Resistance to severe babesiosis in young calves has frequently been ascribed to an unknown serum factor(s) which inhibits growth of Babesia bovis in vitro. Our experiments show that young calf sera are as suitable as adult bovine sera for the in vitro culture of Babesia divergens, indicating that in this species at least inverse age resistance is due to alternative mechanisms. The suitability of commercial foetal calf sera for B. divergens cultures seems highly variable.  相似文献

The effects of continuous oxytetracycline administration on the development of parasitaemia of Babesia divergens during both natural and artificial infections were studied. During natural exposure on grazing heavily infested with Ixodes ricinus, seven out of 42 cattle with no previous exposure to tick-borne diseases were injected every four days with a long acting preparation of oxytetracycline at a dose rate of 20 mg/kg. During the six week grazing period 21 untreated cattle developed a patent parasitaemia of B divergens and all became seropositive by the fluorescent antibody test. In contrast, no parasites were observed in treated cattle and antibody titres remained low. Artificial infections were studied with different dose levels of oxytetracycline and their effects on antibody stimulation noted. First, four groups of cows were infected with 10(8) erythrocytes infected with B divergens, three groups being injected every four days with the long acting oxytetracycline formulation at dose levels of 20, 10 and 5 mg/kg, respectively. The highest level completely inhibited parasite replication and antibody formation; the same was observed in one animal dosed at 10 mg/kg but the remainder, plus those treated at 5 mg/kg, developed both low parasitaemia and high antibody titres. The untreated cows developed severe babesiosis. A further untreated control group was added and three weeks after cessation of oxytetracycline treatment all were infected with 10(9) erythrocytes infected with a homologous isolate of B divergens. The controls, plus those in which the previous infection had been completely inhibited, developed severe clinical babesiosis but the remainder were refractory to parasite development.  相似文献

In splenectomised calves infected with Babesia divergens IFA titres appeared after peak parasitaemia and maximum titres were observed 35 days after infection: the IFA response of adult non-splenectomised cattle was similar. It was concluded that the absence of a spleen did not markedly inhibit the IFA response of cattle to B divergens infection.  相似文献

A total of 61 intact Holstein-Friesian calves were exposed to Babesia bovis (= B argentina) by the injection of infected blood or the application of infected Boophilus microplus larvae. Tick-induced infections were uniformly severe, even when induced by relatively small numbers of infected ticks. In contrast, calves infected with carrier blood experienced mild, subclinical reactions despite detectable parasitaemia. The greater severity of tick-induced reactions appeared to be due to the large number of infective doses injected by each infected tick rather than greater virulence of tick versus blood-origin babesiae. The severity of tick-induced babesiosis was related to the age of the experimental calves, with more severe reactions and high mortality occurring among older animals. The mean daily temperature rise increased with age and was highest in 15 animals dying of babesiosis. No relationship could be found between peripheral blood parasitaemia, packed cell volume reduction and mortality in tick-induced infections. One thousand larvae from infected colonies induced babesiosis in all of 29 animals exposed. Although larval transmission of B bovis is difficult to quantitate, there would seem to be no objection to using a controlled tick-borne challenge in the laboratory for assessment of susceptibility.  相似文献

Forty Friesian one-year-old calves were vaccinated simultaneously with live Babesia bovis and B bigemina vaccines. Three groups of 10 calves each were treated with two, three or four doses of 20 mg kg-1 long-acting oxytetracycline (OTC/LA) at six- to seven-day intervals starting from day 6 after vaccination. Ten animals remained untreated. The treated calves showed considerably fewer days of patency and higher packed cell volumes than the vaccinated untreated calves. All calves developed serum antibodies to both parasites following vaccination. Five months later the 40 vaccinated and 30 new calves were challenged with syringe-transferred virulent parasites of both species. The vaccinated calves showed no parasites or clinical manifestations while calves of the new group exhibited severe clinical babesiosis. These results show that when OTC/LA is administered following anti-babesial vaccination, parasitaemia and red blood cell destruction are significantly reduced without, however, inhibiting the development of immunity.  相似文献

The objective of this study was to evaluate the therapeutic and prophylactic efficacy of imidocarb dipropionate (IMDP) against babesiosis and to determine specific antibodies against Babesia ovis in experimentally infected lambs. Thirty-six 6-month-old splenectomized lambs were used. The lambs were randomly divided into six groups with six animals each, and were intravenously inoculated with 50 mL B. ovis-infected erythrocytes as follows: group I (therapy group) was treated with IMDP (1.2 mg/kg body weight) starting on the day of onset of clinical signs of babesiosis after the inoculation; group II (untreated control animals) was not treated with any therapeutic treatment after the inoculation; groups III, IV, V and VI (prophylaxis groups) were administered IMDP (2.4 mg/kg body weight) 1, 2, 3 and 4 weeks before the inoculation, respectively. The animals were housed in a tick-proof room with water and food ad libitum up to the 30th day post-inoculation (PI). The lambs were monitored from the first day PI by recording the manifestation of clinical disease, rectal temperature, and the degree of parasitaemia. All the lambs became infected with B. ovis, except five animals from group III, which were treated 1 week prior to experimental infection. Other animals showed signs of acute clinical babesiosis. The animals treated with IMDP (group I) were able to clear the parasite from the blood circulation after 48 h post-treatment. The recrudescence of B. ovis was observed in two lambs 7 days after treatment, and they were treated with the second similar dose of the drug. Six lambs (1, 1, 2 and 2 lambs in group III, IV, V and VI, respectively) from the prophylaxis groups died within 7-17 days after showing high parasitaemia and clinical symptoms of the disease. Regardless of the clinical symptoms, 83.30% and 66.66% of the lambs which were administered IMDP 1-2 and 3-4 weeks before, were determined to be protected against the virulent field strain of B. ovis.  相似文献

THE EFFECT OF AGE ON RESISTANCE OF CATTLE TO BABESIA BOVIS   总被引:1,自引:0,他引:1  
SUMMARY Susceptible Hereford cattle of different ages were inoculated with 2 times 108Babesia bovis organisms. Experiment 1 consisted of cows aged 6 to 7 years, steers aged 17 to 18 months and calves aged 5 to 6 months, while experiment 2 consisted of cows aged 6.5 to 7.5 years, steers aged 23 to 24 months and yearlings aged 11 to 12 months. Daily measurements of temperature, parasitsemia and packed cell volume were made in order to determine susceptibility of the different ages. Twenty-four of the 36 animals in experiment 1, which included all 12 cows, required treatment. One cow died as a result of an enlarged ruptured spleen, and 2 steers and 1 calf died with classical babesiosis symptoms. No treatment was given to experiment 11 animals, and 5 of the 12 cows died, but the steers and yearlings underwent relatively mild reactions. Statistical analysis confirmed the high susceptibility to B. bovis of the aged cows in both experiments, and the innate resistance of 5 to 6 month old calves in experiment 1. The reaction of the 18-month-old steers in experiment 1 was significantly greater than that of the calves, but significantly less severe than that of the aged cows. Two-year-old steers and yearlings in experiment 2 underwent similar mild reactions, suggesting that innate immunity may persist for longer periods when compared to aged cows. Age groups showing reduced susceptibility were found to reach peak parasitaemia, temperature and anaemia before the more susceptible age groups. Heterologous challenge of the remaining experiment 1 and experiment 2 animals at 6 and 8 months respectively after primary inoculation, revealed all animals of all ages had a solid resistance to B. bovis.  相似文献

Babesiosis is caused by a haemotropic protozoal parasite of the genus Babesia, member of the phylum Apicomplexa and transmitted by the bite of an infected tick. There are many Babesia species affecting livestock, dogs, horses and rodents which are of economic significance. Infections can occur without producing symptoms, but babesiosis may also be severe and sometimes fatal caused by the intraerythrocytic parasite development. The disease can cause fever, fatigue and haemolytic anemia lasting from several days to several months. There are a number of effective babesiacides, but imidocarb dipropionate (which consistently clears the parasitaemia; often the only available drug on the market) and diminazene aceturate are the most widely used. Some Babesia spp. can infect humans, particularly Babesia microti and Babesia divergens, and human babesiosis is a significant emerging tick-borne zoonotic disease. Clinical manifestations differ markedly between European and North American diseases. In clinical cases, a combination of clindamycin and quinine is administered as the standard treatment, but also administration of atovaquone-azithromycin is successful. Supportive therapy such as intravenous fluids and blood transfusions are employed when necessary. More specific fast-acting new treatments for babesiosis have now to be developed. This should be facilitated by the knowledge of the Babesia spp. genome and increased interest for this malaria-like parasite.  相似文献

To assess the epidemiology of Babesia divergens in a veterinary practice based in the mid-east of France ("Monts du Lyonnais"), blood was collected from 254 cattle belonging to 24 herds. To assess the dynamics of the carrier state, six carriers were identified, treated with flumethrin and sampled once every 3 weeks during 6 months. Two different DNA extraction methods were compared. Each sample was tested for the presence of parasites using a PCR-RFLP test based on the 18S rRNA gene. The sensitivity of the test was equivalent to a parasitaemia as low as 10(-5)% (in "Filter Paper" samples) and 10(-6)% in 1 ml blood (extracted using "Matrix"). With the latter method, the rate of detection diminishes in the low parasitaemia range but could probably be improved. This test proved to be very useful in the detection of B. divergens carriers. Serology using IFAT showed 7% of the cattle seropositive, which is suggestive of a disease situation with a low clinical risk level. Analysis of the PCR results suggests a 20% prevalence rate of carriers in the cattle population. The use of the mean parasitaemia is proposed to serve as a babesiosis clinical risk indicator. This approach could also be used in other babesia infections provided the lowest detectable parasitaemia level (threshold level) could be resolved for each parasite species.  相似文献

Bovine babesiosis is a tick-borne disease caused by Babesia spp. haemoprotozoans. The disease is of great importance at tick enzootic unstable areas and hampers cattle production in several developing countries. The available immunisation alternatives are pre-immunition and attenuated vaccines. Despite being efficient and protective, they are unsafe as they use cattle blood cells as inoculum and may potentially spread other diseases. Another alternative to help in babesiosis control would be the identification of genetically resistant cattle to Babesia bovis infection. The objective of this work was to phenotype cattle based on primary response against B. bovis infection. Two-hundred and forty half-sib Hereford and Aberdeen Angus heifers (120 animals from each breed), 12-18-month-old na?ve cattle, originated from a tick-free area in Southern Brazil, were used in the experiment. Animals were monitored following an inoculation with 1x10(7)B. bovis parasitised erythrocytes. Results showed three different phenotypes: 1-'susceptible', animals with babesiosis clinical signs that received treatment to avoid death; 2-'intermediate', animals with clinical signs: parasitaemia, >or=21.5% reduction in packed cell volume (PCV) and increase in body temperature when compared to their pre-challenge physiological parameters, no specific treatment was needed as animals self recovered from the disease, and 3-'resistant', animals without clinical signs that showed B. bovis presence in blood smears, <21.5% PCV reductions, with little or no increase in body temperature and no need for babesiosis treatment. The frequencies of each phenotype were: 45.4, 26.7, and 27.9%, respectively, demonstrating the existence of phenotypic variation for B. bovis in Bos taurus cattle.  相似文献

Splenectomised calves infected with Babesia major were shown to have no resistance to challenge with Babesia divergens; however, initial infection with B divergens provided a good protection against subsequent challenge with B major. Previously unpublished data from 1958 to 1967 attest the occurrence of Babesia divergens in cattle in several endemic foci of Northeast Hungary. During that period the number of clinical cases showed fluctuation with intervals of 4-5 years and monophasic seasonality (peaking in June). In order to assess the current status of bovine babesiosis in that region, blood samples were collected from 654 cattle on 44 farms of 36 settlements in or near the endemic area during 2005, and serum levels of IgG antibodies to B. divergens were measured by indirect fluorescent antibody test (IFAT). Only 2 samples (0.3%) showed positivity. In one village clinical babesiosis was observed over the past few years. Animals brought into the endemic area during the spring developed haemoglobinuria in the summer of the same year, but those introduced during the summer or autumn showed clinical signs only after two years. Sampled animals born and raised locally had neither haemoglobinuria nor seroconversion. Reduction in the number of cases during the past decades may have been influenced by the availability of hosts (i.e. decrease of cattle breeding) and the activity of vectors associated with climate-related changes (e.g. increase of annual sunlight hours in the endemic area). This is the first report on the prevalence of antibodies to B. divergens in cattle in Hungary.  相似文献

Outbreaks of acute bovine babesiosis affecting several individuals within a herd are well known among veterinary practitioners and farmers within different parts of Sweden. Although outbreaks may not occur every year, they are feared by the farmers due to the unpredictability of the disease, its suddeness of onset, its rapid course, and the high mortality unless treated. Sporadic outbreaks of acute bovine babesiosis comprising several animals within a limited time period have been previously reported in Sweden (Linnaeus 1747, Bergman & Waxberg 1915, Heijbel 1928), in Finland (Hinderson 1928), in Norway as cited by Tambs-Lyche (1943), in the United Kingdom (Donnelly et al. 1970) and elsewhere within the area where B. divergens is distributed (Tambs-Lyche 1943, Purnell 1980). In all of these cases groups of animals had been transferred onto pastures where clinical cases of bovine babesiosis were known to occur from areas where the disease was rare or had never occurred; i.e. unexposed animals were transferred onto pastures with Babesia infected ticks. The farmers first became aware of this situation after the outbreak had occurred.  相似文献

Babesia divergens was cultivated in sheep erythrocytes in RPMI 1640 supplemented with 10% Fetal Calf Serum (FCS) or sheep serum. In vitro cultures in sheep red blood cells were initiated with human erythrocytes infected in vitro with B. divergens Rouen 1987 or with gerbil blood infected with several isolates from bovine origin. After the first subcultures on sheep erythrocytes, a ten-fold multiplication of the parasites was obtained within 48 h. Erythrocytes from three splenectomized sheep were infected in vitro with B. divergens; when parasitaemia reached 10%, the animals were inoculated with homologous parasitized erythrocytes. All sheep expressed hyperthermia with a peak between the 6th and the 9th day post-infection (p-i) and a transitory parasitaemia 10 days p-i. In vitro primary cultures were performed on two of these sheep, demonstrating the parasite persistence at very low parasitaemia in the infected animals. Splenectomized sheep can be used as a new model for B. divergens chronic infection.  相似文献

Thirty-eight dual purpose calves in the 3 agricultural zones of the state of Yucatan, Mexico were monitored for antibodies to Babesia bovis and B. bigemina from close to birth up to 12 months of age.The results showed that infection with both parasites is widespread throughout the 3 zones. Many of the animals had produced antibody to both parasites by 6 months of age indicating high transmission rates resulting in enzootic stability for both parasites.Any changes to agricultural practices in the state that could affect disease stability through either reduction in numbers of tick vectors or the introduction of susceptible adults will have to be accompanied with measures such as vaccination to limit losses due to clinical babesiosis.  相似文献

Babesia divergens, the main causative agent of bovine babesiosis in Western Europe, was isolated from naturally infected cattle. Ninety-six blood samples were examined by means of an in vitro culture technique in sheep erythrocytes: 19 of them were collected from animals in the acute phase of the disease with visible parasitemia on blood smears, while the 77 remaining animals showed no microscopically detectable parasites. B. divergens was cultured from the 19 first blood samples as well as from 31 samples collected from asymptomatic animals. The time period before parasites could be detected in the culture varied in the latter samples from 6 to 20 days. The effects of sampling condition (anticoagulant used) and storage length were tested. A good correlation was obtained between immunofluorescent antibody test and culture, with identical results (positive or negative) for 89.6% of the samples collected from asymptomatic animals. The sensitivity of the in vitro culture method was determined and was about 10 parasites/mL of whole blood from three independent experiments performed with three different isolates, confirming its suitability to detect and culture diverse B. divergens isolates from carrier cattle. The parasites could indeed be isolated 9 months after the acute babesiosis phase in the blood of naturally infected animals. The 50 isolates collected in this study were successfully subcultured, cryopreserved and resuscitated using the same culture medium. The in vitro isolation of B. divergens from asymptomatic carrier cattle was achieved and will allow the analysis of parasite diversity within cattle herds.  相似文献

Bovine babesiosis is an important tick-borne disease caused by Babesia bovis, B. bigemina and B. divergens. The first steps taken in the development of an effective vaccination strategy against bovine babesiosis followed the observations that animals, recovered from natural infection with Babesia were strongly protected against subsequent challenge. Further investigation indicated that the use of donor blood from recovered animals to infect recipient animals did not produce the severe form of the disease. The past century has seen a refinement of this original carrier-donor system to one using attenuated less virulent strains with standardized doses of known parasite concentration to ensure reliability. With the implementation of good manufacturing practices further changes were necessary in the production of these vaccines, such as freezing for long-term storage to allow sufficient time for pre-release safety and effectivity testing. Regardless of these improvements the vaccines are not without problems and breakdowns and breakthroughs occur from time to time. Despite considerable research efforts into the development of alternative more consumer friendly vaccines, none is immediately forthcoming and the live attenuated babesiosis vaccines are still used in many countries.  相似文献

Polyclonal bovine IgM-rheumatoid factors (IgM-RFs) were examined in sera of cattle immunized against babesiosis. An enzyme-linked immunosorbent assay (ELISA) was used enabling rapid screening of serum samples. Results obtained indicate a rise of serum IgM-RF levels with age in healthy bovines. However when animals of similar age and pertaining to the same herd were examined, levels of serum IgM-RF exhibited a wide distribution range. Mean values in 60 sera of 2 yrs old clinically healthy heifers originating from a single herd were of 452.60 +/- 201.26 e.u. at 1 in 1,000 serum dilution and of 202.37 +/- 137.86 e.u. at 1 in 4,000 serum dilution. In a herd where repeated vaccination of dams against babesiosis was carried out 3 to 6 weeks before delivery either with live Babesia bovis parasites or soluble antigens of in vitro grown organisms, no significant differences in mean IgM-RF values were found between revaccinated animals and a control group. Nor did the mean values of serum IgM-RF of calves born to the respective groups of dams exhibit significant differences. It thus appears that immunization of healthy cattle against this parasite does not affect mean serum IgM-RF levels.  相似文献

Clinical observations of Babesia canis infection in 63 dogs during a 1-year period are summarised, demonstrating the pathogenicity of the Babesia strain endemic in Hungary. Most patients had babesiosis in the spring and autumn, correlating with the seasonal activity of ticks. Male animals appeared in higher numbers, probably due to an overrepresentation of outdoor dogs. Uncomplicated babesiosis was diagnosed in 32 cases. The disease affected dogs of any age in this study. Symptoms were similar to those published from other parts of the world: lethargy, fever, splenomegaly, pallor, icterus, haemoglobinuria and presence of ticks were the most common observations. Thrombocytopenia, lymphopenia and neutropenia were frequent haemogram changes. Imidocarb appeared to be highly effective in eliminating the Babesia infection. Thirty-one animals demonstrated babesiosis with complications. Most Rottweilers (7/9) developed complicated disease. Old age was a risk factor for multiple complications. Multiple organ manifestations had poor prognosis. Hepatopathy (44%), pancreatitis (33%), acute renal failure (ARF; 31%) and disseminated intravascular coagulation (DIC; 24%) were frequent complications, while immune-mediated haemolytic anaemia (IMHA; 10%), acute respiratory distress syndrome (ARDS; 6%) and cerebral babesiosis (3%) were rarely observed. There was a significant difference between the mean age of dogs having uncomplicated disease, babesiosis with a single complication and babesiosis with multiple complications (3.4, 4.8 and 8.6 years, respectively, p < 0.001). The recovery rate (78, 68 and 25%, respectively, p = 0.005) and mortality rate (3, 21 and 67%, respectively, p < 0.001) also tended to differ significantly in these groups. Systemic inflammatory response syndrome (SIRS) and DIC are two possible pathways leading to multiple organ dysfunction syndrome (MODS) in babesiosis. DIC was found to predict MODS more sensitively in this study than SIRS: there were 6 animals developing MODS out of 11 identified with DIC, while only 5 dogs developed MODS out of 22 having SIRS.  相似文献

Tick fever is an important disease of cattle where Rhipicephalus (Boophilus) microplus acts as a vector for the three causal organisms Babesia bovis, Babesia bigemina and Anaplasma marginale. Bos indicus cattle and their crosses are more resistant to the clinical effects of infection with B. bovis and B. bigemina than are Bos taurus cattle. Resistance is not complete, however, and herds of B. indicus-cross cattle are still at risk of babesiosis in environments where exposure to B. bovis is light in most years but occasionally high. The susceptibility of B. indicus cattle and their crosses to infection with A. marginale is similar to that of B. taurus cattle. In herds of B. indicus cattle and their crosses the infection rate of Babesia spp. and A. marginale is lowered because fewer ticks are likely to attach per day due to reduced numbers of ticks in the field (long-term effect on population, arising from high host resistance) and because a smaller proportion of ticks that do develop to feed on infected cattle will in turn be infected (due to lower parasitaemia). As a consequence, herds of B. indicus cattle are less likely than herds of B. taurus cattle to have high levels of population immunity to babesiosis or anaplasmosis. The effects of acaricide application on the probability of clinical disease due to anaplasmosis and babesiosis are unpredictable and dependent on the prevalence of infection in ticks and in cattle at the time of application. Attempting to manipulate population immunity through the toleration of specific threshold numbers of ticks with the aim of controlling tick fever is not reliable and the justification for acaricide application should be for the control of ticks rather than for tick fever. Vaccination of B. indicus cattle and their crosses is advisable in all areas where ticks exist, although vaccination against B. bigemina is probably not essential in pure B. indicus animals.  相似文献
