为了探讨不同品种绵羊肌肉生长抑制素(myostatin,MSTN)表达水平及其与生长性状(体重、体高、体长、胸围和管围)的关联性,试验采用实时荧光定量 PCR方法比较阿勒泰羊、吐鲁番黑羊、也木勒白羊、哈萨克羊、巴什拜羊不同月龄(初生及1、2、3、4、5月龄)肌肉和脂肪MSTN mRNA的表达水平,运用SPSS 19.0软件分析与其生长性状的相关性.在肌肉中:初生时,阿勒泰羊、哈萨克羊、巴什拜羊MSTN基因mRNA表达水平均明显低于其他各月龄;也木勒白羊3月龄MSTN基因mRNA水平均显著低于其他各月龄(P<0.05);阿勒泰羊、吐鲁番黑羊MSTN基因mRNA在4月龄表达水平最高;哈萨克羊MSTN基因mRNA初生时表达水平最低,5月龄表达水平最高.在脂肪中:也木勒白羊MSTN基因mRNA表达水平初生时显著低于其他各月龄(P<0.05);阿勒泰羊、也木勒白羊、巴什拜羊MSTN基因mRNA 5月龄表达水平最高,巴什拜羊MSTN基因mRNA 5月龄显著高于其他各月龄(P<0.05);哈萨克羊MSTN基因mRNA 2月龄表达水平最高,显著高于5月龄(P<0.05).相关性分析结果发现,MSTN基因mRNA表达水平与生长性状多呈负相关关系.阿勒泰羊在肌肉和脂肪中MSTN基因mRNA表达水平与其生长性状均呈负相关关系;吐鲁番黑羊MSTN基因mRNA在肌肉中的表达水平与体高、体长呈负相关,其余均呈正相关;也木勒白羊MSTN基因mRNA在肌肉中的表达水平与胸围、管围呈正相关,在脂肪中表达水平与体长呈正相关,其余均呈负相关;哈萨克羊在肌肉和脂肪中除与体长呈正相关外,其余均呈负相关;巴什拜羊在肌肉中MSTN基因mRNA表达水平与体长、管围呈正相关,其余均呈负相关.MSTN基因mRNA在不同月龄不同品种间的表达水平存在差异性,无固定表达模式;且与生长性状多呈负相关关系,与部分体尺性状呈正相关关系.MSTN基因mRNA表达水平可能与生长性状和骨骼生长有直接关系.  相似文献

通过实时荧光定量也木勒白羊MSTN基因在不同组织(心、肝、脾、肺、肠、肾、尾脂、半膜、半键、股二、股三、背最长肌)表达量的多少,分析MSTN基因在不同组织的表达规律,为也木勒白羊肌肉生长性状的选育提供遗传学资料。结果表明:也木勒白羊MSTN基因在肌肉半膜、半键、股二、股三的表达量显著高于其他各组织(P0.05),背最长肌表达量显著高于各内脏组织和尾脂(P0.05),尾脂、肝、脾、肠、肾中表达量显著低于其他各组织(P0.05),内脏组织中肺的表达量最高,心居于第二,其余各内脏组织表达量均差异不显著(P0.05)。说明MSTN基因在12种组织中均有表达,但在内脏和尾脂中的表达量低于肌肉组织。  相似文献

肌肉生长抑制素(MSTN)是一种骨骼肌生长发育的负调控因子.试验以不同生长阶段的阿勒泰羊羔羊为研究对象,采用实时荧光定量PCR技术测定了肌肉和脂肪组织中MSTN mRNA相对表达量.比较该基因在不同月龄阿勒泰羊羔羊肌肉和脂肪组织中的表达差异.结果表明:阿勒泰羊0~6月龄羔羊MSTN mRNA在肌肉和脂肪组织中均有表达.4月龄羔羊MSTN mRNA表达量在肌肉中最高,0~4月龄的表达量逐渐上升,4~6月龄逐渐下降.6月龄羔羊脂肪中的MSTN mRNA表达量最高.不同月龄阶段阿勒泰羊羔羊肌肉和脂肪MSTN mRNA表达量存在差异,这些差异可能会影响其生长发育和产肉性能等指标,这对进一步理解MSTN基因的调控作用奠定了基础.  相似文献

运用荧光定量PCR技术对不同月龄(0日龄或初生、1月龄、2月龄、3月龄、4月龄、5月龄)新疆巴什拜羊肌肉和脂肪MSTN基因的表达量进行定量,2-ΔΔCT方法分析不同生长阶段MSTN基因表达量的差异,以期获取MSTN基因在不同月龄巴什拜羊的表达规律,同时丰富与肌肉生长相关基因的研究。结果表明:初生、3月龄新疆巴什拜羊肌肉中MSTN基因的表达量极显著低于1、2、4、5月龄(P0.01),1月龄的表达量又极显著高于其他各月龄(P0.01),2月龄显著低于5月龄(P0.05);初生时新疆巴什拜羊脂肪中MSTN基因的表达量极显著低于4月龄和5月龄(P0.01),1月龄MSTN基因的表达量极显著高于2月龄和3月龄(P0.01)、极显著低于4月龄和5月龄(P0.01),5月龄MSTN基因的表达量极显著高于4月龄(P0.01)。表明巴什拜羊肌肉生长发育在初生时较快,在1月龄和2月龄生长发育较慢,3月龄生长发育最快,随后生长发育趋于平稳;脂肪的生长发育在初生后略快于1月龄,2月龄和3月龄的生长发育较快,4月龄较慢,5月龄生长发育慢于其他各月龄。  相似文献

探索MSTN(生长抑制素)基因在脂肪组织中不同生长阶段的表达差异,为哈萨克羔羊的良种选育提供遗传学资料。采用荧光定量1~6月龄哈萨克羔羊脂肪组织中MSTN基因的相对表达量,2-ΔΔCt处理不同月龄相对表达量数据,SPSS 17.0软件分析表达量差异。结果表明:MSTN基因在1~6月龄哈萨克羔羊尾脂中的相对表达量6月龄最高,1、2、3、4各月龄之间无显著性差异(P0.05),5月龄最低。说明MSTN基因在脂肪中的表达量在生长发育的前期差异不显著(P0.05),在5月龄显著降到最低点,随后在6月龄达到最高。  相似文献

试验旨在分析MSTN基因在1~6月龄哈萨克羔羊肌肉组织中表达量及其与各项生长指标的相关性,为哈萨克羊的良种选育提供科学依据。随机选取5只哈萨克公羔羊,每月定时在羔羊活体腿部取肌肉组织,软尺测量体长、体高、体重、管围、胸围等各项生长指标。实时荧光定量PCR分析1~6月龄哈萨克羔羊肌肉组织中MSTN的表达量,相关性分析MSTN基因在羔羊肌肉中的表达量与各项生长指标的关联性。结果显示:哈萨克羔羊各项生长指标在1、2月龄快速增长,体重从4月龄开始增重缓慢,体长、体高、胸围在3月龄出现增长缓慢,管围在3月龄起基本停止增长。MSTN基因在1~4月龄哈萨克羔羊肌肉组织中表达量均无显著性差异(P0.05),呈现略微上升的趋势,5月龄达到最高峰,6月龄迅速下降。MSTN基因在肌肉中表达量与体长呈显著负相关(P0.05),相关系数为-0.472,与体重、胸围呈弱负相关,与管围、体高呈弱正相关。  相似文献

本试验旨在探究肌肉生长抑制素(myostatin,MSTN)基因在小鼠不同发育阶段心肌组织中的表达情况,分别选取幼年(7日龄)、性成熟(21~28日龄)、体成熟(42~56日龄)和老年(84日龄以上)4个发育阶段的健康小鼠(各3只)为研究对象,采用实时荧光定量PCR、HE染色、免疫组织化学等方法研究小鼠不同发育时期MSTN基因在心肌组织中的表达及不同时期心肌组织的发育。实时荧光定量PCR结果显示,MSTN基因在小鼠不同发育阶段心肌组织中均有表达,且其在小鼠老年时期的表达量最高,体成熟时期的表达量最低;幼年小鼠和性成熟小鼠心肌组织中MSTN基因表达水平显著低于老年小鼠(P0.05),而体成熟小鼠则极显著性低于老年小鼠(P0.01)。HE染色结果显示,小鼠心肌细胞核及肌纤维的生长在不同发育时期明显不同,其中幼年时期的细胞核最多,肌纤维排列相对较紧密,而在性成熟、体成熟、老年时期心肌的发育指标依次降低。免疫组织化学结果显示,MSTN基因主要在心肌细胞核中表达,其各个时期表达量与实时荧光定量PCR的结果相符合。  相似文献

比较分析HSL和PPARγ基因在湘西黄牛不同组织中的发育性表达规律,以探讨HSL和PPARγ基因与湘西黄牛脂肪酸代谢的关系。利用实时荧光定量PCR技术检测HSL和PPARγ基因在6,18,30月龄湘西黄牛的肝脏、肌肉组织(背最长肌)、脂肪组织(皮下脂肪和腹腔脂肪)中mRNA的相对表达量。结果表明:HSL基因在湘西黄牛肝脏、背最长肌、皮下脂肪和腹腔脂肪的4个部位中的表达量随月龄增长依次极显著减少(P0.01),而PPARγ基因正好相反。6月龄时,肝脏中HSL基因的表达量显著高于其他3个组织(P0.05),PPARγ基因皮下脂肪表达量显著高于肝脏(P0.05); 18月龄时,HSL基因在4个组织中的表达量按大小关系依次为肝脏≈背最长肌皮下脂肪腹腔脂肪; PPARγ基因在4个组织中的表达量按大小关系依次为皮下脂肪肝脏≈腹腔脂肪背最长肌; 30月龄时,HSL基因在4个组织中的表达量按大小关系依次为肝脏背最长肌皮下脂肪≈腹腔脂肪; PPARγ基因在4个组织间的表达呈现两两差异显著(P0.05),其表达量依次为肝脏皮下脂肪腹腔脂肪背最长肌。30月龄的粗脂肪(EE)含量和饱和脂肪酸(SFA)含量显著高于6月龄和18月龄的(P0.05),18月龄和30月龄湘西黄牛肉多不饱和脂肪酸(PUFA)含量显著高于6月龄的(P0.05); 30月龄时单不饱和脂肪酸(MUFA)的含量较高,但显著低于18月龄的(P0.05)。由此可见,HSL和PPARγ基因在湘西黄牛不同部位脂肪组织和肌肉组织中均有表达且随月龄与部位的不同表达不同,存在月龄和组织差异性,且二者的表达规律呈负相关。在动物生产应用中,建议湘西黄牛在30月龄左右再屠宰食用,口感及营养价值更好。  相似文献

研究旨在比较分析固醇调节元件结合蛋白-1(Sterol regulatory element-binding protein 1,SREBP-1)基因在湘西黄牛不同组织中的发育性表达规律,以探讨SREBP-1基因与肉牛脂肪代谢的关系。选取湖南德农牧业集团有限公司国家级保种场不同月龄(6月龄、18月龄、30月龄)湘西黄牛各4头,利用实时荧光定量PCR检测肝脏、背最长肌、皮下和腹腔脂肪组织SREBP-1基因的相对表达量。结果表明:①SREBP-1基因在湘西黄牛肝脏、背最长肌、皮下和腹腔脂肪的4个部位中的表达量随月龄增加依次显著增加(P0.01);②在6月龄时,皮下脂肪SREBP-1基因的表达量显著高于肝脏与腹腔脂肪(P0.05);③18月龄时,SREBP-1基因在肝脏中的表达量显著高于背最长肌与腹腔脂肪(P0.05);④30月龄时,SREBP-1基因在肝脏组织中的表达量显著高于其他3个组织(P0.05),在皮下脂肪中的表达量显著高于背最长肌与腹腔脂肪(P0.05)。由此可见,SREBP-1基因在湘西黄牛不同部位脂肪组织和肌肉组织中均有表达,且其表达随月龄与部位的不同而不同,存在月龄和部位的差异性。  相似文献

To explore the different varieties of five Xinjiang local sheep myostatin (MSTN) expression levels and their correlation with growth traits (body weight,body height,body length,heart girth and cannon circumference).The MSTN mRNA expression level in different month(Birth,1 months,2 months,3 months,4 months,5 months) of muscle and fat in Altay sheep,Turpan Black sheep,Emil White sheep,Kazakh sheep,Bashbay sheep by quantitative Real-time PCR method,using SPSS 19.0 software to analyze the correlation with growth traits.The MSTN mRNA expression levels in muscle:The MSTN gene mRNA expression levels of Altay sheep,Kazakh sheep and Bashbay sheep at birth were lower than other months;The MSTN gene mRNA expression level of Emil White sheep in 3 months was significantly lower than other months (P<0.05);The MSTN gene mRNA expression levels of Altay sheep,Turpan Black sheep were the highest in 4 months;The MSTN gene mRNA expression level of Kazakh sheep was the lowest at birth,and was the highest in 5 months.The MSTN gene mRNA expression levels in fat:The MSTN gene mRNA expression level of Emil White sheep at birth was significantly lower than other months (P<0.05); The MSTN gene mRNA expression levels of Altay sheep,Emil White sheep and Bashbay sheep were the highest in 5 months,the MSTN gene mRNA expression level of Bashbay sheep in 5 months was significantly higher than other months (P<0.05);The MSTN gene mRNA expression level of Kazakh sheep was the highest in 2 month,it was significantly higher than 5 months (P<0.05).Correlation analysis revealed that the MSTN gene mRNA expression levels were mostly negatively correlated with growth traits.The MSTN gene mRNA expression levels in muscle and fat of Altay sheep were all negatively correlated with growth traits;The MSTN gene mRNA expression levels in muscle of Turpan Black sheep were negative correlated with body length and body height,others were positively;The MSTN gene mRNA expression levels in muscle of Emil White sheep were positive correlated with heart girth and cannon circumference;The MSTN gene mRNA expression levels in fat were positive correlated with body length,others were negatively correlation;The MSTN gene mRNA expression levels in muscle and fat of Kazakh sheep were positive correlated with body length,others were negatively correlation;The MSTN gene mRNA expression levels in muscle of Bashbay sheep were positive correlated with body length and cannon circumference,others were negatively correlation.The MSTN gene mRNA expression level was discrepancy in different month of different varieties,and was no fixed expression pattern;The MSTN gene mRNA expression levels were positively correlated with the part of the body size.The MSTN gene mRNA expression levels might have a direct relationship with the growth traits and bone growth.  相似文献

为了探究肌肉生长抑制素(myostatin,MSTN)基因在肌肉和脂肪中的DNA甲基化模式及mRNA表达水平,试验以5月龄巴什拜羊羔羊为研究对象,采用亚硫酸氢盐测序法(bisulfite sequencing PCR,BSP)检测MSTN基因启动子区和第1外显子甲基化模式,并通过实时荧光定量PCR检测MSTN基因在巴什拜羊股二头肌、股三头肌、半腱肌、半膜肌、背最长肌和尾脂中的mRNA表达水平。结果显示,肌肉组织甲基化概率高于脂肪组织,其中,股二头肌、股三头肌、半腱肌、半膜肌、背最长肌和尾脂的甲基化概率分别为74.2%、74.2%、83.2%、83.7%、82.1%和25.3%,MSTN基因在股二头肌、股三头肌、半腱肌、半膜肌、背最长肌中的表达水平显著低于尾脂(P < 0.05),股二头肌、股三头肌、半腱肌、半膜肌和背最长肌之间差异不显著(P > 0.05),巴什拜羊肌肉和脂肪MSTN基因DNA甲基化水平与MSTN基因表达量呈显著负相关(r=-0.886,P < 0.05)。  相似文献

In order to investigate DNA methylation and expression levels of myostatin (MSTN) gene in mscule and fat, 5 months of age of Bashiby sheep were selected, the promoter region and exon 1 methylation levels of MSTN gene was analyzed using bisulfite sequencing PCR (BSP). Real-time PCR was used to detect the expression level of MSTN gene in biceps femoris, femoral triceps, semitendinosus, semimembranosus, longissimus dorsi muscle and tail fat of Bashiby sheep. The results showed that the methylation probability of muscle was higher than fat, the methylation probability of biceps femoris, femoral triceps, semitendinosus, semimembranosus, longissimus dorsi muscle and tail fat were 74.2%, 74.2%, 83.2%, 83.7%, 82.1% and 25.3%, respectively. The expression levels of MSTN gene in biceps femoris, femoral triceps, semitendinosus, semimembranosus, longissimus dorsi muscle were significantly lower than tail fat in Bashiby sheep (P < 0.05), but there were no significant difference among biceps femoris, femoral triceps, semitendinosus, semimembranosus and longissimus dorsi muscle (P > 0.05).The DNA methylation was negatively correlated with the expression levels in muscle and fat of Bashiby sheep (r=-0.886, P < 0.05).  相似文献

To explore the influence of Gaopo pig myostatin (MSTN) gene polymorphism on meat quanlity traits (moisture,crude fat,crude protein,crude ash, loin muscle area, marbling score, pH value, color of meat,tenderness,drip loss, water loss rate), MSTN gene was chosen as a candidate gene for research meat quality traits in this study. 50 Gaopo pigs at 10 months old were selected,the single nucleotide polymorphism (SNP) of three exons of MSTN gene in Gaopo pig were detected by direct sequencing of PCR amplification products, and the correlation between MSTN gene SNP and meat quality traits was analyzed using SPSS 20.0 software. The results showed that a C/T mutation had been found at 63 bp of MSTN gene exon 3,the site was silent mutation, and did not caused changes of encoding amino acid. Genotype analysis showed only three samples had mutations in the site, CC genotype was the dominant type and allele C was the dominant gene, but there was no TT homozygous genotype. The test of Hardy-Weinberg equilibrium showed the research group obeyed the genetic equilibrium (P>0.05). Population genetic parameter analysis showed the heterozygosity (He) of C63T was lower, and had a low variation in Gaopo pig population. Polymorphism information content (PIC) analysis showed that it was a low polymorphic loci (PIC<0.25), and the site could provide a little genetic imformation. There was no significant difference between the different genotypes and meat quality traits (P>0.05).Given the above, the polymorphism of MSTNgene exon was found in Gaopo pig, with less variation and relatively conservative.It's needed to expand the number of sample for further explore whether the site could be considered as genetic markers for meat quality traits of Gaopo pig or not.  相似文献

为了探讨肌肉生长抑制素(myostatin,MSTN)基因的多态性对高坡猪肉质性状(水分、粗脂肪、粗蛋白质、粗灰分、眼肌面积、大理石纹、pH、肉色、嫩度、滴水损失、失水率)的影响,试验以MSTN基因作为肉质性状候选基因,以10月龄的50头高坡猪为研究对象,采用PCR扩增产物直接测序的方法对高坡猪MSTN基因3个外显子的单核苷酸多态性(SNP)进行研究,运用SPSS 20.0软件分析MSTN基因SNP与肉质性状的关联性。结果表明,仅在高坡猪MSTN基因第3外显子63 bp处检测到1个C/T突变位点,该突变为同义突变,未引起编码氨基酸的改变。基因型分析发现,仅有3个个体在该位点发生突变,CC基因型为优势基因型,C为优势等位基因,无TT纯合基因型。经χ2适合性检验分析,该SNP在研究群体中处于Hardy-Weinberg平衡状态(P>0.05)。群体遗传参数分析发现,C63T标记位点的杂合度(He)相对较低,表明其在高坡猪群体中的变异较小;就多态信息含量(PIC)而言,该位点属于低度多态(PIC<0.25),说明该遗传标记能够提供少量的遗传信息。将该位点的不同基因型与肉质性状指标进行关联分析表明,所有肉质性状指标在不同基因型间差异均不显著(P>0.05)。综上所述,MSTN基因外显子在高坡猪中存在多态性,但变异较少,相对保守,能否作为高坡猪肉质性状的遗传标记有待于扩大样本数量进一步研究。  相似文献

[目的]研究MS删基因在不同月龄滩羊不同肌肉组织中的表达差异。[方法]根据GenBank已发表的绵羊MSTN基因序列(NM001009428.1)设计l对引物,以滩羊肌肉组织cDNA为模板,采用实时荧光定量PCR技术(Real—timePCR)检测MS7Yv基因在不同月龄滩羊的背最长肌、腿肌和胸肌中的表达量。[结果]MS驯基因在背最长肌中的表达量最高,其次为胸肌,腿肌中的表达量最低;随着月龄的增加,滩羊3种肌肉组织中的表达量均呈先升高、后降低、再升高的趋势。[结论]不同月龄滩羊肌肉中的MSTNmRNA表达量存在差异.这些差异可能会影响其生长发育和产肉性能等指标。  相似文献

The aim of this study was to investigate the effects of MSTN and MyoG genes on goose skeletal muscle growth.In this study,MSTN and MyoG genes expression were detected in breast and leg muscle of Zi and Rhine goose by Real-time PCR,and the correlations between genes expression levels and carcass traits were investigated.The results showed that the breast muscle weight and breast muscle rate of Rhine goose were extremely significant higher than Zi goose (P<0.01).MSTN and MyoG mRNA expression in breast muscle of Zi goose were significantly higher than that of Rhine goose,and the mRNA level of MSTN in leg muscle of Rhine was extremely significant higher than that of Zi goose (P<0.01),there was no significant difference of MyoG mRNA between Zi goose and Rhine goose (P>0.05).There was extremely significant difference between MSTN mRNA expression in breast muscle and leg muscle of Zi goose (P<0.01).MSTN mRNA expression in leg muscle was significantly higher than that of breast muscle of Rhine goose (P<0.05).There was extremely significant difference between MyoG mRNA expression in breast muscle and leg muscle of Zi goose and Rhine goose (P<0.01).There was a extremely significant negative correlation between MSTN mRNA expression in breast muscle with body weight,breast muscle weight and breast muscle percentage (P<0.01).There was a extremely significant positive correlation between MyoG mRNA expression in breast muscle and leg muscle with body weight (P<0.01).MSTN and MyoG gene might have positive and negative regulation effect on muscle growth.  相似文献

This study was aimed to analyze the association of myostatin (MSTN) gene polymorphisms with meat quality traits in Chishui Black-bone chicken. The DNA pooling was constructed and the direct sequencing technology of PCR product was used to detect the single nucleotide polymorphism (SNP) of MSTN gene. The results showed that C324T were detected in exon 1 of MSTN gene in Chishui Black-bone chicken. However, there was not found SNP site in exon 2 and exon 3 of MSTN gene. The C324T site was devised into three genotypes of CC, CT and TT. The association relationships between the genotype of MSTN gene and meat quality traits revealed that chest muscle pH and leg muscle pH value of Chishui Black-bone chicken with CT genotype were significantly higher than that of CC and TT genotypes (P<0.05), the shear force of CT genotype was significantly higher than that of TT genotype (P<0.05), there was no significant difference in other meat quality traits (P>0.05). This results suggested that the pH value and shear force of MSTN gene had a larger genetic effect in Black-bone chicken,MSTN gene was one of genes which could control the preliminary inference, might affect the pH value and shear force or with the main gene chain in Black-bone chicken, and could be used as pH value and shear force of molecular markers of Black-bone chicken.  相似文献
