AIM To explore the anti-atherosclerotic mechanism of Wendan decoction based on formation of foam cells. METHODS The optimal concentrations of Wendan decoction without cytotoxity to cells were selected by MTT assay. After Wendan decoction treatment, the formation of foam cells was examined by oil red O staining. The cholesterol efflux, cholesterol level, free cholesterol level and cholesterol esterification rate were analyzed using cholesterol efflux assay, total cholesterol assay and free cholesterol assay. The expression levels of macrophage membrane proteins, including CD36, scavenger receptor class A (SR-A), ATP-binding cassette transporter A1 (ABCA1) and scavenger receptor class B type I (SR-BI), were quantified by Western blot. RESULTS The optimal concentrations of Wendan decoction without cytotoxity to the cells were 0~6 g/L. Wendan decoction at the concentrations of 1.5, 3 and 6 g/L were selected for the experiments. Wendan decoction at these concentrations inhibited the formation of foam cells induced by oxidized low-density lipoprotein (ox-LDL), and reduced the accumulation of intracellular lipids in a concentration-dependent manner (P<0.05 or P<0.01). Wendan decoction also reduced intracellular total cholesterol level, cholesterol ester level and cholesterol esterification rate (P<0.05 or P<0.01), promoted efflux of intracellular cholesterol (P<0.01), and decreased the protein level of CD36 in THP-1 cell-derived macrophages (P<0.01) in a concentration-dependent manner. Wendan decoction at the concentration of 6 g/L significantly reduced the protein level of SR-A in THP-1 cell-derived macrophages (P<0.05). At the concentrations of 3 and 6 g/L, Wendan decoction significantly increased the protein levels of ABCA1 and SR-BI in THP-1 cell-derived macrophages (P<0.05 or P<0.01). CONCLUSION Wendan decoction significantly inhibits ox-LDL-induced formation of foam cells by reducing cholesterol deposition and promoting cholesterol efflux, and its mechanism may be related to the down-regulation of CD36 and SR-A and the up-regulation of ABCA1 and SR-BI.  相似文献

AIM To explore the effect of continuous renal replacement therapy (CRRT) on mRNA expression of autophagy-related molecules and the prognosis in the patients with acute kidney injury (AKI). METHODS A total of 174 patients from our hospital who were diagnosed to have AKI and underwent CRRT between February 2015 and March 2018 were involved in this study. The plasma levels of interleukin-1β (IL-1β) and interleukin-6 (IL-6), the serum creatinine (SCr) level, and the mRNA expression levels of autophagy-related molecules, including microtubule-associated protein 1 light chain 3-II (LC3-II), autophagy-related protein 5 (Atg5) and beclin-1, in the monocytes from peripheral blood were compared before and after CRRT. According to the survival of AKI patients after 4 weeks of CRRT, the enrolled patients were divided into death group (n=43) and survival group (n=131), and the mRNA expression levels of the above molecules were compared between the 2 groups. RESULTS After CRRT treatment, the plasma levels of IL-1β and IL-6, the level of SCr, and the mRNA expression levels of LC3-II, Atg5 and beclin-1 in the monocytes were significantly lower than those before CRRT treatment (P<0.05). The mRNA expression levels of LC3-II, Atg5 and beclin-1 in death group were significantly higher than those in survival group (P<0.05). The positive correlation between SCr and IL-1β, IL-6, LC3-II or beclin-1 was observed (P<0.05), and no correlation between SCr and Atg5 was found (P>0.05). CONCLUSION CRRT decreases the mRNA expression levels of autophagy-related molecules in the patients with AKI and reduces the autophagy activity, which is protective for the patients. These autophagy-related molecules may be applied as a potential markers to predict the prognosis of CRRT.  相似文献

AIMTo investigate the role of soluble Klotho protein in THP-1-derived foam cell formation. METHODSTHP-1 monocytes were induced into macrophages by treatment with 160 nmol/L phorbol myristate acetate for 48 h, and then were divided into 6 groups: negative control group (THP-1-derived macrophages), positive control group [THP-1-derived foam cells induced by oxidized low-density lipoprotein (ox-LDL) for 48 h], and 25, 50, 100 and 200 μg/L soluble Klotho protein groups (THP-1-derived macrophages pretreated with soluble Klotho protein at the indicat?ed concentraions for 2 h and then induced by ox-LDL for 48 h). Lipid droplets in cytoplasm were observed by oil red O staining. The cholesterol outflow rate was detected by scintillation counting technique. The content of intracellular total cholesterol, free cholesterol and cholesterol ester was detected by enzyme fluorescence analysis. The expression of acyl-coenzyme A:cholesterol acyltransferase 1 (ACAT1) and ATP-binding cassette transport?er A1 (ABCA1) at mRNA and protein levels was determined by RT-qPCR and Western blot, respectively. RESULTSOil red O staining and lipid mass quantification showed that THP-1-derived foam cell formation was dose-dependently suppressed by soluble Klotho protein. The cholesterol efflux rate of THP-1-derived foam cells was increased by soluble Klotho protein in a dose-dependent manner (P<0.05). In addition, soluble Klotho protein decreased the expression of ACAT1 and increased the expression of ABCA1 in a dose-dependent manner (P<0.05). CONCLUSION The soluble Klotho protein inhibits THP-1-derived foam cell formation in a dose-dependent manner by down-regulating the expression of ACAT1 and up-regulating the expression of ABCA1.  相似文献

AIM To investigate the effect of β1-adrenergic receptor autoantibodies (β1-AA) on the rhythm of autophagy marker microtubule-associated protein 1 light chain 3 (LC3), and the underlying mechanism of cardiomyocyte death. METHODS The test materials were Sprague-Dawley (SD) rats and H9c2 rat cardiomyocytes. The SD rats were randomly divided into immunization group and control group with 6 rats in each group. The H9c2 cells were randomly divided into control group, β1-AA group, lentivirus (LV)-NC group, and LV-shPer2 group (n=6). Affinity chromatography was used for purification of β1-AA from rat serum. CCK-8 assay was used to observe the viability of cardiomyocytes treated with β1-AA for 24 h. The cells were synchronized by dexamethasone and then treated with β1-AA. The mRNA and protein levels of LC3 at different time points were determined by real-time PCR and Western blot, respectively. The Per2 protein level at different time points was also determined by by Western blot. JTK_CYCLE algorithm was used to estimate the circadian rhythm parameters. After destruction of LC3 circadian rhythm via LV-shPer2, CCK-8 assay was used to measure the viability of H9c2 cells. RESULTS High level of β1-AA in rat serum was found after active immunization compared with control group (P<0.05). The viability of H9c2 cells in β1-AA group was significantly lower than that in control group (P<0.05). The LC3 and Per2 rhythms were both disrupted in H9c2 cells induced by β1-AA (JTK_CYCLE P<0.05). After LV-shPer2 infection, the LC3 rhythm was disrupted (JTK_CYCLE P<0.05) and the cell viability was reduced (P<0.05). CONCLUSION β1-AA may induce the destruction of autophagy marker LC3 rhythm in rat cardiomyocytes and then promote cell death.  相似文献

AIMTo investigate the effects of calpain-2 and autophagy-related protein 5 (Atg5) on apoptosis of BRL-3A rat normal liver cells during endoplasmic reticulum stress (ERS) induced by dithiothreitol (DTT). METH?ODS: BRL-3A cells were treated with DTT at 2.0 mmol/L for 0, 6, 12 and 24 h to induce ERS. Real-time cell analysis (RTCA) was used to measure the effect of DTT on BRL-3A cell proliferation. Apoptosis and cell cycle distribution were analyzed by flow cytometry. The mRNA expression of calpain-2 and Atg5 was detected by real-time PCR. The protein levels of calpain-2, Atg5, Atg7, Atg12 and microtubule-associated protein 1 light chain 3 (LC3) were determined by Western blot. The interaction between calpain-2 and Atg5 was investigated by co-immunoprecipitation (Co-IP). RESULTSThe proliferation of BRL-3A cells treated with DTT was significantly inhibited. The apoptosis of BRL-3A cells was significantly increased after DTT treatment for 6, 12 and 24 h as compared with 0 h group (P<0.05). The cell cycle was arrested in G1 phase after DTT treatment (P<0.05). After DTT treatment for 6, 12 and 24 h, the mRNA expression of calpain-2 and Atg5 in the BRL-3A cells was significantly increased as compared with 0 h group (P<0.05). The protein levels of calpain-2, Atg12 and Atg7 in the cells treated with DTT for 6, 12 and 24 h were significantly higher than those in 0 h group, and the ratio of LC3-II/LC3-I was also significantly higher than that in 0 h group, while Atg5 expression was significantly lower than that in 0 h group (P<0.05). The results of Co-IP found that the anti-calpain-2 antibody precipitated Atg5 protein from the cell lysates, and the anti-Atg5 antibody also precipitated calpain-2 from the cell lysates, which confirmed the interaction between calpain-2 and Atg5. CONCLUSION Calpain-2 may participate in ERS-induced hepatocyte apoptosis by interacting with Atg5.  相似文献

AIM To investigate the regulatory effect of retinoic acid X receptor (RXR) on autophagy induced by hypoxia/reoxygenation (H/R) in rat alveolar type Ⅱ epithelial cells (AEC Ⅱ) and its molecular mechanism. METHODS AEC Ⅱ were cultured in normoxia. The cells growing to logarithmic growth phase were randomly divided into 5 groups: (1) control (Con) group: cells were cultured for 30 h under normal operation; (2) H/R group: cells were cultured in hypoxia condition for 6 h and then in reoxygenation condition for 24 h; (3) DMSO group: cells were pretreated 1.5 h with medium containing less than 0.1% DMSO before modeling, and the rest were treated the same as the H/R group; (4) 9-cis-retinoic acid (9-RA) group: cells were pretreated for 1 h with 9-RA (100 nmol/L) before hypoxia; (5) HX531 group: cells were treated with 9-RA (100 nmol/L) for 0.5 h, then treatment with HX531 (2.5 μmol/L) for 1 h. CCK-8 assay was used to detect the cell viability. Immunofluorescence staining was used to observe the expression of RXRα. Transmission electron microscope was used to observe the changes of intracellular ultrastructure, and the mRNA expression of adenosine AMP-activated protein kinase (AMPK), beclin 1, LC3, mammalian target of rapamycin (mTOR) and P62 was detected by RT-PCR. Western blot was used to detected the protein levels of p-AMPK, beclin 1, LC3-Ⅱ, p-mTOR and P62. RESULTS Compared with Con group, the cell viability in H/R, DMSO, 9-RA and HX531 groups were significantly decreased. The mRNA expression of AMPK, beclin 1 and LC3 was significantly increased, and the protein levels of p-AMPK, beclin 1 and LC3-Ⅱ were also increased. The mRNA expression of mTOR and P62 was decreased, and the protein levels of p-mTOR and P62 were also decreased (P<0.05). The cell injury in 9-RA group was alleviated and autophagy level was significantly lower than that in H/R, DMSO and HX531 groups (P<0.05), and no significant difference among H/R, DMSO and HX531 groups was observed (P>0.05). CONCLUSION H/R induces autophagy of AEC Ⅱ. Activating RXR reduce the damage of AEC Ⅱ cells induced by H/R, and its mechanism may be related to the inhibition of autophagy.  相似文献

AIM To investigate the effect of niflumic acid (NFA) on human glioma U87 cells and to clarify the potential mechanism. METHODS The U87 cells were cultured in vitro and divided into blank control group, and 50, 100 and 200 μmol/L NFA groups. MTT assay was performed to determine the viability of cells in various groups. Migration and invasion abilities were measured by real-time cell analysis (RTCA). RESULTS The results of MTT assay showed that compared with blank control group, the viability of U87 cells was increased after treatment with NFA for 12 h (P<0.05 or P<0.01), while the viability was significantly decreased after treatment with NFA for 24 and 48 h (P<0.05 or P<0.01) in a concentration-dependent manner. The results of RTCA showed that compared with control group, the cell migration and invasion abilities were inhibited in 100 and 200 μmol/L NFA groups (P<0.05 or P<0.01) and the inhibitory effects were more obvious in 200 μmol/L NFA group (P<0.01). CONCLUSION NFA inhibits the viability, migration and invasion of human glioma U87 cells.  相似文献

AIM To investigate the mechanism of long noncoding RNA (lncRNA) FEZF1-AS1 regulating microRNA-363-3p (miR-363-3p) on the viability and apoptosis of lipopolysaocharide (LPS)-induced vascular endothelial cells. METHODS Human umbilical vein endothelial cells (HUVECs) were cultured in vitro. pcDNA-NC, pcDNA-FEZF1-AS1, anti-miR-NC, anti-miR-363-3p, miR-NC and miR-363-3p mimics were transfected into the HUVECs and LPS stimulation was applied for 24 h. RT-qPCR was used to detect the expression of FEZF1-AS1 and miR-363-3p. The cell viability was measured by MTT assay. The apoptotic rate was analyzed by flow cytometry. The dual-luciferase reporter experiment was used to verify the targeted regulation of FEZF1-AS1 and miR-363-3p. Western blot was used to determined the expression of cyclin D1, Ki67 and cleaved caspase-3. RESULTS Compared with control group, the expression level of FEZF1-AS1 in LPS group was significantly reduced (P<0.05), and the expression level of miR-363-3p was significantly increased (P<0.05). Compared with pcDNA-NC+LPS group, the cell viability in pcDNA-FEZF1-AS1+LPS group was significantly increased (P<0.05), the apoptotic rate was significantly reduced (P<0.05), the protein levels of cyclin D1 and Ki67 were significantly increased (P<0.05), and the protein level of cleaved caspase-3 was significantly reduced (P<0.05). Compared with anti-miR-NC+LPS group, the cell viability in anti-miR-363-3p+LPS group was significantly increased (P<0.05), the apoptotic rate was significantly reduced (P<0.05), the protein levels of cyclin D1 and Ki67 were significantly increased (P<0.05), and the protein level of cleaved caspase-3 was significantly reduced (P<0.05). Dual-luciferase reporter experiment confirmed that FEZF1-AS1 targeted miR-363-3p. Compared with miR-NC+pcDNA-FEZF1-AS1+LPS group, the cell viability in miR-363-3p+pcDNA-FEZF1-AS1+LPS group was significantly reduced (P<0.05), the apoptotic rate was significantly increased (P<0.05), the protein levels of cyclin D1 and Ki67 were significantly reduced (P<0.05), and the protein level of cleaved caspase-3 was significantly increased (P<0.05). CONCLUSION Over-expression of FEZF1-AS1 promotes the viability and inhibits apoptosis of LPS induced vascular endothelial cells by inhibiting the expression of miR-363-3p.  相似文献

AIM To investigate the expression of histone chaperone anti-silencing function 1B (ASF1B) in prostate cancer cells and its effect on cell viability in vitro. METHODS Human prostate cancer PC-3 cells were used, and knockdown of ASF1B was conducted by small interfering RNA (siRNA) transfection into the cells. The cells were divided into control group, siRNA negative control vector (mock) group and siRNA-ASF1B group. The viability of the PC-3 cells treated with ASF1B-siRNA for 12, 24 and 48 h was measured by MTT assay. Flow cytometry was used to analyze cell apoptosis and cell cycle distribution. The mRNA expression of apoptosis-related molecules was detected by RT-qPCR, and the expression levels of MAPK/JNK/ERK signaling pathway-related proteins were determined by Western blot. RESULTS The protein level of ASF1B in the normal cells (benign prostatic hyperplasia) was significantly lower than that in the PC-3 cells (P<0.01). Compared with control group and mock group, the protein expression level of ASF1B in the PC-3 cells transfected with siRNA-ASF1B plasmid and the viability of the PC-3 cells were significantly decreased (P<0.01). The apoptotic rate of the PC-3 cells transfected with siRNA-ASF1B plasmid was significantly higher than that in control group (P<0.01), and the cell cycle was arrested at G1 phase. The mRNA levels of p53, caspase-3, Bax and PARP-1 in the PC-3 cells transfected with siRNA-ASF1B plasmid were up-regulated compared with those in control group and Mack group (P<0.01). In addition, the protein levels of MAP2K4 and p-JNK in the PC-3 cells in siRNA-ASF1B group were significantly higher than those in mock group (P<0.01), while the protein level of p-ERK was significantly lower than that in mock group (P<0.01). CONCLUSION ASF1B silencing induces G1 arrest and promotes apoptosis of PC-3 cells. Activating MAPK/JNK/ERK signaling pathway may be a possible contributor to the anti-prostate cancer effect of siRNA-ASF1B.  相似文献

AIM To investigate the effect of microRNA-22 (miR-22) secreted by macrophage exosomes on the autophagy of H9c2 cardiomyocytes under uremic toxin stimulation. METHODS The macrophage-derived exosomes stimulated by indoxyl sulfate (IS) were collected and co-cultured with H9c2 cells. The levels of miR-22 in the macrophages, macrophage-derived exosomes and H9c2 cells were detected by RT-qPCR. The viability of H9c2 cells was measured by CCK-8 assay. The expression of exosome surface marker protein CD63 and autophagy-related proteins LC3 and P62 was determined by Western blot. RESULTS Under IS stimulation, the expression of exosome surface marker protein CD63 in the macrophages was significantly higher than that in control group (P<0.05), and the levels of miR-22 in the macrophages and macrophage-derived exosomes were significantly increased (P<0.01). With the increase in macrophage exosome concentration, the viability of H9c2 cells was decreased gradually (P<0.05), and the stimulation of macrophage exosomes reduced P62 expression and promoted the conversion of LC3-I to LC3-II in a dose-dependent manner (P<0.05). Macrophage-derived exosomes increased the ratio of LC3-II to LC3-I but decreased P62 protein expression in the H9c2 cells transfected with miR-22 mimic compared with the cells transfected with corresponding negative control miRNAs (P<0.05). However, miR-22 inhibitor yielded contrasting results. CONCLUSION IS-stimulated macrophages increase expression of miR-22 in cardiomyocytes through exosomes, and promote autophagy of the cardiomyocytes.  相似文献

AIM To establish a suitable cell model for the study of ovarian function through comparing the isolation and primary culture effect of human ovarian mural granulosa cells (MGCs) and cumulus cells (CCs). METHODS The follicular fluid of 16 patients who underwent assisted reproductive technique and their cumulus oocyte complexes (n=223) were collected. Density gradient centrifugation was used to isolate the MGCs and the methods of mechanical cutting plus enzyme hydrolysis were used to isolate the CCs. The cell counts and survival rates were analyzed by trypan blue staining and the expression of follicle stimulating hormone receptor (FSHR) was analyzed by flow cytometry to identify the purity. The expression of microtubule-associated protein 1 light chain 3 (LC3), P62 and Bax at mRNA and protein levels was determined by qPCR and Western blot, respectively. RESULTS There had less isolation time, higher survival rate (P<0.05) and better tractility in vitro of CCs compared with MGCs. The results of flow cytometry showed that the FSHR expression of CCs and MGCs after isolation was (92.23±2.66)% and (81.33±6.57)%, respectively, with significant differences (P<0.05). The mRNA level of LC3 in CCs was significantly lower than that in MGCs (P<0.01), and the mRNA level of Bax was significantly higher than that in MGCs (P<0.05). There was no significant difference in P62 mRNA expression between CCS and MGCs(P>0.05). The difference of protein expressions of these molecules in the 2 kinds of cells were consistent with that in mRNA. CONCLUSION Mechanical cutting method plus enzyme hydrolysis is a simple way to isolate the CCs, with high purity and good cellular state in vitro, which can be used as a cell model for ovarian function research.  相似文献

AIM To explore the anti-atherosclerotic mechanism of Wendan decoction based on reverse cholesterol transport. METHODS Eight-week-old apolipoprotein E gene knockout (ApoE-/-) mice with high-fat diet and daily drug gavage were randomly divided into model group, simvastatin group, and low-, middle- and high-dose Wendan decoction groups, with 15 mice in each group. The C57BL/6 mice of the same age served as control group. The mice were weighed once every week. After 10 weeks, the mice were anesthetized with chloral hydrate. The serum were collected for lipid level examination. The atherosclerotic plaque buildup in aortic root and whole aorta was observed by HE staining and oil red O staining, respectively. The levels of proteins related to cholesterol transport, ATP-binding cassette transporter A1 (ABCA1) and caveolin-1 in the aorta, and scavenger receptor class B type I (SR-BI) and CD36 in the liver, were quantified by Western blot. RESULTS Wendan decoction at middle dose inhibited the increase in the body weight of ApoE-/- mice fed with high-fat diet (P<0.05). Wendan decoction at different doses significantly reduced the serum levels of triglyceride, total cholesterol and low-density lipoprotein cholesterol in the ApoE-/- mice (P<0.05 or P<0.01), but had no effect on serum high-density lipoprotein cholesterol level (P>0.05). Wendan decoction at different doses inhibited the formation of atherosclerotic plaques in whole aorta of the ApoE-/- mice (P<0.05 or P<0.01). Middle- and high-dose Wendan decoction significantly inhibited the formation of atherosclerotic plaques in the aortic root (P<0.05). Bedsides, Wendan decoction at different doses increased the protein level of ABCA1 and decreased the protein level of caveolin-1 in the aorta of the ApoE-/- mice (P<0.01). Middle- and high-dose Wendan decoction increased the liver protein level of SR-BI in the ApoE-/- mice (P<0.01). However, Wendan decoction at different doses had no effect on the liver protein level of CD36 in the ApoE-/- mice (P>0.05). CONCLUSION Wendan decoction reduces the body weight, serum lipid levels and formation of atherosclerotic plaques in ApoE-/- mice fed with high-fat diet, and its mechanism is related to up-regulation of ABCA1 protein level in the aorta and SR-BI protein level in the liver as well as down-regulation of caveolin-1 protein level in the aorta.  相似文献

AIM To investigate the expression of Wnt1 and LGR5 in gastric mucosa with stress ulcer after traumatic brain injury (TBI) in rats, and to study the relationship between Wnt1/LGR5 expression and stress ulcer after TBI. METHODS Healthy 7-week-old male SD rats (n=30) were randomly divided into 3 groups: sham operation (sham) group, mild TBI (mTBI) group, and severe TBI (sTBI) group. The mTBI and sTBI were induced by electronic cortical contusion impactor. Neurological severity score (NSS) was calculated 24 and 48 h after modeling. Mucosal blood flow in gastric fundus, greater curvature, pylorus and cardia of anesthetized rats 48 h after injury was measured by Doppler flowmeter. All rats were sacrificed, and their gastric tissues were harvested after 48 h and then stained with hematoxylin and eosin. The protein expression of Wnt1 and LGR5 was analyzed by Western blot and immunohistochemistry. RESULTS Compared with sham group, the NSS in mTBI group and sTBI group was significantly increased (P<0.01). Compared with sham group, the gastric mucosal blood flow of the rats after TBI was significantly decreased (P<0.01), and the decrease in sTBI group was more significant than that in mTBI group (P<0.05). The protein expression of Wnt1 and LGR5 in mTBI group was significantly higher than that in sham group (P<0.05), and that in sTBI group was significantly higher than that in mTBI group (P<0.05). Normal glandular gastric tissue was observed, and no abnormal change was found in sham group, while the infiltration of inflammatory cells in gastric lamina propria, mucosa and submucosa was obvious in mTBI group and sTBI group. CONCLUSION Traumatic brain injury activates Wnt1 and LGR5 expression to induce inflammatory changes in gastric mucosa, and then induces stress ulcer. This results provides experimental basis for the pathogenesis and treatment of stress ulcer after trauma.  相似文献

AIM To investigate the effect of 27nt-miRNA (27nt-miR) on apoptosis of human umbilical vein endothelial cells (HUVECs) induced by oxidized low-density lipoprotein (Ox-LDL) and its underlying mechanism. METHODS HUVECs were cultured in vitro and grouped as below: normal control group, Ox-LDL group, 27nt-miR+Ox-LDL group, anti-27nt-miR+Ox-LDL group and negative control+Ox-LDL group. The cells in Ox-LDL group were treated with Ox-LDL at 40 mg/L for 48 h, while those in normal control group were untreated but cultured normally. The cells in 27nt-miR+Ox-LDL group, anti-27nt-miR+Ox-LDL group and negative control+Ox-LDL group were transfected with their corresponding lentiviral vectors under the same procedure, followed by treatment with Ox-LDL at 40 mg/L for 48 h to induce apoptosis. The cell viability was measured by CCK-8 assay. The migration capacity was detected by scratch assay. The caspase-3 activity was measured by caspase-3 activity assay kit. The apoptotic rate was analyzed by Hoechst 33258 and flow cytometry. The mRNA and protein expression levels of Bcl-2, Bax and caspase-3 were determined by RT-qPCR and Western blot. RESUITS: Compared with negative control+Ox-LDL group, the cell viability and migration ability were significantly decreased by over-expression of 27nt-miR in the HUVECs (P<0.05), while the activity of caspase-3 and apoptosis induced by Ox-LDL were significantly increased (P<0.05). Furthermore, the mRNA and protein expression levels of Bax and caspase-3 were significantly up-regulated (P<0.05), and the mRNA and protein expression level of Bcl-2 was down-regulated in 27nt-miR+Ox-LDL group (P<0.05). Meanwhile, all the above indexes showed an opposite tendency in anti-27nt-miR+Ox-LDL group. CONCLUSION 27nt-miR promotes Ox-LDL-induced apoptosis and inhibits the viability and migration of HUVECs in vitro, possibly through regulating the expression of apoptotic/anti-apoptotic proteins such as Bax,caspase-3 and Bcl-2.  相似文献

AIM To observe the effect of Chaihu-Shugan decoction (CHSGD) on atherosclerosis in spontaneously hypertensive rats (SHR) and its possible mechanism. METHODS The male SHR (n=50) were randomly divided into model group (gavage of normal saline), compound kendir leaves (CKL) group (gavage of 0.5 g/kg CKL), and low-, medium- and high-dose CHSGD (CHSGD-L, CHSGD-M and CHSGD-H) groups (gavage of 2.5, 5 and 10 g/kg CHSGD, respectively), and another 10 male Wistar rats of the same origin were selected as normal control (NC) group (gavage of normal saline). The blood pressure was measured by intelligent noninvasive sphygmomanometer. The levels of blood lipids were measured by automatic biochemical analyzer. The expression of oxidative stress-related indexes, nitric oxide (NO), malondialdehyde (MDA) and superoxide dismutase (SOD), were detected by colorimetry. HE staining was used to detect the degree of atherosclerosis, and Western blot was used to detect the expression of Rho-associated kinase (ROCK)/c-Jun N-terminal kinase (JNK) signaling pathway-related proteins, RhoA, ROCK1 and JNK. RESULTS After 4 weeks of treatment, compared with NC group, the blood pressure, the serum levels of triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and MDA, and the protein expression of RhoA, ROCK1 and JNK in aortic tissues of the rats in model group were significantly increased (P<0.05), and the serum levels of high-density lipoprotein cholesterol, NO and SOD were significantly decreased (P<0.05). HE staining showed that the diameter of aortas in the rats was thickened, a large number of foam cells were formed under the endothelium, and the proliferation of smooth muscle cells was observed. Compared with model group, the blood pressure, the serum levels of TG, TC, LDL-C and MDA, and the protein expression of RhoA, ROCK1 and JNK in aortic tissues of the rats in CKL, CHSGD-L, CHSGD-M and CHSGD-H groups were significantly decreased (P<0.05), and the serum levels of NO and SOD were significantly increased (P<0.05). HE staining showed that the structure of each layer of rat aortas gradually returned to normal, the vascular cells were in good order, and the inflammatory cell infiltration was slight. Compared with CKL group, the blood pressure, the serum levels of TG, TC, LDL-C and MDA, and the protein expression of RhoA, ROCK1 and JNK in aortic tissues of the rats in CHSGD-L and CHSGD-M groups were significantly increased (P<0.05), and the serum levels of NO and SOD were significantly decreased (P<0.05). No significant difference of the above indexes between CHSGD-H group and CKL group was observed (P>0.05). CONCLUSION Chaihu-Shugan decoction may attenuate the oxidative stress response via inhibition of ROCK/JNK signaling pathway, thus alleviating the symptoms of atherosclerosis in SHR.  相似文献

AIM To investigate the alleviating effect of exenatide (Exe), a glucagon-like peptide-1 (GLP-1) receptor agonist, on the ectopic lipid accumulation in skeletal muscle of ob/ob mice and its mechanism. METHODS Eight-week-old male ob/ob mice and their wild-type (WT) littermates were randomly divided into 3 groups, ob/ob group, ob/ob+Exe group and WT group, and treated with Exe at 24 nmol/kg or the same volume of saline intraperitoneally once daily for 4 weeks. The body weight, fasting blood glucose (FBG) and fat content were measured after the 4-week treatment. The oil red O staining and the quantification of triglyceride (TG) were performed on the skeletal muscle. The serum levels of TG, total cholesterol and free fatty acid (FFA) were also measured by ELISA. The expression levels of AMP-activated protein kinase (AMPK) and lipid metabolism-related proteins were determined by Western blot. Mouse myoblast C2C12 cells were used as an in vitro model to further investigate the effects of Exe. RESULTS As compared with the ob/ob mice treated with saline, 4-week Exe treatment did not reduce body weight, FBG, food intake and fat content in ob/ob mice (P>0.05). However, serum FFA was decreased (P<0.05). Oil red O staining and the quantification of TG showed that 4-week Exe treatment significantly attenuated the ectopic lipid accumulation in the skeletal muscle of ob/ob mice (P<0.05). The results of Western blot showed that the levels of phosphorylated AMPK (p-AMPK) and lipolysis-related proteins were up-regulated, while the lipid synthesis-related proteins were down-regulated by Exe (P<0.05). Treatment with Exe alleviated the lipid accumulation in the C2C12 cells induced by sodium palmate (P<0.05), and the effects of Exe on the levels of p-AMPK and lipid metabolism-related proteins in the C2C12 cells were consistent with those in the ob/ob mice (P<0.05). Treatment with Exe also up-regulated the protein expression of glucose transporter 4 and improved the ability of glucose uptake in the C2C12 cells (P<0.05). CONCLUSION Short-term Exe treatment attenuates the ectopic lipid accumulation in skeletal muscle of ob/ob mice by up-regulating lipolysis-related proteins and down-regulating lipid synthesis-related proteins, which is independent on body weight loss.  相似文献

AIM To investigate the relationship between the expression level of galectin-3 and the stability of plaque structure in human atherosclerotic plaques. METHODS The coronary specimens from autopsy cases (n=84) were collected. Among them, 22 cases had coronary atherosclerotic lesions without sudden death of coronary heart disease (A1 group), 20 cases were sudden death of coronary heart disease without secondary lesions (A2 group), 24 cases were sudden death of coronary heart disease with secondary lesions (A3 group), and 18 cases without heart disease were used as normal control group (control group). The intimal thickness, necrotic lesion thickness, fibrous cap thickness and the degree of lumen stenosis were measured by routine HE staining in all coronary arteries. The foam cells in the lesion were marked by CD68 and counted. The expression of galectin-3, CD68 and matrix metalloproteinase-2 (MMP-2) in coronary artery intima was detected by immunohistochemical staining, Western blot and RT-qPCR. The correlation between above factors and the structural stability of atherosclerotic plaques was also analyzed. RESULTS Compared with control group, the intima and necrotic lesions were thickened, the fiber cap was thinned, and the degree of lumen stenosis were increased in A1~3 groups (P<0.05). The number of foam cells in the atherosclerotic focus was increased (P<0.05). The protein and mRNA levels of galectin-3, CD68 and MMP-2 in the lesions showed an increasing trend from normal group to A1~3 groups (P<0.05). The expression of galectin-3, CD68 and MMP-2 in atherosclerotic lesions was positively correlated with intimal thickness and necrotic lesion thickness, and negatively correlated with fibrous cap thickness. CONCLUSION The expression of galectin-3 in human coronary atherosclerotic lesions is increased, which is related to the stability of atherosclerotic plaques.  相似文献

AIM To study the effect of pterostilbene (PTE) on autophagy and SIRT1-FoxO signaling pathway in cervical cancer cells. METHODS Human cervical cancer HeLa cells were used as the study objects. CCK-8 assay and flow cytometry were used to determine the effects of PTE at different concentrations on the viability and apoptotic rate of HeLa cells. The mRNA expression of SIRT1 and FoxO in the cells was detected by qPCR. The number of autophagosomes in the cells was observed under transmission electron microscope.The protein levels of SIRT1, FoxO, LC3-Ⅰ,LC3-Ⅱ, p62, Bax and Bcl-2 in the cells were determined by Western blot. RESULTS After treatment with PTE for 24 and 48 h, the viability inhibitory rate of the HeLa cells was increased with increasing PTE concentration. Compared with 0 μmol/L PTE, after 15, 30 and 60 μmol/L PTE treatment, the apoptotic rate of the HeLa cells, the number of autophagosomes in the cells, and the protein levels of Bax, LC3-Ⅱ/LC3-Ⅰ, SIRT1 and FoxO were increased (P<0.05), while the protein levels of Bcl-2 and p62 were decreased (P<0.05) in a concentration-dependent manner. CONCLUSION PTE may induce autophagy and apoptosis of HeLa cells by activating SIRT1-FoxO signaling pathway, and inhibit HeLa cell viability.  相似文献

AIM To explore the effect of dasatinib on the viability, apoptosis and migration of human renal carcinoma cell lines 786-O and 769-P, as well as the molecular mechanism in vitro. METHODS 786-O cells and 769-P cells were treated with different concentrations (0~2 μmol/L) of dasatinib, and 0 μmol/L dasatinib was used as blank control group. MTT method was used to detect cell viability. Wound healing assay was used to detect the effect of dasatinib on migration. Hoechst 33258 staining was used to observe the effect of dasatinib on apoptosis. Flow cytometry was used to detect the effect of dasatinib on cell cycle. Western blot method was used to detected cell cycle- and apoptosis-related protein levels. RESULTS Dasatinib inhibited viability and migration of 786-O and 769-P cells, and the inhibitory effect of dasatinib increased with the concentration of dasatinib (P<0.05). The IC50 values of dasatinib against 786-O and 769-P cell lines were (0.958 7±0.028 8) μmol/L and (0.784 3±0.066 0) μmol/L, respectively. After treatment with dasatinib for 24 h, the expression of pro-apoptotic proteins cleaved caspase-3 and cleaved caspase-9 increased significantly (P<0.01), while the expression of cyclin D1 decreased (P<0.05). The cycle-related pathway proteins p53 and p21 increased (P<0.05), while the level of p-AKT was decreased (P<0.05). CONCLUSION Dasatinib impaired the viability and migration ability of human renal carcinoma cell lines 786-O and 769-P by up-regulating p53 expression and down-regulating AKT phosphorylation.  相似文献
