The objective of this study was to investigate the relationship between molecular marker diversity and heterosis in both intra-and inter-sub-specific hybrids of rice to evaluate the feasibility of predicting hybrid performance using molecular markers. Eleven elite lines were intermated resulting in a diallel set including 10 indica × indica, 15 japonica × japonica and 30 indica × japonica crosses. The F1 hybrids and parents were evaluated for agronomic performance in a replicated field trial. The parental lines were tested for DNA polymorphisms with 113 restriction fragment length polymorphism (RFLP) probes covering the 12 rice chromosomes. Inter-subspecific crosses showed better performance and higher heterosis than intrasubspecific hybrids. Correlations of marker heterozygosity with hybrid performance and heterosis differed considerably between the two subspecies; they were higher in crosses within japonica subspecies than within indica subspecies. Very little correlation was detected in intersubspecific crosses. It was concluded that relationships between marker heterozygosity and hybrid performance were complex owing to germplasm diversity and the complexity of the genetic basis of heterosis. The implications of the results in predicting hybrid performance using molecular markers are discussed.  相似文献

An investigation was undertaken to develop new stable thermo‐sensitive genic male sterility (TGMS) rice lines in intermated progenies (IMPs) of TGMS lines by using an anther clearing technique. The results indicate that both pre‐ and post‐meiotic genetic systems operate during anther development for the expression of sterility in TGMS lines. In all the TGMS lines, sterile anthers were small with empty pollen grains of irregular shape, except for TS 16, which showed pollen‐free anthers. This indicates that the sensitive stage of TS 16 is around stage IV (stamen and pistil primordia) of panicle development. Distinct differences were observed between sterile and fertile phases with respect to anther size, shape and colour of the pollen grains in TS 18 and TS 29. The pollen grains at the sterile phase were small and irregular in shape while in the fertile phase they were plump and larger with a yellow colour, establishing that the occurrence of sterility in TS 18 and TS 29 is post‐meiotic. Three distinct classes of pollen fertility percentage viz. <20%, 50‐70% and >90% were observed in IMPs. Anther clearing in IMPs showed distinct developmental patterns of pollen production with respect to distinct classes of pollen fertility. Less than 20% pollen fertility was observed in hybrids such as TS 15 × TS 16, TS 15 × Co 47, TS 18 × TS 16 and TS 18 × Co 47 which hold promise for developing new TGMS lines with a good plant type and acceptable quality.  相似文献

Summary Wide compatible varieties (WCVs) show normal spikelet fertility in crosses with Indica and Japonica rice varieties. Crosses of Indica and Japonica varieties frequently show high spikelet sterility which prevents exploitation of heterosis for grain yield. We screened 41 rice varieties for the wide compatibility trait by crossing each with three Indica and three Japonica testers. Varieties giving fertile F1 hybrids with both groups of testers were classified as WCVs. Seven varieties viz., BPI-76 (Indica); N 22; Lambayeque-1 and Dular (Aus); Moroberekan, Palawan and Fossa HV (Japonicas), were identified as WCVs. The frequency of WCVs was higher among Aus and Japonicas. The wide compatibility trait in varieties: Dular and Moroberekan was controlled by a single dominant gene linked with the Est-2 and Amp-3 loci (mean recombination 32.0%). Est-2 and Amp-3 showed complete linkage. Pgi-2 was found to be linked with Est-2 and Amp-3 (mean recombination 16.1%). Est-2 and Amp-3, showed a tighter linkage with C + (mean recombination 4.1%). Pgi-2 showed a lower linkage with C + (mean recombination 17.3%). The recombination values between the WC gene in Dular and C + was much higher than those reported in Japan for the WC gene (S5 n) from Ketan Nangka. It is possible that the WC gene from Dular is different from that in Ketan Nangka. Linkage intensities with the WC gene were not strong enough to be of use for indirect selection for the wide compatibility trait. A search for a more closely linked isozyme or DNA marker was proposed.  相似文献

Presence of substantial heterosis and economic hybrid seed production are two most desirable components for success of any commercial hybrid breeding programme. Thermosensitive genic male sterile (TGMS) lines of rice, in this regard, have tremendous potential in realizing further quantum jump in yield and economical hybrid seed cost. Analyses for combining ability and heterosis over optimum (120N : 60P2O5 : 40K2O kg/ha) and high (200N : 90P2O5 : 60K2O kg/ha) fertility environments for six traits were made in 2 years (2001 and 2002) using 120 hybrids of inter‐ and intra‐subspecific nature derived from hybridization of 30 elite indica TGMS lines and four cultivars, viz., ‘Pant Dhan 4’ and ‘Ajaya’ (I = indica), ‘Taichung 65’ (J = japonica) and ‘IR 65598‐112‐2’ (TJ = tropical japonica) in line × tester mating design. Predominance of non‐additive genetic variance suggested good prospects of hybrid breeding. Pooled analysis revealed highly significant variances for lines, general combining ability (GCA), specific combining ability (SCA) and line x tester. TGMS line 365‐8S was the best general combiner for all the six traits including grain yield. Trend of relative mid‐parent heterosis for grain yield, panicle length, grain number per panicle and earliness in flowering was I/TJ > I/J > I/I. For panicle number per plant and 1000‐grain weight, trends were I/TJ > I/I > I/J and I/I > I/TJ > I/J, respectively. Grain yield recorded heterosis of 49.3%, 71.9% and 92.7% for I/I, I/J and I/TJ hybrid groups respectively. Effect of environments on the hybrid performance indicated better response of hybrids at high fertilizer dose. Study suggests greater prospects of combining improved japonica and tropical japonica germplasms having wide compatible gene with indica TGMS lines for exploitation of intersubspecific heterosis.  相似文献

Anther culture of recalcitrant indica × Basmati rice hybrids   总被引:1,自引:1,他引:0  
Fertile, green, di-haploid plants were obtained at high frequencies from several indica × Basmati rice F1 hybrids and/or F2 plant populations using an improved anther culture procedure. Anthers from cold-pretreated (10 °C for 10 d) panicles of six indica (HKR120, HKR86-3, HKR86-217, PR106, Gobind andCH2 double dwarf) and two Basmati rice (Basmati 370,Taraori Basmati) varieties and 14 heterotic indica ×Basmati F1/F2 hybrids were cultured in modified agarose-solidified N6M, Heh5M and RZM media. Best callus induction frequencies (2.6–78%) were obtained in RZM medium containing 4% (w/v) maltose,2,4-D, NAA and kinetin. F2 plants compared to F1 hybrids and parental rice varieties, were more responsive to anther culture. Androgenesis frequencies of 31–78% were obtained for indica × Basmati F2 plants in RZM medium in just 30 d which are comparable to or higher than that reported for japonica rice varieties and hybrids involving japonica rice parent(s). Agarose (1.0% w/v)-solidified MS medium containing 3.0% maltose, kinetin, BAP, and NAA, induced green shoot regeneration in 0–51% of the anther-derived callide pending upon the genotype. High plant regeneration frequencies (67–337 green plants per 1000 anthers)were obtained from anther calli of several F1hybrids (Gobind × Basmati 370 and HKR120 ×Taraori Basmati) and F2 plants (Gobind × Basmati370, Gobind × Taraori Basmati, HKR86-3 × TaraoriBasmati). A sample of 498 plants obtained from the above hybrids, were transferred to pots with>90% survival; 8–78% of these plants had >5%spikelet fertility and were diploid. In addition,18% of the haploid plants could be diploidized by submerging in 0.1% colchicine solution for 16–18 h. The improved anther culture procedure reported here, resulted in several fold increase in the recovery of green plants from recalcitrant indica × Basmati rice hybrids compared to previous published procedures. The study may accelerate the introgression of desirable genes from indica into Basmati rice using anther culture as a breeding tool. Embryo sac abortion occurs frequently in the spikelet of indica/japonica hybrids in rice. To provide a comprehensive understanding of embryo sac sterility in typical indica/japonica hybrids, a simple eosin B staining procedure for use with confocal microscopy was developed to observe the mature embryo sacs of 35 indica/japonica F1s making up an incomplete diallel cross design (5 × 7). A diversity of abnormal embryo sacs existed in the hybrids. The main types were classified into five categories: (i) embryo sac degeneration (ESD), (ii) embryo sac without egg apparatus, (iii) embryo sac without female germ unit, (iv) embryo sac with abnormal polar nuclei and (v) abnormal small embryo sac (ASES). The frequencies of abnormal embryo sacs in the 35 hybrids ranged from 26.0% to 76.3%, with an average of 48.6%. The highest frequency of abnormality was 31.3% ESD. The frequencies of various abnormalities varied with parental genotype and environment.  相似文献

The purpose of the study was to evaluate the relationship between the genetic distances (GD) and phenotypic distances (PD) of parents and the specific combining ability (SCA) and heterosis effects. The experiment comprised 18 parental genotypes of wheat (Triticum aestivum L.) and 76 F2 hybrids, obtained after crossing in a line × tester scheme. Parents and hybrids were examined in a field experiment conducted in a block design with three replications. SCA as well as mid-parent heterosis effects were estimated for selected morphological and technological traits. PDs and GDs were investigated between pairs of parental genotypes. GD between parental genotypes was evaluated by using randomly amplified polymorphic DNA markers. Heterosis was observed in all hybrids, and protein content exhibited the highest heterosis among the seven examined traits. The relationship between PD and GD as well as the SCA and mid-parent heterosis effects were evaluated using correlation coefficient. The correlation between PD, SCA and heterosis were low and not significant for the examined traits, whereas the correlation between SCA, heterosis and GDs were significant for protein content and rheological properties. The results indicate that GDs between parents can be used to predict performance of hybrids for selected technological traits.  相似文献

Reproductive fertility traits were studied in the reciprocal hybrids of the eggplant(Solanum melongena L.) and S. aethiopicum L. Gilo Group, and in synthetic amphidiploids to discover whether fertility in these reciprocal hybrids was restored by chromosome doubling. Isozyme and RAPD analyses confirmed hybridity of the hybrids and amphidiploids. Analyses of chloroplast and mitochondrial DNAs confirmed that the cytoplasm of each of the hybrids and amphidiploids was from the maternal parent. Pollen sterility of S. melongena × S. aethiopicum Gilo Group [F1 (Mel × Aet)] was restored by chromosome doubling, while the reciprocal hybrid S. aethiopicum Gilo Group ×S. melongena [F1 (Aet × Mel)]and its amphidiploid did not produce any pollen grains; their microspores degenerated without being released from tetrads. Hence the cytoplasm of S. aethiopicum Gilo Group seems to beresponsible for their pollen-non-formation type sterility of the hybrid. Both the F1 hybrids did not set any fruits by either selfing or backcrossing, while their amphidiploids set fruits after pollinating with pollen from the amphidiploid of F1 (Mel × Aet). Seeds obtained from both the amphidiploids germinated normally. Chromosome doubling has been effective in restoring fertility of the hybrids. A breeding program was initiated at the International Rice Research Institute (IRRI) in 1990 to develop thermosensitive genic male sterile (TGMS) rice lines for developing two-line rice hybrids for the tropics. The TGMS trait was transferred from a temperate japonica TGMS mutant, Norin PL 12, to indica and tropical japonica rice varieties using the pedigree selection procedure. Six new TGMS rice lines adapted to tropical conditions were developed which showed complete pollen and spikelet sterility when maximum temperature was higher than 30 °C 1–2 week after panicle initiation. However, up to 85.5% spikelet fertility was observed when these lines were exposed to 26–29 °C during the critical stage. Using two of these TGMS lines, some heterotic rice hybrids showing 1–1.6 t/ha higher grain yield than the inbred check varieties were identified in unreplicated observational yield trial conducted at IRRI. Two of the six two-line hybrids yielded significantly higher than the check variety in a replicated preliminary yield trial. An eosin B staining procedure for use with confocal microscopy (WE‐CLSM) was used to examine the mature embryo sacs in 24 typical autotetraploid (2n = 4x = 48) indica/japonica hybrids and to compare with their original diploid (2n = 2x = 24) indica/japonica hybrids in rice. Four of the 24 hybrids showed >80% embryo sac fertility, and 13 hybrids over 70%. The average embryo sac fertility of the 24 autotetraploid indica/japonica hybrids was 68.08%, which was 17.33% higher than that in diploid hybrids. When compared with the diploid hybrids, 79.17% of the autotetraploid hybrids had higher embryo sac fertility. A diversity of abnormal embryo sacs occurred in autotetraploid indica/japonica hybrids. Embryo sac abnormalities were similar for both autotetraploid and diploid hybrid, but their frequencies were different. Some novel abnormal types were found in autotetraploid hybrids, such as enlarged egg apparatus. The embryo sac fertilities and frequencies of various abnormalities in autotetraploid hybrids varied with parental genotype and environment. The average seed set of the autotetraploid hybrids were higher than their diploid hybrids in different growing seasons.  相似文献

The thermosensitive genic male sterility (TGMS) system has great potential for revolutionizing hybrid rice production through simple, less expensive and more efficient seed production technology. For the successful utilization of this novel male sterility system, knowledge of the breeding and fertility behaviour of a TGMS line is essential. In this study, the fertility transformation behaviour, the critical fertility and sterility temperatures and the mode of inheritance of male sterility were studied for a new TGMS line, TS6, identified at Tamil Nadu Agricultural University, Coimbatore, India. The pollen and spikelet fertilities recorded on plants raised at fortnightly intervals revealed that this line was completely sterile for 78 consecutive days (35/22 to 32/23°C, maximum/minimum temperatures) and reverted to fertile when the temperature was 30/18°C. It remained fertile continuously for 69 days and the maximum pollen and spikelet fertilities recorded were 75 and 70%, respectively. The fertility was highly influenced by daily maximum temperature followed by average and minimum temperatures. It was not influenced by relative humidity, sunshine hours or photoperiod. The critical temperature inducing sterility and fertility was 26.7 and 25.5°C, respectively. The male sterility in TS6 was inherited as a monogenic recessive in the F2 and BC1 populations of TS6 × MRST9 as well as TS6 × IR68281B. Using bulked segregant analysis on an F2 population of TS6 × MRST9, an RAPD marker, OPC052962, was identified to be associated with TGMS in TS6.  相似文献

Cardamom is an important high value spice crop. Hybrid breeding is discussed for some time, but there is no information for cardamom. Eight genetically diverse cardamom lines were crossed in a diallel to evaluate the performance of hybrids and determine heterosis over mid-parent, better-parent and standard control for yield and yield components. The study was undertaken for a period of 3 years from 1988 to 2001 (Experiment I). Hybrids generally showed good overall performance for most of the characteristics compared with parents. Ten of the 56 hybrids significantly out-yielded the standard control (RR 1). Substantial mid-parent (MPH), better-parent (BPH) and standard (SH) heterosis were observed for the majority of the characteristics studied. Further evaluation of selected hybrids for yield and disease resistance during 2002–2007 (Experiment II) led to the identification of two cross combinations (CCS 1 × NKE 19 and RR 1 × NKE 12) with high yield and mosaic resistance. The study revealed that cardamom hybrids with high yield potential, desirable quality characteristics and mosaic resistance can be developed from appropriate parents through heterosis breeding. This is the first report of heterosis in cardamom for yield and yield contributing characteristics.  相似文献

Heterotic response for total yield among five cultivar-groups of summer squash, Cocozelle, Vegetable Marrow, Zucchini, Crookneck and Straightneck, was studied in two sets of diallel crosses evaluated in two harvest seasons. Each diallel cross included two representatives of each the five cultivar-groups. Griffing and diallel GGE biplot (genotype main effect and interaction) analyses were carried out. The relative importance of the specific combining ability variance on total variance was greater than that reported earlier for intragroup crosses. Highly significant heterosis was manifested only in combinations of Crooknecks or Straightnecks crossed to Cocozelles, Vegetable Marrows or Zucchinis. GGE biplots showed a consistent arrangement where testers of ssp. texana interacted with cultivars of ssp. pepo and vice versa. We propose that the heterotic pattern of texana-pepo crosses could be potentially used to achieve increased yields in hybrids by introgressing one group into another in a manner that would maintain fruit attributes of the cultivar-group. This study quantified the magnitude of heterosis in pearl millet (Pennisetum glaucum) topcross hybrids produced by crossing 16 diverse landraces and three high yielding open-pollinating varieties on two homozygous male-sterile lines. Hybrids and pollinators were grown in 12 year ×;location combinations in India that were grouped into three zones. Genetic components of variance quantifying the differences among these hybrids were estimated. The hybrids showed a conspicuous heterosis for grain yield, earliness and biomass yield but not for straw yield. The level and direction of heterosis for time to flowering depended strongly on the earliness of the male-sterile line. In the terminal drought stress zone hybrids made on the early maturing male-sterile line 843A had the highest level of heterosis for grain yield (88%). This was partly due to escape from terminal stress. In the other two zones the heterosis for grain yield was on average 30%. Heterosis for biomass yield and biomass yield per day was on average also positive in all three zones. For all traits, except time to flowering and biomass yield per day, pollinator effects were the only significant source of variation. Differences between hybrids were mostly caused by additive genetic effects. Significant amount of heterosis observed in landrace-based topcross hybrids for grain yield and other productivity-related traits suggested that substantial improvement in pearl millet productivity in and environments can be obtained by topcrossing locally adapted landraces on suitable male-sterile lines. Interspecific hybrids from crosses between H. syriacus × H. paramutabilis and H. syriacus × H. sinosyriacus were obtained. In both cases unilateral incongruity was observed and reciprocal crosses yielded no fruits. In vitro embryo rescue, 11 weeks after pollination, increased the germination capacity of H. syriacus × H. sinosyriacus embryos, while this was not the case for H. syriacus × H. paramutabilis embryos. However, a lot of the generated H. syriacus × H. sinosyriacus seedlings were lost due to variegated and total albinism. In the progeny of H. syriacus ‘Oiseau Bleu’ × H. paramutabilis about 95% of the seedlings had an intermediate flower and leaf morphology compared to both parent plants. Leaves on the adult F1 hybrids showed a polymorphism. In total four different leaf types could be observed on the same plant. This leaf polymorphism also was seen in the progeny of H. syriacus ‘Melwhite’ × H. sinosyriacus ‘Lilac Queen’. In this progeny about 50% of the seedlings had an intermediate flower and leaf morphology compared with the parent plants. The hybrid nature of the seedlings of both progenies was also confirmed by AFLP analysis. Despite the low germination vigour of the pollen of the hybrids, a small F2 generation was obtained from H. syriacus ‘Oiseau Bleu’ × H. paramutabilis.  相似文献

The objectives of this study were to determine the crossing performance of highland maize inbred lines for grain yield, days to silk and plant height; estimate genetic distance (GD) among the inbred lines and in association with tester parents, and to investigate the relationship of GD with hybrid performance and midparent heterosis (MPH). A total of 26 inbred lines were crossed with six (population and line) testers in a factorial-mating scheme. The F1’s and the parents were evaluated at five locations in Ethiopia. Nine amplified fragment length polymorphism (AFLP) primer pairs were used to genotype all the parents. The F1’s were found to vary widely for grain yield and other traits measured. Yield superiority of more than 30% over the best hybrid check was obtained for some testcross hybrids. Midparent heterosis on average was moderate for grain yield and, plant height. And for days to silking, MPH values were mostly negative. Mean GD values determined from the inbred lines by population tester (0.680) and line tester (0.661) combinations were not significantly different. Cluster analysis separated the tester parents from the corresponding inbred lines. AFLP grouping of the inbred lines was in agreement with their pedigree records. Genetic distances derived from the inbred lines × all testers and from the population testers’ sub-group were not positively correlated with hybrid performance and MPH for most traits. In contrast, correlations of GDs involving the line testers’ sub-group with F1’s and MPH were significantly positive but with low magnitude to be of predictive value.  相似文献

The Indica‐Japonica differentiation of three photoperiod‐sensitive and/or thermosensitive genetic male‐sterile rice (PGMS or TGMS, respectively) lines and 47 male parental lines from seven ecotypes were studied for their restriction fragment length polymorphism marker data to determine which ecotype crosses with the three PGMS and/or TGMS lines could lead to higher yield potential, and to estimate the relationship between the Indica‐Japonica differentiation of parents and heterosis in grain yield and its components. The results indicated that hybrids derived from ‘N422s’ and the early‐middle ripening Indica varieties from southern China, and hybrids between ‘Pei'ai64s’ and three Japonica ecotypes, including North‐eastern Japonica varieties, restoring lines of Japonica hybrid rice and north China Japonica varieties, showed the highest grain yields. There was less variation of yield among the F1s between ‘108s’ and the seven ecotypes than among the other F1s. Highly significant positive correlations between heterosis of the F1 yield and genetic distance of the parents were detected, although the correlation between F1 yield performance and genetic distance did not reach a significant level. Considerable variation of correlation between heterosis and genetic distance was also detected in the Indica × Indica crosses and Indica × Japonica crosses. There was much higher correlation (r = 0.63) between the F1 yield performance and the genetic distance of parents in the Indica × Indica crosses than in the others. It is proposed that a genetic distance of 0.4‐0.8 between the two parents of hybrid rice might be appropriate not only for F1 performance, but also for heterosis.  相似文献

Summary Genetic divergence was investigated among 17 rice varieties known to possess some degree of cold tolerance at different growth stages. The 17 varieties and their 102 F1 hybrids with 6 male testers were studied for isozyme variation at 15 loci, spikelet fertility, and degree of cold tolerance at various stages. Multivariate analyses of the data provided several schemes of divergence based on various sources of evidence. All schemes gave similar results, and separated the varieties into japonica and indica groups. The japonica group displayed specific isozymes, a low F1 fertility with indica testers, and a high degree of cold tolerance which was expressed in the F1 progenies. The indica group displayed contrasting specific isozymes, a high F1 fertility with indica testers, and a moderate to low degree of cold tolerance which was not expressed in the F1 progenies. One variety, ARC 6000, displayed unique traits in most schemes and was classified into a distinct type based on the isozymes. The results emphasize that cold tolerance is a major trait for classification of rice into two varietal groups and that proper characterization of potential donors is essential in breeding. Isozyme studies are useful tools for this purpose.  相似文献

Sorghum head bug, Calocoris angustatus Lethiery is one of the most important pests of grain sorghum in India. Head bug damage increases the severity of grain molds, which renders the grain unfit for human consumption. Therefore, we studied the gene action for resistance to head bugs and grain molds in a diverse array of male-sterile lines and testers in a line × tester mating design under natural infestation. Mean squares for parents, parents vs crosses, lines, testers, and lines × testers were significant for head bug damage and grain mold severity. General combining ability (GCA) effects were significant and negative for ICSA 88019 for head bug damage, and ICSA 88019 and ICSA 88020 for grain molds (except for ICSA 88020 in 1993). General combining ability effects were positive for ICSA 42 and 296 A. GCA effects of lines and testers for head bug damage and grain mold severity were in the same direction (+ve or −ve). Head bug damage in the grain was significantly correlated with grain mold severity. Testers IS 8891, IS 15107, and TAM 2566 (with colored grain and less susceptibility to molds) produced mold-resistant hybrids in combination with all the male-sterile lines, while the reverse was true in the case of Swarna and ICSV 112. Resistance to head bugs showed dominance to partial dominance type of gene action, while in the case of grain molds, it showed dominance to over dominance. Resistance to these pests is governed by both additive and nonadditive types of gene action. The implications of these results are discussed in relation to need for crop improvement in sorghum. Fourteen genetically diverse inbred lines comprising of four genic male sterile lines and 10 often pollinated lines as testers were crossed following ‘line × tester’ mating design to generate 40 F1s. These F1s, parents and check ‘CH-27’ were evaluated in α-lattice square design, replicated thrice during summer seasons of 2020 and 2021. The male sterile line DPChMS 9-2 was good general combiner for marketable fruit yield and eight other component traits while testers namely, DPCh 10, PBC 535, VVG, Him Palam Mirch-2 and DPCh 40 showed significant GCA for majority of yield related traits. The hybrids DPChMS 9-2 × HPM-2, DPChMS 9-2 × DPCh 40, DPChMS 9-2 × DPCh 101, DPChMS 9-2× VVG and DPChMS 26-1× HPM-1 were the most desirable based on specific combining ability (SCA) and economic heterosis for fruit yield and other important traits. Heterosis was positively correlated with SCA indicating prediction of potential hybrids can be made based on both these parameters simultaneously. Present study provides an opportunity to exploit top hybrids for stability and commercial cultivation.  相似文献
