The essential oils obtained by hydrodistillation from the aerial parts of Tunisian native Hypericum perfoliatum L. (sect. Drosocarpium Spach.) and Hypericum tomentosum (sect. Adenosepalum Spach.) were analyzed by GC and GC–MS. Thirty-two compounds were identified in the essential oils of H. perfoliatum with α-pinene (13.1%), allo-aromadendrene (11.4%), germacrene-D (10.6%), n-octane (7.3%), α-selinene (6.5%) and β-selinene (5.5%) as main constituents. Sixty-seven components were identified in the oil of H. tomentosum with menthone (17.0%), n-octane (9.9%), β-caryophyllene (5.3%), α-pinene (5.2%), lauric acid (4.1%) and β-pinene (3.7%) as the most abundant components. Both oils were characterized by the presence of many components which could have numerous applications in food, pharmaceutical and perfume industries.  相似文献

The variation in the content and composition of Origanum majorana L. essential oil has been studied. Plant material has been harvested at four phenological stages (early vegetative, late vegetative, budding and full-flowering). Essential oil yield varied from 0.04 to 0.09% reached during the full-flowering stage. Analysis of the essential oils by GC and GC/MS revealed the presence of 38 components represented mainly by oxygenated monoterpenes (64.01–71.4%), monoterpene hydrocarbons (21.73–29.92%) and sesquiterpene hydrocarbons (1.47–4.05%). The main components were terpinen-4-ol (29.13–32.57%), cis-sabinene hydrate (19.9–29.27%), trans-sabinene hydrate (3.5–11.61%), γ-terpinene (2.11–8.20%), bornyl acetate (1.52–2.94%) and linalool (1.05–1.39%). On the other hand, phenolic contents varied from 2.706 to 6.834 mg/g of dry weight obtained during the later vegetative stage. RP-HPLC analysis of the methanolic extract of O. majorana L. dried aerial parts showed the predominance of phenolic acids during the early vegetative stage whereas flavonoids predominate during the other stages of growth. The main phenolic acids identified were trans-2-hydroxycinnamic, rosmarinic, vanillic, chlorogenic, gallic and cinnamic whereas the main flavonoids were amentoflavone, apigenin, quercetin, luteolin, coumarin and rutin. Results obtained showed that later vegetative stage is characterized by the highest contents of bioactive compounds and therefore it could be considered as the best stage for harvesting marjoram plants.  相似文献

Hydrodistilled volatile oil obtained from the aerial parts of Artemisia annua L., cultivated near Sarajevo, Bosnia, was analyzed by GC-MS. More than one hundred compounds were identi?ed, representing 95.5% of the total oil. The major constituents of essential oil were oxygenated monoterpenes, artemisia ketone (30.7%) and camphor (15.8%). Isolated essential oil was tested for radical-scavenging ability using the stable DPPH radical, the ABTS radical, for reducing power ability with a test based on the reduction of ferric cations, for reducing ability of hydroxy radical in ORAC assay, and for metal chelating ability using the ferrozine assay. In all tests oil did not show a prominent antioxidant activity, but still comparable with thymol, an already known antioxidant. The screening of antimicrobial activity of oil was individually evaluated against representatives of Gram-positive, Gram-negative bacteria and fungi, using the agar diffusion method. All tested microorganisms were inhibited by essential oil. To the best of our knowledge, this is the first report of antimicrobial activity of essential oil of A. annua against Haemophilus influenzae, Enterococcus faecalis, Streptococcus pneumoniae, Micrococcus luteus and Candida krusei microbial strains. The antioxidant, antibacterial and antifungal activity of essential oil of A. annua from Bosnia is presented here for the first time and extends our knowledge in the range of valuable biological activities and possible roles in therapy associated with this medicinal herb.  相似文献

The direct esterification of chlorogenic acid (5-CGA) by immobilized Candida antarctica lipase B (Novozym 435) in supercritical CO2/t-butanol has been studied. The Taguchi approach was applied to evaluate the effects of temperature (35–55 °C), pressure (150–250 bar), t-butanol (2–10%, v/v), and the enzyme amount (10–30 mg/ml), on the ester concentration and overall conversion. Optimum reaction conditions were established at: 150 bar, 55 °C, 10% t-butanol (v/v), 20 mg/ml of lipase. Addition of 20 mg/ml of molecular sieves (3 Å) was also necessary to minimize the inhibiting effect of the increasing water concentration. Maximum conversions reached 77, 82 and 85% in 25 h using geraniol, pentanol and heptanol as aliphatic chain donors, respectively. HPLC analysis demonstrated the selective esterification of 5-CGA from a coffee pulp aqueous-methanolic extract, which reached a conversion to heptyl ester of 65% at the optimal condition. The supercritical CO2 selectivity towards the esterified product was the working principle of this study, by which minimized interphase transport limitations and enhanced mass-transfer phenomena substantially improved the reaction kinetics.  相似文献

A laboratory experiment was conducted with the poultry red mite Dermanysuss gallinae (De Geer) to assess the toxicity of a range of essential oils obtained from wild-growing plants in Tunisia to this pest. Details of the percentage essential oil yield from these plants were also recorded. For comparison, commercially sourced essential oil from Thymus vulgaris (L.) was also tested against D. gallinae after work elsewhere found this product to be acaricidal. Recently fed adult female D. gallinae were exposed to the essential oils at 0.1 mg oil/cm2 in Petri-dishes at 22 °C over a period of 24 h.Results showed that the yield of essential oil varied considerably depending upon the source plant. Whilst maximum yields of 0.5% were achieved, three of the seven wild plants selected provided yields of less than 0.1%. Similar variability was recorded with respect to the toxicity of the essential oils to D. gallinae. Three of the essential oils tested did not cause significant D. gallinae mortality (in comparison to the control). However, all other selected oils provided mortality levels statistically similar to the 90% mortality achieved with commercial T. vulgaris essential oil, with the oil from Pelargonium graveolens (L’Hér.) killing 100% of D. gallinae exposed to it. Essential oil from P. graveolens in particular may be suitable for further development as a D. gallinae acaricide alongside or in place of commercial thyme essential oil.  相似文献

Comparison of the seed oil of Moringa pterygosperma (moringa) and Sclerocarya birrea (marula) showed a marked difference in fatty acid composition and oxidative stability. Moringa, with 1000 ppm tocopherols, had an oil stability index (OSI) of 133 h at 110 °C while marula with 1000 ppm tocopherols yielded 37 h at 110 °C. This correlated well with the fatty acid composition of these two oils. Moringa had less than 1% polyunsaturates and marula had 6.7% of these oxidatively unstable materials. In addition, fatty acid compositions of seven species of moringa are presented. All of these species had levels of behenic acid ranging from 1 to 7% with oleic acid levels from 68 to 79%. The highest amount of polyunsatures was found in the moringa species was in Moringa drouhardii with 3.6%.  相似文献

The hydrodistilled essential leaves and stems oils of Orthosiphon stamineus Benth were analysed by GC–MS/MS. Sixty nine compounds representing 97.6 and 97.4% of the total leaves and stems oils, respectively were identified, of which β-caryophyllene (24.0 and 35.1%), α-humulene (14.2 and 18.4%), β-elemene (11.1 and 8.5%), 1-octen-3-ol (8.2 and 7.0%), β-bourbonene (3.4 and 3.0%), β-pinene (2.1 and 1.7%), caryophyllene oxide (1.6 and 2.2%), camphene (1.6 and 1.3%) and limonene (1.2 and 1.1%) were the major compounds. Thus, the monoterpenes and sesquiterpenes were the predominant portions of the oils. Essential oils and methanol extract of O. stamineus and the derived fractions of hexane, chloroform, and ethyl acetate were tested for anti-fungal activity, which was determined by disc diffusion and minimum inhibitory concentration (MIC) determination methods. The oils, methanol extract and derived fractions of methanol extract displayed great potential of anti-fungal activity as a mycelial growth inhibitor against the tested phytopathogenic fungi such as Botrytis cinerea, Rhizoctonia solani, Fusarium solani, Colletotricum capsici and Phytophthora capsici, in the range of 49.3–70.3% and minimum inhibitory concentration ranging from 500 to 1000 μg/ml.  相似文献

Essential oils from 22 aromatic plant species were tested for mortality of the mosquito larvae Culex quinquefasciatus. Lethal concentrations were determined for individual essential oils. Essential oils obtained from Thymus vulgaris, Satureja hortensis and Thymus satureioides plants showed the highest effect, with LC50 found lower than 50 μg/ml (33, 36 and 44 μg/ml, respectively). Analyses showed that majority substances for T. vulgaris were thymol and p-cymene (60.3 and 10.1%, respectively); carvacrol and γ-terpinene for S. hortensis (48.1 and 36.7%, respectively), and borneol and thymol for T. satureioides (30.3 and 32.5%, respectively).The selected essential oils also showed very good effectiveness with respect to mortality and percentage of adult emergence upon short-term exposure in water contaminated with lethal doses of individual oils. While there was 77% adult emergence from the larvae in the control, in T. vulgaris, T. satureoides and S. hortensis there was only 12.3, 15.3 and 16.0% adult emergence, respectively. High antioviposition effectiveness was found in all selected oils. Almost 100% deterrence of female oviposition was determined for all oils in concentrations of 0.02%. Significant differences were seen with tested concentrations of 0.01 and 0.005%, where the oil of T. vulgaris proved most effective (repellency about 99.8 and 62.3%, respectively).  相似文献

Optimal conditions were developed for hydrothermal processing of whole barley kernels (cv. Blenheim) to degrade phytate (myo-inositol hexaphosphate) and to increase the content of freemyo-inositol. The hydrothermal treatment comprised of two wet steeps, where lactic acid solution of different concentrations was used, and two dry steeps followed by successive drying. Experiments were performed as a central composite design and evaluated by multiple linear regression. The variables in the experiments were temperature in the first wet and dry steep (T1), temperature in the second wet and dry steep (T2) and lactic acid solution concentration in both wet steeps (C) and mathematical models were developed in these variables. Optimal conditions for maximal phytate degradation and for maximal increase of freemyo-inositol wereT1=48 °C,T2=48–50 °C andC=0·8%, at these conditions the amount of phytate was reduced by 95–96% and the freemyo-inositol concentration was increased from 0·56 to 2·45 μmol/g d.m. We conclude that this hydrothermal process can be used to produce a barley product (cv. Blenheim) with a low phytate content and a high level of freemyo-inositol.  相似文献

Barley alpha-amylase isozymes 1 (AMY1) and 2 (AMY2) have 80% sequence identity but possess different physico-chemical properties. By incubation in the range 37–85 °C T50 is 75.2 °C of AMY1 and 79.2 °C of AMY2. While AMY2 is also most stable in urea at pH 6.7, [urea]50 being 8.2 M compared to 7.9 M for AMY1, AMY1 has highest stability in urea below pH 6 or in the presence of NaCl. Moreover AMY1 is most stable in guanidinium chloride. Charge screening thus destabilises AMY2 but stabilises AMY1. Isozyme sequence comparison suggests that AMY1 lacks four of the 20 salt-bridges identified in the crystal structure of AMY2. The four residues that differ comprise Lys67AMY2 and Asp267AMY2, forming salt-bridges on the surface of the catalytic (β/α)8-barrel (domain A), and Glu96AMY2 and His344AMY2 that participate in charged networks between domain A and the small domain B and the C-terminal domain, respectively. Four corresponding AMY2 mimics A68K; D97E; Q269D; N346H were made in AMY1 by site-directed mutagenesis. While D97E and Q269D have slightly improved stability compared to AMY1 wild-type, N346H and, under certain conditions, A68K are destabilised. The four mutants show 22–176% activity (kcat/Km) toward 2-chloro-4-nitrophenol β- -maltoheptaoside and amylose DP17 and 43–117% activity for insoluble starch.  相似文献

The effects of modified atmosphere packaging (MAP) [bioriented polypropylene (BOPP-1 or BOPP-2)] in combination with antimicrobial agents Bacillus subtilis, 107 colony-forming units (cfu) ml−1; ethylenediamine tetraacetic acid, calcium disodium salt hydrate (EDTA) (0.1%); or 4-hexylresorcinol (4-HR) (0.15%) on postharvest decay control and quality retention of litchi cv. McLean's Red were assessed as possible replacements for commercial SO2 fumigation. Fruits dipped in B. subtilis, EDTA or 4-HR (5 min) separately, blow dried (25 °C, 3 min), packed in BOPP-1, held for 18 d at 2 °C, 95% RH, and 2 d at 14 °C, 75% RH were significantly less decayed. The antagonist–BOPP-1 combination also promoted the best bacterial survival during storage. B. subtilis was observed to survive effectively in BOPP-1 (16% O2, 6% CO2; 90% RH), but its survival was adversely affected in BOPP-2 (5% O2, 8% CO2; 93% RH). Alternaria alternata and Cladosporium spp. were the major decay-causing fungi in BOPP-1 treatments, and Candida, Cryptococcus and Zygosaccharomyces were the predominant yeasts in BOPP-2 treatments. Combination treatments EDTA, 4-HR or B. subtilis in BOPP-1 inhibited polyphenol oxidase (PPO) and significantly reduced pericarp browning and severity. Although the combination treatments EDTA, 4-HR or B. subtilis in BOPP were equally effective in controlling decay and browning, the EDTA and 4-HR affected the natural pinkish-red colour of the pericarp by showing higher h° values (orange–pink). Among the combination treatments, B. subtilis+BOPP-1 had the best potential to control decay, retain the colour and the overall litchi fruit quality during a marketing chain of 20 d.  相似文献

Soluble sugar contents and antioxidant enzyme activities were measured in the embryo of wheat (Triticum aestivum L., cv Charger) seeds throughout their storage under two conditions (45 °C and 100% relative humidity (RH), and 30 °C and 75% RH) to investigate whether grain deterioration during ageing was related to lipid peroxidation resulting in a decrease in the efficiency of the antioxidant defence system, and in changes in sugar metabolism. The half-viability period (P50) was only 6 d at 45 °C and 100% RH against about 3.75 months at 30 °C and 75% RH. However, sensitivity of grains to accelerated ageing carried out at 45 °C and 100% RH increased during their pre-storage at 30 °C and 75% RH. Loss of grain viability at 45 °C and 100% RH was associated with an accumulation of H2O2 which was concomitant with a progressive decrease in catalase (CAT) and superoxide dismutase (SOD) activities, and with an increase in glutathione reductase (GR) activity. However, malondialdehyde content did not change, suggesting that there was no lipid peroxidation during such an ageing. In return, ageing of grains at 30 °C and 75% RH was not associated either with strong changes in CAT, SOD and GR activities or with an accumulation of H2O2. When there was no correlation between CAT, SOD and GR activities and seed viability, there was a linear correlation between CAT activity and seed vigour evaluated by the T50 during ageing at 45 °C and 100% RH. Sugar metabolism in wheat embryo largely depended on ageing conditions. Loss of grain viability at 45 °C and 100% RH was associated with a marked decrease in sucrose (Su) and a slight increase in raffinose (Ra), and subsequently in the Ra/Su ratio. At 30 °C and 75% RH, seed ageing was associated with an increase in both sugars but with no strong increase in Su/Ra ratio. Our results suggest that ageing was associated with various mechanisms depending on the conditions of ageing, and that accelerated ageing at 45 °C and 100% RH was not the only model to consider in order to understand the mechanisms involved in seed deterioration.  相似文献

Study of the effect of hydrothermal process conditions on pasta quality   总被引:1,自引:1,他引:1  
The effect of hydrothermal treatment on the pasting, hydration properties and colour quality of commercial fresh pasta were studied following an Instantaneous Controlled Pressure Drop treatment. This hydrothermal procedure involves a physical modification at high temperature (<144 °C) and restricted moisture content (<30%) during a very short time (15–40 s) followed by a rapid pressure drop to a vacuum (50 mbar). Two process variables (steam pressure level and processing time) were investigated using response surface methodology. Steam pressure level had the greatest effect on hydration and pasting parameters. Increased pressure resulted in elevation of mass ratio between cooked and uncooked pasta (Wi/W0)100 °C and reduction of for all treated pasta. The optimum cooking time of untreated pasta was 7 min (W/W0=2.4), whereas for the same ratio it was less than 1 min for pasta treated at pressures ranging between 2.5 and 3.5 bar. The viscosity at 10 min (V10) increased linearly when the pressure level increased. V10 was 146 cP for untreated pasta whereas it reached 2659 cP for pasta treated at 3.9 bar (144 °C) for 25 s. The cold paste viscosity and setback viscosity decreased when the pressure varied from 1 to 3.9 bar.  相似文献

Lesquerella fendleri is a developing oilseed crop suitable for temperate growing regions in the US. The seed oil is rich in lesquerolic acid (57%) and could serve as a domestic source of hydroxy fatty acids. A method for the analysis of fatty acids and total oil content of a half or single lesquerella seed was developed. Lesquerella seeds are small with 1000 seed weights around 0.6 g (half seed mass of 200–500 μg). Conventional analytical balances provide mass accuracy to ±40 μg which fails to provide sufficient accuracy/precession (4–40% mass error) for the initial seed mass. A microbalance which measures to ±2 μg provided good reproducibility in initial seed weights but was not suitable for mass balance of the extracted oil. A normal phase HPLC coupled to an evaporative light scattering detector gave good response for oil in the mass range of 22 μg to 110 μg/mL. Therefore, micelle concentrations from single or half seed extractions could be determined with good reproducibility. This method was then evaluated on bulk seed that had been fractionated on a gravity table. Gravity table fractionation of L. fendleri seeds obtained from a large field plot provided seven fractions with increasing bulk density. These fractions were then analyzed in sets of 30 individual seeds and as an aggregate of 50 seeds. Oil content for individual seeds varied widely (15.6–44.2%) as did lesquerolic acid content (42.2–63.7%). The mean oil content increased (27–33%) with increasing bulk density (684–745 g/L). The mean lesquerolic acid content did not correlate with bulk density.  相似文献

In the present investigation, seed methanolic extracts of three Vitis vinifera (Muscat d’Italie, Syrah and Carignan) were assayed for their antioxidant activities. Results showed that there are strong variations in the contents of total phenols (440.97–121.94 mg GAE g−1 DW), flavonoids (48.07–16.81 mg EC g−1 DW) and tanins (37.15–14.9 mg EC g−1 DW) from the studied seeds. The phenolic composition of these extracts was determined by RP–HPLC after acid hydrolysis. The main phenolic compound was quercetin with 27.2% in Muscat d’Italie, 48.8% in Syrah and 28.4% in Carignan. Besides, all seed extracts showed remarkable DPPH radical scavenging activity with IC50 values ranged from 1.8 to 30 μg ml−1. EC50 values of reducing power activity ranged from 100 to 120 μg ml−1. The high phenolic content and the considerable antioxidant activities of vitis seed extracts could potentially be considered as an expensive source of natural antioxidants.  相似文献

Artemisia annua L. is an aromatic-antibacterial herb that destroys malarial parasites, lowers fevers and checks bleeding, and of which the secondary compound of interest is artemisinin. The objective of the present study was to determine yield, yield components and artemisinin content of A. annua L. grown under four nitrogen applications (0, 40, 80 and 120 kg ha−1) in the Çukurova region of Turkey in 2004 and 2005. Field trials were conducted at Çukurova University, Agricultural Faculty Field Crops Department. In the study, plant height, number of branches, fresh herbage yield, dry herbage yield, fresh leaf yield, dry leaf yield, essential oil content and artemisinin content (by high performance liquid chromatography, HPLC) were examined. By analysis of variance, nitrogen doses had no any statistical effect on the traits investigated except for artemisinin content. Artemisinin content of the dried leaves were significantly affected by nitrogen applications, which varied from 6.32 to 27.50 mg 100 g−1. Contents were from 120 and 80 kg ha−1 nitrogen for the years of 2004 and 2005, respectively.  相似文献

Six waxy rice samples (grown at the same site during the same season) were investigated to identify those aspects of amylopectin structure and granule composition which differentiate the starches into high (Tp 77·8 °C on average) or low (Tp 66·2 °C on average) gelatinisation temperature. All starches contained less than 2·5% amylose with negligible lipid. Granule dimensions were similar (mean diameter 4·9–5·7 μm) as was the β-amylolysis limit (59·9–63·0%). However, the structural elements of amylopectins within the two groups varied. The low gelatinisation temperature starches contained two major chain fractions upon debranching with iso-amylase, F3 (DP 16) and F1 (DP 51) on average while the high gelatinisation temperature starches contained three fractions, F3 (DP 16), F2 (DP 19) and F1 (DP 40). The proportion of F1/(F2+F3) for the low and high gelatinisation starches representing on average 20·2 and 10·5% respectively. Hence, the higher proportion of exterior chains and especially those with DP 19 appear to confer the higher gelatinisation temperature characteristic of these starches. Debranched β-limit dextrins were prepared from these starches. Regardless of their origin, (high or low gelatinisation temperature), they contained three fractions: F (DP 5), F (DP 13) and F (DP 19). Hence, the high versus low gelatinisation characteristic was imparted by the exterior chains. Using 13-C CP-MAS/NMR it was confirmed that the number of double helices within the starches were approximately constant although small differences in crystallinity were identified by X-ray diffraction. Retrogradation of the solubilised native high and low gelatinisation temperature amylopectin molecules confirmed that the ‘high temperature gelatinisation amylopectin’ molecules form higher dissociation temperature and enthalpy crystalline domains. This confirms that the higher proportion of short and especially relatively longer short chains promotes crystallite formation.  相似文献

Amino acid surfactants are high-value surfactants which have excellent emulsifying characteristics and minimal toxicity to the living body. Enzymatic synthesis of palm kernel amino acid surfactant was optimized by response surface methodology (RSM) using palm kernel olein (PKO) and l(+)-lysine catalyzed by Lipozyme RM IM. The reaction was performed in batch mode stirred tank reactor (STR) with one multi-bladed impeller. A central composite rotatable design (CCRD) was employed to evaluate the interactive effects of various parameters. The parameters were temperature (A): (40.00–70.00 °C), impeller speed (B): (100.00–400.00 rpm), substrates ratio (C): (1.00–4.00 mmol) and enzyme amount (D): (5.00–8.00 g). The optimum condition derived via RSM at fixed reaction time of 24 h was temperature; 47.50 °C, impeller speed; 323.96 rpm, substrates ratio; 3.25 mmol and enzyme amount; 7.25 g. The experimental yield was 89.03% under the optimum condition, which compared well with the maximum predicted value of 93.77%.  相似文献

The aim of this research was to evaluate acute toxicity of the essential oil of leaves of Chinese chives, Allium tuberosum Rottler ex Spreng (Asparagales: Alliaceae) and its major constituents against Apolygus lucorum Meyer-Dür (Hemiptera: Miridae). The essential oil of A. tuberosum leaves was obtained by hydrodistillation and analyzed by gas chromatography and gas chromatography-mass spectrometry. The major constituents of the oil were sulfur-containing compounds, including allyl methyl trisulfide (36.24%), diallyl disulfide (27.26%), diallyl trisulfide (18.68%), and dimethyl trisulfide (9.23%). The essential oil of A. tuberosum leaves exhibited acute toxicity against Ap. lucorum with an LD50 value of 20.03 μg per adult. Among the main compounds, diallyl trisulfide (LD50 = 10.13 μg per adult) showed stronger acute toxicity than allyl methyl trisulfide (LD50 = 21.10 μg per adult) and dimethyl trisulfide (LD50 = 21.65 μg per adult). The LD50 value of diallyl disulfide against Ap. lucorum was 28.10 μg per adult. The results indicated that the essential oil of A. tuberosum and its major constituents may have a potential to be developed as botanical insecticides against Ap. lucorum.  相似文献

Grain characteristics and groat composition have been evaluated in 35 genotypes from nine taxonomic species of Avena, including three species (A. agadiriana, A. atlantica, A. damascena) for which no previous data are available. There was substantial interspecific and intraspecific variation for all characteristics measured. The proportion of groat in the grain ranged from 32·7–62·1%, and mean groat weight from 2·4–37·4 mg. Groat protein concentrations ranged from 13·9–41·3%, and exceeded 32% in one A. atlantica, two A. damascena and one A. murphyi genotype. Groat β-glucan concentration showed very wide variation (2·2–11·3%) are there were substantial interspecific and intraspecific differences. The highest β-glucan concentrations were found in genotypes of A. atlantica. Although there were interspecific and intraspecific variations in groat oil concentration (4·2–10·6%), and in fatty acid composition, data were within previously reported ranges for A. sativa. Overall these data indicate that some of the genotypes of the wild species studied may be of value for breeding oats with improved levels of β-glucan and protein, and that further studies are warranted into both interspecific and intraspecific variations in grain quality factors in wild oat species.  相似文献
