Ureaplasma diversum infection in bovine females may result in various reproductive problems, including granular vulvovaginitis, abortion, weak calves, salpingitis, and spontaneous abortion. The presence of U. diversum in a dairy bovine population from midwestern Brazil has not been established. The aim of this study was to determine whether U. diversum was present in dairy cattle from midwestern Brazil using polymerase chain reaction (PCR). Vulvovaginal mucus was analyzed from 203 cows located in six municipalities in the north region of Mato Grosso State, Brazil. A total of 25% of dairy cows with vulvovaginitis were positive for U. diversum. The factors evaluated were included in a multivariable logistic regression model with the presence of at least one positive cow in the herd serving as the dependent variable. Three variables were significantly associated with a U. diversum-positive PCR and were included in the final multivariable model: number of parities, vulvar lesions, and reproductive problems. For each new parity, the chance of U. diversum infection decreased 0.03-fold, indicating that cows with the highest number of parities were more protected. The presence of vulvar lesions was increased 17.6-fold in females positive for U. diversum, suggesting that this bacterium could be related to the red granular lesions in the vulvar mucosa, whereas reproductive problems were increased 7.6-fold. However, further investigations should be conducted to ascertain the effects of U. diversum in association with other mycoplasma species in the herds studied.  相似文献

We report herein a survey in which cultures of bovine reproductive tracts for Ureaplasma diversum and mycoplasmas were carried out in order to better understand the role of these organisms in granular vulvitis (GV). Samples cultured were vulvar swabs from clinically normal cows or ones with GV, preputial swabs or raw semen from bulls, and abomasal contents of aborted fetuses.

Ureaplasma diversum was isolated from 104 (43.3%) of 240 dairy cows, 32 (27.1%) of 118 beef cows, 43 (47.2%) of 91 beef heifers, 23 (67.6%) of 34 beef bulls, and three (60%) of five dairy bulls. Mycoplasmas were isolated from 18 (7.5%) dairy cows, two (1.6%) beef cows, three (8.8%) beef bulls, and one dairy bull. No isolation was made from 97 aborted fetuses. For 65 dairy cows and 30 beef heifers with vulvar lesions, the isolation rates for ureaplasmas of 62.5% and 69.7%, respectively, were significantly higher (X2) than those for normal animals (37.5% and 30.3%). On immunofluorescent serotyping of 137 of the 205 isolates, there were 66 in serogroup C (strain T44), 18 in serogroup B (strain D48), eight in serogroup A (strain A417 or strain 2312), 14 cross-reacting, and 31 that were not identified. It was concluded that U. diversum is commonly present in the lower reproductive tract of beef/dairy cattle in Saskatchewan and is associated with granular vulvitis.

Background Ureaplasma diversum has been associated with various reproductive problems in cattle, including granular vulvovaginitis, endometritis, salpingitis, early embryonic death, weak calves, decreased conception rates, balanoprosthitis, impaired spermatozoids and seminal vesiculitis in bulls. Methods This study briefly outlines the use of polymerase chain reaction (PCR) for the rapid detection of U. diversum directly from urogenital swabs collected from Australian beef cattle. Results The 16S ribosomal RNA gene sequences obtained from the PCR products of the clinical samples were closely related to U. diversum strain A417. Conclusion The present test enabled detection of the organism directly from clinical swabs collected from animals with or without lesions.  相似文献

In cattle, Ureaplasma diversum has been associated with decreased fertility, granular vulvovaginitis, endometritis, salpingitis and spontaneous abortion in cows and seminal vesiculitis, balanoposthitis and changes in bull sperm. The presence of U. diversum within the Australian cattle population has not been established. One of the aims of this study was to determine if U. diversum was present in Australian cattle, using culture and polymerase chain reaction (PCR), both of which are considered to be gold standards for bacterial identification. Of 100 samples collected from 66 male and 34 female cattle, 15 were positive for U. diversum. Therefore, Australia can no longer be considered free of U. diversum. Further studies should be conducted to ascertain the effects of U. diversum within Australian cattle herds and, if warranted, to investigate prevention, treatment and eradication protocols.  相似文献

In order to determine the role of Mycoplasma spp, Ureaplasma diversum and BHV-1 as causal agents of Granular Vulvovaginitis Syndrome in Nelore heifers raised under tropical conditions and based on the hypothesis that stressful conditions during puberty or breeding season would be a determinant factor for the infection, 340 heifers not vaccinated against BHV-1 were divided in Post-pubertal, in the beginning of the first breeding season, and Pubertal heifers. The vaginal lesion score (VLS) Grade 1 to 4 was giving according to lesion area and severity. Vaginal mucus was used to isolate Mycoplasma spp., Ureaplasma diversum and BHV-1. The predominant VLS was 2. No sample was positive for BHV-1; 48% were positive for Mycoplasma spp., Ureaplasma diversum, or both, with predominance of Ureaplasma diversum. Serum neutralization for BHV-1 showed more positive animals in pubertal group (23%); 3 of the paired sera demonstrated seroconversion. These data indicated that post-pubertal and pubertal Nelore heifers raised under extensive conditions are more susceptible to Mycoplasma spp. and Ureaplasma diversum. The hypothesis that the stress of pubertal period could lead to an acute vaginal infection by HBV-1 was not proofed.  相似文献

We evaluated the prevalence of mycobacterial infections (i.e., Mycobacterium bovis and non-tuberculous mycobacteria [NTM]) and their associated risk factors among cattle herds and trade cattle in southwestern Nigeria. Through cross-sectional study design, cattle herds from three locations were screened using the single intradermal comparative cervical tuberculin test based on two diagnostic standards; more than 4 mm (? 4 mm) and more than 2 mm (? 2 mm) cut-off points. Abattoir study involved screening trade cattle for tuberculous lesions. Overall, 515 cattle from 45 herds were screened. Using >?4 mm, animal level and herd prevalence of 11.7 and 46.7% were recorded, respectively. Applying the ? 2 mm cut-off, animal level and herd prevalence increased to 31.1 and 60.0%, respectively. Significantly, using the ? 2 mm cut-off, cattle in medium size herds/extensive management system (OR?=?1.6; 95% CI 1.1–2.5) and Sokoto Gudali (OR?=?2.3; 95% CI 1.4–3.8) were more at risk of being positive reactors, while Rahaji (OR?=?0.3; 95% CI 0.1–0.7) breeds of cattle and cows in the peri-urban area (OR?=?0.4; 95% CI 0.2–0.9) were less at risk of being positive reactors. Again, M. avium reactor of 21.7% was observed. In the abattoir, 1797 cattle were examined with 126 lesions suggestive of tuberculosis (TB). Culture/molecular analyses confirmed 2.2% M. bovis and 0.9% NTM infections. Risk factors associated with bovine TB among trade cattle were sex (OR?=?4.0; 95% CI 1.2–13.5) and age (OR?=?0.3; 95% CI 0.1–0.9). We confirm 11.7% prevalence of mycobacterial infections among populations of cattle screened with breed and herd size being major risk factors.  相似文献

Background

The aim of this study was to detect the associations between bovine herpesvirus 1 (BHV-1) status of a herd and respiratory disease (BRD) occurrence and reproductive performance in pregnant heifers and cows. The association between management-related factors and higher BRD occurrence was also estimated.

Methods

Serum samples, collected from cows and youngstock from 103 dairy cattle herds, were analyzed for antibodies against BHV-1, bovine respiratory syncytial virus (BRSV), bovine viral diarrhoea virus (BVDV), and Mycoplasma bovis. A questionnaire was used to collect data concerning herd management factors and reproductive performance, as well as the occurrence of clinical signs of respiratory disease in the last two years, as evaluated by the veterinarian or farm manager. Multiple correspondence analysis (MCA) and logistic regression analysis were performed to identify and quantify the risk factors.

Results

A low to moderate prevalence (1-49%) of BRSV antibodies among youngstock was associated with a high occurrence of respiratory disease (OR = 6.2, p = 0.010) in cows and in-calf heifers. Employees of the farm may participate in the spread of such disease. Larger herd size, loose-housing of cows, housing youngstock separately from cows until pregnancy, and purchasing new animals were factors possibly related to a high occurrence of respiratory disease symptoms in pregnant heifers and cows. The highest risk of abortions (> 1.3%) and increased insemination index (number of inseminations per pregnancy) (> 1.9) occurred in herds with a moderate prevalence of BHV-1 antibodies (1-49%) in cows.

Conclusions

BHV-1 was not associated with acute respiratory disease in adult dairy cattle, however was significantly related to reproductive performance. BRSV possesses the main role in respiratory disease complex in adult dairy cattle.  相似文献

The objective of the present experiment is to study the effect of feed intake restriction on the reproductive performance and pregnancy rate in Egyptian buffalo heifers. Thirty anestrus buffalo heifers were randomly divided into two equal groups. The low feed intake (LFI, n?=?15, 50 % restriction) group was fed a diet that consists of 3 kg concentrate, 1 kg wheat straw, and 3 kg fresh alfalfa, while the high feed intake (HFI, n?=?15) group was fed double the amount given to the LFI group for 4 months. All animals were weighed, transrectally examined, and visually checked for the signs of estrus, and blood samples were collected. Heifers in heat were mated with one fertile bull. The number of heifers showing estrus activity was 93.3 % in HFI vs. 20 % in LFI (P?<?0.01). Ovarian activity started earlier (P?=?0.03) in the HFI than LFI group. The weight at breeding, the diameter of the dominant follicle, number of heifers showing ovulations, number of services per conception, pregnancy rate, and overall mean of progesterone and estrogen concentrations were significantly higher (P?<?0.01) in the HFI than in the LFI group. The level of serum total protein, albumin, globulin, glucose, total cholesterol, and calcium were significantly higher (P?<?0.05) in the HFI group. Restriction of the daily feed intake to 50 % from NRC recommendations impair reproductive performance in terms of increasing the age at first service and reducing the pregnancy rate in buffalo heifers. In conclusion, feed intake could be effective in improvement of reproductive performance in buffalo heifers and further studies should be done on large scale of buffaloes in this point.  相似文献

The objective of this study was to determine the changes in biochemical indicators for nutritional stress from a herd of Boran and Nguni cows. A total of 40 cows (20 from each herd) were randomly selected for the study. The animals were identified according to their parities as follows: parity 1 (n = 8), parity 2 (n = 16), parity 3 (n = 8) and parity 4 (n = 8). Serum chemistry levels of glucose, total cholesterol, urea, creatinine, albumin, globulin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), gamma glutylaminotransferase (GGT), leukocytes, erythrocytes, haemoglobin, packed cell volume (PCV) and platelet counts were determined for 12 consecutive months spanning across the wet and dry seasons. The Boran cows had different creatinine concentration levels at different parities. The Boran cows in parity 1 had the highest (P < 0.05) concentration of creatinine 87.2 ± 5.17 μmol/L than other cows in different parities within the herd. There were significant differences in enzymes such as AST, ALP and ALT among the herd and parities. Boran cows in parity 3 had the lowest (P < 0.05) AST concentration levels of 52.6 ± 3.48 U/L, Nguni cows in parity 4 had the highest concentration of ALP of 161.3 ± 8.10 U/L while Nguni cows in parity 1 had the highest concentration level of ALT 55.1 ± 1.56 U/L than all the cows within the same herd. The Nguni herd had significantly higher (P < 0.05) levels of creatinine in both the wet (97.8 ± 3.27 μmol/L) and dry seasons (108.7 ± 3.29 μmol/L) compared with the Boran herd. Cows from the Nguni herd maintained significantly higher amount of urea, creatinine, albumin and total protein in both the wet and dry seasons as compared with cows from the Boran herd. Cows from the Nguni herd maintained significantly higher amount of urea, creatinine, albumin and total protein in both the wet and dry seasons as compared with those from the Boran herd.  相似文献

Monthly incidence rates for the detection of bovine leukemia virus (BLV) infection were calculated for a 15-month period for cattle over 12 months of age in a 200-cow dairy herd. Detection of BLV infection was based on the presence of persisting antibodies as measured by agar-gel immunodiffusion using the glycoprotein-51 antigen. A non-parametric procedure was used to examine 6-month trends in cumulative monthly incidence rates. Over the 15-month study, 25 heifers were detected as becoming infected with BLV after 12 months of age. No differences were found between the 12 monthly incidence rates (p = 0.77). Cumulative incidence rates between June and September (the fly season) were not higher than those for any other 6-month period (p = 0.29). Similarly, there was no evidence that BLV infection was transmitted during artificial insemination (p = 0.18) or during routine vaccination procedures (p = 1.0). Cumulative rates for a period corresponding to exposure of heifers to the dry herd were excessively high (p = 0.01). These findings suggest that transmission of BLV infection in the cattle studied was associated with close physical contact between susceptible and infected cattle, and not with biting flies, artificial insemination, or vaccination procedures.  相似文献

Background

Heifers not used as breeding stock are often implanted with steroids to increase growth efficiency thereby altering hormone profiles and potentially changing the environment in which ovarian follicles develop. Because bovine granulosa cell culture is a commonly used technique and often bovine ovaries are collected from abattoirs with no record of implant status, the objective of this study was to determine if the presence of an implant during bovine granulosa cell development impacts follicle stimulating hormone-regulated steroidogenic enzyme expression. Paired ovaries were collected from 16 feedlot heifers subjected to 1 of 3 treatments: non-implanted (n?=?5), Revalor 200 for 28 d (n?=?5), or Revalor 200 for 84 d (n?=?6). Small follicle (1 to 5 mm) granulosa cells were isolated from each pair and incubated with phosphate buffered saline (n?=?16) or 100 ng/mL follicle stimulating hormone (n?=?16) for 24 h.

Results

Granulosa cells of implanted heifers treated with follicle stimulating hormone produced medium concentrations of progesterone similar (P?=?0.22) to non-implanted heifers, while medium estradiol concentrations were increased (P?<?0.10) at 28 and 84 d compared to non-implanted heifers indicating efficacy of treatment. Additionally, real-time PCR analysis in response to follicle stimulating hormone treatment demonstrated a decrease in steroidogenic acute regulatory protein (P?=?0.05) mRNA expression in heifers implanted for 84 d and an increase in P450 side chain cleavage mRNA in granulosa cells of heifers implanted for 28 (P?<?0.10) or 84 d (P?<?0.05) compared to non-implanted females. However, no difference in expression of 3-beta-hydroxysteroid dehydrogenase (P?=?0.57) and aromatase (P?=?0.23) were demonstrated in implanted or non-implanted heifers.

Conclusions

These results indicate follicles which develop in the presence of high concentrations of androgenic and estrogenic steroids via an implant tend to demonstrate an altered capacity to respond to follicle stimulating hormone stimulation. Thus, efforts should be made to avoid the use of implanted heifers to study steroidogenesis in small follicle granulosa cell culture systems.  相似文献

A cross-sectional study was conducted in three counties (Damxung, Maizhokunggar and Yadong) in Tibet in April and May 2015. A total of 1,523 yaks owned by 181 herders were randomly selected and blood sampled. Sera were tested using the rose bengal test (RBT) and a competitive immune-enzymatic assay (C-ELISA) and the test results interpreted in parallel. The individual yak prevalence was 2.8% (95% CI 2.0–3.7) with a herd prevalence of 18.2% (95% CI 12.9–24.6). At the individual level, two predictor variables, age and production system, were significantly associated with seropositivity by a binary logistic regression analysis. The odds of Brucella infection were significantly higher in older Yaks (3–5 years old, OR = 4.51; 95% CI 1.53–19.29; ≥6 years old, OR = 3.89; 95% CI 1.23–17.21) compared to those of younger yaks (≤2 years old). The odds of seropositivity for yaks managed under an agro-pastoral production system were 2.9 (95% CI 1.48–5.86) times higher compared to those managed under a pastoral production system. At the herd level, an association between the infection with Brucella and a history of abortions in the herd was observed (OR = 4.98, 95% CI 1.48–16.62). Surprisingly, vaccination was not associated with a lower level of infection (p = 0.49 and p = 0.99 for individual and herd level data, respectively). The results of the survey indicate that bovine brucellosis is endemic among the yak population in the plateau region of China, and the risk factors identified in the study should be considered in the epidemiology of the disease and when developing control programs for the disease.  相似文献

This study investigated the association between pre-breeding blood urea nitrogen (BUN) concentration and reproductive performance of beef heifers within different management systems in South Africa. Bonsmara heifers (n?=?369) from five herds with different estimated levels of nitrogen intake during the month prior to the commencement of the breeding season were sampled in November and December 2010 to determine BUN concentrations. Body mass, age, body condition score (BCS) and reproductive tract score (RTS) were recorded at study enrolment. Trans-rectal ultrasound and/or palpation was performed 4–8 weeks after a 3-month breeding season to estimate the stage of pregnancy. Days to pregnancy (DTP) was defined as the number of days from the start of the breeding season until the estimated conception date. Logistic regression and Cox proportional hazards survival analysis were performed to estimate the association of pre-breeding BUN concentration with subsequent pregnancy and DTP, respectively. After stratifying for herd and adjusting for age, heifers with relatively higher pre-breeding BUN concentration took longer to become pregnant when compared to those with relatively lower BUN concentration (P?=?0.011). In the herd with the highest estimated nitrogen intake (n?=?143), heifers with relatively higher BUN were less likely to become pregnant (P?=?0.013) and if they did, it was only later during the breeding season (P?=?0.017), after adjusting for body mass. These associations were not present in the herd (n?=?106) with the lowest estimated nitrogen intake (P?>?0.500). It is concluded that Bonsmara heifers with relatively higher pre-breeding BUN concentration, might be at a disadvantage because of this negative impact on reproductive performance, particularly when the production system includes high levels of nitrogen intake.  相似文献

Background

Bovine brucellosis remains one of the most prevalent zoonotic infections affecting dairy cattle in developing countries where the applied control programs often fail. We analyzed the epidemiologic pattern of bovine brucellosis in a dairy cattle herd that showed several cases of abortions after regular vaccination with RB51 (B. abortus vaccine). In 2013 thirty dairy cows, from a Holstein-Friesian dairy herd with a population of 600 cattle, aborted five months post vaccination by a regular RB51 vaccine. Blood samples were drawn from milking cows and growing heifers, as well as heifers and cows pregnant up to 6 months. These samples were collected in June 2013 (n?=?257) and May 2014 (n?=?263) and were tested by real time (rt)-PCR as well as serological tests, in particular Rose Bengal Test (RBT), Enzyme-Linked Immunosorbent Assays (ELISA) and Fluorescence Polarization Assay. Tissue specimens were also collected from an aborted fetus and cultured. Isolates were subjected to bacteriological typing tests at the genus and species levels.

Results

Five months post vaccination with RB51 vaccine, Brucella (B.) DNA was detected in blood samples of cows by rt-PCR. The serological tests also revealed the spread of Brucella field strains within the herd in 2013. Four Brucella isolates were recovered from specimens collected from the aborted fetus. These isolates were typed as follows: one B. abortus RB51 vaccine strain and three isolates of B. abortus field strain. The seropositive cows with positive rt-PCR might indicate an infection by the Brucella field strain; while the positive rt-PCR results from seronegative animals may either be due to circulating RB51 vaccine DNA in vaccinated animals or to circulating field strain in infected animals before seroconversion.

Conclusion

The results herein suggest that PCR can be a good supplementary tool in an outbreak situation, if an assay is available that can differentiate vaccine and field strains with a high analytical sensitivity. We recommend using RBT and ELISA in parallel in outbreak situations, to identify as many infected animals as possible during the initial screenings. This test procedure should be repeated for at least three successive negative tests, with one month interval.

The present study was conducted to evaluate the effects of high and low supplementation levels pre- and post-weaning on performance, endocrine, metabolic, and reproductive responses of Nellore heifers. Fifty Nellore heifers with 132?±?9.9 kg average body weight (BW) and 138?±?19 days of age were supplemented from 4 to 14 months. The heifers were distributed into five supplementation plans: HH—6 g/kg of BW of supplement pre- and post-weaning, HL—6 g/kg of BW of supplement pre-weaning and 3 g/kg post-weaning, LH—3 g/kg of BW pre-weaning and 6 g/kg of BW post-weaning, LL—3 g/kg of BW pre- and post-weaning, and CC—control, no supplementation. Interactions were not significant (P?>?0.10). The level of supplement fed pre-weaning did not affect any of the performance variables evaluated at the end of the experiment (P?>?0.10). There was a significant effect of supplementation and level of supplementation fed post-weaning on average daily gain (ADG) and final BW (P?<?0.05). Overall ADG was also affected only by supplementation and level of supplement fed post-weaning (P?<?0.05) with animals receiving 6 g/kg of BW post-weaning gaining more weight. Follicular diameter was greater in animals that received 6 g/kg of BW post-weaning (P?<?0.05). In summary, performance, endocrine, metabolic, and reproductive variables evaluated in the current study were improved by the level of supplement fed post-weaning. Heifers receiving supplementation of 6 g/kg of BW post-weaning had greater responses, independent of the level received during the pre-weaning phase.  相似文献

Bull semen is commonly contaminated with mycoplasmas. To determine the source of contamination, semen and the genital tracts of 45 artificial insemination bulls were cultured for these organisms. The results indicate that mycoplasmas colonize the prepuce and the distal part of the urethra. Only rarely were they found in the ampullae or seminal vesicles. In 92% of the bulls with contaminated semen the same Mycoplasma species or Ureaplasma diversum was isolated from the prepuce and urethral orifice as was found in the semen. This suggests that the prepuce and distal urethra is the source of contamination. Colonization of the genital tracts with Mycoplasmas or U. diversum was not associated with histological changes.  相似文献

Bovine respiratory disease (BRD) exerts a major impact on the beef cattle industry nationally and worldwide, with a range of aetiological factors impacting its pathogenesis. Previous research has focussed on an increasing number of bacteria and viruses that have been shown to play a role in eliciting disease. Recently, additional agents have been emerging as potential contributors to BRD, including the opportunistic pathogen Ureaplasma diversum. To determine if U. diversum was present in Australian feedlot cattle and if that presence was linked to BRD, nasal swabs were collected from a cohort of 34 hospital pen animals and compared to 216 apparently healthy animals sampled contemporaneously at feedlot induction and again after 14 days on feed at an Australian feedlot. All samples were subjected to a de novo polymerase chain reaction (PCR) assay targeting U. diversum in combination with other BRD agents. U. diversum was detected at a low prevalence in cattle at induction (Day 0: 6.9%, Day 14: 9.7%), but in a significantly greater proportion of cattle sampled from the hospital pen (58.8%). When considering the presence of other BRD-associated agents, co-detection of U. diversum and Mycoplasma bovis was most common in hospital pen animals receiving treatment for BRD. These findings suggest that U. diversum may be an opportunistic pathogen involved in the aetiology of BRD in Australian feedlot cattle, in combination with other agents, with further studies are warranted to identify if a causal relationship exists.  相似文献

The associations between herd bovine herpesvirus 1 (BHV-1) seroprevalence, along with other infectious and farm management factors with bovine respiratory disease (BRD) in dairy calves and heifers, were investigated. Serum samples from 103 dairy cattle herds were analyzed for antibodies against BHV-1, bovine respiratory syncytial virus (BRSV), bovine viral diarrhea virus (BVDV), and Mycoplasma bovis (M. bovis). A questionnaire was used to record herd management practices. A high occurrence of respiratory disease in unweaned calves was associated with low to moderate and high prevalence of BHV-1 among cows (OR=14.8, p=0.005 and OR=19.2, p=0.002, respectively) and positive BVDV status of a herd (OR=5.1, p=0.02). The presence of BVDV in a herd was related to a high incidence of respiratory disease in heifers 3-16 months old (OR=4.3, p=0.027). Based on the results of multiple correspondence analysis, holding youngstock separately from cows until pregnancy, introducing new animals and the activities of on-farm employees may contribute to a higher incidence of BRD.  相似文献
