In a context of the global major changes, it is mandatory to enlarge the range of crops supporting food security and pay great attention to neglected and underutilized species. However, basic knowledge of the biology of many neglected and underutilized species is still lacking to increase their yields. In this study, the mating system of white fonio [Digitaria exilis (Kippist.) Stapf], a West African minor and promising cereal, is assessed. Progenies arrays from both homozygous and heterozygous mothers were genotyped with microsatellites markers. The rate of genotyping errors in the experiments was assessed and a likelihood framework was used to determine the probability of different mating systems: outcrossing, self-fertilization and apomixis. The results suggested that white fonio has a highly selfing reproductive system with a possible outcrossing rate of 1.7%. Understanding the reproduction system of white fonio opens up opportunities for more effective breeding programs and a wider use of this cereal for food security improvement.  相似文献

South Italy is still a centre of diversity for melons but new cultivars progressively replace its traditional landraces. The area of Salento is important for several typical old crops as the cucumber melon ‘Meloncella’ (Cucumis melo L. var. chate (Hasselq.) Filov) traditionally cultivated for its unripe fruits. Information on cultivation, quality and variation of this Apulian landrace (probably the last relic of a wider cultivation in Europe) is reported. Runner bean (Phaseolus coccineus L.) is traditionally cultivated in Poland for dry seeds. The national collection of runner bean maintained in the National Center for Plant Genetic Resources gathers 152 accessions, which are mainly landraces originated in Poland (68%), Ukraine (17%) and Slovakia (10%). The collection contains valuable genetic resources for bean breeders and research. The aim of this study was to describe the level and structure of genetic diversity of three landraces and two commercial cultivars of runner bean from the national collection in order to assess their genetic potential for breeding. Amplified Fragment Length Polymorphism analysis included five combinations of selective primers. Analysis of seven genetic diversity parameters reveled fair amount of genetic variation both in landraces and cultivars. High genetic diversity of commercial cultivars relative those of landraces suggests that the breeding process leading to their release was rather moderate and most likely included domestic gene pool of runner bean. Low gene diversity and low Nei’s genetic distance values as well as intergradations among accessions in the PCoA may indicate reduced variability P. coccineus grown in Poland as a result of its migration pathways.  相似文献

Summary The genetic diversity of 95. representative Chilean common bean (Phaseolus vulgaris L.) landraces was analyzed using phaseolin seed protein and eight isozyme systems as genetic markers. Four types of phaseolin were found, “C”, “T”, “S” and “H”, in decreasing order of frequency. Each type had a different distribution between the Northern and Southern regions of the country. Nei’s genetic distance based on isozyme diversity indicated that a high percentage of the total variation found in this sample occurred between landraces and only a small percentage of the variation was detected within populations. Cluster analysis based on Nei’s genetic distance and a principal component analysis of isozyme frequencies did not detect a clear association between the geographic distribution of the landraces and their isozyme constitution. However, Nei’s genetic distance analysis clustered the bean landraces into two major groups which had a specific isozyme pattern, seed color, and seed size. The genetic analysis also detected a rare polymorphism for theMdh-2 locus, a null allele at theDiap-2 locus, and polymorphism for theAco-2 locus. The principal component analysis of isozyme frequencies showed that only 30% of the genotypes analyzed were similar to the Andean check and 5% of the samples were similar to Middle American check. This finding suggests a high frequency of hybridization between the Middle America and Andean gene pools in cultivated common bean from Chile.  相似文献

‘Domestication’ is a traditional farmers’ practice reported for yams (Dioscorea sp.) in Benin (West Africa). It involves introducing ‘spontaneous’ (naturally occurring) yams, supposedly wild (D. abyssinica and D. praehensilis), in varieties of the D. cayenensis–D. rotundata cultivated species complex. In this study, we established the genetic nature of ‘predomesticated’ yam plants using the amplified fragment length polymorphism (AFLP) technique. A total of 213 accessions, consisting of 32 predomesticated yams, 70 D. cayenensis–D. rotundata, 86 D. abyssinica and 25 D. praehensilis yams were analysed. Using 91 AFLP markers, three groups of accessions were distinguished, broadly corresponding to the above botanical species. Of the 32 predomesticated accessions, 16% were clustered with D. praehensilis, 37% with D. abyssinica and the remaining 47% with D. cayenensis–D. rotundata. These results demonstrated the use of wild plants by farmers in their domestication process, and suggested that plants derived from intervarietal and interspecific hybridisation may also be subject to this process. This study has shown that through domestication farmers influence and increase the genetic diversity in yam by using sexual reproduction of wild and possibly cultivated yams.  相似文献

Bambara groundnut (Vigna subterranea (L.) Verdc.) is a neglected and less exploited species in Benin. However, the rational use of the different landraces available involves an appropriate knowledge of their characteristics. This study examined the characteristics of 52 local landraces of Bambara groundnut traditionally cultivated in Benin based on 12 qualitative and 11 quantitative variables, in order to expose those of good qualities. After a classification based on the morphological variables of the seeds, the experimental field was installed in a completely randomized device of blocks repeated three times. The collected data was analyzed with Statistica 7.1 and NTSYS PC 2.2. softwares using qualitative characteristics (Growth’s type, Stem’s hairiness, Flower’s colour, Leave’s shape etc.). As result, the number of morphotypes increased from 18 (out of classification) to 47. Therefore, it appears that the Benin collection of Bambara groundnut is broad in morphological variability. The analysis of the principal component of the quantitative variables revealed three specific classes within the collection. The most efficient variety which belonged to the second class, had a high field yield per hectare (> 1 t/ha). The Souhouloumankpa variety yielded the earliest (103 days after sowing) in addition to the high field yield. The study also showed a positive relationship between 100 seed’s weight, their length, the number of seeds per plant and the seed’s field yield in kg/ha. For a sustainable preservation and an improvement of the available resources, a molecular characterization of the Bambara groundnut landraces in Benin Republic needs to be done.  相似文献

Melon (Cucumis melo L.) landraces of the Madrid provenance, Spain, have received national distinction for their high fruit quality and sensorial attributes. More specifically, a unique array of Group Inodorus landraces have been continuously cultivated and conserved by farmers in the municipality of Villaconejos since the 19th century. Their genetic relationships to other Group Inodorus and Flexuous melon market classes is not known, and, thus, a study was designed to determine their genetic relationships using 52 simple sequence repeat (SSR) markers, and then make genetic comparisons between these accessions and a previously published “Standard Reference Germplasm Array” (RA) containing Group Inodorus (14 Spanish and one USA), Flexuosus (1 Spanish), and Cantalupensis ( 2 USA) melon accessions. This subset consisted of 15 Spanish Group Inodorus landraces that circumscribed the genetic variation of major Spanish melon market classes (Groups Inodorus and Flexuosus), and USA commercial varieties (Groups Cantalupensis and Inodorus). Based on genetic distances, Villaconejos (Madrid) genotypes differed substantially from RA subset accessions, thus defining their genetic uniqueness. Principal component analysis (PCA) partitioned the accessions examined into four distinct groups revealing that Villaconejos black epidermis melons (landraces ‘Largo’, ‘Largo Negro Escrito’ and ‘Puchero’) were distinctly different from all other accessions examined, as cluster analysis separated Rochet market type Villaconejos’ accessions (landraces ‘Mochuelo’, ‘Mochuelo Tradicional’ and ‘Melón de Villaconejos’) from RA of the same market type. Genetic assessment of principal Spanish market classes revealed comparatively low intra-market heterogeneity in Piel de Sapo type accessions and high heterogeneity in Black and Yellow market type accessions. While a relatively high level of genetic introgression was detected between Yellow and Green market types, black epidermis market types were genetically unique. Given the uniqueness and high genetic diversity resident in Villaconejos landraces, this germplasm pool should be considered as a genetic source for broadening the comparatively narrow genetic base of Group Cantalupensis and Inodorus melon market types, especially standard commercial Spanish Group Inodorus market types (e.g., Piel de Sapo, Rochet, and Canari).  相似文献

Eight genomic SSR markers with a M13 tail attached were used to assess the genetic diversity of 72 Ussurian Pear accessions (Pyrus ussuriensis Maxim.) in China. The M13-tailed method was effective in discriminating all the 32 wild accessions. All the 40 Ussurian Pear cultivars could be successfully discriminated with the exception of 4 sets of synonymies or spots. A total of 108 alleles were obtained with an average of 13.5 per locus. The expected heterozygosity, observed heterozygosity, and power of discrimination were 0.78, 0.63, and 0.86 respectively. Three triploid cultivars (‘Anli’, ‘Ruan’er’, and ‘Pitaiguo’), and one wild accession, P. ussuriensis ‘Xilin-3’, showed three alleles at some SSRs. The number of alleles and observed heterozygosity per locus for 40 Ussurian Pear cultivars were 9.1 and 0.62, respectively, lower than the values of 32 wild accessions which were 11.3 and 0.65, respectively. A dendrogram based on the SSR genotypes was obtained, showing two major groups corresponding to cultivated group and wild group. All the cultivars fell into the cultivated group. Some subgroups (Nanguoli subgroup, Zhibazi subgroup, Xiangshuili subgroup, Balixiang subgroup, Anli subgroup) could be found in the cultivated group. A very close relationship between ‘Huagaili’ and ‘Miansuan’, and a close relationship between ‘Anli’ and a wild accession, P. ussuriensis ‘Huangshanli’ could be found in Anli subgroup. ‘Nanguoli’ and ‘Xiaowuxiang’ showed a close relationship with at least one identical allele at each locus with the exception of NH015a.  相似文献

Microstructure of the mature caryopsis of Digitaria exilis Stapf was studied by light and scanning electron microscopy and compared to chemical composition. The general structure of the caryopsis was similar to that of other grains, notably the millets. Thin bracts (the palea and lemma) and two glumes encased the caryopsis which consists of the thin, compressed layers of pericarp, testa, and cuticle surrounding the endosperm and embryonic tissues. The endosperm consisted of a single layer of aleurone cells and the starchy endosperm. The aleurone layer was thin over most of the starchy endosperm and thicker at the junction of the embryo and starchy endosperm. Aleurone cells contained lipid droplets and protein bodies. The cell contents of the starchy endosperm consisted of simple, polyhedral starch granules, lipid droplets, and protein bodies. Protein bodies were more abundant toward the periphery, and diminished toward the central portion of the starchy endosperm. Cells in certain regions of the embryo contained few, small, spherical starch granules and an abundance of protein bodies. Protein bodies containing phytic acid inclusions were located in the scutellum of the embryo. Compositional analyses revealed that the grain contained 8.2% protein, 2.1% fat, 0.48% fiber, and 1.4% ash.  相似文献

Little is known about genetic diversity and geographic origin of wheat landraces from Oman, an ancient area of wheat cultivation. The objectives of this study were to investigate the genetic relationships and levels of diversity of six wheat landraces collected in Oman with a set of 30 evenly distributed SSR markers. The total gene diversity, (HT), conserved in the three durum wheat (Triticum durum desf.) landraces (HT = 0.46) was higher than in the three bread wheat (Triticum aestivum L.) landraces (HT = 0.37), which were similar to Turkish and Mexican bread wheat landraces calculated in previous studies. Genetic variation partitioning (GST) showed that variation was mainly distributed within rather than among the durum (GST = 0.30) and bread wheat (GST = 0.19) landraces. Based on modified Rogers’ distance (MRD), the durum and bread wheat landraces were distinct from each other except for a few individuals according to principal coordinate analysis (PCoA). One bread wheat landrace (Greda) was separated into two distinct sub-populations. A joint cluster analysis with other landraces of worldwide origin revealed that Omani bread wheat landraces were different from other landraces. However, two landraces from Pakistan were grouped somewhat closer to Omani landraces indicating a possible, previously unknown relationship. Implications of these results for future wheat landrace collection, evaluation and conservation are discussed.  相似文献

Eighty-nine accessions of wild and cultivated peas (12 Pisum fulvum Sibth. et Smith., 7 P. abyssinicum A. Br., 31 wild and 42 cultivated forms of P. sativum L.) were analysed for presence of the variants of three functionally unrelated polymorphic markers referring to different cellular genomes. The plastid gene rbcL either contains or not the recognition site for restriction endonuclease AspLEI (rbcL+ vs. rbcL−); the mitochondrial gene cox1 either contains or not the recognition site for restriction endonuclease PsiI (cox1+ vs. cox1−); the nuclear encoded seed albumin SCA is represented by slow (SCAS) or fast (SCAF) variant. Most of the accessions possessed either of two marker combinations: 24 had SCAF cox1+ rbcL+ (combination A) and 49 accessions had SCAS cox1rbcL− (combination B), 16 accessions represented 5 of the rest 6 possible combinations. All accessions of P. fulvum and P. abyssinicum had combination A, the overwhelming majority of cultivated forms of P. sativum had combination B while wild representatives of P. sativum had both combinations A and B, as well as rare combinations. This pattern indicates that combination A is the ancestral state in the genus Pisum L., inherited by P. fulvum and P. abyssinicum, while combination B seems to have arisen in some lineage of wild P. sativum which rapidly fixed mutational transitions of the three markers studied, most probably via a bottleneck effect during the Pleistocene. Then this ‘lineage B’ spread over Mediterranean and also gave rise to cultivated forms of P. sativum. Rare combinations may have resulted from occasional crosses between ‘lineage A’ and ‘lineage B’ in nature or during cultivation, or represent intermediate evolutionary lineages. The latter explanation seems relevant for an Egyptian cultivated form ‘Pisum jomardii Schrank’ (SCAF cox1rbcL−) which is here given a subspecies rank. Wild representatives of P. sativum could be subdivided in two subspecies corresponding to ‘lineage A’ and ‘lineage B’ but all available subspecies names seem to belong to lineage B only. Presently all wild forms would better be considered within a fuzzy paraphyletic subspecies P. sativum subsp. elatius (Bieb.) Schmalh. s. l.  相似文献

Variability study of agro-morphological characteristics is important for varietal improvement and plant breeding program. For this, 148 chili pepper landraces (Capsicum annuum L.) collected from Northern Benin were characterized using 39 (21 qualitative and 18 quantitative) morphological characters. The trial was conducted in a Fisher block design with 3 replicates at the experimental station of the Faculty of Science and Technology (FAST) of Dassa based in Central Benin. The results revealed considerable morphological variability between landraces. Twenty nine (16 quantitative and 13 qualitative) variables among the 39 examined were discriminatory and group the landraces into 4 classes when considering the qualitative variables. With quantitative variables, 9 groups were obtained. The correlations between the studied quantitative variables show that the yield parameters are correlated with the variables related to the vegetative part of the plant. This study will help breeders to better select their parents for crossing in order to produce desirable and high yielding varieties.  相似文献

Squash (Cucurbita spp.) is a common component in traditional cropping systems in Mexico, mainly in the agroecosystem known as the “milpa”, in which squash is cultivated in association with maize (Zea mays), the main crop. Using a questionnaire, 80 farmers were interviewed about crop production and selection practices in order to understand how these factors affect genetic diversity of local squash populations. We found that the most of the farmers who cultivate squash were elderly 59.8 ± 14.5 (mean ± SD; n = 78) years old. Squash varieties in the area were exclusively locally adapted landraces, and had not been replaced by modern squash cultivars. Two cultivated squash species, C. argyrosperma ssp. argyrosperma and C. moschata, had been grown intercropped with maize by 97.5% of the interviewed farmers, but only 50.0% were still producing squash at the time of the study. Farmers recognize typical characteristics of particular varieties within each of the local cultivated squash species, and selection is directed to maintain their identity. Nearly two thirds of the farmers (62.0%) had exchanged seeds of squash for planting, a practice that serves to increase genetic variability in the populations. All of the interviewed farmers were conscious of the possible hybridization between the wild gourd (C. argyrosperma ssp. sororia) and their cultivated squash. Despite various natural and human managed factors identified as contributing to enhancement of genetic diversity in these populations, results of the study show that genetic erosion of Cucurbita is likely in the region in the near future.  相似文献

Plant breeders are interested in strawberry species as donors of volatile compounds in breeding programmes because of the diversity and intensity of wild strawberry aroma. Therefore, the topic of this paper is the prospective analysis of four accessions of four wild strawberry accessions in comparison to a standard cultivar of Fragaria × ananassa Duch. by using human sensory, gas chromatography mass spectrometry (GCMS) and gas chromatography–olfactometry (GCO). The wild species have higher aroma intensities compared with the cultivated one. The flavour quality differs significantly. Semiquantitative GC analysis revealed that F. × ananassa cv. ‘Elsanta’ has the lowest content of volatile compounds whereas Fragaria moschata L. ‘Cotta’ has the highest. The aroma impressions, measured by GCO, support the findings of GCMS analyses. The nasal impact frequency (NIF)-profiles of the wild types are more manifold and of higher intensities than those of the cultivated F. × ananassa cv. ‘Elsanta’ which corresponds with the overall flavour impression when tasting the fresh fruits.  相似文献

An overview of ex situ collections of cultivated flax (Linum usitatissimum L.) and 53 other species of the genus Linum L. is presented. The names of 33 genebanks in 23 countries preserving Linum germplasm and the species preserved at these places are provided. World genebanks engage in ex situ preservation of about 48,000 accessions of cultivated flax (Linum usitatissimum). However, possibly only 10,000 accessions of these are unique. Of the ca. 200 species of the genus Linum, other than L. usitatissimum, 53 are reported to be represented in the world’s ex situ collections with a total of about 900 accessions. Of these, 279 accessions alone refer to the wild progenitor of cultivated flax, Linum bienne Mill. However, L. bienne from the region of origin of cultivated flax is rarely represented in genebank collections. References to publications describing the techniques for ex situ conservation and the infraspecific variation of cultivated flax are made. It is suggested that taxonomic species delimitation within the genus Linum is in need of clarification and that the variation in Linum species for ornamental use or use in breeding is investigated.  相似文献

To establish the genetic relationship among Sahelian sorghum [Sorghum bicolor (L.) Moench S.L.] landraces from Burkina Faso were submitted to electrophoretic analysis for 10 enzymatic systems and 18 loci. Four enzymatic systems (ADH, LAP, MDH, PGD) and five loci revealed polymorphism both within and among landraces. Thirty-eight per cent of the landraces were monomorphic in all the 18 loci. The genotypic frequencies in most of the landraces deviated markedly from Hardy–Weinberg proportions due to a major heterozygote deficit, the landrace being homozygous or a mixture of homozygotes. Multivariate analysis yielded three main groups, containing native landraces and five minors, containing introduced cultivars, randomly distributed over the territory. The pattern of allelic occurrence was random and unrelated to external selection pressures. The major diversity among landraces appears to be from genetic shift caused by farmers’ selection of their seeds. It could also be due to the low rates of outcrossing (19%) and migration (0.06) prevailing in the set.  相似文献

Cowpea landraces belonging to Vigna unguiculata (L.) Walp. subsp. unguiculata cv.-gr. unguiculata and cv.-gr. sesquipedalis collected from part of the Deccan Plateau and West Coast of India were evaluated to (i) identify the diverse source(s) of variation for improved characters like pods/peduncle and seed index (ii) study the response of landraces for adaptation to drought and heat stress and (iii) understand the breeding value of a landrace in the genetic improvement of a popular cowpea cultivar. Landraces were evaluated for various morphological characters, pods/peduncle, seed index and other economically important agronomic traits, rust resistance and drought and heat tolerance in different years and environments. Landraces were found as an important source of genetic variability for pods/plant, pods/peduncle, better pod filling ability (seed index), grain yield/plant as well as drought and heat tolerance and rust resistance. Hybridization between C 152 (cv.) and DWDCC 016 (landrace) resulted in release of new variation not present in the two parents. Thus the landrace, DWDCC 016, can be utilized to improve cultivated varieties by transferring to them the economically valuable traits like pods/peduncle and seed index thereby enhancing realisation of sink potential and ultimately grain yield in a sustainable way.  相似文献

Landraces of rice (Oryza sativa L.) are valuable sources of genetic variation that have been lost in advanced cultivars. Seeds of a rice landrace stored for almost 100 years were found on Sado Island in Niigata prefecture, Japan. This report aims to present basic data on the genetic variation of this landrace, which was known as ‘Echizen’. Five samples of ‘Echizen’, consisting of two old samples, one sample maintained on farm, and two lines regenerated from old seeds were compared with other advanced cultivars and landraces using 19 microsatellite markers. Among the five samples of Echizen, the two stored samples showed greater diversity than the other samples. Cluster analysis based on the UPGMA method also showed that old Echizen was a diverse landrace that could cover the genetic diversity of most Japanese rice cultivars.  相似文献
