A gas-liquid chromatographic (GLC) method is described for the determination of diethylstilbestrol (DES) in molasses-based liquid feed supplements, using dienestrol diacetate as the internal standard. A sample equivalent to 110 mug DES was diluted, acidified with dilute sulfuric acid, extracted with chloroform, and subjected to basic sodium acetate cleanup. The bis-(trimethylsilyl) acetamide derivative was prepared and determined by GLC. Interfering peaks and/or low recoveries were found to be related to emulsions and the procedure incorporates centrifugation to minimize these.  相似文献

A method is given for determining monosodium glutamate (MSG) in soups and soup bases by gas-liquid chromatography (GLC) of the trimethylsilyl ether derivative of glutamic acid. This method compared favorably with existing methods including GLC of the trifluoroacetate (TFA)/butyl ester derivative, and analysis on an amino acid analyzer. In addition, some samples were analyzed by GLC/mass spectrometry. The MSG content of various products ranged from approximately 0.2% in some condensed soups to 13.1% on bouillon cubes. The method, which can detect as little as 0.05%, requires only a 10 min single step derivatization at room temperature and is preferred to the TFA/butyl ester technique.  相似文献

Biogenic primary alkylamines in wines are toxicologically significant and affect sensory properties. An optimized method for analysis in wines involving derivatization with pentafluorobenzaldehyde (PFB) to corresponding pentafluorobenzylimines, liquid-liquid extraction, and gas chromatography with mass selective detection is presented. Reaction parameters including pH, temperature, time, and derivatizing agent and amine concentration were varied in simulated wine solution (15% ethanol) to determine effect on reaction efficiency. Optimal reaction efficiency was characterized (pH 12, 24 degrees C, 30 min, and 10 mg/mL PFB), and parameters were used for the analysis of 10 biogenic alkylamines in 12 California wines. Alkylamine concentration in wines ranged from 0.048 to 91 mg/L. Amine recoveries from wines at five fortification levels (0.1-85 mg/L) were generally 81-100%.  相似文献

A gas-liquid chromatographic (GLC) method has been developed for the determination of the experimental herbicide 3,6-dichloropicolinic acid (Dowco 290) in soils. The method involves extraction of 1 g soil samples with 1N NaCl at approximately pH 7, methylation with diazomethane utilizing a microgenerator, and detection by electron capture GLC. Interferences are small, so that a cleanup step is not necessary even at the 6 ppb level. The procedure is rapid, requiring only 45 min/sample. Recoveries range from 84 to 94% at the 6-1000 ppb level with a minimum detectable limit of 6 ppb. Standard deviations for the percentage recovery values vary from 10.9 to 2.3 for the tested range of 6.7-670 ppb, respectively.  相似文献

The Onley-Yip procedure for determining ethylenethiourea (ETU) in milk and crops was modified to reduce interferences by the ethylenebisdithiocarbamates (EBDCs). A 20 g crop-methanol extract is cleaned up by adsorbing the sample onto Gas-Chrom S. desorbing ETU, and eluting ETU from aluminum oxide with chloroform containing ethanol. ETU is converted to the S-butyl derivative for gas-liquid chromatography (GLC) and flame photometric detection (sulfur mode). For liquid chromatography (LC), ETU is cleaned up on another aluminum oxide column and injected directly. LC and GLC results are confirmed by thin layer chromatography. A cooking procedure based on conversion of EBDCs to ETU is included for surveying crops for possible EBDC content. Recoveries from 8 crops and milk fortified at 0.05 ppm ETU ranged from 73 to 100%.  相似文献

A rapid quantitative method for determining 8 antioxidants in various food products is described. Two procedures are employed. The first involves the use of a glass wood precolumn to separate 3(2)-tert-butyl-4-hydroxyanisole, 3,5-di-tert-butyl-4-hydroxytoluene, 4-hydroxymethyl-2,6-di-tert-butylphenol, and mono-tert-butylhydroquinone (TBHQ) from nonvolatile residues resulting from direct injection of diluted sample or sample extracts into the gas-liquid chromatographic (GLC) column. In the second procedure, the antioxidants TBHQ, 3,3'-thiodipropionic acid, n-propyl gallate, 2,4,5-trihydroxybutyrophenone, and nordihydroguaiaretic acid are isolated from food products by extraction with 70% ethanol. The antioxidant residues are then converted to trimethylsilyl derivatives, and determined by GLC, using a flame ionization detector. Recoveries of all 8 antioxidants from 28 food samples fortified at either 10 or 100 ppm ranged from 70 to 105%.  相似文献

Simplified methodology is presented for the micro determination of polychlorinated biphenyls (PCBs) in biological samples, by conversion to the decachlorobiphenyl (DCB) derivative. Beef adipose tissue and human milk extracts were fortified with PCB standards at 0.1--5.0 ppm, and perchlorinated with antimony pentachloride (SbCl5). Several Aroclors representing various degrees of chlorine content were investigated to assess the efficiency of conversion to DCB. Samples were cleaned up on a Florisil mini column and the PCBs were quantitated by electron capture GLC. Several chlorinated pesticides which were subjected to the perchlorination procedure did not interfere. As little as 0.1 ppm PCBs in 500 mg tissue extract can be recovered at 79-99%. The background DCB content of several brands of SbCl5 was determined. The levels of PCBs in human milk obtained by the perchlorination technique are compared with data acquired by electron capture gas-liquid chromatography in which the individual chlorobiphenyls in the sample are measured.  相似文献

A liquid chromatographic (LC) multiresidue method for determining residues of N-methylcarbamate insecticides in crops was collaboratively studied in 6 laboratories. Methanol and a mechanical ultrasonic homogenizer are used to extract the carbamates. Water-soluble plant coextractives and nonpolar plant lipid materials are removed from the carbamate residues by liquid-liquid partitioning. Additional crop coextractives (e.g., carotenes, chlorophylls) are removed with a Nuchar SN-silanized Celite column. The carbamate residues are then separated on a reverse phase LC column, using an acetonitrile-water gradient mobile phase. Eluted residues are detected by an in-line post-column fluorometric detection technique. Seven carbamates and 2 carbamate metabolites were included in the collaborative study. Each collaborator determined all the carbamates at 2 levels (approximately 0.05 ppm and United States tolerance) in blind duplicate samples of grapes and potatoes. Fortified and control samples were analyzed. Repeatability coefficients of variation for all the carbamates on the 2 crops averaged 4.7% and ranged from 2.4 to 7.1%. Reproducibility coefficients of variation for all the carbamates on the 2 crops averaged 8.7% and ranged from 5.3 to 12.4%. Accuracy, measured by comparison with fortification values, averaged 95% and ranged from 79 to 103%. The estimated limit of quantitation is 0.01 ppm. A method for the quantitative determination of fluoroacetate (FAC) residues in animal tissues is described. The procedure involves tungstic acid extraction, partitioning into ethyl acetate, evaporation of ethyl acetate, derivatization with pentafluorobenzyl bromide (PFB), and analysis of the resulting derivative (PFB-FAC) by capillary gas chromatography-mass spectrometry (CGC-MS) with specific ion monitoring (SIM). The tungstic acid system extracted 96.8 +/- 4.2% of the endogenous 14C-1080 residues in rat tissues. Recovery of FAC during the extraction, purification, and derivatization procedures is established by use of a 14C-FAC spike. 1,2-Dibromobenzene is used as an internal standard for the CGC-MS analysis. PFB-FAC is identified on the basis of comparative retention times and the relative intensities of m/z 257.9 and 181.0. PFB-FAC is quantitated by comparing the response at m/z 257.9 to a PFB-FAC standard curve. Routine sensitivity of the method allows determination of 10 ppb fluoroacetate in tissue.  相似文献

Abstract

Measurement of total soil cadmium (Cd) is difficult due to calcium (Ca) and other chemicals which cause high background absorbance when trace levels of Cd are to be determined. When soil Cd is low, even use of deuterium background correction with flame atomic absorption spectroscopy (AAS) cannot provide accurate Cd results. Use of furnace atomic absorption with method of standard additions can circumvent these interferences, but the cost and time required are substantial. We desired a more rapid, convenient, and reliable alternative to extraction using dithizone and back‐extraction into acid, or to ammonium pyrollidinedithiocarbamate (APDC) which does not require close pH adjustment nor have many sources of potantial contamination. We evaluated analysis of these complex soil extracts with the method of Viets (1978) which extracts metals from 1N acid solutions using Aliquat‐336 in methylisobutyl‐ketone (MIBK). We tested the use of the less toxic and less water soluble 3‐heptanone as an organic solvent alternative to MIBK which can be directly analyzed by flame atomic absorption. A series of extraction experiments were conducted to determine if Cd was extracted from standard solutions and from total metal digests of calcareous soils into an Aliquat‐336/3‐heptanone solution, and under what conditions extraction was optimum. In the optimum method, Cd was extracted from aqua regia soil digests by 10% Aliquat‐336 in 3‐heptanone without addition of ascorbic acid or potassium iodide (KI) used by Viets. Excellent recovery of Cd was obtained for both standard reference soils and low Cd highly calcareous soils from North Dakota and Minnesota. Addition of ascorbic acid and KI did not increase the efficiency of extraction indicating that the extraction system used was free of ferric‐iron [Fe(III)] interference. The ion‐association complex of Cd remained stable for at least 24 hr after extraction, providing a very convenient method to analyze low levels of total Cd in soils and other geologic materials.  相似文献

A limited interlaboratory study was carried out to determine the effect of lighting conditions on the release of cadmium from ceramic glazes by 4% acetic acid. Cadmium release increased with increased exposure to light. Further interlaboratory study on plates manufactured under controlled conditions showed that if temperature, intensity of illumination, and time of exposure are specified, reproducible results for leaching of both lead and cadmium can be obtained. A modification of the official AOAC method, 25.031-25.034, to increase sensitivity was collaboratively studied by 8 laboratories. Each received 6 solutions of lead in 4% acetic acid (3 sets of blind duplicates at the 0.1, 0.5, and 0.8 mug/ml levels) plus a reagent blank. Average recoveries were 0.1006, 0.5056, and 0.8194 mug/ml, respectively, with coefficients of variation of 11.4, 3.3, and 2.8%, respectively. The proposed modification has been adopted as official first action, and a parameter for exposure to light during extraction has been included in the method.  相似文献

Reduction of aflatoxin B1 and aflatoxin B2 with sodium borohydride quantitatively yielded new fluorescent derivatives, designated as aflatoxin RB1 and aflatoxin RB2. Mass spectrometric data showed that RB1 and RB2 were trihydroxy derivatives of B1 and B2, respectively. Nuclear magnetic resonance analysis revealed that new chemical shifts were present in aflatoxins RB1 and RB2 in addition to those of the parent aflatoxins. The new compounds had lower melting points and different ultraviolet and infrared spectra compared to aflatoxins B1 and B2 and the monohydroxy derivative aflatoxicol. They were lses toxic to chick embryos than the parent toxins. Since the reduction yields were quantitative and since the reduction products could be detected at low levels comparable to those for B1 and B2, the reduction reaction could be used as a confirmatory test for both aflatoxins B1 and B2. Preliminary results obtained from gas-liquid chromatography (GLC) analysis of the trimethylsilyl derivatives of aflatoxins RB1 and RB2 indicated that these compounds could furnish the basis for developing an analytical GLC method for aflatoxins B1 and B2.  相似文献

A modification of the official method for ochratoxins and a screening method for zearalenone, aflatoxin, and ochratoxin is described and expanded to include citrinin and penicillic acid. The method uses 0.5N phosphoric acidchloroform (1+10) in the initial extraction; the extract is divided and eluted from 2 columns to provide a quantitative thin layer chromatographic (TLC) method for aflatoxin and ochratoxin in corn and dried beans. Aflatoxin and zearalenone are eluted from one column and ochratoxin, penicillic acid, and citrinin from the other. Ochratoxin A recoveries are low (50%) in peanuts. Zearalenone, penicillic acid, and citrinin were qualitatively recovered from corn and beans; zearalenone and penicillic acid were recovered from peanuts but citrinin was not. Several TLC solvents were used to separate interferences.  相似文献

Quantitation of o- and p-sulfamoylbenzoic acid residues in saccharin and its sodium salt is achieved by a method comprising methanolic extraction and high-performance ion exchange chromatography. A commercially available anion exchange column was employed with an aqueous buffered (pH 9.2) mobile phase. As little as 80 ppm of the ortho-isomer and 25 ppm of the para-isomer can be accurately determined. The levels of detectability (2 times noise) are estimated as 8 ppm (0.16 mug on column) and 2.5 ppm (0.05 mug on column), respectively. Recoveries from saccharin ranged from 92.7 to 96.5% (ortho) and from 92.2 to 103.3% (para). Recoveries from the sodium salt ranged from 93.1 to 104.4% (ortho) and from 93.5 to 97.8% (para). Of 9 other potential saccharin impurities tested separately, only one was found to interfere slightly in the chromatographic part of the procedure.  相似文献

A liquid chromatographic (LC) method using a 2-step purification technique for the simultaneous determination of 10 carbamates in bovine, swine, and duck livers has been developed. Carbamates are extracted from liver samples with methylene chloride. After evaporation, the residues from the extract are dissolved in methylene chloride-cyclohexane (1 + 1) and cleaned up by gel permeation chromatography. The eluate containing carbamate residues is evaporated to dryness, reconstituted in methylene chloride, further purified by passing it through an aminopropyl Bond Elut extraction cartridge, and analyzed by liquid chromatography using post-column derivatization with orthophthalaldehyde and fluorescence detection. Excitation and emission are set at 340 and 418 nm, respectively. Liver samples for beef, pork, and duck were fortified with 5, 10, and 20 ppb of mixed carbamate standards. The average of 10 recoveries of 10 carbamates at all 3 levels of fortification was greater than 80% with coefficients of variation less than 17%.  相似文献

The 2-chloroethyl esters of 5 fatty acids have been identified in spice and food samples by gas-liquid chromatography-mass spectrometry (GLC/MS). Twenty-four spice samples were analyzed for the 2-chloroethyl esters of fatty acids by AOAC official multiple residues pesticide procedure using GLC with microcoulometric detection. The esters of capric, lauric, myristic, palmitic, and linoleic acids have been identified at levels up to 1400 ppm. 2-Chloroethyl linoleate was the most abundant ester in all samples. Several foods analyzed by the same procedures showed levels of 2-chloroethyl linoleate as high as 35 ppm. Recoveries from fortified samples ranged from 84 to 98% for the various esters. A method using an acid-catalyzed esterification reaction was developed to rapidly determine the fatty acid content of these spices. GLC analysis with microcoulometric detection was used. Recoveries from fortified samples ranged from 92 to 110%. After 2 spice samples found to be free of 2-chloroethyl esters were fumigated with ethylene oxide, the level of 2-chloroethyl linoleate reached 77 ppm. All levels of 2-chloroethyl esters were confirmed by GLC/MS.  相似文献

A gas-liquid chromatographic (GLC) method was developed for the detection and determination of thiourea in citrus peels. After the peel is extracted with ethyl ether, the ether extract is adsorbed on sodium sulfate together with water. Thiourea is recoverd from both the sodium sulfate and the peel residue with ethyl acetate-acetone(2+1). The extracted mixture is cleaned on an alumina column, the eluate is concentrated under vacuum, and thiourea is extracted from the concentrate with sodium carbonate solution. GLC was carried out on the prepared benzoyl derivative of thiourea. The average recoveries of thiourea from lemon peel were 85.3, 93.1, and 97.6% at the fortification levels of 1, 10, and 100 ppm, respectively. The detection limit was low as 0.08 ppm.  相似文献

An international collaborative study of IUPAC methods II.D.19 and II.D.25 for preparation and GLC analysis of fatty acid methyl esters was begun in 1976. The IUPAC methodology, applicable to animal and vegetable oils and fats and fatty acids from all sources, contains special instructions for preparation and analysis of methyl esters of fatty acids containing 4 or more carbon atoms (analysis of milk fat). Twenty-three collaborators participated in the analysis of 5 known mixtures, 4 vegetable oils, 1 fish oil, and 2 butterfats. Several blind duplicate samples were included. The experimental data were subjected to statistical analysis to examine intra- and interlaboratory variation. Reproducibility and accuracy data for the higher fatty acid (14:0-22:1) mixtures and fish and vegetable oils were satisfactory and were in good agreement with results from an AOCS Smalley Committee check sample program involving analysis of the same samples. Typical coefficients of variation (%) at various concentrations were 15 (2% level), 8.5 (5% level), 7 (10% level), and 3 (50% level). Low recoveries and poor reproducibility were characteristics of results obtained for butyric acid in the butterfat and related known mixtures. A coefficient of variation of about 19% was found for analysis of butyric acid in butterfat, vs coefficients of variation in the range of 4-13% for similar levels of other components in butterfat and other samples. The IUPAC methodology for GLC analysis of fats and oils other than milk fats has been adopted by the AOAC as official first action to replace the current GLC method, 28.063-28.067.  相似文献

A method is described for the electron capture gas-liquid chromatographic determination of thiabendazole and methyl 2-benzimidazole carbamate (MBC) after derivatization with pentafluorobenzyl bromide (PFB-Br). The samples are extracted with ethyl acetate, and the residual thiabendazole, benomyl, and MBC are isolated by liquid-liquid extraction into dilute HCl. After neutralization and re-extraction with ethyl acetate, thiabendazole and MBC are reacted with PFB-Br to form the PFB derivatives. Alumina column chromatography was used to clean up extracts, and the derivatives could be detected as low as 5-10 pg. Recoveries were 95-98% from fruits fortified with 0.3-2.0 ppm thiabenzadole; recoveries were 91-97% when 0.05-1.0 ppm benomyl/MBC were added to fruits/crops. The PFB derivatives were identified by gas-liquid chromatography-mass spectrometry.  相似文献

Corn samples spiked at levels of 100, 300, 1000, and 2000 mug zearalenone/kg were sent to 22 collaborators for analysis by the Eppley method. All samples were yellow corn except one white corn sample spiked at 2000 mug/kg. Results from 16 collaborators were statistically analyzed. Only 4 of 16 collaborators detected zearalenone in the sample containing 100 mu/kg, but 11 detected the toxin in the sample containing 300 mug/kg. Average recoveries from all samples were 129% at 300 mug/kg, 101% at 1000 mug/kg, and 88% at 2000 mug/kg. The between-laboratory coefficients of variation were 53.0% at 300 mug/kg, 38.2% at 1000 mug/kg, and 27.0% at 2000 mug/kg. Five naturally contaminated corn samples, one in triplicate, were also provided. The mean level of zearalenone in the naturally contaminated samples ranged from 431 to 7622 mug/kg. The mean coefficient of variation for all samples was 40.5%. Two collaborators measured quantities of zearalenone on thin layer chromatographic plates densitometrically. Their results were not included in the statistical analysis, but the results indicated that densitometric measurement, given proper dilutions of solutions, could be used.
