The present study examined localization of cholecystokinin receptor (CCK-R) mRNA in the muscle layer of the ovine omasum and role of CCK-R type 1 (CCK-1R) in the regulation of muscle contraction of the omasum. We demonstrated that not only CCK-R type 2 (CCK-2R) mRNA but also CCK-1R mRNA is highly expressed in the muscle layer of the ovine omasum. Application of CCK-8 to muscle strips of the greater curvature of the ovine omasum at 1-100 nM induced tonic contraction in a concentration-dependent manner, and the contractile effect of CCK-8 was inhibited by both CCK-1R antagonist lorglumide (IC(50) 2.7 and 7.9 microM in the longitudinal and circular muscle, respectively) and CCK-2R antagonist PD135,158 (IC(50) 51.4 microM in the longitudinal muscle), indicating that not only CCK-2R but also CCK-1R is functionally expressed in the plasma membrane of smooth muscles in the omasum and mediates action of exogenous CCK. Contractile effect of intravenous infusion of CCK-8 (1-30 pmol/kg/min) on omasal contraction was also confirmed in the in vivo experiments using conscious sheep in the absence and presence of atropine infusion (14.4 nmol/kg/min), and showed that circulating CCK increases omasal electromyographic (EMG) activity at lower plasma concentration than that it inhibits ruminal contractions. Taking account of our previous results in the in vivo study using other CCK-1R antagonist, it is suggested that circulating CCK, even at normal range of plasma concentration, plays a physiological role as a regulator of omasal contractions in sheep and CCK-1R mediates the action of CCK.  相似文献

The present study was planned to evaluate a role of nitric oxide (NO) in the regulation of regular ruminal contractions in conscious sheep. Intravenous infusion of S-nitroso-acetyl-DL-penicillamine (SNAP) at doses of 3-30 nmol kg(-1) min(-1)for 30 minutes inhibited both the amplitude and frequency of ruminal contractions in a dose-dependent manner. However, intravenous infusion of Nomega-nitro-L-arginine-methyl ester (L-NAME) at doses of 0.3-3.0 micromol kg(-1) min(-1)did not alter the basal tone of intraruminal pressure and the amplitude of ruminal contractions. The frequency of contractions was slightly inhibited by L-NAME infusion at 1.0 micromol kg(-1)min(-1). The effects of L-NAME were abolished by simultaneous infusion of L -arginine at 30 micromol kg(-1) min(-1). These results suggest that exogenous NO can diminish the ruminal contractions, while endogenous NO is not involved in the regulatory mechanism of basal tone and regular phasic contractions of the rumen in healthy sheep.  相似文献

The omasums of 30 healthy cows and 55 cows with various gastrointestinal disorders (10 with left displacement and eight with right displacement of the abomasum, 10 with abomasal volvulus, 10 with traumatic reticuloperitonitis, nine with ileus of the small intestines and eight with reticulo-omasal stenosis) were examined ultrasonographically on the right side of the body with a 3.5 MHz linear transducer. The dorsal and ventral margins of the omasum and its size in the fifth to 11th intercostal spaces were determined. Generally, the ultrasonographic appearance of the omasum did not differ between the healthy and abnormal cows. The omasum appeared as a semicircle, and the omasal wall closest to the transducer was visible as a thick echogenic line. In a few of the abnormal cows, the omasal laminae were visible and the omasum appeared to have motility. In the cows with left and right displacement of the abomasum and abomasal volvulus, the dorsal margin of the omasum was significantly further from the dorsal midline in some intercostal spaces than in the healthy cows. In the cows with left displacement of the abomasum, the ventral margin of the omasum was significantly further from the dorsal midline in the 7th intercostal space than in the healthy cows. In the cows with reticulo-omasal stenosis, traumatic reticuloperitonitis and ileus of the small intestine, the ventral margin of the omasum was significantly closer to the dorsal midline in some intercostal spaces than in the healthy cows. The mean (sd) size of the omasum in the healthy cows varied from 16.3 (1.5) cm to 56.9 (10.0) cm, depending on the intercostal space, and the omasum was significantly smaller in some intercostal spaces in the cows with reticulo-omasal stenosis, right displacement of the omasum, abomasal volvulus and ileus of the small intestine.  相似文献

In five conscious adult ewes at rest, chronically implanted with electrodes in the musculature of the omasal wall, intravenous (i.v.) infusion for 30 min of alpha 1- or alpha 2-adrenergic receptor blockers, prazosin (20 micrograms/kg/min) and yohimbine (30 micrograms/kg/min), respectively, had no significant effects on omasal myoelectrical activity. The i.v. administration for 15 min of alpha 1- or alpha 2-agonists phenylephrine (4 micrograms/kg/min) or naphazoline (2.5 micrograms/kg/min), respectively, increased the frequency and the amplitude of groups of myoelectrical discharges of omasum, as well as the duration of its activity. Pretreatment of animals with prazosin blocked the responses to phenylephrine. Yohimbine prevented the effects of naphazoline dose-dependently. It is suggested that both alpha 1- and alpha 2-adrenoreceptors are involved in regulation of the sheep's omasal notility. This regulation did not seem to be a simple consequence of the changes in the reticular motility.  相似文献

The mucosal relief of the omasum in cattle, sheep and goats was examined by the naked eye, stereoscopic microscope and scanning electron microscope. The mucosal relief of the omasum showed wide variations both between and within these species. Omasal papillae on the laminae varied highly in shape, i. e. conical, rounded, claw-like, wart-like, finger-like etc. They decreased gradually in size from the oral to the aboral region of the omasal lamina. Ridge-like structures and linear arrangements of the papillae were also seen on the omasal laminae of cattle and sheep. Many grooves and folds were found on the lateral surface of the omasal papillae in adult cattle and on the inter-papillar surface of the laminae of all three species. The mucosal relief of the omasum in cattle seemed to be more well-developed than those of sheep and goats. Dietary effects on the omasal mucosa were also discussed.  相似文献

Vagally-mediated regulation of motility in the abomasal body by duodenal and abomasal antral motility was demonstrated in acutely prepared anaesthetized sheep. The enteric plexuses between the abomasal body, antrum and duodenum were interrupted by transection. Antral contractions were more effective than duodenal contractions at causing inhibition of the abomasal body, and antral isometric conditions were more effective than antral isotonic conditions. Inhibition of motility in the abomasal body was reduced by unilateral cervical vagotomy, was abolished by bilateral cervical vagotomy, and was reversibly inhibited by cervical vagus cold block. The objectives of this study were to evaluate the role of nitric oxide (NO) synthase isoforms (nNOS, eNOS, and iNOS) in the regulation of the migrating motor complex (MMC) in sheep using electromyography and their expression in the gastrointestinal (GI) tract by Western blot (WB) and immunohistochemistry. Intravenous administration of L-NAME or the nNOS inhibitor 7-nitroindazole (7-NI) decreased the MMC interval. Myoelectric activity of intestinal phase II was increased, whereas antral activity was reduced. These effects were blocked by L-arginine. Inhibitors of either iNOS (aminoguanidine and S-methylisothiourea) or eNOS (L-NIO) were ineffective. The NO donor sodium nitroprusside decreased GI myoelectric activity, inhibited the MMC pattern, and prevented the effects induced by L-NAME and 7-NI in the intestine. Intracerebroventricular administration of these agents did not modify GI motility. In the rumen, abomasal antrum, duodenum, and jejunum, WB showed three bands at about 155, 145, and 135kDa corresponding to nNOS, and a 140-kDa band (eNOS); however iNOS was not detected. Positive nNOS immunostaining was observed in neurons of the myenteric and submucous plexus of all GI tissues, while eNOS was found in the endothelial cells, ruminal and intestinal epithelium, as well as in some enteric neurons and in endocrine-like cells of the duodenal Brunner's glands. In contrast, only weak iNOS immunoreactivity was found in ruminal epithelium. Taken together, our results suggest that NO, synthesized at a peripheral level by nNOS, is tonically inhibiting the MMC pattern and intestinal motility in sheep.  相似文献

The study was conducted to establish the ultrasonographic features of the healthy and impacted omasum in cows and buffaloes. Scanning was done using a 3.5 MHz microconvex transducer. In healthy buffaloes, the omasum could be scanned at the eighth to ninth intercostal space as a round or oval structure having thick echogenic wall with echogenic leaves. Gradual slow movements of omasal leaves could also be seen in real-time B-mode. The omasum appeared to be very clear, large, and close to the transducer at the start of the omasal contraction, and as the contraction progressed the omasum retracted away from the transducer and became very small. In healthy cows the omasum was seen as a crescent-shaped structure with an echogenic wall. The contents of the omasum or omasal leaves could not be visualized. Omasal contractility was not as prominent as in buffaloes. In buffaloes, the impacted omasum appeared amotile, the omasal leaves were not visible, and the omasum as a whole gave a prominent distal acoustic shadow. In cows, the impaction could be diagnosed based on amotile omasum covering a large area on the right side. Ultrasonography was found to be helpful in subjective assessment of omasal impaction but could not aid in diagnosing the severity of impaction.  相似文献

Three ruminating calves were each provided with a rumen cannula (from which reticulum samples could be obtained), an abomasal cannula and a sleeve sutured at the omasal-abomasal orifice that permitted digesta flowing from the omasum to be diverted and collected. The calves were each given in turn a high roughage (HR) and high concentrate (HC) diet consisting mainly of dried grass and flaked maize in the proportions by weight of 3:2 and 1:2 respectively. A water soluble non-absorbable marker (polyethylene glycol) and a particulate marker (103Ru-phenanthroline complex) were also given and the diets were sometimes supplemented with MgSO4. For each diet (HR(O), HR(Mg), HC(O), HC(Mg)) samples were taken from the reticulum and omasal outflow. Magnesium, phosphorus, PEG and 103Ru were measured in the samples as taken and liquid-rich and solid-rich fractions prepared from them. From their distributions relative to the two markers, Mg and P flows into and out of the omasum were estimated. Results were too few to show significant differences in absorption efficiency between diets but overall approximately 10-40% of the Mg entering the omasum was absorbed as a net effect implying that this organ may be the major site of Mg absorption in young cattle. For P also substantial amounts entering the omasum (approximately 10-40%) were absorbed in that organ.  相似文献

The effects of intravenous infusion of proglumide on regular ruminal contractions were examined in conscious sheep using doses that inhibit pancreatic exocrine secretion. After a control period of 20 min, proglumide was infused intravenously for 40 min at a dose of 15, 30 or 60 μmol/kg per min and venous blood was collected. The intravenous infusion of proglumide significantly increased the frequency of ruminal contractions at 15 μmol/kg per min without altering the amplitude, while it significantly decreased the frequency and amplitude of ruminal contractions at 30 and 60 μmol/kg per min in a dose-dependent manner. Proglumide did not increase contractile activity of the omasum, abomasum and duodenum or the plasma concentration of immunoreactive cholecystokinin (CCK). Application of proglumide at 1–30 mmol/L inhibited bethanechol-induced contraction in both longitudinal and circular muscle strips of the dorsal sac of the rumen. These results suggest that proglumide at a low dose acts indirectly on the rumen as a CCK receptor antagonist to increase the frequency of contractions, whereas at higher doses it inhibits cholinergic-induced contraction of the ruminal muscles or acts as an agonist to inhibit contractions in sheep. Hence, proglumide at high doses seems unsuitable for research or therapeutic use as a CCK receptor blockade in sheep.  相似文献

The relationship between plasma calcium level and rumen motility in cows and sheep and abomasal motility in cows was investigated by inducing hypocalcaemia in seven cows and five sheep by the infusion of Na2 EDTA over a period of approximately two hours. Rates and amplitudes of rumen and abomasal contractions were markedly reduced by the reduction of plasma calcium level to approximately 50% of normal. There were significant positive linear relationships (P less than 0.05) between rate and amplitude of rumen contractions in both sheep and cows over a plasma calcium range of 1-3 mmol/L. There was also a significant linear relationship (P less than 0.05) between plasma calcium and abomasal rate of contraction over the same range in cows, but the relationship with amplitude of abomasal contraction was not quite significant (P less than 0.1 greater than 0.05).  相似文献

OBJECTIVE: To determine and compare the effects of erythromycin, neostigmine, and metoclopramide on abomasal motility and emptying rate in suckling calves. ANIMALS: 6 male Holstein calves (15 to 40 days of age). PROCEDURE: Calves were monitored for 1 hour before being fed milk replacer (60 mL/kg; time, 0 minutes) and then were monitored for another 3 hours. Calves received 6 treatments in randomized order: erythromycin (8.8 mg/kg, IM) at -30 minutes; low-dose erythromycin (0.88 mg/kg, IM) at -30 minutes; erythromycin (8.8 mg/kg, IM) at -30 minutes and neostigmine (0.02 mg/kg, SC) at -30 and 90 minutes; neostigmine (0.02 mg/kg, SC) at -30 and 90 minutes; metoclopramide (0.1 mg/kg, IM) at-30 and 90 minutes; and placebo (2 mL of saline [0.9% NaCl] solution, SC) at -30 minutes. Abomasal volume was calculated from ultrasonographic measurements of abomasal width, length, and height. Abomasal motility and emptying rate were assessed by measuring luminal pressure and change in abomasal volume over time. RESULTS: Administration of erythromycin (8.8 mg/kg) increased the frequency of abomasal luminal pressure waves and the mean abomasal luminal pressure and decreased the half-time of abomasal emptying by 37%. Administration of metoclopramide, neostigmine, and low-dose erythromycin (0.88 mg/kg) did not alter abomasal motility, mean luminal pressure, or emptying rate. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that administration of erythromycin at the labeled antimicrobial dose (8.8 mg/kg, IM) exerted an immediate, marked prokinetic effect in healthy suckling calves, whereas administration of metoclopramide or neostigmine did not alter abomasal motility or emptying rate.  相似文献

The distribution of nicotinamide adenine dinucleotide phosphate diaphorase (NADPHd)-positive neurones was investigated in the chicken caecum. Double staining combined NADPHd histochemistry with immunohistochemistry for neural nitric oxide synthase (nNOS) indicated that NADPHd-positive neurones also showed immunoreactivity for nNOS. NADPHd-positive nerve cell bodies were observed in both the myenteric and the submucous plexuses. Nerve fibres showing enzyme activity were mainly distributed in the circular muscle layer, but only a few fibres in the mucosal layer. Fine nerve fibres showing NADPHd activity were found running between germinal centres in the caecal tonsil. Quantitative analysis showed no significant differences in the number of enzyme-positive nerve cell bodies per ganglion of the myenteric and the submucous plexuses among three different caecal regions; proximal, middle and distal regions. Larger numbers of ganglia were detected in the submucous plexus than the myenteric plexus at all three regions. These data indicate that nitrergic neurones in the submucous plexus mainly project to the circular muscle layer in the chicken caecum. It is possible that nitrergic nerves regulate the motility of the chicken caecum.  相似文献

Detailed studies on the distribution of the muscle coat at the omasoabomasal junction in cattle, especially in the pila omasi, were carried out in order to clarify the mechanism of closing of the ostium omasoabomasicum. Anatomical and histological observations revealed that the muscle coat forming the circumference of the ostium omasoabomasicum is composed of inner circular and outer longitudinal layers. The former was particularly thickened at the end of the sulcus omasi (pila omasi). Joined to the pila omasi was a thick muscle bundle which extended from the labium sinistrum (left lip) of the sulcus reticuli and ran obliquely along the floor of the sulcus omasi. Moreover, on the abomasal side of the ostium omasoabomasicum, vela abomasi were formed in such a manner as to surround the ostium omasoabomasicum. These were continuous with the edges of the sulcus omasi. Judging from its location and muscular structure, the pila omasi may contract in accordance with the contraction of the reticulum. As a result, the ostium omasoabomasicum may be narrowed and the vela abomasi pulled toward the omasum, perhaps obstructing the ostium omasoabomasicum. Accordingly, it is presumed that the retention of contents in the omasum may effectively prevent abomasal contents from moving backward into the omasum.  相似文献

The hindlimb arteriovenous difference (AVD) model was used to determine whether 30 mg/ kg of the nitric oxide synthase (NOS) inhibitor L-NGnitroarginine methyl ester (hydrochloride; L-NAME) inhibited ovine NO synthesis and influenced muscle metabolism. Eight Border Leicester x Merino cross lambs (50 to 55 kg BW) were infused with saline (control) or saline containing L-NAME via an indwelling jugular vein catheter in a balanced randomized crossover design with 3 d between treatments. The abdominal aorta and deep femoral vein were catheterized for assessment of AVD of hind limb metabolism. Arterial hematocrit and insulin concentration and both arterial and venous concentrations of nitrate/nitrite (NOx), glucose, lactate, NEFA, and urea were determined. Infusion of L-NAME decreased arterial NOx concentrations (P = 0.049), indicating inhibition of systemic NO synthesis. Treatment had no effect on arterial (3.5 vs. 3.6 +/- 0.19 mmol/L for control and L-NAME lambs, respectively; P = 0.39) or venous (3.3 vs. 3.4 +/- 0.16 mmol/L, P = 0.55) plasma glucose concentrations or on glucose AVD (0.19 vs. 0.27 +/- 0.065 mmol/L, P = 0.20). There was an interaction (P = 0.038) between time and treatment, such that L-NAME initially increased the AVD of glucose (up to 180 m) divergent from control lambs. The response was then decreased before a possible inflection beyond 240 min. Infusion of L-NAME increased hindlimb venous NEFA (222 vs. 272 +/- 13.2 micromol/L, P = 0.007) and NEFA AVD (79.4 vs. -13.3 +/- 31.5 micromol/L, P = 0.018). These metabolic changes were independent of plasma insulin concentrations, which were not affected by L-NAME infusion (25.3 vs. 27.8 +/- 3.62 mU/L, P = 0.85). The increase in hindlimb lipolysis after L-NAME infusion does not seem to be due to increased lipolysis of plasma triacylglycerol because circulating arterial (155 vs. 142 +/- 20.8 micromol/L, P = 0.58), venous (154 vs. 140 +/- 20.5 micromol/L, P = 0.50), and AVD (1.0 vs. 2.9 +/- 3.17 micromol/L, P = 0.38) triacylglycerol concentrations were unaffected by L-NAME infusion. In conclusion, these data indicate that infusion of 30 mg of L-NAME/kg inhibits NO synthesis, which in turn influences fat and carbohydrate metabolism in the ovine hindlimb independently of plasma insulin concentrations.  相似文献

The objective of the current study was to evaluate the effect of unrepresentative sampling of digesta particulate matter entering the omasal canal on the accuracy of fiber flow measurements. The experimental design comprised one period, one diet, and three cows as experimental units. Within each cow, the physical and chemical composition of digesta particulate matter was assessed at seven sites within the digestive tract. Three Finnish Ayshire dairy cows, equipped with ruminal and simple-T duodenal cannulas, in extended lactation were offered grass silage twice daily on an ad libitum basis. Digesta samples were collected from the rumen (dorsal and ventral sac), reticulum, omasal canal, omasum, duodenum, and rectum to determine particle size distribution in digesta, chemical composition of various particle size fractions, and distribution of two flow markers (Cr-labeled straw and indigestible NDF [INDF]) among particle size fractions. Digesta samples were wet-sieved using sieves of 2.50, 1.25, 0.630, 0.315, 0.160, and 0.080 mm. Particulate matter was analyzed for OM, NDF, and Cr concentrations, and INDF concentration was determined based on 12-d ruminal incubation. The particle size of digesta entering the omasal canal was larger compared with the omasum or the duodenum, suggesting that omasal canal samples were not representative of particle size distribution truly escaping the rumen. The concentration of potentially digestible NDF (PDNDF) decreased with decreasing particle size. The PDNDF concentration of particulate matter retained on all sieves was greatest in the rumen and gradually decreased along the digestive tract. From the reticulorumen to the omasum, the decrease was associated with decreased particle size, reflecting selective passage of particulate matter. In contrast, from the omasum to the duodenum and rectum, the PDNDF concentration decreased within each particle size fraction without effect on particle size, indicating a nonselective passage of particulate matter between these sites. Variation between particle size fractions was slightly greater for Cr concentration than for INDF concentration, indicating that unrepresentative sampling of particulate matter had a greater effect on Cr concentration compared with that of INDF. Owing to unrepresentative sampling, NDF entering the omasal canal was overestimated by 5% using INDF and underestimated by 7% using Cr as a particle phase marker. Of total NDF digestibility, proportionally 0.90, 0.07, and 0.03 occurred in the reticulorumen, omasum, and intestines, respectively. The current results indicate that, despite unrepresentative sampling of digesta particulate matter entering the omasal canal, the errors in determined NDF flow were small. The omasum may have a greater role in postruminal NDF digestion than the intestines.  相似文献

Alimet (Novus Inter., Inc., St. Louis, MO) feed supplement (an 88% aqueous solution of 2-hydroxy-4-(methylthio) butanoic acid; HMB) is a source of L-Met commonly used in nonruminants and ruminants. The absorption of HMB across ovine omasal and ruminal epithelia was evaluated in this study. Ruminal and omasal epithelia were collected from eight lambs (BW = 67.6 kg +/- 9.1) and mounted in parabiotic chambers that were repeatedly sampled throughout a 60-min incubation. The appearance of HMB (using DL-[5-14C]-HMB as a radiolabeled marker) in serosal buffers increased quadratically (P < .004) with time in both tissues. More (P < .001) HMB appeared in the serosal buffers with omasal than with ruminal epithelia. Both tissues responded similarly, and, after 60 min of incubation, the accumulation of HMB within the tissues increased linearly (P < .001) as substrate concentration (.375, .75, 1.5, 3.0, 6.0, and 12.0 mM) increased in mucosal buffers. As the concentration of HMB in the mucosal buffers increased, there was a quadratic (P < .001) increase in the appearance of HMB in the serosal buffer of the omasal epithelium, indicating some saturation of the system. The increase in serosal appearance of HMB was linear (P < .001) with ruminal tissue. The results indicate that there are probably multiple mechanisms involved in the absorption of HMB. Because saturation was observed in the omasum, it is likely that mediated transport accounts for at least a portion of the absorption of HMB in the omasum. Other mechanisms (e.g., diffusion and(or) paracellular absorption) are responsible for the balance of the absorption. Omasal epithelium appears to have a greater capacity for HMB absorption than ruminal epithelium. The enzymes involved in the conversion of HMB to 2-keto-4-(methylthio)butanoic acid were found in ruminal and omasal epithelia, liver and kidney. These results indicate that HMB can be absorbed across ruminal and omasal epithelium and that HMB can be used as a source of L-methionine.  相似文献

An 11-month-old Japanese Black steer with chronic bloat underwent clinical and histological analyses. During the observation period, it showed normal appetite and fecal volume but persistent chronic bloat symptoms. Compared to controls, the steer's feces contained undigested large straws. Necropsy revealed normal rumen, reticulum, and abomasum but a small omasum. The rumen, reticulum, and abomasum mucosa was normal, with well-developed ruminal papillae. However, severe hypoplasia of the omasal laminae was observed along with hypoplasia reticular groove and ruminoreticular fold. The contents of the reticulum, omasum, and abomasums comprised undigested large sized hay particles. The omasum papillae showed no pathological abnormalities. This is a rare case of a steer with chronic bloat probably caused by severe hypoplasia of the omasal laminae.  相似文献
