为明确镰刀菌毒素在我国不同小麦产区的污染与分布,利用超高效液相色谱-串联质谱仪对全国214个小麦品种籽粒中的7种镰刀菌毒素含量进行了测定和比较分析。结果表明:7种镰刀菌毒素的平均回收率在95.02%~116.52%之间,相对标准差为6.32%~21.50%。所有测试小麦样品均被镰刀菌毒素污染,7种毒素中以雪腐镰刀菌烯醇(nivalenol,NIV)含量最高,即使在NIV含量最低的长江中下游冬麦区,平均含量也高达182.56μg/kg,虽然目前并没有关于NIV的相关限量标准,但也应该予以足够重视;脱氧雪腐镰刀菌烯醇(deoxynivalenol,DON)含量较高,在8.07~337.23μg/kg之间,低于国家限量标准1 000μg/kg;T-2含量在4.05~78.05μg/kg之间,低于现有国际限量标准100μg/kg;玉米赤霉烯酮(zearalmone,ZEA)含量在5.30~12.50μg/kg之间,低于国家限量标准60μg/kg;其它3种毒素含量较低,均低于17.73μg/kg。不同麦区间毒素比较发现,DON和3-乙酰脱氧雪腐镰刀菌烯醇的含量在长江中下游冬麦区最高,分别为43.09μg/kg和6.39μg/kg,且显著高于其它麦区;来自长江中下游冬麦组和黄淮冬麦区南片水地组的小麦品种籽粒中毒素T-2和HT-2的含量显著高于其它麦区;其余毒素在不同麦区间均无显著差异。  相似文献

小麦四种镰刀菌毒素的ELISA检测及毒素污染分析   总被引:3,自引:1,他引:2  
为明确镰刀菌毒素在小麦中的污染与分布状况,选取以四种镰刀菌毒素的特异性抗体制备的ELISA试剂盒,检测了来自全国范围内的183个冬小麦、46个春小麦品种(系)样品中的镰刀菌毒素含量,并对不同麦区小麦样品中脱氧雪腐镰刀菌烯醇(DON)、伏马(fumonisin)、单端孢霉烯醇(T-2)和玉米赤霉烯酮(ZON)四种毒素含量及污染率进行比较分析.结果表明,我国小麦品种(系)中镰刀菌毒素污染比较普遍,但污染水平较低,均符合国家规定标准.四种镰刀菌毒素中以DON毒素为主,其平均含量为73.87μg/kg.不同麦区间DON、fumonisin和T-2毒素的含量存在显著差异(P<0.01),而ZON毒素的含量在麦区间差异不显著(P>0.05),东北春麦晚熟组、长江中下游冬麦组和黄淮冬麦区南片冬水组镰刀菌毒素总体含量较高.DON与ZON毒素污染率在麦区间存在显著差异(P<0.01),而fumonisin与T-2毒素污染率差异不显著(P>0.05).在不同麦区间,东北春麦晚熟组的DON毒素的含量和污染率均为最高.  相似文献

本研究明确了黄淮冬麦区主栽小麦品种对赤霉病的抗侵染、抗扩展、抗毒素积累和抗籽粒侵染能力以及几种抗性之间的相互关系,并检测了各品种是否带有FHB1抗性基因,旨在为该区小麦赤霉病抗性鉴定评价、抗性品种培育和利用提供科学依据。小麦赤霉病综合抗性鉴定结果表明:22个黄淮冬麦区主栽品种中,有20个为感病品种,只有‘郑麦9023’和‘西农979’为中感品种,所有品种均不含FHB1基因;长江中下游麦区的9个品种中,‘扬麦17’和‘宁麦9号’等6个品种表现中抗,‘扬麦23’表现中感,‘苏麦3号’和‘扬麦21’表现抗,‘扬麦14’‘扬麦17’和‘扬麦23’不含有FHB1基因,其他品种均含FHB1基因。小麦品种的抗扩展能力与抗侵染能力无显著相关性(r=0.27,P0.05);两种接种条件下小麦品种的病粒率与抗脱氧雪腐镰刀菌烯醇(DON)毒素积累能力呈极显著正相关(r=0.86,P0.01;r=0.88,P0.01);单小花滴注法接种条件下,小麦品种的平均病级与病粒率和籽粒中DON含量都呈极显著正相关(r=0.71,P0.01;r=0.81,P0.01);喷雾接种条件下,小麦品种的病小穗率与平均病级、病粒率、籽粒中DON含量和ZEN含量都呈极显著正相关(r=0.78,P0.01;r=0.73,P0.01;r=0.78,P0.01;r=0.63,P0.01)。在毒素积累抗性上,DON含量和ZEN含量呈极显著正相关(r=0.70,P0.01)。在目前黄淮冬麦区没有中抗品种的情况下,可以增加育种和鉴定目标为抗籽粒侵染和抗毒素积累的品种,在小麦品种推广过程中加以运用,可以达到较好的效果。  相似文献

脱氧雪腐镰刀菌烯醇( DON) 在小麦籽粒中的积累分析   总被引:2,自引:0,他引:2  
 镰刀菌产生的脱氧雪腐镰刀菌烯醇(deoxynivalenol,DON)污染小麦后对人畜具有较大的毒害作用。为明确DON毒素在小麦籽粒中的积累数量及其与镰刀菌菌株、接菌数量、小麦品种和病害严重度的关系,试验采用单花滴注方法在5个抗病性不同的小麦品种上分别接种9 个禾谷镰刀菌菌株,每个菌株接种1 × 106 、1 × 105 和1 × 104 个/ mL 3 个分生孢子浓度,并利用ELISA 法测定收获麦粒中DON 毒素含量。结果表明,麦粒中DON 毒素含量差异主要是由于不同禾谷镰刀菌菌株产生毒素能力不同所致。接种菌株8003、4020 的所有麦粒中DON 毒素含量均显著高于相同条件下接种其他7 个菌株的小麦。当接种产毒素能力强的菌株时,小麦抗病品种表现出在一定程度上降低DON 毒素积累的能力。不同接菌浓度对小麦赤霉病的发病程度和麦粒中DON 毒素含量有显著影响,在相同条件下,接菌浓度越高,病害严重度越高,DON 毒素含量也越高;反之,接菌浓度越低,病害严重度越低,DON 毒素含量也越低。  相似文献

为探明在小麦不同生育期施用新型杀菌剂丙硫菌唑及施用次数对小麦赤霉病及籽粒DON毒素总量[包括脱氧雪腐镰刀菌烯醇(DON)?3-乙酰脱氧雪腐镰刀菌烯醇(3-ADON)?15-乙酰脱氧雪腐镰刀菌烯醇(15-ADON)]的控制效果, 2020年-2021年通过田间自然发病和人工接种发病试验的方法, 研究了小麦不同生育期施用丙硫菌唑及施用次数对小麦赤霉病的防效, 并通过液相色谱-串联质谱法测定了不同处理小麦籽粒中对DON毒素总含量?结果表明, 30%丙硫菌唑可分散油悬浮剂(OD)防治2次对小麦赤霉病的防效和对DON毒素的控制效果均显著高于防治1次; 防治2次时, 其首次最佳防治时期为小麦齐穗期至扬花20%, 防效为90.25%~95.13%, 毒素控制效果为77.35%~79.97%?30%丙硫菌唑OD作为防控小麦赤霉病的新型药剂具有良好的应用前景, 本研究为该药剂推广应用于小麦赤霉病及籽粒DON毒素的防控提供了科学依据?  相似文献

 为明确不同小麦品种(系)对赤霉病的抗性和麦穗组织中DON毒素积累水平,培育和利用抗赤霉病和DON毒素积累的品种提供资源和依据,本研究采用单小花滴注接种法对河南省的106个小麦品种(系)抗赤霉病性进行鉴定分析,并用ELISA测定了病穗组织中DON毒素水平。结果表明不同小麦品种(系)对赤霉病的抗性有显著差异,106个小麦品种(系)中未发现抗病和中抗材料,中感品种(系)有华育198、郑麦103和春丰0021等14个,占13.2%;感病的有曌式2010-06、百农898和中麦63等92个,占86.8%。不同小麦品种(系)籽粒、颖壳和穗轴中DON毒素积累水平有显著差异,籽粒中DON毒素水平在(0.70~287.63)mg/kg之间,其中郑03876、豫保1号和中麦63 的DON毒素水平在2 mg/kg 以下,为抗毒素材料;其他的103个品种DON毒素水平大于2 mg/kg;颖壳和穗轴中的DON毒素水平在(51.03~392.87)mg/kg之间,普遍比籽粒中DON毒素含量高。籽粒中DON毒素水平与小麦品种(系)的平均病害严重度间呈极显著正相关。  相似文献

 为明确黄淮麦区主栽小麦品种对茎基腐病的抗性水平、不同抗性指标的相关性,以及茎杆和籽粒中镰刀菌毒素积累情况,在采用苗期茎基部滴注法和成株期混合播种法进行抗性鉴定的基础上,本研究还利用超高效液相色谱高分辨质谱联用技术,测定了小麦茎杆和籽粒中6种常见镰刀菌毒素的含量。结果表明,供试的20个小麦品种中,苗期抗病、中抗、感病和高感的分别有‘百农207'等4个、‘郑麦7698'等7个、‘周麦18'等 4个和‘矮抗58'等5个,分别占20%、35%、20%和25%。成株期表现抗病、中抗和感病的分别有‘周麦18'和‘中麦895'共2个、‘济麦23'等 8个和‘矮抗58'等 10个,分别占10%、40%和50%。抗性鉴定数据的统计分析发现,苗期和成株期抗性水平无显著相关性(P>0.05)。毒素检测结果表明,供试品种籽粒中均未检测到毒素,茎秆中均检测到脱氧雪腐镰刀菌烯醇(DON)、玉米赤霉烯酮(ZEN)及隐蔽型真菌毒素脱氧雪腐镰刀菌烯醇-3-葡萄糖苷(D3G),含量分别为4.03~17.65 mg·kg-1、0.09~1.28 mg·kg-1和1.93~16.82 mg·kg-1。根据研究结果,认为‘矮抗58'和‘郑麦7698'可作为感病和中抗对照品种。本研究结果为小麦茎基腐病抗性鉴定评价、抗性品种培育和利用提供了科学依据。  相似文献

小麦赤霉病流行区镰刀菌致病种及毒素化学型分析   总被引:5,自引:0,他引:5  
 为从分子水平上明确小麦赤霉病流行区镰刀菌致病种及其B 型毒素化学型的分布特点,本研究对2008 年度采自四川、重庆、湖北、安徽、江苏、河南6 省33 县市的赤霉病穗上分离获得的433 个镰刀菌单孢菌株,用鉴定种和鉴定B 型毒素化学型的特异性引物进行了鉴定分析。致病种检测结果表明,四川病穗检测到Fusarium asiaticum、F. graminearum、F.avenaceum 和F. meridionale 4 个镰刀菌种,重庆、湖北、安徽和江苏病穗检测到F. asiaticum 和F. graminearum 2 个种,河南病穗仅检测到F. graminearum 1 个种。毒素化学型检测结果表明,Nivalenol(NIV)是四川和重庆镰刀菌主要毒素化学型,Deoxynivalenol(DON)是湖北、河南、安徽和江苏镰刀菌主要毒素化学型;将DON 化学型进一步划分为3-AcDON 和15-AcDON 显示,四川、湖北、江苏镰刀菌毒素以3-AcDON 为主,安徽镰刀菌毒素为3-AcDON 和15-AcDON 两者参半,河南镰刀菌全部产生15-AcDON。结果揭示,F. asiaticum 是四川、重庆、湖北和江苏等赤霉病流行麦区的优势致病种;镰刀菌产生的DON 和NIV 毒素化学型存在明显的地域分布,长江上游的麦区以NIV 为优势化学型,长江中下游麦区以DON 为优势化学型;镰刀菌致病种与DON 毒素的化学型间存在一定关系。  相似文献

储存对小麦中DON毒素含量的变化规律研究   总被引:1,自引:0,他引:1  
本研究的目的是调查南京地区含有脱氧雪腐镰刀菌烯醇(Deoxynivaleno,DON,又称致吐毒素)的小麦中常温储存条件下DON毒素浓度的变化情况。采用耦联电子捕获器气谱色谱仪对小麦储存期内面粉中的DON含量逐月进行检测。小麦面粉储存前DON浓度是8.83±1.75 mg/kg,储存期DON毒素平均浓度分别为6.66、7.33、6.08、7.90、5.45、6.56、7.29、8.02、6.82和8.60 mg/kg,结果表明在储存期间小麦面粉中的DON毒素没有明显变化。研究结果揭示了在调查地区常温储存条件下,小麦中的致吐毒素没有发生显著的变化。  相似文献

小麦赤霉病是小麦穗期的主要病害之一。化学防控一直是小麦主产区防控赤霉病的主要措施。为明确几种新型杀菌剂对小麦赤霉病的防效和对小麦籽粒DON毒素含量的影响,于2018年进行了氰基丙烯酸酯类和三唑类杀菌剂单剂及其复配剂对赤霉病的防效试验。结果表明:30%戊唑·多菌灵悬浮剂(SC)1500 mL/hm^2处理对赤霉病病穗防效达92.40%,病指防效达93.20%,小麦籽粒DON毒素检出量较不用药对照降低80.38%;25%氰烯菌酯SC 2000 mL/hm^2处理对赤霉病的病穗防效达86.80%,病指防效达88.78%,小麦籽粒DON毒素检出量较不用药对照降低88.19%;48%氰烯·戊唑醇SC 900 mL/hm^2和40%丙硫·戊唑醇SC 600 mL/hm^2对小麦赤霉病的病穗防效分别为77.20%、78.00%,病指防效分别为80.27%和79.59%,对籽粒DON毒素检出量较不用药对照分别降低73.87%和81.42%。在小麦赤霉病较重发生的情况下,上述4种杀菌剂单剂或复配剂1次用药既能高效控制病情,又能有效控制小麦籽粒DON毒素不超标。本试验研究进一步阐明,氰烯菌酯、戊唑醇、丙硫菌唑等杀菌剂及其复配剂均能有效控制小麦赤霉病的危害,并能有效降低小麦籽粒DON毒素含量;吡唑醚菌酯单剂及其复配剂虽然对小麦赤霉病的病穗和病指防效也较高,但控制小麦籽粒DON毒素含量效果相对较差。研究结果为小麦穗期赤霉病化学防控提供了科学参考。  相似文献

Grain samples of 15 naturally contaminated barley cultivars, collected after harvest in southeastern Poland, were analysed for occurrence of Fusarium trichothecenes and zearalenone (ZEA). Barley kernels were contaminated with the following toxic metabolites: deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), 15-acetyldeoxynivalenol (15-AcDON), nivalenol (NIV), HT-2 toxin (HT-2), T-2 toxin (T-2), diacetoxyscirpenol (DAS), T-2 tetraol and ZEA. Significant correlations between concentrations of individual toxins and the dominant Fusarium species were found. Moreover, significant differences in toxin concentrations between cultivars were detected. Distribution of these mycotoxins was studied in two fractions of kernels (diameter > 2.5 mm and < 2.5 mm). A two-factor analysis of variance revealed significant differences between the two fractions, and between the analysed cultivars. Most of the interactions between fractions and cultivars were also significant. The highest concentration of the analysed toxins was in the fraction of small kernels. Kernel fraction <2.5 mm, although accounting for only 12.8% of sample weight, contained high proportions of the total toxin content: 80% of DON, 94% of NIV, 85% of ZEA, 83% of T-2 tetraol, 80% of DAS, 68% of HT-2 toxin and 81% of T-2 toxin. The results indicate that the level of contamination with Fusarium trichothecenes and ZEA, can be reduced by rejection of small kernels.  相似文献

Fusarium head blight (FHB), caused principally by Gibberella zeae (Fusarium graminearum), is a devastating disease of small grains such as wheat and barley worldwide. Grain infected with G. zeae may be contaminated with trichothecene mycotoxins such as deoxynivalenol (DON) and nivalenol (NIV). Strains of G. zeae that produce DON may also produce acetylated derivatives of DON: 3‐acetyl‐DON (3‐ADON) and 15‐acetyl‐DON (15‐ADON). Gradients (clines) of 3‐ADON genotypes in Canada have raised questions about the distribution of G. zeae trichothecene genotypes in wheat fields in the eastern USA. Tri3 and Tri12 genotypes were evaluated in 998 isolates of G. zeae collected from 39 winter wheat fields in New York (NY), Pennsylvania (PA), Maryland (MD), Virginia (VA), Kentucky (KY) and North Carolina (NC). Ninety‐two percent (919/998) of the isolates were 15‐ADON, 7% (69/998) were 3‐ADON, and 1% (10/998) was NIV. A phylogenetic analysis based on portions of three genes (PHO, RED and URA) from 23 isolates revealed two species of Fusarium (F. graminearum sensu stricto and one isolate of F. cerealis (synonym F. crookwellense)). An increasing trend of 3‐ADON genotypes was observed from NC (south) to NY (north). Punctuated episodes of atmospheric transport may favour a higher frequency of 3‐ADON genotypes in the northeastern USA, near Canada, compared with the mid‐Atlantic states. Discoveries of the NIV genotype in NY and NC indicate the need for more intensive sampling in the surrounding regions.  相似文献

Fusarium head blight (FHB) in small grain cereals is primarily caused by the members of the Fusarium graminearum species complex. These produce mycotoxins in infected grains, primarily deoxynivalenol (DON); acetylated derivatives of DON, 3‐acetyl‐DON (3‐ADON) and 15‐acetyl‐DON (15‐ADON); and nivalenol (NIV). This study reports the isolation of Fusarium cerealis in infected winter wheat heads for the first time in Canada. A phylogenetic analysis based on the TRI101 gene and F. graminearum species‐specific primers revealed two species of Fusarium: F. graminearum sensu stricto (127 isolates) and F. cerealis (five isolates). Chemotype determination based on the TRI3 gene revealed that 65% of the isolates were 3‐ADON, 31% were 15‐ADON and 4% were NIV producers. All the F. cerealis isolates were of NIV chemotype. Fusarium cerealis isolates can often be misidentified as F. graminearum as the morphological characteristics are similar. Although the cultural and macroconidial characteristics of F. graminearum and F. cerealis isolates were similar, the aggressiveness of these isolates on susceptible wheat cultivar Roblin and moderately resistant cultivar Carberry differed significantly. The F. graminearum 3‐ADON isolates were most aggressive, followed by F. graminearum 15‐ADON and F. cerealis NIV isolates. The findings from this study confirm the continuous shift of chemotypes from 15‐ADON to 3‐ADON in North America. In Canada, the presence of NIV is limited to barley samples and the discovery of NIV‐producing F. cerealis species in Canadian wheat fields may pose a serious concern to the Canadian wheat industry in the future.  相似文献

Quantitative real-time polymerase chain reaction differentiating 10 Fusarium spp. and Microdochium nivale or M. majus was applied to a total of 396 grain samples of wheat, barley, triticale, oat, and rye sampled across Denmark from 2003 to 2007, along with selected samples of wheat and barley from 1957 to 2000, to determine incidence and abundance of individual Fusarium spp. The mycotoxins deoxynivalenol (DON), nivalenol, zearalenone, T-2, and HT-2 were quantified using liquid chromatography-double mass spectrometry. Major differences in the Fusarium species complex among the five cereals as well as great yearly variation were seen. Fusarium graminearum, F. culmorum, and F. avenaceum were dominant in wheat, with DON as the dominant mycotoxin. F. langsethiae, F. culmorum, and F. avenaceum were dominant in barley and oat, leading to relatively high levels of the mycotoxins T-2 and HT-2. F. graminearum, F. culmorum, and F. avenaceum dominated in triticale and rye. The nontoxigenic M. nivale/majus were present in significant amounts in all cereal species. Wheat and barley samples from 1957 to 1996 exhibited no or very low amounts of F. graminearum, indicating a recent increase of this pathogen. Biomass and mycotoxin data exhibited good correlations between Fusarium spp. and their corresponding mycotoxins under field conditions.  相似文献

Combined analyses of the natural occurrence of fusarium head blight (FHB), mycotoxins and mycotoxin‐producing isolates of Fusarium spp. in fields of wheat revealed FHB epidemics in 12 of 14 regions in Hubei in 2009. Mycotoxin contamination ranged from 0·59 to 15·28 μg g?1 in grains. Of the causal agents associated with symptoms of FHB, 84% were Fusarium asiaticum and 9·5% were Fusarium graminearum, while the remaining 6·5% were other Fusarium species. Genetic chemotyping demonstrated that F. asiaticum comprised deoxynivalenol (DON), 3‐acetyldeoxynivalenol (3‐AcDON), 15‐acetyldeoxynivalenol (15‐AcDON) and nivalenol (NIV) producers, whereas F. graminearum only included DON and 15‐AcDON producers. Compared with the chemotype patterns in 1999, there appeared to be a modest shift towards 3‐AcDON chemotypes in field populations during the following decade. However, isolates genetically chemotyped as 3‐AcDON were present in all regions, whereas the chemical 3‐AcDON was only detected in three of the 14 regions where 3‐AcDON accounted for 15–20% of the DON and acetylated forms. NIV mycotoxins were detected in seven regions, six of which also yielded NIV chemotypes. The number of genetic 3‐AcDON producers was positively correlated with amounts of total mycotoxins (DON, NIV and acetylated forms) or DON in wheat grains. Chemical analyses of wheat grains and rice cultures inoculated with different isolates from the fields confirmed their genetic chemotypes and revealed a preferential biosynthesis of 3‐AcDON and 4‐AcNIV in rice. These findings suggest the importance of chemotyping coupled with species identification for improved prediction of mycotoxin contamination in wheat.  相似文献

Experiments were conducted to determine the extent of Fusarium langsethiae infection in wheat, barley and oats grown under identical experimental conditions. In total, four experiments were conducted with both winter and spring sown experiments at two locations. The amount of F. langsethiae infection was determined by quantifying F. langsethiae DNA and quantifying the combined concentration of the trichothecene mycotoxins HT-2 and T-2 (HT-2 + T-2) in cereal head fractions (grain and rest of the head) after threshing at harvest. Results of the study showed that under identical experimental conditions, oats had the highest F. langsethiae DNA and HT-2 + T-2 concentrations compared to wheat and barley. This indicates that the high levels detected on UK oats compared to wheat and barley from surveys of commercial crops is a consequence of genetic differences rather than differences in agronomy applied to the cereal species. The concentration of HT-2 and T-2 per unit of F. langsethiae DNA in oats compared to wheat and barley was also significantly higher indicating host differences in either the stimulation of HT-2 and T-2 production or in the metabolism of HT-2 and T-2. The study also showed that the proportion of F. langsethiae DNA in threshed grains was significantly lower than that in the rest of the cereal head.  相似文献

Fusarium head blight (FHB) is an important disease of wheat, which can result in the contamination of grains with mycotoxins such as deoxynivalenol (DON). Artificial inoculation of flowering ears with conidial suspensions is widely used to study FHB diseases. Our goal was to compare four inoculation treatments in which a conidial suspension was sprayed on flowering ears and to study the effect of the application of moisture during kernel setting and filling with a mist-irrigation system. Ten wheat genotypes were inoculated with a DON-producing Fusarium culmorum strain. Inoculation treatments varied in time of application of the inoculum (morning or evening) and in the method of controlling humidity during inoculation (bagging or mist irrigation). A wet season was simulated with a mist-irrigation system, keeping the crop canopy wet for at least 26 days after flowering. The severity of FHB symptoms (area under disease progress curve (AUDPC)), yield loss and DON contamination in the grains were determined. AUDPC data obtained with the different inoculation treatments were highly correlated (r=0.85–0.95). Mist irrigation after inoculation resulted in a higher mean disease severity, but in a overall lower toxin contamination as compared to the non-irrigated treatments. Genotypic differences in DON accumulation were present: for one wheat line toxin contamination significantly increased when irrigated, while two genotypes accumulated significantly less toxin. The closest relationships (r=0.73–0.89) between the visual symptoms and the DON content were obtained under moderate mean infection pressure. This relation between visual symptoms and the DON content deteriorated at higher infection levels.  相似文献

ABSTRACT A meta-analysis of the effect of tebuconazole (e.g., Folicur 3.6F) on Fusarium head blight and deoxynivalenol (DON) content of wheat grain was performed using data collected from uniform fungicide trials (UFTs) conducted at multiple locations across U.S. wheat-growing regions. Response ratios (mean disease and DON levels from tebuconazole-treated plots, divided by mean disease and DON levels from untreated check plots) were calculated for each of 139 studies for tebuconazole effect on Fusarium head blight index (IND; field or plot-level disease severity, i.e., mean proportion of diseased spikelets per spike) and 101 studies for tebuconazole effect on DON contamination of harvested grain. A random-effects meta-analysis was performed on the log-transformed ratios, and the estimated mean log ratios were transformed to estimate the mean (expected) percent control for IND ( C(IND) ) and DON ( C(DON)). A mixed effects meta-analysis was then done to determine the effects of wheat type (spring versus winter wheat) and disease and DON levels in the controls on the log ratios. Tebuconazole was more effective at limiting IND than DON, with C(IND) and C(DON) values of 40.3 and 21.6%, respectively. The efficacy of tebuconazole as determined by the impact on both IND and DON was greater in spring wheat than in winter wheat (P < 0.01), with a 13.2% higher C(IND) and a 12.4% higher C(DON) in spring wheat than in winter wheat. Fusarium culmorum (Fc) and F. graminearum (Fg) belong to the predominant causal agents of fusarium crown and root rot (FCR) in wheat. While many studies have been done to investigate crown rot, including stem base infection, root colonization and mycotoxin production associated with root rot is not well understood. In this study the impact of mycotoxins on the colonization of wheat roots and stem bases was analysed by using Fc and Fg isolates that varied in both quantity and types of trichothecenes they produce. Seedling inoculations in growth chambers with a high deoxynivalenol (DON)- and 3-acetyldeoxynivalenol (3ADON)-producing isolate led to more severe symptoms and 20-times greater colonization of the stem base, as measured by Fc DNA accumulation, than isolates that produced less DON/3ADON. In contrast to stem base colonization, in vitro inoculations of roots with a Tri5 deletion mutant deficient in Fg trichothecene production led to three-times higher colonization than the wildtype. Furthermore, an Fc isolate that produced low levels of zearalenone resulted in twice the level of colonization of a high DON/3ADON-producing isolate included in the study. When root inoculation with a low DON/3ADON-producing Fc isolate was supplemented with exogenous DON, DON production decreased by more than half per unit weight of Fc DNA, and root colonization doubled compared to the untreated control. Therefore, in contrast to its potential role as an aggressiveness factor in stem base infection, trichothecene production by Fc and Fg is detrimental to the early stages of wheat root colonization in FCR.  相似文献

Summary An infection of bread wheat by fusarium head blight contaminates the crop with mycotoxins, particularly deoxynivalenol (DON) and nivalenol (NIV). The toxicity and natural occurrence of these mycotoxins in wheat are reviewed. Based on 8 years data of fusarium head blight epidemics of wheat in the Netherlands, DON contamination of the grain was estimated. Fusarium head blight ratings averaged an infection of 1.7% of all spikelets; estimates for DON contamination averaged 0.9 mg kg–1. Taking a guideline level for DON in uncleaned bread wheat of 2 mg kg–1, in 1979 and 1982 a wheat crop was produced with estimated DON concentrations above the limit of tolerance. Human and animal exposure to mycotoxins in the Netherlands appears to be small but chronic. The information presented in this paper illustrates the need for an annual evaluation of the crop for fusarium head blight incidence and mycotoxin content, and the necessity of fusarium head blight resistant wheat cultivars.Samenvatting Aaraantasting van tarwe doorFusarium culmorum enFusarium graminearum leidt tot vorming van mycotoxinen in het graan, waarvan deoxynivalenol (DON) en nivalenol (NIV) de belangrijkste toxinen zijn. Gebaseerd op gegevens vanFusarium epidemieën in de jaren 1979–1986 wordt een schatting gegeven van de concentratie DON in Nederlandse tarwe. Rekening houdend met de herkomst en verwerking van tarwe, blijken zowel in dierlijk als menselijk voedsel lage concentraties DON chronisch voor te komen. Op basis van een maximaal toelaatbare dagelijkse dosis DON van 3 g kg–1 lichaamsgewicht is de schatting van de dagelijkse opname van DON in het jaar volgend op de oogst van 1982 net op de grens. Zowel een jaarlijkse inventarisatie vanFusarium aantasting en DON besmetting van het graan, als de ontwikkeling vanFusarium-resistente rassen zijn noodzakelijk.  相似文献
