In this study, the effect of an arbuscular mycorrhizal fungus (AMF) and two migratory endoparasitic nematodes on Musa plant growth, including the root system, were examined. In addition, the AMF-nematode interaction was studied. Seven Musa genotypes with different root systems were selected. Based on their relative mycorrhizal dependency, two genotypes (Calcutta 4 and Obino l'Ewai) were selected for AMF-nematode interaction studies. The experiments were performed under greenhouse conditions. Mycorrhization with Glomus mosseae resulted in a significantly better plant growth even in the presence of nematodes. The effect of AMF on the root system was genotype-dependent and seemed to be related to the relative mycorrhizal dependency of the genotype. The nematodes also affected the root system, decreasing branching. Nematode population densities were significantly reduced in the presence of AMF, except for Pratylenchus coffeae in Obino l'Ewai. In the root system, it appeared that the decreased branching caused by the nematodes was counterbalanced by the increased branching caused by the AMF.

Defoliation-induced changes in grass growth and C allocation are known to affect soil organisms, but how much these effects in turn mediate grass responses to defoliation is not fully understood. Here, we present results from a microcosm study that assessed the role of arbuscular mycorrhizal (AM) fungi and soil decomposers in the response of a common forage grass, Phleum pratense L., to defoliation at two nutrient availabilities (added inorganic nutrients or no added nutrients). We measured the growth and C and N allocations of P. pratense plants as well as the abundance of soil organisms in the plant rhizosphere 5 and 19 d after defoliation. To examine whether defoliation affected the availability of organic N to plants, we added 15N-labelled root litter to the soil and tracked the movement of mineralized 15N from the litter to the plant shoots. When inorganic nutrients were not added, defoliation reduced P. pratense growth and root C allocation, but increased the shoot N concentration, shoot N yield (amount of N in clipped plus harvested shoot mass) and relative shoot N allocation. Defoliation also reduced N uptake from the litter but did not affect total plant N uptake. Among soil organisms, defoliation reduced the root colonization rates of AM fungi but did not affect soil microbial respiration or the abundance of microbe-grazing nematodes. These results indicate that interactions with soil organisms were not responsible for the increased shoot N concentration and shoot N yield of defoliated P. pratense plants. Instead, these effects apparently reflect a higher efficiency in N uptake per unit plant mass and increased relative allocation of N to shoots in defoliated plants. The role of soil organisms did not change when additional nutrients were available at the moment of defoliation, but the effects of defoliation on shoot N concentration and yield became negative, apparently due to the reduced ability of defoliated plants to compete for the pulse of inorganic nutrients added at the moment of defoliation. Our results show that the typical grass responses to defoliation—increased shoot N concentration and shoot N yield—are not necessarily mediated by soil organisms. We also found that these responses followed defoliation even when the ability of plants to utilize N from organic sources, such as plant litter, was diminished, because defoliated plants showed higher N-uptake efficiency per unit plant mass and allocated relatively more N to shoots than non-defoliated plants.

Summary A lysimetric experiment was performed in a greenhouse to evalute root deposition and net release of soluble organic compounds after 1 and 2 years from pine and beech seedlings inoculated with an ectomycorrhizal fungus (Laccaria laccata) and/or rhizobacteria (Agrobacterium radiobacter for beech and Agrobacterium sp. for pine). Total C compounds released in the rhizosphere of both plants increased after inoculation with the bacteria or ectomycorrhizal fungus. The rhizobacteria increased root and plant growth and rhizodeposition, but the mycorrhizal fungi appeared to increase only root deposition. Soluble C compounds, collected after 2 years, represented only 0.1–0.3% of the total C compounds released into the rhizosphere, and were modified by inoculation with the microorganisms. After inoculation with the bacteria, levels of sugars and amino acids decreased in pine and beech rhizospheres, whereas organic acids increased, especially in the pine rhizosphere. In the rhizosphere of mycorrhizal beeches, sugar and amino acids increased, and organic acids differed from those released from non-mycorrhizal beeches. In the mycorrhizal pine rhizosphere, however, all compounds decreased. Following dual inoculations, mycorrhizal colonization increased, no effect on plant growth was observed, and virtually no organic acids were detected.

The P efficiency, crop yield, and response of maize to arbuscular mycorrhizal fungus (AMF) Glomus caledonium were tested in an experimental field with long-term (18-year) fertilizer management. The experiment included five fertilizer treatments: organic amendment (OA), half organic amendment plus half mineral fertilizer (1/2 OM), mineral fertilizer NPK, mineral fertilizer NK, and the control (without fertilization). AMF inoculation responsiveness (MIRs) of plant growth and P-uptake of maize were estimated by comparing plants grown in unsterilized soil inoculated with G. caledonium and in untreated soil containing indigenous AMF. Soil total P, available P, microbial biomass P, alkaline phosphatase activity, plant biomass, crop yield and total P-uptake of maize were all significantly increased (P < 0.05) by the application of OA, 1/2 OM, and NPK, but not by the application of NK. Specifically, the individual crop yield of maize approached zero in the NK-fertilized soils, as well as in the control soils. All maize plants were colonized by indigenous AMF, and the root colonization at harvest time was not significantly influenced by fertilization. G. caledonium inoculation increased mycorrhizal colonization significantly (P < 0.05) only with the NK treatment, and produced low but demiurgic crop yield in the control and NK-fertilized soils. Compared to the inoculation in balanced-fertilized soils, G. caledonium inoculation in either the NK-fertilized soils or the control soils had significantly greater (P < 0.05) impacts on soil alkaline phosphatase activity, stem length, plant biomass, and total P-uptake of maize, indicating that AMF inoculation was likely more efficient in extremely P-limited soils. These results also showed that balanced mineral fertilizers and organic amendments did not differ significantly in their effects on MIRs in these soils.

The effects of low temperature and reduced light on a Glycine-Bradyrhizobium-Glomus spp. symbiosis were examined in pot experiments. Soybean plants, Glycine max L. Merr. cv. Tachiyutaka, were grown with N fertilization or inoculation with Bradyrhizobium japonicum plus P fertilization or inoculation with Glomus mosseae in the glasshouse. After the flowering stage, half the pots with soybean plants were subjected to low temperature (15°C 14h/13°C 10 h) with light reduced by shading. At 0, 7, 16, and 28 days after the application of the treatments, the growth, nodulation, vesicular-arbuscular mycorrhizal (VAM) infection and the N and P contents of the soybean plants were measured. In all symbiont-fertilization combinations, the low-temperature treatment reduced the production of dry matter by the soybeans. Nodulation (weight and number) was slightly reduced by this treatment but the proportion of larger nodules was increased. The root length infected by the VAM fungus was little affected by the low-temperature treatment. Both the nodule weight and the infected root length were linearly related to shoot dry weight regardless of treatment and of the symbiont-fertilization combination used. These results suggest that the growth of the symbionts on the root was in balance with the shoot growth of the host, irrespective of climatic conditions, and imply a considerable degree of host control. P inflows to root systems were greatly affected by low-temperature treatment regardless of the symbiont-fertilization combination. This suggests that a simple comparison of P inflows between mycorrhizal and non-mycorrhizal plants may give misleading information on the effects of low temperature or reduced light conditions on P uptake by mycorrhizal plants.

To overcome soil nutrient limitation, many plants have developed complex nutrient acquisition strategies including altering root morphology, root hair formation or colonization by arbuscular mycorrhizal fungi (AMF). The interactions of these strategies and their plasticity are, however, affected by soil nutrient status throughout plant growth. Such plasticity is decisive for plant phosphorus (P) acquisition in P‐limited soils. We investigated the P acquisition strategies and their plasticity of two maize genotypes characterized by the presence or absence of root hairs. We hypothesized that in the absence of root hairs plant growth is facilitated by traits with complementary functions, e.g., by higher root mycorrhizal colonization. This dependence on complementary traits will decrease in P fertilized soils. At early growth stages, root hairs are of little benefit for nutrient uptake. Regardless of the presence or absence of root hairs, plants produced average root biomass of 0.14 g per plant and exhibited 23% root mycorrhizal colonization. At later growth stages of maize, contrasting mechanisms with functional complementarity explained similar plant biomass production under P limitation: the presence of root hairs versus higher root mycorrhizal colonization (67%) favored by increased fine root diameter in absence of root hairs. P fertilization decreased the dependence of plant on specific root traits for nutrient acquisition. Through root trait plasticity, plants can minimize trade‐offs for developing and maintaining functional traits, while increasing the benefit in terms of nutrient acquisition and plant growth. The present study highlights the plasticity of functional root traits for efficient nutrient acquisition strategies in agricultural systems with low nutrient availability.

The interactions between soil P availability and mycorrhizal fungi could potentially impact the activity of soil microorganisms and enzymes involved in nutrient turnover and cycling, and subsequent plant growth. However, much remains to be known of the possible interactions among phosphorus availability and mycorrhizal fungi in the rhizosphere of berseem clover (Trifolium alexandrinum L.) grown in calcareous soils deficient in available P. The primary purpose of this study was to look at the interaction between P availability and an arbuscular mycorrhizal (AM) fungus (Glomus intraradices) on the growth of berseem clover and on soil microbial activity associated with plant growth. Berseem clover was grown in P unfertilized soil (−P) and P fertilized soil (+P), inoculated (+M) and non-inoculated (−M) with the mycorrhizal fungus for 70 days under greenhouse conditions. We found an increased biomass production of shoot and root for AM fungus-inoculated berseem relative to uninoculated berseem grown at low P levels. AM fungus inoculation led to an improvement of P and N uptake. Soil respiration (SR) responded positively to P addition, but negatively to AM fungus inoculation, suggesting that P limitation may be responsible for stimulating effects on microbial activity by P fertilization. Results showed decreases in microbial respiration and biomass C in mycorrhizal treatments, implying that reduced availability of C may account for the suppressive effects of AM fungus inoculation on microbial activity. However, both AM fungus inoculation and P fertilization affected neither substrate-induced respiration (SIR) nor microbial metabolic quotients (qCO2). So, both P and C availability may concurrently limit the microbial activity in these calcareous P-fixing soils. On the contrary, the activities of alkaline phosphatase (ALP) and acid phosphatase (ACP) enzymes responded negatively to P addition, but positively to AM fungus inoculation, indicating that AM fungus may only contribute to plant P nutrition without a significant contribution from the total microbial activity in the rhizosphere. Therefore, the contrasting effects of P and AM fungus on the soil microbial activity and biomass C and enzymes may have a positive or negative feedback to C dynamics and decomposition, and subsequently to nutrient cycling in these calcareous soils. In conclusion, soil microbial activity depended on the addition of P and/or the presence of AM fungus, which could affect either P or C availability.

We have evaluated the effectiveness of arbuscular mycorrhizal fungi (AMF) inoculation (+M and ?M) at 0, 60, and 120 kg ?ha?1 of P fertilizer on crop growth (IEg), plant P nutrition and yield (IEy), and on mycorrhization occurrence in a processing tomato crop. Two experiments were carried out in calcareous soil under field conditions. Phosphorus fertilization had no effect on crop growth and yield. At harvests, +M plants showed higher aerial dry weight, fruit fresh weight, and P concentration. Inoculated plants produced larger inflorescences, higher flower number, and total and marketable fruit number compared with ?M plants. At P0 and P60, plants associated with exogenous AMF were able to enhance P recovery, nevertheless factors other than the P uptake improvement concurred to make the inoculation effective. In both years, P fertilization enhanced IEg and IEy, and the application of 60 kg ?ha?1 of P in inoculated soil was enough to reach high production level (134 Mg ?ha?1). In the first trial, due to earlier root mycorrhization in inoculated and P fertilized soil, higher IEg and IEy were obtained compared with the second experiment. In the latter, during the initial phase, plant growth was more affected by P fertilization than by soil arbuscular mycorrhizal (AM) inoculation. Root mycorrhization by native AM fungi indicates that the intensive management of the investigated agro-system did not depress fungi infectivity; however, it caused the selection of less effective AMF. The application of selected AMF as a biofertilizer may represent an innovative ecosustainable practice for improving the crop profitability for growers while reducing the need for P fertilization.

Defoliation of plants is known to have effects on soil organisms and nutrient availability in grassland communities, but few studies have examined whether changes in soil attributes can further feed back to plant growth and plant nutrient content. To examine defoliation-induced soil feedbacks, we established replicated miniecosystems with a grass Phleum pratense, defoliated half of the systems, collected soil from both defoliated and non-defoliated systems and planted new seedlings into each soil. The two soils did not differ in promoting shoot and root growth. However, seedlings that grew in the soil collected from defoliated systems had higher shoot N content, allocated relatively more N to shoots and had lower root N concentration than those growing in the soil collected from non-defoliated systems. Our study provides novel evidence that defoliation can generate long-lasting changes in grassland soil that in turn can affect plant N allocation.

Soil replant disease is the main bottleneck interfering with tree growth of peach in soils with poor traits. A potted study was conducted to evaluate the effects of inoculation with an arbuscular mycorrhizal fungus (AMF), Acauloapora scrobiculata, on plant growth, mineral nutrients, soil enzyme activities, soil microbial populations, and root exudate compositions of peach (Prunus persica L. Batsch) seedlings grown in replant soil and non-replant soil. After 15 weeks in AMF inoculation, replant soil heavily inhibited root mycorrhizal colonization. In replant soil, AMF inoculation significantly increased shoot biomass and root phosphorus, potassium, calcium, copper, zinc, iron, and boron concentrations. Mycorrhizal peach seedlings showed a higher number of soil bacteria and total microbes but a lower number of soil fungi under replant conditions, as well higher soil urease and acid phosphatase activity and lower soil sucrase and catalase activity. Greater soil aggregate stability was observed in mycorrhiza-inoculated replant soil than in non-mycorrhizal soil due to the increase of water-stable aggregates in 2–4 mm and 1–2 mm size. In addition, a total of 92 substances were identified in root exudates, and the mycorrhizosphere had considerably more root exudate compositions. AMF inoculation had a significantly inhibitive effect on the relative abundance of allelochemical substances, including benzoic acid, benzaldehyde, diisooctyl phthalate, phenols, and sterols, while there was an increase in diphenyl-ethanedione and à-(benzoyloxy)-benzeneacetonitrile in replanted peach. It was concluded that AMF inoculation could partly mitigate soil replant disease of peach through modulating soil microbe balance, improving soil aggregate stability, and changing root exudate compositions.

Cadmium (Cd) contamination in soil poses a huge threat to plants even at low concentrations; Broussonetia papyrifera has great potential in remediation of soil heavy metal contamination. However, whether exogenous indole-3-acetic acid (IAA) application and arbuscular mycorrhizal fungi (AMF) have synergistic effects on Cd tolerance of B. papyrifera remains unclear. To investigate the effects of AMF inoculation and IAA application on the tolerance of B. papyrifera to Cd stress, two experiments were conducted: the first to investigate the effect of AMF (Rhizophagus irregularis) inoculation on the tolerance of B. papyrifera to Cd stress and the second to investigate the combined effects of AMF inoculation and IAA application on the tolerance of B. papyrifera to Cd stress. Parameters including endogenous hormone concentration, antioxidant defense response, malondialdehyde (MDA) content, and gene expression related to antioxidant enzyme system and hormone were measured. The results indicated that AMF alleviated Cd toxicity of B. papyrifera by reducing MDA content and improving antioxidant enzyme activities and Cd absorption capacity. Furthermore, the combination of AMF inoculation and IAA application had a synergetic effect on the tolerance of B. papyrifera to Cd stress through upregulating BpAUX1 and BpAUX2, which might contribute to root growth and root xylem synthesis, and by upregulating BpSOD2 and BpPOD34 to enhance the antioxidant enzyme system. This work provides a new insight into the application of IAA in the remediation of soil Cd pollution by mycorrhizal plants.

Summary The effects of P, N and Ca+Mg fertilization on biomass production, leaf area, root length, vesiculararbuscular mycorrhizal (VAM) colonization, and shoot and root nutrient concentrations of pretransplant rice (Oryza sativa L.) plants were investigated. Mycorrhizal plants generally had a higher biomass and P, N, K, Ca, Mn, Fe, Cu, Na, B, Zn, Al, Mg, and S shoot-tissue nutrient concentrations than non-mycorrhizal plants. Although mycorrhizal plants always had higher root-tissue nutrient concentrations than non-mycorrhizal plants, they were not significantly different, except for Mn. N fertilization stimulated colonization of the root system (colonized root length), and increased biomass production and nutrient concentrations of mycorrhizal plants. Biomass increases due to N were larger when the plants were not fertilized with additional P. P fertilization reduced the colonized root length and biomass production of mycorrhizal plants. The base treatment (Ca+Mg) did not significantly affect biomass production but increased the colonized root length. These results stress the importance of evaluating the VAM rice symbiosis under various fertilization regimes. The results of this study suggest that pretransplant mycorrhizal rice plants may have a potential for better field establishment than non-mycorrhizal plants.

A field experiment was conducted to study and compare the effectiveness of two arbuscular mycorrhizal fungi (AMF), Glomus macrocarpum (GM) and Glomus fasciculatum (GF) on three accessions of Artemisia annua. The AM inoculation significantly increased the production of herbage, dry weight of shoot, nutrient status (P, Zn and Fe) of shoot, concentration of essential oil and artemisinin in leaves as compared to non-inoculated plants. The extent of growth, nutrient concentration and production of secondary plant metabolites varied with the fungus–plant accession combination. The mycorrhizal dependency of the three accessions was related to the shoot: root ratio. Comparing the two fungal inoculants in regard to increase in essential oil concentration in shoot, the effectiveness of GF was more than that of GM. While in two accessions, GM was more effective in enhancing artemisinin concentration than GF. Increase in concentration of essential oil was found to be positively correlated to P-status of the plant. Conversely, no correlation was found between shoot-P and artemisinin concentration.

Maize (Zea mays L.) and sorghum (Sorghum bicolor L.) Moench (local variety called Masakwat) plants were grown in a sterilized low-P soil in the greenhouse for 12 weeks. Each plant species was either mycorrhizal with vesicular-arbuscular mycorrhizal (VAM) fungi, non-mycorrhizal but minimally fertilized with soluble P, or non-mycorrhizal but highly fertilized with soluble P. Drought stress was imposed after 4 weeks at weekly intervals. Under unstressed conditions, leaf area, shoot dry weights, xylem pressure, and soil water potentials were similar for VAM and the two non-mycorrhizal P-fertilized treatments but each of the VAM-infected species had a greater total root length. Total P uptake was similar for the maize treatments but higher for VAM than non-mycorrhizal P-fertilized sorghum treatments. Under drought-stressed conditions, the growth parameters and soil water potential were similar for all maize treatments but they were reduced by mycorrhizal inoculation in sorghum. Greater water extraction occurred in drought-stressed mycorrhizal sorghum. In both plant species, total P uptake and P uptake per unit root length (including unstressed species) were significantly enhanced in non-mycorrhizal P-fertilized treatments compared with the mycorrhizal treatment. Except for the root dry weight of sorghum plants, there were no differences in the growth parameters and P uptake between minimally and highly P-fertilized non-mycorrhizal treatments for either maize or sorghum. The increased total root length in drought-stressed mycorrhizal sorghum plants and the similar infected root lengths in unstressed and drought-stressed sorghum plants may have caused high C partitioning to drought-stressed mycorrhizal roots and therefore caused the reduced growth parameters in mycorrhizal plants compared to the non-mycorrhizal P-fertilized counterparts. The results indicate that P fertilization in addition to mycorrhizal inoculation may improve the drought tolerance of maize and sorghum plants.

Aims : The aim of this study was to explore interactive effects between quality (types) and quantity (application rates) of biochar as well as of arbuscular mycorrhiza (AM) symbiosis on the growth of potato plants. Methods : A low P sandy loam soil was amended with 0%, 1.5%, or 2.5% (w/w) of either of 4 types of biochar, which were produced from wheat straw pellets (WSP) or miscanthus straw pellets (MSP) pyrolyzed at temperatures of either 550°C or 700°C. Potato plants grown in pots containing the soils or soil biochar mixture were inoculated with or without AM fungus (AMF), Rhizophagus irregularis. The experiment was carried out under fully irrigated semi‐field conditions and plants were harvested 101 days after planting. Results : Application of high temperature biochar decreased growth, biomass and tuber yield of potato plants, while the low temperature biochar had a similar effect on yield as plants grown without biochar amendment. Total biomass of potato plants were decreased with the increasing rate of biochar. Arbuscular mycorrhizal fungus inoculation stimulated the growth of potato plants in all organs, increased tuber biomass significantly in 1.5% MSP700 amended plants, and to a lesser degree for WSP700, MSP550, and WSP550. In addition, plant biomass gain was linearly related to N, P, and K uptake, the ratio of P to N in the leaf of plants indicated that all treatments were mainly P‐limited. A multiple linear regression using P uptake and biochar rate as independent variables explained 91% of the variation in total biomass. The single effect of AMF inoculation, type and rate of biochar affected plant N, P and K uptake similarly. While AMF inoculation significantly increased P uptake in potato plants grown in soil with WSP700 or MSP700 despite of the rate of biochar. In general, application of biochar significantly increased AMF root colonization of potato plants. Conclusions : The application of MSP550 at 1.5% combined with AMF stimulated growth of potato the most. Furthermore, the results indicated that the interactive effect of AMF inoculation, biochar type and application rate on potato growth to a large extent could be explained by effects on plant nutrient uptake.

The effect of manure and mineral fertilization on the arbuscular mycorrhizal (AM) fungal community structure of sunflower (Helianthus annuus L.) plants was studied. Soils were collected from a field experiment treated for 12 years with equivalent nitrogen (N) doses of inorganic N, dairy manure slurry, or without N fertilization. Fresh roots of tall fescue (Festuca arundinacea Schreb.) grass collected from the field plots without N fertilization and unfumigated field soils were used as native microbial inoculum sources. Sunflower plants were sown in pots containing these soils, and three different means of manipulating the microbial community were set: unfumigated soil with fresh grass roots, fumigated soil with fresh grass roots, or fumigated soil with sterilized grass roots. Assessing the implications with respect to plant productivity and mycorrhizal community structure was investigated. Twelve AM fungal OTUs were identified from root or soil samples as different taxa of Acaulospora, Claroideoglomus, Funneliformis, Rhizophagus, and uncultured Glomus, using PCR-DGGE and sequencing of an 18S rRNA gene fragment. Sunflower plants grown in manure-fertilized soils had a distinct AMF community structure from plants either fertilized with mineral N or unfertilized, with an abundance of Rhizophagus intraradices-like (B2). The results also showed that AM inoculation increased P and N contents in inorganic N-fertilized or unfertilized plants, but not in manure-fertilized plants.

Sečovlje salterns are an important protected area of biotic diversity in the Mediterranean. They represent an extreme environment with high salinity and drought that severely influence the growth of organisms. In the present study, diversity of plant halophytes and their mycorrhizal status were screened at eight different locations, which were mostly dikes and salt ponds, and which were deliberately selected for their distinct properties (e.g. soil salinity ranging from 105 to 2627 μS cm−1, vegetation type and management practice of the salterns).Twelve different halophytic plant species were recorded, of which eleven are designated as vulnerable. With few exceptions, they were found at the abandoned (Fontanigge) and sustained (Lera) locations of the Sečovlje salterns, distributed according to their tolerance to the salinity and waterlogging. The highest diversity of halophytes was listed at Fontanigge, in the abandoned, periodically flooded and gradually overgrown salt ponds. All of the examined species were colonised with either arbuscular mycorrhizal fungi (AMF) and/or dark septate endophytes (DSEs). High levels of colonisation were however detected only for species belonging to the Asteraceae and Plantaginaceae families. Higher root colonisation frequencies were generally seen for plants growing in the abandoned parts, when compared to the managed parts, whereas there was little correlation of the colonisation parameters with physicochemical parameters of rhizospheric soil properties.Molecular analysis by temporal temperature gradient gel electrophoresis (TTGE) of roots of halophytic plant species with confirmed AMF colonisation (arbuscules present) revealed the occurrence of at least six different AMF species, related to Glomus geosporum, Glomus caledonium and Glomus intraradices, and to different Glomus sp. clades and the Diversispora clade. This is to the best of our knowledge the first report of AMF and DSE mycorrhizal status of most of the halophyte plant species examined and of the brother scale identification of AMF species based on molecular analyses of roots of diverse halophytes from high saline environments.

We examined the effect of a vesicular-arbuscular mycorrhizal (VAM) fungus Glomus pallidum Hall on the phosphatase activity and cytokinin concentration in cowpea [Vigna unguiculata (L.) Walp] roots at successive stages of plant growth. Both acid and alkaline phosphatase activity were significantly (P=0.05) higher in mycorrhizal than in non-mycorrhizal roots 30 days after inoculation. Similarly, the cytokinin content was significantly increased in mycorrhizal roots compared to non-mycorrhizal roots. Our study suggests that these biochemical changes may improve the growth of mycorrhizal cowpea plants.
