A total of 349 Salmonella enterica subspecies enterica
serovar Choleraesuis (S. Choleraesuis) strains, which were isolated
between 2008 and 2012 from 349 pigs at two slaughterhouses in Okinawa Prefecture, Japan,
were investigated for antimicrobial susceptibility and the presence of antimicrobial
resistance genes. All isolates were resistant to at least four antimicrobial agents. The
antimicrobial agents for which isolates showed a high incidence of resistance were as
follows: ampicillin (100%) and streptomycin (100%), followed by gentamicin (99.7%),
oxytetracycline (99.7%), sulfamethoxazole/trimethoprim (99.4%), nalidixic acid (40.1%) and
oxolinic acid (40.1%). All isolates were sensitive to cefuroxime, ceftiofur, colistin,
fosfomycin, enrofloxacin, orbifloxacin and danofloxacin. The predominant resistance
phenotypes and genotypes were: resistance to ampicillin, streptomycin, gentamicin,
oxytetracycline and sulfamethoxazole/trimethoprim (58.5%, 204/349) and
blaTEM-strA-strB-aadA1-aadA2-aacC2-tet
(B)-sul1-sul2-dhfrXII-dhfrXIII (36.1%, 126/349). The quinolone
resistance-determining regions (QRDRs) of gyrA, gyrB, parC and
parE of the quinolone-resistant isolates (n=12) showed amino acid
substitutions of Ser-83→Phe or Asp-87→Tyr in GyrA and Ser-107→Ala in ParC. To our
knowledge, this is the first report on the molecular characterization of antimicrobial
resistance among S. Choleraesuis strains in Japan. 相似文献

This study determined the
antimicrobial resistance profiles of Escherichia coli isolates from dogs
with a presumptive diagnosis of urinary tract infection (UTI). Urine samples from 201 dogs
with UTI diagnosed through clinical examination and urinalysis were processed for
isolation of Escherichia coli. Colonies from pure cultures were
identified by biochemical reactions (n=114) and were tested for susceptibility to 18
antimicrobials. The two most frequent antimicrobials showing resistance in Urinary
E. coli isolates were oxytetracycline and ampicillin. Among the
resistant isolates, 17 resistance patterns were observed, with 12 patterns involving
multidrug resistance (MDR). Of the 69 tetracycline-resistant E. coli
isolates, tet(B) was the predominant resistance determinant and was
detected in 50.9% of the isolates, whereas the remaining 25.5% isolates carried the
tet(A) determinant. Most ampicillin and/or amoxicillin-resistant
E. coli isolates carried blaTEM-1 genes.
Class 1 integrons were prevalent (28.9%) and contained previously described gene cassettes
that are implicated primarily in resistance to aminoglycosides and trimethoprim
(dfrA1, dfrA17-aadA5). Of the 44 quinolone-resistant
E. coli isolates, 38 were resistant to nalidixic acid, and 6 were
resistant to nalidixic acid, ciprofloxacin and enrofloxacin. Chromosomal point mutations
were found in the GyrA (Ser83Leu) and ParC (Ser80Ile) genes. Furthermore, the
aminoglycoside resistance gene aacC2, the chloramphenicol resistant gene
cmlA and the florfenicol resistant gene floR were also
identified. This study revealed an alarming rate of antimicrobial resistance among

Thirty-eight Aerococcus viridans isolates were obtained from milk from
478 cows with clinical mastitis in a farm during the periods between November 2011 and
February 2012, and between December 2012 and March 2013. Additional isolates were obtained
from processed manure (a mixture of composted manure, straw and hydrated lime) and bedding
materials. The processed manure was later used to cover the floor of the stalls in barns
as bedding materials. The temperatures recorded in the composted and processed manure were
not as high as those generally observed during satisfactory composting. To reveal the
association of A. viridans in manure-related products with intramammary
infection in cows, isolates were characterized by their DNA fragment patterns as
determined by pulsed-field gel electrophoresis (PFGE) and antimicrobial susceptibility
testing. Isolates obtained from milk, processed manure and bedding materials had identical
DNA fragment patterns. Antimicrobial susceptibilities were determined for 29 isolates from
milk, processed manure and bedding materials. Of these, 26 (89.7%) were resistant to
clindamycin, whereas virtually all the isolates were susceptible to 12 other
antimicrobials including cefalosporins that have been used to treat bovine mastitis in
Japan. In vitro, three A. viridans isolates from milk
and an isolate from processed manure survived for 3 hr in Good’s buffer (pH 9) at high
temperature (50°C). The results suggest that the processed manure and bedding materials in
this farm were possible sources of A. viridans that caused infection in

Present study was undertaken to study the prevalence of β-haemolytic streptococci in
equine of northern temperate region of Jammu and Kashmir, India. One hundred and forty one
samples were collected in duplicate from nasopharyngeal tract of diseased (53) and
apparently healthy equine (88) for isolation and direct PCR. A total of 77 isolates of
streptococci were recovered from 141 samples with an overall prevalence rate of 54.60%.
Out of these 77 isolates, 52 were from diseased and 25 from apparently healthy animals. Of
the 77 isolates, 4 were identified as Streptococcus equi subsp.
equi, 56 as S. equi subsp.
zooepidemicus and 17 as S. dysgalactiae subsp.
equisimilis. Thus the overall prevalence of S. equi subsp.
equi, S. equi subsp. zooepidemicus
and S. dysgalactiae subsp. equisimilis was 2.83, 39.71
and 12.05% respectively. The sensitivity of the PCR for the detection of S.

We analyzed the correlation between minimum inhibitory concentrations (MICs) of antimicrobials used in humans and those used in animals to enable comparison of antimicrobial susceptibility between Escherichia coli isolated from humans and those from animals. We compared the following pairs of MIC data: piperacillin (PIPC) to ampicillin (ABPC), amikacin (AMK) to kanamycin (KM), minocycline (MINO) to oxytetracycline (OTC), and levofloxacin (LVFX) to enrofloxacin (ERFX) using 103 isolates of E. coli from healthy livestock (cattle, pigs, broiler chickens, and layer chickens). Kappa analysis of the agreement for resistance and susceptibility between PIPC and ABPC, AMK and KM, MINO and OTC, and LVFX and ERFX showed almost perfect (κ = 0.81), slight (κ = 0.12), fair (κ = 0.37), and moderate (κ = 0.46) agreement, respectively. Within the antimicrobial pairs, all isolates resistant to the human antimicrobial were also resistant to the veterinary antimicrobial. However, there was less agreement within the pairs for those isolates that were sensitive to the human antimicrobial. The percentage agreement for susceptibility, defined as the percentage of isolates sensitive to both antimicrobials compared with isolates sensitive to both antimicrobials, as well as those sensitive only to the human antimicrobial, was 89.9%, 87.3%, 64.0%, and 89.9% for PIPC and ABPC, AMK and KM, MINO and OTC, and LVFX and ERFX, respectively. Our results suggest that the possibility of missing the resistance for antimicrobials used in human medicine by examining MICs for the equivalent antimicrobials used in veterinary medicine is low. 相似文献

A total of 156 Shiga-like toxin producing Escherichia coli (STEC) were isolated from fecal samples of Korean native (100/568, 18%) and Holstein dairy cattle (56/524, 11%) in Korea between September 2010 and July 2011. Fifty-two STEC isolates (33%) harbored both of shiga toxin1 (stx1) and shiga toxin2 (stx2) genes encoding enterohemolysin (EhxA) and autoagglutinating adhesion (Saa) were detected by PCR in 83 (53%) and 65 (42%) isolates, respectively. By serotyping, six STEC from native cattle and four STEC from dairy cattle were identified as O-serotypes (O26, O111, O104, and O157) that can cause human disease. Multilocus sequence typing and pulsed-field gel electrophoresis patterns highlighted the genetic diversity of the STEC strains and difference between strains collected during different years. Antimicrobial susceptibility tests showed that the multidrug resistance rate increased from 12% in 2010 to 42% in 2011. Differences between isolates collected in 2010 and 2011 may have resulted from seasonal variations or large-scale slaughtering in Korea performed to control a foot and mouth disease outbreak that occurred in early 2011. However, continuous epidemiologic studies will be needed to understand mechanisms. More public health efforts are required to minimize STEC infection transmitted via dairy products and the prevalence of these bacteria in dairy cattle. 相似文献

The minimum inhibitory
concentrations (MICs), mutation prevention concentrations (MPCs) and contribution of
quinolone resistance-determining region (QRDR) mutations to fluoroquinolone
(ciprofloxacin, enrofloxacin and orbifloxacin) susceptibility in 23 Pasteurella
multocida (Pm) isolates were investigated.
Fluoroquinolone-susceptible isolates (MICs ≤0.25 µg/ml,
9 isolates) had no QRDR mutations, and their respective MPCs were low.
Fluoroquinolone-intermediate isolates (MICs=0.5 µg/ml,
14 isolates) had QRDR mutations (Asp87 to Asn or Ala84 to Pro in gyrA),
and their respective MPCs were high (4–32 µg/ml).
First-step mutants (n=5) and laboratory-derived highly resistant fluoroquinolone mutants
(n=5) also had QRDR mutations. The MICs of fluoroquinolones for mutant-derived strains
were decreased in the presence of efflux inhibitors. The results indicated that the
fluoroquinolone resistance of Pm is mainly due to multiple target gene
mutations in gyrA and parC and the overexpression of

Background
The anaerobic spirochetes Brachyspira hyodysenteriae and Brachyspira pilosicoli cause diarrheal diseases in pigs. Their fastidious nature has hampered standardization of methods for antimicrobial susceptibility testing. For monitoring of antimicrobial susceptibility wild type cutoff values are needed to define where the wild type distribution of MICs ends and no approved cutoffs are available for Brachyspira spp. In this study antimicrobial susceptibility data for both species (in total 906 isolates) were compiled and analyzed and wild type cut off values for B. hyodysenteriae proposed.Methods
The MICs of tiamulin, valnemulin, tylosin, tylvalosin, doxycycline and lincomycin were determined by broth dilution in brain heart infusion broth supplemented with 10% fetal calf serum.Results
The compiled MICs from the broth dilution tests of the B. hyodysenteriae type strain, B78T (ATCC® 27164T), showed that the method yields reproducible results. In an international perspective the frequencies of isolates with decreased antimicrobial susceptibility were low among both B. hyodysenteriae and B. pilosicoli. However, in B. pilosicoli a constant level of 10-15% isolates with tiamulin MICs >4 μg/ml was detected between 2002 and 2010 and in B. hyodysenteriae a gradual increase in tiamulin MICs was seen between 1990 and 2003 although this increase has ceased during the last years. The wild type cutoff values proposed for B. hyodysenteriae are: tiamulin >0.25 μg/ml, valnemulin >0.125 μg/ml, tylosin >16 μg/ml, tylvalosin >1 μg/ml, lincomycin >1 μg/ml and doxycycline >0.5 μg/ml.Conclusions
The broth dilution method used in this study has over the years generated tightly grouped MIC populations for the field isolates and reproducible results for the control strain B78T and is therefore a suitable antimicrobial susceptibility test method for monitoring of Brachyspira spp. Here we propose wild type cutoff values for six antimicrobial agents for B. hyodysenteriae tested by broth dilution based on MIC distributions and the current knowledge on mechanisms of resistance in this species. There are few studies on antimicrobial resistance mechanisms and MIC distributions in B. pilosicoli but to some extent the cutoff values proposed for B. hyodysenteriae may be applicable also for monitoring of antimicrobial susceptibility in B. pilosicoli. 相似文献9.

Enterococcus faecium and E. faecalis are important human pathogens and also served as sentinel organisms for monitoring systems of antimicrobial resistance in both animals and humans. In this study, 106 E. faecium and 56 E. faecalis isolates were collected from 61 pig farms in 18 proveinces of China. Antimicrobial susceptibility was determined for 9 clinically important antibiotics and 3 antimicrobial growth promoters. The Enterococcus isolates showed high prevalence of resistance to medically important antibiotics, such as ampicillin (50.9% for E. faecium and 19.6% for E. faecalis), chloramphenicol (24.5% for E. faecium and 41.1% for E. faecalis), erythromycin (83.0% for E. faecium and 91.1% for E. faecalis), tetracycline (79.2% for E. faecium and 100% for E. faecalis), quinupristin/dalfopristin (26.4% for E. faecium) and ciprofloxacin (73.6% for E. faecium and 66.1% for E. faecalis). Resistance to tigecycline, linezolid and vancomycin was very rare. The resistance status of three representative in-feed antibiotics bacitracin, nosiheptide and enramycin was firstly investigated with Enterococcus as indicator bacteria. The Enterococcus isolates showed extremely high frequency of bacitracin resistance (96.7% for E. faecium and 87.8% for E. faecalis), while no nosiheptide and enramycin resistance was observed. Pulsed-field gel electrophoresis (PFGE) analysis showed that a majority of E. faecium and E. faecalis strains showed unrelated profiles, indicating high heterogeneity among the Enterococcus isolates. In this study, the mechanism conferring multiple drug resistance in several strains of flavobacteria isolated from the ovarian fluids of hatchery reared 3-year old brook trout Salvelinus fontinalis was investigated. Metabolic fingerprinting and 16S rRNA gene sequences identified the isolates as Flavobacterium johnsoniae. The isolates exhibited multiple resistances to a wide range of antimicrobial classes including penicillin, cephem, monobactam, aminoglycoside, and phenicol. Although plasmids and other transposable elements containing antimicrobial resistance genes were not detected, the isolates did contain a genomic sequence for a chloramphenicol-inducible resistance-nodulation-division family multidrug efflux pump system. Efflux pumps are non-specific multidrug efflux systems. They are also a component of cell-cell communication systems, and respond specifically to cell membrane stressors such as oxidative or nitrosative stress. Understanding of efflux pump mediated antibiotic resistances will affect efficacy of clinical treatments of fishes associated with F. johnsoniae epizootics. 相似文献

We examined the antimicrobial susceptibility of 848 Escherichia coli isolates from 237 feces samples of wild sika deer (Cervus nippon) captured between 2016 and 2019 in 39 of the 47 prefectures of Japan. Five of the 237 wild sika deer (2.1%) carried E. coli with resistance to at least one antimicrobial, and all the resistant isolates showed resistance to tetracycline. The resistant isolates contained antimicrobial resistance genes that were similar to those in E. coli derived from humans and farm animals. Although wild sika deer are not currently likely to be a source for the transmission of antimicrobial resistance in Japan, they can potentially mediate antimicrobial resistance spread by coming into contact with humans, animals, and their surroundings. 相似文献

The prevalence of antimicrobial resistant Escherichia coli was tested in symbovine flies and sympatric house martins (Delichon urbica) at a dairy farm. Antimicrobial resistant E. coli was detected in 89% (n = 147) of isolates from flies within a calf barn. Isolates with the same antimicrobial resistance phenotypes, genes, and pulsotypes were found between both fly and calf E. coli isolates, suggesting that the calves were the initial source of the antimicrobial resistant strains in fly isolates. Symbovine flies were considered as important reservoirs of antimicrobial resistant E. coli strains at a dairy farm, due to their intensive contact with cattle feces and manure. House martin fecal samples from the same farm contained 4.5% (n = 393) of antimicrobial resistant E. coli. House martin isolates displayed different macrorestriction profiles than fly isolates and the significance of house martins as a reservoir and vector of antimicrobial resistant E. coli appears low. 相似文献

Background
We describe coagulase-negative staphylococci (CoNS) isolates collected from ducklings exhibiting tremor in South Korea over the period of 2010 to 2011. Screening of antimicrobial susceptibility and analysis of SCCmec elements of CoNS were also investigated.Results
Staphylococcus cohnii was the most frequent staphylococcus (9 isolates) and S. sciuri (4 isolates), S. lentus (3 isolate), S. simulans (1 isolate) and S. epidermidis (1 isolate) were also detected. Among the 15 antimicrobials tested in this study, resistance against oxacillin (15 isolates, 83.3%) was most frequently observed, but only one isolate (SNUDS-1) possessed mecA. This isolate was shown to possess SCCmec type III; the type 3 ccr complex and the class A mec complex.Conclusions
Based on these results, isolate SNUDS-1 was shown to possess SCCmec type III; the type 3 ccr complex and the class A mec complex. Although the SCCmec type III is not predominant in human, MR-CoNS (Methicillin resistance Coagulase-negative staphylococci) in food animals should be monitored to prevent the dissemination of antimicrobial resistance genes and resistant pathogens to the community. 相似文献14.

To search natural compounds having inhibitory effect on bacterial growth is important,
particularly in view of growing multidrug resistant (MDR) strains of bacterial pathogens.
Like other bacterial pathogens, MDR Vibrio cholerae, the causative agent
of diarrheal disease cholera, is becoming a great concern. As an approach of searching new
antimicrobial agents, here, we show that anethole, a well-studied natural component of
sweet fennel and star anise seeds, could potentially inhibit the growth of MDR O1 El Tor
biotype, the ongoing 7th cholera pandemic variant strains of toxigenic V.
cholerae. The minimum inhibitory concentration (MIC) of anethole against
diverse O1 El Tor biotype strains is evaluated as 200
µg/ml. Moreover, the effect of anethole is
bactericidal and exerts rapid-killing action on V.
cholerae cells. This study is the first report which demonstrates
that anethole, purified from natural compound, is a potent inhibitor of growth of
toxigenic V. cholerae. Our data suggest that anethole could be a
potential antimicrobial drug candidate, particularly against MDR V.

The aim of this study was to assess the prevalence, the genotypic diversity, the antimicrobial resistance traits of canine and feline clinical methicillin-resistant Staphylococcus pseudintermedius (MRSP) isolates in a diagnostic laboratory in Italy during 2015-2016.All isolates were characterized by multilocus sequence typing (MLST), staphylococcal cassette chromosome (SCC)-mec typing and staphylococcal protein A (spa)-typing. The resistance profiles were assessed by antimicrobial susceptibility testing and confirmed genotypically by the detection of mecA gene and by microarray analyses.The prevalence of MRSP isolates was high (31.6%). All the strains were multidrug resistant and the most frequent clone was ST71-SCCmec type II-III. These results confirm a high prevalence of MRSP amongst clinical samples from pets in Italy. These isolates show multidrug resistance features that are of concern both in veterinary and human medicine for clinical and epidemiological reasons. 相似文献

Our objectives were to describe the antimicrobial susceptibility of Escherichia coli isolates from dogs in the northeastern USA and to identify temporal trends in resistance to selected antimicrobial agents. Data were collected retrospectively for all canine E. coli isolates from clinical samples submitted to Cornell University’s Animal Health Diagnostic Center between January 1, 2004 and December 31, 2011. Antimicrobial susceptibility testing was performed on 3519 canine E. coli isolates; frequency of resistance to each agent ranged from 0.4% (amikacin) to 34.3% (ampicillin). No trends were evident among urinary isolates, but cephalosporin resistance remained consistently high. Among non-urinary isolates, there was evidence of a significantly increasing trend in prevalence of resistance to several agents, including cephalosporins, enrofloxacin, and tetracycline. These data suggest that some of the most commonly used antimicrobial agents in companion animal practice are becoming less effective against canine E. coli infections outside the urinary tract. 相似文献

Background
The bacterial genus Staphylococcus consists of many species that causes infections in pet animals. Antimicrobial resistant staphylococci cause infections that are difficult to treat and they are important from the point of one health perspective. The aim of this study was to determine the prevalence of methicillin-resistant Staphylococcus (MRS) species, including methicillin-resistant S. aureus (MRSA) in diseased pet animals (Group A) and kennel dogs (Group B) in Lithuania and to characterize the isolates according to their antimicrobial resistance.Results
Twenty-one MRS isolates were obtained from 395 clinical samples (5.3 %; CI 95 % 3.5-8.0) of Group A animals. Sixteen, four and one isolates were from dogs, cats and a pet rabbit, respectively. The mecA gene was present in 20 isolates, whereas one isolate was positive for the mecC gene. Twenty-one MRS isolates (20.0 %; CI 95 % 13.5-28.6) were obtained from the vagina of female dogs (n = 105) (Group B). All isolates carried the mecA gene. Twelve MRS species were isolated of which S. pseudintermedius was the most common (18/42) followed by S. haemolyticus (8/42) and S. lentus (4/42). MRSA was not found. All MRS strains were susceptible to vancomycin, linezolid, daptomycin and quinupristin/dalfopristin. Resistance to tetracycline (16/21), clindamycin (15/21) and erythromycin (14/21) was the most common types of resistance in Group A animals. Three isolates also demonstrated resistance to rifampin. Resistance toward gentamicin (16/21), ciprofloxacin (15/21), macrolides (15/21) and tetracycline (12/21) was the most common in kennel dogs (Group B). The most common genes encoding resistance to antimicrobials (excluding beta-lactams) in isolates from Group A pets were tetK (21/42), aph(3′)-IIIa (11/42) and aac(6'')-Ie-aph(2'''')-Ia (9/42).Conclusions
A wide range of MRS species were found in pet animals in Lithuania. MRSA was not found. 相似文献18.

Mycoplasma bovis has spread widely throughout the world via animal movement and has become
an important pathogen of bovine respiratory disease. However, the minimum inhibitory concentrations of
antimicrobials for Mycoplasma bovis have not been studied in China. The objective of this
study was to determine the prevalence and antibiotic resistance of Mycoplasma bovis isolated
from young cattle with respiratory infection in China. Mycoplasma bovis was detected in 32/45
bovine respiratory infection outbreaks at beef farms in 8 provinces in China. The isolates were susceptible or
had medium sensitivity to ciprofloxacin, enrofloxacin and doxycycline, but were frequently resistant to
macrolides (13/32, 41%). An A2058G (Escherichia coli Numbering) mutation located in the
rrnA operon in domain V of 23S rRNA was observed in strains that were resistant to
macrolides. This single mutations at the rrnA operon in domain V of 23S rRNA may play an important role in the
resistance of Mycoplasma bovis strains to macrolides. 相似文献

This study was undertaken to identify and characterize amino acid substitutions in gyrA and parC related with quinolone resistance of 27 nalidixic acid-resistant (NaR) Salmonella isolates collected in poultry slaughterhouses in Korea. A total of 51 Salmonella isolates were detected from 44.8% (47/105) of the total samples from 15 poultry slaughterhouses examined, among which 27 (52.9%) NaR isolates were detected while ciprofloxacin (Cip) resistance was not present in the isolates. These 27 NaR isolates of DNA sequencing revealed that it contained three types of gyrA mutations in only D87 codon. Mutations in the D87 codon resulted in substitutions to G in most of the isolates, but D87Y and D87N exchanges were also detected. Although Cip resistance was absent, reduced susceptibility characterized by mutations in gyrA was apparent among Salmonella isolates from poultry slaughterhouses in Korea. 相似文献
