The resistance to Fusarium oxysporum f.sp. melonis (Fom) race 1.2 has been studied in melons, such as the Portuguese accession ‘BG-5384’ and in the Japanese ‘Shiro Uri Okayama’, ‘Kogane Nashi Makuwa’, and ‘C-211’, since a good characterization of the resistance is necessary before its introgression into commercial varieties. These four melon accessions showed a high level of resistance to races 0, 1, and 2 of Fom, indicating that the partial resistance to the race 1.2 previously detected may not have been race specific. To determine the mode of inheritance of the resistance to Fom race 1.2, the F1, F2, BCPR, and BCPS generations from the crosses between the four resistant accessions above and ‘Piel de Sapo’, a Fom race 1.2 susceptible melon, were developed. They were subsequently inoculated with two Fom isolates, one from the pathotype 1.2Y and the other from the pathotype 1.2W. The area under the disease progress curve was determined for each inoculated plant, and the data were analyzed. We show that the resistance seen in these accessions is polygenically inherited with a complex genetic control because many epistatic interactions were detected. The three epistatic effects; additivity × additivity, dominance × dominance, and dominance × additivity are present and significant, with differing magnitudes from one cross to the next. The relatively low heritabilities, and these epistatic effects make difficult the improvement of the resistance, from these sources, through a standard selection procedure.  相似文献

Nicotiana wuttkei Clarkson and Symon discovered in the 1990s in Australia may be of potential interest to breeders as it carries resistance to Peronospora hyoscyami de Bary. The crossability between N. wuttkei (2n = 4x = 32) and three N. tabacum (2n = 4x = 48) cultivars (‘Puławski 66’, ‘Wiślica’ and ‘TN 90’) and the morphology and cytology of their amphihaploid hybrids (2n = 4x = 40) were studied. Seeds were produced only when N. wuttkei was used as the maternal parent, but under normal germination all seedlings died. Viable F1 hybrids of N. wuttkei × N. tabacum cv. ‘Puławski’ and N. wuttkei × N. tabacum cv. ‘Wiślica’ were obtained only by in vitro cotyledon culture. The amphihaploid plants were intermediate between the parents for most morphological traits. In 46.4% of the PMC’s, only univalents were present. The remainder of the cells had 1–5 bivalents and 1–2 trivalents. In spite of a detectable frequency of monads (2.6%), dyads (2.6%) and triads (4.5%), the hybrids were self and cross sterile.  相似文献

The objective of this work was to check the possible allelism between two sources of resistance to the root-knot nematode Meloidogyne incognita race 1 in lettuce (‘Grand Rapids’ and ‘Salinas-88’). The experiments were carried out in greenhouses, in expanded 128-cell polystyrene trays filled with commercial substrate. Lettuce cultivars ‘Salinas 88’ and ‘Grand Rapids’ were tested along with the populations F1 (‘Grand Rapids’ × ‘Salinas-88’), F2 (‘Grand Rapids’ × ‘Salinas-88’), F3 (‘Grand Rapids’ × ‘Salinas-88’), and with F4 families derived from the latter population. Seedlings were inoculated 15 days after sowing with a nematode egg suspension equivalent to 30 eggs ml−1 of substrate. Plants were evaluated for apparent gall incidence, gall scores, egg mass scores and extracted egg numbers 45 days after the inoculation date. There was evidence that two different genes are involved in control of resistance to M. incognita race 1 in lettuce cultivars Grand Rapids and Salinas-88. Lines with higher levels of nematode resistance than either Grand Rapids or Salinas-88 could be selected in the F4 generation of the cross between these resistant parental lines.  相似文献

Fusarium wilt, caused by Fusarium oxysporum f. sp. melonis (F.o.m), is a worldwide soil-borne disease of melon (Cucumis melo L.). The most effective control measure available is the use of resistant varieties. Resistance to races 0 and 2 of this fungal pathogen is conditioned by the dominant gene Fom-1. An F2 population derived from the ‘Charentais-Fom1’ × ‘TRG-1551’ cross was used in combination with bulked segregant analysis utilizing the random amplified polymorphic DNA (RAPD) markers, in order to develop molecular markers linked to the locus Fom-1. Four hundred decamer primers were screened to identify three RAPD markers (B17649, V01578, and V061092) linked to Fom-1 locus. Fragments amplified by primers B17649 and V01578 were linked in coupling phase to Fom1, at 3.5 and 4 cM respectively, whereas V061092 marker was linked in repulsion to the same dominant resistant allele at 15.1 cM from the Fom-1 locus. These RAPDs were cloned and sequenced in order to design primers that would amplify only the target fragment. The derived sequence characterized amplified region (SCAR) markers SB17645 and SV01574 (645 and 574 bp, respectively) were present only in the resistant parent. The SV061092 marker amplified a band of 1092 bp only in the susceptible parent. These markers are more universal than the CAPS markers developed by Brotman et al. (Theor Appl Genet 10:337–345, 2005). The analysis of 24 melon accessions, representing several melon types, with these markers revealed that different melon types behaved differently with the developed markers supporting the theory of multiple, independent origins of resistance to races 0 and 2 of F.o.m.  相似文献

Pseudomonas syringae is the main pathogen responsible for bacterial blight disease in pea and can cause yield losses of 70%. P. syringae pv. pisi is prevalent in most countries but the importance of P. syringae pv. syringae (Psy) is increasing. Several sources of resistance to Psy have been identified but genetics of the resistance is unknown. In this study the inheritance of resistance to Psy was studied in the pea recombinant inbred line population P665 × ‘Messire’. Results suggest a polygenic control of the resistance and two quantitative trait loci (QTL) associated with resistance, Psy1 and Psy2, were identified. The QTL explained individually 22.2 and 8.6% of the phenotypic variation, respectively. In addition 21 SSR markers were included in the P665 × ‘Messire’ map, of which six had not been mapped on the pea genome in previous studies.  相似文献

Pomegranate cultivation is one of the most attractive farming enterprises in the Indian arid tropics. However, the quality of the fruit is often severely affected by a physiological disorder called ‘aril browning’ in which a part or all the arils show discolouration (browning) and such fruits are unfit for consumption. This has become a serious concern to consumers, growers and researchers in the recent times. In order to understand the genotypic variation for aril browning and its association with other fruit traits, 158 progenies obtained by selfing two pomegranate multiple hybrids viz., {(‘Ganesh’ × ‘Kabul’) × ‘Yercaud’} × {(‘Ganesh’ × ‘Gulsha Rose Pink’)-F2} and {(‘Yercaud’ × ‘Jyothi’) × (‘Ganesh’ × ‘Gulsha Rose Pink’)-F2} × {(‘Ganesh’ × ‘Kabul’) × ‘Yercaud’} were studied. Because of heterozygous nature of the crop and diverse genetic base of parents, a wide array of recombinants were produced which were scored for aril browning, fruit skin colour, aril colour, total soluble solids (TSS) and seed mellowness. Results of Spearman’s correlation analysis revealed that aril browning is inversely related with aril colour (r = −0.41). A statistical model constructed to study the reasons for the observed variation in aril browning showed that about 82.9% of it was accounted collectively by skin colour, aril colour, TSS and seed mellowness. Further, a refined model represented by Y (aril browning severity) = 0.78 − 0.52 X1 (aril colour) + 0.23 X2 (TSS) was found to contribute to 73.5% of the observed variability in aril browning with least error in prediction. Analysis of data further showed that every unit increase in intensity of aril colour amounted to decrease in severity of aril browning by 0.52 units. However, for every unit increase in TSS there was an increase of 0.23 units in severity of aril browning. Thus, with the increase in intensity of aril colour there was a reduction in severity of aril browning while with raise in TSS, aril browning incidence was higher, an association often not favourable in selection of desirable genotypes. The results of the present study suggested that while developing varieties free from aril browning it is important to strike a balance between aril colour and TSS level.  相似文献

In a previously made cross Brassica napus cv. Oro (2n = 38) × Capsella bursa-pastoris (2n = 4x = 32), one F1 hybrid with 2n = 38 was totally male sterile. The hybrid contained no complete chromosomes from C. bursa-pastoris, but some specific AFLP (amplified fragment length polymorphism) bands of C. bursa-pastoris were detected. The hybrid was morphologically quite similar to ‘Oro’ except for smaller flowers with rudimentary stamens but normal pistils, and showed good seed-set after pollination by ‘Oro’ and other B. napus cultivars. The fertility segregation ratios (3:1, 1:1) in its progenies indicated that the male sterility was controlled by a single recessive gene. In the pollen mother cells of the male sterile hybrid, chromosome pairing and segregation were normal. Histological sectioning of its anthers showed that the tapetum was multiple layers and was hypertrophic from the stage of sporogenic cells, and that the tetrads were compressed by the vacuolated and disaggregated tapetum and no mature pollen grains were formed in anther sacs, thus resulting in male sterility. The possible mechanisms for the production of the male sterile hybrid and its potential in breeding are discussed.  相似文献

Powdery mildew is one of the most important melon pathogens all over the world. So far, many genes conferring resistance to powdery mildew of melon have been described, but few of these have been finely mapped or cloned. Two F2 populations derived from Ano2 × Hami413 and Ano2 × Queen were used to map the powdery mildew resistance gene by methods of Bulked Segregation Analysis (BSA), comparative genomics and Resistance Gene Analogues (RGA) mapping. It was found that the resistance to powdery mildew in Ano2 was conferred by a dominant gene, and the gene was named Pm-AN. The genetic analysis revealed that Pm-AN located between two codominant markers RPW and MRGH63B in linkage groupV. The genetic distances between Pm-AN and these two markers were 1.4–1.8 and 1.6–2 cM. No recombination was found between Pm-AN and markers ME/E1, SRAP23. Pm-AN was located in a RGA-rich region and cosegregated with the RGA marker MRGH5 and the resistance gene Vat. Synteny analysis showed that markers in this region were collinear between melon and cucumber. Segregation distortion was found in this region using both Ano2 × Hami413 and Ano2 × Queen F2 populations, and the distortion was more distinct in Ano2 × Hami413 F2 population. The center of segregation distortion was located in the RGA rich region harboring Pm-AN.  相似文献

The common bacterial blight pathogen [Xanthomonas axonopodis pv. phaseoli (Xap)] is a limiting factor for common bean (Phaseolus vulgaris L.) production worldwide and resistance to the pathogen in most commercial cultivars is inadequate. Variability in virulence of the bacterial pathogen has been observed in strains isolated from Puerto Rico and Central America. A few common bean lines show a differential reaction when inoculated with different Xap strains, indicating the presence of pathogenic races. In order to study the inheritance of resistance to common bacterial blight in common bean, a breeding line that showed a differential foliar reaction to Xap strains was selected and was crossed with a susceptible parent. The inheritance of resistance to one of the selected Xap races was determined by analysis of segregation patterns in the F1, F2, F3 and F4 generations from the cross between the resistant parent PR0313-58 and the susceptible parent ‘Rosada Nativa’. The F1, F2 and F3 generations were tested under greenhouse conditions. Resistant and susceptible F3:4 sister lines were tested in the field. The statistical analysis of all generations followed the model for a dominant resistance gene. The resistant phenotype was found to co-segregate with the SCAR SAP6 marker, located on LG 10. These results fit the hypothesis that resistance is controlled by a single dominant gene. The symbol proposed for the resistance gene is Xap-1 and for the bacterial race, XapV1.  相似文献

Interspecific hybridization was carried out between Lilium longiflorum and L. lophophorum var. linearifolium by using the cut style method of pollination, as a contrast, intraspecific hybridization between L. longiflorum ‘Gelria’ and L. longiflorum was also made, but no mature seeds and offspring were obtained from the two combinations under in vivo condition. Ovules excised from each carpel 5–35 days after pollination (DAP) were cultured on B5 or half-strength B5 medium containing sucrose at different concentrations in vitro. In L. longiflorum × L. lophophorum var. linearifolium, only 1.17% of ovules excised at 10 DAP developed into seedlings, and in L. longiflorum ‘Gelria’ × L. longiflorum, only 0.99% of ovules excised at 25 DAP developed into seedlings; none of the ovules excised at other different DAP in the two cross combinations produced any seedlings. The results showed that interspecific hybridization had a more serious post-fertilization barrier than the intraspecific hybridization, and that a lower concentration (3%) of sucrose led to better embryo development and higher percentage of seedlings in ovule cultures. All hybrid seedlings obtained were successfully transplanted to soil and grew normally. The progenies investigated were identified as true hybrids based on inter-simple sequence repeat (ISSR) analysis.  相似文献

Broad tolerance to phytophthora root rot (PRR) caused by Phytophthora sojae has become an important goal for the improvement of soybean (Glycine max) because of the rapid spread of races that defeat the available resistance genes. The aim of this research was to identify the location of quantitative trait loci (QTL) in ‘Conrad’, a soybean cultivar with broad tolerance to many races of P. sojae. A PRR susceptible breeding line ‘OX760-6-1’was crossed with Conrad. Through single-seed-descent, 112, F2 derived, F7 recombinant inbred lines (RILs) were advanced. A total of 39 random amplified polymorphic DNA bands (RAPDs) and 89 type 1 microsatellite (simple sequence repeat; SSR) markers were used to construct a genetic linkage map. In the greenhouse, RILs were inoculated with four P. sojae isolates (three from China and one from Canada). Disease was measured as the percent of dead plants 20 days after germination in P. sojae inoculated vermiculite in the greenhouse. Three QTLs (QGP1, QGP2, QGP3) for PRR tolerance in the greenhouse were detected using WinQTLCart 2.0 with a log-likelihood (LOD) score 27.14 acquired through permutations (1,000 at P ≤ 0.05). QGP1 (near Satt509) was located at linkage group F and explained 13.2%, 5.9%, and 6.7% of the phenotypic variance for tolerance to the JiXi, JianSanJiang and ShuangYaShan isolates, respectively. QGP2 (near Satt334) was located in a different interval on linkage group F and explained 5.1% and 2.4% of the phenotypic variance for JiXi and ShuangYaShan isolates, respectively. QGP3 was located on linkage group D1b + W (near OPL18800/SCL18659) and explained 10.2% of the phenotypic variance for Woodslee isolate. QGP1 and QGP2 appeared to be associated with PRR tolerance across a range of isolates but QGP3 was active only against the Woodslee isolate. At Woodslee and Weaver (in Ontario) in 2000, the interval associated with QGP3 explained 21.6% and 16.7% of phenotypic variance in resistance to PRR, respectively and was referred as QFP1. The identified QTLs would be beneficial for marker assistant selection of PRR tolerance varieties against both China and North America P. sojae races. Cytoplasmic male sterility (CMS) is an indispensable trait for F1 hybrid seed production in bunching onion (Allium fistulosum L.). Expansion of the cytoplasmic diversity of F1 hybrid cultivars by introduction of various CMS resources has great potential to eliminate vulnerability to cytoplasm type-specific diseases. This study aimed to evaluate appearance frequency of male sterile plants in several bunching onion accessions and to identify CMS resources. In eight (‘Nogiwa Aigara’, ‘Bansei Hanegi’, ‘Amarume’, ‘Kimnung’, ‘Zhangqiu’, ‘INT/CHN/1990/GOTOU’, ‘Natsunegi’ and ‘Guangzhou’) of 135 accessions collected from Japan, China, Mongolia, Korea and Taiwan, male sterile plants appeared with varied frequencies from 1.7% (‘Nogiwa Aigara’ and ‘Bansei Hanegi’) to 24.5% (‘Zhangqiu’). The inheritance mode of Zhangqiu- and Guangzhou-derived male sterility was confirmed to be CMS by sib-crossings and interbreed crossings. Microscopic examination of microsporogenesis in the CMS plants revealed that microspore protoplasm rapidly degenerated without mitotic division after the release of microspores from tetrads. The CMS germplasm described here would be useful for the development of “A” lines to be used in F1 hybrid seed production of bunching onion. Male fertility in ‘Nogiwa Aigara’, ‘Bansei Hanegi’, ‘Kimnung’, ‘INT/CHN/1990/GOTOU’ and ‘Natsunegi’ was verified to be controlled by a single fertility restoration locus.  相似文献

Zingiberene (ZGB) and acylsugars (AS) are allelochemicals responsible for high levels of arthropod resistance found in Solanum habrochaites (= Lycopersicon hirsutum) var. hirsutum ‘PI 127826’ and S. (= L.) pennelli ‘LA 716’, respectively. These accessions were used to develop commercial lines with good levels of pest resistance. The objective of the present work was to assess the ZGB and AS contents and the levels of resistance to Tuta absoluta in tomato hybrids between high ZGB × high AS lines, as compared with their parental lines and with commercial checks. High AS homozygous lines [TOM-688 and TOM-689, both originated from the interspecific cross S. lycopersicum (= L. esculentum) × S. pennelli], high ZGB homozygous lines (ZGB-703 and ZGB-704, both derived from the interspecific cross S. lycopersicum × S. habrochaites var. hirsutum), double heterozygotes for both ZGB and AS, single heterozygotes for ZGB, and single heterozygotes for AS were assessed for AS and ZGB contents. Low-ZGB low-AS checks ‘Débora Max’ and ‘TOM-684’ were used, as well as checks with high ZGB (PI 127826) and high AS (LA 716). The genotypes were submitted to infestation with South American tomato pinworm adults in a screenhouse, and oviposition counts were taken 10 days after the initial infestation date. Plants were scored for overall plant damage and percent leaflets attacked up to the 38th day after infestation. Genotypes heterozygous for ZGB or AS showed allelochemical contents intermediate to those of their high and low content parents, indicating incompletely dominant gene action for contents of each of the allelochemicals. There were no significant differences in T. absoluta oviposition between high-AS homozygous genotypes, high-ZGB homozygotes, single heterozygotes for AS, single heterozygotes for ZGB and double heterozygotes for ZGB and AS, but all these genotypes showed egg counts significantly lower than the low-ZGB low-AS checks. Feeding damage of T. absoluta was higher in the low-ZGB low-AS checks than in any other ZGB-rich or AS-rich tomato genotype. Relative to ZGB or AS single heterozygotes, the heterozygotes for both ZGB and AS showed higher levels of resistance to the insect, as measured by overall plant damage, indicating a synergic effect of the allelochemicals on resistance.  相似文献

Flowering dogwood (Cornus florida L.) is an ornamental tree valued for its showy white, pink, or red spring bract display and red fall color. A “pseudo” F2 flowering dogwood population was recently developed from a honeybee mediated cross of ‘Cherokee Brave’ × ‘Appalachian Spring’. The foliage color of 94 “pseudo” F2 plants segregated into green- and red- leaved phenotypes and was visually rated for color on five spring dates over 3 years (2007–2009). Chi-square analyses of observed segregation of phenotypes indicated that a complementary gene interaction form of epistasis controls foliage color with a 9:7 two gene ratio. We propose the symbols rl 1 and rl 2 for the genes controlling this trait.  相似文献

Sponge gourd is a popular vegetable grown throughout India. Tomato leaf curl New Delhi virus, the causal virus of tomato leaf curl disease, has recently been reported to be associated with sponge gourd, causing up to 100% crop loss under epidemic conditions. We have collected 30 genotypically diverse genotypes of sponge gourd from different parts of India, screened these for resistance under natural epiphytotic conditions, and then confirmed the results through challenge inoculation with a purified strain of the virus under insect-proof greenhouse conditions. The minimum vulnerability index was recorded in genotype DSG-6 (3.33), followed by DSG 7 (6.0) under the challenge (whitefly-populated) inoculation conditions. Two susceptible genotypes (‘Pusa Sneha’ and NSG-1-11), both possessing desirable fruit characters, were crossed with the two most promising resistant lines (DSG-6 and DSG-7) and the disease reaction of segregating and backcross generations studied through challenge inoculation with a purified strain of virus under insect-proof greenhouse conditions. A chi-square (χ2) test of frequency distribution based on the vulnerability index of the F2 progenies of the two resistant × susceptible crosses revealed monogenic dominant Mendelian ratio 3(R):1(S) to be the best fit in all crosses. This monogenic dominant model was further confirmed by the 1(R):1(S) ratio found to be best fit for the test cross with the susceptible parent. These results reveal that resistance to Tomato leaf curl New Delhi virus associated with yellow mosaic disease of sponge gourd is controlled by a single dominant gene in the genetic background of the resistant parents (DSG-6 and DSG-7) and that these two lines can be effectively utilized for the development of high-yielding and yellow mosaic disease-resistant varieties/hybrids of sponge gourd. This is the first conclusive identification of a resistant source and the inheritance of resistance against Tomato leaf curl New Delhi virus in sponge gourd.  相似文献

Near isogenic lines (NILs) varying for reduced height (Rht) and photoperiod insensitivity (Ppd-D1) alleles in a cv. Mercia background (rht (tall), Rht-B1b, Rht-D1b, Rht-B1c, Rht8c+Ppd-D1a, Rht-D1c, Rht12) were compared for interception of photosynthetically active radiation (PAR), radiation use efficiency (RUE), above-ground biomass (AGB), harvest index (HI), height, weed prevalence, lodging and grain yield, at one field site but within contrasting (‘organic’ vs. ‘conventional’) rotational and agronomic contexts, in each of 3 years. In the final year, further NILs (rht (tall), Rht-B1b, Rht-D1b, Rht-B1c, Rht-B1b+Rht-D1b, Rht-D1b+Rht-B1c) in Maris Huntsman and Maris Widgeon backgrounds were added together with 64 lines of a doubled haploid (DH) population [Savannah (Rht-D1b) × Renesansa (Rht-8c+Ppd-D1a)]. There were highly significant genotype × system interactions for grain yield, mostly because differences were greater in the conventional system than in the organic system. Quadratic fits of NIL grain yield against height were appropriate for both systems when all NILs and years were included. Extreme dwarfing was associated with reduced PAR, RUE, AGB, HI, and increased weed prevalence. Intermediate dwarfing was often associated with improved HI in the conventional system, but not in the organic system. Heights in excess of the optimum for yield were associated particularly with reduced HI and, in the conventional system, lodging. There was no statistical evidence that optimum height for grain yield varied with system although fits peaked at 85 and 96 cm in the conventional and organic systems, respectively. Amongst the DH lines, the marker for Ppd-D1a was associated with earlier flowering, and just in the conventional system also with reduced PAR, AGB and grain yield. The marker for Rht-D1b was associated with reduced height, and again just in the conventional system, with increased HI and grain yield. The marker for Rht8c reduced height, and in the conventional system only, increased HI. When using the System × DH line means as observations grain yield was associated with height and early vegetative growth in the organic system, but not in the conventional system. In the conventional system, PAR interception after anthesis correlated with yield. Savannah was the highest yielding line in the conventional system, producing significantly more grain than several lines that out yielded it in the organic system.  相似文献

The nutritional value of cucumber (Cucumis sativus L.) can be improved by the introgression of β-carotene (i.e., provitamin A and/or orange flesh) genes from “Xishuangbanna gourd” (XIS; Cucumis sativus var. xishuangbannanesis Qi et Yuan) into US pickling cucumber. However, the genetics of β-carotene content has not been clearly defined in this US market type. Thus, three previous populations derived from a US pickling cucumber (‘Addis’) × XIS mating were evaluated for β-carotene content, from which the high β-carotene inbred line (S4), ‘EOM 402-10’, was developed. A cross was then made between the US pickling cucumber inbred line ‘Gy7’ [gynoecious, no β-carotene, white flesh; P1] and ‘EOM 402-10’ [monoecious, possessing β-carotene, orange flesh; P2] to determine the inheritance of β-carotene in fruit mesocarp and endocarp tissue. Parents and derived cross-progenies (F1, F2, BC1P1, and BC1P2) were evaluated for β-carotene content in a greenhouse in Madison, Wisconsin. While F1 and BC1P1 progeny produced mature fruits possessing white, light-green, and green (0.01–0.02 μg g−1 β-carotene) mesocarp, the F2 and BC1P2 progeny mesocarp segregated in various hues of white, green, yellow (0.01–0.34 μg g−1 β-carotene), and orange (1.90–2.72 μg g−1 β-carotene). Mesocarp and endocarp F2 segregation adequately fit a 15:1 [low-β-carotene (0.01–0.34 μg g−1): high-β-carotene (1.90–2.72 μg g−1)] and 3:1 (low-β-carotene: high-β-carotene) ratio, respectively. Likewise, segregation of carotene concentration in mesocarp and endocarp tissues in BC1P2 progeny adequately fit a 3:1 (low-β-carotene: high-β-carotene) and 1:1 (low-β-carotene: high-β-carotene) ratio, respectively. Progeny segregations indicate that two recessive genes control the β-carotene content in the mesocarp, while one recessive gene controls β-carotene content in the endocarp. Single marker analysis of F2 progeny using the carotenoid biosynthesis gene Phytoene synthase determined that there was no association between this gene and the observed β-carotene variation in either fruit mesocarp or endocarp.  相似文献

Phytophthora root and stem rot caused by Phytophthora sojae, is one of the most damaging diseases of soybean, for which management is principally done by planting resistant cultivars with race specific resistance which are conferred by Rps (Resistance to Phytophthora sojae) genes. The Rps8 locus, identified in the South Korean landrace PI 399073, is located in a 2.23 Mbp region on soybean chromosome 13. In eight cv. Williams (rps8/rps8) × PI 399073 (Rps8/Rps8) populations, this region exhibited strong segregation distortion. In a cross between the South Korean lines PI 399073 (Rps8/Rps8) and PI 408211B (multiple Rps genes) this region segregated in a Mendelian fashion. In this study, microsporogenesis was evaluated to identify meiotic abnormalities that may be associated with the segregation distortion of the Rps8 region. Pollen was collected from greenhouse-grown plants of the parental genotypes: Williams, PI 399073, and PI 408211B; as well as selected Rps8/rps8 RILs from Williams × PI 399073 BC4F2:3 and PI 399073 × PI 408211B F4:5 populations. There were no differences for pollen viability among the genotypes. However, for PI 399073, a mix of dyads, triads, tetrads and pentads was observed. A high frequency of meiotic abnormalities including fragments, laggards, multinucleated microspores; and microcytes containing DNA was also observed in Rps8/rps8 Williams × PI 399073 BC4F2:3 RILs. These meiotic abnormalities may contribute to the high degree of segregation distortion present in the Williams × PI 399073 populations.  相似文献

Polish apple cvs: ‘Ligol’, ‘Odra’ and ‘Primula’ served for studies of self-incompatibility. Basing on available sequence data, a new set of primers upstream and downstream of the hypervariable (HV) region of apple S-RNases were designed. Using the RT-PCR method, cDNA was amplified on RNA isolated from styles. PCR products were cloned and sequenced. A new trans-generic S-RNase allele, designated as Skb (GenBank accession no. EU443101), was discovered in cvs ‘Odra’ and ‘Primula’. Nucleotide sequence alignment revealed that Skb-RNase shows 98% identity to SaucS19-RNase from Sorbus aucuparia and 97% identity to CmonS17-RNase from Crataegus monogyna. The occurrence of extensive intergeneric hybridization among extant Pyrinae is considered since the deduced amino acid sequence of Skb-RNase from M. × domestica showed higher similarity to CmonS17 from C. monogyna, SaucS19-RNase from S. aucuparia, St from Malus transitoria, S5-RNase and S3-RNase from Pyrus pyrifolia, and S40-RNase from P. ussuriensis than to S-alleles from Malus × domestica and all of them are grouped in the same cluster of phylogenetic tree. In respect to extremely high similarities between aforementioned S-RNases it could be possible that these alleles existed before the separation of Malus, Pyrus, Sorbus and Crataegus genera. Within Malus, the Skb-RNase from M. × domestica and St-RNase from M. transitoria show 100% identity of the HV region at the deduced amino acid level, suggesting that these S-RNases diverged more recently than the other Malus S-RNases. In ‘Ligol’, the agronomically most important cultivar in Poland, the S2 and S9 were identified.  相似文献
