The objective of this study was to evaluate oxidative stress in lambs experimentally infected with Haemonchus contortus and supplemented with selenium and vitamin E. Twenty male Corriedale lambs were divided into four experimental groups with five animals each: G1 consisted of animals infected and supplemented with 0.2 mg/kg of live weight (LW) sodium selenite by intramuscular injection (IM); G2 consisted of animals infected with larvae and supplemented with 0.2 mg/kg LW sodium selenite IM and 2000 IU per animal of Vitamin E IM; G3 consisted of animals infected with larvae and supplemented with 2000 IU per animal of Vitamin E IM; and G4 consisted of animals infected with larvae. The animals were infected orally with 500 H. contortus larvae (L3) every 48 hours for 20 days. For biochemical analyses and eggs per grams of feces (EPG) evaluation, blood and feces were both collected at zero (T0), 20 (T1), 40 (T2) and 60 (T3) days. The weight of the animals was also measured at these times. Lower TBARS values were observed in the supplemented animals compared to the control group. The groups supplemented with Selenium exhibited blood GSH-Px activity higher than that of non-supplemented animals. Supplementation with selenium provided greater antioxidant protection against oxidative stress generated from experimental infection of lambs with H. contortus.  相似文献

Effect of selenium on sheep lymphocyte responses to mitogens   总被引:1,自引:0,他引:1  
The effect of selenium (Se) on sheep lymphocyte response to mitogens was studied. In an indoor experiment lambs were fed a basal diet containing 0.13 mg Se kg-1, and supplemented with, respectively, 0.1 or 0.5 mg Se kg-1, either as sodium selenite or as selenomethionine. Enhancement of the proliferative response of lymphocytes to phytohaemagglutinin (PHA), pokeweed mitogen (PWM) and concanavalin A was found in lambs following selenium supplementation at the lower levels. The highest dietary selenium content, however, induced decreased mitogen response. Transient increases in lymphocyte response to PHA and PWM by ewes supplemented with selenium was demonstrated in one field study and a combined effect of selenium and vitamin E was seen in another. There was no stimulatory effect on the mitogen response of lymphocytes from sheep supplemented with dietary vitamin E alone.  相似文献

This study was conducted to exploit ameliorative effect of additional vitamin E and/or zinc supplementation on immune response of peripartum Sahiwal cows. Thirty‐two pregnant dry Sahiwal cows were blocked into four treatment groups (n = 8), namely control, zinc (Zn), vitamin E (Vit E) and zinc + vitamin E (Zn + Vit E). Feeding regimen was same in all the groups except that the Sahiwal cows in the zinc‐, vitamin E‐ and zinc + vitamin E‐fed groups were additionally supplemented with 60 mg Zn/kg DM, 1000 IU vitamin E and 60 mg/kg + 1000 IU Zn + vitamin E, respectively, from day 60 pre‐partum to day 90 post‐partum. Blood samples were collected on days ?60, ?45, ?30, ?15, ?7, ?3, 0, 3, 7, 15, 30, 45, 60, 90 and 120 with respect to day of parturition and analysed for total immunoglobulin (TIG), immunoglobulin G (IgG), interleukin‐2 (IL‐2), vitamin E (Vit E) and zinc (Zn) status. Before calving, cows showed a decrease in plasma TIG, IgG, IL‐2, Vit E and Zn levels. However, increased levels of plasma TIG, IgG, IL‐2, Vit E and Zn were observed after calving. After calving, Sahiwal cows supplemented with Zn + Vit E had higher plasma TIG, IgG and IL‐2 in comparison with cows of control and Zn + Vit E‐fed groups. In the present study, plasma vitamin E level was higher in Vit E‐fed and Zn + Vit E‐fed cows; however, zinc level was higher in Zn‐ and Zn + Vit E‐supplemented cows. In conclusion, a reduced immune response during peripartum period in Sahiwal cows was ameliorated by dietary vitamin E and zinc supplementation.  相似文献

The LD50-values of sodium selenite after i/m injection to rabbits was determined by means of probit-regression straight line. Simultaneously a combination of vitamin E and sodium selenite was tested for toxicity in order to examine the effect of vitamin E on the selenium toxicity. The LD50/24h was 2.53 mg sodium selenite/kg body mass and in combination with vitamin E (30 IU vitamin E/5 mg NaSeO3) 2.73 mg/kg. Furthermore the selenium concentration was determined in blood, liver, kidney and muscles of animals which died or were killed after 24 h respectively. Not only chemical analysis but also relations of time-dose-efficiency and significantly increased LD50-values show a certain protective effect of vitamin E on selenium intoxication.  相似文献

Three experiments were carried out, using sheep fed a marginally low selenium diet, to study the effect of selenium supplementation on the antibody response to tetanus toxoid and on the serum IgG concentration. Six groups of three six-month-old lambs were fed a basal diet containing 0.13 mg Se kg-1 supplemented with either 0.1, 0.5 or 1.0 mg Se kg-1, as sodium selenite or as selenomethionine. These animals generally showed enhanced antibody response to tetanus toxoid, parainfluenza-3 virus and Corynebacterium pseudotuberculosis, and their total serum IgG concentrations were higher than in unsupplemented control animals although few responses were statistically significant. In two field studies significantly higher titres to tetanus toxoid were detected in ewes injected with 100 mg selenium as barium selenate, although no influence on serum IgG concentrations was detected. Lambs from selenium supplemented ewes had significantly higher titres to tetanus toxoid than lambs from ewes in the control group. Dietary vitamin E supplementation had a similar effect on the antibody response to tetanus toxoid in ewes, though no additive effect was seen when vitamin E was given together with selenium.  相似文献

为探索不同剂量的Se和VE对蛋鸡生产性能及蛋品质的影响,我们选用32周龄罗曼蛋鸡72只,随机分成6组。在基础日粮含Se0.1mg/kg基础上,添加Na2SeO3-VE粉,各组Se的添加水平分别是每公斤日粮0、0.5、1.0、1.5、2.0、3.0mg,VE添加水平分别是0、41.66、83.32、124.98、166.64、249.96IU。实验期8周。结果表明,添加1mg/kgSe、83.32IUVE组鸡产蛋率显著高于其他各组;各组鸡蛋重随Se水平的提高有下降趋势,但差异不显著;蛋品质各组差异不显著。  相似文献

1. The influence of different forms of dietary selenium (Se) on vitamin E (alpha-tocopherol) and Se contents of egg yolk and chicken meat was investigated. 2. Eggs were collected from laying hens subjected to 4 different dietary treatments after 24 weeks of age. Treatments compared the effects of inorganic Se supplementation (selenite) to those of organic (Se-enriched yeast, Se-enriched alga Chlorella) supplements. In a second experiment the effect of the above organic dietary Se supplementation on the alpha-tocopherol contents of meat from broiler chickens was evaluated. 3. Dietary Se supplementation increased the alpha-tocopherol content of egg yolks from 297 mg/kg dry matter in treatment without supplementation to 311 mg/kg when selenium was supplemented as selenite, and to 370-375 mg/kg when organic supplements were used. The Se and alpha-tocopherol contents of breast and thigh meat in broilers were significantly increased by organic dietary Se supplementation. 4. The inclusion of organic dietary Se sources in the diets of laying hens and broilers would enhance the nutritional value (vitamin E and Se contents) of products (eggs and meat) for human consumption.  相似文献

This study was planned to investigate the comparative effect of vitamins C (L‐ascorbic acid), E (DL‐α‐tocopherol acetate), probiotics, lower than normal protein level (14%) and combination of these treatments on immune response of male broiler breeders after zinc‐induced moulting. One hundred and eighty birds at the age of 65 weeks were induced to moult by mixing zinc oxide (ZnO) in feed at the rate 3000 IU/kg of feed. Upon completion of moulting, birds were divided into six groups (five replicates per group) in a completely randomized design and were fed vitamin C (500 IU/kg), vitamin E (100 IU/kg), lower protein level, probiotics (50 mg/l), and a combination of these components, while one group was kept as control. After completion of moulting phase (5 weeks), the treatment effects were tested as in vitro macrophages engulfment percentage, nitric oxide (NO) production, serum antibody titres against Newcastle disease (ND) and infectious bronchitis (IB). The results showed that in vitro macrophage engulfment percentage in unopsonized conditions was significantly higher in vitamin E‐supplemented group. In addition, in opsonized condition, the macrophage engulfment percentage was significantly higher in both vitamin E‐ and C‐supplemented groups. The NO (opsonized and unopsonized) production and antibody titre against ND and IB were significantly higher in vitamin E‐supplemented group. It was concluded that vitamin E is a better option for enhanced immune response in broiler breeders after zinc‐induced moulting.  相似文献

在3×5的两因子试验中,研究了不同添加水平维生素E(4、14和24mg/kg)与有机和无机硒(亚硒酸钠0.20、0.35和0.50mg/kg,酵母硒0.35和0.50mg/kg)对褐壳蛋种母鸡在20~40周龄阶段生产繁殖性能和有关血液指标的影响。结果表明,产蛋母鸡的采食量、产蛋率、蛋重、产蛋量、饲料转化率和死淘率在3个维生素E添加水平间无差异;除了添加酵母硒0.50mg/kg组蛋鸡的蛋重和产蛋量显著低于酵母硒0.35mg/kg组以外,其它各项生产性能指标在各硒处理组间差异不明显。维生素E和硒可增加彼此在血液中的存留;当日粮中只补加维生素E4mg/kg时,将日粮亚硒酸钠硒添加水平由0.20mg/kg提高到0.35~0.50mg/kg可使血浆α-生育酚含量由0.50μg/ml上升到接近1.0μg/ml左右。  相似文献

Excess consumption of selenium (Se) accumulator plants can result in selenium intoxication. The objective of the study reported here was to compare the acute toxicosis caused by organic selenium (selenomethionine) found in plants with that caused by the supplemental, inorganic form of selenium (sodium selenite). Lambs were orally administered a single dose of selenium as either sodium selenite or selenomethionine and were monitored for 7 days, after which they were euthanized and necropsied. Twelve randomly assigned treatment groups consisted of animals given 0, 1, 2, 3, or 4 mg of Se/kg of body weight as sodium selenite, or 0, 1, 2, 3, 4, 6, or 8 mg of Se/kg as selenomethionine. Sodium selenite at dosages of 2, 3, and 4 mg/kg, as well as selenomethionine at dosages of 4, 6, and 8 mg/kg resulted in tachypnea and/or respiratory distress following minimal exercise. Severity and time to recovery varied, and were dose dependent. Major histopathologic findings in animals of the high-dose groups included multifocal myocardial necrosis and pulmonary alveolar vasculitis with pulmonary edema and hemorrhage. Analysis of liver, kidney cortex, heart, blood, and serum revealed linear, dose-dependent increases in selenium concentration. However, tissue selenium concentration in selenomethionine-treated lambs were significantly greater than that in lambs treated with equivalent doses of sodium selenite. To estimate the oxidative effects of these selenium compounds in vivo, liver vitamin E concentration also was measured. Sodium selenite, but not selenomethionine administration resulted in decreased liver vitamin E concentration. Results of this study indicate that the chemical form of the ingested Se must be known to adequately interpret tissue, blood, and serum Se concentrations.  相似文献

Forty-eight weanling S.P.F. Yorshire pigs were used to study the influence of supplemental vitamin E (25 IU per kg of diet) selenium (0.5 ppm in diet) and methionine (0.1% in diet) on the incidence of hepatosis dietetica and mulberry heart disease when fed a torula yeast-corn diet. Vitamin E and/or selenium increased pig survival. Supplemental selenium resulted in increased liver selenium concentrations. No hepatosis dietetica was observed in any of the pigs. The addition of vitamin E and/or selenium at the levels used did not reduce the frequency of myocardial lesions; however, they prevented skeletal muscular dystrophy and exudative diathesis. The myocardial lesions were less severe in supplemented pigs compared with unsupplemented controls.  相似文献

OBJECTIVE: To evaluate the respiratory excretion and elimination kinetics of organic and inorganic selenium after oral administration in sheep. ANIMALS: 38 crossbred sheep. PROCEDURES: Selenium was administered PO to sheep as a single dose of 0, 1, 2, 3, or 4 mg/kg as sodium selenite or selenomethionine. Expired air was collected and analyzed from all sheep at 4, 8, and 16 hours after administration. RESULTS: Clinical signs consistent with selenium intoxication were seen in treatment groups given sodium selenite but not in treatment groups given the equivalent amount of selenium as selenomethionine. However, a distinct garlic-like odor was evident in the breath of all sheep receiving 2 to 4 mg of selenium/kg. The intensity of odor in the breath did not correlate with clinical signs in affected animals receiving sodium selenite treatment. CONCLUSIONS AND CLINICAL RELEVANCE: The concentration of selenium in expired air was greater in sheep receiving selenium as selenomethionine than sodium selenite. The concentration of selenium in expired air from sheep receiving high doses of selenium (3 and 4 mg of selenium/kg) was larger and selenium was expired for a longer duration than the concentration of selenium in expired air from sheep receiving low doses of selenium (1 and 2 mg of selenium/kg).  相似文献

Seventy-two Hereford X Simmental cows, averaging 498 kg in body weight and 5.2 yr of age, were used in a 2-yr study to ascertain if selenium (Se)-vitamin E (E) injections and winter protein supplementation would affect growth, reproduction and health of beef cattle maintained year-round on feedstuffs marginally deficient in Se (.03 to .05 mg/kg). Cows received either no injection or a mixture of 30 mg Se (as sodium selenite) and 408 IU E injected subcutaneously beginning 3 to 4 mo prepartum and at 60-d intervals throughout the 2-yr period. Calves born to Se-E treated cows were injected with 5.5 mg Se and 75 IU E/100 kg body weight at 60-d intervals beginning at 1 mo of age. Calves were born between December 30 and February 20 and cows were bred between March 20 and May 20. Cattle grazed pasture (.05 mg Se/kg) that consisted of orchardgrass, bluegrass and white clover during the fall, spring and summer. During winter (December 15 to May 2), cattle were fed corn silage (.03 mg Se/kg) supplemented with either: no protein supplement (control), soybean meal or a urea-corn mixture. Cows and calves receiving Se-E had higher (P less than .01) whole blood glutathione peroxidase (GSH-Px) activity and plasma Se concentrations than controls. Selenium-E injections reduced (P less than .05) calf death losses from 15.3% to 4.2% and slightly increased (P less than .10) adjusted calf weaning weights. Background

Increasing the polyunsaturated fatty acid (PUFA) content and decreasing the saturated fatty acid (SFA) content of mutton can help to improve its nutritional value for consumers. Several laboratories have evaluated the effects of vitamin E on the fatty acid (FA) composition of muscle in sheep. However, little information is available on wool sheep, even though wool sheep breeds are an important source of mutton, especially in northern China where sheep are extensively farmed. The present study was designed to address the effects of vitamin E on muscle FA composition in male Aohan fine-wool sheep.

Methods

Forty-two male Aohan fine-wool lambs (5 mo old) with similar initial body weight were randomly divided into seven groups and fed diets supplemented with 0 (control group), 20, 100, 200, 1,000, 2,000, or 2,400 IU/sheep/d vitamin E for 12 mo. Three lambs from each group were slaughtered to measure vitamin E and FA content in the longissimus lumborum (LL) and gluteus medius (GM) muscles.

Results

Vitamin E concentrations in the LL and GM increased significantly after 12 mo of vitamin E supplementation (P < 0.05). However, this increase did not occur in a dose-dependent manner because the muscle vitamin E concentration was highest in the 200 IU/sheep/d group. Dietary vitamin E supplementation also caused a significant reduction in SFA content and an increase in monounsaturated FA (MUFA) content in the LL and GM (P < 0.05). All six doses of vitamin E significantly increased cis9 trans11-conjugated linoleic acid (c9t11-CLA) content in the LL compared with the control group (P < 0.05).

Conclusions

Dietary supplementation with vitamin E increased muscle vitamin E content and improved the nutritional value of mutton by decreasing SFA content and increasing MUFA and c9t11-CLA contents in Aohan fine-wool sheep. These effects were greatest in sheep fed a diet containing 200 IU/sheep/d vitamin E.  相似文献

Lymphocytes of lambs on a low selenium/vitamin E diet were isolated from peripheral blood, and mitogenic responses to phytohaemagglutinin (PHA) tested in the presence of different doses of sodium selenite and emulsified vitamin E added in vitro. An enhancing effect of selenium was observed at doses of 1 ng/ml or less, and reached a plateau at about 10 ng/ml. Toxic effects were evident beyond 1 micrograms/ml. The stimulatory potential of selenium among lambs was inversely related to their ability to respond to PHA in control cultures but was not related to the blood glutathione peroxidase activity of the animals concerned. Optimal doses of vitamin E added to culture (0.15-1.5 micrograms/ml) elevated responses beyond those seen with selenium, but synergistic effects were not apparent. Similar results were obtained when lymphocytes from deficient, myopathic lambs were cultured with serum from lambs supplemented in vivo, and when PHA responses of untreated and treated lambs were compared. Tests with other phytolectins (concanavalin A and pokeweed mitogen) suggested that the two micronutrients exert a differential influence on lymphocyte sub-populations. It was also concluded that the poor lymphocyte responses seen in myopathic lambs can be readily and rapidly reversed by injection of these nutrients, and that prophylaxis is most effective during the first 6 weeks of life.  相似文献

Thirty-five crossbred wethers were used to determine the concentrations of alpha-tocopherol in serum and tissues after oral supplementation of six different vitamin E product forms. Five wethers were assigned to each of the following treatments: 1) control, no supplemental vitamin E (C), 2) emulsifiable DL-alpha-tocopheryl acetate-dry (Rovimix E-50% SD), 3) nonemulsifiable DL-alpha-tocopheryl acetate-dry (Rovimix E-50% Ads), 4) emulsifiable DL-alpha-tocopheryl acetate-liquid (Rovimix E-40% Dispersible Liquid Concentrate [DLC]); 5) emulsifiable DL-alpha-tocopherol-liquid (Hoffmann-La Roche, E-40% DLC alcohol), 6) micellized DL-alpha-tocopheryl acetate-liquid (Bioglan, Inc., E-20%); and 7) micellized DL-alpha-tocopherol-liquid (Bioglan, Inc., E-20%). Animals were supplemented daily with 1,000 IU of their respective vitamin E sources for 56 d. Blood samples were collected daily from d 0 to 7 and weekly until d 56. Animals were subsequently killed by exsanguination after stunning and eight different tissues were collected for alpha-tocopherol analysis. There were effects of day, treatment, and day x treatment interaction on serum alpha-tocopherol. All supplemented groups were higher in serum alpha-tocopherol concentration than were the C wethers. The emulsifiable vitamin E alcohol liquid product form (Treatment 5) yielded higher (P less than .01) serum alpha-tocopherol concentration than the emulsifiable acetate liquid product (Treatment 4). Sheep on Treatment 5 reached maximum concentration on d 1, sheep on Treatment 6 on d 2, and the sheep on the remaining Treatments by d 3. This study was conducted to investigate the effect of vitamin A and E supplementation on the antioxidant defences of broiler chickens against carbon tetrachloride (CCl(4))-induced oxidative stress at 4 weeks of age. Superoxide dismutase (SOD) and glutathione peroxidase (GPX) activities as well as total antioxidant (TAO) level were analysed before and after CCl(4) challenge. Day-old Lohman broiler chickens (n = 144) were randomly assigned to six factorially arranged dietary treatments consisting of vitamin A [1.35 (control) or +20 mg/kg] and vitamin E [20 (control), +40 or +60 mg/kg]. The background of vitamins A and E in the basal diet was 4500 IU (1.35 mg) and 30 IU (20 mg) respectively. At 4 weeks of age, eight chickens from each treatment were bled before interperitoneal injection with 1 ml of CCl(4) (mixed with olive oil in a ratio of 1:1) and bled again 24 h post-injection. Vitamin E supplementation decreased (p < 0.05) the activity of both SOD and GPX and showed a tendency (p = 0.07) for TAO reduction. CCl(4) attenuated SOD and GPX activities as well as TAO level. The decrease was profound (p < 0.05) in chickens fed the basal diet as well as those fed basal diet supplemented with 20 mg vitamin A. TAO levels behaved similarly when chickens were challenged with CCl(4). After CCl(4) injection, SOD activities of all experimental groups were equivalent. The presence of vitamin A decreased (p < 0.05) plasma GPX activity in chickens fed the basal diet supplemented with 40 mg/kg of vitamin E. Results of this experiment suggested that vitamin E supplementation elevated antioxidant enzyme activities while vitamin A supplementation attenuated this effect. Vitamin E supplementation improved the total reducing power by maintaining comparable levels of TAO upon CCl(4) challenge. Further experiments need to be carried out to investigate the role of vitamin A in oxidative stress and to evaluate the lipid peroxidation products.  相似文献

This study assessed the effect of a vitamin E supplement given to pregnant mares on immunoglobulins (Ig) levels in foals. In addition, the fatty acid (FA) content and composition of the mares’ milk was assessed. Milk α‐tocopherol concentrations were compared between pregnant Danish Warmblood mares (n = 17) given a daily oral supplement of 2500 international units (IU) RRR‐α‐tocopherol in the last 4 weeks of pregnancy and a group of unsupplemented mares (n = 17) receiving 170–320 IU vitamin E daily originating from the feed. Milk α‐tocopherol was higher in supplemented mares (36.7, 12.4 and 9.8 μmol/l respectively) in relation to control mares (13.1, 6.4 and 5.8 μmol/l on days 1, 2 and 3 respectively; p < 0.001). Milk IgG was higher on days 2 and 3 post‐partum (PP) in supplemented mares (1.03 and 0.73 mg/ml respectively) in relation to control mares (0.79 and 0.56 mg/ml respectively; p < 0.05). Milk IgM was higher on days 2 and 3 post‐partum (PP) in supplemented mares (0.19 and 0.17 mg/ml) in relation to control mares (0.13 and 0.11 mg/ml respectively; p < 0.05). Plasma α‐tocopherol in foals was higher from supplemented mares on days 1, 2 and 3 (5.7, 14.8 and 19.2 μmol/l respectively) in relation to foals from control mares (3.6, 6.1 and 7.6 respectively; p < 0.001). Foal plasma IgM was higher from supplemented mares on day 3 (0.50 mg/ml) in relation to foals from control mares (0.32 mg/ml; p < 0.001). The total FA content in milk was highest on day 1 (21.6 g FA/kg milk) in relation to days 2 and 3 (13.6 and 13.5 g FA/kg milk respectively; p < 0.001). In conclusion, a daily oral supplement of 2500 IU RRR‐α‐tocopherol increased α‐tocopherol content in mare milk and foal plasma, IgG and IgM in mare milk and IgM in foal plasma.  相似文献

Selenium and vitamin E concentrations were measured in the blood sera of 287 cows in 91 herds. Herds with chronic mastitis and muscle diseases had significantly lower selenium concentrations (10.4 +/- 5.26 and 11.7 +/- 8.63 micrograms/litre serum, respectively) than healthy control herds (17.7 +/- 15.00 micrograms/litre). The selenium concentrations in herds with low fertility did not differ from control herds. The vitamin E concentrations were similar in all the herds. Supplementation of the mineral mixture with 20 mg sodium selenite and 2 g vitamin E/kg on 15 farms with a deficient selenium supply increased the selenium concentrations in the blood within two months in 12 of the herds. The vitamin E concentrations were hardly influenced by the supplementation.  相似文献
