为加快窄叶台湾榕种苗的人工培育,以扦插基质、插穗直径、生根剂浓度及浸泡时间为试验因素,每个因素设置3个水平,采用正交设计的试验方法对窄叶台湾榕进行扦插试验,并对试验结果进行验证。结果表明,各试验因素对窄叶台湾榕扦插成活率的影响顺序为:插穗直径大小吲哚乙酸浓度插穗浸泡时间扦插基质;以100%黄心土为扦插基质,选用直径为0.4~0.8 cm、长度为10~12 cm的穗条在吲哚乙酸400 mg·L~(-1)溶液浸泡30 min后扦插,窄叶台湾榕扦插成活率最高,为68.89%;而使用ABT代替吲哚乙酸作为生根剂,可以使窄叶台湾榕扦插成活率进一步提升至81.11%。 相似文献

为了提高黄刺玫成活率、生根率、缩短生根时间和育苗周期,以野外黄刺玫为材料,研究不同浓度的吲哚-3-丁酸、吲哚乙酸和a-奈乙酸对硬枝扦插黄刺玫枝条生根的影响.结果表明:不同浓度生长剂对重瓣黄刺玫硬枝插穗的生根影响差异显著.其中用100 mg/L吲哚丁酸浸泡3h生根率最高,平均生根率达72.33%;1 000 mg/L生长剂速蘸10 s平均生根率最低,为18.33%.不同生长剂对黄刺玫生根率影响差异并不显著. 相似文献

通过使用高锰酸钾、醋酸、双氧水等3种氧化剂和吲哚乙酸、吲哚丁酸、萘乙酸等3种外源激素不同浓度对新疆杨插穗进行处理,以探讨新疆杨扦插生根促进技术。结果表明:虽然不同氧化剂试验处理对新疆杨生根率与对照相比差异是显著的,但没有达到生产上对生根率的要求;不同的激素浓度水平会明显影响新疆杨的生根率,以吲哚丁酸浸泡插穗6h最佳浓度是200mg·L-1,扦插生根效果较好,生根率达到3.65%。 相似文献

Somatic embryogenesis (SE) in Eucalyptus spp. has been limited to germinated seeds, flowers, lignotubers or zygotic embryos. The low yield of somatic embryos from leaf explants has hampered progress, even though leaves offer a more viable source of clonal explants from superior selected genotypes. It was hypothesised that SE from leaf explants could be enhanced through pairing of synergistic exogenous plant growth regulators, such as natural auxins with natural cytokinins. Callus and embryo induction using 2,4-dichlorophenoxyacetic acid (2,4-D), α-naphthalene acetic acid (NAA), indole acetic acid (IAA), and indole butyric acid (IBA), each at either 1.0 or 3.0?mg L?1, indicated that IAA and IBA favoured significantly higher numbers of embryos compared with 2,4-D or NAA. Hence, IAA and IBA were used for subsequent experiments, combining them (at 1.0?mg L?1) with either the synthetic cytokinin, kinetin, or the natural cytokinin, trans-zeatin, both at 0.1?mg L?1. The combination of trans-zeatin and either IAA or IBA resulted in a significant increase in SE (e.g. 86 ± 17.2% and 23 ± 3.2% for IAA with trans-zeatin and kinetin, respectively), compared with kinetin, or with these auxins alone. Embryo maturation and plantlet regeneration was highest in those calli that were induced with IAA and trans-zeatin, indicating that maturation was dependent on auxin depletion, based on the stability of the analogue used for induction. For the E. grandis clone under study, the use of synergistic plant growth regulators significantly enhanced SE from leaf explants, thus presenting the opportunity to benefit from the advantages that SE offers over conventional organogenesis. 相似文献

以MS为基本培养基,添加不同浓度的2,4-D、NAA、KT或其组合,研究库拉索芦荟愈伤组织的诱导及生长情况。结果表明,两种生长素均可诱导出愈伤组织,但较高浓度的2,4-D易导致褐化,而高浓度的NAA诱导出的愈伤组织含水量较高,以两种生长素配合使用再辅以低浓度的KT效果较好,其中15mg/L的KT 4mg/L的2,4-D 0.1mg/L的KT的组合,愈伤组织诱导率达到90%,且愈伤组织生长较旺盛。 相似文献

以白林2号杨优树萌发枝条(已木质化)为外植体进行组织培养快繁技术研究,结果表明:初代分化最适培养基为MS+6-BA0.3 mg·L-1+IAA0.5 mg·L-1,分化仅为7 d,并有丛生芽发生;使用MS+6-BA0.3 mg·L-1+NAA0.01 mg·L-1+IAA0.3 mg·L-1进行继代培养效果最好,丛生芽粗壮,数量较多;以附加不同含量NAA和IAA诱导生根,可获得再生植株,其中1/2MS+NAA0.01 mg·L-1+IAA0.2 mg·L-1培养基生根率达到88%。 相似文献

尾叶桉MLA无性系 (简称MLA)是扦插难生根无性系 ,尾叶桉U6 无性系 (简称U6 )和刚果 12号桉W5无性系 (简称W5)是相对易生根无性系。分别用吲哚乙酸 (IAA)、吲哚丁酸 (IBA)、萘乙酸 (NAA)处理MLA、U6 和W5插条后 ,各无性系插条体内的过氧化物酶 (POD)、多酚氧化酶 (PPO)、吲哚乙酸氧化酶 (IAAO)活性呈现规律性的升高。在生根过程的不同时期 ,各酶的活性差异极显著 ,表明桉树插条的不定根发生和发展与POD、PPO、IAAO活性有密切联系。而IAA、IBA、NAA对不同无性系和不同的氧化酶活性的促进效果也有显著差异。讨论了生长素促进桉树插条氧化酶活性和不定根发生与发展的机理 相似文献

以楸树无菌苗茎段为材料,研究了不同生长素(NAA、IAA、IBA、2,4-D)在不同浓度下(0.05、0.1、0.2mg/L)对不定芽诱导、增殖和生根的影响。结果表明:在添加IAA激素的培养基中,不定芽的诱导生长最好。IAA0.1mg/L和6-BA0.8mg/L纽合不定芽诱导率高达100%,而且生长健壮;在NAA0.1mg/L和6-BA2.0mg/L的培养基中,IAA0.1mg/L和6-BA0.8mg/L组合诱导出的不定芽增殖倍数最高,为3.23,且不定芽健壮;添加I从0.Img/L对不定芽的生根也有利。 相似文献

Incubation with acetone and dichloromethane had little effect on the germination of different lots of Norway spruce seeds. This was not the case with Scots pine where almost half the seed lots tested were adversely affected by acetone and dichloromethane. The auxins indole‐3‐acetic acid (IAA) and indole‐3‐butyric acid (IBA) were applied to Scots pine seeds in acetone, dichloromethane and phosphate‐citrate buffer. The uptake of the two compounds was most rapid from acetone. Although the uptake from dichloromethane was slower than from acetone it reached the same level after 6 h. Uptake of the growth regulators from buffer was significantly lower than from the organic solvents. The accumulation of IAA and IBA into the seeds from both acetone and dichloromethane was linear between 0.5 and 10 mM, and between 0.5 and 15 mM from buffer. IAA and IBA levels in infused seeds declined over a 5‐day germination period, the former more rapidly than the latter. The metabolic activity of the IAA degradation system was as effective in seeds after immersion in the organic solvents as seeds immersed in buffer. 相似文献
