Two experiments were conducted to evaluate the effects of early feed restriction on lipid peroxidation, pulmonary vascular remodelling and ascites incidence in broilers under normal and low ambient temperature. In experiment 1, the restricted birds were fed 8h per day either from 7 to 14 d or from 7 to 21 d, while the controlled birds were fed ad libitum. In experiment 2, the restricted birds were fed 80 or 60% of the previous 24-h feed consumption of full-fed controls for 7 d from 7 to 14 d. On d 14, half of the birds in each treatment both in experiment 1 and experiment 2 were exposed to low ambient temperature to induce ascites. Body weight and feed conversion ratio were measured weekly. The incidences of ascites and other disease were recorded to determine ascites morbidity and total mortality. Blood samples were taken on d 14, 21, 28, 35 and 42 to measure the plasma malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px). On d 42, samples were taken to determine the right/total ventricular weight ratio (RV/TV), vessel wall area/vessel total area ratio (WA/TA) and mean media thickness in pulmonary arterioles (mMTPA). Low-temperature treatment increased plasma MDA concentration. When broilers were exposed to a cool environment for 3 weeks, plasma SOD and GSH-Px activity were decreased compared with normal-temperature chicks. RV/TV, WA/TA and mMTPA on d 42 were increased in birds exposed to cold, consistent with the increased pulmonary hypertension and ascites morbidity. Early feed restriction markedly decreased plasma MDA concentration. The plasma SOD and GSH-Px activity of feed-restricted birds were markedly higher than those fed ad libitum on d 35 and d 42. All early feed restriction treatments reduced ascites morbidity and total mortality. On d 42, the RV/TV, WA/TA and mMTPA of feed-restricted broilers were lower than that of the ad libitum-fed broilers. The results suggested that early feed restriction alleviated the lipid peroxidation, promoted the activity of enzymatic antioxidant and inhibited pulmonary vascular remodelling. These changes might be associated with reduced ascites incidence.  相似文献

A study was conducted to examine the effect of nutritional stress on the development of the thymus, bursa, and pancreas of 7-to-14-day-old commercial meat-type chickens. One group of 7-day-old chickens was given access to food for only 30 minutes daily for 7 days. The birds were necropsied, and the thymus, pancreas, and bursa were compared with those of the control chickens fed ad libitum and necropsied at 7 and 14 days of age. The thymuses from birds on the restricted diet were atrophied (0.45 +/- 0.08 g) and congested compared with the thymuses from chickens fed ad libitum (1.32 +/- 0.31 g). The bursae from chickens on a restricted diet were also smaller (0.26 +/- 0.08 g) than the bursae from birds with free access to food (0.74 +/- 0.11 g). The restricted diet did not appear to cause any gross or histological pancreatic changes. The thymic lesions produced by nutritional stress were similar to those observed in the runting/stunting syndrome.  相似文献

1. The increase in the numbers of yellow and of white ovarian follicles was studied from 10 weeks of age to sexual maturity in broiler breeder pullets fed ad libitum and on a restricted intake. The growth of follicles greater than 1 mm diameter commenced at 14 to 16 weeks of age in ad libitum birds and at 22 to 24 weeks in those which were restricted. 2. In a second experiment, groups of birds restricted from 4 d of age were transferred to ad libitum feeding at 2-week intervals and examined at first egg. The numbers of yellow follicles at first egg were not affected by restriction to 14 weeks of age but, thereafter, declined linearly with time on restriction. 3. In the second experiment there were no comparable responses in the numbers of white follicles or in the rates of atresia. 4. Food restriction to 22 weeks of age produced about half the response in numbers of yellow follicles as restriction continued to point of lay (27 to 32 weeks of age). In practical situations, the results suggest that food intake should be restricted up to the onset of lay and then increased in relation to the rise in egg production. 5. Atresia was greatest in white follicles 3.15 to 6.30 mm diameter and was less outside this range.  相似文献

The left and right antitrochanters of 80 female turkeys of four strains were examined for histopathological changes at sexual maturity. Ten birds of each strain were fed ad libitum and 10 restricted to achieve 0.6 of the bodyweight of the ad libitum fed birds at 24 weeks old when the birds were photostimulated. Bodyweights at sexual maturity (28 to 31 weeks) for the four strains were 5.4, 6.5, 7.6 and 13.2 kg for restricted and 7.5, 9.0, 11.1 and 17.1 kg for ad libitum fed birds. The prevalence and severity of cartilage change increased with mature size and was lower in restricted turkeys then in turkeys fed ad libitum. The results showed that abnormal cartilage changes forming the basis of clinical disease occur in females. The prevalence and severity of lesions were directly related to bodyweight and were not limited to particular genotypes. Lesions were more severe in the centre of the antitrochanter, possibly because of forces exerted by the trochanter. Concentrations of basophilic cells in the hyaline cartilage were associated with the increased prevalence and severity of lesions in large turkeys and may be of significance in the development of osteochondrotic lesions in the antitrochanter.  相似文献

1. The Richards function was used to describe the growth curves (n = 989) of 9 broiler lines. Chickens were fed ad libitum and body weight was recorded every second week from hatching to 26 weeks of age. 2. The accuracy of curve fit measured by the coefficient of determination (R2) was better for males than for females (0.9986-0.9995 vs 0.9972-0.9988, respectively). 3. The estimation of the asymptotic final weight (A) for different lines enabled the degree of maturity (ut = yt/A) to be determined at any fixed point of the curve. At the age of 7 weeks this had a value of 0.318-0.369 for cockerels and 0.325-0.377 for pullets and represented the slaughter maturity of individual lines. The ratio of inflection/asymptotic weight (y+/A = 0.370-0.388) indicated that in some cases chicken growth can be described approximately by the Gompertz function (y+/A = 0.368). 4. It was found that the age at the inflection point of curves (t+ 48.2-55.7 d for cockerels and t+ = 47.8-52.8 d for pullets) roughly corresponds to the slaughter age of the chickens. 5. The interline differences in the parameters of maturation rate for weight (y+/A, k, t+, u7) are low in comparison with the differences in body weight (A, y+, y7) and absolute growth rate (v, v+). 6. The intragroup phenotypic correlation among growth parameters and the importance of the mathematical models are discussed.  相似文献

Broiler breeder pullets are fully fed for several weeks to give chicks a vigorous start, to establish an adequate frame size, and to build increased flock BW uniformity. This study was designed to determine whether reducing the length of the initial ad libitum feeding period of pullets would be detrimental to subsequent fleshing, skeletal development, and BW variation. A total of 720 Ross 308 pullets were placed in 8 pens on the day of hatch and provided ad libitum access to feed at 1 wk (1WK) or 3 wk (3WK) of age, at which time a 5:2 restriction program began. Individual BW and external fleshing scores, and flock BW variation (CV and uniformity) were monitored. At 4, 8, 12, and 16 wk, 60 randomly selected birds per treatment were dissected for assessment of breast muscle, fatness, and reproductive development. At 3 wk of age, BW of the 3WK pullets (471 g) was greater than that of the 1WK pullets (312 g), and the daily rate of gain was double. Although feed allocation was decreased markedly at 3 wk in 3WK birds, by 4 wk they weighed 30% more, and had a greater frame size and proportion of breast muscle than the 1WK birds. At 8 wk of age, the 3WK birds were still heavier (973 g for 3WK vs. 899 g for 1WK). Most carcass measures were similar between treatments at 12 wk of age, by which time BW profiles were similar. At 16 wk of age, frame size and proportion of breast muscle were not different between groups. The BW variation did not differ through the initial 12 wk, but was superior at 14 and 16 wk of age in 1WK birds, possibly because of greater feed allocation between 8 to 16 wk, which is the most intense feed restriction period. The reduced feed intake of 3WK birds at the onset of feed restriction reduced their ME requirement for maintenance, likely contributing to this result. Increasing the length of the ad libitum feed access period after hatch altered growth and conformation traits to 8 wk of age and did not affect frame size or proportion of breast muscle, but increased BW variability late in the rearing period.  相似文献

1. Comparisons of age, food intake, body weight and body composition at the onset of sexual maturity were made between dwarf and normal chickens from lines selected for high or low juvenile body weight.

2. At the same age birds which had started to lay were significantly heavier and contained significantly more abdominal and carcass fat than those which were not laying.

3. Differences between normal and dwarf pullets within lines varied with genetic background, indicating differences between lines in meeting a body weight or a body composition requirement, or both, for the onset of sexual maturity.

采用低温诱发肉鸡腹水综合征(AS),并观察早期限饲对常温和低温饲养时肉鸡AS的发病率、肉鸡体内脂质过氧化作用和抗氧化酶活性的影响。试验1,限饲处理组肉鸡分别于7~14日龄或7~21日龄每天给料8h,对照组肉鸡全程自由采食。试验2,限饲组肉鸡分别于7~14日龄给予对照组肉鸡前一天饲料消耗量的60%或80%,对照组肉鸡自由采食。试验过程中连续测定肉鸡血浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)、丙二醛(MDA)水平和腹水心脏指数(RV/TV值)。结果显示低温显著升高了肉鸡血浆MDA浓度,降低了血浆SOD和GSH—Px的活性;低温饲养肉鸡5~6周龄时的RV/TV值显著升高,AS的发病率显著增加。限饲处理显著降低了肉鸡血浆MDA含量,并且限饲处理肉鸡生长后期血浆中SOD和GSH—Px活性显著高于自由采食组肉鸡。所有的限饲处理均能降低肉鸡AS的发病率,并且早期限饲处理肉鸡42日龄的RV/TV值显著低于非限饲肉鸡。这表明早期限饲能够减轻肉鸡体内脂质过氧化作用,提高机体抗氧化酶的活性,从而降低AS的发病率。  相似文献

The influence of two methods of food restriction during rearing on the body and liver composition of layer-type birds was investigated during and after restriction. In each of two experiments groups of birds were reared on one of three treatments: ad libitum food intake (A), time-limited food restriction (TR) or proportional food restriction (PR). All birds were fed ad libitum during the laying phase. In experiment 1, six birds from each treatment were selected at random for estimation of body and liver composition and killed at 70, 101, 162, 218 and 337 d of age. In experiment 2 four birds were selected from each treatment at 280 and 467 d of age. Body composition was predicted in experiment 2 at various times such as sexual maturity, using equations based on deuterium oxide dilution techniques. Body composition changes caused by food restriction were related to the length of restriction. Towards its end the major changes were: decreased body fat, increased total body water and increased water content of the fat-free mass for TR and PR birds compared to A birds. Some changes were not due to a reduced liveweight. Food restriction during rearing did not change body composition permanently, although predicted body fat at sexual maturity (experiment 2) was lower and protein content higher for TR and PR than for A birds. There was no demonstrable relationship between the body fat content predicted at sexual maturity and subsequent rate of lay (experiment 2).  相似文献

1. Broiler breeder pullets were maintained on 10-, 11-, 12-, 13-, 14- or 16-h photoperiods to determine the effect of constant photoperiods on sexual development in broiler breeders. The birds were fed to achieve a 2100 g body weight at approximately 17 or 20 weeks to see if the photosexual response was modified by rate of growth. 2. In both body weight groups, pullets maintained on 10h were the first to reach sexual maturity (50 eggs/100 bird-d), and these and the 11-h pullets matured significantly earlier than any of the other photoperiod groups. Pullets maintained on 13 or 14 h matured latest, at about 3 weeks after the 10-h pullets, though both were only marginally later than the 12- or 16-h birds. These differences in maturation probably reflect the different rates at which photorefractoriness is dissipated in broiler breeders reared on photoperiods that vary in their degree of stimulatory competence. 3. There were no significant interactions among the photoperiods and the ages at 2100 g; faster-growing birds consistently matured about 10 d earlier than conventionally grown pullets.  相似文献

1. Females from a medium body weight strain of turkeys were fed ad libitum or restricted to 0.6 of ad libitum-fed body weight at sexual maturity and were photostimulated at 18, 24 or 30 weeks of age. Ovarian follicular structure was assessed in 8 birds from both rearing treatments at photostimulation, after photostimulation for 6 or 12 weeks and at 55 weeks of age. 2. The patterns of small white follicles and of larger yellow follicles were similar in restricted and in ad libitum-fed turkeys for birds in lay at the same age, regardless of the time elapsed since photostimulation. 3. Among birds in lay the number of normal yellow follicles and the proportion of multiple yellow follicles declined with age. A relatively low proportion of restricted birds photostimulated at 18 and 24 weeks were in lay after 12 weeks of photostimulation. 4. For birds in lay the number of white follicles 1.4 to less than 1.8 mm in diameter increased and the number greater than 1.8 mm declined with age. There was a corresponding increase in the rates of atresia in follicles 1.8 to less than 5.0 mm in diameter.  相似文献

The relationship between water intake and water concentration in the body and in the gastrointestinal contents was studied in meat-type chickens fed either intermittently or ad libitum, as controls. In experiment 1, male broilers were reared from 12 to 33 d of age and were fed every second day or ad libitum. The water consumption of the intermittently-fed birds on days of food removal was 31% of the consumption of the control group. On days of food restoration their water consumption exceeded that of the controls, by 27%. No significant differences in water:food ratios were found between birds fed intermittently or the controls on days of food restoration. The restricted birds had a lower fat and a higher protein concentration in the body. No relationship was found between water:food ratio and body composition. In experiment 2, birds were kept on the same feeding regimes from 12 to 46 d of age. The water content of the pancreas increased after food restoration compared with ad libitum feeding or food deprivation. The amount of water in the gastrointestinal contents increased progressively from crop to ileum. The feeding regimens had no effect on the water content of the intestine.  相似文献

1. Ovarian function was assessed at first egg and at 44 and 54 weeks of age in traditional turkeys and in a male-line fed ad libitum or restricted to 0.5 of ad libitum during rearing. Egg production was recorded daily and fertility and hatchability were determined at 2 ages. 2. Age at sexual maturity did not differ between the 3 treatments. 3. The number of follicles at first egg in the male line averaged 16.3 compared with 8.9 in traditional turkeys. Food restriction in the male line decreased the number of yellow follicles to 14.6 at first egg and restricted birds were largely in a non-reproductive state from 44 to 54 weeks of age. 4. The total number of recorded ovipositions to 54 weeks of age in male line fed ad libitum or restricted and traditional turkeys respectively were 40.1, 25.2 and 70.7; comparable productions of normal shelled eggs were 28, 20 and 66.3. 5. There were no differences between treatments in fertility after a single insemination at 2 different ages. Hatch of fertiles after the first inseminations were 71.4, 45.5 and 90.3 respectively for male line fed ad libitum or restricted and traditional turkeys.  相似文献

Female chicks from 6 pure lines of broiler breeders were obtained from international broiler breeder companies. The lines were from three female and two male lines and a relaxed selection line derived from one of the male lines (M2) and maintained without selection for 25 years (generations). Replicate groups from each line were reared in floor pens and fed ad libitum. An additional group of the M2 male line was feed restricted and housed in similar pens. Twenty birds from each group were photostimulated after housing them in individual cages at 14 weeks of age. Ovarian follicular dynamics and follicular sensitivity were determined after the onset of lay. Female lines had a greater proportion of atretic follicles and more groups of multiple follicles than male lines. Body weights, fatness and age at puberty were similar in male and female lines. The relaxed selection line had more abdominal fat than any of the selected lines and the ovary contained a similar number of normal yellow follicles as the selected male line. Feed restriction decreased body weight, the numbers of yellow follicles, multiple yellow follicles and the proportion of atretic yellow follicles, and increased the age at onset of lay. The response of ovarian follicles of three different sizes to exogenous gonadotrophin stimulation with PMSG was similar in small yellow follicles, 5 to 6 and 2 to 3 mm white follicles in a female line, a male line and its relaxed line fed ad libitum. The results suggest that the original lines from which modern male-line broiler stocks are derived had a relatively large number of yellow follicles and a high propensity for multiple ovulation at the onset of lay. Separate selection for different traits in male and female lines has not had a dramatic effect on ovarian function or sensitivity to gonadotrophin stimulation.  相似文献

1. The Guinea Savanna in the tropics is characterised by high diurnal temperatures, often beyond the thermo-neutral zone of modern poultry, which imposes heat stress on them. 2. An experiment was conducted to determine the effect of partially (12 h wet mash in the day and 12 h dry mash in the night) or wholly (24 h) feeding wet mash on the growth performance of broiler chickens. 3. Three treatments: dry mash (control), wet mash (day) + dry mash (night) and wet mash (day + night) were tested. At 28 d of age, a total of 120 broiler chickens (Hybro, Netherlands) were randomly divided, using a completely randomised design, into 12 groups of 10 birds, each with mean live weight of approximately 800 g/bird. A maize-soybean based grower mash (200 g CP/kg and 12·5 MJ/kg ME) was fed. The wet mash was prepared daily by addition of 1·3 parts of water to 1 part of dry mash and fed from 28 to 56 d of age. The birds were housed in raised-floor pens. Feed and water were given ad libitum and light provided 24 h. Mean daily room temperature was 28-29°C in the morning and 35-41°C in the afternoon. 4. Mean feed intake of birds fed the wet mash (174 g/day) or wet + dry mash (166 g/day) was higher than that of birds fed the dry mash (152 g/day). Mean live weight gain of birds fed the wet mash (64 g/day) or wet + dry mash (64 g/day) was higher than that of birds fed the dry mash (58 g/day). There were no differences in gain/feed ratios (0·38-0·39) of any of the treatments, neither were there any differences between the feeding of wet mash partly or wholly for all variables. 5. It was concluded that wet feeding, particularly during day-time, had the potential to improve growth performance of broiler chickens in a hot climate.  相似文献

Hy‐Line Gray commercial pullets were maintained under 8‐h photoperiods, 16‐h photoperiods and 16‐h photoperiods supplemented with a diet containing 20 or 200 mg/kg melatonin (MEL) to investigate the role of MEL in sexual development. A total of 256 Hy‐Line Gray commercial pullets were placed, four birds to a cage, in four similar light‐proof rooms (8‐h photoperiod) at 6 weeks of age. At 70 day, three rooms containing a total of 192 birds were transferred to a 16‐h photoperiod, whereas 64 birds were maintained under the 8‐h photoperiod. Diets containing MEL at 20 and 200 mg/kg were fed to birds in two of the rooms under 16‐h photoperiods. Birds maintained under an 8‐h photoperiod matured 11.25 day later than those maintained under a 16‐h photoperiod (p < 0.05). The group of birds receiving 20 mg/kg MEL matured 1.19 day later than those maintained under the 16‐h photoperiod and 10.06 day earlier than those maintained under the 8‐h photoperiod. The group of birds receiving 200 mg/kg MEL matured 3.13 day later than those maintained under a 16‐h photoperiod and 8.12 day earlier than those maintained under an 8‐h photoperiod. The average body weight of birds maintained under the 8‐h photoperiod was greater than that of birds maintained under the 16‐h photoperiod (p < 0.05) and was similar between the different MEL groups. The abdominal fat weight was lower in 16L:8D group compared with 8L:16D group (p < 0.05). The concentrations of follicle‐stimulating hormone, luteinizing hormone, oestrogen and insulin did not differ significantly among the groups. The melatonin concentration in 200 mg/kg melatonin group was higher than that observed in the other groups; however, this concentration did not differ significantly (p > 0.05). These data suggest that the birds did not perceive the final 8‐h photoperiod as being part of the night when they were given the MEL diets; continuously high plasma MEL was not observed in birds that responded as if they were in constant darkness. However, the later maturity of the groups administered MEL diets compared with the groups maintained under a constant 16‐h photoperiod clearly indicated that MEL has some influence on the sexual maturity of pullets.  相似文献

1. Lohmann Brown pullets, in one trial, and Hyline Brown pullets in another, were reared from day 2 on short daylengths, and from week 8 in trial 1 (week 16 in trial 2) on food restriction. These restrictions were lifted at various times during the rearing period as a means of determining the relative importance of the day length and food restriction stimuli on the attainment of sexual maturity and subsequent laying performance. 2. A total of 2304 pullets were used in each trial. The birds were reared in light proof rooms, and subjected to 8L:16D until they were moved to a laying facility where a light stimulus of 16L:8D was applied. In trial 1 the six ages at which light stimulation was applied were 115, 122, 129, 136, 143 and 171 d. Within each light treatment, food restriction of pullets, which consisted of feeding 72 g of food/bird d, was lifted at six different ages, namely, 115, 129, 143, 157, 171 and 185 d. In trial 2 both the light stimulation and the lifting of food restriction occured at 111, 125, 139, 153, 167 and 181 d of age, producing 6x6=36 treatments in both trials. 3. The first trial was terminated when the pullets were 28 weeks old, soon after all the birds had commenced laying, because of an outbreak of Egg Drop Syndrome. However, because age at maturity was the variable of major interest, data from this experiment could be used in the analysis. The second trial ended when the birds reached 40 weeks of age. Variables measured were age at maturity, food intake and body weight gain subsequent to the lifting of restrictions and, in the second experiment, rate of lay, peak rate of lay and egg weight at various ages. 4. The mean age at sexual maturity was influenced by the date of release from light restriction (P<0.001) and from food restriction (P<0.001) in both trials. In addition, the interaction between the age at release from light and from food restriction was significant. Regression equations were produced for each trial to describe the relationships between the age at sexual maturity and the age at release from light restriction and food restriction. 5. There was an effect of both light restriction (P<0.001) and of food restriction (P<0.001) on the increase in food intake (g/bird d) in the week following release from food restriction in both experiments. These effects were not independent: the effect of the interaction of light and food restriction on this increase in food intake was also highly significant. The longer the birds were subjected to light restriction, the less dramatic the increase in food intake when food restriction was lifted. The more sustained the period of food restriction, the higher the increase in food intake in the week following release from the restriction. 6. Mean egg weight was 4 g heavier at 22 weeks of age in birds released from food restriction at 16 and 18 weeks, than from those released at 24 and 26 weeks of age. However, by 30 weeks of age, birds restricted for longer produced heavier eggs than their earlier-maturing counterparts. This effect continued to the end of the trial at 40 weeks of age, at which time there was a 2.3 g difference in egg weight between these treatments. 7. Both light and food restriction have an effect on the age of maturity in laying hens. The length of time between the release from light or from food restriction to the onset of laying depended on the age of the pullets when the release occurred. Egg weight at a given age was significantly affected by the age at release from food restriction, but not from light restriction.  相似文献

1. The first objective was to evaluate baseline heart rate (HR) responses in fast growing broilers fed ad libitum from 1 to 45d or subjected to a food restriction (85%, 70% and 55% of ad libitum from 7 to 21d) and a refeeding (22 to 45d) regimen in a normal thermal environment. The second was to evaluate HR and haematocrit responses in fast growing broilers reared at a low environmental temperature and fed ad libitum (A-L) from 1 to 42d, subjected to food restriction (70% ad libitum ) between 7 to 21d and refed thereafter (R-R), or subjected to food restriction between 7 to 42d (R), and to follow the changes in normal birds and those prone to ascites. 2. The baseline HR in the normal thermal environment at 21 and 45d in birds fed ad libitum was lower (P < 0.001) than at 7d. The food-restricted groups had higher HRs at 21d than at 7d, with the differences significant (P < 0.01) for the 70% and 55% treatments. At 21d the HRs in restricted groups were significantly higher (P < 0.01) than in ad libitum -fed birds. After returning to ad libitum feeding the HRs in these birds tended to decline, with the differences significant (P < 0.05) for the 70% and 55% treatments. 3. In the birds exposed to cold, each feeding regimen produced distinct patterns of change in HR and haematocrit. Both feeding regimen and age had significant (P < 0.001) effects on the changes of both variables. Relative to 7d, on 14d and 21d the HRs decreased and haematocrits increased in all groups, but the rates of change were highest in the A-L and R-R birds. The first fulminant cases of ascites were observed during the third week in the A-L group and during the fourth week in the R-R group. None of the chickens from the R group developed ascites. Altogether, the incidence of ascites was 48% in the A-L group and 28% in the R-R group. 4. Among the birds exposed to cold, the ascitic birds had significantly (P < 0.001) lower HRs and higher haematocrits than normal birds from the same feeding regimen group. The time trends in the rates of change in HR and haematocrit were distinctly different (P < 0.001) for ascitic and normal birds. In view of the evidence presented, it appears that hypoxaemia and tissue hypoxia in broiler chickens may be a result of circulatory insufficiency associated with progressive bradycardia rather than, as commonly believed, pulmonary insufficiency. Circulating concentrations of leptin and IGF-I, leptin gene expression, and serum binding of [126I]ovine leptin in cattle during pubertal development, as well as leptin gene expression and circulating concentrations of leptin during the estrous cycle and different calendar seasons, were investigated. Multivariate regression analysis was utilized to evaluate temporal changes in BW, leptin mRNA, and serum concentrations of IGF-I and leptin normalized to the week of puberty (Exp. 1). Body weight accounted for most of the variation associated with the onset of puberty in the full regression model (R2 = 0.99; P < 0.01). However, serum leptin was closely related to changes in BW (r = 0.85; P < 0.02) and in the absence of BW was most predictive of pubertal onset (r2 = 0.73; P < 0.01). Mean concentrations of leptin increased (P < 0.0001) linearly from 16 wk before until the wk of pubertal ovulation in yearling heifers reaching sexual maturation from early spring to midsummer. Leptin mRNA transformed to a percent of the value at puberty increased (P < 0.02) as puberty approached, but serum leptin and leptin mRNA values were not well correlated. We found no evidence of leptin-binding proteins in serum of developing heifers. Combined mean serum concentrations of IGF-I (ng/mL) during periods III and IV (-9 wk to wk of puberty; 216.6 +/- 9) were 21% higher (P < 0.0001) than combined mean concentrations of IGF-I during periods I and II (-19 to wk of puberty; 193 +/- 10). In mature heifers and cows (Exp. 2), serum leptin tended to decrease (P = 0.10) during the late luteal/early follicular phase of the estrous cycle, which corresponded to a reduction (P < 0.03) in adipocyte leptin gene expression. In mature ovariectomized cows, serum concentrations of leptin increased (P < 0.001) by 34% from early winter to the summer solstice and remained unchanged throughout the remainder of the year (Exp. 3). Results from these studies indicate that marked increases in both circulating leptin and leptin gene expression occur in developing heifers during pubertal development and are associated with increases in serum IGF-I and BW. Seasonal effects on circulating leptin observed in mature cows from winter to summer could also plausibly account for a portion of the prepubertal rise in serum leptin observed in heifers.  相似文献
