Biochemical changes associated with flowering in Bambusa arundinacea Linn and Bambusa nutans Wall. ex Munro were analyzed. Gregarious flowering was initiated in natural areas and plantations of B. arundinacea in late 2014 and reached full bloom in early 2015, whereas sporadic flowering recorded during 2014–2015 in vegetatively propagated plants of B. nutans. Leaf and nodal shoot samples from flowering culms/shoots and nonflowering culms/shoots were collected in February 2015 at the initiation of fruit filling, then analyzed and compared for soluble sugars, phenols and peroxidase activity. In both species sugars were higher in leaves of flowering culms/shoots and lower in nodal shoots of flowering culms/shoots compared to the nonflowering. Phenols were lower in leaves and nodal shoots of flowering B. arundinacea, but higher in leaves and decreases in nodal shoots of B. nutans. Peroxidase activity increases in leaves and nodal shoots of B. arundinacea after flowering but increases in nodal shoots and decreases in leaves of B. nutans.  相似文献

Taxus chinensis and T. wallichiana in have been threatened in their distribution areas in recent decades because of their over-exploitation and reduction and destruction of native habitats. Determining the genetic diversity in populations of the two species will provide guidelines for their protection and preservation. Two hundred and fifteen trees from six populations of T. chinensis and 150 sampled trees of T. wallichiana were sampled. Six microsatellite primer pairs selected from 16 primer pairs were used to investigate genetic variation at the population and species levels. Five yielded polymorphic alleles, and among the 13 putative alleles amplified, 11 were polymorphic (accounting for 76.33 %).Shannon’s information index (I) and percentage of polymorphic bands (PPB) (I = 0.202 and PPB = 67.22 % for T. chinensis; I = 0.217 and PPB = 65.03 % for T. wallichiana). Both species had low levels of genetic diversity (mean H o = 0.107, H e = 0.121 for T. chinensis; H o = 0.095, H e = 0.109 for T. wallichiana). Genetic differentiation among populations was higher (F ST = 0.189) for T. chinensis and lower (0.156) for T. wallichiana, indicating limited gene flow (Nm) among populations for T. chinensis (0.68) and T. wallichiana (0.65). Variation among individuals of T. chinensis was 63.59 and 73.12 % for T. wallichiana. Thus, the threatened status of the two conifers is related to a lack of genetic diversity. All populations are isolated in small forest remnants. An ex situ conservation site should be established with a new population for these species that comprises all the genetic groups for the best chance to improve their fitness under environmental stresses.  相似文献

Nontimber forest products are a source of income for women in rural African communities. However, these products are frequently damaged by insect pests. The present study investigates the diversity and damage rates of insect pests that attack Carapa procera seeds and Lophira lanceolata fruits. The experiment was set up in western Burkina Faso and, for C. carapa, consisted of pests collected from seeds that had fallen to the ground and from stockpiled seeds. For L. lanceolata, pests were collected from fruits on the trees, and on the ground. The collected samples were sent to the laboratory to estimate the proportion of damaged seeds/fruits and rear the insects. The results showed that Ephestia spp., Tribolium castaneum, Oryzeaphilus spp., and Tenebroides mauritanicus were the pests of Carapa procera seeds and Lophira lanceolata fruits. Ephestia spp. was recorded as the main pest of both C. procera and L. lanceolata, whereas T. castaneum was only detected from seeds of L. lanceolata. For C. procera, the stocks were the most infested (29 %) by Ephestia spp. The infestation rate of fruits of L. lanceolata by Ephestia spp. on trees (31.42 ± 3.75 %) was less than the rate of fruits by T. castaneum on the ground (44.00 ± 3.5 %). The different body sizes of Ephestia spp. may indicate the occurrence of two putative species, one from C. procera and another one from L. lanceolata. This work provides important information that could contribute to the setting up of a local-scale sustainable management framework for oil tree pests in Burkina Faso and surrounding countries.  相似文献

The large white butterfly, Pieris brassicae (L.), is an important pest of Indian mustard, Brassica juncea (L.) Czern., and inflicts heavy damage to all the above ground plant parts with strong yield reducing impacts. Farmers have few practical options other than to spray insecticides to protect their crop. In this study, an attempt was made during 2012–2013 and 2013–2014 crop seasons at Ludhiana, India, to evaluate Ethiopian mustard, Brassica carinata A. Braun as a trap crop to manage this pest as an alternate pest management strategy. B. carinata borders surrounding B. juncea were compared to chemically protected B. juncea and control (without B. carinata borders) plots for their effects on P. brassicae infestation and grain yield. Oviposition preference of adult butterflies was studied in a two-choice test in field cages, while the effect of two host plants on larval performance was studied under laboratory conditions under no choice conditions. B. juncea plots bordered with B. carinata harbored significantly lower larval population compared to control B. juncea plots during both the seasons. The grain yield in the bordered plots was also significantly higher than that from control plots and was statistically non-significant from that obtained in chemically protected plots. Female butterflies showed distinct oviposition preference for B. carinata over B. juncea and the larvae reared on B. carinata completed development in shorter period and grew bigger and heavier than those reared on B. juncea. Results indicated that B. carinata has potential to be used as trap crop to manage P. brassicae.  相似文献

Key message

This paper presents a greenhouse study for assessing the genetic variation in maritime pine (Pinus pinaster Aiton) in response to pinewood nematode (Bursaphelenchus xylophilus (Steiner et Buhren) Nickle), which is a causal agent of pine wilt disease. Fifteen out of 96 half-sib families were selected as less susceptible. This experiment is an important first step for creating a resistance breeding program.

Context

Pine wilt disease is caused by the pinewood nematode (Bursaphelenchus xylophilus (Steiner et Buhren) Nickle), a quarantine pest, and is a concern to maritime pine (Pinus pinaster) in Portugal due to its economic, environmental, and social impacts. This disease is regarded as a major threat to European forests.

Aims

This paper aimed to evaluate the genetic variation in maritime pine families that were inoculated with pinewood nematode, identify the most resistant families, and establish the guidelines for a resistance improvement program.

Methods

Two-year-old half-sib progenies obtained from 96 plus trees were inoculated. The plants were monitored for survival on four different dates. The statistical analysis followed the mixed model theory.

Results

Genetic variability of the susceptibility to pine wilt disease was observed. At 157 days after inoculation, the 15 highest genetic ranking families out of 96 were selected, having a predicted survival mean of 15.6% instead of 11.0% on average for the all 96 families.

Conclusion

This study allows for the implementation of an improvement program to help control pine wilt disease.

Lecanicillium lecanii has been developed as biopesticides and widely used in the biological control of several insects in agricultural practice. However, the poor efficacy has blocked its application. The genetic manipulation has been proved as a useful tool to improve the virulence of entomopathogenic fungus. In this study, a scorpion toxin gene (BmKit) from Buthus martensi was cloned and transferred into L. lecanii and then its resulting activity against cotton aphids (Aphis gossypii) was assessed. The results showed that the engineered strain BmKit-12 grew significantly quicker than WT in host insects. The median lethal concentration (LC50) for BmKit-12 was 7.1-fold lower than that for WT, and the median survival time (LT50) for BmKit-12 was reduced by 26.5 % compared with that for WT. Although the conidial yield was reduced by 31.4 % on insect cadavers, the mycelia growth, sporulation, and conidia germination of BmKit-12 on plates were not significantly different from WT. Considering the lower amount of mycelia for BmKit-12 in host insects in late growth stage (fungal sporulation stage), we think the expression of BmKit may not affect the capability of sporulation. In conclusion, the expression of BmKit gene can significantly enhance the pathogenicity of L. lecanii against cotton aphids in a relatively cost-effective way.  相似文献

The dynamics of mixed stands are more complex and less studied than those of monospecific stands. The objective of this work was to analyze the variables involved in seedling occurrence and seedling survival in mixed stands of Pinus pinaster and P. pinea in Mediterranean areas. From 2011 to 2016, regeneration of both species was monitored at two sites located in Central Spain. We installed 72 regeneration plots where seedling dynamics were monitored. All the trees in the study areas were measured and mapped. Additionally, we took hemispherical photographs in each regeneration plot. The average density of P. pinea seedlings over the study period was almost 20 times larger than that of P. pinaster. Our results indicate that the seedlings of both species grow under moderate light conditions. In addition, we found that the occurrence of seedlings of both species was related to the structure of the stand. P. pinea seedlings grew where the density and size of P. pinaster trees were low and where P. pinea trees provided moderately sheltered conditions, whereas the number of P. pinaster seedlings was related to under intermediate densities of P. pinaster trees. Furthermore, seedling survival was positively associated with age of the seedlings and negatively with the August average maximum temperature. The temporal continuity of mixed stands of P. pinea and P. pinaster in the study area is compromised by the observed lack of regeneration of P. pinaster.  相似文献

Geometric morphometric analyses (GMMs) of the leaf shape can distinguish two congeneric oak species Quercus dentata Thunberg and Quercus aliena Blume in sympatric areas.

Contexts

High genetic and morphological variation in different Quercus species hinder efforts to distinguish them. In China, Q. dentata and Q. aliena are generally sympatrically distributed in warm temperate forests, and share some leaf morphological characteristics.

Aims

The aim of this study was to use the morphometric methods to discriminate these sympatric Chinese oaks preliminarily identified from molecular markers.

Methods

Three hundred sixty-seven trees of seven sympatric Q. dentata and Q. aliena populations were genetically assigned to one of the two species or hybrids using Bayesian clustering analysis based on nSSR. This grouping served as a priori classification of the trees. Shapes of 1835 leaves from the 367 trees were analyzed in terms of 13 characters (landmarks) by GMMs. Correlations between environmental and leaf morphology parameters were studied using linear regression analyses.

Results

The two species were efficiently discriminated by the leaf morphology analyses (96.9 and 95.9% of sampled Q. aliena trees and Q. dentata trees were correctly identified), while putative hybrids between the two species were found to be morphologically intermediate. Moreover, we demonstrated that the leaf morphological variations of Q. aliena, Q. dentata, and their putative hybrids are correlated with environmental factors, possibly because the variation of leaf morphology is part of the response to different habitats and environmental disturbances.

Conclusion

GMMs were able to correctly classify individuals from the two species preliminary identified as Q. dentata or Q. aliena by nSSR. The high degree of classification accuracy provided by this approach may be exploited to discriminate other problematic species and highlight its utility in plant ecology and evolution studies.

Information on population genetic structure and crop genetic diversity is important for genetically improving crop species and conserving threatened species. The PAL gene sequence is part of a multigene family that can be utilized to design DNA marker systems for genetic diversity and population structure investigation. In the current study, genetic diversity and population structure of 100 accessions of wild Pistacia species were investigated with 78 PAL markers. A protocol for using PAL sequences as DNA markers was developed. A total of 313 PAL loci were recognized, showing 100% polymorphism for PAL markers. The PAL markers produced relatively more observed and effective alleles in Pistacia falcata and Pistacia atlantica, with a higher Shannon’s information index and expected heterozygosity in P. atlantica, Pistacia vera and Pistacia mutica. Pairwise assessment of Nei’s genetic distance and genetic identity between populations revealed a close association between geographically isolated populations of Pistacia khinjuk and Pistacia chinensis. The accessions of wild Pistacia species had more genetic relationship among studied groups of species. Analysis of molecular variance indicated 19% among-population variation and 81% within-population variation for the PAL gene based DNA marker. Population structure analysis based on PAL revealed four groups with high genetic admixture among populations. The results establish PAL markers as a functional DNA marker system and provide important genetic information about accessions from wild populations of Pistacia species.  相似文献

The naturally occurring Verticillium nonalfalfae shows promise for biocontrol of the highly invasive Tree of Heaven (Ailanthus altissima), but might also bear a risk for non-target tree species. In this study, we conducted inoculations on potted seedlings of A. altissima as well as on eight indigenous and two invasive tree species associated with Tree of Heaven in Austria. Although vascular discolourations developed in all inoculated tree species, V. nonalfalfae was reisolated from Ailanthus and eight of the ten non-target-species, whereas typical disease symptoms and mortality only occurred on A. altissima. Results confirmed high susceptibility (S) of A. altissima to V. nonalfalfae but indicated tolerance (T) of Acer campestre, Acer pseudoplatanus and Quercus robur, possible resistance (PR) of Fraxinus excelsior, Populus nigra, Tilia cordata, Ulmus laevis and Ulmus minor and resistance (R) of Fraxinus pennsylvanica and Robinia pseudoacacia to this potential biocontrol agent. Results from seedling inoculations were confirmed by cursory field observations in Ailanthus-inoculated forest stands, where admixed A. campestre, A. pseudoplatanus, F. excelsior, Populus alba, R. pseudoacacia and U. laevis canopy trees remained asymptomatic, while mortality was induced in Ailanthus.  相似文献

The fall armyworm, Spodoptera frugiperda (J. E. Smith) and the corn earworm, Helicoverpa zea (Boddie) are among the main pests of maize. Both species exhibit cannibalistic behavior and quite often share the same feeding guild in maize (maize ears), which can result in several interspecific and intraspecific interactions. Paired interaction scenarios of intraspecific and interspecific larvae were assessed in arenas in the presence and absence of food to characterize movements resulting from interactions of these insects. There was a difference in the frequency of behavioral movements in all the interactions, except for S. frugiperda in the presence of food. Head touching and recoiling were the predominant movements in most of the interaction scenarios. Spodoptera frugiperda exhibited a predominance of defensive movements when competing against H. zea in the same instars. Cannibalism and predation occurred frequently in interactions involving 6th instar of H. zea against opponents in 4th instar. Larvae of H. zea show a higher aggressive movement than S. frugiperda. The larvae of S. frugiperda take advantage during the interactions, although they present more defensive movements compared to H. zea. This study provides relevant information regarding the interaction of these species and intraguild interaction, which might influence the population dynamics and the competitive displacement of pest species that share the same ecological niche.  相似文献

Our previous studies have revealed that the ThCAP gene plays a vital role in transgenic Populus (P. davidiana × P. bolleana) in response to cold stress. However, the regulatory mechanism of ThCAP gene expression has been unclear. In this study, the 5′ flanking region of the ThCAP promoter (PThCAP) was cloned using a genome-walking method. By analyzing cis-acting regulatory elements of PThCAP, a DRE motif and MYC and MYB elements were found to be located in the promoter. To identify the regulatory elements that control the expression of the ThCAP gene promoter, a series of deletion derivatives of PThCAP, P1–P5, from the translation start code (?1538, ?1190, ?900, ?718 and ?375 bp), were fused to the GUS reporter gene, and then each deletion was stably introduced into Arabidopsis thaliana plants. Deletion analysis of the promoter suggested that only the P2 fragment had strong GUS expression in leaves and roots of A. thaliana exposed to low temperature stress. These results suggest that this 290-bp region (?1190 to ?900 bp), as an important part in PThCAP, was associated with cold tolerance of A. thaliana. Our results provide evidence for the regulatory mechanism of ThCAP gene involved in the response to cold stress, and that the gene is promising candidate gene for genetic improvement of crops.  相似文献

Canningia tomici sp. n. (Microsporidia, Unikaryonidae) infects the midgut epithelium, the gut muscules, Malpighian tubules, connective tissues, adipose tissues and the gonads of the pine shoot beetle, Tomicus piniperda (L.) (Coleoptera, Scolytidae). The infection is present in populations of Tomicus piniperda in Europe and in the United States. Uninucleate oval single spores occur in two sizes: 2.8?±?0.4?× 1.4?±?0.4?μm and 3.8?±?0.3?×?2.0?±?0.2?μm. The polar filament of this microsporidium is fixed subapically in a flat anchoring disc. The relationship between Xenorhabdus nematophilus and Photorhabdus luminescens, the bacterial symbionts of the entomopathogenic nematodes Steinernema carpocapsae and Heterorhabditis bacteriophora, against the diamondback moth, P. xylostella (L.), pupae showed the pathogenic capability of P. luminescens to be over that of X. nematophilus. They gave 60 and 40?% mortality with LC50 values of 5?×?104 and 5.5?×?105 cells/ml, respectively.The number of bacterial cells influences the rate of killing the pupae of P. xylostella and a direct correlation between mortality of the pupae, percentage of deformed adults and the spraying dose with significant differences were observed. An inverse correlation found between the no. of eggs laid/female, percentage of hatching, adult survival and the spraying dose. These bacterial effects have been attributed to different toxic bacterial enzymes which damage the haemocytes and inhibit activation of the humoral immune system of the insect.  相似文献

Pinus pinaster Ait. susceptibility to pinewood nematode significantly differed among provenances, and the two Atlantic provenances of the Iberian Peninsula being the most affected. However, significant provenance × environment interaction was found. Provenance susceptibility was related to basal diameter, number of branches and oleoresin flow, and some climatic parameters.

Context

The pinewood nematode Bursaphelenchus xylophilus, native to North America, is an important pest affecting pine forests throughout Eurasia. In Europe, it has been detected in Portugal and Spain and is primarily associated with Pinus pinaster, an important Mediterranean tree species.

Aims

We have investigated the differences in susceptibility among several P. pinaster provenances in the Iberian Peninsula and France, as well as their relationship to certain growth traits and physiological parameters.

Methods

Three independent inoculation tests were performed on 3 to 4-year-old trees, followed by assessment of growth traits and physiological variables, along with time course destructive sampling for nematode quantification.

Results

The results showed significant differences among provenances for almost all growth traits, wilting, and mortality, though a significant provenance × environment interaction was also detected. Two Atlantic provenances, Noroeste-Litoral and Leiria, displayed the largest susceptibility to pinewood nematode. Changes in susceptibility to B. xylophilus between experiments were influenced by temperature and seasonality. Autumn precipitation and mean maximum temperature during summer at the original provenance sites could be related to provenance susceptibility.

Conclusion

Noroeste-Litoral and Leiria were the most disease-affected provenances. This study emphasizes the need for further research on how tree growth stage influences susceptibility and on the possibility of cross-breeding among provenances.

The insecticidal effect of low temperatures for the control of all life stages of two stored-product insects, Plodia interpunctella and Liposcelis bostrychophila, was evaluated under laboratory conditions. The temperatures tested were 0, ??5, ??10 and ??15 °C, and, at these temperatures, the insects were exposed for 2, 4 and 8 h and also for 1, 2, 3 and 7 days. Regarding P. interpunctella, the most cold-tolerant life stage was larvae, given that 2 days of exposure were needed for complete (100%) mortality at ??10 °C. Moreover, all larvae were dead at ??15 °C even after 2 h of exposure. Eggs of P. interpunctella were susceptible to cold, as mortality was complete after 7, 1 day, 2 and 2 h at 0, ??5, ??10 and ??15 °C, respectively. L. bostrychophila was by far less susceptible than P. interpunctella. For this species, adults were still alive even after 7 days at ??10 °C, while complete mortality was achieved only after 1 day at ??15 °C. Eggs of L. bostrychophila were the most cold-tolerant life stage, as survival was observed even after 3 days at ??15 °C. The results of the present study provide the first data set on which all life stages of P. interpunctella and L. bostrychophila are evaluated for their susceptibility to cold treatments in a standardized series of exposures and temperatures. These results are expected to further encourage the “real world” application of cold treatments for the disinfestation of durable stored-products.  相似文献

The invasive stink bug Halyomorpha halys is established in many European and American agro-ecosystems, where it causes severe crop losses. Potential control measures might include enhancement of plant defences. When attacked by herbivorous insects that oviposit on it, the plant may respond by priming direct defences, which might affect the development of future brood. Halyomorpha halys attacks numerous plant species in the invaded areas. Here, we investigated whether Vicia faba plants challenged by H. halys females can impair the development of its offspring through the activation of induced direct defences. We measured the weight and dimension of nymphs that developed on oviposition-experienced plants after 7 and 17 days from hatching. Nymphs that developed on oviposition-experienced plants weighed less compared to those that developed either on control plants or on plants solely subjected to H. halys feeding, and third instars showed shorter dimensions (tibia length). In addition, when oviposition-experienced plants were attacked by nymphs, higher and more rapid expression of two jasmonic acid-dependent genes (cysteine proteinase inhibitor gene and NAI1) was detected, possibly due to a mechanism of priming. Increased expression of the salicylic acid-responsive PR1 gene was also detected in egg-experienced plants, although the response was delayed compared to JA-dependent genes. Our results suggest that V. faba plants recognize H. halys oviposition as a warning signal and pre-activate defences against future nymphal herbivory.  相似文献

Tomicus minor Hartig (Col., Curculionidae, Scolytinae), occurring on Pinus sylvestris L., is a species which demonstrates high reproductive capability on weakened stands, accelerating the process of forest death. In protected areas, T. minor is regarded as a sensitive bioindicator that reacts to decline in the health and vitality of forests. Although there have been many publications concerning T. minor, no precise method has yet been given for estimating its population so as to enable the monitoring of forest vitality and assessment of the role played by T. minor in the forest ecosystem. The aim of the present work is to develop a statistical method for estimating populations of T. minor, requiring minimum work and interference with the forest ecosystem and permitting the computation of estimation errors. Research was carried out in the years 1992–2011 in pine stands aged over 80 years, growing in a variety of habitats and situated at varying distances from sawmill timber yards. Attack density of T. minor was measured on trap logs made from uninfested living trees. The population of T. minor on the trap logs was described using a multiple linear regression model with two explanatory variables. Among the features investigated, the T. minor population was found to depend significantly on the number of egg galleries on the fifth metre of the trap log counted from the thinner end (p < 0.001) and on the diameter of the trap log in bark at the thinner end (p < 0.05). The model explains approximately 85% (R 2 = 0.8564) of the variation in the total number of T. minor egg galleries on the trap logs. The numbering of units beginning from the thinner end of the log enabled increased precision in determining the model parameter resulting from the concentration of egg galleries on certain units of the log. In all validated plots, the mean real and model values for the number of T. minor egg galleries on the trap logs are similar (p > 0.5), confirming the high accuracy of the developed model.  相似文献

Halyomorpha halys, originating from East Asia, has become an important agricultural pest in northern temperate regions since its accidental introduction into North America in the mid-1990s and Europe in 2007. Trissolcus japonicus is the predominant egg parasitoid of this pest in its native range. Here, we tested the hypothesis that T. japonicus uses volatiles associated with H. halys adults during host location. The effects of these volatile compounds on H. halys adults were also assessed. Chemical analysis with GC-MS and electrophysiological analysis with GC-EAD revealed the presence of two bioactive volatile compounds from H. halys adults, n-tridecane and (E)-2-decenal. In the Y-tube assays, female T. japonicus were attracted by n-tridecane but strongly repelled by (E)-2-decenal. Furthermore, host searching time of female T. japonicus was significantly reduced when an H. halys egg mass was treated with n-tridecane and prolonged when treated with (E)-2-decenal or a mixture of (E)-2-decenal and n-tridecane. Male H. halys were significantly attracted by n-tridecane, whereas females had no response to this compound in the Y-tube assays. (E)-2-Decenal strongly repelled male and female H. halys in the Y-tube assays. We discuss the multifunctional kairomonal and other possible roles of these semiochemicals at different trophic levels in the context of intra- and inter-specific chemical communication, in the H. halysT. japonicus system.  相似文献
