The aim of the present study was to determine the combined effect of both stress and EFA deficiency on several biological and biochemical parameters. Fish were fed during 15 weeks two isocaloric and isoproteic diets: a control diet based on fish oil and formulated to meet the n-3 HUFA requirements for this species (1.5% of n-3 HUFA) and a deficient diet containing beef tallow and formulated to be deficient in n-3 HUFA. Each experimental diet was evaluated both at high and low stocking densities (10 and 3.2 kg m–3 of initial density, respectively).High stocking density produced a chronic stress situation with elevation of plasma cortisol levels. It also caused a reduction in hepatosomatic index and liver lipid contents, increasing the oleic acid/n-3 HUFA ratios in the polar lipids. Fish fed the EFA deficient diet at low stocking density showed common deficiency symptoms. High stocking density in fish fed the EFA deficient diet induced a higher degree of EFA deficiency symptoms leading to mortality, liver steatosis, liver lipid deposition, reduced muscle lipid and reduced n-3 HUFA contents, which particularly affected EPA, but not DHA, suggesting a preferential retention of the latter fatty acid, specially in the phosphoglycerides fraction.  相似文献

An experiment was conducted to determine the effect of an essential fatty acid (EFA) deficient diet on growth, immune status and renal morphology of juvenile gilthead sea bream, using two diets: a control diet containing 2% (DW) of n-3 high unsaturated fatty acid (n-3 HUFA) and a diet formulated to be deficient in EFA for this species and containing 0.5% DW of n-3 HUFA (diet NFA). After 9 weeks of feeding the EFA-deficient diet fish showed a reduction in growth compared with the control group (107.48 +/- 9.14 and 123.14 +/-11.87 g final weight respectively). Fish fed the NFA diet showed a reduction in the erythrocyte volume together with increased erythrocyte fragility, haemoglobin content and red blood cell count in comparison with fish fed the control diet. EFA deficiency also reduced cellular immunity in terms of neutrophil activity and the number of circulating lymphocytes. The serum alternative complement pathway was markedly reduced in fish fed the EFA deficient diet. Fish fed the control diet showed glomeruli with a well-defined Bowman's space and normal renal tubes. However, up to 88% of fish fed the EFA-deficient diet showed alterations in renal morphology affecting at least 50% of the glomeruli, which showed extreme dilation of capillaries and occlusion of Bowsman's capsule. Mesangial proliferation and diffuse thickening of the capillary walls, as well as renal tube degeneration, were also observed.  相似文献

A feeding experiment was conducted on gilthead seabream (Sparus aurata) broodstock to investigate the incidence of n-3 highly unsaturated fatty acids (n-3 HUFA) dietary deficiencies on the lipid composition of female liver, gonads and eggs, in relation to spawning quality. Broodstock were fed a control (C) diet or a n-3 HUFA deficient (D) but linolenic acid rich diet. After 20 weeks of feeding, the results showed that levels of total neutral (TNL) and total polar (TPL) lipids of female gonads and eggs were independent of diet. However the fatty acid composition of phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylinositol (PI) of female liver, gonads and eggs in the two groups of fish showed marked differences, reflecting the influence of fatty acid levels in the broodstock diets. This influence was even higher in TNL than in the phospholipid classes examined. In fish fed n-3 HUFA deficient diet, fatty acid composition of TNL of female gonads and eggs reflected the diet more than liver. A higher egg production in broodstock fed C diet (1.8% n-3 HUFA in diet) was extended to spawning quality such as percentages of fertilised and hatched eggs.  相似文献

Feeding experiments and laboratory analyses were conducted to establish the essential fatty acid (EFA) requirement of red drum (Sciaenops ocellatus). Juvenile red drum were maintained in aquaria containing brackish water (5 ± 2‰ total dissolved solids) for two 6-week experiments. Semipurified diets contained a total of 70% lipid consisting of different combinations of tristearin [predominantly 18:0] and the following fatty acid ethyl esters: oleate, linoleate, linolenate, and a mixture of highly unsaturated fatty acids (HUFA) containing approximately 60% eicosapentaenoate plus docosahexaenoate. EFA-deficient diets (containing only tristearin or oleate) rapidly reduced fish growth and feed efficiency, and increased mortality. Fin erosion and a “shock syndrome” also occurred in association with EFA deficiency. Of the diets containing fatty acid ethyl esters, those with 0.5–1% (n-3) HUFA (0.3–0.6% eicosapentaenoate plus docosahexaenoate) promoted the best growth, survival, and feed efficiency; however, the control diet containing 7% menhaden fish oil provided the best performance. Excess (n-3) HUFA suppressed fish weight gain; suppression became evident at 1.5% (n-3) HUFA, and was pronounced at 2.5%. Fatty acid compositions of whole-body, muscle and liver tissues from red drum fed the various diets generally reflected dietary fatty acids, but modifications of these patterns also were evident. Levels of saturated fatty acids appeared to be regulated independent of diet. In fish fed EFA-deficient diets (containing only tristearin or oleate), monoenes increased and (n-3) HUFA were preferentially conserved in polar lipid fractions. Eicosatrienoic acid [20:3(n-9)] was not elevated in EFA-deficient red drum, apparently due to their limited ability to transform fatty acids. Red drum exhibited some limited ability to elongate and desaturate linoleic acid [18:2(n-6)] and linolenic acid [18:3(n-3)]; however, metabolism of 18:3(n-3) did not generally result in increased tissue levels of (n-3) HUFA. Based on these responses, the red drum required approximately 0.5% (n-3) HUFA in the diet (approximately 7% of dietary lipid) for proper growth and health.  相似文献

The effects of dietary n-3 highly unsaturated fatty acid (n-3 HUFA) on eggs and larval quality were investigated in the Chilean flounder Paralichthys adspersus . Broodstock were fed with three formulated diets with similar proximate compositions but different n-3 HUFA (2.1%, 3.1% or 4.1%) estimated levels from 5 months before and during the spawning period. The diet with an intermediate n-3 HUFA level resulted in a significantly higher ( P <0.05) percentage of buoyant eggs (68.2 ± 2.9%), fertilization (92.8 ± 3.9%), normal cell cleavages (93.5 ± 1.9%), hatching rate (87.7 ± 4.1%) and normal larvae (76.3 ± 3.7%) compared with the other two diets. In contrast, high levels of n-3 HUFA produced larvae with a higher survival capacity when subjected to fasting. The diet with the lowest content of n-3 HUFA produces lower quality eggs and larvae. The n-3 HUFA level in eggs increased with an increase in the dietary level, and the n-3/n-6 ratios were 1:1, 2:1 and 3:1. The DHA/EPA and EPA/ARA ratios of 2 and 4 in eggs, respectively, were associated with improved egg and larval quality and were similar to the ratios found in eggs from wild broodstock. Attainment of optimal fatty acid contents in broodstock diets is one of the key factors for producing the high-quality spawning required for managed culture of this flounder.  相似文献

Two hundred and twenty rainbow trouts (IBW: 700 g) were randomly allotted to four tanks, with a male/female ratio of 0.56. Fish were fed for 168 d with four experimental diets containing herring oil, cod liver oil and coconut oil with the following inclusion rates: diet A: 12-1-0% respectively; diet B: 6-1-6%; diet C: 0-1-12%; diet D: 0-0-13%. Irrespective of the dietary treatment, weight gains of broodstocks were high (> 3 g/d) and FCR below 2. No significant difference was observed concerning the total amount of eggs spawn, egg average weight (82.5 mg/egg) and lipid content (5.4 mg/egg). However, the fatty acid profile of eggs was significantly affected by the dietary treatments. The content of unsaturated fatty acids, particularly the n-3 fatty acid series (EPA and DHA) significantly decreased with increasing levels of coconut oil in the diet.  相似文献

A feeding experiment was carried out to determine the effect of a diet lacking n-3 and n-6 highly unsaturated fatty acids (HUFA) on lipid and fatty acid content in intestine and gills of male gilthead seabream (Sparus aurata L.) broodstock at different stages of the reproductive cycle: November (pre-spermatogenesis), March (spermatogenesis), and June (post-spermatogenesis). For this purpose, gilthead seabream broodstock were fed either a control diet (C) or an n-3 and n-6 HUFA-deficient diet (D). The results showed no changes in fatty acid content of polar lipids of intestine and gills from fish fed diet C at different stages of the reproductive cycle. However, significant changes were observed in the fatty acid content of neutral lipids in intestine but not in gills in this group. Thus, between November and March, saturates and n-3 HUFA decreased while monoenes increased. In June, the contents of these fatty acids had returned to their initial values (November). Moreover, in fish fed diet D, the fatty acid content of neutral lipid changed in both intestine and gills. In intestine NL, a decrease in saturates and n-3 HUFA and an increment in monoenes were observed from November to June. In gills, a decrease was also observed in n-3 HUFA from NL along the cycle. Nevertheless, n-6 HUFA content remained unchanged. These results show both tissue specificity in seasonal mobilization of fatty acids linked to reproductive processes and the influence of dietary fatty acids on body composition.  相似文献

The influence of broodstock dietary lipids on egg quality and egg fatty acid composition throughout the spawning season of gilthead seabream was investigated. For this purpose, the fish were fed for 7 months either a control diet (diet C) or a diet deficient in n−3 highly unsaturated fatty acids (n−3 HUFA) but rich in both oleic (18:1n−9) and linolenic (18:3n−3) acids (diet D). Eggs spawned by both groups of fish were sampled at the beginning, middle and end of the spawning season and the fatty acid composition of their neutral (NL) and polar lipids (PL) determined. In the early season, percentages of fertilized and hatched eggs, relative proportions of NL and PL as well as their fatty acid compositions, were not affected by the lipid composition of the broodstock diet. However, the eggs spawned during the middle and late seasons showed marked differences among the two groups of fish, clearly reflecting the influence of dietary fatty acids. This influence was more evident in the neutral lipid fraction than in the polar lipids. No correlation was found between the number of buoyant eggs and eicosapentaenoic (20:5n−3, EPA), docosahexaenoic (22:6n−3, DHA) fatty acids or total n−3 HUFA contents in egg phospholipids. However, a negative correlation was detected when percentages of fertilized eggs were compared with the levels of 18:1n−9, 18:3n−3 and with the ratio 18:1n−9/n−3 HUFA present in the phospholipids. Our results indicate the importance of maintaining not only the level of n−3 HUFA in egg membrane phospholipids, but also the balance between n−3 HUFA and other fatty acids such as 18:1n−9 and 18:3n−3, in order to obtain a high spawning quality.  相似文献

Total lipid (TL), lipid classes and fatty acid composition of neutral (NL) and polar (PL) lipids were studied in the gonads, liver and muscle of gilthead seabream males ( Sparus aurata ) fed a control diet (diet C) or an n-3 and n-6 highly unsaturated fatty acids (HUFA)-deficient diet (diet D), at different stages of the reproductive cycle. Between pre-spermatogenesis (November) and spermatogenesis (March), the lipid content was high and particularly rich in cholesterol, phosphatidylcholine and phosphatidylethanolamine in gonads from both dietary groups. At post-spermatogenesis (June), TL and especially PL dramatically decreased in the gonads from both groups. However, at this period diet C fish gonads were richer in triacylglycerides (TAG) than those from diet D fish. The liver lipid contents and particularly TAG were over 200% lower in June than in March for both groups. Nevertheless, the most noteworthy depletion of lipids during this period was achieved by the n-3 HUFA in diet D fish. Conversely, arachidonic acid (20:4n-6) did not decrease in NL or PL from gonads and liver in groups C and D. Muscle lipids from diet C fish were relatively insensitive to seasonal influences. However, in June, the muscle TAG content was significantly reduced in diet D fish.  相似文献

Long-chain polyunsaturated fatty acids (LCPUFA) with 20 or 22 carbons are considered important to the development of infants and sometimes added to infant formulae. In this study, two characteristic sources of n-3 LCPUFA (fish oil and microalgal oil) were orally administrated to rat pups of mildly n-3 PUFA — deficient dams to compare the consequences of the administration. The milk from the dams fed a n-3 PUFA — restricted diet contained less n-3 LCPUFA than that of the dams fed a control diet. Pups were administered 1 mg/g weight of the test oil at the age of 5–7 days. At the age of 7 days, they were sacrificed before or after the administration and fatty acid compositions of the stomach and serum lipid were studied. The administration changed docosahexaenoic acid (DHA; 22:6n−3) levels in the stomach contents and serum lipids with time. Eicosapentaenoic acid (EPA; 20:5n−3) levels increased immediately after the administration of fish oil. The administration of microalgal oil also affected the serum lipid EPA level, in spite of a lack of EPA. In this study, both oils effectively supplemented DHA. Fish oil returned the serum EPA level close to the control value while microalgae oil had little effect.  相似文献

SUMMARY: The effect of Japanese catfish Silurus asotus lipid intake on the lipid metabolism of male adult mice was determined by measuring the total cholesterol, triacylglycerol, phospholipid, and fatty acid content of plasma and liver. Mice were fed diets containing 5% lard, 5% Thai catfish oil, 5% Japanese catfish lipids, or 5% sardine lipids for 4 weeks. The arachidonic acid (20 : 4n-6) content was highest in the livers of mice fed the Japanese catfish lipid diet and docosahexaenoic acid (22 : 6n-3) content was higher in livers of the Japanese catfish lipid and sardine lipid diet groups than those of the lard and Thai catfish diet groups. However, no significant differences were observed in the 22 : 6n-3 levels in either plasma or liver between the Japanese catfish lipid and sardine lipid diet groups. Although the Japanese catfish lipid diet contains, by far, the highest cholesterol content (143 mg/100 g), there was no significant difference in the plasma cholesterol levels between the lard, Thai catfish oil and Japanese catfish lipid diet groups. However, the liver cholesterol levels of mice fed the Japanese catfish lipid diet were lower than those of mice fed the Thai catfish oil diet, and were similar to those of animals fed a sardine lipid diet. These results suggest that Japanese catfish lipids are suitable components of the diet for ingestion of 20 : 4n-6 and 22 : 6n-3, and for decreasing the cholesterol content of liver.  相似文献

European sea bass juveniles (14.4±0.1 g mean weight) were fed diets containing different levels of fish oil then of n-3 highly unsaturated fatty acids (n-3 HUFA) for 12 weeks. The fish performance as well as fatty acid (FA) composition of neutral and polar lipids from whole body after 7 and 12 weeks feeding were studied. The requirements of juvenile sea bass for n-3 highly unsaturated fatty acids (n-3 HUFA) were studied by feeding fish diets containing six different levels of n-3 HUFA ranging from 0.2% to 1.9% of the diet, with approximately the same DHA/EPA ratio (1.5:1).

The growth rate at the end of the trial showed significant differences. Fish fed low dietary n-3 HUFA (0.2% DM of the diet) showed significantly lower growth than the diet 3 (0.7%), then no further improvement (P>0.05) of growth performance was seen by elevating the n-3 HUFA level in the diet up to 1.9% (diet 6). No difference in feed efficiency, protein efficiency ratio or protein retention was observed among treatments, nor in protein and total lipid content. However, the n-3 HUFA levels in diets highly influenced fish fatty acid composition in neutral lipid, while polar lipid composition was less affected. Comparison of polar lipid content after 7 or 12 weeks indicated that DHA remained stable at the requirement level, while arachidonic acid decreased with time. Results of this experiment suggest that the requirement for growth of n-3 HUFA of juvenile sea bass of 14 g weight is at least 0.7% of the dry diet.  相似文献

真鲷幼鱼摄食缺乏多不饱和脂肪酸(PUFA)饵料时,其生长增重率和存活率比摄食富含PUFA配合饵料或玉筋鱼鱼糜的明显下降。幼鱼摄食含有不同脂肪酸饵料时,其肌肉和肝脏组极性或非极性脂中脂肪酸组成都有变化。肌肉和肝脏极性脂中的PUFA比非极性脂中PUFA含量高,并且当饵料中不能提供足够的PUFA时,非极性脂中的PUFA下降明显,而极性脂中的PUFA变化较小,被优先贮存。真鲷幼鱼肌肉和肝脏组织缺乏n-9、n-6和n-3相互转化的能力。因此,为提高幼鱼的生长增重率和存活率,必须保证饵料中含有足够的PUFA。  相似文献

In this study, the semen production and quality, hepato‐somatic index, haemato‐immunologic values, oxidative stress and the fatty acid contents in liver, muscle and semen of rainbow trout fed diets supplemented n‐3 series long‐chain polyunsaturated fatty acids (LC‐PUFAs) under regular stripping condition were investigated. For this aim, three diets (Control, D1 and D2) were prepared. These diets were contained n‐3 LC‐PUFAs (as a percentage of dietary total fatty acid) at 3.14%, 7.84% and 13.63% respectively. Experimental fish were fed with the control and test diets. The highest hepato‐somatic index, spermatologic (semen pH and volume, sperm motility and density), haematologic (haematocrit value, haemoglobin, erythrocyte count, corpuscular volume, haemoglobin and its concentration in corpuscular), immunologic (nitroblue tetrazolium activity, leucocyte count, phagocytic index, protein and immunoglobulin [IgM] in total plasma) and antioxidants (reduced glutathione, catalase, glutathione peroxidase and superoxide dismutase) values were found in fish fed the D2, D1 and control diets respectively (p < 0.01). Increase in the dietary n‐3 LC‐PUFAs was not significantly (p > 0.01) increased the oxidative stress (malondialdehyde) in fish. The results indicated that the n‐3 LC‐PUFAs at 13.63% level of total fatty acid in the diet could increase the semen production and quality, hepato‐somatic index, haematologic and immunologic values, and the n‐3 LC‐PUFA contents in liver, muscle and semen of rainbow trout broodstock under regular stripping condition.  相似文献

ABSTRACT:   Changes in lipid content and fatty acid composition of muscle, liver and ovary of captive-reared and wild silver Japanese eel Anguilla japonica were examined during artificial maturation induced by salmon pituitary homogenate (SPH) injections. Although the relative levels of n-3 and n-6 highly unsaturated fatty acids (HUFA) in liver and ovary were higher than in muscle in both captive and wild silver eels before SPH injection, these tended to decrease with maturation. The relative levels of n-6 HUFA in muscle, liver, ovary and eggs of wild silver eels were remarkably higher than those in captive eels. Therefore, we attempted to alter the ratio of n-6 HUFA in eggs by feeding eels a diet supplemented with linoleic acid-rich plant oil. Although the percentage of n-6 polyunsaturated fatty acids to total fatty acids in eggs of eels fed the supplemented diet was similar to wild silver eels, the percentage of n-6 HUFA remained remarkably lower than in wild silver eels. Hence, it appears that the supplemented diet affected the fatty acid composition of eggs, but did not result in much conversion of linoleic acid to its higher homologs in eels.  相似文献

Arctic charr (Salvelinus alpinus L.) were fed either a commercial diet or six experimental test diets containing coconut oil and different polyunsaturated fatty acids (PUFA) at a level of 1% by dry weight. Best growth rates were observed with the commercial diet, worst with diet containing coconut oil with no PUFA. An increase in hepatic lipid, hepatic sterol esters and muscular moisture content, and a decrease in muscular lipid was generally found in fish fed the test diets compared to those maintained on the commercial diet.Phosphatidylcholine was the dominant polar lipid (PL) class in all tissues examined. Extensive modification of dietary saturated fatty acids into 18:1 (n-9) was observed in tissue triacylglycerols (TAG) of fish fed test diets. No changes occurred with the commercial diet.Dietary PUFA were essentially incorporated unchanged into tissue TAG of all fish in the present study. PUFA composition of hepatic phospholipids was significantly influenced by that contained in the diets. However both 18:2 (n-6) and 18:3 (n-3) in the test diets were extensively elongated and desaturated prior to incorporation into PL. The (n-9) PUFA content was always higher in liver of fish fed the test diets. When 18:2 (n-6) and 18:3 (n-3) were supplied together, the level of (n-3) PUFA exceeded those of (n-6) PUFA. Muscle PL were less influenced by diet than liver. In muscle (n-3) PUFA were always the predominant PUFA irrespective of diet. Only low amounts of (n-9) PUFA were found. It is suggested that (n-3) PUFA are the prime essential fatty acids for Arctic charr, and that they are used in preference to (n-6) PUFA for elongation, desaturation and incorporation into PL. The results suggest that the quantitative requirement of Arctic charr for EFA is may be higher than that of other salmonids.  相似文献

Tilapia (Oreochromis) nilotica were fed either a commercial diet containing 2.2% (n-3) and 0.5% (n-6) polyunsaturated fatty acids (PUFA), or a diet containing 1.0% methyl linoleate as the only PUFA. The fatty acid composition of tissue lipids generally reflected that of the diet. Fish from both dietary groups were injected intraperitoneally with 14C-labelled linoleic acid, 18:2 (n-6), or linolenic acid, 18:3 (n-3), and the distribution of radioactivity in tissue lipids examined. The conversion of both 18:2 (n-6) and 18:3 (n-3) to longer chain PUFA was lower in fish fed the commercial diet than in those fed the diet containing only 18:2 (n-6). Half of the radioactivity from both substrates recovered in liver polar lipids was present in C20 and C22 PUFA with fish maintained on the experimental diet. It is concluded that T. nilotica is capable of elongating and desaturating both 18:2 (n-6) and 18:3 (n-3), but that this conversion is suppressed by dietary longer chain PUFA. Sea urchin eggs and larvae have been suggested as potential live prey for marine fish larval feeding. This study evaluated the fatty acid composition of Paracentrotus lividus eggs, prisms and four-armed plutei, obtained from wild and captive broodstocks fed on raw diets: maize, seaweed and a combination of maize and seaweed. Amounts of essential fatty acids (EFA) for marine fish larvae [arachidonic acid (ARA), eicosapentaenoic acid (EPA) and docosahexanoic acid (DHA)] were determined in eggs and endotrophic larvae. ARA ranged from 3.93% in eggs from combination to 18.7% in plutei from maize diets. In any developmental stage, EPA amounts were always lower than 5% for the raw diets, and DHA showed null or trace amounts including the wild diet. Thus, broodstock-prepared diets had to be formulated based on different lipid sources (Algamac, linseed oil, cod liver oil and olive oil) in order to test eggs and larvae EFA enhancement. EFA improvement was possible for all tested prepared diets. Algamac diet lead to superior EFA enhancement mainly in DHA (7.24%, 4.92% and 6.09% for eggs, prisms and plutei, respectively) followed by cod liver oil diet. Only these two lipid sources should be considered for prepared broodstock diets in order to obtain suitable live prey for fish larval feeding.  相似文献

研究了黄颡鱼受精卵孵化期间和仔鱼发育阶段脂肪含量和脂肪酸的组成变化规律.采用常规化学分析方法和气相色谱法对黄颡鱼从鱼卵受精开始至仔鱼孵化后未投饵的7 d内的脂肪含量和脂肪酸组成进行测定.结果表明,受精卵在整个孵化期间脂肪含量有下降趋势.受精卵中不饱和脂肪酸含量大于饱和脂肪酸含量.受精卵在整个孵化期间各种脂肪酸含量无明显变化.仔鱼孵化后,鱼体总脂肪含量急剧下降,总脂含量从0日龄的4.57%降低到7日龄的0.75%.仔鱼在饥饿期间鱼体脂肪酸组成发生明显变化,单不饱和脂肪酸含量下降最为明显,尤其是C18:1.仔鱼在饥饿期间,脂肪酸按n-9>n-6>n-3顺序被先后利用,黄颡鱼仔鱼发育阶段主要以单不饱和脂肪酸作为能量代谢基质,而C20:4n6(AA)和C22:6n3(DHA)优先于C20:5n3(EPA)被保存下来.  相似文献

Atlantic salmon fry (4 g) were fed a semi-synthetic diet devoid of n-3 and n-6 polyunsaturated fatty acids (PUFA) for 4 months. External signs of nutritional imbalance during the period were slow growth rate and increased mortality. Some symptoms of essential fatty acid deficiency appeared in the fatty acid composition of the blood and liver during the fourth month. At that time, the percentages of n-3 and n-6 PUFA of triacylglycerols (TAG) were nearly exhausted, and the percentages of 22:6 n-3 and of 20:5 n-3 in phospholipids (PL) showed a marked decrease. This decrease in the PUFA level of the PL was paralleled by the appearance of 20:3 n-9, whereas in the TAG an increase in the percentage of 18:1 n-9 was observed. After 4 months the monounsaturated fatty acid 18:1 n-9 constituted nearly 50% of the fatty acids in the TAG fraction of the liver. The time course of the changes in fatty acid composition of liver and blood lipids was quite similar, whereas the carcass lipid composition appeared to respond slowly to a diet devoid of essential fatty acids.  相似文献
