To determine the effects of BCS at parturition and postpartum lipid supplementation on blood metabolite and hormone concentrations, 3-yr-old Angus x Gelbvieh beef cows, which were nutritionally managed to achieve a BCS of 4 +/- 0.07 (479.3 +/- 36.3 kg of BW) or 6 +/- 0.07 (579.6 +/- 53.1 kg of BW) at parturition, were used in a 2-yr experiment (n = 36/yr). Beginning at 3 d postpartum, cows within each BCS were assigned randomly to be fed hay and a low-fat control supplement or lipid supplements with either cracked high-linoleate or high-oleate safflower seeds until d 61 of lactation. The diets were formulated to be isonitrogenous and isocaloric, and the safflower seed supplements were formulated to achieve 5% DMI as fat. On d 31 and 61 of lactation, blood samples were collected preprandially and then hourly postprandially (at 0, 1, 2, 3, and 4 h). Serum insulin (P = 0.27) and glucose (P = 0.64) were not affected by BCS at parturition. The mean concentrations of plasma NEFA (P = 0.08) and beta-hydroxybutyrate (P = 0.08) tended to be greater, and serum IGF-I was greater (P < 0.001) in BCS 6 than BCS 4 cows. Conversely, serum GH was greater (P = 0.003) for BCS 4 cows, indicating that regulation of IGF by GH may have been uncoupled in BCS 4 cows. The postpartum diet did not affect NEFA (P = 0.94), glucose (P = 0.15), IGF-I (P = 0.33), or GH (P = 0.62) concentrations. Oleate-supplemented cows had greater (P = 0.03) serum insulin concentrations, whereas control cows had greater (P = 0.01) plasma beta-hydroxybutyrate concentrations. Concentrations of NEFA (P = 0.05) and glucose (P < 0.001) were greater, and beta-hydroxybutyrate tended (P = 0.07), to be greater at d 3, whereas serum IGF-I was greater (P = 0.003) at d 6 of lactation. Similar concentrations of NEFA, glucose, GH, and IGF-I indicate that the nutritional status of beef cows during early lactation was not influenced by lipid supplementation. However, perturbations of the somatotropic axis in BCS 4 cows indicate that the influence of energy balance and BCS of the cow at parturition on postpartum performance should be considered when making managerial decisions.  相似文献

Percutaneous liver biopsies and blood samples were obtained from 80 dairy cows in nine Michigan herds over the peripartum period. Thirty-nine cows were fed 17 g of supplemental inositol and 41 were fed a placebo. Liver biopsies were assayed for total myoinositol and triglyceride (TG) concentrations. Blood samples were assayed for serum dextran precipitable cholesterol, nonesterified fatty acids (NEFA), insulin, thyroxine (T4), free (FT4), triiodothyronine (T3) and free T3 (FT3) concentrations. Serum concentrations of insulin and the thyroid hormones decreased near parturition, with lowest concentrations occurring in the immediate postpartum period. Concentrations of T3 correlated well with T4, and the concentrations of free thyroid hormones reflected concentrations of total thyroid hormones. The percentage of hormone in the free fraction remained constant over time. Serum insulin, T3 and T4 were negatively correlated with serum NEFA and liver TG concentrations. Thyroid hormone concentrations were positively correlated with serum dextran precipitable cholesterol concentrations. Inositol supplementation was associated with reduced circulating T3 and FT3 concentrations, but not T4 and FT4 concentrations. Changes in hormone concentrations at parturition and their relationship to liver TG and serum NEFA concentrations were consistent with a metabolic adaptation by the dairy cow to the negative energy balance of early lactation.  相似文献

We determined the effects of short-term fasting and refeeding on temporal changes in plasma concentrations of leptin, insulin, insulin-like growth factor- 1 (IGF-1), growth hormone (GH), glucose, and nonesterified fatty acids (NEFA), in early lactating cows, non-lactating pregnant cows, and postpubertal heifers. In experiment 1, Holstein cows in early lactation were either fed ad libitum (Control, n=5) or feed deprived for 48 h (Fasted, n=6). Plasma leptin, insulin, and glucose concentrations rapidly declined (P<0.05) within 6h, and IGF-1 by 12h, but all these variables sharply returned to control levels (P>0.10) within 2h of refeeding. Plasma NEFA and GH concentrations were elevated (P<0.05) by 4 and 36 h of fasting and returned to control levels (P>0.10) by 8 and 24h after refeeding, respectively. In experiment 2, four ruminally cannulated pregnant non-lactating Holstein cows were used in a cross-over design and were fasted for 48 h (Fasted) or fasted with partial evacuation of rumen contents (Fasted-Evac). The plasma variables measured did not differ (P>0.10) between Fasted and Fasted-Evac cows. Plasma leptin, insulin, and IGF-1 concentrations were reduced by 10, 6, and 24h of fasting, respectively, in Fasted-Evac cows; and these variables were reduced by 24h in Fasted cows (P<0.05). Plasma glucose levels were reduced (P<0.05) by 48 h of fasting in both groups of fasted animals. Plasma NEFA and GH levels were increased (P<0.05) by 12 and 48 h of fasting, respectively. In experiment 3, postpubertal Holstein heifers were either fed ad libitum (Control, n=4) or feed deprived for 72 h (Fasted, n=5). Concentrations of leptin, insulin, IGF-1, and glucose in plasma were reduced (P<0.05) by 24, 10, 24, and 48 h of fasting, respectively. Plasma NEFA concentrations increased (P<0.05) by 4h, of fasting while GH levels were not significantly (P>0.10) affected by fasting. Collectively, our data provide evidence that plasma leptin concentrations are reduced with short-term fasting and rebound on refeeding in dairy cattle with the response dependent on the physiological state of the animals. Compared to the rapid induction of hypoleptinemia with fasting of early lactation cows, the fasting-induced hypoleptinemia was delayed in non-lactating cows and postpubertal heifers.  相似文献

Leptin may play a role in the endocrine-metabolic processes that guarantee the physiological course of lactation in dairy cattle. This study was aimed at determining the changes in plasma concentrations of leptin and some of the main hormones and metabolites involved in the lactogenetic process in high-yielding dairy cows throughout lactation; we also wanted to assess whether leptin secretion is subjected to seasonal influences. Blood samples were collected from 23 Italian Friesian dairy cows from the end of a lactation to the ninth month of the subsequent one; in addition, blood was sampled from 47 dairy cows in different phases of lactation during February and July. Plasma concentrations of leptin, growth hormone (GH), insulin, prolactin (PRL), glucose, non-esterified fatty acids (NEFA) and urea were quantified by either validated radioimmunoassay (RIA) or enzymatic colorimetric methods. At the beginning of lactation, GH concentrations significantly increased, while a significant reduction occurred in leptin and insulin. This endocrine condition, such as the significant increase in NEFA plasma concentrations, is indicative of a marked lipid mobilization. In the more advanced stages of lactation, when both energy and protein balances become positive, leptin plasma concentrations increased, whereas GH and NEFA concentrations declined. During the summer months, a significant increase in leptin plasma concentrations, irrespective of the phase of lactation, was observed. Collectively, our findings suggest that, in dairy cows, leptin may represent a 'metabolic signal' of animal's status of fattening and nutritional level; in addition, leptin seems to be influenced by photoperiod and environmental temperature.  相似文献

The impact of closely controlled energy management during the dry period upon general peripartum metabolism and the subsequent lactation was evaluated in multiparous dairy cows. Eight lactating, pregnant Holstein cows were paired according to current milk production and body condition score and assigned to either the control or the stair-step compensatory nutrition (SSCN) regimen 16 weeks prior to expected calving date. Control cows were fed according to National Research Council [National Research Council., 2001. Nutrient Requirements of Dairy Cattle. 7th rev. ed. Natl. Acad. Sci., Washington, DC.] recommendations and for ad libitum intake during late lactation and the far-off dry period. The SSCN cows were subjected to dietary energy restriction [80% of net energy for lactation requirements (NEL)] during late lactation in order to maintain body weight, followed by realimentation (130% of NEL) during the far-off dry period to induce a compensatory response. A common diet was fed thereafter. The SSCN cows gained less body condition during late lactation and tended to gain more body condition during the far-off dry and transition periods than control cows. There was no difference in dry matter intake during the prepartum transition period and early lactation. Milk yield was not affected by treatment. Serum nonesterified fatty acids (NEFA) were not altered by dietary treatment during late lactation and the dry period; however NEFA were lower in SSCN cows at week 4 of the subsequent lactation. Serum insulin was higher in SSCN cows 8 weeks prior to parturition (end of restriction), tended to be higher at 4 weeks (end of realimentation) before calving, and was also higher at week 12 of early lactation. An SSCN regimen stimulated body condition gain of mature dairy cows during the dry period, did not affect periparturient nutrient metabolism, and led to modest improvements in metabolic parameters later in the subsequent lactation.  相似文献

Background

The present study hypothesized that GH-AluI and IGF-I-SnabI polymorphisms do change the metabolic/endocrine profiles in Holstein cows during the transition period, which in turn are associated with productive and reproductive parameters.

Methods

Holstein cows (Farm 1, primiparous cows, n = 110, and Farm 2, multiparous cows, n = 76) under grazing conditions were selected and GH and IGF-I genotypes were determined. Blood samples for metabolic/endocrine determinations were taken during the transition period and early lactation in both farms. Data was analyzed by farm using a repeated measures analyses including GH and IGF-I genotypes, days and interactions as fixed effects, sire and cow as random effects and calving date as covariate.

Results and Discussion

Frequencies of GH and IGF-I alleles were L:0.84, V:0.16 and A:0.60, B:0.40, respectively. The GH genotype was not associated with productive or reproductive variables, but interaction with days affected FCM yield in multiparous (farm 2) cows (LL yielded more than LV cows) in early lactation. The GH genotype affected NEFA and IGF-I concentrations in farm 1 (LV had higher NEFA and lower IGF-I than LL cows) suggesting a better energy status of LL cows.There was no effect of IGF-I genotype on productive variables, but a trend was found for FCM in farm 2 (AB cows yielded more than AA cows). IGF-I genotype affected calving first service interval in farm 1, and the interaction with days tended to affect FCM yield (AB cows had a shorter interval and yielded more FCM than BB cows). IGF-I genotype affected BHB, NEFA, and insulin concentrations in farm 1: primiparous BB cows had lower NEFA and BHB and higher insulin concentrations. In farm 2, there was no effect of IGF-I genotype, but there was an interaction with days on IGF-I concentration, suggesting a greater uncoupling somatropic axis in AB and BB than AA cows, being in accordance with greater FCM yield in AB cows.

Conclusion

The GH and IGF-I genotypes had no substantial effect on productive parameters, although IGF-I genotype affected calving-first service interval in primiparous cows. Besides, these genotypes may modify the endocrine/metabolic profiles of the transition dairy cow under grazing conditions.  相似文献

The present study examined the Serum 3-methylhistidine concentrations and energy-associated variables of 5 healthy Holstein cows and 5 Holstein cows with ketosis. The serum total cholesterol and apolipoprotein B-100 concentrations and lecithin-cholesterol acyltransferase (LCAT) activity of the ketotic cows were lower than those of the healthy cows 14 days before parturition. The serum non-esterified fatty acid (NEFA) concentration on the day of parturition and 3-methylhistidine concentration 14 days after parturition were higher in the ketotic cows. The serum 3-methylhistidine concentration 14 days after parturition was negatively correlated with the serum LCAT activity 14 days before parturition and was positively correlated with the serum NEFA concentration on the day of parturition. Insufficiency of cholesterol metabolism and acceleration of body fat degradation occur before parturition in cows with ketosis, and these characteristics are correlated with acceleration of protein degradation after parturition.  相似文献

试验旨在揭示围产期奶牛血清代谢物和相关激素水平的动态变化规律,选取临床健康经产奶牛 20 头,分别于产前 14、7 d 及产后 0、7、14 d 采血并分离血清,检测血清葡萄糖、甘油三酯、非酯化脂肪酸、β-羟丁酸等代谢物浓度和胰岛素、生长激素、脂联素和瘦素等激素水平,并根据相应公式计算胰岛素敏感指数。结果表明:奶牛分娩当天的血糖浓度较高,而后逐步恢复至产前水平;奶牛分娩后甘油三酯浓度下降,非酯化脂肪酸和β-羟丁酸浓度升高;胰岛素浓度在产前 14 d 至产后 7 d 各时间点无显著变化;产前 7 d 时的生长激素浓度高于其他时间点(P<0.01);脂联素浓度在整个试验期无显著变化;瘦素水平在产前 7 d 和产后 0 d升高(P<0.01),但产后 7 d 时下降(P<0.01);胰岛素敏感性指数在分娩当天显著降低,随后逐步恢复。结果表明,奶牛产后体脂动员明显,可能与生长激素、瘦素等激素的调控有关。  相似文献

We investigated the influence of rumen-protected choline (RPC) supplementation on milk production, lipid metabolism and vitamin E status in dairy cows receiving a silage-based diet. Twenty-six Italian Holstein multiparous cows were assigned by weight and average production in the previous lactation, to one of two groups: control (no RPC supplementation) and RPC (supplemented with 20 g/day rumen-protected choline chloride). Treatment began 14 days before expected calving and continued for 30 days after parturition. Choline administration significantly increased milk production during the first month of lactation and also the concentration (and total secretion) of choline in milk, but did not affect fat or protein concentrations in milk, or plasma levels of glucose, beta-hydroxybutyrate, cholesterol and non-esterified fatty acids (NEFA). However, around parturition, NEFA concentrations in plasma were lower in treated animals than in controls, suggesting improved lipid metabolism as a result of choline supplementation. Choline supplementation also increased alpha-tocopherol plasma concentrations, suggesting a novel aspect in dairy cows.  相似文献

The periparturient period of dairy cows is characterized by intense lipid mobilization from adipose tissue leading to increased plasma concentrations of nonesterified fatty acids (NEFA). High NEFA are a predisposing factor for inflammatory based diseases. A major component of these diseases is uncontrolled macrophage/monocyte inflammatory responses. Changes in the endocrine activity of adipose tissue during the periparturient period could impact macrophage function by modifying the secretion of adipokines including adiponectin. Currently, the effects of adiponectin on monocyte activation in dairy cattle are unknown. In humans and rodents, this adipokine regulates monocyte phenotype and alterations in its plasma levels are linked with the development of inflammatory diseases. The objectives of this study were to establish associations between plasma adiponectin expression dynamics and different markers of lipid mobilization during the periparturient period of dairy cows and to characterize the effects of adiponectin on the inflammatory response of bovine monocytes. Plasma adiponectin, NEFA, BHB, albumin, and subcutaneous and retroperitoneal fat depots depth were measured during the periparturient period of dairy cows. In vitro, bovine monocytes were cultured with adiponectin to assess changes in pro-inflammatory responses following LPS stimulation. Results from this study demonstrate that alterations in plasma adiponectin levels in periparturient cattle are inversely correlated with the concentrations of plasma NEFA, an important marker of lipid mobilization. Furthermore, adiponectin exposure significantly decreased monocyte expression of TNFα after LPS stimulation thus markedly reducing their inflammatory response. Reduced plasma adiponectin during the periparturient period could predispose dairy cows to the development of uncontrolled monocyte inflammatory responses.  相似文献

The effects of energy density in the diet [low = 0.86 SFU/kg dry matter (DM) or high = 1.06 SFU/kg DM] and daily milking frequency (two or three times) in early lactation on plasma concentrations of metabolites and hormones were evaluated in 40 Holstein dairy cows arranged in a 2 x 2 factorial block design. The four treatment combinations were L2, L3, H2 and H3, and the experimental period comprised the first 8 weeks of lactation. Plasma glucose, insulin and insulin-like growth factor (IGF)-I concentrations were on average 8 (3.43 versus 3.19 mmol/l), 114 (41.6 versus 19.4 pmol/l) and 60% (91.9 versus 57.4 ng/ml) higher, whereas beta-hydroxybutyrate (BOHB), plasma urea nitrogen (PUN) and growth hormone (GH) concentrations were on average 18 (0.73 versus 0.89 mmol/l), 14 (7.18 versus 8.35 mmol/l), and 63% (1.0 versus 2.6 ng/ml) lower for cows fed diet H than for cows fed diet L. Cows milked three times daily had a 6% (3.20 versus 3.42 mmol/l) lower plasma glucose concentration and a 19% (0.88 versus 0.74 mmol/l) higher plasma concentration of BOHB compared with cows milked two times daily. Plasma non-esterified fatty acid (NEFA) concentration was not affected by either treatment. Overall, it is concluded that increasing the daily milking frequency creates a higher metabolic imbalance in early lactation. Cows in early lactation will benefit from receiving a high energy density diet and thereby avoid a too high metabolic imbalance when mobilizing body tissue in support of milk production.  相似文献

Blood indicators are used as a tool to diagnose metabolic disorders. The present work was conducted to study the relationships among blood indicators of lipomobilization and hepatic function in high-yielding dairy cows. Two groups of Holstein cows were studied: 27 early lactation cows and 14 mid lactation cows from four different herds with similar husbandry characteristics in Galicia, Spain. Blood samples were obtained to measure beta-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA), triglycerides (TG), and the activity of aspartate transaminase (AST) and gamma-glutamyl transferase. Cows in early lactation had higher levels of BHB and NEFA than mid lactation cows. High lipomobilization (NEFA > 400 µmol/L) was detected in 67% and 7% of early lactation and mid lactation cows, respectively, while subclinical ketosis (BHB > 1.2 mmol/L) was detected in 41% and 28% of the early lactation and lactation cows, respectively. TG concentrations were low in all cows suffering subclinical ketosis and in 61% of the cows with high lipomobilization. During early lactation, 30% of cows suffered hepatic lipidosis as detected by levels of AST. Compromised hepatic function was observed in early lactation cows as shown by lower concentrations of glucose, total protein, and urea.  相似文献

为了研究甘油对围产期奶牛能量平衡和肝脏糖原合成的影响,选用24头体重(674.6±13.8) kg、胎次(2.5±0.5)、上一泌乳期305 d产奶量(7 412±15) kg和预产期(23.8±0.3) d的经产奶牛,随机分为4组,从分娩前19 d开始分别在基础日粮中添加甘油0,100,200和300 g/d。结果表明,与对照组相比,添加甘油200和300 g/d,显著提高了奶牛分娩后7,21和35 d体况评分,显著改善了奶牛分娩前14 d和分娩后7,21和35 d的能量平衡。200和300 g/d组奶牛分娩前14 d胰岛素浓度和分娩后7,21和35 d奶牛血糖和胰岛素浓度显著高于对照组(P<0.05)。200和300 g/d组奶牛分娩后7,21和35 d血浆游离脂肪酸和β-羟丁酸浓度显著低于对照组和100 g/d组(P<0.05)。200和300 g/d组奶牛分娩后7,21和35 d肝脏糖原含量和糖异生途径限速酶活性显著高于对照组(P<0.05)。结果显示在日粮中添加甘油能够加强肝脏葡萄糖异生作用,改善奶牛能量平衡,适宜添加量为200 g/d。  相似文献

Parasitism in cattle is known to impair growth and development. Recent findings suggest that productivity of adult animals is also affected, but little is known about the physiological mechanisms involved. Furthermore, development of nematode resistance to drugs makes imperative the search of management practices that avoid whole herd treatment. We undertook an epidemiological and endocrine study in a grass based dairy farm in Argentina to study the effect of parasites on milk production and the underlying mechanisms involved, and identify individual animals that would benefit from antiparasitic treatment. All the cows in the dairy were followed monthly for egg parasite output in feces. Samples were cultured for genera determination. Milk production and reproductive results were recorded and periodical bleedings for hormone determination were performed. Nematode egg output (EPG) was maximal in late Summer and Autumn and minimal in Spring in coincidence with the Ostertagia inhibition-disinhibition cycle as this genus had the highest prevalence in all the study. The highest proportion of positive samples was found in the high producing herd and maximal counts were found in the peripartal period. Milk production did not correlate with EPG mean values but, when cows were grouped by EPG positivity around parturition, a significant difference in total milk production between EPG null and positive cows was observed. Positive cows produced 7%, 12% or 15% less milk than null EPG cows, depending on the sampling month/s chosen for classification. The highest difference was seen when both prepartum and postpartum samples were taken into account. No difference in lactation length and a marginal effect on partum to first service interval were encountered. Endocrine studies revealed a decrease in serum growth hormone (GH), type I insulin-like growth factor (IGF-I) and prolactin during lactation in cows with positive EPG in the first postpartum sample with respect to null EPG cows at that time. GH levels decreased and prolactin and IGF-I levels increased in both groups of cows from month 0 to 6 in milk. Serum insulin levels remained stable throughout lactation and were similar in both groups of cows. In conclusion, EPG around parturition may be a useful tool for identifying cows that will have a decrease in productivity due to parasite effects and would possibly benefit from an antiparasitic treatment. Besides, our results suggest that detrimental effect of parasites on milk production may be mediated by GH, IGF-I and prolactin serum levels.  相似文献

Metabolic hormones affect ovarian function in the cow. However, the relationship between metabolic factors and ovarian function is not clear in the postpartum primiparous cow because they are still growing. The aim of the present study was to investigate in detail the time-dependent profile of the metabolic hormones, metabolites, and milk yields of ovulatory and anovulatory primiparous cows during the first follicular wave postpartum. We used 16 primiparous Holstein cows and obtained blood samples for the profiles of metabolites (glucose; non-esterified fatty acid, NEFA; ketone body; total cholesterol; and aspartate aminotransferase), metabolic hormones (growth hormone, GH; insulin-like growth factor-I, IGF-1; and insulin), and progesterone every other day from 1 to 21 days postpartum. In addition, all ovaries were observed using ultrasound. Dairy milk yield was recorded during the experimental period. In all cows, the first follicular wave postpartum was observed and 6 of the cows ovulated. The plasma glucose (P<0.0001) and IGF-1 (P<0.001) concentrations were lower and the plasma NEFA (P<0.0001) and ketone bodies (P<0.0001) concentrations and daily milk yield (P<0.0001) were higher in the anovulatory cows compared to the ovulatory cows. However, the GH levels, which enhance lipolysis for milk production, insulin and other metabolites did not differ between the two groups. In conclusion, the present study suggests that anovulation of the dominant follicle during the first follicular wave postpartum in primiparous cows is induced by low IGF-1 levels that are similar to those of multiparous cows. In addition, anovulatory cows are likely to mobilize body fat stores for milk production more easily than ovulatory cows.  相似文献

BACKGROUND: There is insufficient evidence in the literature concerning the fluctuations and correlations of serum fat-soluble vitamin concentrations during lactation in dairy cows and the potential effect of age of the animals on these values. OBJECTIVES: The objectives of this study were to determine fluctuations in mean serum concentrations of the fat-soluble vitamins A and E and b-carotene and the correlations among them in clinically healthy dairy cows, from the last month of the dry period until the end of lactation, and to investigate whether age was a factor that affects these serum concentrations. METHODS: Forty-five clinically healthy Holstein cows were assigned to 2 groups based on age. Group A consisted of 23 cows 4 years old (mean +/- SD, 3.24 +/- 0.44 years), and group B consisted of 22 cows >4 years old (6.68 +/- 1.66 years). Blood samples for determination of serum beta-carotene, vitamin A, and vitamin E concentrations were collected from each animal just before the start of study (1 month prior to expected parturition), at parturition, and thereafter, at monthly intervals until the end of lactation. RESULTS: There was a significant positive correlation between beta-carotene, vitamin A, and vitamin E values. Mean serum concentrations of beta-carotene, vitamin A, and vitamin E were significantly higher in younger animals. CONCLUSIONS: To our knowledge, this is the first evidence that age is a factor affecting serum concentrations of fat-soluble vitamins in dairy cows.  相似文献

The objective of this study was to evaluate the effects of malic acid (MA) on feed intake, milk yield and composition, blood metabolites and energy balance in early lactation Holstein dairy cows from 1 to 63 day in milk (DIM). Twenty-eight multiparous Holstein dairy cows, blocked by lactation number, previous 305-d mature equivalent milk production, and expected calving date, were arranged into four groups in a randomized block design. Treatments were: control (without MA), LMA, MMA and HMA with 70, 140 and 210 g malic acid per cow per day, respectively. The supplement of food grade MA (99.8% of MA) was hand-mixed into the top one-third of the daily ration. Cows were fed ad libitum a total mixed ration consisting of equal proportion of forage and concentrate. Milk yield increased (P = 0.04), but feed intake and milk components were not affected (P > 0.05) by MA supplementation. The energy balance, expressed as the difference between energy input and output, tended to be higher (P = 0.08) for MA supplemented cows during the 63-DIM period MA and supplemented cows showed a trend (P = 0.07) toward less loss of BW during the 63-day period, especially during the first 21-day of lactation. Concentrations of plasma glucose and serum insulin were higher for cows fed LMA, MMA, and HMA relative to control and linearly (P < 0.01) increased with increasing MA supplementation. Concentrations of non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHBA) and urine ketones were lower for MA-supplemented cows at 7, 14 and 21 DIM of lactation and linearly (P < 0.01) decreased with increasing MA supplementation. Although feed intake was not affected, milk yield increased, MA-supplemented cows experienced higher concentrations of plasma glucose and serum insulin, lower concentrations of plasma BHBA and NEFA, and lower concentrations of urine ketones, suggesting that nutrient digestibilities and energy availability may have been improved.  相似文献

Changes in ACTH challenge test characteristics in dairy cows changing their physiological status at different lactational stages and different feeding levels were not investigated in terms of repeatability yet. In 23 multiparous Holstein cows (10 cows fed a sole fresh herbage diet without concentrate, 13 cows fed with concentrate), three ACTH challenge tests were performed: once during pregnancy shortly prior to drying off ( T1 ), and in week 3 ( T2 ) and 8 ( T3 ) after parturition. Test characteristics were correlated to performance and metabolic parameters: DMI, BW, energy balance (EB), plasma concentrations of free fatty acids (NEFA) and beta‐hydroxybutyrate (BHB). Basal plasma cortisol concentrations were higher at T1 compared with T2 and T3 (p < .05). The adrenal cortex sensitivity (expressed as total AUC (AUCt) of cortisol response after ACTH application) was lowest at T2 compared with T1 and T3 (p < .05). Ranking of the individual animals’ responses was not repeatable between time points of the ACTH tests. Enhancing the energy deficiency during early lactation by omission of concentrate did not affect baseline cortisol concentrations in plasma, but decreased peak height at T2 (p < .05). Baseline plasma cortisol concentrations were positively correlated with cortisol peak values after ACTH application, previous lactation performance, milk yield and BW (p < .05). The AUCt was positively correlated with baseline cortisol concentrations, EB and DMI. Cortisol release after ACTH injection was lower in animals with high plasma concentrations of NEFA, BHB and with higher contents of fat and free fatty acids in milk (p < .05). Cortisol peak height after ACTH administration was higher in cows with a more positive EB, higher DMI and lower plasma concentrations of NEFA and BHB. In summary, cortisol responses to ACTH challenges in this study were not repeatable in dairy cows changing their physiological status.  相似文献

To assess the leptin response to metabolic challenges, three Italian Simmental cows were infused for 6 h: with (a) saline (control); (b) glucose; and (c) amino acid solutions according to a 3 x 3 Latin square experiment. The infusions were carried out at the 36th week of pregnancy, and the second and 12th week of the following lactation. At each of the three infusion periods, blood samples were collected from the jugular vein before and 15, 30, 120, 180, 240, 300 and 360 min after the beginning of each infusion. All samples were analysed for leptin, insulin, glucagon, growth hormone (GH), glucose, non-esterified fatty acids (NEFA) and urea. The physiological phase of the cows significantly affected the basal concentrations of insulin, glucagon, urea and NEFA. The infusion of both glucose and the amino acid solutions did not affect leptin concentrations. Insulin response was significantly increased when animals were infused with the glucose solution and, within treatment, the greatest response was observed at the 12th week of lactation. The greatest glucagon response was observed when infusing the amino acid solution. Urea response to all treatments increased from the dry period to the 12th week of lactation. The GH and NEFA responses were not affected by treatments. The Multi Species radio-immunoassay used in this study showed a lower sensitivity for ruminant leptin which may partially explain the lack of significant leptin variations. However, it can be hypothesized that leptin variations around parturition can be affected by the negative energy balance, and leptin release is not acutely affected by glucose and amino acid availability. In addition, no short-term relationship were found between insulin, glucagon and GH and leptin release in Italian Simmental cows during the dry period and early lactation.  相似文献

The objective of this report is to review the literature on elevated blood concentrations of nonesterified fatty acids (NEFA) before and after parturition in high-yielding dairy cows. It highlights the factors that influence serum NEFA production and their circulation before and after parturition, such as adaptation for nutrient partitioning for fetal needs, onset of lactogenesis, stress of calving and numerous changes in physiological, metabolic, and endocrine status to accommodate parturition and lactogenesis. The role of NEFA in the liver and peripheral tissues and its toxic effects when in excess are discussed. The cow's adaptive physiologic mechanisms to prevent or decrease excessive values of serum NEFA and preventive and therapeutic interventions to enhance these mechanisms are categorized as natural and artificial antidotes respectively. Feeding systems during the dry period and daily exercise or walking activity which may burn excessive NEFA through beta-oxidation in the muscles are considered as more useful antidotes to managing the NEFA metabolism. This will minimize accumulation of lipids in the liver during early lactation and alleviate the negative effects of plasma NEFA leading to more optimal metabolic health and productivity of dairy cows.  相似文献
