The precise level of environmental control in vitro may aid in identifying genetically superior plant germplasm for rooting characteristics (RC) linked to increased foraging for plant nitrogen (N). The objectives of this research were to determine the phenotypic variation in root morphological responses of 49 Solanum chacoense (chc), 30 Solanum tuberosum Group Phureja – Solanum tuberosum Group Stenotomum (phu-stn), and three Solanum tuberosum (tbr) genotypes to 1.0 and 0.5 N rate in vitro for 28 d, and identify genotypes with superior RC. The 0.5 N significantly increased density of root length, surface area, and tips. All RC were significantly greater in chc than in either phu-stn or tbr. Based upon clustering on root length, surface area, and volume, the cluster with the greatest rooting values consisted of eight chc genotypes that may be utilized to initiate a breeding program to improve RC in potato.  相似文献

Background

The DEFECTIVE IN OUTER CELL LAYER SPECIFICATION 1 (DOCS1) gene belongs to the Leucine-Rich Repeat Receptor-Like Kinase (LRR-RLK) subfamily. It has been discovered few years ago in Oryza sativa (rice) in a screen to isolate mutants with defects in sensitivity to aluminum. The c68 (docs1–1) mutant possessed a nonsense mutation in the C-terminal part of the DOCS1 kinase domain.

Findings

We have generated a new loss-of-function mutation in the DOCS1 gene (docs1–2) using the CRISPR-Cas9 technology. This new loss-of-function mutant and docs1–1 present similar phenotypes suggesting the original docs1–1 was a null allele. Besides the aluminum sensitivity phenotype, both docs1 mutants shared also several root phenotypes described previously: less root hairs and mixed identities of the outer cell layers. Moreover, our new results suggest that DOCS1 could also play a role in root cap development. We hypothesized these docs1 root phenotypes may affect gravity responses. As expected, in seedlings, the early gravitropic response was delayed. Furthermore, at adult stage, the root gravitropic set angle of docs1 mutants was also affected since docs1 mutant plants displayed larger root cone angles.

Conclusions

All these observations add new insights into the DOCS1 gene function in gravitropic responses at several stages of plant development.

Powdery scab caused by Spongospora subterranea f. sp. subterranea (Sss) causes extensive losses in potato production systems globally. Two pot experiments were established in the greenhouse in summer 2013 and winter 2014 to evaluate the effectiveness of different soil chemicals, fumigant, amendments and biological control agents (BCAs) against Sss in the rhizospheric soil, potato roots and tubers. The study used visual assessment methods to assess the effect of treatments on root galling and zoosporangia production, and qPCR to measure Sss concentration in the soil and in the potato roots and tubers. All six soil treatments, namely metam sodium, fluazinam, ZincMax, calcium cyanamide, Biocult and a combination of Bacillus subtilis and Trichoderma asperellum recorded significantly (P < 0.05) lower numbers of zoosporangia in the roots compared to the untreated control. The same effect was observed on the concentration of Sss DNA in the roots at tuber initiation. A more diverse picture was obtained when root gall scores at tuber initiation and Sss DNA in the rhizospheric soil at tuber initiation and harvesting were compared. Significant differences (P < 0.05) were also noted in disease severity, disease incidence, and tuber yield between metam sodium, fluazinam, ZincMax, calcium cyanamide and the untreated control. Calcium cyanamide gave the highest tuber yield. The study demonstrated the potential of soil treatments such as metam sodium, fluazinam, ZincMax and calcium cyanamide in managing Sss in potatoes by reducing the pathogen both in the rhizospheric soil and the roots of the potato plant.  相似文献

Lupinus albus seeds contain conglutin gamma (Cγ) protein, which exerts a hypoglycemic effect and positively modifies proteins involved in glucose homeostasis. Cγ could potentially be used to manage patients with impaired glucose metabolism, but there remains a need to evaluate its effects on hepatic glucose production. The present study aimed to analyze G6pc, Fbp1, and Pck1 gene expressions in two experimental animal models of impaired glucose metabolism. We also evaluated hepatic and renal tissue integrity following Cγ treatment. To generate an insulin resistance model, male Wistar rats were provided 30% sucrose solution ad libitum for 20 weeks. To generate a type 2 diabetes model (STZ), five-day-old rats were intraperitoneally injected with streptozotocin (150 mg/kg). Each animal model was randomized into three subgroups that received the following oral treatments daily for one week: 0.9% w/v NaCl (vehicle; IR-Ctrl and STZ-Ctrl); metformin 300 mg/kg (IR-Met and STZ-Met); and Cγ 150 mg/kg (IR-Cγ and STZ-Cγ). Biochemical parameters were assessed pre- and post-treatment using colorimetric or enzymatic methods. We also performed histological analysis of hepatic and renal tissue. G6pc, Fbp1, and Pck1 gene expressions were quantified using real-time PCR. No histological changes were observed in any group. Post-treatment G6pc gene expression was decreased in the IR-Cγ and STZ-Cγ groups. Post-treatment Fbp1 and Pck1 gene expressions were reduced in the IR-Cγ group but increased in STZ-Cγ animals. Overall, these findings suggest that Cγ is involved in reducing hepatic glucose production, mainly through G6pc inhibition in impaired glucose metabolism disorders.  相似文献

Late blight, caused by the oomycete Phytophthora infestans (Mont.) de Bary, is a devastating disease in potato and tomato and causes yield and quality losses worldwide. The disease first emerged in central America and has since spread in North America including the United States and Canada. Several new genotypes of P. infestans have recently emerged, including US-22, US-23 and US-24. Due to significant economic and environmental impacts, there has been an increasing interest in the rapid identification of P. infestans genotypes. In addition to providing details regarding the various phenotypic characteristics such as fungicide resistance, host preference, and pathogenicity associated with various P. infestans genotypes, information related to pathogen movement and potential recombination may also be determined from the genetic analyses. Restriction fragment length polymorphism (RFLP) analysis with the RG57 loci is one of the most reliable procedures used to genotype P. infestans. However, the RFLP procedure requires propagation and isolation of the pathogen and relatively large amounts of DNA. Isolation of the late blight pathogen is sometimes impossible due to the poor condition of the infected tissues or the presence of fungicide residues. In this study, we describe a procedure to identify P. infestans at the molecular level in planta using terminal restriction fragment length polymorphism (T-RFLP) of the RG57 loci. This T-RFLP assay is sufficiently sensitive to detect and differentiate P. infestans genotypes directly in planta without propagation and isolation of the pathogen, to facilitate the timely implementation of best management practices.  相似文献

The dietary intake and control of blood glucose levels are very important in hyperglycemic patients and α-glucosidase inhibitors are a cost-effective means to preventing the progression of diabetes. In search of a natural inhibitor from food materials, α-glucosidase inhibitory activity and the anti-hyperglycemic effects of a brown alga, Ecklonia stolonifera, were investigated using non-insulin dependent diabetic mice. Methanolic extract of E. stolonifera (MEE), which contains a high content of polyphenols, showed strong inhibition of α-glucosidase in vitro. Male KK-Ay mice, a genetically non-insulin dependent diabetic model, showed hyperglycemia with aging, but the ingestion of MEE suppressed the increase in plasma glucose and lipid peroxidation levels in unfasted KK-Ay mice dose dependently. In KK-Ay mice, which were fed the MEE diet for 4 weeks, MEE moderated the elevation of plasma glucose levels after the oral administration of maltose. The polyphenols in MEE were estimated to be phlorotannins by HPLC-PDA and LC/MS analyses. These results demonstrate that E. stolonifera, seaweed typically used as a health food, has strong antidiabetic and antioxidant effects in vivo, thus, it may have beneficial properties in the prevention of diabetes and could be useful in the development of an antidiabetic pharmaceutical and functional food.  相似文献

Platycodon grandiflorum (PG), a species of herbaceous flowering perennial plant of the family Campanulaceae, has been used as a traditional oriental medicine for bronchitis, asthma, pulmonary tuberculosis, diabetes, hepatic fibrosis, bone disorders and many others similar diseases and as a food supplement. For the primary profiling of PG gas chromatography coupled with high resolution – time of flight mass spectrometry (GC/HR-TOF MS) was used as an analytical tool. A comparison of optimal extraction of metabolites was carried out with a number of solvents [hexane, methylene chloride, methanol, ethanol, methanol: ethanol (70:30, v:v)]. In extracts with methanol: ethanol (70:30 v:v) were detected higher amounts of metabolites than with other solvents. Principal component analysis (PCA) and partial least-squares discriminant analysis (PLS-DA) plots showed significant differences between the diploid and tetraploid metabolite profiles. Extracts of tetraploid showed higher amounts of amino acids, while extracts of diploid contained more organic acids and sugars.

Potato mop top virus (PMTV) induced necrosis can cause tuber quality loss at harvest and storage. Stored potatoes help maintain a constant supply of potatoes to the market and processing industry. PMTV-induced necrosis continues to develop during storage and appropriate timelines are needed for growers to make marketing decisions of their potatoes before incurring any significant quality losses. Survival analysis was used to estimate the time to event occurrence (PMTV-induced necrosis) in four (red-, russet-, white-, and yellow-skinned) potato market types across six post-harvest assessments conducted over two years. At each assessment the presence and absence of PMTV-induced necrosis was recorded and probability of tuber survival was estimated. Survival curves were significantly different among the four market type potato cultivars (Log-Rank test, P < 0.0005). Red- and russet-market type cultivars showed low and high survival probability, respectively, demonstrating that red cultivars need priority marketing. The survival probabilities decreased with increased storage time during both years, indicating that PMTV-induced necrosis development is dependent on potato cultivar and post-harvest storage. The median (50% of tubers with symptoms) survival times were estimated as 167 and 214 days for red- and other market type potato cultivars. The information from this study could potentially help growers regulate storage times for their cultivars to minimize tuber quality loss due to PMTV-induced tuber necrosis.  相似文献

Background

Combining ability is a measure for selecting elite parents that make the highest contributions to hybrid performance. However, the genetic bases of combining ability and how they contributed to heterosis is seldomly known.

Results

We constructed a both NCII and NCIII population derived from an indica-japonica cross to study the relationship among parental performance, combining ability and hybrid performance of 11 agronomic traits. Among them, specific combining ability is more important to grain yield than parental performance and general combining ability. We performed linkage analyses to phenotypic values and combining ability of all 11 traits in Doubled haploid lines and its two backcross populations and identified 108 QTLs in total. Among these QTLs, four known loci, Sd1, Ghd7, Ghd8 and DEP1 contribute a lot to GCA effects of agronomic traits except grain yield and seed setting rate. Three QTLs, Ghd8, S5 and qS12, contribute a lot to SCA effects of grain yield and present overdominace.

Conclusions

Our study provides insights into the genetic bases of combining ability and heterosis and will promote the improvements of indica-japonica hybrid breeding.

Use of nutrient efficient crop species or cultivars within species in combination with improved crop production practices offer the best option for meeting the future food requirements of growing world populations. Efficiency improvement has a key role for nutrient use and it improves both productivity of yield and minimizes risks of losses that potentially harm the environment. To improve nutrient use efficiency (NUE), one strategy is to select cultivars of crops with high NUE. Both nitrogen (N) and phosphorus (P) are among essential macronutrients commonly applied to agricultural crops to optimize yields. Evaluation of potato cultivars with high N and P efficiency is essential for sustainable production of the crop. In Ethiopia, however, little research has been done to elucidate better NUE of potato crop until recently. This study was, thus, designed to evaluate selected potato cultivars (Belete, Gudene, Jalene, Marachere and one local check) for NP use efficiency under low levels of the nutrients in Wolaita Sodo University, Ethiopia during 2014 and 2015. Factorial combination of 3 levels of each N (0, 55.5 and 111 kg N ha?1 ) and P (0, 19.5 and 39 kg P ha?1 ) were assigned to the main plots and the 5 cultivars to subplots of split-plot design in three replications. Efficiency indices such as agronomic efficiency, physiological efficiency and recovery efficiency were computed for combined NP nutrients. Results showed that agronomic nitrogen use efficiency, apparent phosphorus use efficiency and phosphorus use efficiencies were significantly influenced by the interaction effect of rates of NP nutrition and variety. Application of NP more than 55.5:19.5 kg ha?1 will not beneficial due to their less or equal use and recovery efficiencies to 55.5:19.5 kg NP ha?1. Belete was the most NP efficient variety followed by Jalene and Gudene in all efficiency indices. Therefore, Belete, Jalene and Gudene return better for NP at 55.5:19.5 kg ha?1 for economical use of fertilizes while obtaining satisfactory yield. It can be concluded that yield response variation of the varieties was related to their differences in NP uptake and use efficiencies; and the improved potato varieties were superior to local check in using NP nutrients efficiently.  相似文献

Poly(2-hydroxyethylmethacrylate) (PHEMA)/hydroxyapatite (HAP) nanocomposites were synthesized through a new route involving nano-sized HAP (nHAP) particles or modified nHAP mixed with monomer 2-hydroxyethylmethacrylate via in situ polymerization in supercritical carbon dioxide (scCO2). Fourier-transform infrared spectroscopy showed phosphate peak increased with nHAP content in composite. X-ray diffraction patterns of PHEMA/nHAP revealed the presence of crystallized nHAP. Thermogravimetric analysis showed that the ultimate nHAP content in PHEMA/nHAP composites is consistent with its initial amount. Scanning electron microscopy revealed that nanocomposite particles are much smaller than PHEMA particles. PHEMA/nHAP composites with average diameter of approximately 600 nm were obtained in scCO2 with 94 % yield. Mechanical properties of PHEMA/nHAP nanocomposites were better than those of PHEMA, and compressive modulus and strength of composites with 30 wt.% nHAP were 193 and 29 MPa, respectively. Nanocomposite adsorption toward bovine serum albumin was evaluated, and results indicated that analyte adsorption amount can reach up to 282 mg/g.  相似文献

In an experimental breeding scheme to improve late blight (Phytophthora infestans) and white potato cyst nematode (Globodera pallida) resistance of tetraploid potato over three generations of crossing and selection, 15 clones survived the final selection, and these were derived from 15 great-grandparents. There was no direct selection for resistance to Potato virus Y (PVY), but 14 out of the 15 great-grandparents were resistant to PVY and three had extreme resistance. Thirteen of the 15 descendants had PVY resistance and one extreme resistance. This was within the range expected for a random (unselected) sample from the genotypes of the great-grandparents. Hence, we found no evidence for any positive or negative association between PVY resistance and the attributes selected. The conclusion is that laborious selection is not required in every generation when many parents have PVY resistance, including some with more than one copy of a PVY resistance gene or resistance at more than one locus. However, in the future, determining the major virus resistance genes present in potential parents in each generation using diagnostic molecular markers would prevent susceptible × susceptible crosses being made and maximise the number of resistant × resistant ones.  相似文献

Macropores resulting from soil pedogenesis, biological activity, and agricultural practices play important roles in soil water, chemical and gas transport; however, seldom studies focus on the effect of soil macropores on CO2 behavior. In this study, a 150-day soil column incubation experiment was conducted to investigate the CO2 behavior in a homogeneous soil column and a soil column with an artificial macropore, which have the same total porosity. The results showed that the cumulative CO2 fluxes observed in the soil with a macropore (57.2 g m?2) were higher than those in the homogeneous soil (52.7 g m?2). The soil cumulative CO2 fluxes measured using column incubation can fit the kinetic model, and a higher carbon mineralization rate in the soil with the macropore was found. The results of the incubation experiment also suggest that macropore increased the gas diffusivities, and thus decreased the CO2 concentrations in the soil profile. This study proposed a simulation experiment and quantified the effect of macropore on soil CO2 behavior, which could help to understand the mechanism of CO2 emission from soil with macropores especially caused by agricultural practices.  相似文献

Camellia euphlebia (family, Theaceae) has been used for the prevention and treatment of cardiovascular diseases in Southern China. However, there has been no report on the hypolipidemic activity of Camellia euphlebia flower. This study evaluated the hypolipidemic activity of different preparation of Camellia euphlebia flower extracts using in vivo models. Mice intragastrically administered aqueous extract at 400 mg/kg dose or ethanol extract at 100 and 400 mg/kg doses of Camellia euphlebia flower for 28 days exhibited significant decreases in the levels of total cholesterol, triglycerides and low-density lipoprotein cholesterol, while displaying increased level of high-density lipoprotein cholesterol in the serum. The Camellia euphlebia flower extracts also improved the antioxidant ability of hyperlipidemic mice as well as protecting the animals against liver damage by lowering the level of glutamic-pyruvic transaminase activity. Furthermore, 400 mg/kg ethanol extract effectively down-regulated the mRNA levels of fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and glycerol-3-phosphate acyl transferase, suggesting that Camellia euphlebia flower extract may potentially inhibit lipid accumulation in the liver by regulating the expression of fatty acid synthase, 3-hydroxy-3-methylglutaryl CoA reductase and glycerol-3-phosphate acyl transferase. These results provided support for the potential hypolipidemic activity of Camellia euphlebia flower and could partly explain the basis of using Camellia euphlebia for the treatment of hyperlipidemia.  相似文献

Wild species-related germplasm is widely used to introduce new alleles and/or increase heterozygosity in cultivated species. Twenty-four SSR markers, specifically designed for cultivated potatoes, were evaluated to determine the extent of genetic variation within and among ten accessions of Solanum chacoense (chc). Fifteen of these markers were informative: there was no polymorphism in one of the markers, four of the markers showed evidence that more than one locus was being amplified, and the other four markers failed to consistently amplify products. Heterozygosity in these 10 accessions ranged from 33% to 87%. Variation among accessions was the largest proportion of variance for three markers, variation among genotypes within accessions was the largest proportion for three markers, and for the other nine markers variation within genotypes (chromosome to chromosome) was the largest proportion. Genetic similarity averaged 29.5% across markers. Where accessions have already been screened and found to possess the trait of interest, multiple genotypes from those accessions should be evaluated to identify genotypes with the greatest expression of the trait.  相似文献

Causing potato brown rot, Ralstonia solanacearum (R. solanacearum) strains are reported as one of the most destructive bacteria to potato (Solanum tuberosum L.) in China. In this study, 113 strains were isolated from potato, collected in the four major agroecological zones in China. The study showed that 102 strains belonged to the phylotype IIB sequevar 1 (race 3 biovar 2). The 11 remaining strains belonged to the phylotype I, sequevar 13, 17, 18, 16 or 14 M, a new sequevar closely related to sequevar 14. Thirty-four strains were further characterized according to their virulence at low temperature on three wild potato species. IIB-1 strains all belonged to high and moderate virulence, while others belonged to the low virulence group, which had limited pathogenicity.  相似文献

Background

The castor bean tick (Ixodes ricinus) transmits infectious diseases such as Lyme borreliosis, which constitutes an important ecosystem disservice. Despite many local studies, a comprehensive understanding of the key drivers of tick abundance at the continental scale is still lacking. We analyze a large set of environmental factors as potential drivers of I. ricinus abundance. Our multi-scale study was carried out in deciduous forest fragments dispersed within two contrasting rural landscapes of eight regions, along a macroclimatic gradient stretching from southern France to central Sweden and Estonia. We surveyed the abundance of I. ricinus, plant community composition, forest structure and soil properties and compiled data on landscape structure, macroclimate and habitat properties. We used linear mixed models to analyze patterns and derived the relative importance of the significant drivers.

Results

Many drivers had, on their own, either a moderate or small explanatory value for the abundance of I. ricinus, but combined they explained a substantial part of variation. This emphasizes the complex ecology of I. ricinus and the relevance of environmental factors for tick abundance. Macroclimate only explained a small fraction of variation, while properties of macro- and microhabitat, which buffer macroclimate, had a considerable impact on tick abundance. The amount of forest and the composition of the surrounding rural landscape were additionally important drivers of tick abundance. Functional (dispersules) and structural (density of tree and shrub layers) properties of the habitat patch played an important role. Various diversity metrics had only a small relative importance. Ontogenetic tick stages showed pronounced differences in their response. The abundance of nymphs and adults is explained by the preceding stage with a positive relationship, indicating a cumulative effect of drivers.

Conclusions

Our findings suggest that the ecosystem disservices of tick-borne diseases, via the abundance of ticks, strongly depends on habitat properties and thus on how humans manage ecosystems from the scale of the microhabitat to the landscape. This study stresses the need to further evaluate the interaction between climate change and ecosystem management on I. ricinus abundance.
