Sodium iron ethylenediaminetetraacetate (NaFeEDTA) has superior iron bioavailability especially in foods containing iron absorption inhibitors. However, mechanisms involved in the absorption and subsequent partitioning of iron complexed with EDTA are poorly understood. Our objectives were to compare retention and tissue distribution of iron administered to rats either as FeSO4 or NaFeEDTA, either orally (OR) or subcutaneously (SC). Weanling rats were fed semipurified diets supplemented with either FeSO4 or NaFeEDTA for 7 days. They were then given a meal containing 59Fe-labeled FeSO4 or NaFeEDTA, or they were injected SC with these two forms of radiolabeled Fe. 59Fe retention was measured by whole body counting. Urine was collected and counted at 24 h intervals throughout the counting period. Tissue samples were analyzed for nonheme iron and 59Fe activity. Absorption of iron from FeSO4 or NaFeEDTA was similar (57.7 and 53.4%, respectively). Seventy-seven percent of the injected Na59FeEDTA was excreted in the urine within 24 h, whereas only 0.5, 0.8, and 1.4% of the injected 59FeSO4, oral 59FeSO4, and oral Na59FeEDTA, respectively, was excreted in the urine. The nonheme iron content was lower in the liver and spleen, by 56.8 and 28.4%, respectively, among rats consuming the NaFeEDTA diet as compared to rats fed FeSO4. We conclude that iron is dissociated from EDTA prior to or during intestinal absorption and that some fraction of the dissociated EDTA is absorbed separately from the iron.  相似文献

Although it has been shown that iron absorption from NaFeEDTA, a promising iron fortificant, is effectively down-regulated in iron-loaded rats, effects of prolonged exposure to high dietary levels of NaFeEDTA are not well understood. The objectives of this study were to determine whether rats can adapt to a high dietary level of NaFeEDTA by down-regulating iron absorption, and to determine effects on tissue iron distribution, with or without an iron absorption inhibitor. Male Sprague-Dawley rats were exposed to diets supplemented with FeSO4 or NaFeEDTA at 1200 mg of Fe/kg of diet, with or without tea, for 27 days. Iron absorption measured by whole-body counting before and after exposure showed that rats adapted to the high dietary level of FeSO4 or NaFeEDTA by down-regulating iron absorption to a similar extent. However, nonheme iron concentrations in liver and spleen were about 35-50% lower, whereas the concentration in kidney was about 300% higher in rats fed NaFeEDTA, compared to rats fed FeSO4. Tea had no major impact on iron absorption or iron status, regardless of iron source. Our results showed that although iron absorption was down-regulated similarly, body iron distribution was markedly different between rats exposed to FeSO4 and those exposed to NaFeEDTA. Further studies are warranted to determine the effects of prolonged exposure to dietary NaFeEDTA on kidney iron accumulation and kidney function.  相似文献

Sodium iron(III) ethylenediaminetetraacetate (NaFeEDTA) has considerable promise as an iron fortificant because of its high bioavailability in foods containing iron absorption inhibitors. In this study, uptakes of iron from NaFeEDTA, FeSO4, and FeCl3 by Caco-2 cells were compared in the absence or presence of ascorbic acid (AA), an iron absorption enhancer; at selected pH levels; and in the absence or presence of an iron absorption inhibitor, bathophenanthroline disulfonic acid (BPDS). Ferritin formation in the cells was used as the indicator of iron uptake. Uptake from all three Fe sources was similar in the absence of AA. Adding AA at a 5:1 molar excess as compared to Fe increased uptake by 5.4-, 5.1-, and 2.8-fold for FeSO4, FeCl3, and NaFeEDTA, respectively. The smaller effect of AA on uptake from NaFeEDTA may be related to the higher solubility of NaFeEDTA and/or the strong binding affinity of EDTA for Fe3+, which may prevent AA and duodenal cytochrome b from effectively reducing EDTA-bound Fe. Uptake was inversely related to the pH of the media over a range of 5.8-7.2. Because uptake by DMT-1 is proton-coupled, the inverse relationship between pH and Fe uptake in all three iron sources suggests that they all follow the DMT-1 pathway into the cell. Adding BPDS to the media inhibited uptake from all three iron compounds equally. Because BPDS binds Fe2+ but not Fe3+ and because only Fe2+ is transported by DMT-1, the finding that BPDS inhibited uptake from NaFeEDTA suggests that at least some iron dissociates from EDTA and is reduced just as simple inorganic iron at the brush border membrane of the enterocyte. Taken together, these results suggest that uptake of iron from NaFeEDTA by intestinal enterocytes is regulated similarly to uptake from iron salts.  相似文献

Caseinophosphopeptides (CPP) issued from enzyme digestion of caseins bind cations and keep them soluble in the digestive tract. They could be used as ligands to improve iron (Fe) bioavailability. Fe-deficient young rats were repleted with Fe (40 or 200 mg/kg of diet) bound either to the beta-CN (1-25) of beta-casein or to whole beta-casein or as FeSO(4). A control pair-fed group was given 200 mg of Fe (FeSO(4))/kg of diet for 6 weeks. After repletion, hemoglobin concentration of the control group was reached only by the ) animals fed 200 mg of Fe/kg; beta-CN (1-25) bound Fe (40 and 200 mg) produced higher Fe liver and spleen stores than FeSO(4). Binding Fe to the whole, nonhydrolyzed beta-casein gave results intermediate between the other experimental groups. Binding Fe to phosphoserine residues of low molecular weight CPP improved its ability to cure anemia and to restore iron tissue stores, as compared to Fe bound to the whole casein and to inorganic salts.  相似文献

Elemental iron powders are widely used to fortify flour and other cereal products. Our objective was to test the hypothesis that baking enhances the bioavailability of elemental iron powders by oxidizing Fe(0) to Fe(2+) or Fe(3+). An in vitro digestion/Caco-2 cell culture model and a piglet model were used to measure bioavailability. Bread flour, either unfortified or fortified with hydrogen-reduced (HR) iron powder or FeSO(4) (300 mg Fe/kg flour), was baked into bread. For the in vitro studies, bread samples were treated with pepsin at pH 2, 3, 4, 5, 6, or 7 and subsequently incubated with pancreatic enzymes at pH 7 in a chamber positioned above monolayers of cultured Caco-2 cells. Ferritin formation in the cells was used as an index of iron bioavailability. Ferritin formation in cells fed HR Fe bread was similar to cells fed FeSO(4) bread when the peptic digestion was conducted at a pH 2 but lower when the peptic phase was conducted at pH 3, 4, 5, 6, or 7 (P < 0.05). Pig diets containing 35% dried bread were prepared and fed to cross-bred (Hampshire x Landrace x Yorkshire) anemic pigs in two studies. The rate of increase in hemoglobin Fe over the feeding period was used to calculate relative biological value (RBV), an index of iron bioavailability. In the first pig study, RBV of HR Fe added to flour prior to baking was 47.9% when compared to FeSO(4) fortified flour (P < 0.05). In the second pig study, a third treatment consisting of unfortified bread with HR iron added during diet mixing (after bread baking) was included. RBVs of the HR Fe diet (Fe added after baking) and HR Fe diet (Fe added before baking) were 40.1% and 53.5%, respectively, compared to the FeSO(4) diet. Differences in RBV between the HR Fe (before and after baking) and FeSO(4) (before baking) treatment groups were significant, but the difference between the before and after HR treatment groups was not significant. We conclude that bread baking does not enhance the bioavailability of elemental iron powders.  相似文献

Extrinsic and intrinsic iron and zinc labels were used to test iron and zinc absorption from two bean (Phaseolus vulgaris) genotypes, containing normal (common beans, CB) or higher (HFeZnB) iron and zinc concentrations, fed in single meals to young women with low iron reserves. The women were divided into two groups, with one receiving a CB test meal (n = 12) and the other, an HFeZnB test meal (n = 11). The beans were intrinsically labeled hydroponically with (55)Fe (CB and HFeZnB) and with (70)Zn (HFeZnB). Concentrations of zinc and iron were 98 and 65% higher, respectively, in HFeZnB as compared to CB, but phytic acid contents were similar. Extrinsic labels were (59)Fe (CB and HFeZnB), (67)Zn (CB), and (68)Zn (HFeZnB). Iron and zinc percent absorption levels were calculated from radio-iron activity in red blood cells and from urinary excretion of zinc isotopes. Intrinsic and extrinsic iron absorption measures were highly correlated (R (2) = 0.986) (average extrinsic/intrinsic ratio was 1.00). Iron absorption was low (geometric mean < 2%) in both bean types, and total iron absorbed was not different between types. Intrinsic zinc absorption from the HFeZn beans was higher than extrinsic absorption (15.2% vs 13.4%, p < 0.05) (average extrinsic/intrinsic was 0.90). The correlation between intrinsic and extrinsic zinc measures was not as high as that for iron (R (2) = 0.719). Percent zinc absorption levels were similar in both bean types, but total extrinsic zinc absorbed was 90% higher (p < 0.05) from the HFeZnB meal. Thus, the less expensive and time-consuming extrinsic labeling may be used to screen various varieties of beans for iron bioavailability in humans, but it underestimates zinc absorption by approximately 10%. Selective breeding for high-zinc bean genotypes may improve zinc status. However, high-iron genotypes appear to have little effect on iron status when fed alone in single meals to women with low iron reserves.  相似文献

Abstract

The chemical behaviour of iron chelates, incorporated in compound fertilizers with inorganic micronutrient compounds, was studied. Pot experiments were conducted to evaluate the effectiveness of these products in controlling iron deficiency. A commercial compound fertilizer, containing 0.09% Fe as Fe‐DTPA, 0.12% Cu, 0.16% Mn and 0.04% Zn as sulfates, proved to be ineffective in preventing iron chlorosis in Chamaecyparis lawsoniana ’Alumii’, growing on sphagnum peat at two lime levels. The same fertilizer formulation with 0.09%‐ Fe as Fe‐EDDHA did prevent iron chlorosis at both lime levels. Fe‐EDTA, incorporated in the compound fertilizer, gave good results at the lower but not at the higher lime level, due to the low stability of Fe‐EDTA at high pH. Data from laboratory experiments showed that copper replaced the chelated iron in the compound fertilizer containing Fe‐DTPA, causing the iron to precipitate. The strong competition between copper and iron for the organic ligand is due to a specific affinity of copper for DTPA, resulting in a copper chelate with high stability constant and a molar ratio of copper to chelating agent of 2 : 1.

In the case of Fe‐EDDHA and Fe‐EDTA the competition between iron and copper is much weaker. In contrast to Fe‐DTPA, these chelates remain rather stable when incorporated in fertilizers containing micronutrients.  相似文献

用营养液培养方法研究了铁和两种形态氮素对玉米植株吸收铁、锰、铜、锌等微量元素及其在体内分布的影响。结果表明:与硝态氮(NO3--N)相比,铵态氮(NH4+-N)显著提高了玉米对铁的吸收,降低了对锰、铜及锌的吸收。供铁也明显提高了植株地上部铁的吸收总量,降低了锰及锌的吸收量,尤其是在供应No3--N时这种作用更为明显。在缺铁条件下,NH4+-N处理的玉米新叶中铁的含量明显高于NO3--N处理;而新叶、老叶、茎中锰、锌、铜含量以及根中锰、锌含量都明显低于NO3--N处理。但使用NH4+-N时,根中铜的含量较高。在供铁条件下,NH4+-N处理的玉米植株四个不同器官中锰和锌的含量显著低于NO3--N处理的植株,而铜的含量正好相反。在缺铁条件下,玉米新叶中活性锰、活性锌的含量显著高于供铁处理;与NO3--N相比,NH4+-N的供应也显著降低了玉米新叶中活性锰以及活性锌的含量。  相似文献

本试验利用营养液和土壤培养系统,研究不同Fe、 Cd处理下根表铁膜对水稻吸收Cd、 Mn、 Cu、 Zn的影响。土壤中Fe的水平为0、 1、 2 g/kg Fe(以FeSO47H2O的形式供应),Cd 的水平为0、 2、 10 mg/kg Cd(以3CdSO48H2O的形式供应)。营养液中Fe和Cd的水平分别为0、 10、 30、 50、 80、 100 mg/L Fe 和 0、 0.1、 1.0 mg/L Cd。收获后测定水稻根表、 根中和地上部Cd、 Fe、 Mn、 Cu、 Zn 含量。试验结果表明,两种培养方式下,随着介质中Fe浓度的增加,水稻根表铁膜(DCB-Fe)逐渐增多。土壤培养方式下,根表铁膜中Cd 和 Mn 含量随铁膜量增加而略有增加,所有元素含量均表现为根中大于铁膜中。营养液培养条件下,根表铁膜中Mn和Cu含量在高量 Fe 供应时有所增加, Mn、 Cu、 Zn表现为铁膜中大于根中。根表铁膜中Zn含量在两种培养方式下均未呈现一定规律性变化。根中和地上部 Cd、 Mn、 Cu、 Zn 含量一般都随介质中Fe浓度的增加而下降,Cu和Zn含量在加Cd处理中下降。以上结果证明,铁膜对Cd 的吸附阻挡能力有限,对Mn、 Cu、 Zn 的吸附作用因培养方式和元素种类不同而有所差异,植株体内微量元素含量的下降主要与它们之间的相互抑制作用有关。  相似文献

A "whole-body" radioassay procedure was used to assess retention and absorption by rats of Zn in mature kernels of whole grain wheat harvested from 28 genotypes (Triticum spp.) grown in nutrient solution supplied with 2 microM ZnSO4 radiolabeled with 65Zn. Grain-Zn concentration differed among genotypes and ranged from 33 to 149 microg g(-1) of dry weight (DW); similarly, grain-Fe concentration varied approximately 4-fold, from 80 to 368 microg g(-1) of DW. Concentrations of Zn and Fe in the grain were positively correlated. Therefore, selecting genotypes high in grain-Zn also tends to increase grain-Fe concentration. Concentrations of myo-inositolhexaphosphate (phytate) in the wheat grain varied from 8.6 to 26.1 micromol g(-1) of DW. Grain intrinsically labeled with 65Zn was incorporated into test meals fed to Zn-depleted rats. All rats readily ate the test meals, so that Zn intake varied directly with grain-Zn concentration. As determined by the percentage of 65Zn absorbed from the test meal, the bioavailability to rats of Zn in the wheat genotypes ranged from about 60 to 82%. The amount of bioavailable Zn (micrograms) in the grain was positively correlated to the amount of Zn accumulated in the grain. There was a significant negative correlation between grain-phytate levels and percentage of Zn absorbed from the wheat grain, but the effect was not large. These results demonstrate that concentrations of Zn in whole-wheat grain, as well as amounts of bioavailable Zn in the grain, can be increased significantly by using traditional plant-breeding programs to select genotypes with high grain-Zn levels. Increasing the amount of Zn in wheat grain through plant-breeding contrivances may contribute significantly to improving the Zn status of individuals dependent on whole grain wheat as a staple food.  相似文献

To study the response of date palm trees to Fe fertilization, two date palm cultivars, Khlas and Ruzaiz, were fertilized with different levels of FeEDDHA (trunk injected and through soil) and FeSO4 · 7H2O(trunk injected). Leaves were collected 100 days after iron application and analyzed for micronutrients. The injection with 100 g FeSO4 · 7H2O/tree increased the Fe contents in the leaves of both cultivars, and with 100 g FeEDDHA/tree in the leaves of Ruzaiz cultivar. Soil application of FeEDDHA was not effective. The contents of Cu, Mn and Zn in the leaves were not affected by Fe fertilization.  相似文献

Iron deficiency is the most common nutritional disorder worldwide. Iron fortification of foods is considered to be the most cost-effective long-term approach to reduce iron deficiency. However, for fortified foods to be effective in reducing iron deficiency, the added iron must be sufficiently bioavailable. In this study, fortification of whole-grain wheat flour with different sources of iron was evaluated in vitro by measuring the amount of dialyzable iron after simulated gastrointestinal digestion of flour baked into chapatis and subsequent intestinal absorption of the released iron using Caco-2 cell layers. The dialyzability of iron from iron-fortified wheat flour was extremely low. Additions of 50 mg/kg iron to the flour in the form of ferrous sulfate, Ferrochel amino acid chelate, ferric amino acid chelate taste free (TF), Lipofer, ferrous lactate, ferrous fumarate, ferric pyrophosphate, carbonyl iron, or electrolytic iron did not significantly increase the amount of in vitro dialyzable iron after simulated gastrointestinal digestion. In contrast, fortification of flour with SunActive Fe or NaFeEDTA resulted in a significant increase in the amount of in vitro dialyzable iron. Relative to iron from ferrous sulfate, iron from SunActive Fe and NaFeEDTA appeared to be 2 and 7 times more available in the in vitro assay, respectively. Caco-2 cell iron absorption from digested chapatis fortified with NaFeEDTA, but not from those fortified with SunActive Fe, was significantly higher than from digested chapatis fortified with ferrous sulfate. On the basis of these results it appears that fortification with NaFeEDTA may result in whole-grain wheat flour that effectively improves the iron status.  相似文献

The presence of ethylenediamine-N-(o-hydroxyphenylacetic)-N'-(p-hydroxyphenylacetic) acid (o,p-EDDHA) as the second largest component in commercial EDDHA iron chelates has recently been demonstrated. Here is reported the speciation of o,p-EDDHA by the application of a novel methodology through the determination of the complexing capacity, protonation, and Ca(2+), Mg(2+), Cu(2+), and Fe(3+) stability constants. The pM values and species distribution in solution, hydroponic, and soil conditions were obtained. Due to the para position of one phenol group in o,p-EDDHA, the protonation constants and Ca and Mg stability constants have different values from those of o,o-EDDHA and p,p-EDDHA regioisomers. o,p-EDDHA/Fe(3+) stability constants are higher than those of EDTA/Fe(3+) but lower than those of o,o-EDDHA/Fe(3+). The sequence obtained for pFe is o,o-EDDHA/Fe(3+) >/= o,p-EDDHA/Fe(3+) > EDTA/Fe(3+). o,p-EDDHA/Fe(3+) can be used as an iron chelate in hydroponic conditions. A way to decrease iron and zinc deficiency in humans is to biofortify foods by increasing the bioavailable contents in these elements. The aim of this work was to study if chelating agents could be used to increase the capture of Fe and Zn by wheat grains. Zn and/or Fe in combination with the chelating agents ethylenediaminedisuccinic acid (EDDS) or ethylenediaminetetraacetic acid (EDTA) were added at various times (i.e., at flower head formation, anthesis, and postanthesis) to spring wheat ( Triticum aestivum var. Greina) grown in nutrient solution. Treatments lasted for 2 weeks, and the plants were harvested at grain maturity. The shoots of treated plants accumulated higher Zn and/or Fe concentrations than untreated plants, depending on the treatment. The plants also accumulated significant concentrations of EDDS or EDTA in their shoots. Elevated Zn and Fe concentrations in the shoots did in most cases not lead to significantly higher Zn and Fe concentrations in the grains. The grains of plants treated with EDDS during flower head formation accumulated elevated Fe and Zn concentrations but at the cost of a reduction in yield. The control plants transferred higher percentages of Fe and Zn from the shoot into the grain than the treated plants. This indicates that EDTA and EDDS inhibited in most cases the translocation of Fe and Zn from the shoots into the grains. The amounts of EDDS and EDTA found in the grains of treated plants were very small. This indicates that there was little transfer of the chelates into the symplast and that the apoplastic pathway, which is important for the transport of chelants into the shoots, is efficiently blocked between shoots and seeds.  相似文献

Heme iron has been identified in many plant sources-most commonly in the root nodules of leguminous plants, such as soy. Our objective was to test the effectiveness of soy root nodule (SRN) and purified soy hemoglobin (LHb) in improving iron bioavailability using an in vitro Caco-2 cell model, with ferritin response as the bioavailability index. We assessed bioavailability of iron from LHb (either partially purified (LHbA) or purified (LHbD)) with and without food matrix and compared it with that from bovine hemoglobin (BHb), ferrous sulfate (FeSO4), or SRN. Bioavailability of each treatment was normalized to 100% of the FeSO4 treatment. When iron sources were tested alone (100 ug iron/mL), ferritin synthesis by LHbD and BHb were 19% (P > 0.05) and 113% (P < 0.001) higher than FeSO4, respectively. However, when iron sources were used for fortification of maize tortillas (50 ppm), LHbA and BHb showed similar bioavailability, being 27% (P < 0.05) and 33% (P < 0.05) higher than FeSO4. Heat treatment had no effect on heme iron but had a significant reduction on FeSO4 bioavailability. Adding heme (LHbA) iron with nonheme (FeSO4) had no enhancement on nonheme iron absorption. Our data suggest that heme iron from plant sources may be a novel value-added product that can provide highly bioavailable iron as a food fortificant.  相似文献

研究了干旱条件下不同N、P水平对微量元素Zn、Cu、Mn、Fe有效性的影响。结果表明 ,在中壤质土壤含水量 11%~ 14%的情况下 ,不同N、P配比明显增加了土壤剖面中速效P的含量 ,但随着土壤深度的增加 ,土壤速效P呈不断下降的趋势。大田和土壤培养试验表明 ,施P有增加土壤Zn、Cu、Mn有效性的趋势 ,但有效Fe则有下降的趋势。土壤速效P、Zn、Cu、Mn、Fe之间的相关分析表明 ,土壤速效P与有效Zn之间存在显著的正相关关系 (r =0.65) ,而与有效Fe之间则存在显著的负相关关系 (r =- 0.63)。  相似文献

Iron (Fe) plaque on roots of rice was found to lessen the toxic effects of zinc (Zn). With excess Zn, plants with Fe plaque had significantly greater dry mass of roots and foliage than plants without plaque. In the excess of Zn, plants with Fe plaque had higher concentrations of Fe but lower Zn than plants without plaque, although the differences were not significant. In the excess of copper (Cu) or nickel (Ni), plants with Fe plaque had significantly lower concentration of phosphorus (P) in leaves than plants without plaque. It is suggested that the low concentration of P in leaves of plants with plaque will increase the concentration of “active iron”; inside leaves, and therefore, increase their tolerance against heavy metal toxicity.  相似文献

Abstract

Soil extraction techniques to measure the status of available micronutrients for plants are important in the diagnosis of deficiency or toxicity. Mehlich 3 (M3), EDTA (pH=8.2), DTPA‐TEA, and Soltanpour and Schwab (SS) solutions were confronted for their ability to extract simultaneously copper (Cu), zinc (Zn), manganese (Mn), and iron (Fe). Argentinean soils from different taxonomic orders with widely varying properties were investigated. The values obtained showed that DTPA‐TEA and SS solutions extracted similar amounts of Zn, Fe, and Mn, while EDTA dissolved comparatively higher amounts of Fe and Mn. Mehlich 3 yielded the highest extractions for the four micronutrients. Soil pH not only affected the extraction of Mn by DTPA‐TEA, SS, and EDTA extractions, but also the extraction of Fe by EDTA. The organic carbon affected the determination of Fe and Zn in all cases. The correlations of the different tests for Cu, Zn, Mn, and Fe were significant. The results suggest that for the determination of the bioavailable status of micronutrients, any of the studied tests could be applied using the soil edaphic properties as factors to improve the correlations between them and standardize the methods.  相似文献

The reduced iron powder has considerable potential for use as an iron fortificant because it does not change organoleptically during storage or food preparation for cereal flour, and its bioavailability is scarcely influenced by iron absorption inhibitors in foods. The objective of this article is to study the effects of ascorbic acid, phytic acid, and pH on iron uptake from reduced iron powder (43 microm) and FeSO 4, and to compare iron bioavailability of reduced iron powders among four selected granularity levels. The cell ferritin formation is used as a marker of iron uptake. Obviously, iron uptake of reduced iron powder is increased with decreasing of powder granularity and is much lower than FeSO 4 when the size is above 43 microm, but significantly higher at 40-60 nm. In the presence of ascorbic acid or phytic acid, Caco-2 cell iron absorption from reduced iron powder (43 microm) is significantly higher than that from FeSO 4. And iron uptake of Caco-2 cells is decreased with increasing of pH from 5.5 to 7.5. Moreover, the decrease trend is more obvious for reduced iron powder than for FeSO 4. Our results indicated that iron bioavailability of reduced iron powder by intestinal enterocytes is similar to that of iron salts, and reduced iron powder is more excellent than FeSO 4 as food fortificant, especially at ultramicroscopic granularity.  相似文献
