The objective of the experimental study was to examine whether aerial ammonia alone could predispose the respiratory system of pigs to infection with toxigenic Pasteurella multocida type A. Two groups of 5 pigs each were continuously exposed to 50 ppm ammonia and less than 5 ppm ammonia, respectively, for a 59-day period (from 37 kg to 90 kg bodyweight) followed by necropsy. In an aerosol chamber all pigs were exposed to an aerosol of toxigenic P. multocida type A (mean bacterial concentration in the aerosol-exposure chamber: 10(5) colony forming units/m3; exposure period: 25 min) at day 10, 21, 35 and 49 after the onset of ammonia exposure. During the experiment none of the pigs showed clinical signs of pneumonia nor did they develop visible distortion of the snout. None of the pigs had gross lesions in the lungs at necropsy and toxigenic P. multocida was not detected by culture from the lungs from any of the pigs. The chance of recovering toxigenic P. multocida from nasal swabs (collected during experiment) was 2-4 times greater in the test group compared to the control group. The average daily weight gain was lower for the ammonia exposed pigs compared to the control group. In conclusion the results from this study suggest that ammonia in concentrations of 50 ppm is unlikely to predispose growing pigs to pulmonary infection with toxigenic P. multocida.  相似文献

In two trials, 60 male commercial broilers were placed in each of eight environmentally controlled chambers receiving 0, 25, 50, or 75 ppm aerial ammonia from 1 to 28 days. Birds exposed to 25 ppm (lower concentration) ammonia gas developed ocular abnormalities but at a slower rate when compared with birds exposed to 50 and 75 ppm (higher concentrations). Birds exposed to higher concentrations also developed more severe lesions. With little atmospheric ammonia present after 28 days of the grow-out stage, the corneas indicated signs of healing. Lymphocytes and heterophils were seen in the iris at 49 days in ammonia-exposed birds even when ammonia exposure was terminated at 28 days. The lower ammonia concentrations resulted in abnormalities that were slight when compared with those seen at the higher ammonia concentrations. As measured by the incidence of inflammatory infiltrates in the trachea, lung, and air sacs, respiratory tract tissues did not appear to be affected by any tested level of aerial ammonia. The findings in this investigation represent the first report indicating that ammonia-induced uveitis in chickens clears rapidly after exposure to ammonia ceases.  相似文献

The effects of atmospheric ammonia (NH3) on the nasal and tracheal mucosa of pigs were investigated by morphometric and functional methods. Pigs were exposed to four concentrations of NH3 [5 (control), 25, 50 and 100 ppm] for 6 days in a specially designed air-pollutant exposure chamber. Samples were taken from the turbinates and the trachea, and the respiratory mucosa was examined by light and scanning electron microscopy. Dose-response curves to carbachol and isoproterenol were constructed using isolated strips of tracheal smooth muscle, with or without epithelium. In pigs exposed to ammonia, considerable mucosal injuries were observed in the turbinates but not in the trachea. The number of neutrophils in the epithelial layer and in the lamina propria, and epithelial hyperplasia were closely and significantly correlated with the concentrations of ammonia (r=0.894, p<0.001; r=0.727, p<0.001; and r=0.818, p<0.001, respectively). Except for the lamina propria, all these changes were significant (p<0.05) at ammonia concentrations as low as 25 ppm. The percentage of the surface of the turbinate mucosa that was ciliated tended to decrease with increasing ammonia concentration (r=0.439, p<0.082). Ammonia induced smooth-muscle hyperresponsiveness to carbachol with a close linear correlation between individual values of the carbachol-induced maximal effect and the NH3 concentrations (r=0.526, p<0.003). While mechanical destruction of the epithelium induced an increase in E max in the control group, no difference was observed between the intact and denuded strips from animals exposed to ammonia. The response to isoproterenol was not influenced by ammonia. It was concluded that quantitative histological analysis of the inflammatory infiltration and epithelial hyperplasia in the turbinates is a useful tool for quantifying the effects of atmospheric pollutants in pigs; a 6-day exposure to ammonia induces nasal irritation and functional disturbances of the tracheal smooth-muscle contractions at concentrations as low as 25 ppm.Abbreviations E max maximal effect - IM intramuscularly - IV intravenously - NAL nasal lavage - pD 2 negative logarithm of the molar concentration of agonist which induces an increase in tension equivalent to 50% of the maximal effect E max expressed as grams force per mg of dry weight - r correlation coefficient  相似文献

Two 4-wk trials (preliminary study) and three 5-wk trials (major study) were conducted to determine the effects of adding Yucca schidigera extract or anhydrous calcium chloride to nursery diets on the growth performance of nursery pigs and aerial ammonia concentration. The pigs were weaned between 13 and 15 d of age and had an initial BW of 3 to 6 kg. In each trial, pigs were allotted to three identical pig nursery rooms that were environmentally regulated. There were three diets (one diet per room): 1) control, containing 23% CP; 2) control plus 125 ppm of Yucca schidigera extract; and 3) control plus 1.95% anhydrous calcium chloride. Growth performance was recorded weekly. Aerial ammonia concentration was measured daily using aspiration detector tubes and during the last week of each trial using diffusion tubes. Manure samples were collected twice a week during the experimental period to determine ammonia and N concentrations and pH. Plasma urea concentration was determined in blood samples collected from the pigs at the end of each trial. Data were analyzed using split-plot and Latin square designs for the preliminary and major studies, respectively. Feed intake was similar among pigs fed all three diets. There were no differences in ADG and ADG/ADFI (G/F) between pigs fed the control diet and pigs fed the yucca extract diet (P > or = 0.41). In all trials, pigs fed the calcium chloride diet had lower ADG and G/F than pigs fed the other two diets (P < 0.05). In the preliminary study, aerial ammonia tended to be greater in the rooms in which pigs were fed the control diet than in the rooms with pigs fed the yucca extract diet (P = 0.08) and the calcium chloride diet (P = 0.11). In the major study, aerial ammonia increased weekly (diet x week; P < 0.001) in all rooms. In the 4th wk, ammonia concentrations were greater (P < 0.001) in the rooms in which pigs were fed the control diet than in the rooms in which the other two diets were fed. Dietary treatment had no effect on plasma urea concentration (P > or = 0.10), manure ammonia and N concentrations (P > or = 0.50), and manure pH (P > or = 0.78). Although aerial ammonia concentrations were relatively low, the addition of Yucca schidigera extract or calcium chloride to the diet of nursery pigs reduced ammonia concentrations in the nursery rooms.  相似文献

A cross-sectional epidemiologic study associating air quality with swine health was conducted on 28 swine farms in southern Sweden. Correlation of housing air environment to swine diseases and productivity (data collected over the preceding 12 months) were investigated. The most prevalent swine health problems detected at slaughter were pneumonia and pleuritis. In farrowing and nursery operations, the most prevalent problem was neonatal pig mortality. Several air contaminants (dust, ammonia carbon dioxide, and microbes) were found to be correlated with these swine health problems. Maximal safe concentrations of air contaminants were estimated on the basis of dose-response correlation to swine health or human health problems. Recommended maximal concentrations of contaminant were: dust, 2.4 mg/m3; ammonia, 7 ppm; endotoxin, 0.08 mg/m3; total microbes, 10(5) colony-forming units/m3; and carbon dioxide, 1,540 ppm. The overall quality of the ventilation system was correlated with lower concentration of ammonia, carbon dioxide, microorganisms, and endotoxin, but not with dust concentrations. High animal density was related to high ammonia and air microbe concentrations. Animal density measured as kilograms of swine per cubic meter (compared with kilograms of pig weight or swine per square meter) had the highest correlation to animal health and air contaminants.  相似文献

1. The natural history of air hygiene in a pullet house was assessed at three-weekly intervals using a combination of in vitro and in vivo assays. The performance of an internal air filter was also examined as an experimental technique for providing clean air. 2. Overall, air hygiene was poor by comparison with occupational standards for human health. The mass concentrations of respirable and inspirable dust were 1.4 and 11.3 mg/m3 compared to human exposure limits of 5 and 10 mg/m3 respectively. The concentration of ammonia was typically about 20 ppm. 3. The majority (greater than 99%) of airborne particles were non-viable. Commensal bacteria from the skin were the most numerous airborne bacteria. Scopulariopsis and Aspergillus spp. were the most prevalent fungi recovered from the air and bird's lungs respectively. The concentrations of airborne and lung fungi were positively correlated with ammonia concentrations. 4. The differences between in vivo and in vitro assays of airborne microorganisms suggest that an aerosol sampler should be devised which better mimics the physical and biochemical environment of the respiratory tract.  相似文献

Three air contaminants that may have serious health consequences to humans and poultry are ammonia, dust, and aerosolized bacteria. This study measured ammonia concentrations, dust concentrations, and the presence or absence of aerosolized Salmonella spp. and Escherichia coli inside and outside five commercial layer facilities. The average outside ammonia concentration measurements decreased as the distance from the facility increased from 10 to 40 ft. The measurements at 10 ft from the facilities were consistently higher than the average concentrations inside the facilities. The ammonia measurement trends inside of the facilities were affected by the temperature-dependent ventilation systems. Average dust concentrations inside the five facilities were consistently below 2 mg/m3. Three facilities also experienced average outside dust concentrations at all measured distances below 2 mg/m3. Two facilities had relatively high average dust measurements at 10 ft from ventilation fans (32.12 mg/m3 and 75.18 mg/m3). Escherichia coli and Salmonella were isolated from the air inside all five facilities and outside the facilities up to 40 ft from the ventilation fans. In condusion, dust concentrations may pose the largest risk to human and animal health at 10 ft away from the poultry facilities; risks associated with ammonia inhalation are greatest inside facilities during the coolest months of the year; and aerosolized bacteria are found inside and outside poultry facilities, but further work is needed to quantify the bacteria to further assess the health risk related to this issue.  相似文献

1. The occupational exposure to inhalable dust and ammonia of 12 stockmen working in barn systems conforming to the UK Royal Society for the Prevention of Cruelty to Animals (RSPCA) Freedom Foods requirements was measured and compared with established limits for occupational exposure. The influence of the type of task upon the daily exposure was assessed and comparisons of daily exposure were made with that of battery cage system stockmen. 2. The mean inhalable dust exposures experienced by poultry stockmen working in barn systems was 10.8 mg/m3 (9.5 mg/m3, time-weighted average), for a complete working day. This was greater (P < 0.01) than the mean dust exposure measured in current cage systems of 4.8 mg/m3 (4.4 mg/m3, time-weighted average). The individual values of dust exposure for 4 of the 12 barn stockmen exceeded the occupational exposure standard (OES) of 10.0 mg/m3 for inhalable dust, whereas that of all of the individual cage stockmen taking part in this study did not. 3. The mean ammonia exposure experienced by poultry stockmen working in barn systems was 14 ppm (11 ppm, time-weighted average), for a complete working day. This also was greater (P < 0.01) than the mean ammonia exposure measured in current cage systems of 8 ppm (7 ppm, time-weighted average). None of the individual values of ammonia exposures for both barn and cage system stockmen taking part in this study exceeded the OESs of 25 ppm, although the short-term limit of 35 ppm for 15 min was exceeded on a number of occasions. 4. Current methods for assessing the combined effects of dust and ammonia indicated that 8 of the 12 barn stockmen were at risk compared with 1 of the 8 cage system stockmen. 5. The average dust burden in daily common tasks such as maintenance and collecting floor eggs ranged from 5 to 15 mg/m3, while intermittent common tasks such as nest box checks and litter spreading ranged from 13 to 35 mg/m3. Tasks that had the highest exposure values included sweeping and partition removal, ranging from 63.3 to 70.7 mg/m3. 6. Designs of the typical barn systems investigated offer little scope for reducing airborne dust for common daily tasks. Personal respiratory protection while carrying out tasks that result in high dust concentrations in barns is advisable.  相似文献

Tylvalosin (TVN) is a water soluble macrolide used in swine production to treat enteric, respiratory, and arthritic pathogens. There is limited data on its distribution to synovial fluid beyond gavage studies, which do not represent field conditions. This study measured water disappearance, TVN concentration in the medicated water, daily dose, and concentrations of TVN and 3‐O‐acetyltylosin (3AT) in the synovial fluid and plasma of treated pigs over the administration period. The study emphasized understanding variation in tissue TVN concentrations within the context of a field setting. Sixty finisher pigs were housed individually with individual waterers. Six pigs were randomly allocated to the following time points for sample collection: 0, 48, 60, 72, 84, 96, 102, 108, 114, and 120 hr on medication. TVN was administered daily in the water for 5 days. Water disappearance and medicated water concentration were measured daily. At each time point, six pigs were euthanized and plasma and synovial fluid were collected for analysis. Median TVN synovial fluid concentrations ranged between <1 ng/ml (hour 0) to 3.6 ng/ml (hour 84). There was substantial variation between individual pigs for water disappearance (mean 4.36L and range 0–7.84). Median TVN water concentration was 59 ppm (range 38–75 ppm).  相似文献

In each of two experiments, 924 pigs (4.99 kg BW; 16 to 18 d of age) were assigned to 1 of 42 pens based on BW and gender. Pens were allotted randomly to dietary copper (Cu) treatments that consisted of control (10 ppm Cu as cupric sulfate, CuSO4 x 5H2O) and supplemental dietary Cu concentrations of 15, 31, 62, or 125 ppm as cupric citrate (CuCit), or 62 (Exp. 2 only), 125 (Exp. 1 only), or 250 ppm as CuSO4. Live animal performance was determined at the end of the 45-d nursery phase in each experiment. On d 40 of Exp. 2, blood and fecal samples were collected from two randomly selected pigs per pen for evaluation of plasma and fecal Cu concentrations and fecal odor characteristics. In Exp. 1, ADG, ADFI, and G:F were increased (P < 0.05), relative to controls, when pigs were fed diets containing 250 ppm Cu as CuSO4. Pigs fed diets containing 125 ppm Cu as CuCit had increased (P < 0.05) ADG compared with pigs fed diets supplemented with 15 or 62 ppm Cu as CuCit. The ADG, ADFI, and G:F did not differ among pigs fed diets containing 125 and 250 ppm Cu as CuSO4 or 125 ppm Cu as CuCit. In Exp. 2, pigs fed diets containing 250 ppm Cu as CuSO4 had improved (P < 0.05) ADG, ADFI, and G:F compared with controls. In addition, ADG, ADFI, and G:F were similar when pigs were fed diets containing either 250 ppm Cu as CuSO4 or 125 ppm Cu as CuCit. Pigs fed diets containing 62 ppm Cu as CuSO4 or CuCit had similar ADG, ADFI, and G:F. Plasma Cu concentrations were not affected by dietary Cu source or concentration, but fecal Cu concentrations were increased (P < 0.05) as the dietary concentration of Cu increased. Pigs consuming diets supplemented with 125 ppm Cu as CuCit had fecal Cu concentrations that were lower (P < 0.05) than pigs consuming diets supplemented with 250 ppm Cu as CuSO4. Fecal Cu did not differ in pigs receiving diets supplemented with 62 ppm Cu as CuSO4 or CuCit. Odor characteristics of feces were not affected by Cu supplementation or source. These data indicate that 125 and 250 ppm Cu gave similar responses in growth, and that CuCit and CuSO4 were equally effective at stimulating growth and improving G:F in weanling pigs. Fecal Cu excretion was decreased when 125 ppm Cu as CuCit was fed compared with 250 ppm Cu as CuSO4. Therefore, 125 ppm of dietary Cu, regardless of source, may provide an effective environmental alternative to 250 ppm Cu as CuSO4 in weanling pigs.  相似文献

During a 2-year period from January 1, 1999, to December 31, 2000, 77 diagnoses of mulberry heart disease (MHD) were documented at the Iowa State University Veterinary Diagnostic Laboratory. Mean (+/- SD) liver vitamin E concentrations were lower (P < 0.05) in pigs with MHD (3.12 +/- 1.12 ppm, wet weight) than in pigs that died of causes other than MHD (4.80 +/- 3.2 ppm, wet weight). The majority of the pigs affected with MHD ranged in age from 3 to 7 weeks. Statistical influence of age was found on the concentration of vitamin E (P < 0.01) but not on concentration of selenium in liver in pigs with MHD. Concentrations of vitamin E below 2 ppm were considered deficient. Hepatic vitamin E concentrations below 2 ppm were measured in 25% of the pigs with gross and microscopic lesions of MHD. In contrast, liver selenium concentrations were adequate in all pigs.  相似文献

In May 1996, 150 grower pigs in 5 California counties were exposed to selenium-contaminated feed distributed by a single feed company. Feed samples from 20 herds had a mean selenium concentration of 121.7 ppm dry weight (range, 22.1-531 ppm). In San Luis Obispo County, 52 pigs in 24 herds were exposed to the feed, and 8 pigs died with signs of paralysis. Bilateral symmetrical poliomyelomalacia involving the ventral horns of the cervical and lumbar intumescence was evident on histologic examination of spinal cord from affected pigs. Of 44 surviving exposed pigs, 33 (75%) exhibited signs of selenosis, including anorexia, alopecia, and hoof lesions. Thirty-nine of 44 pigs (88.6%) had elevated (>1 ppm) blood selenium concentrations. Surviving exposed pigs were changed to a standard commercial ration containing approximately 0.5 ppm (dry weight) selenium. Blood selenium concentrations were determined weekly for 46 days following removal of the contaminated feed and were compared with values of 20 control pigs fed a standard commercial ration. Mean (+/-SD) blood selenium concentrations of exposed pigs were 3.2 +/- 2.6 ppm at the initial sampling and 0.4 +/- 0.1 ppm after 46 days. Mean blood selenium concentrations of < or = 0.3 ppm for control pigs at all samplings were significantly lower (P < 0.001) than concentrations for exposed pigs. Muscle and liver samples of 22 of the 44 exposed pigs were collected at slaughter approximately 72 days after withdrawal of the selenium-contaminated feed. Muscle samples had a mean selenium concentration of 0.36 ppm (wet weight). Liver samples had a mean selenium concentration of 1.26 ppm (wet weight). One liver sample had a selenium value in the toxic range for pigs (3.3 ppm wet weight; reference range, 0.4-1.2 ppm). A 1-compartment pharmacokinetic model of selenium elimination in exposed pigs was generated, and the geometric mean blood selenium elimination half-life was estimated to be 12 days. The 60-day withdrawal time recommended by the Food Animal Residue Avoidance Database was considered sufficient to allow safe human consumption of tissues from exposed pigs.  相似文献

The growth performance and endocrine responses of male weaner pigs (3 to 8 weeks of age) was evaluated in two different environments (clean and dirty) and housing (single or groups of 10 pigs/pen) conditions. The dirty environment contained significantly elevated ammonia, carbon dioxide and dust levels compared with the clean environment. Pigs grew faster and consumed more feed in the clean environment and this was associated with reduced plasma cortisol concentrations compared with pigs in the dirty environment. Pigs housed in groups in the dirty environment had increased β-endorphin and decreased IGF-I concentrations compared to group housed pigs in the clean environment. Feed conversion efficiency did not differ due to environment or group housing. Plasma concentration of cortisol, β-endorphin, IGF-I and IGF-II did not differ between single and group housed pigs. Activity of the hypothalamic-pituitary-adrenal (HPA) axis was greater in response to environmental conditions than group housing, and this was associated with reduced growth in weaner pigs.  相似文献

The physiological effect on the equine respiratory tract of recycled phone book paper vs. sawdust bedding was investigated in 12 mares with no clinical signs of respiratory disease. Mares were housed in a 20-stall barn and randomly assigned to one of two treatment groups (Group 1=sawdust; Group 2=phone book paper) for 14 days. After a 14-day turnout, Group 1 was assigned to phone book paper and Group 2 was assigned to sawdust for 14 days using a crossover experimental design. Endoscopic examinations of the upper respiratory tract and trachea along with tracheal washes (TW) were performed on days 0 and 14 of each trial. Cytologic examinations were performed on TW samples. Cell type and laboratory interpretation of TW samples were noted. There was no difference (P>0.05) in cell type between treatment groups within or across trials. Laboratory interpretation indicated no abnormalities in 30 of 48 TW samples. Stall conditions were visually rated and graded daily for drinking water cleanliness, depth of bedding and absorbency, overall cleanliness, ammonia odor, and aerial ammonia levels. The phone book paper bedding was more absorbent (P<0.01), had a cleaner appearance (P<0.01), and had lower aerial ammonia levels (P<0.01) than the sawdust bedding. The phone book paper was effective in controlling aerial ammonia and did not adversely affect the respiratory health of clinically normal horses based on the diagnostic tests performed in this study. Phone book paper did effect the outward appearance of the horses, as horses housed on phone book paper had a dustier outward appearance. In addition, pigment was seen in the TW samples of 12 horses bedded on phone book paper, whereas it was rarely noted in horses bedded on sawdust. Overall, phone book paper appeared to be a viable option for horse bedding when compared to sawdust.  相似文献

A commercial doxycycline formulation was administered in drinking water to 12 pigs at the recommended dose of 10 mg/kg daily for 5 days. The mean plasma concentration at steady-state was 1.37 +/- 1.21 microg/mL, which was reached at 68 +/- 27.2 h postadministration. Absorption and elimination half-life values were 7.20 +/- 2.42 and 7.01 +/- 2.10 h, respectively. Most plasma concentrations during dosing were higher than the minimum inhibitory concentrations (MICs) described for the main porcine bacterial pathogens of the respiratory tract (Pasteurella multocida, Actinobacillus pleuropneumoniae, Bordetella bronchiseptica and Mycoplasma hyopneumoniae). It is concluded that when pigs were treated with doxycycline in drinking water at the recommended rate, therapeutically effective concentrations were achieved throughout the treatment period, supporting the clinical use of this tetracycline in the control of respiratory infections. However, inter-animal differences were marked.  相似文献

A total of 400 barrows from Dekalb EB and 83 terminal sires mated to 43 and 45 maternal lines were used to evaluate the effects of dietary ractopamine (RAC; Paylean, Elanco Animal Health, Greenfield, IN) concentrations (0, 5, 10, or 20 ppm; as-fed basis) and feeding durations (6 to 34 d) on growth, efficiency, carcass, and meat quality characteristics of finishing pigs. Barrows were weighed and sorted into five weight blocks, each block consisting of 16 pens (five pigs per pen). Weight blocks were allocated to feeding duration treatments and assigned consecutively by weight from lightest to heaviest to represent 34, 27, 20, 13, and 6 d on test, respectively. The lightest and heaviest blocks averaged 79.8 and 103.8 kg, respectively, at the start of the test. Within a weight block, pens (four per treatment) were randomly assigned to one of four dietary concentrations of RAC in a basal diet containing 18.5% CP and 1.13% lysine. The experiment-wide target slaughter weight was 109 kg, and pigs and feeders were weighed weekly. Weight blocks (80 barrows per block) were slaughtered at a commercial packing plant after 6, 13, 20, 27, or 34 d on test. Overall, RAC supplementation improved (P < 0.05) ADG; however, ADG was not different (P > 0.08) from controls for pigs fed 5, 10, and 20 ppm RAC for 27, 34, and 6 d, respectively. During each feeding period, RAC-fed pigs had improved (P < 0.05) G:F, and, after 20, 27, and 34 d on test, pigs fed 20 ppm RAC had greater (P < 0.05) G:F compared with those fed 0 or 5 ppm RAC. Hot carcass weight was increased (P < 0.05) by RAC feeding after 13 and 27 d of feeding, and by feeding 10 and 20 ppm RAC after 20 d of feeding. After 34 d, pigs fed 20 ppm RAC had heavier (P < 0.05) hot carcass weight than pigs fed 10 ppm RAC. Fat-free lean estimates and the 10th-rib LM area were increased (P < 0.05) by feeding 10 and 20 ppm RAC after 27 d, and by feeding 20 ppm RAC after 34 d compared with controls. Japanese and American color scores, as well as L*, a*, and b* values of the LM, were not affected (P > 0.11) by 5 and 10 ppm RAC compared with controls during each feeding period. Visual marbling score for the LM was decreased (P < 0.05) when RAC was fed at 10 and 20 ppm compared with 0 ppm RAC when fed for 34 d. Dietary RAC improved growth performance at all feeding durations, whereas carcass composition was improved at longer feeding durations. In addition, 5 and 10 ppm RAC did not affect objective and subjective measures of pork quality.  相似文献

Objective To investigate the protective efficacy of a pool of denatured membrane protein antigens of Mycoplasma hyopneumoniae (J strain) in the molecular size range 70 to 85 kDa (F3 antigen) in combination with adjuvants for pigs challenged with M hyopneumoniae .
Design A vaccine efficacy experiment with assessment of serum and respiratory tract antibody responses.
Procedure F3 antigens were emulsified with five different adjuvants. To groups of three pigs per vaccine, four vaccines were given by intramuscular injection, and two vaccines, including one of those given intramuscularly, were given by intraperitoneal injection.
Results Compared to six unvaccinated pigs, animals vaccinated with F3 antigen displayed significantly reduced pneumonia (54% reduction in mean lung score) following experimental challenge. Analysis of post-vaccination, pre-challenge IgG and IgA ELISA antibody absorbances in serum and respiratory tract washings revealed no correlation with lung score. Six weeks after challenge, pigs previously vaccinated intramuscularly mostly demonstrated greater IgG and IgA responses in respiratory tract washings, and greater IgG serum antibody responses, than those vaccinated by intraperitoneal injection.
Conclusion Pigs vaccinated with M hyopneumoniae antigens in the molecular size range of 70 to 85 kDa showed a significant reduction in lung lesions compared with unvaccinated control animals after experimental challenge. IgG and IgA antibody concentrations in serum and respiratory tract washings after vaccination do not provide a useful prognostic indicator of protection from enzootic pneumonia.  相似文献

A field trial was conducted to study Mycoplasma hyopneumoniae (Mh) infection dynamics by nested polymerase chain reaction (nPCR) and serology in pigs of a farm affected by enzootic pneumonia (EP). Moreover, correlation of Mh detection at different respiratory tract sites with presence of EP gross and microscopic lung lesions was assessed. These parameters were studied and compared between vaccinated (two doses at 1 and 3 weeks of age versus one dose at 6 weeks of age) and non-vaccinated pigs. Animals were monitored from birth to slaughter by nPCR from nasal swabs and by serology. From 3 to 22 weeks of age, an average of three pigs per treatment and per batch were necropsied (n = 302). The remaining pigs were sent to the slaughter (n = 103). Nasal, bronchial and tonsillar swabs were taken from the necropsied/slaughtered pigs; gross and microscopic EP-suggestive lung lesions were also assessed. Single and double vaccination resulted in earlier seroconversion and higher percentage of Mh seropositive pigs compared to control group. At slaughter, double vaccinated pigs showed lower percentage of EP-compatible gross lung lesions and lower Mh prevalence at upper respiratory tract sites (nasal cavity and tonsil) than control pigs.  相似文献

1. A survey of the concentration and emission rates of aerial ammonia, nitrous oxide, methane, carbon dioxide, dust and endotoxin was undertaken in 4 examples each of typical UK broiler, cage and perchery houses over 24 h during winter and summer.

2. Overall the air quality within the poultry houses was unsatisfactory as judged by the dual criteria of farmer health and bird performance.

3. Mean concentrations of ammonia ranged from 12.3 to 24.2 ppm while concentrations of methane and nitrous oxide were close to ambient levels. Mass concentrations of aerial dust ranged from 2 to 10 rag/m3 and 0.3 to 1.2 mg/m3 for inspirable and respirable fractions respectively, while endotoxin concentration was typically about 0.1 ^g/m3 (inspirable fraction).

4. Emission rates of gaseous ammonia were rapid (9.2 g (NH3/h per 500 kg live body weight) and uniform across the three types of building, while emissions of methane and nitrous oxide were slow. Rates of dust emission ranged from 0.86 to 8.24 g/h per 500 kg live body weight in the inspirable size fraction.  相似文献

OBJECTIVE: To assess the effects of inhalation of feed flour dust and dustborne endotoxin on respiratory tracts of pigs. ANIMALS: 29 healthy Belgian Landrace pigs. PROCEDURE: Pigs housed in an environmental chamber were exposed for 6 days to feed flour dust (1 to 15 mg/m3) and dustborne endotoxins (50 to 2,500 ng/m3). Effects were evaluated by measuring albumin concentration, lactate dehydrogenase (LDH) activity, cell composition of nasal lavage (NL) and bronchoalveolar lavage (BAL) fluids and blood, and percentages of CD4+ and CD8+ T lymphocytes in blood and lavage fluids. Dustborne endotoxin was obtained by mixing endotoxins from Escherichia coli (serotype O127:B8) with feed flour before spraying the flour in the environmental chamber. RESULTS: Exposure did not affect cell composition of NL fluid or blood. Total cell counts of BAL fluids were increased in all groups exposed to dust. Macrophage counts were increased in pigs exposed to inhalable dust concentrations as low as 4.4 mg/m3, and lymphocyte counts were increased in groups exposed to high dust concentrations. Percentages of CD4+ and CD8+ T lymphocytes in blood and lavage fluids were unchanged. In all dust-exposed groups, albumin content of BAL fluid was increased, whereas LDH activity was unaffected. Macrophage and lymphocyte infiltration and edema in the bronchi were identified by light microscopy. Effects attributable to E. coli endotoxin exposure were not identified. CONCLUSIONS: Inhalation of feed flour dust did not affect nasal mucosa but did induce bronchial airway inflammation. Dustborne endotoxins did not have effects attributable to endotoxin alone.  相似文献
