This study was designed to test the possibility that Streptococcus iniae enters through the gills and causes infection in hybrid striped bass. To determine the dose response, four groups of fish were inoculated with S. iniae via the gills with a dose of 5.0×105, 2.6×106, 5.0×106, or 1.0×108 CFU/fish. One group of fish was inoculated with tryptic soy broth (TSB) via the gills to serve as controls. The cumulative percent mortality was 13%, 27%, 100% and 100% for 5.0×105, 2.6×106, 5.0×106 and 1.0×108 CFU/fish, respectively. We also examined the tissue dissemination of S. iniae at 0.5, 4, 8, 12, 24, 48 and 72 h after experimental gill inoculation. Fish were inoculated with 2.6×106 or 5.0×106 CFU/fish, which caused low and high mortality, respectively. Within 48 h, fish inoculated with the 2.6×106 dose were culture positive on the gill surface, blood of the first and second gill arches, blood of the third and fourth gill arches and the nares. However, for the dose of 5.0×106 CFU/fish, S. iniae was also isolated from the olfactory, optic and cerebellum regions of the brain, eye, head kidney, trunk kidney, spleen and liver at 48 h. For the 2.6×106 dose, S. iniae was not isolated until 48 h post-inoculation, but was isolated at 12 h for the 5.0×106 dose. The results of this study indicate that S. iniae can enter hybrid striped bass through the gills. However, mortality at similar S. iniae doses was lower than we previously observed by inoculation of the nares.  相似文献

The bacterial flora occurring in brackish pond water, sediment, gills and intestine of healthy tilapia cultured in Saudi Arabia were estimated both quantitatively and qualitatively, and the isolates were identified to genus or species level. Total viable count of bacteria ranged from 1.4±1.5×103 to 8.6±2.7×103 cfu ml−1; 1.2±3.1×106 to 7.3±1.1×107 cfu g−1; 8.7±1.9×105 to 2.1±0.9×106 cfu g−1; and 2.8±2.4×107 to 1.0±1.6×108 cfu g−1 in the pond water, sediment, gills and intestine of brackish water tilapia, respectively. In total, 19 bacterial species were identified. The bacteria were predominantly Gram-negative rods (87%). Pond water and sediment bacteria influenced the bacterial composition of gills and intestine of tilapia. In contrast to gill bacteria, more diversification was observed in intestinal bacteria. The predominant (prevalence >10%) bacterial species were Vibrio parahaemolyticus, Vibrio carchariae, Vibrio alginolyticus, Chryseomonas sp., Vibrio vulnificus, and Streptococcus sp. in all the populations with the exception of the sediment population where Streptococcus sp. was replaced by Shewanella putrefaciens. Vibrio spp. (58% of the total isolates) dominated the total bacterial population.  相似文献

Four studies examined shipping factors of packing technique, density, duration, type of water and use of habitat material for shipping juvenile and adult prawns Macrobrachium rosenbergii. Prawns were shipped in double polyethylene lined 38 × 38 × 20 cm deep styrofoam boxes containing oxygenated water. At temperatures of 19–20°C, 17 g prawns could be shipped safely for 42 h at a density of 10–12 prawns per box (12–15 g liter−1 shipping water). Juveniles, mean size about 6 g, could be shipped at a density of 40 per box (18 g liter−1) for 24 h or 20–25 per box (9–11 g liter−1) for 48 h. Use of mesh material to increase surface area in the box did not appear beneficial nor did shipping in brackish water (salinity8‰). Adults packed unrestricted resulted in survival rates substantially higher than those obtained from immobilized prawns wrapped in mesh.

During the shipment, pH and dissolved oxygen concentrations decreased whereas ammonia concentrations increased. The decreased pH levels may have reduced the ammonia toxicity by decreasing the amount of toxic unionized ammonia (NH3) in solution. In general, dissolved oxygen concentrations appeared more closely related to survival rates than did other water quality parameters which were measured.  相似文献

The potential of bacteriophage therapy to control bacterial disease in farmed fish was tested using, as an example, furunculosis of Atlantic salmon, caused by Aeromonas salmonicida subsp. salmonicida.

In vivo testing with Atlantic salmon and rainbow trout (Oncorhynchus mykiss Walbaum) showed no adverse effects, with bacteriophage generally cleared within 96 h of administration by either intraperitoneal (i.p.) injection or oral in-feed.

Juvenile Atlantic salmon were administered a combination of bacteriophage O, R and B (1.9 × 108 pfu fish− 1) by i.p. injection, after they had been challenged with A. salmonicida subsp. salmonicida 78027, also by i.p. injection. The fish that were injected with bacteriophage immediately after challenge died at a significantly slower rate then those that were either not treated with bacteriophage, or treated 24 h post-challenge. However, the end result (100% mortality) was not affected.

In further experiments the effects of oral (1.88 × 105 pfu g− 1 fish− 1 daily for 30 days), bath (1.04 × 105 ml− 1 daily for 30 days) and i.p. (6.25 × 107 pfu fish− 1) phage treatment to control furunculosis in experimentally infected Atlantic salmon were compared with antibiotherapy (treatment with 10 mg kg− 1 bw− 1 day− 1 oxolinic acid for 10 days), using an indirect cohabitation challenge. No protection was offered by any of the bacteriophage treatments, compared to the positive challenge group, although significant protection was offered by the oxolinic acid treatment. Analysis of samples taken from the trials demonstrated that bacteriophage were correctly administered to the fish and, on occasion, were isolated from fish that had succumbed to furunculosis. It was also shown that bacteriophage resistant A. salmonicida subsp. salmonicida isolates could be recovered from mortalities in all the treatment groups.

The results suggest that, although there were no safety problems associated with the approach, furunculosis in Atlantic salmon is not readily controllable by application of bacteriophage.  相似文献

White shrimp Litopenaeus vannamei held in 25‰ seawater at 27 °C or 28 °C were injected with TSB-grown Vibrio alginolyticus at 1 × 104 colony-forming units (cfu) shrimp− 1 or 1 × 105 cfu shrimp− 1, and then cultivated onward at water temperatures varying from 20 to 34 °C. Over 24–144 h, mortality of V. alginolyticus-injected shrimp held at 34 °C or 32 °C was significantly higher than that of shrimp held at lower temperatures. In a separate experiment, shrimp held in 25‰ seawater at 28 °C and then cultured onward at 20 to 32 °C were examined for immune parameters at 24–96 h. THC, phenoloxidase activity, respiratory burst, and SOD activity decreased significantly at 24 h after transfer to 32 °C. Shrimp held in 25‰ seawater at 27 °C and then cultured onward at 20 to 34 °C showed a significant reduction in phagocytic activity and clearance efficiency for V. alginolyticus at 24 h after transfer to 34 °C. It was concluded that transfer of shrimp from 27 or 28 °C to higher temperatures (32 and 34 °C) reduced their immune capability and decreased resistance to V. alginolyticus infection.  相似文献

Preparation of protoplasts and their subsequent applications for both basic and applied research of marine macroalgae remains largely under developed due to lack of development of reliable methods with consistent yields of viable protoplasts. An improved enzyme preparation with a single commercial enzyme, e.g. 2% Cellulase Onozuka R-10 in 1% NaCl solution, was developed to produce protoplasts rapidly from different green algal genera of Ulva, Enteromorpha and Monostroma. The simple dissolution of enzyme powder in 1% NaCl resulted in exclusion of 2% Macerozyme R-10 from the mixture consisting of 2% Cellulase Onozuka R-10 with 3% NaCl earlier reported as superior for the same algae. Optimal conditions for the isolation of maximum yields of viable protoplasts were found to be with 2% Cellulase Onozuka R-10 incubated at 20 °C for 2 h in 1% NaCl solution with 0.8 M mannitol adjusted to pH 6.0. The protoplast yield with optimized enzyme mixture was as high as 102.8 × 106 cells g− 1 f. wt for M. oxyspermum while it was in the range of 74.4–88.6 × 106 cells g− 1 f. wt thallus for seven species of Ulva, and 82.5–95.4 × 106 cells g− 1 f. wt for three species of Enteromorpha. The regeneration rate of protoplasts isolated using this method ranged from 89 to 92% with normal morphogenesis. The seeding of nylon threads with isolated protoplasts of M. oxyspermum was successful and after 3–4 weeks the entire frame with nylon threads became thick green in color with tiny germlings in laboratory culture. Thus, the method described in the present study allow for rapid mass production of viable protoplasts that could be potentially used as a source for seed material for mariculture and for other applied phycological research.  相似文献

Levels of glucose, lactate, pO2, pCO2, HCO3, TCO2, Na+, K+, Cl, protein, and oxyhemocyanin in the hemolymph and its osmolality and pH were measured when tiger shrimp, Penaeus monodon (13.5 ± 1.5 g body weight), were individually injected with saline or dopamine at 10 8, 10 7, or 10 6 mol shrimp 1. Results showed that hemolymph glucose, lactate, pCO2, HCO3, and TCO2 values increased from 2 to 4 h; hemolymph osmolality, Na+, and total protein had increased at 2 h; and hemolymph K+ decreased from 2 to 8 h after the dopamine injection. All physiological parameters returned to the control values 4–16 h after receiving dopamine. The dopamine injection also significantly decreased the oxyhemocyanin/protein ratio of P. monodon which occurred at 2 h, resulting from an elevation of hemolymph protein and a slight decrease of oxyhemocyanin. These results suggest that stress-inducing dopamine caused a transient period of modulation of energy metabolism, osmoregulation, respiration, and the acid–base balance in P. monodon in adapting to this environmental stress.  相似文献

As indicated by market demand and cultivation prospects, the whitefish (Coregonus lavaretus L.) has the potential to become an important alternative to rainbow trout in fish farming in Finland. The fish processing industry has called for a fish species having non-pigmented flesh, and there is a long tradition of whitefish farming for stocking purposes in Finland. However, cultivation in net cages in the brackish water of the coastal area exposes fish to vibriosis (Listonella (Vibrio) anguillarum) and furunculosis (Aeromonas salmonicida salmonicida). Hence, profitable whitefish farming requires the efficient control of both of these diseases. The efficacy of vaccination (Apoject 1800®) was studied in the laboratory using challenge tests and by monitoring specific antibody production (ELISA) and the blood leucocyte pattern. The fish (22 g) were vaccinated at 15°C and the immunity developed during 8 weeks prior to challenge with vibriosis (1.8×105 cfu) and 12 weeks prior to challenge with furunculosis (4.9×102 cfu). The challenge was performed by i.p. injection. Side-effects around the injection site were studied 16 weeks post-vaccination. The immune system of the whitefish responded well and vaccination gave good protection against vibriosis and furunculosis. The RPS was 100% following the Vibrio challenge and 99% following the Aeromonas challenge. The antibody levels continued increasing in the vaccinated group throughout the study period. Lymphocyte and neutrophil counts were elevated in vaccinated compared to non-vaccinated fish at 8 weeks post-vaccination, but not after 16 weeks. Vaccination produced weak adhesions around the injection site in 42% and minor pigmentation in 9% of the 65 fish examined.  相似文献

A towable netpen unit based on commercial marina float components was developed to serve dual purposes of transport and holding of live Pacific herring in inshore waters of British Columbia. The rectangular netpen unit measures approximately 28×9×7 m deep and holds 25–30 tonnes of herring at densities of 8–16 kg m−3. The design was evaluated under field conditions including a comparison with two industry methods of live fish transport. Results over 4 years indicate the design to be seaworthy and flexible in application. As a mode of fish transport, it offers high survival (97% to 8 weeks) and improved cost-efficiency over other methods tested. The towable netpen is considered useful for culture and transport of other fish species such as salmonids and gadids.  相似文献

Filtration rates of hatchery-reared king scallop (Pecten maximus L.) juveniles, fed a single species alga diet (Pavlova lutheri (Droop) Green), were measured at a range of temperatures (6–21 °C). Weight specific filtration rate (ml min−1 g−1 (live weight)) of juveniles of a selected size range of 17–19 mm shell height (0.26–0.36 g live weight) increased with temperature above 16 °C and decreased below 11 °C, but was not significantly different between these two temperatures. Measurements at 16 °C using juveniles with a wider size range of 10–25 mm shell height (0.05–0.8 g live weight) gave the allometric equation: filtration rate (ml min−1)=12.19×weight (g)0.887. Filtration rate decreased significantly when the cell concentration was greater than 200 cells μl−1 (4.25 mg (organic weight) l−1). With six other algae food species, filtration rates similar to those with P. lutheri were only achieved with Chaetoceros calcitrans (Paulsen) Takano. All other algae species tested were cleared from suspension at significantly lower rates. Experiments with diet mixtures of P. lutheri and these other algae suggested that this was usually a reflection of lowered filtration activity, rather than pre-ingestive rejection of cells. In experimental outdoor nursery rearing systems, the filtration rate was inversely proportional to the concentration of cells in the inflow, in the range 5–210 cells μl−1. It was not affected by flow rate (2–130 l h−1, equivalent to 0.12–28.38 l h−1 g−1 (live weight)) with scallop juveniles stocked from 2 to 62 g l−1. The results are discussed in relation to on-growing scallops at field sites.  相似文献

Several studies have shown that food ration can affect the growth of cultured fish. Determining the optimal food ration would help to achieve better growth and also provide direct economic benefits due to reduced food wastage, which would lead to commercial success. Therefore, we studied the effects of ration levels on growth performance of 0+ juvenile yellowtail flounder to determine the optimal food ration. Two experiments were conducted; the first experiment as a preliminary using ration levels of 1%, 2%, 4%, 6% body weight per day (% bw day−1) held at 7.0 °C with a stocking density of 0.95 kg m−2 (45% bottom coverage). Results of this preliminary experiment indicated that fish fed with 1% bw day−1 had significantly lower growth (weight, length, body depth and specific growth rates (SGR)) than those fed with 2%, 4% and 6% ration. However, fish fed with rations of 1% and 2% showed significantly lower gross food conversion ratios (GFCR) than fish fed with 4% and 6% rations. Survival was not significantly affected by different ration levels. Based on these preliminary results, we used ration levels of 1%, 1.5%, 2% and 3% for the main experiment. Fish were held at 10 °C with a stocking density of 1.45 kg m−2 (34% bottom coverage). Results indicated that fish fed with 1%, 1.5% and 2% bw day−1 had significantly lower growth than fish fed with 3% bw day−1. GFCR was significantly different for all four rations. It was lower for 1% than 1.5%, 2% and 3% rations. Survival was not significantly different between any treatments. We discuss our results with emphasis on growth and economics (i.e., feed wastage) and stress the need to balance both components in a commercial operation.  相似文献

The technical features of a laboratory scale water recycling unit for experimental small scale tilapia breeding are described. Two units (1 and 2) were operated during a 6 month period, carrying a similar fish load (7·5 kg) and feeding rate (2% fish body weight/day). Unit 1 received natural illumination, while unit 2 was artificially illuminated (14/10 - light/dark cycle). Both units were equipped with a biological filter bed (substrate surface area, 3500 cm2). In unit 1, total ammonium and nitrite concentrations ranged from 0·05 to 0·5 mg liter−1, while nitrate varied between 10–40 mg liter−1. In unit 2 corresponding values were 0·15-3 mg liter−1, 0·05–0·8 mg liter−1 and 10–40 mg liter−1. Temperatures ranged between 20–29°C and pH values between 7·5–6·9 in both units. Dissolved oxygen concentrations decreased gradually from 5·6 to 3·4 mg liter−1 in unit 1 and from 5·6 to 2·6 mg liter−1 in unit 2. Twenty-six spawnings occurred in unit 1 in March and April, while only eight spawnings occurred in unit 2, possibly because of the absence of sunlight. The aim was to assess whether selection for increased growth rate in Atlantic salmon (Salmo salar) is associated with increased feed intake and/or better feed utilization. Growth responses of offspring from selected and wild lines of Atlantic salmon (initial weight 814±14 g and 533±12 g, respectively) were tested in a 14-week experiment. Selected and wild salmon increased body weight by 79 and 39%, respectively, during the experiment. Relative feed intake (DFI), thermal growth coefficient (TGC) and feed efficiency ratio (FER) were significantly higher in the selected (DFI: 0.67% BW d−1, TGC: 2.96×10−3 and FER: 1.16), than in the wild, line (DFI: 0.48% BW d−1, TGC: 1.39×10−3 and FER: 0.93). FER was positively correlated with mean growth rate (r=0.90, n=6, P<0.05), and differences between the lines indicated a 4.6% increase in FER per generation of selection. Fish of the selected line had a significantly lower intake of protein and energy per kilogram gain, so the higher growth rate of the selected line was the result of both greater feed consumption and more efficient feed utilization for growth. This implies that selection for increased growth rate in Atlantic salmon may improve both of these traits.  相似文献

A continuous, underway fish egg sampler (CUFES) was employed to assess the abundance and distribution of eggs of both sardine, Sardinops sagax , and round herring, Etrumeus whiteheadi , on the Western Agulhas Bank, South Africa, during September 1996. Samples were collected while underway along six inshore/offshore transects, and at stations along the transects. Volumetric estimates of egg density (eggs m−3) from on-station CUFES samples were highly correlated with both volumetric and areal (eggs m−2) estimates of egg density from samples collected from CalVET net hauls at these stations, demonstrating the validity of this novel sampling technique. Sardine and round herring eggs were encountered in a band running parallel to the coast and extending from 10 to 30 nautical miles offshore to the shelf edge, and highest egg densities were associated with strong north-west-flowing currents in the region of the shelf edge. Collecting samples while underway increased the precision of the estimate of mean egg density for sardine eggs but not for round herring eggs. The use of CUFES in obtaining a fine-scale resolution of sardine egg distribution, and as a tool for stock assessment, are discussed.  相似文献

Gastrointestinal lipid absorption in Atlantic salmon was studied following force-feeding with a standard grower diet to which isotope-labelled free fatty acid (FFA) [1-14C]-decanoic acid (14C-10:0) and triacylglycerol (TAG) [9,10-3H(N)]-triolein (trioleic acid; 3H-18:1) were added. Following force-feeding, the 15 fish, with an average weight of 704 g, were kept in five circular tanks, three fish per tank, supplied with seawater (salinity, 32 g l−1). At 3, 6, 12, 18 and 48 h, the fish from one tank were killed, and the gastrointestinal (GI) tract was removed and divided into six segments: stomach (ST), anterior, mid, and distal pyloric ceca (PCA, PCM, and PCD, respectively), mid intestine (MI), and distal intestine (DI). The levels of radioactivity (dpm [disintegrations per minute] μg−1 tissue) from the 14C-10:0 and the 3H-18:1 substrates were measured in tissue samples from each segment, allowing an estimation of the amounts of fatty acid substrates absorbed from the lumen into the intestinal cells. The highest levels of radioactivity from both fatty acid substrates were measured in tissue from pyloric ceca (PC) 12 to 18 h after force-feeding. The level of radioactivity from the 14C-10:0 substrate was significantly higher in the PCA than in the PCM and PCD, while there was no clear difference in the level of radioactivity from the 3H-18:1 substrate measured among the different PC tissue segments and the level measured in MI. The level of radioactivity in tissue from DI was negligible to low for both fatty acid substrates. Total lipid accumulation in tissue from PCM, MI and DI was quantified morphometrically at each sampling point using light microscopy and image analysis. These results confirmed the findings from the isotope study, where the highest levels of fat were present at 12 and 18 h following force-feeding. In conclusion, PC was the most important site for fatty acid absorption in the GI tract of Atlantic salmon.  相似文献

This study was carried out for three successive years during 1999–2001 to evaluate growth and yield performance of fish, prawn and paddy under rice–fish integration system in rainfed medium land ecosystem. Irrespective of stocking density, faster growth rate was recorded for Catla catla followed by Cyprinus carpio, Cirrhinus mrigala, Labeo rohita and Macrobrachium rosenbergii. C. carpio and C. mrigala performed better growth rate against that of L. rohita probably due to the fact that being bottom dwellers, C. carpio and C. mrigala are more tolerant to fluctuation of oxygen concentration. Productivity of fish and prawn was, however, higher (p<0.05) in refuges with 10-cm weir height plots, irrespective of stocking density, while overall yield performance was good at stocking density of 25,000 ha−1. It was observed that, even with supplemental feeding, with increase in stocking density, biomass yield increased up to an optimum and then decreased. An average minimum and maximum yield of 906.6–1282.3 kg ha−1 of fish and prawn has been achieved, which was much higher than the earlier recorded productivity in a season under rice–fish integration system. Highest grain yield was recorded at 15-cm weir height plot (3629 kg ha−1), probably contributed by higher number of panicles per square meter (235.5) and number of filled grains per panicle (121.7). Percentage increase in rice yield under rice–fish integration system was 7.9–8.6% against control, where paddy was cultivated without integration of fish and prawn probably due to better aeration of water, greater tillering effect and additional supply of fertilizer in form of leftover feed and fish excreta. Irrespective of stocking density, the overall rice equivalent yield (REY) of the system was high (4.22–4.55 tons ha−1) at 12.5-cm weir height plots-cum-refuge, without using any pesticide, herbicide, etc.  相似文献

This present study was designed to investigate the effects of stocking density and water exchange on the growth rate, survival and performance index of L. setiferus postlarvae under controlled laboratory conditions. The experiment was done with postlarvae (PL10 to PL40) at densities of 50, 150, 250 and 350 shrimp/m2 and various different water exchanges rate per day (0, 6, 12 and 18%). The maximum growth rate was obtained for shrimp with 12% water exchange per day at all densities. A reduction of the maximum growth rate was observed in relation to density with the highest values in shrimp stocked in a density of 50 and 150 shrimp/m2 (mean value of 0.53 mg/d) and the lowest in shrimp stocked in a density of 350 shrimp/m2 (0.24 mg/d). The multiple regression equation obtained to relate performance index (growth rate* survival : PI), shrimp density (X1) and water exchange (X2) was: PI = 0.31 + (0.001) X1+ 0.039 X2+ 2.28 × 10−6 X12+ (−0.0017) X22+ (0.000026)X1X2, R 2= 0.78; P > 0.03. According to this equation the optimum shrimp density-water exchange comhination was between 5 to 12% of water exchange at stocking density of between 50 and 150 shrimp/m2. Salinity, ammonia-N and nitrite-N increased according to the time spent in tanks without water exchange. With no (0%) water exchange, water quality parameters measured were outside the optimum for L. setiferus postlarvae. The use of optimum density and water exchange in a nursery system for L. setiferus with optimum variables established is proposed.  相似文献

Assessment of the fish stock in the Theodorushaven was conducted with a dual-beam hydroacoustic system. Species composition determined by a seine and trawl fishery showed large concentrations of fish in the harbour during winter. The hydroacoustic survey showed that the distribution of fish in the harbour area was stratified with fish near the bottom. The overall density in the harbour area was 0.67 fish m–3. The density in the associated channel area was estimated at 0.1 fish m–3. The coefficient of variation for the estimated total fish abundance was >25%.Fish >25 cm were not caught in the Theodorushaven fishery, although analysis of the sonar data indicated their presence. Fishery data from the adjacent Volkerak-Zoommeer showed that bream was the dominant species. Therefore, the biomass estimate of the stock >25 cm in the Theodorushaven was based on the standard length–weight relationship of bream. This resulted in maximum biomass estimates of 5800 kg ha–1 for the harbour area and 1100 kg ha–1 in the channel area. The minimum biomass estimate, based on the assumption that the more slender zander was the dominant species >25 cm, gave values of 4100 kg ha–1 in the harbour area and 780 kg ha–1 in the channel area.  相似文献

The optimum feeding rate of the rotifer Bruchionus plicutilis was investigated to determine the best conditions for growth of the rotifer, and also in order to maintain good water quality of the culture. Fifty rotifers of a large size strain were cultured individually at five food density levels of Nannoehloropsis sp. (0.5 ± 106, 1.5 ± 106, 3 ± 106, 5 ± 106, and 8 ± 106 cells/ml). At each level, daily survival and offspring production were recorded until the death of the final individual. The data obtained were analyzed by the life table method. The maximum value of three growth indices (the net reproduction rate, mean life expectancy at age 0, and intrinsic rate of increase) was obtdned at the food density of 1.5 ± 106 cells/ml. The ration size was calculated to be 325% (dry weight)/ day, which is equivalent to about 70% of the saturated feeding rate. It was suggested that the feeding rate should be controlled to lower than the saturated value for an efficient mass culture.  相似文献

Growth performance, muscle cellularity and flesh quality were investigated in Atlantic salmon (Salmo salar L.) fed either of two diet ranges [high protein (HP), or low protein (LP)], which differed in digestible protein/digestible energy ratios but were of equivalent digestible energy content (21.4 MJ kg−1 wet weight). Smolts from an early maturing (Lochy) and a late maturing (Mowi) strain were PIT-tagged and reared together in duplicate 5×5×5-m sea cages for each diet. The Lochy and Mowi fish were harvested in May and August, respectively, after 417 and 515 days in seawater. The average body weight of fish in each cage at harvest was in the range 3.8–5.4 kg, with no significant difference between diets. The total cross-sectional area of white muscle and the number and diameter of muscle fibres was determined at the level of the first dorsal fin ray. The distribution of muscle fibre diameters was investigated using nonparametric smoothing and bootstrapping techniques. Diet had no effect on fibre size distribution or fibre number in the Mowi strain, and small but significant effects for the Lochy strain. At harvest, in Lochy salmon of average fork length 69 cm there were around 15% more fibres in fish fed the HP than LP ratio diets. However, the 50th percentile of fibre diameter was 20% greater in fish fed the LP than HP diets, such that the total muscle cross-sectional area was similar. The lipid content (14.1–15.3% wet mass), astaxanthin pigment concentration (7.0–8.5 mg kg−1 wet mass) and colour (RocheSalmoFan™ and Minolta Chromatometer readings) of the flesh were similar for both strains and diets. There was no significant difference in the average muscle fibre density between strains and diet, which varied between 60 and 140 fibres mm−2 muscle cross-sectional area. Gaping during processing of the fillet was in part related to muscle cellularity. Little or no gaping was observed in any fish with a fibre density in excess of 95 fibres mm−2 muscle. It was concluded that individual variation in fibre density is important in the development of gaping, but that muscle cellularity and flesh quality are relatively insensitive to the protein to energy ratio in the diet over the range studied.  相似文献
