Pomegranate bacterial blight disease caused by Xanthomonas axonopodis pv. punicae is one of the most serious threat to the pomegranate industry in India. So far, the chemicals used to control this pathogen alone are less effective. In this article, an attempt was made to supplement chemical control through improving the systemic acquired resistance of plant using nutrients such as nitrogen (N) and salicylic acid (SA) as elicitor. The study revealed that foliar application of SA at the rate of 300 ppm increased above-ground dry mass of plant by 64.97%, improved chlorophyll content of leaves and resulted higher concentration of macro-elements and micro-elements, particularly Mn and Zn, in leaves. It also significantly increased the activities of nitrate reductase (NR), superoxide dismutase (SOD), catalase and peroxidase. However, the maximum response of SA was recorded when it was sprayed after soil application of N at 100% of the recommended dose. As a consequence, this combination recorded the least bacterial blight infection with average severity of 11.58% followed by without-N application under challenge inoculation of the pathogen. However, sustainable production cannot be realized without N application. The integration of this strategy for activation of defence mechanism of plant with the bactericide streptocycline resulted in further reduction of bacterial blight disease incidence and severity under field condition to very low levels (3.84% and 18.57% respectively).  相似文献

Bacterial diseases of bean cause economically important losses worldwide. The most important method for managing bacterial diseases on bean is the use of pathogen-free seed. In this study, 198 different dry bean seed samples of six different cultivars including Dermason, Cali, Sira, Battal, Bombay and Seker, were collected from 12 provinces of Central Anatolia Region of Turkey. All were tested to investigate the seedlots as primary inoculum sources of the major bacterial diseases. The data revealed that 22,72 %, 13,63 %, 11,11 %, 1,51 % and 0.5 % of seed samples tested were contaminated with five seedborne bacterial pathogens, Pseudomonas savastanoi pv. phaseolicola (Psp), Pseudomonas syringae pv. syringae (Pss), Xanthomonas axonopodis pv. phaseoli (Xap), X. axonopodis pv. phaseoli var. fuscans (Xapf) and Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff), respectively. All bacterial strains isolated were identified based on morphological, physiological, biochemical, molecular and pathogenicity tests. The results showed that Psp and Pss were found together on cv. Cali; Psp and Xap on cv. Dermason and cv. Sira; and Pss and Xap on cv. Seker, cv. Dermason, and cv. Cali. Therefore, the results in the present study suggested that evaluation and selection of pathogen-free seeds are very important for preventing the spread of pathogens and effective management of seed borne bacterial diseases prevalent in bean growing regions; in addition to implementation of integrated crop production strategies such as crop rotation, sanitation, seed treatment, tolerant/resistant cultivar selection and proper bactericide application.  相似文献

Xanthomonas axonopodis pv. manihotis (Xam) is the causal agent of cassava bacterial blight (CBB) disease. CBB is a major constraint to cassava cultivation in Ghana. In this study, a survey was conducted in eight regions of Ghana to assess the presence of CBB disease. Out of the eight regions visited, CBB, though at different prevalence, was observed in five regions. Cassava plants samples showing suspected bacterial blight symptoms were collected for analysis by Polymerase Chain Reaction (PCR). The results of the analysis showed that Ashanti region had the highest prevalence in percentage of CBB, which recorded (70%), followed by Volta region (60%); Brong Ahafo region (40%); Eastern region (40%) and Greater Accra region (20%). Morphological examination of the putative pathogen was carried out on Cefazolin trehalose agar (CTA) and Nutrient agar (NA) media. The isolates were subjected to conventional PCR using Xanthomonas genus specific primer, RST2/RST3, Xam specific Variable Number Tandem Repeat (VNTRs) loci, XaG1_67F/R and X-gumD primers, which produced 840, 446 and 402 bp, respectively. The isolates also tested positive with SYBR Green fluorescent dye, using Real-time PCR. The resulting PCR products were sequenced and analyzed using a BLASTn program, which revealed homology between 93 and 100% with several other Xam strains retrieved from GenBank nucleotide database. The pathogenicity test of the isolates on the susceptible Esam cassava variety produced symptoms typical of Xam and the pathogen was consistently re-isolated from the inoculated cassava plants and thereby satisfying the Koch’s postulates.  相似文献

Bacterial leaf/fruit spot and canker of stone fruits, caused by Xanthomonas arboricola pv. pruni, is a recurrent disease in Italy. A set of 23 strains has been isolated in peach and plum orchards in an intensively stone fruit cultivated area located in north-eastern Italy. They were all identified as X. arboricola pv. pruni by means of phytopathological and serological features: hypersensitive reaction on bean pods, pathogenicity test on immature peach or plum fruitlets, identification by immunofluorescence assay and conventional PCR. Phylogenetic analysis based on sequencing of the gyrB housekeeping gene of the isolates showed that they formed a unique clade, well characterised and separated from other xanthomonads. An insight into the genetic population features was attempted by rep-PCR analysis, using the ERIC, REP and BOX primers. The combined rep-PCR fingerprints showed a slight intra-pathovar variation within our isolates, which grouped in five close clusters. Copper resistance has been assessed in vitro for our whole X. arboricola pv. pruni collection, highlighting that two isolates show a level of resistance in vitro up to 200 ppm of copper. Nonetheless, the copLAB gene cluster, present in many other species of Xanthomonads, was not detected in any isolate, confirming the presence of a still unknown mechanism of copper detoxification in our Xanthomonads arboricola pv. pruni tolerant/resistant strains.  相似文献

Bacterial blight (BB) of rice caused by Xanthomonas oryzae pv. oryzae (Xoo), remains a major production constraint in rice cultivation especially in irrigated and rainfed lowland ecosystems in India. The pathogen is highly dynamic in nature and knowledge on pathotype composition among the Xoo population is imperative for designing a scientific resistance breeding program. In this study, four hundred isolates of Xoo collected from diverse rice growing regions of India were analyzed for their virulence and genetic composition. Virulence profiling was carried out on a set of differentials consisting of 22 near isogenic lines (NILs) of IR24 possessing different BB resistance genes and their combinations along with the checks. It was observed that different NILs possessing single BB resistance gene were susceptible to about 59–94% of the Xoo isolates except IRBB 13 (containing BB resistance gene xa13), which showed susceptibility to about 35% of the isolates. Based on the reaction of the Xoo isolates on the differentials, they were categorized into 22 pathotypes. Among the 22 pathotypes, IXoPt-1 and IXoPt-2 were least virulent and IXoPt # 18–22 were highly virulent. Pathotype IXoPt-19 which was virulent on all single BB resistance genes except xa13 constituted the major pathotype (22.5% isolates) and was widely distributed throughout India (16 states). This was followed by pathotype IXoPt-22 (17.25%) which was virulent on all the NILs possessing single BB resistance genes. Molecular analysis was carried out using two outwardly directed primers complementary to sequence of IS1112, a repetitive element of Xoo. A high level of genetic polymorphism was detected among these isolates and the isolates were grouped into 12 major clusters. The data indicated complex nature of evolution of the Xoo pathotypes and there was no strong correlation between pathotypes and genetic clusters as each genetic cluster was composed of Xoo isolates belonging to different pathotypes. The study indicated that none of the single BB resistance genes can provide broad spectrum resistance in India. However, two-gene combinations like xa5 + xa13 and different 3 or 4 genes combination like Xa4 + xa5 + xa13, Xa4 + xa13 + Xa21, xa5 + xa13 + Xa21 and Xa4 + xa5 + xa13 + Xa21 are broadly effective throughout India.  相似文献

Rhizoctonia solani, a devastating soil borne fungus inciting banded leaf and sheath blight (BLSB) disease is a constraint in maize production and improvement program. Rhizoctonia isolates collected from seven diverse maize cropping zones of India were examined for morphological and molecular variability. All the tested isolates caused symptoms of BLSB on maize and were also cross infective on rice and sugarcane hosts, but showed significant variability in hyphal diameter, mean hyphal cell size, weight, size and distribution of scleorotia, culture pigmentation, incubation period, pathogenicity and expression of symptoms. Neighbour joining cluster analysis placed the 62 isolates of R. solani into four major groups, A, B, C and D. Group A was more diverse and included isolates of diverse agro-ecological zones. The cluster analysis corresponded well with principle component analysis. Pathogenicity testing of R. solani isolates on maize genotype (CM 501) revealed highly variable virulence pattern of the pathogen population suggesting its high evolutionary potential, and hence adaptability to diverse geographical regions. The study reveals a strong evidence of inherent potential of the R. solani isolates to survive in diverse ecological zones and its probable spread to other maize cultivars across India. Sequence comparisons of the internal transcribed sequence-ribosomal DNA region of 62 isolates did not reveal much diversity among the isolates. Majority of the isolates (n?=?61) clustered together with anastomosis group (AG) AG1-IA used as reference strain in the phylogram, distinct from AG1-IB, AG2–2IIIB and Waitea circinata used as reference strains. BLSB isolates representing distinct geographical locations shared identical sequences indicating long-distance dispersal of the pathogen. The study confirms that the genetic flexibility of the pathogen allows for its adaptation to variable ecological niches and long-distance introduction of new genotypes into the region. The study emphasizes that epidemiological studies may complement the molecular studies.  相似文献

Whole genome sequencing of a copper resistant (CuR) black rot strain of Xanthomonas campestris pv. campestris (Xcc) isolated from a broccoli plant in Trinidad revealed a unique operon for copper resistance. The cop genes of strain Xcc-BrA1 were determined to be present on a 160 to 180 kb plasmid shown to be non-conjugative with other xanthomonads. While nucleotide comparison of a putative 8.0 Kbp copLABMGF gene cluster identified in Xcc-BrA1 genome did not reveal any homologous region with other known CuR Xanthomonas strains from diverse origins, the comparison of the translated amino acid sequence indicated similarity with X. citri, X. c. pv. citrumelonis and X. vesicatoria Cop proteins. Cloning of the copLAB gene cluster from Xcc-BrA1 conferred copper resistance to other copper-sensitive xanthomonads. Although Xcc-BrA1 harbors copLAB genes with similar sizes and organization and is able to grow on Cu-amended medium as other CuR xanthomonads, the phylogenetic analysis of nucleotide sequences indicates that the cop cluster in Xcc-BrA1 is unique and distantly related to other copLAB genes from Xanthomonas and Stenotrophomonas. The origin of copper resistance genes in Xcc-BrA1 is likely a result of horizontal gene acquisition from a still unknown phylloplane cohabitant. The findings of this study have implications for the management of crop diseases caused by CuR xanthomonads. Future studies could focus on and determining the distribution, overall importance and appropriate control measures for strains harbouring these unique genes.  相似文献

In 1972, bacterial leaf spot of onion (BLSO) was first recorded in Japan by Goto. The pathogen was considered as a pathovar of Pseudomonas syringae specifically causing disease on onion and Welsh onion, but it has not been taxonomically investigated in detail. In 2012 and 2014, a disease suspected as BLSO re-emerged on onion in Shizuoka and Hyogo Prefectures, Japan, respectively. A pathogenic bacterium isolated from the infected onions was thought to be the BLSO agent after preliminary examinations. Strains isolated from BLSO in 1969, 1986, 1987, 2012 and 2014 were characterized and compared with the causal agent of bacterial blight of leek (P. syringae pv. porri), which causes similar symptoms on Allium plants. The result of rep-PCR distinguished the BLSO agent from P. syringae pv. porri. Multilocus sequence analysis on housekeeping genes and hrp genes encoding the type-III secretion system revealed that the strains of the BLSO agent clustered independently of P. syringae pv. porri. The BLSO agent and P. syringae pv. porri also differed in utilization of erythritol, dl-homoserine, glutaric acid and other bacteriological characteristics and caused different reactions on onion, Welsh onions, chives, shallot, rakkyo, leek, garlic and Chinese chive. Thus, the BLSO agent clearly differs from P. syringae pv. porri and is considered to be a new pathovar of P. syringae. Common bacterial blight (CBB) caused by Xanthomonas axonopodis pv. phaseoli (Xap) is the main bacterial disease of snap bean. The present work aimed to select snap bean genotypes that are resistant to CBB based on three components of resistance: area under the disease progress curve, latent period, and lesion diameter on pods (DL). A completely randomized two-factor factorial design with six replications was used to evaluate leaves and pods of 14 snap bean and three common bean genotypes (PI 207262, BAC-6 and A-794) with high resistance to CBB. Two Xap isolates, 1394-98 and 775-90, were used to inoculate leaves and pods. Data were analyzed using nonparametric statistics. Significant differences were observed among the evaluated genotypes for all of the components of resistance, except for DL. The snap bean UENF 1482 demonstrated good performance in two disease resistance components. For this reason, this genotype can be recommended for use in breeding programs that aim to generate snap bean genotypes resistant to Xanthomonas axonopodis pv. phaseoli.  相似文献

Xanthomonas oryzae pv. oryzae, the causal agent of bacterial blight in rice, has developed resistant strains due to rampant usage of chemical pesticides and disease resistant rice cultivars. Virulence attenuating agents could be an alternative strategy to combat this pathogen; therefore, the main objective of the present study was to screen phenolic compounds of plant origin as well as a thiadiazole derivative, acibenzolar S methyl for their effect on various virulence determinants of Xanthomonas oryzae pv. oryzae such as the type three secretion system, motility, extracellular polysaccharide production and secretion of exoenzymes through the type two secretion system. From the compounds screened, cinnamic acid and its derivatives like ferulic acid, caffiec acid and p-coumaric acid showed significant reduction in promoter activity of xopQ, an effector secreted through type three secretion system. None of the compounds used in this study showed substantial inhibitory effect on other virulence determinants indicating that their effect was specific on the type three secretion system except acibenzolar S methyl which, along with inhibitory effects on promoter activity of xopQ, also showed reduction in motility of bacteria. Wheat-bran-derived natural medium was effective for induction of type three and type two secretion systems at detectable levels. We also performed virulence assay that showed significant reduction in disease symptoms confirming cinnamic acid, ferulic acid, caffeic acid, p-coumaric acid as effective virulence attenuating agents of Xanthomonas oryzae pv. oryzae BXO43. These studies demonstrate the potential of selected small molecules for development of new alternative strategies to control the bacterial blight of rice.  相似文献

Plant volatile compounds, including terpenes, are known to be involved in the rice defense system. In the present analysis of a terpene synthase, OsTPS18, in rice, we found that OsTPS18 was localized in the cytoplasm and synthesized the sesquiterpenes (E)-nerolidol and (E)-β-farnesene. The amounts of (E)-nerolidol and (E)-β-farnesene increased after jasmonic acid (JA) treatment. (E)-Nerolidol had significant antibacterial activity against Xanthomonas oryzae pv. oryzae (Xoo). These results suggest that (E)-nerolidol plays an important role in JA-induced resistance against Xoo and that it functions as an antibacterial compound in rice.  相似文献

Plant response to one type of stress can be affected by simultaneous exposure to a second stress, for example when abiotic and biotic stresses occur together. Ten rice genotypes comprising those with bacterial blight (BB) resistance (R) genes, drought quantitative trait loci (QTLs) plus a BB R gene, and BB susceptible genotypes, were subjected to mild and moderate drought stress and plants were inoculated with two Xoo strains (PXO99 and PXO145) to simulate the challenges rice crops face under simultaneous stress of drought and BB. Plant height and dry shoot biomass were significantly reduced by drought stress treatments. The BB disease lesion lengths varied according to rice genotypes and PXO99 Xoo multiplication and spread in planta was higher compared to that of PXO145, which generally decreased under mild drought stress. Rice genotype IRBB7 (Xa7) showed less Xoo spread and a reduced Xoo multiplication under drought stress compared to the well-watered control with PXO145. In contrast, in genotypes with a different BB R gene and/or drought QTLs [IRBB4 (Xa4), IR87705–6-9-B (Xa4 + qDYT 2.2 ), IR87707–445-B-B-B (Xa4 + qDYT 2.2  + qDYT 4.1 ) and IR87707–446-B-B-B (Xa4 + qDYT 2.2  + qDYT 4.1 )], Xoo multiplication and spread in planta was higher with drought stress. This study has shown that drought stress affected rice response to the BB pathogen and the response varied according to the rice genotype. It is concluded that evaluating rice varieties under combined abiotic and biotic stresses will be the best strategy to determine biotic stress resistance durability under climate change.  相似文献

The virulence spectrum of 300 isolates of Xanthomonas oryzae pv. oryzae (Xoo), representing 17 districts of Punjab, Pakistan was elucidated through inoculation on a set of six rice IRRI-differentials. The virulence level was assessed by using principal component and cluster analysis. Among six principal components (PCs), PC-1 exhibited 59.3 % of the total variance. The highly virulent isolates clusters on the positive side of the ordination away from the point of intersection of PC1 and PC2 and classifies the Xoo isolates from slow disease to the highest disease causing entities. The 300 isolates were categorized into 29 pathotypes (Pt1-29) wherein the highly virulent pathotype (Pt-1), comprises of 39 Xoo isolates were widespread in 12 districts. The majority of Xoo isolates were moderately to least virulent (21.7–43 %) and average disease progress curves confirmed the field reactions of these pathotype clusters for an efficient recognition of Xoo isolates. Interaction of the pathogen with differentials harboring different resistant genes was well investigated in the current study for future management approaches for which the surveillance of the new Xoo pathotypes may expedite the disease resistant rice breeding programme in the country.  相似文献

This study evaluated the role of oxidative stress on the expression of Pht cluster genes involved in phaseolotoxin synthesis in Pseudomonas syringae pv. phaseolicola. Results demonstrate that the expression of Pht cluster genes is regulated by oxidative stress in a manner dependent of the ROS present in the cell. The presence of H2O2 and Paraquat, influences on the expression of the Pht cluster genes in function of the compound and of the concentration evaluated, demonstrating that expression of Pht genes is part of the oxidative stress response in P. syringae pv. phaseolicola NPS3121.  相似文献

The plant pathogenic oomycete, Phytophthora infestans, is the causal agent of late blight disease in tomato and potato. For characterizing Egyptian P. infestans isolates by DNA marker analysis, 40 isolates of P. infestans were collected from different locations in Egypt during two growing seasons (2012/2013 and 2013/2014). The 40 isolates were grouped into seven genotypes, in which 24 alleles were detected. The identified genotypes were not completely associated with geographic location and sample collection years. These results provide genetic and geographical information for developing a program to manage late blight disease.  相似文献

General studies of the bacterial pathogenXanthomonas campestris have revealed many classes of gene which are essential for pathogenicity to cruciferous plants. ‘Pathogenicity genes’ include structural genes encoding extracellular plant-degrading enzymes, genes required for the export (or secretion) of the enzymes from the bacterial cell, several independent sets of regulatory genes, a cluster ofhrp genes, and several genes of unknown function. Plants respond to infection by synthesizing characteristic pathogenesis-related proteins. The spectrum of proteins produced depends on the genotype of the challenging bacterium.Arabidopsis is susceptible to infection byXanthomonas, which should make it feasible to analyze plant resistance and response by molucular genetic techniques.  相似文献

Olive knot disease in Japan was first reported in Shizuoka Prefecture in 2014, and the causal agent was identified as Pseudomonas savastanoi pv. savastanoi. Subsequently, olive trees having knots were also found in Aichi and Kanagawa Prefectures in 2015, and the isolates from knots were also suspected to be P. savastanoi pv. savastanoi through preliminary examinations. Therefore, the Aichi and Kanagawa isolates were identified through comparison of isolates from three prefectures. Phylogenic analysis based on 16S rDNA and housekeeping genes (gyrB, rpoD, gltA and gap1) revealed that the isolates belonged to the same cluster as the pathotype strain, ICMP4352PT. The iaaM, H and L genes, which are involved in promotion of symptoms, and the ina gene coding the ice nucleation protein, were detected by PCR from all the isolates. In rep-PCR (ERIC and REP) analyses, the isolates yielded DNA fragment-banding patterns that were nearly identical to that of ICMP4352PT, but slight variations in banding patterns were observed among them. In a pathogenicity test, the isolates formed distinct knots on olive and pink jasmine. Phenotypic properties of the isolates were almost identical to those of ICMP4352PT, with the exception of d-sorbitol utilization. Consequently, Aichi and Kanagawa isolates from olive were identified as P. savastanoi pv. savastanoi, and several genetic diversities in terms of rep-PCR were found in the Japanese population of P. savastanoi pv. savastanoi, indicating their heterogeneity.  相似文献

Bacterial pathogens of onion (Allium cepa) plants and their undetected presence in seed can cause substantial losses to onion producers. In this study, 23 Pseudomonas syringae strains were isolated from five onion plants and 18 onion seeds. The symptoms on leaves and seed stalks were irregular lesions with necrotic centres and water soaked margins. The aim of the study was to characterize these P. syringae strains using Biolog GN III carbon source utilization, multilocus sequence typing (MLST) based on partial sequences of four housekeeping genes (cts, gapA, gyrB and rpoD), and to determine whether or not the strains were pathogenic on onion (cv. Granex 33), chive (Allium schoenoprasum cv. Grasiue), leek (Allium porrum cv. Giant Italian) and spring onion (Allium fistulosum cv. Salotte) plants. Both Biolog analysis and MLST analysis separated onion strains into two clusters, one supporting the existence of a new pathovar of P. syringae, and the other corresponding to P. syringae pv. porri. Pseudomonas syringae strains belonging to the new pathovar we pathogenic only on onion plants of the Allium spp. tested. The results of this study revealed that bacterial blight of onion in South Africa is caused by two pathovars of P. syringae sensu lato, namely, the newly described pathovar, allii, and P. syringae pv. porri. The symptoms caused by these two pathovars in the field were indistinguishable.  相似文献

Bacterial spot disease of stone fruits, caused by Xanthomonas arboricola pv. pruni, is of high economic importance in the major stone-fruit-producing areas worldwide. A better understanding of disease epidemiology can be valuable in developing disease management strategies. The effects of weather variables (temperature and wet/dry period) on epiphytic growth of X. arboricola pv. pruni on Prunus leaves were analyzed, and the relationship between inoculum density and temperature on disease development was determined and modeled. The information generated in this study, performed under controlled environmental conditions, will be useful to develop a forecasting system for X. arboricola pv. pruni. Optimal temperature for growth of epiphytic populations ranged from 20 to 30 °C under leaf wetness. In contrast, multiplication of epiphytic populations was not only interrupted under low relative humidity (RH) (< 40%) at 25 °C, but also resulted in cell inactivation, with only 0.001% initial cells recovered after 72 h incubation. A significant effect of inoculum density on disease severity was observed and 106 CFU/ml was determined as the minimal infective dose for X. arboricola pv. pruni on Prunus. Infections occurred at temperatures from 15 to 35 °C, but incubation at 25 and 30 °C gave the shortest incubation periods (7.7 and 5.9 days respectively). A model for predicting disease symptom development was generated and successfully evaluated, based on the relationship between disease severity and the accumulated heat expressed in cumulative degree day (CDD). Incubation periods of 150, 175 and 280 CDD were required for 5, 10 and 50% of disease severity, respectively.  相似文献
