptomeria japonica D. Don) to investigate the radial movement of minerals in their stems in the resting period. Eight trees of four cultivars, two of which genetically form wet heartwood, were treated near the end of the growing period. At 40 days after the treatment, Rb was detected in the outer heartwood, whereas Eu was not. Radial movement of Rb was more rapid in trees with wet heartwood than in those with normal heartwood. At 204 days after the treatment, more Rb was detected in the heartwood than was found on the first sampling, whereas no Eu was detected in the heartwood. The difference in radial movement between Rb and Eu was considered mainly to be the result of selective transport of beneficial minerals by Japanese cedar. The difference in the rate of radial movement of Rb between wet and normal heartwood became more conspicuous at 204 days after treatment. We concluded that the movement of Rb from the sapwood to the outer heartwood was by active transport through the rays, whereas that in the heartwood was by diffusion due to the gradient of Rb concentration.  相似文献

分别采用抗坏血酸还原法和光化学沉积法,将Ag掺杂到TiO2中,以聚丙烯无纺布为基膜,制备了Ag/TiO2-PVA复合膜以减少TiO2的流失和提高对太阳光的吸收,并对Ag/TiO2-PVA复合膜进行了SEM和XRD表征,以紫外光、模拟太阳光为光源,分别研究了Ag/TiO2-PVA复合膜对有机染料活性翠兰、甲基橙的光催化降解效果,以考察Ag/TiO2-PVA复合膜的催化作用。SEM和XRD的分析结果表明:上述两种方法均能有效地在聚丙烯无纺布表面形成Ag/TiO2-PVA复合膜,抗坏血酸法制备的Ag/TiO2-PVA复合膜的Ag/TiO2负载量高于光化学沉积法。染料废水处理实验结果表明:TiO2/Ag-PVA复合膜对活性翠蓝废水及甲基橙废水的紫外光降解,具有很好的催化作用;对活性翠蓝废水的自然光降解的催化作用不明显,但是对甲基橙废水的自然光降解催化作用较明显。  相似文献

Vegetative propagation techniques are recognized as indispensable tools for mass multiplication of important multipurpose trees adopted in different agroforestry systems. Albizia procera, one among important species, is difficult to propagate commercially either by stem / root cuttings or layering. A study was undertaken to develop procedure for its in vitro regeneration through organogenesis. Explants collected from 15±2 yr-old mature plus trees and from 15 days old juvenile seedlings were regenerated with exogenous application of different hormones. Epicotyl and hypocotyl explants excised from juvenile seedlings showed higher callusing than axillary bud and shoot tip explants derived from mature trees. Benzylaminopurine (BA) at 3 μg/l was most effective, which induced hundred percent callusing in epicotyl and hypocotyl explants in 1/2 Murashige and Skoog (MS) medium. Callus originated from axillary buds and apical shoot tips of mature trees failed to form organs, however callus derived from epicotyl and hypocotyl explants proliferated and formed de novo shoots and leaflets. A concentration of 3 μg/l of BA was found effective for shoot proliferation. Shoots grew vigorously in 2 μg/l gibberellic acid (GA3) treatment and rooted in 1/2 MS medium supplemented with indole-3-butyric acid (IBA) and indole-3-acetic acid (IAA). Rooting was most successful on medium supplemented with 6 μg/l IBA alone on which 93.3% of the shoots formed roots. Sand or vermiculite supplemented with 4 ml of yoshida solution proved as best hardening media, which recorded 70-80% survival of plantlets. One year old tissue culture raised plants had comparatively more height, collar diameter, biomass, and root shoot ratio than plants raised from cuttings and seeds of the same age. The procedures enumerated provide a basis for the development of in vitro techniques for rapid multiplication of A. procera. Rubidium solution was injected in the sapwood of a Japanese cedar cultivar in the growing period, and its radial movement in stem was traced to investigate the accumulation of alkali metals in the heartwood. Sapwood-injected Rb was detected in outer heartwood at 10 days after the treatment, and continued increasing at 20 days after. Radial movement of Rb toward heartwood was considered to occur soon after the treatment, and to decline at a certain point of the time after Rb injection ceased. However, Rb continued moving in heartwood probably by diffusion even after the cease of Rb injection. In a series of injection experiment, radial movement of injected Rb is not corresponding to the seasonality of both cambial activity and cytological changes of ray parenchyma accompanied with heartwood formation. From the results on Rb’s behavior, we conclude that accumulation of K and other alkali metals in heartwood of Japanese cedar has two steps, active transport from sapwood to outer heartwood via ray, and diffusion in heartwood, and that these processes proceed independently from both cambial activity and cytological changes of ray parenchyma.  相似文献

Quercus ilex and Quercus suber trees growing at several sites in Extremadura, Western Spain, that were showing symptoms of oak decline were injected with potassium phosphonate, quinosol or carbendazim using a low-pressure method of trunk injection composed of a pressurized capsule system. A team of four people injected between 120 and 189 trees per day, depending upon the density of the undergrowth vegetation. This labour cost represented, approximately, 15–20% of the total cost of the treatment. The potassium phosphonate-injected trees showed a significant improvement in vegetative growth within 2 years of the injection treatment, and they also showed some recovery from the decline symptoms during the third year. Only one injection treatment of an average of 3.5 capsules (corresponding to 24.5 g phosphonic acid) per tree of approximately 36 cm in diameter, was necessary to reduce the disease severity significantly. Indirectly, these results corroborate the implication of Phytophthora cinnamomi in oak decline within Spanish Quercus woodlands.  相似文献
